The purpose of this study was to explore the effect of acute mild stress (12–48 hour food and water deprivation) and acute severe stress (12 hour food and water deprivation followed by 10 min swim in water at 4°) on the intake of different isocaloric dietary regimes. Each group of experimental animals was given only one particular diet. Rats subjected to mild stress showed very little preference of dietary regimes. When the food intake was measured during 3 hour period, following 48 hours of fasting, animals showed 2 to 3 fold increase in the food and water intake but no particular dietary preference. However, when rats were subjected to severe stress, there was an increase in the food intake of 154% (control diet); 174% (high-carbohydrate diet); 310% (high protein diet) and 423% (high fat diet) compared to animals subjected to mild stress. In terms of the absolute quantity of food, the animals subjected to severe stress ate more high-fat diet than any other diet; the consumption of high fat diet was 142% more than high-protein diet, 180% more than control diet and 258% more than high carbohydrate diet. Animals subjected to severe stress and given high-carbohydrate and high fat diet also showed 80% increase in the water intake. Prior administration of naloxone (1 mg/kg body weight, i.p.) reduced the stress induced increase in the intake of food and water. Naloxone inhibited the intake of high-fat diet more than any other diet. The ability of naloxone to block the increase in the intake of high-fat diet, and the reported increase in the concentration of β-endorphin in the different regions of brain of the animals subjected to the cold swim, suggest that endogenous opioid system in body is activated during stress. An activation of the endogenous opioid system leads to a preferential increase in the intake of palatable foods.  相似文献

The effect of subchronic metformin treatment on food intake, weight gain and plasma and tissue hormone levels was investigated in genetically obese male Zucker rats and in their lean controls. Metformin hydrochloride (320 mg/kg/day for 14 days in the drinking water) significantly reduced 24 hour food intake both after one and two weeks treatment in obese rats. In contrast, metformin had only a transient effect on food intake in lean animals. The reduced food intake was associated with body weight decrease, particularly in obese rats. Metformin markedly reduced also the hyperinsulinemia of the obese animals without altering their plasma glucose or pancreatic insulin content which may reflect an improved insulin sensitivity after metformin treatment. Metformin did not change plasma corticosterone levels or insulin and somatostatin concentrations in the pancreas. Metformin reduced pyloric region somatostatin content in lean rats. It is concluded that metformin has an anorectic effect and reduces body weight and hyperinsulinemia in genetically obese Zucker rat.  相似文献

Glucagon-like peptide-1 (GLP-1) is produced by and released from the small intestine following ingestion of nutrients. GLP-1 receptor (GLP-1R) agonists applied peripherally or centrally decrease food intake and increase glucose-stimulated insulin secretion. These effects make the GLP-1 system an attractive target for the treatment of type 2 diabetes mellitus and obesity. In addition to these more frequently studied effects of GLP-1R stimulation, previous reports indicate that GLP-1R agonists suppress water intake. The present experiments were designed to provide greater temporal resolution and site specificity for the effect of GLP-1 and the long-acting GLP-1R agonists, exendin-4 and liraglutide, on unstimulated water intake when food was and was not available. All three GLP-1R ligands suppressed water intake after peripheral intraperitoneal administration, both in the presence of and the absence of food; however, the magnitude and time frame of water intake suppression varied by drug. GLP-1 had an immediate, but transient, hypodipsic effect when administered peripherally, whereas the water intake suppression by IP exendin-4 and liraglutide was much more persistent. Additionally, intracerebroventricular administration of GLP-1R agonists suppressed water intake when food was absent, but the suppression of intake showed modest differences depending on whether the drug was administered to the lateral or fourth ventricle. To the best of our knowledge, this is the first demonstration of GLP-1 receptor agonists affecting unstimulated, overnight intake in the absence of food, the first test for antidipsogenic effects of hindbrain application of GLP-1 receptor agonists, and the first test of a central effect (forebrain or hindbrain) of liraglutide on water intake. Overall, these results show that GLP-1R agonists have a hypodipsic effect that is independent of GLP-1R-mediated effects on food intake, and this occurs, in part, through central nervous system GLP-1R activation.  相似文献

The effects of centrally administered Angiotensin II (Ang II) on water and food intake in rodent models are well known. However, most studies have focused on the acute effects of intracranial Ang II. In the current study, we evaluated the effects of intracerebroventricular Ang II on food and water intake as well as locomotor activity over the entire dark phase of the murine diurnal cycle. Consistent with the previous reports, centrally administered Ang II rapidly stimulated water intake over the initial 1-hour period following treatment. However, this acute increase was immediately followed by a marked reduction in water intake resulting in decreased cumulative water intake approximately 7h after Ang II treatment. Pretreating animals with an Ang II type 1 receptor blocker, Losartan, completely antagonized the acute effect of Ang II and abolished initial water intake. In contrast, application of an Ang II type 2 receptor blocker, PD123319, abrogated the prolonged inhibitory effect of Ang II on drinking behavior and partially suppressed the initial increases in water intake. The suppressive effects of Ang II on cumulative food intake and spontaneous physical activity were also evident throughout the entire dark phase of diurnal cycle. These experiments are the first to suggest that the stimulatory effect of central Ang II treatment on water consumption is very temporary and that it causes a sustained suppressive effect on voluntary locomotion and food intake behavior in mice.  相似文献

Twenty-four hour basal food and water intakes were recorded in Wistar rats. Diabetes was produced in a group of rats by injecting streptozotocin (STZ, 75 mg/kg, b.w., IP) and their post-diabetic basal food and water intakes were recorded. Noradrenaline (2 microg) and dopamine (2 microg) were injected separately into the nucleus accumbens through the implanted cannula in non-diabetic and diabetic animals and their 24 hr food and water intakes were recorded. Food and water intakes were also recorded following bilateral electrolytic lesions of nucleus accumbens in both the groups of rats. In diabetic rats, basal food and water intakes were significantly increased in comparison to basal intakes of non-diabetic rats. Following injection of noradrenaline, a significant increase in water intake but not food intake was seen in non-diabetic rats, whereas food and water intakes remained unchanged in diabetic rats. Following injection of dopamine, a significant increase in food and water intakes was observed in non-diabetic rats, whereas dopamine-induced increase in food intake was absent in diabetic rats. The bilateral lesions of nucleus accumbens resulted in a significant inhibition of food and water intakes in non-diabetic rats, whereas inhibition of water intake without change in food intake observed in diabetic rats. However, no difference was observed in the pattern of change in water intake following lesions or dopamine injections between non-diabetic and diabetic rats, whereas difference was observed for food intake. The results suggest that nucleus accumbens activity changes for food intake, but not for water intake in diabetes.  相似文献

Male Wistar rats received two i.p. injections of morphine-HCl, 2.5 mg/kg at 8.00 a.m. and 2.00 p.m. on the 1st day: the dose was doubled every other day to reach a total daily dose of 40 mg/kg on the 4th day. This schedule was maintained for 12 days. On day 16 the animals received the last injection of morphine, 20 mg/kg. One hour later (9.00 a.m.) six rats were decapitated and PRA, PAC and ACTH were measured by radioimmunoassay. Groups of six rats were killed at 9.00 a.m. on the 1st, 2nd, 5th and the 8th day after morphine withdrawal. Control data for PRA, PAC and ACTH were obtained from eighteen saline-injected rats. Nine out of morphine-treated animals were kept in metabolism cages to investigate simultaneously food and water intake. and renal excretion. Morphine withdrawal after chronic morphine treatment in the rat resulted in antidiuresis and a reduction of electrolyte excretion which were not due to a reduction in water and food intake. The simultaneous increase of PRA and PAC associated with decreased electrolyte excretion indicates that, in addition to antidiuretic hormone, also the renin-aldosterone-system probably play a relevant role in the renal excretory changes after morphine withdrawal.  相似文献

This study examined the effect of subcutaneous administration of the neurohormone oxytocin on water intake of ad lib-fed (with or without sodium availability in the diet) and food-deprived animals. Results of the first experiment showed that oxytocin increased water intake and urine excretion in food-deprived but not in ad lib-fed animals. However, oxytocin treatment did not modify the reduced water "balance" (fluid intake minus urine volume) resulting from food deprivation or the daily food intake (Experiment 1). The dose-dependent polydipsic effect of oxytocin on food-deprived rats was always preceded by an increase in sodium and fluid urine excretion (Experiment 2). Oxytocin also increased the water intake of animals fed ad lib with a low sodium diet (Experiment 3). These results suggest that the effect of oxytocin on water intake is dependent on the presence or absence of sodium in the diet and that the excretion of sodium is the main mechanism of oxytocinergic polydipsia in food-deprived male rats.  相似文献

Previous studies have shown that naloxone causes a decrease in food and water intake; however, the site of this action has not been determined. We investigated this problem by giving bilateral injections of 15 μg/rat of naloxone into the lateral ventricles of cannulated, food and water deprived rats. This treatment caused a significant decrease in food intake when compared to saline injected controls. Water intake in naloxone-treated animals did not differ significantly from that of saline-treated controls during the one hour test period. The total dose of naloxone given centrally, 15 μg, did not produce a change in eating or drinking if given peripherally. The findings imply that naloxone exerts its effect on food intake at a central site. A dose-related and significant suppression of water intake was seen after treatment with nalaxone peripherally (1, 3, and 10 mg/kg, i.p.) in rats with either subdiaphragmatic vagotomy (vag) or a sham vagotomy (sham). Although a significant suppression of food intake was seen in the sham rats, no supression of food intake was seeen in the vag rats at any dose of naloxone tested. In rats pretreated with methyl atropine (5 mg/kg, i.p.), naloxone (3 mg/kg, i.p.) was equivalent to saline in that it did not decrease food intake. However, nalaxone did cause a significant decrease in water intake in methylatropine pre-treated rats. These results suggest that the suppression of food intake by naloxone has a central site of action which is mediated by the vagus, and specifically by vagal efferents, since the effect was blocked by methylatropine. The results also suggest that naloxone's effect on water intake is mediated by a different mechanism than that involved with food intake.  相似文献

Ghrelin is the only peripheral orexigenic peptide of gastrointestinal origin. Its preprandial increase is supposed to initiate food intake. This assumption is based on studies with intravenously infused ghrelin in rather high doses and the correlation between ghrelin levels and hunger sensations. As yet it is unclear whether or not low dose ghrelin resulting in physiological and moderately supraphysiological plasma levels has an effect on hunger sensations, the wish for food intake and / or the quantity of the meal consumed. We examined 20 normal-weight males (age 25±1.7 years, BMI 24±0.5 kg/m(2)) in a prospective double-blind randomized fashion. On two different days they obtained a ghrelin infusion 1 ng/kg/min or intravenous saline starting one hour after a standardized meal. Hunger and satiety ratings were documented by visual analogue scales. A second meal was served on demand and consumed until feeling satiated. Time point of the second meal as well as ingested calories were registered. Prior to the start of i.v. ghrelin the postprandial decrease of active plasma ghrelin by 30 pg/ml was comparable. In the controls the postprandial reduction was significant until 210 min compared to basal. With i.v. ghrelin basal levels were reached within 10 min. The maximal rise was twice basal. No effect was observed on hunger and satiety ratings. The time period between the meals and the food quantity of the second meal were similar. During ghrelin infusion glucose and growth hormone but not insulin and cortisol levels were significantly higher after the second meal compared to saline. The present data demonstrate for the first time the effect of a low dose ghrelin infusion on food intake. Neither physiological nor moderably supraphysiological ghrelin levels were associated with any change of the various food intake parameters determined. These data do not favour a hormonal role of peripheral ghrelin in the regulation of food intake.  相似文献

Insulin and C-peptide are released in equimolecular concentrations in the circulation after food ingestion as they result from the enzymatic cleavage of proinsulin. In the brain, insulin inhibits food intake through hypothalamic receptors. In the present study, we tested the ability of C-peptide to modulate food intake when injected in the brain lateral ventricle of Long-Evans rats. For this purpose, 10 adult male rats (BW 320 - 350 g) were deprived of food overnight. They were intra-cerebroventricularly injected with 10 microg C-pepitde or vehicle (artificial cerebrospinal fluid [CSF]) during the first hour of the light period and chow intake was measured 1, 3, 6 and 24 hours after injection. Chow availability immediately triggered food consumption. Food intake was not different between CSF- and C-peptide-injected rats either after one hour (5.7 +/- 0.6 g [CSF] vs. 6.7 g +/- 0.5 g; ns) or after 24 hrs (23.3 +/- 1.4 g [CSF] vs. 25.1 g +/- 1.4 g; ns). In addition, a higher dose (20 microg/rat) had no effect at all in satiated rats one hour after injection or later contrary to the 100 % increase measured after injection of 2 microg of neuropeptide Y. Thus, we conclude that contrary to insulin, C-peptide does not regulate feeding behaviour in normal rats whatever their insulin status.  相似文献

在食物中含10 %和20%蛋白质的条件下, 采用食物平衡法测定了单宁酸对根田鼠食物摄入量和蛋白质消化率的作用。食物蛋白质含量为10 %时, 第1~5 天, 单宁酸对根田鼠食物摄入量具有显著的抑制作用, 自第6天, 其作用不明显, 以3 %和6 %单宁酸处理的根田鼠, 其食物蛋白质消化率较对照组分别降低22 %和47.67 %;在食物蛋白质含量为20 %的条件下, 单宁酸对根田鼠的食物摄入量和蛋白质消化率无显著作用。上述结果验证了植物次生化合物能抑制植食性小哺乳动物食物摄入量及蛋白质消化率的假设。  相似文献

Food intake is stimulated following administration of orexin-A into the perifornical region of the lateral hypothalamus (LH/PFA). Orexin neurons originating in the LH/PFA interact with a number of hypothalamic systems known to influence food intake, including glutamatergic neurons. Glutamatergic systems in the LH/PFA were demonstrated to initiate feeding through N-methyl-d-aspartic acid (NMDA) receptors. Male Sprague-Dawley rats fitted with brain guide cannulas to the LH/PFA were used in two experiments. In the first experiment, a combination microdialysis/microinjection probe was used to deliver artificial cerebrospinal fluid (aCSF) or 500 pmol of orexin-A into the LH/PFA. Orexin-A increased interstitial glutamate to 143 +/- 12% of baseline (P < 0.05), which remained elevated over the 120-min collection period. In the second experiment, the NMDA receptor antagonist d-2-amino-5-phosphonopentanoic acid (d-AP5; 10 nmol) was administered before orexin-A. The orexin-induced increase in food intake (from 1.1 +/- 0.4 to 3.2 +/- 0.5 g, P < 0.05) during the first hour was absent in rats receiving d-AP5 + orexin-A (1.2 +/- 0.5 g). There was no effect of d-AP5 alone on food intake. These data support glutamatergic systems in the LH/PFA mediating the feeding response to orexin-A through NMDA receptors.  相似文献

The effect of two cholecystokinin (CCK) receptor antagonists (L-364718 and L-365260) and of diazepam on voluntary food intake in chicken was studied. L-365260 significantly increased cumulative food intake at 30 and 60 min, whereas L-364718 had no effect. Diazepam also increased food intake at 30 minutes. It was therefore concluded that anxiolytic effects could account for an increase in food intake in the conditions under which the study was carried out. Consequently, the effect of L-365260 could either be due to its anxiolytic properties or to its direct effects on food intake control.  相似文献

The effects of dynorphin A (1-13) (DYN), injected into the preoptic area, was investigated on water intake in rats. DYN at both doses of 2 and 10 nmoles significantly increased water intake for two and four hours after the injection in a dose related fashion. However, no significant change was observed in food intake. Naloxone pretreatment (0.3 mg/kg, s.c.) completely attenuated the DYN-induced stimulation of water intake. The present studies suggest that DYN in the preoptic area may play an important role in the regulation of drinking behavior, but have no effect on food intake.  相似文献

Seasonal variation in daily food intake is a well-documented phenomenon in many organisms including wild-type coho salmon where the appetite is noticeably reduced during periods of decreased day length and low water temperature. This reduction may in part be explained by altered production of cholecystokinin (CCK) and growth hormone (GH). CCK is a hormone produced in the brain and gut that mediates a feeling of satiety and thus has an inhibitory effect on food intake and foraging behaviour. Growth hormone (GH) enhances feeding behaviour and consequently growth, but its production is reduced during winter. The objectives of this study were: first, to compare the seasonal feeding behaviour of wild and GH-transgenic coho salmon; second, to determine the behavioural effect of blocking the action of CCK (by using devazepide) on the seasonal food intake; and third, to measure CCK expression in brain and gut tissues between the two genotypes across seasons. We found that, in contrast to wild salmon, food intake in transgenic salmon was not reduced during winter indicating that seasonal control of appetite regulation has been disrupted by constitutive production of GH in transgenic animals. Blocking of CCK increased food intake in both genotypes in all seasons. The increase was stronger in wild genotypes than transgenic fish; however blocking CCK in wild-type fish in winter did not elevate appetites to levels observed in the summer. The response to devazepide was generally faster in transgenic than in wild salmon with more rapid effects observed during summer than during winter, possibly due to a higher temperature in summer. Overall, a seasonal effect on CCK mRNA levels was observed in telencephalon with levels during winter being higher compared to the summer in wild fish, but with no seasonal effect in transgenic fish. No differences in seasonal CCK expression were found in hypothalamus. Higher levels of CCK were detected in the gut of both genotypes in winter compared to summer. Thus, CCK appears to mediate food intake among seasons in both wild-type and GH-transgenic salmon, and an altered CCK regulation may be responsible at least in part for the seasonal regulation of food intake.  相似文献

The objective of the present study was to determine the rostrocaudal distribution and the effect of reduced food intake (60% of the average daily food intake for 1-4 weeks) on the amount of leucine-enkephalin (Leu-enk), neuropeptide Y (NPY) and galanin (Gal) in the lateral septum of male rat brain. Using pre-embedding immunocytochemistry combined with densitometry on 60 microm serial vibratome sections we found that in control animals Leu-enk-immunoreactive elements showed an increasing density from rostral towards the medial part of the septum, then a gradual decrease towards the caudal direction. The distribution of NPY proved to be rather even along the examined sequence of sections with two smaller peaks roughly at the 1/3 and 2/3 of the rostrocaudal axis. Gal showed similar distribution but the peaks were shifted to more caudal direction. We also found that Leu-enk forms the most dense plexus followed by a moderate amount of NPY-positive axonal meshwork. Gal was present in the lowest amount along the lateral septal nuclei. The effect of reduced food intake was marked and differential in the case of the three examined peptides. During the first 2 weeks of reduced food intake NPY-immunoreactivity was upregulated as compared to the control, then it was reduced close to the control value by the 4th week. The changes in Gal immunoreactivity showed similar pattern. The average relative density of Leu-enk-immunoreactive elements immediately decreased as a result of reduced food intake for 1 week and it gradually decreased by the end of the 4th week. These results indicate that reduced food intake affects the expression of NPY, Gal and Leu-enk not only in the relevant hypothalamic neuroendocrine centres, but also in the lateral septal area.  相似文献

Cholecystokinin octapeptide (CCK-8, 1, 190 pmol/5 min) decreased food intake and water consumption in two models of ingestive behavior, i.e., food deprivation-induced feeding and insulin-induced feeding, when administered into the third (3V) and lateral (LV) cerebral ventricles. In fasted dogs, the suppression of food intake was more prominent after 3V CCK-8, whereas intravenously administered CCK-8 was without effect. Neuropeptide Y (NPY, 1, 190 pmol) had no significant stimulatory effect on food intake and water consumption in fasted as well as satiated dogs, and actually reduced both food and water intake in insulin-treated dogs. There was a slight but significant decrease in food and water intake after 275 nmol naloxone administration in both feeding models, and some of the dogs vomited. In insulin-treated animals, CCK-8 reversed, but NPY potentiated the hypothermic phase of temperature response observed after saline administration, whereas naloxone failed to alter rectal temperature. These results suggest that the effect of CCK-8 on feeding seems to involve central mechanisms in the dog, and that the mechanisms by which CCK-8, NPY and naloxone affect feeding behavior are different.  相似文献

While prolactin (PRL) has been reported to increase food intake by virgin female rats, its effects on food intake by male rats are relatively unexplored. The present studies examined the possibility that PRL has sex-specific effects on food intake by rats. In the first study, intact female and male rats were given subcutaneous injections of saline vehicle or ovine (o) PRL (1.0 mg/kg) twice daily at 08:00 and 20:00 h for 10 days. Food intake, body weight, and water intake were measured daily. Results indicate that oPRL administration increased food intake by an average of 4.5 g per day in female subjects, but did not significantly alter body weight or water intake. Male rats treated with oPRL did not significantly alter their food intake, even after an additional five days of treatment. In the second study, a wide range of oPRL doses (vehicle, 0.02, 0.2, 2.0, and 20.0 mg/kg/day) were tested in gonadectomized female and male rats. The results indicate that female rats responded to increasingly larger doses of oPRL with greater increases in food intake, with a maximum increase of approximately 6. 1 g per day at a dose of 20.0 mg/kg. In contrast, male rats maintained baseline levels of intake across all oPRL doses tested. These data suggest that PRL has sex-specific effects on food intake.  相似文献

It was previously shown that sodium nitroprusside (NP) stimulates food (FI) and water (WI) intakes when injected intraperitoneally (i.p.) in male rats deprived for 1-h of both food and water during day time. The present work shows that: 1) intramuscular NP increased only water intake; 2) when NP was i.p. injected simultaneously without 1-h deprivation it slightly reduced both intakes in the first 30 min but stimulated significantly FI between 30 and 60 min, and 3) it increased significantly FI even in absence of available water, while WI in absence of food was practically not affected. In conclusion NP effect on FI does originate mainly in the splanchnic area, it requires a latency of about 30 min, and stimulation of FI by NP triggers an increase in WI and not the other way around.  相似文献

Neuropeptide Y (NPY), a peptide contained within numerous presynaptic terminals in the hypothalamic paraventricular nucleus (PVN), was injected directly into the PVN of satiated, brain-cannulated rats, and food and water intake were measured 0.5, 1, 2 and 4 hrs postinjection. Neuropeptide Y (24 and 78 pmoles/0.3 microliter isotonic saline) caused a dose-dependent increase in food intake, as well as a small, dose-dependent increase in water intake. This effect on feeding occurred even when food was not presented until 4 hrs postinjection. To determine the behavioral specificity of this effect, the impact of PVN injection of NPY (78 pmoles) on various behaviors was observed. With food available, only feeding and drinking behavior were affected. No change in other behaviors, including grooming, rearing, sleeping, resting or different levels of activity, was observed. With food absent, NPY still elicited drinking, suggesting that this is a primary effect, rather than secondary to the feeding. In addition to drinking, NPY reliably increased activity while decreasing sleep and grooming. These results suggest an important role for hypothalamic NPY, or a structurally-related peptide, in the regulation of feeding and drinking behavior.  相似文献
