A lipase gene (lip) and its activator gene (act) on a 2.9 kb BglII-EcoRI fragment from Pseudomonas sp. KWI-56 were cloned in Escherichia coli using pUC19 as a vector plasmid. From the sequencing results, the open reading frames of the lip and the act were found to contain 1092 and 1032 nucleotides, respectively. The act existed downstream of the lip with the same orientation. When the lip was expressed in E. coli using the lac promoter on the pUC plasmid vector, the lipase activity of E. coli carrying both the lip and the act was 200-fold greater than that carrying only the lip. This result suggested the act was important in the expression of the lip in E. coli.  相似文献

(±)-Bornyl acetate and (±)-isobornyl acetate were asymmetrically hydrolyzed by microorganisms to give (?)-borneol with (+)-bornyl acetate and (?)-isoborneol with (+)-isobornyl acetate although in low hydrolytic ratios. On the contrary (±)-bornyl chloroacetate and (±)-isobornyl chloroacetate were easily hydrolyzed by microorganisms to afford (?)-borneol and (?)-isoborneol with the acetate of their antipodes. Also, enzymatic hydrolysis of (±)-menthyl chloroacetate proceeded faster than that of (±)-menthyl acetate.  相似文献

By selected microorganisms the acetates of the racemic sec-alcohols were asymmetrically hydrolyzed to optically active sec-alcohols and the acetates of their antipodes. Microbial hydrolysis of the acetates of the racemic prim-alcohols having an asymmetrical carbon atom at the β-position gave lower optically active alcohols and the acetates of their antipodes, but the acetates having an asymmetrical carbon atom at the γ-position were hydrolyzed only to give racemic alcohols.  相似文献

The lipase-catalyzed kinetic resolution of trans- and cis-2-azidocycloalkanols and the preparation of enantiomerically pure trans- and cis-2-aminocycloalkanols are described.

Four kinds of lipases were screened for the acetylation of trans- and cis-2-azidocycloalkanols. Among them, Pseudomonas sp. lipases (lipase PS and lipase AK, Amamo Pharmaceutical Co.) showed the highest enantioselectivity. These products were converted to the corresponding 2-aminocycloalkanols to determine their enantiomeric excess (ee) and absolute configurations by HPLC and CD analyses, using (S)-TBMB carboxylic acid [(S)-2-tert-butyl-2-methyl-1,3-benzodioxole-4-carboxylic acid] as the chiral conversion reagent. The results of the CD analysis proved N,O-bis-(S)-TBMB carboxylated cis-2-aminocycloalkanols to adopt a predominantly N-equatorial conformation.

The partially resolved trans- and cis-2-aminocycloalkanols, except for trans-2-aminocyclopentanol, were recrystallized from ethyl acetate to give enantiomerically pure forms.  相似文献

We examined whether developing cotyledons of soybean seed had photosynthetic activities. The cotyledons evolved oxygen under illumination and the activity was inhibited by 3-(3′,4′-dichlorophenyl)-1,1-dimethylurea. The rate of oxygen evolution decreased during the development of seeds; about 30μ miol O2 · mg chlorophyll-1 · hr-1 at the early developing stage and about 10μ miol O2-mg chlorophyll-1 · hr-1 at the late developing stage. The rate of oxygen uptake remained at an almost constant level of 40^mol 02-mg chlorophyll-1 · hr-1 throughout the development. Photosynthetic 14CO2-fixation by the cotyledon was observed. Ribulose bisphosphate carboxylase was immunochemically detected in the developing cotyledons. These results show that functional photosynthetic apparatus is present in the developing cotyledons of soybean seeds.  相似文献

Asymmetric hydrolysis of (±)-1-pentyl-2-propynyl and 1-pentyl-2-propenyl acetates by selected microorganisms produced chiral 1-octyn-3-ol and 1-octen-3-ol, respectively, with high optical purities and acetates of their antipodes. Enantioselectivity of microbial hydrolysis changed with the microorganisms used. Also, (±)-1-ethylhexyl acetate was asymmetrically hydrolyzed by microorganisms to give (S)-3-octanol and (R)-1-ethylhexyl acetate of relatively low optical purity and hydrolytic ratio, compared with those of (±)-1-pentyl-2-propynyl acetate.  相似文献

This paper reports on a total synthesis of racemic δ-cadinene (2), which had been obtained previously in optically active form by acid-catalyzed cyclization of (–)-germacrene D. Total synthesis of (±)-deguelin was described. Thermal rearrangement of 1,1-dimethyl-2-propynyl ethers of β-resorcinaldehyde and 2,3-dimethoxy-9-hydroxy-rotoxen-12(6H)-one afforded β-tubaaldehyde and dehydrodeguelin respectively. 2,2-Dimethyl-5-hydroxybenzopyran-6-yl 3-(3,4-dimethoxyphenoxy)-1-propynyl ketone was converted to the corresponding benzopyran-6-yl benzopyran-4-yl ketone, which was treated with sodium acetate to give (±)-deguelin.  相似文献

The enzymatic properties of P2-2 enzyme were determined by using cells of M. radiodurans. The enzyme was: most active at 60°C incubation temperature, stable at 40°C in neutral buffer, and inactivated by heating at 80°C for 15min. Maximal lytic activity occurred at pH 8.5 in Tris-HCl buffer. The range of enzyme stability was between pH 5.5 and 8. Bivalent metal ions, p-chloromercuribenzoate and monoiodo acetate inhibited lytic activity. The molecular weight was estimated to be 16,000 daltons by gel filtration on Sephadex G-75. The enzymatic digestion of peptidoglycans from the cell walls of M. radiodurans and M. lysodeikticus liberated free amino groups, but neither reducing groups nor N-acetylhexosamine, indicating that the enzyme was an endopeptidase. From analysis of the N-terminal amino acids of the digests, it is suggested that the P2-2 enzyme cleaves the peptide bond at the carboxyl group of D-alanine in peptidoglycan.  相似文献

Boron is an essential nutrient for plants, but it is toxic in excess. Transgenic rice plants expressing an Arabidopsis thaliana borate efflux transporter gene, AtBOR4, at a low level exhibited increased tolerance to excess boron. Those lines with high levels of expression exhibited reduced growth. These findings suggest a potential of the borate transporter BOR4 for the generation of high-boron tolerant rice.  相似文献

14C-labelled methionine, xanthosine, and 7-methylxan-thosine were given to excised tea shoots. The methyl group of methionine was incorporated into 7-methylxanthosine (ca. 10%) in the earlier period of incubation after the uptake. About 50% of the radioactivity of xanthosine was rapidly incorporated into caffeine via 7-methylxanthosine, 7-methylxanthine, and theobromine within 24 hr. 7-Methylxanthosine was also converted into caffeine at a high rate. The results suggest that the pathway for caffeine biosynthesis is as follows: xanthosine → 7-methylxanthosine → 7-methylxanthine → theobromine → caffeine.  相似文献
