Firefly luciferase can catalyze the formation of fatty acyl-CoA via fatty acyl-adenylate from fatty acid in the presence of ATP, Mg2+ and coenzyme A (CoA). A long chain fatty acyl-CoA (C16–C20), produced by luciferase from a North American firefly (Photinus pyralis) and a Japanese firefly (Luciola cruciata), was isolated and identified by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry analysis. Of a number of substrates tested, linolenic acid (C18:3) and arachidonic acid (C20:4) appear to be suitable for acyl-CoA synthesis. This evidence suggests that firefly luciferase within peroxisomes of the cells in the photogenic organ may be a bifunctional enzyme, catalyzing not only the bioluminescence reaction but also the fatty acyl-CoA synthetic reaction.  相似文献

The characteristic surface lipid compositions of several C3 and C4 plants are discussed. C4 plants produce surface lipids (epicuticular waxes) made up of the ubiquitous classes of aliphatic compounds: free fatty acids, aldehydes, primary alcohols, alkanes and aliphatic linear esters. C3 plants synthesize surface lipids comprising the ubiquitous classes and either of the two following groups of compound: (i) lβ-diketones, hydroxy lβ-diketones, alkan-2-ol esters; (il) ketones and secondary alcohols with the functional group in the middle of the hydrocarbon chain. These features are suggested to represent physioIogical characteristics of the plant and to be related to ecological adaptations. Wax class compositions might also be an ancillary method for defining the C3 or C4 mechanism of CO2 assimilation in cases where uncertainty exists.  相似文献

A rapid and sample procedure was developed to determine, by gas-liquid chromatography, the concentrations of C2---C4 alcohols, C2---C6 volatile fatty acids (VFA) and lactic acid or 2,3-butanediol in fermentation liquids. both lactic acid and 2,3-butanediol are oxidized to acetaldehyde by periodic acid and acetaldehyde was eluted before ethanol. A complete separation of the alcohols and acids was performed in <15 min on a column packed with 80/100 Chromosorb WAW, having GP 10% SP-1200/1% H3PO4 as the liquid phase. The method was suitable for the analysis of rumen fluid and fermentation products from microbial cultures. The detection limits for all compounds were <0.13 nmol · injection−1.  相似文献

为了研究许氏平鲉(Sebastes schlegelii)雌鱼体内受精后仔鱼开口前和仔鱼开口后两个阶段氨基酸和脂肪酸的组成变化规律, 采用生化常规方法定量测定并分析了许氏平鲉发育早期的受精卵(FE)、胚胎期(ES)、初产仔鱼(PL1)、前仔鱼期(PL2)、后仔鱼期(PL3)和稚鱼期(J)6个阶段的氨基酸和脂肪酸组成特点及含量变动。结果表明: 总氨基酸含量从FE至PL1显著下降, 至PL3显著上升, 至J又显著下降(P<0.05); 游离氨基酸含量以FE最低(12.77 mg/g), 从FE到PL1显著上升(P<0.05), 并在PL1含量达到最高值(92.19 mg/g), PL1发育到J呈现先下降后上升再下降的变化趋势(P<0.05), 游离氨基酸与总氨基酸的比值范围在2.37%—19.66%。在各发育阶段干样中检出碳链长度在C14-C24的29种脂肪酸, 分别为9种饱和脂肪酸(SFA)、9种单不饱和脂肪酸(MUFA)和11种多不饱和脂肪酸为(PUFA), 受精卵中主要脂肪酸依次为C22:6n-3(DHA)、C18:ln-9c、C16:0和C20:5n-3(EPA)。胚胎期(FE-ES)的脂肪酸利用率顺序为SFA、MUFA、n-6PUFA、n-3PUFA, 主要以C18:3n-3、C18:0、C16:1n-7及C20:5n-3(EPA)作为胚胎期的能量来源, C22:6n-3(DHA)的实际利用率最低(9.71%), 被优先保存下来, C16:0的实际利用量最高(10.94mg/g); 仔鱼内源营养阶段(ES-PL1)脂肪酸利用率顺序为MUFA、n-6PUFA、SFA、n-3PUFA, 主要以C16:1n-7、C18:0、C20:4n-6(ARA)及C18:1n-9c作为开口前仔鱼的主要能量来源, 其中仔鱼对DHA实际利用量最高(18.23 mg/g)。PL1-PL3阶段DHA相对于EPA和ARA被选择性利用; PL3至J阶段ARA相对于EPA和DHA被选择性利用。研究表明: 许氏平鲉仔鱼开口前阶段总氨基酸含量与游离氨基酸含量的变化趋势截然相反, 胚胎期与仔鱼内源营养阶段脂肪酸利用率和利用量均有所不同, 仔鱼期DHA优先被利用, 过渡至稚鱼期ARA优先被利用。建议在仔鱼开口后添加富含DHA生物性饵料, 仔鱼过渡到稚鱼期在配合饵料中添加ARA营养物质, 防止苗种营养不足, 保证成活率。  相似文献

The objective of this study was to determine whether the conversion of free, very long chain fatty acids (C22–C26) to their CoA-esters are involved in cerebroside synthesis, since cerebrosides are uniquely rich in very long chain fatty acids including lignoceric acid (C24:0). We have studied lignoceroyl-CoA synthetase activity in the microsomes isolated from normal and jimpy mouse brain. The jimpy mouse lacks the ability to make myelin and is deficient in enzyme activities involved in the synthesis of myelin components, including cerebrosides. Unexpectedly, the lignoceroyl-CoA synthetase activity in jimpy brain microsomes was slightly higher than that in control microsomes. The palmitoyl (C16:0)-CoA synthetase activity in jimpy brain was not different from the control. The level of cerebrosides in microsomes was grossly lower in jimpy brain. The implication of these findings and the involvement of lignoceric acid activation in cerebroside synthesis is discussed.  相似文献

The effect of both endogenous and exogenous unsaturated free fatty acids on manganese release from chloroplasts of chill-resistant (spinach) and chill-sensitive (tomato, bean) plants was studied. The level of endogenous free fatty acids increased 2–3-fold during cold and dark storage of leaves of chill-sensitive plants and was accompanied by depletion of about 60% of total chloroplast manganese content. Similar effects were observed when accumulation of free fatty acids in chloroplasts was achieved by storage of growing tomato plants for a few days in the dark at room temperature. In contrast, the cold and dark treatment of leaves of chill-resistant plant (spinach) affected neither free fatty acid, manganese levels nor Hill-reaction activity in chloroplasts. Incubation of chloroplasts of both chill-sensitive and chill-resistant plants with bean leaf galactolipase resulted in an accumulation of free fatty acids and a release of approx. 60% of total manganese content. The same amount of total manganese content was released following 3 h incubation of chloroplasts with linolenic acid at fatty acid/chlorophyll ratio (w/w, 2:1–10:1). The efficiency of C18 unsaturated fatty acids/linolenic, linoleic, oleic on manganese release from chloroplasts was established in decreasing order C18:3 > C18:2 > C18:1. The results indicate that the inhibitory effect of both endogenous and exogenous fatty acids on Hill reaction depends on the release from chloroplasts of functionally active, loosely bound manganese. Thus, similarly to both Tris and hydroxylamine treatments of chloroplasts, the incubation of chloroplast preparations with unsaturated fatty acids may be a useful tool for manganese depletion of chloroplasts.  相似文献

A chlorogenate hydrolase (EC 3.1.1.42) synthesized 2-phenylethyl caffeate (2-CAPE) from 5-chlorogenic acid (5-CQA) and 2-phenylethyl alcohol (2-PA) (by transesterification), from 5-CQA and 2-phenylethyl bromide (2-PBr) (by substitution of bromine), and from caffeic acid (CA) and 2-PA or 2-PBr (by condensation) as well as hydrolysis of 5-CQA. Some reaction conditions including pH, temperature, substrate and solvent concentrates, and reaction time were optimized for the production of 2-CAPE. A maximal molar yield of 50% was achieved by transesterification, 4.7% by substitution of bromine, and 13% by condensation. Among the parameters studied for optimization, the pH of the buffer solution and concentration of 2-PA or 2-PBr affected the production of 2-CAPE. The optimum pH for the hydrolysis reaction was within the neutral range (pH 6.5), whereas the residual three reactions were only catalyzed within the acidic range (pH 3.0–4.0). The optimum concentrations of 2-PA and 2-PBr for three reactions were 5–70 vol% and no 2-CAPE was produced in the 2-PA or 2-PBr solutions containing powdered enzyme. The enzyme may bind to the caffeoyl moiety of 5-CQA or CA to form an enzyme–substrate complex. It then catalyzes four different reactions corresponding to the reaction conditions.  相似文献

The activity of lipases from Rhizopus delemar, Rhizopus arrhizus, and Penicillium simplicissimum entrapped in microemulsions formulated by bis-(2-ethylhexyl)sulfo-succinate sodium salt (AOT) in isooctane has been studied in esterification reactions of various aliphatic alcohols with fatty acids. The effect of the nature of the fatty acids (chain length) and of the alcohols (primary, secondary, or tertiary; chain length; cyclic structures) on the lipase activities was investigated in relation to the reverse micellar structure. The lipases tested showed a selectivity regarding the structure of the substrates used when hosted in the AOT/isooctane microemulsion systems. Penicillium simplicissimum lipase showed higher reaction rates in the esterification of long chain alcohols as well as secondary alcohols. Primary alcohols had a low reaction rate and tertiary a very slow rate of esterification. Long chain fatty acids were better catalyzed as compared to the shorter ones. Rhizopus delemar and R. arrhizus lipases showed a preference for the esterification of short chain primary alcohols, while the secondary alcohols had a low rate of esterification and the tertiary ones could not be converted. The reaction of medium chain length fatty acids was also better catalyzed than in the case of the long ones. The observed lipase selectivity appeared to be related to the localization of the enzyme molecule within the micellar microstructure due to the hydrophobic/hydrophilic character of the protein. Epicuticular wax from Juniperus scopulorum contains hydrocarbons (16%), esters (11%), free acids (1%), nonacosan-10-ol (27%), nonacosane-diols (7%) and estolides (16%). The major hydrocarbon is tritriacontane; the principal esters are C34–C46, mainly octyl and decyl esters of C28-C36 acids; and the diols consist of nonacosane-4,10-diol (57%),5,10-diol (28%), 7,10-diol (11%) and 10,13-diol (4%). Hydrolysis of the estolides gave a mixture of acids, ω-hydroxy acids, , ω-diols, alcohols and hydroxy acids. The hydroxy acids are a new class of C28–C36 acids with the OH attached to either the eighteenth or twentieth carbon from the terminal methyl end; the major component is 13-hydroxydotriacontanoic acid. Syntheses of this acid and of nonacosane-4,10-dione and nonacosane-4,10-diol are described.  相似文献

Cp#2Yb (Cp#=C5H4(CH2)2NMe2) has been obtained by reaction of YbI2(THF)2 with 2 equiv. of C5H4(CH2CH2NMe2)K in THF. The X-ray structure analysis shows a bent structure with intramolecular coordination of both nitrogen atoms to ytterbium. The reaction of C60-fullerene with Cp#2Yb leads to the formation of the fullerenide derivative [Cp#2Yb]2C60, which shows an ESR signal in the solid state and in THF solution at room temperature (solid: ΔH = 50 G, G = 1.9992; solution: ΔH = 10 G, G = 2.0001) and a magnetic moment of 3.6 BM. The lutetium fullerenides CpLu(C60)(DME) (3) and Cp*Lu(C60)(DME)(C6H5CH3) (4), (Cp = η5−C5H5, Cp* = η5−C5Me5), were obtained by reaction of C60 with CpLu(C10H8) (DME) and Cp*Lu(C10H8) (DME) in toluene. Both complexes are paramagnetic (μeff = 1.4 and 0.9 BM) and exhibit temperature-dependent ESR signals (293 K: g = 1.992 and 2.0002 respectively).  相似文献

The triglyceride compositions of extracts and cornicle secretions of specimens from a range of aphid families were examined by mass spectrometry and found to support earlier evidence that there is little correlation between taxonomic position and chemical constitution.

Parasites developing within Myzus persicae preferentially consumed the typical aphid triglycerides which contain hexanoic (C6), sorbic (C6:2), myristic (C14), and palmitic (C16) acid moieties, so that when parasitism was advanced, only triglycerides with three long-chain fatty acids per molecule remained. The adult parasites themselves contained very little triglyceride.

Triglycerides of Aphis evonymi and Aphis fabae were similar and varied in composition regularly throughout the year. In the apterous and alate (summer) viviparae, there were equal amounts of myristoyl (C14) and palmitoyl (C16) triglycerides, while in fundatrices and fundatrigeniae (spring forms) myristoyl was predominant. Males collected in autumn resembled the spring forms. However, oviparae and eggs of these and other species differed considerably from the other seasonal forms, in having more hexenoic (C6:1), sorbic (C6:2), and myristoleic (C14:1) acid moieties.  相似文献

Atromentic acid was isolated from culture liquids of Clitocybe illudens. Identification, based on analyses and spectra of the compound and a number of new derivatives, was confirmed by direct comparison of its tetramethylated derivative with a synthetic sample. Products, giving a blueing reaction have been detected in the same culture liquids, and an as yet unidentified compound has been isolated in crystalline form as its methylation product, C42H36O12. The presence of pulvinic acid derivatives in Basidiomycetes other than the Boletaceae has been reported in only one other instance.  相似文献

Ethyl esters (EE) of C2 to C14 saturated acids were interesterified with tripalmitin using papaya (Carica papaya) lipase to produce structured triacylglycerols (TG) with palmitoyl moieties in the secondary (sn-2), and short-chain or medium-chain acyl moieties in the primary (sn-1,3) positions. It was found that the incorporation of the acyl moieties rose with time and chain length of the ethyl ester. Little reaction occurred with ethyl acetate. The positional analysis of the structured TG formed revealed an increase in preference of the lipase for the primary positions as compared to the secondary position with increasing chain length of the acyl donor from C2 to C14.  相似文献

Epoxy alkylstearates were synthesized by lipase catalysed esterification and perhydrolysis followed by epoxidation of oleic acid in a one-pot process. Immobilized Candida antarctica lipase (Novozym®435) was used as the catalyst. The esterification reaction occurred relatively quickly and was followed by epoxidation of the alkyl ester and the remaining fatty acid. Higher degree of esterification was achieved with n-octanol, n-hexanol and n-butanol as compared to that with ethanol and iso-propanol. The rate and yield of epoxidation was enhanced with iso-propanol but was lowered with the other alcohols. The lipase suffered significant loss in activity during the reaction primarily due to hydrogen peroxide. The presence of alcohols, in particular ethanol, further contributed to the enzyme inactivation. The epoxidation reaction could be improved by step-wise addition of the lipase.  相似文献

The fatty acid content of 30 species of Porifera, including samples of Hexactinellida and Lithistida for which no fatty acid data previously existed, have been examined. The sponges are unique among animal phyla in diversity of fatty acids with generally high levels of long chain fatty acids (LCFAs; C24–30, high unsaturation (mainly polyunsaturation), high incidence of branched and odd chain fatty acids. Further, peculiarities in proportions of individual acids of particular chain lengths distinguish the phylum. Hexactinellid fatty acid traits corresponded closely to those of Demospongiae while the calcareous species was atypical in exhibiting low levels of LCFAs and unsaturation. Seasonal and geographical influences on components of the fatty acid profile limit the extent to which this information can be utilized in a chemotaxonomic sense.  相似文献

Two gamma- and UV-radiation resistant, Gram-positive, red- or pink-pigmented, rod-shaped, strictly aerobic, oxidase- and catalase-positive bacterial strains, TDMA-25T and TDMA-uv51T, were isolated from fresh water collected at Misasa, a radioactive site in Japan. Phylogenetic analysis based on 16S rRNA gene sequences placed both in a distinct lineage in the family Deinococcaceae, and the highest degrees of sequence similarity determined belonged to Deinococcus maricopensis LB-34T (88.8–89.3%), Deinococcus pimensis KR-235T (86.4–86.7%) and Deinococcus yavapaiensis KR-236T (86.1%). The DNA G+C content of the strains was 53–58 mol%. The major respiratory quinone was MK-8. The predominant fatty acids were C15:0 iso, C16:0 iso, C13:0 iso, C17:0 iso, C16:0, C13:0 anteiso, C15:0 and C12:0 iso. The strains degraded gelatin, casein, starch and Tween 80. Unique physiological characteristics, differences in their fatty acid profiles, and genotypic and phylogenetic features, differentiated strains TDMA-25T and TDMA-uv51T from closely related Deinococcus species. Hence, the two strains are described as novel species of the genus Deinococcus. The names Deinococcus misasensis sp. nov. (type strain TDMA-25T=JCM 14369=NBRC 102116=CCUG 53610) and Deinococcus roseus sp. nov. (type strain TDMA-uv51T=JCM 14370=NBRC 102117=CCUG 53611) are proposed.  相似文献

This study is the first attempt to characterise the chemical composition of the secretion of the smooth pads of the locust Locusta migratoria and to relate this to the composition of the cuticle coverage of the pads and the wings. Gas-chromatography and mass-spectrometry (GC-MS) were the principal techniques used for the characterization of these materials. Secretion droplets were visualised and quantified with the aid of diverse microscopic techniques. The chemical composition of prints is shown to differ from the cuticle coverage, in particular, with respect to the fatty acid distribution: in the secretion, saturated and unsaturated fatty acids with chain lengths between C16 and C20 in both the free form and as glycerides predominate, whereas cuticle coverage contains waxes of long-chained fatty-acids bound to long-chain primary alcohols. The second important difference is the significant amount of glucose and other saccharides found in methanolyzates of the pad fluid. A considerable amount of the amino acids (up to 53%) was detected in the non-volatile portion of the fluid. Data obtained from the shock-freezing, carbon-platinum coating and replica preparation show that the secretory droplets contain nano-droplets on their surfaces. The results lead us to suggest that the pad secretion is an emulsion consisting of lipidic nano-droplets dispersed in an aqueous liquid. According to the chemical composition of the secretion, a high-viscosity of the fluid may be suggested. Presumably, the fluid is a kind of a coupling agent, promoting and strengthening adhesion between otherwise incompatible materials by providing the proximity of contact for intermolecular forces.  相似文献

Phosphatidylcholine containing large amounts of long polyunsaturated fatty acid, eicosapentaenoic acid (C20:5) and docosahexaenoic acid (C22:6), was synthesized in isooctane. Immobilized phospholipase A2 was used as a catalyst. A parallel non-enzymatic esterification reaction was investigated in separate experiments.

The concentrations of lyso-phosphatidylcholine, polyunsaturated fatty acids, water and the enzyme were varied over wide ranges as were the temperature and the reaction time. The type of enzyme, carrier and immobilization procedure were held constant.

The yield of phosphatidylcholine was relatively high (about 21%) when the concentration of polyunsaturated fatty acids was high (300 mg/g of reaction mixture) and the water content was low (below 30% of the dry immobilized enzyme). The highest yield of phosphatidylcholine was found at 80 hours and 75°C. However, at this temperature an extensive non-enzymatic reaction between polyunsaturated fatty acids and lyso-phosphatidylcholine occurred. At 80°C the polyunsaturated fatty acids were partly oxidized. Therefore, a temperature of 45°C to 65°C is probably the optimum temperature for the reaction.  相似文献

A high-performance liquid chromatographic technique for the rapid assessment of fatty acids in cardiac tissue is described. A level of 50.4 ± 14.9 nmol fatty acids per g wet weight of rat myocardial tissue could be monitored. The content of the individual fatty acids C14:0, C16:0, C16:1, C18:0, C18:1, C18:2 and C20:4 amounted to 1.9, 13.5, 0.6, 14.4, 6.1, 6.5 and 7.2 nmol/g wet weight, respectively. A comparison of this method with a well established gas chromatographic technique yielded good agreement. In contrast with time-consuming gas chromatographic techniques, there is no need to isolate (unesterified) fatty acids from the other lipid classes with column chromatography or thin-layer chromatography, because the derivatizing reagent 9-anthryldiazomethane reacts highly specifically with fatty acids.  相似文献

Yolk fatty acid (FA) concentrations and egg quality were evaluated with respect to the inclusion of different levels of marine fish oil (MFO) and linseed oil (LO) in a basal diet. Ten diets were arranged factorially with two levels of MFO addition (15 and 17 g/kg) and five levels of linseed oil (1, 2, 3, 4 and 5 g/kg). Two commercial feeds with no n-3 FA sources added, or containing 15 g MFO/kg served as controls for egg sensorial quality traits. Type of diet did not affect egg production traits, nor total fat content in yolk or saturated and monounsaturated FA concentration in yolk fat. An increase in the inclusion of MFO from 15 to 17 g/kg increased (P<0.05) egg yolk content of total n-3 FA, C20:5 n-3 and C22:6 n-3 by 3.3, 9.8 and 3.5%, respectively. Linseed oil addition increased linearly (P<0.001) the level of total n-3 FA and the proportion of C18:3 n-3, C20:5 n-3, C22:5 n-3 and C22:6 n-3 retained in yolk fat by respectively 131, 12.0, 6.9 and 20.7% between extreme diets. Efficiency of retention of n-3 FA decreased (P<0.001) with dietary level of n-3 FA, and with the dietary ratio of MFO to LO. Both MFO and LO addition decreased (P<0.001) C20:4 n-6 to C18:2 n-6 ratio in yolk. Sensorial quality of eggs was not affected by treatments and did not differ with respect of that obtained in commercial n-3 FA enriched eggs. No interaction was found between LO and MFO addition for any of the traits studied. Regression equations have been calculated in order to predict efficiency of retention and yolk fat content of LC n-3 and n-6 FA from dietary characteristics.  相似文献
