二甲戊灵对离体大蒜染色体的加倍研究   总被引:2,自引:0,他引:2  
在MS分化培养基中分别添加不同浓度的秋水仙素和二甲戊灵,以 "改良蒜" 的蒜瓣基部为外植体诱导愈伤组织染色体变异并再生植株,观察根尖细胞染色体数和叶片下表皮保卫细胞特征检测诱变效果.结果表明:大蒜染色体数变异受诱变剂浓度和处理持续时间的影响,诱变剂高浓度和短时间处理均有利于大蒜愈伤组织的分化;添加0.1%秋水仙素处理5 d,愈伤组织存活率为80.7%,分化率为78.1%,四倍体的诱导率为4%;添加 100 μmol/L二甲戊灵处理5 d,愈伤组织存活率为100%,四倍体的诱导率为6%.对根尖细胞染色体和叶片下表皮气孔数目和大小观察显示,诱变株具有四倍体的基本特征.说明二甲戊灵能在离体条件下有效诱导大蒜产生四倍体,可替代秋水仙素对大蒜染色体的加倍作用.  相似文献

用不同浓度秋水仙素处理野生南荻×芒(Miscanthus lutarioriparia×Miscanthus sinensis)远缘杂交后代以诱导产生多倍体,并对变异株进行形态学和细胞学鉴定,以期获得稳定的四倍体植株并分析其生理特性。结果表明:(1)采用秋水仙素加入培养基处理法和秋水仙素溶液浸泡处理法都可获得一定频率的多倍体植株;胚性愈伤组织以0.2%秋水仙素浸泡处理48h的诱变效果较好,四倍体诱导率达8.7%;芽在0.05%秋水仙素培养基中处理15d较好,四倍体诱导率达10.6%;生根苗在0.1%秋水仙素培养基中处理10d较好,四倍体诱导率达11.1%。(2)经体细胞染色体计数,加倍植株染色体数为2n=4x=76,对照植株的染色体数目为2n=2x=38。(3)生长2年的多倍体植株形态、叶片大小、茎粗、茎壁厚、节间等性状表现出巨大性和超亲优势。  相似文献

对二倍体甜荞长花柱类型 (ES2 s)、二倍体甜荞短花柱类型 (ES2 S)、四倍体甜荞长花柱类型 (ES4s)、四倍体甜荞短花柱类型 (ES4S)、四倍体有翅细野荞 (GR4HI)、四倍体无翅细野荞 (GR4HO)等 6个不同荞麦类型花的花粉形态学进行了电镜观察。结果表明 :这些荞麦类型的花粉粒都是椭圆形 ,都有网状纹饰、3孔沟等特征。二倍体甜荞染色体加倍后形成的四倍体甜荞 ,相对于二倍体甜荞而言 ,花粉粒显著增大和明显变圆。无论二倍体还是四倍体甜荞 ,其长花柱型花的花粉粒都比其对应短花柱型花的花粉粒要小。尽管四倍体甜荞和四倍体细野荞在染色体数目和倍性上一样 ,但是它们在花粉粒的大小上有显著差异。四倍体细野荞有翅类型和无翅类型的花粉粒大小和形态差异不显著  相似文献

采用组织培养方法研究了秋水仙素处理对大花蕙兰类原球茎增殖、分化和四倍体诱导的效应.结果表明,秋水仙素处理明显抑制类原球茎的增殖和分化,提高类原球茎褐变率和死亡率;在试验浓度和时间范围内,秋水仙素浓度越高,处理时间越长,抑制作用越强,解除抑制作用所需的继代次数越多.所有秋水仙素处理均能诱导出四倍体,但不同处理的四倍体诱导率不同,当处理浓度为0.05%、时间为5 d时,四倍体诱导率最高,为23.7%.二倍体及其四倍体的气孔保卫细胞长度和气孔密度均存在极显著的差异.四倍体植株比二倍体矮,茎部较粗壮,株型紧凑,叶片颜色浓绿、质地变硬.秋水仙素处理和组织培养相结合是创建大花蕙兰多倍体的有效方法.  相似文献

以秋水仙素人工诱变获得的不结球白菜同源四倍体Pol CMS及其保持系为材料,采用石蜡切片法研究其花药发育过程.结果显示:四倍体Pol CMS与保持系花药发育差异明显,不育系花药发育受阻于孢原细胞分化期,不形成药室,属无花粉囊型.极少数温敏型花药能产生1~2个花粉囊,但四分体时期绒毡层提早退化,导致成熟小孢子数目减少.四倍体保持系花药发育过程与二倍体基本一致,同一时期的可育小孢子四倍体比二倍体大,二倍体花粉粒都为3个萌发孔,而四倍体多为4个萌发孔.四倍体保持系同一花粉囊中花粉粒发育有不同步现象,花粉粒败育频率比二倍体高.研究表明,不结球白菜同源四倍体Pol CMS比其二倍体Pol CMS不育更彻底,更具应用性.  相似文献

多花黑麦草二倍体种,具有14条染色体。用普通小麦与多花黑麦草杂交,获得了杂种,杂交结实率为0.058%。杂种F1表现为畸形,其体细胞染色体数为28,F1植株产生的花粉败育,雌蕊不孕,用普通小麦和多花黑麦草回交均未获得回交种子,有待于用秋水仙素加倍染色体以获得异源四倍体。  相似文献

该研究以二倍体紫菜薹为材料,采用浓度为0.1%、0.2%和0.3%的秋水仙素溶液,在二倍体紫菜薹生长至子叶期时对植株茎尖生长点进行4次点滴处理,鉴定筛选同源四倍体紫菜薹并进行二倍体和四倍体紫菜薹的营养品质比较。结果表明:(1)使用浓度为0.2%秋水仙素溶液点滴4次对紫菜薹的处理效果最好,四倍体紫菜薹的诱导率为6.62%。(2)在形态学上,四倍体植株的叶片、花簇、花器官、角果和种子与二倍体相比都在巨大性上呈现出明显差异;在解剖学上,四倍体植株在气孔密度减小的同时气孔变大,其花粉表现出矩形、梯形以及其他一些不规则的形状;在细胞学上,四倍体和二倍体植株的染色体数目分别为40条和20条;流式细胞仪鉴定结果显示,四倍体植株的DNA分子荧光强度(2 092 385.03)约为二倍体植株(956 725.15)的2倍。(3)植株营养品质方面,四倍体紫菜薹的游离氨基酸、叶绿素总浓度分别比二倍体植株显著增加了228.58%、110.02%,但四倍体紫菜薹的硝态氮、可溶性蛋白、维生素C、可溶性糖、纤维素总含量分别比二倍体植株显著减少了48.99%、43.20%、45.81%、44.50%、59.97%。(4)与二倍体植株相比,四倍体紫菜薹的单株质量、十叶厚、叶宽、叶柄宽都有不同程度的增加,叶片开展度有一定程度的降低。研究发现,秋水仙素诱导产生四倍体紫菜薹的最适浓度为0.2%;四倍体紫菜薹表现出明显的高产性,但多数营养品质极显著低于二倍体紫菜薹。  相似文献

以胀果甘草成熟种子为材料,消毒后在培养皿中萌动处理12 h、24 h、36 h,用浓度为0.05%、0.075%、0.10%的秋水仙碱处理12 h、24 h、36 h,来进行四倍体的诱导,并对诱导处理后获得的再生植株进行染色体鉴定。结果表明:种子萌动处理24 h后,用浓度为0.075%的秋水仙碱溶液处理24 h诱导效果最好,变异率为41%。其体细胞染色体数目由2n=2x=16变为2n=4x=32。  相似文献

以中国南瓜种‘上海小闸黄狼’南瓜为试材,用秋水仙素为诱变剂,对‘黄狼’种子进行处理,得到了同源四倍体南瓜。在4种浓度处理4h后,发现获得同源四倍体的最佳浓度为0.2%和0.3%。变异材料染色体数加倍(2n=4x=80)。研究发现,与二倍体相比,四倍体单位面积叶面气孔数变少,叶色变深,叶裂变浅,叶尖变钝,叶变大,茎变粗,大量出现畸形雄花,果实变小,果形变直,果柄变短,单瓜种子平均数由340粒减为68粒,抗早衰性更强,脯氨酸含量和过氧化物歧化酶活性更高,丙二醛含量更低。四倍体南瓜伤流量明显比二倍体高,根系活力强于二倍体。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
