Crystals of alkaline phosphatase (EC 3.1.3.1; Mr 94,000) grown at pH 9.5 from 2.25 m-(NH4)2SO4 with 5 × 10?5 m-Zn and 10?2 m-Mg present were analyzed by X-ray diffraction at pH 7.5 in 2.66 m-(NH4)2SO4 with 10?2 m-Zn and 10?2 m-Mg present. The crystals are orthorhombic with a = 195.5 A?, b = 168.3 A?and c = 76.33 A?, and the space group is I222. X-ray phases were determined by the multiple isomorphous replacement and anomalous dispersion method using K2PtCl4, KAu(CN)2 and K2OsO4 derivatives. The electron density maps and analysis of metal binding sites reveal one molecule per asymmetric unit with an internal, non-crystallographic, 2-fold rotation axis relating the subunits. Each subunit contains a major αβ domain with a seven-stranded β-sheet flanked by helices. The sheets are roughly coplanar but the general direction of the strands in each is at 20 ° to the rotation axis and thus 40 ° from each other. The helical content of the αβ domain is approximately 27% of the 459 residues in the monomer and the β content is approximately 7%. The chains in a smaller domain are more convoluted and less easily characterized than in the αβ domain. In both there is extensive monomer-monomer contact.Removal of the zinc and magnesium from the parent crystal produces a stable apoenzyme crystal and addition of cobalt at 10?2 m or cadmium at 10?2 or 5 × 10?2 m reveals seven metal binding sites per dimer. The active centers are 32 Å apart and each is shown by anomalous dispersion data to contain two metal binding sites, A and B. The cadmium derivative refinement determined the A-B separation to be 4.9 Å. Comparison of the parent and apo structures by means of difference maps reveals the double metal site with Zn at A and probably Mg at B. A prominent, partially resolved peak centered 7 Å away is interpreted as a stabilization of the backbone in this position by the metal ion co-ordination to a side-chain. Several negative peaks within 10 Å of the metals indicate local differences between apo and native structures but no significant differences are seen in the other parts of the molecule. At 5 × 10?2 m-Cd two metal sites (D and D′) are found 25.5 Å from the active center, on the surface of the minor domain. They are related to each other by the molecular 2-fold axis with a D-D′ distance of 25 Å. The seventh Cd site, E, is 20 Å from the active center, on the major domain, near a crystalline contact region, and devoid of any molecular symmetry mate.The apparent dissociation constants for cadmium at the A, B and D sites (and A′, B′, D′) are 3 × 10?3 m, 1.5 × 10?1 m and 1.3 × 10?2 m, respectively. Thus in these conditions cadmium is seen to distribute between A and B sites when the combined stoichiometry is two metal ions per dimer.  相似文献

2-Deoxy-2,3-dehydro-N-acetylneuraminic acid and its methyl ester are competitive inhibitors of Arthrobacter sialophilus neuraminidase with Ki = 1.4 × 10?6M and 4.8 × 10?5M, respectively. The Km for the substrate, N-acetylneuraminlactose, is 1.0 × 10?3M. These data, taken together with the conformation of these compounds, indicate that these compounds are transition-state analogs of the enzyme. These results also suggest that the substrate upon binding to neuraminidase is distorted to a conformation approaching that of a half-chair.  相似文献

5-Aminolevulinic acid dehydratase (ALAD) from bovine liver contains zinc that is partially lost during the isolation of the enzyme. ALAD has its maximal activity at 10?5 M ZnCl2. It binds 7.4 Zn per octameric protein with an association constant of 5.3 × 106 M?1. ALAD is inactivated by 1,10-phenanthroline or ethylenediaminetetraacetic acid (EDTA) but not by monodentate anions like cyanide or sulfide. After removal of zinc by chelating agents, the enzyme activity may be restored by Zn2+ or Cd2+. Removal or zinc by EDTA increases KM 60-fold and decreases Vmax to about 12 of its original value. The 113Cd nuclear magnetic resonance spectrum of the enzyme reconstituted with 113Cd-acetate exhibits a single sharp resonance signal at 79 ppm. It does not change by the addition of substrate but disappears when the inhibitor lead acetate is added. Therefore, an immediate interaction between the metal ion of the enzyme and the substrate is excluded, whereas lead changes the environment of cadmium and probably of zinc too.  相似文献

An ATPase is demonstrated in plasma membrane fractions of goldfish gills. This enzyme is stimulated by Cl? and HCO3?, inhibited by SCN?.Biochemical characterization shows that HCO3? stimulation (Km = 2.5 mequiv./l) is specifically inhibited in a competitive fashion by SCN? (Ki = 0.25 mequiv./l). The residual Mg2+-dependent activity is weakly is weakly affected by SCN?.In the microsomal fraction chloride stimulation of the enzyme occurs in the presence of HCO3? (Kmfor chloride = 1 mequiv./l); no stimulation is observed in the absence of HCO3?. Thiocyanate exhibits a mixed type of inhibition (Ki = 0.06 mequiv./l) towards the Cl? stimulation of the enzyme.Bicarbonate-dependent ATPase from the mitochondrial fraction is stimulated by Cl?, but this enzyme has a relatively weak affinity for this substrate (Km = 14 mequiv./l).  相似文献

Mammalian glutamine-dependent carbamoyl phosphate synthetase (EC 2.7.2.9), the first enzyme of de novo pyrimidine nucleotide biosynthesis, was strongly inhibited by polyamines at concentrations of 10?4 to 10?3 M. Spermine was the most effective, followed in order by spermidine and putrescine. The inhibition was partially reversed by increasing the concentration of Mg2+ or MgATP2?, or by adding low concentrations of 5-phosphoribosyl 1-pyrophosphate, an allosteric activator of the enzyme. Polyamines increased the apparent Ka value of the enzyme for phosphoribosyl pyrophosphate. A possible physiological role of polyamines in widening the range of the effective concentrations of phosphoribosyl pyrophosphate as the activator for the enzyme is suggested.  相似文献

The addition of glucagon (10?6 M) to an incubation mixture containing 32Pi and hepatocytes isolated from livers of rats fed ad libitum results in both a 3-fold increased incorporation of 32P into L-type pyruvate kinase and a decreased catalytic activity. The 32P incorporated into pyruvate kinase was covalently bound to the enzyme as evidenced by polyacrylamide gel electrophoresis in sodium dodecyl sulfate. In addition, exogenous cyclic AMP (10?3 M) stimulated the phosphorylation and the suppression of catalytic activity to a similar extent. On the other hand, insulin (10?7 M) had essentially no effect on the incorporation of 32P into pyruvate kinase or on its catalytic activity under the conditions used in this study. These results suggest that phosphorylation of pyruvate kinase invivo is stimulated by glucagon via cyclic AMP and cyclic AMP-dependent protein kinase and that the activity of the enzyme is, at least in part, regulated by a phosphorylation-dephosphorylation mechanism.  相似文献

The rhabdomeres of cephalopod photoreceptors, which are built up mainly of rhodopsin and phospholipid molecules, show a very high alkaline phosphatase activity. The enzyme has been partially characterized in purified rhodopsin vesicle fractions of the rhabdomeres by the following kinetic data: pH optimum 8.7; activation energy 9100 cal·m?1; Vmax = 2.5 μmol·min?1·mg?1; Km = 1.5·10?4M; its activity depends on Mg2+. There is good evidence that the alkaline phosphatase is a membrane-bound enzyme with receptor sites presumably located on the inside of the membrane. This enzyme has not been purified but its high activity compared to that of other known alkalin phosphatases (see Table I) indicates that each mirovillus, the structural unit of the rhabdomere, contains 1–20 enzyme molecules. This finding supports the hypothesis that the alkaline phosphatase is involved in the biochemical amplification process of excitation, or adaptation.  相似文献

Griseofulvin—shown previously to disrupt the mitotic apparatus in vivo—inhibited the in vitro microtubule assembly reaction completely at 8 × 10?4M griseofulvin. In a gel filtration assay, randomly tritiated griseofulvin associated stoichiometrically with purified tubulin, as determined by chromatography on Sephadex G-25. No detectable drug binding was observed when bovine serum albumin was used as a control in an identical column assay. Both gel filtration chromatography and a kinetic analysis of the inhibition of assembly by griseofulvin suggest that the drug interacts directly and stoichimetrically with the tubulin dimer, and that the interaction is both rapid and independent of temperature.  相似文献

myo-Inositol:NAD(P)+ oxidoreductase (myo-inositol oxidoreductase) has been identified in bovine brain. This enzyme elutes from DEAE cellulose with 0.3 M KCl in 50 mM Tris buffer, pH 7.5. Using NADH as cofactor myo-inosose-2 is reduced selectively to myo-inositol. With NADPH the enzyme forms both myo-inositol and scyllo-inositol, however, at a lower rate. The enzyme was chromatographed on G-100 Sephadex and found to have an apparent molecular weight of 74,000. This enzyme differs in DEAE binding, molecular weight and cofactor specificity from the previously described scyllo-inositol oxidoreductase which utilizes NADPH exclusively to produce 3 fold more scyllo-inositol than myo-inositol.  相似文献

The ATPase enzyme system from two-spotted spider mites, Tetranychusurticae (Koch) was sensitive, in vitro, to four acaricides. Tricyclohexylhydroxytin (PlictranR) was an outstanding inhibitor of oligomycin-sensitive (mitochondrial) Mg2+ATPase from fish brain and spider mite homogenates. The I50 values were 6.6×10?11M and 6.2×10?10M, respectively. Less effective were chlorbenside, chlorfenethol and ovotran. Plictran at a higher concentration (2×10?7M) was also more effective on Na+-K+ATPase both in mites and fish brain homogenates as compared to chlorfenethol, chlorbenside and ovotran. Plictran inhibited oligomycin-insensitive Mg2+ATPase at concentrations of 10?8M but stimulated at high concentrations (5×10?6M and higher).  相似文献

Vanadate inhibits the Ca++-ATPase of sarcoplasmic reticulum from pig heart half maximally at about 10?5 M. Mg++ promotes this inhibition by vanadate whereas increasing Ca++-concentrations protect the enzyme against vanadate inhibition. Keeping the ratio Mg++ATP constant there was no influence of ATP on the vanadate inhibition at concentrations up to 5 × 10?3 M ATP. Whenever the ratio Mg++ATP was higher than 1:1 the inhibitory effect of vanadate on the Ca++-ATPase was increased.  相似文献

S-Adenosylhomocysteine (SAH) is a potent product inhibitor for indoleamine-N-methyltransferase (INMT) from rabbit lung. The kinetic studies showed that this inhibition was competitive with respect to S-adenosylmethionine (SAM) and noncompetitive with respect to N-methylserotonin (NMS). The Ki value of 1.0 × 10?5M indicated that SAH had a higher affinity than SAM or NMS for the enzyme. SAH seems to form a reversible complex with the enzyme.  相似文献

The interaction between hydrogen peroxide and oxidized Rhusvernicifera laccase from which the type 2 copper has been removed, was investigated. For that end, the circular dichroic spectrum of the modified enzyme has been measured in the presence of increasing concentrations of hydrogen peroxide. The characteristic band observed upon binding peroxide to native laccase is also observed for the type 2 copper depleted enzyme. However, there are several quantitative differences in the latter one. First, the intensity is lower and band width is larger. Secondly, from the titrations, it becomes apparent that the affinity for H2O2 is markedly lower than that of the native enzyme. While the affinity for the native enzyme is higher than 108 M?1, it decreases to 1·104 M?1 for the type 2 depleted enzyme.  相似文献

Vasoactive intestinal peptide (VIP), secretin, catecholamines and prostaglandin E1 (PGE1) in the presence of a cyclic nucleotide phosphodiesterase inhibitor stimulate the accumulation of cyclic AMP in two colorectal carcinoma cell lines (HT 29 and HRT 18) with subsequent activation of the cyclic AMP-dependent protein kinases. In HT 29 cells incubated without phosphodiesterase inhibitor, 10?9 M VIP promotes a rapid and specific activation of the low Km cyclic AMP phosphodiesterase (1.7-fold); at 25°C the effect is maintained for more than 15 min, while at 37°C the activity returns to basal value within 15 min. As shown by dose-response studies, VIP is by far the most effective inducer (Ka = 4 · 10?10M) of the cyclic AMP phosphodiesterase activity; partial activation of the enzyme is obtained by 3 · 10?7 M secretin, 10?5 M isoproterenol and 10?5 M PGE1; PGE2 and epinephrine are without effect. In HRT 18 cells VIP is less active (Ka = 2 · 10?9M) whereas 10?6 M PGE1, 10?6 M PGE2 and 10?5 M epinephrine are potent inducers of the phosphodiesterase activity. The positive cell response to dibutyryl-cyclic AMP further indicates that cyclic AMP is a mediator in the phosphodiesterase activation process. The incubation kinetics and dose response effects of the various agonists on the cyclic AMP-dependent protein kinase activity determined for both cell types in the same conditions show a striking similarity to those of phosphodiesterase. Thus coordinate regulation of both enzymes by cyclic AMP was observed in all incubation conditions.  相似文献

The present communication describes a comparative study of some enzymatic properties of an early and a late aconitase (EC.4.2.1.3) present in Bacilluscereus T cells of 5 and 12 hr culture age, respectively. The activity of both enzymes increased linearly with increase in enzyme concentration. They demonstrated similar pH *7.5) and temperature (30 C) optima, but differed in their activation energy and affinity for substrate. Late aconitase had higher activation energy (16,100 cal) as compared to early aconitase (9,200 cal). Early aconitase showed a Km value of 100 × 10?4M for sodium citrate and 33.3 × 10?4M for isocitrate. Late aconitase exhibited 5 to 7 times greater affinity for citrate and isocitrate yielding Km values 14 × 10?4M and 7 × 10?4M, respectively. On the basis of available evidence, it is suggested that early and late aconitase present in 5 and 12 hr aged cells of Bacilluscereus T behave as isozymes, and may be designated as aconitase (EC.4.2.1.3) isozyme I and aconitase (EC.4.2.1.3) isozyme II, respectively. The significance of their plausible role during growth and sporulation has been discussed.  相似文献

Varying doses of morphine sulfate (10, 20 or 40 mg/kg daily × 10) were observed to suppress metabolic activities in the mouse prostate gland. Prostate gland fructose, an index of androgenic activity, was significantly reduced by these dose regimes of morphine (P < 0.01). Injections of morphine sulfate (20 mg/kg daily × 10) led to an inhibitition in the in vitro synthesis of both fructose?14C and sorbitol?14C from glucose?14C by the prostate gland, part of which may have been due to decreased uptake of glucose by the gland. The in vitro assimilation of 2-deoxyglucose?14C by the prostate was also reduced by morphine treatment. The in vitro actions of morphine (2 × 10?3M) on the metabolism of radioactive glucose by the mouse prostate gland likewise revealed a significant reduction in the formation of sorbitol?14C, but no decrease in fructose?14C formation. These results indicate that both the in vitro and in vivo actions of morphine can inhibit fructose metabolism in the prostate gland.  相似文献

Substitution of the active site zinc ion of carboxypeptidase A by cadmium yields an enzyme inactive towards ordinary peptide substrates. However, a substrate analog (BzGlyNHCH2CSPheOH) containing a thioamide linkage at the scissile position is cleaved to the thioacid. The kinetic parameters and their pH dependencies are kcatKm = 5.04 × 104 min?1M?1, decreasing with either acid or base (PKE1 = 5.64, pKE2 = 9.55), and kcat = 1.02 × 102 min?1, decreasing with acid (pKES = 6.61). The thiopeptide is less efficiently cleaved by native (zinc) carboxypeptidase A. This cadmium-sulfur synergism supports a mechanism wherein the substrate amide is activated by metal ion coordination to its (thio) carbonyl.  相似文献

We have recently demonstrated that gonadotropin releasing hormone (GnRH) acts directly on ovarian granulosa cells to inhibit the follicle stimulating hormone (FSH)-induced increase in granulosa cell steroidogenesis invitro. A GnRH antagonist, [D-pGlu1, D-Phe2, D-Trp3,6] GnRH (A), which is known to antagonize GnRH-stimulated gonadotropin release by cultured pituitary cells, was tested in the granulosa cell system. GnRH (10?8M) inhibited estrogen and progesterone production by FSH-treated granulosa cells invitro, whereas the antagonist A (10?6M) did not affect FSH stimulation of steroidogenesis. Antagonist A, when added together with GnRH and FSH, blocked the GnRH inhibition of FSH-induced steroidogenesis. Estrogen and progesterone production by granulosa cells was increased by 50% at a molar ratio (IDR50) of 201and121 ([antagonist]/[GnRH]), respectively. At 10?6M, antagonist A completely prevented the GnRH (10?8M) inhibition. A similar effect of antagonist A was seen in FSH-induced increase of luteinizing hormone (LH) receptor content. FSH treatment for 2 days invitro induced an 8-fold increase in LH receptor content in cultured granulosa cells; concomitant treatment with 10?8M GnRH completely inhibited the FSH effect. Antagonist A (10?6M), by itself, had no effect on the FSH action. However, when added together with FSH and GnRH, antagonist A completely abolished the inhibitory effect of GnRH. These results demonstrate that the direct inhibitory effect of GnRH on granulosa cell function can be prevented by a GnRH antagonist and that the GnRH action at the ovarian level may require stringent stereospecific interactions of these peptides with putative GnRH recognition sites.  相似文献

Material released from rat mast cells by compound 4880 gave parallel responses using ACTH and β-endorphin radioimmunoassays. However, incubation of these labeled compounds under conditions of radioimmunoassay with released material and chromatography on Sephadex G-25 provided evidence that neither ACTH nor β-endorphin were present in the material released from mast cells, but represented an artifact produced by the presence of a protease. Analysis of the released enzyme on Sephadex G-75 under non-dissociative conditions yielded an active enzyme complex with a Mr > 150,000. Under dissociative conditions, the Mr of the enzyme was 25,000. The dissociated enzyme reassociated with purified rat mast cell heparin to form the high molecular weight complex. Further investigation of pH, substrate and inhibitor specificity showed that the peptide degradation is due to a chymotrypsin-like protease, the previously described mast cell chymase, which is active in degrading β-endorphin, ACTH, and ACTH1–24.  相似文献
