The midgut protease profiles from 5th instar Mamestra configurata larvae fed various diets (standard artificial diet, low protein diet, low protein diet with soybean trypsin inhibitor [SBTI], or Brassica napus) were characterized by one‐dimensional enzymography in gelatin gels. The gut protease profile of larvae fed B. napus possessed protease activities of molecular masses of approximately 33 and 55 kDa, which were not present in the guts of larvae fed artificial diet. Similarly, larvae fed artificial diet had protease activities of molecular masses of approximately 21, 30, and 100 kDa that were absent in larvae fed B. napus. Protease profiles changed within 12 to 24 h after switching larvae from artificial diet to plant diet and vice versa. The gut protease profiles from larvae fed various other brassicaceous species and lines having different secondary metabolite profiles did not differ despite significant differences in larval growth rates on the different host plants. Genes encoding putative digestive proteolytic enzymes, including four carboxypeptidases, five aminopeptidases, and 48 serine proteases, were identified in cDNA libraries from 4th instar M. configurata midgut tissue. Many of the protease‐encoding genes were expressed at similar levels on all diets; however, three chymoptrypsin‐like genes (McSP23, McSP27, and McSP37) were expressed at much higher levels on standard artificial diet and diet containing SBTI as was the trypsin‐like gene McSP34. The expression of the trypsin‐like gene McSP50 was highest on B. napus. The adaptation of M. configurata digestive biochemistry to different diets is discussed in the context of the flexibility of polyphagous insects to changing diet sources. The effects of diet and different constant temperatures on hemolymph cation concentrations (Na+, K+, Mg2+, Ca2+) have been studied in Morimus funereus larvae collected from natural habitat, fed natural (oak or beech bark) or artificial diet, as well as in larvae reared from hatching on an artificial diet. In the hemolymph of larvae maintained under natural conditions Mg2+ was dominant, whereas Na+ concentration was very low. In their natural diets concentrations of Na+ and K+ were very low, while those of Ca2+ and Mg2+ were high. In larvae continuously reared on an artificial diet, hemolymph Mg2+ concentration was significantly decreased and Na+ concentration increased more than fourfold compared to the results obtained in oak-fed larvae. Na+ and K+ are the dominant cations in the artificial diet. The concentrations of K+ and Ca2+ in the hemolymph of larvae fed natural or artificial diet are nearly identical, suggesting the existence of an internal regulatory mechanism in this insect for these cations. The hemolymph cation concentrations of M. funereus larvae are predominantly dependent upon the diet consumed, much less upon the environmental temperatures. The most stable concentrations of cations were observed in larvae continuously fed an artificial diet and exposed to different constant temperatures. There was much less stability in the hemolymph cation concentration in oak larvae fed either natural or artificial food after their transfer to constant temperatures. Growth, survival, fecundity, and nutrition of Tirathaba rufivena (Walker) fed on two artificial and one natural diet were studied at 29±1°C and 75±5% relative humidity. Larvae reared on the two artificial diets showed faster growth and development, higher pupal survival and weight, and higher adult fecundity than those reared on a natural diet of young coconuts, Cocos nucifera L. The relative growth rate, efficiency of ingested food conversion, and efficiency of digested food conversion in larvae fed on artificial diets were significantly higher than those of larvae fed on young coconuts. The relative consumption rate of larvae fed on artificial diets was significantly lower than that of larvae fed on young coconuts. T. rufivena showed no significant degradation in ability to propagate after being reared on the artificial diet for five successive generations. These results indicated that two artificial diets are suitable for mass rearing of T. rufivena.  相似文献

African catfish, Clarias gariepinus (B.), is one of the promising freshwater fish species in African aquaculture but the expansion of its farming needs more production of its larvae. The use of live food organisms at first feeding for larvae is still obligatory. That increases the cost of larvae production. Hence, the incorporating of exogenous enzymes especially protease in artificial microdiets may provide affordable alternatives for enhancing the larvae performance. The present study was carried out to evaluate the growth and survival of larvae or fingerlings of African catfish fed artificial diets incorporated with different protease levels. Four artificial diets were formulated and enriched with protease enzyme at levels of 0.0, 750, 1,000, and 1,250 unit/kg diet; after that diets were made into crumbles (100–200 µm diameter). After absorption of the yolk sac, diets were offered to fish larvae (3.6 ± 0.2 mg) in triplicates as a starter feed up to apparent satiation every two hours for 30 days. In another treatment, fish larvae were fed on newly hatched Artemia nauplii (2,500 Artemia/L) as a starter food. In another experiment, African catfish fingerlings (10.1 ± 1.6 g) were fed on the same diets up to satiation twice a day for 2 months. It was noticed that the dietary protease improved larval growth and survival but not as Artemia nauplii did where fish larvae fed on Artemia nauplii showed highest growth and survival followed by those fed a diet enriched with 1,250 unit/kg diet of protease. The mortality of larvae fed protease‐enriched diets as well as the control diet was occurred mostly at the first week reaching its maximum at the third week. The poor growth was observed with fish larvae fed the control diet. Meanwhile, catfish fingerlings fed protease‐enriched diets showed higher growth over those fed the control diet. The larvae survival (11.0%–41.7%) was enhanced by increasing protease levels and it was lower than that of fingerlings (95.6%–100.0%). Furthermore, protein retention and digestibility were significantly improved with protease supplementation over the control diet especially at a level of 1,000 unit/kg diet. As compared with the previous studies, live food should be used in larvae rearing for the first week after that a starter diet enriched with protease at levels of 1,250 unit/kg diet should be used. In case of fish fingerlings, the dry diets should be enriched with 1,100 unit/kg diet to improve diet digestibility and subsequently enhance their growth.  相似文献

The growth of river catfish Mystus nemurus (Cuvier & Valenciennes) larvae fed four isocaloric diets (4200 kcal kg?1) with different protein levels during weaning was determined. Diets containing 45, 50, 55, and 60% protein were formulated by linear programming using amino acid profiles based on that of 2‐day‐old river catfish larvae. Artificial diets were fed to the larvae beginning at day 5 after being initially fed Artemia nauplii for 4 days. The larvae thrived solely on artificial diets from day 8 to day 16. On the other hand, the control larvae were fed Artemia nauplii from day 1 to day 16. Results of the feeding trial showed that growth and survival of M. nemurus larvae given the diet containing 60% protein were high and comparable to those of the larvae given only live food (control). Larvae fed the 55% protein diet had significantly lower growth and survival than the larvae on the control and 60% diets but significantly higher growth and survival rates than did larvae fed with 45 and 50% protein diets. Carcass moisture and total lipids after 16 days of feeding did not differ significantly (P > 0.05), but body protein increased with increasing dietary protein. Body protein of the control larvae was similar to that of larvae given the 60% protein diet.  相似文献

Goldspotted oak borer, Agrilus auroguttatus Schaeffer (Coleoptera: Buprestidae), is a new invasive species in southern California, USA. The extent of the host range of this insect is not known, but this knowledge will have a major impact on assessment of the risks that this pest poses to oaks [Quercus spp. (Fagaceae)]. We conducted laboratory tests to determine the potential suitability of native and ornamental oak species for larvae and adults of A. auroguttatus. We infested 179 cut logs (from 163 different trees) with eggs or larvae, measured neonate survival and, after 5 months, counted feeding galleries, and noted the proportion of galleries with late instars. Initial larval survival was generally high when larvae penetrated the phloem (range: 62–98% among oak species), and low by the time larvae began to feed at the phloem/xylem interface (range: 0–25% among oak species). The majority of larvae that established a visible feeding gallery survived to the fourth instar (total of 73% for all oak species). Larval galleries were established with greater frequency in red oaks (Section Lobatae) compared with other oaks (19 vs. 7 or 4%). All red oaks tested (Q. agrifolia Née, Q. kelloggii Newberry, and Q. wislizeni A. DC.) were likely suitable hosts for larvae. Larvae were apparently able to feed on some of the other oaks (Q. chrysolepis Leibmann, Q. suber L., Q. lobata Née, and Q. douglasii Hook & Arn), although it remains unclear whether these species would be preferred hosts under natural conditions. Adult longevity and fecundity varied little by species of oak foliage fed to adults. The host range of A. auroguttatus is likely limited by suitability of oak species for the larval rather than the adult life stage. Results support published field observations that red oaks are more suitable hosts than white oaks.  相似文献

Effects of various single and two species diets on the performance of gypsy moth (Lymantria dispar (L.)) were studied when this insect was reared from hatch to population on intact host trees in the field. The tree species used for this study were red oak (Quercus rubra L.), white oak (Q. alba L.), bigtooth aspen (Populus grandidentata Michaux), and trembling aspen (P. tremuloides Michaux). These are commonly available host trees in the Lake States region. The study spanned two years and was performed at two different field sites in central Michigan. Conclusions drawn from this study include: (1) Large differences in gypsy moth growth and survival can occur even among diet sequences composed of favorable host species. (2) Larvae that spent their first two weeks feeding on red oak performed better during this time period than larvae on all other host species in terms of mean weight, mean relative growth rate (RGR), and mean level of larval development, while larvae on a first host of bigtooth aspen were ranked lowest in terms of mean weight, RGR, and level of larval development. (3) Combination diets do not seem to be inherently better or worse than diets composed of only a single species; rather, insect performance was affected by the types of host species eaten and the time during larval development that these host species were consumed instead of whether larvae ate single species diets or mixed species diets. (4) In diets composed of two host species, measures of gypsy moth performance are affected to different extents in the latter part of the season by the two different hosts; larval weights and development rates show continued effects of the first host fed upon while RGRs, mortality, and pupal weights are affected strongly by the second host type eaten. (5) Of the diets investigated in this study, early feeding on red oak followed by later feeding on an aspen, particularly trembling aspen, is most beneficial to insects in terms of attaining high levels of performance throughout their lives.  相似文献

报道了人工饲料、圆白菜和玉米苗三种食物对甜菜夜蛾Spodoptera exigua发育历期、蛹重以及成虫产卵和飞行能力的影响。幼虫取食人工饲料时发育历期最短,蛹最重(138 mg),产卵量最高(643粒);在12 h的吊飞测试中,成虫飞行距离最远(25.5 km),飞行时间最长(6.2 h),飞行超过10 km的个体占67.9%,超过5 h的占60.7%。幼虫取食玉米苗时发育历期最长,蛹最轻(52 mg),产卵量最低(416粒),成虫仅能飞行3.3 km,1.7 h,飞行超过10 km或5 h的个体均仅占4.2%,各项指标均显著低于人工饲料组。幼虫取食圆白菜时各项指标与人工饲料组基本相似(幼虫历期和蛹重除外),而显著优于玉米苗组(幼虫历期除外)。这些结果表明,幼虫食物对甜菜夜蛾的生长发育、产卵及飞行能力均有显著的影响,因而是影响其个体发育、种群增长及种群动态的重要因素之一。同时,由于取食本人工饲料的各项指标均优于其它两种食料植物,因而可作为饲养甜菜夜蛾的实用饲料。  相似文献

We tested artificial diets for rearing the coconut hispine beetle, Brontispa longissima, a serious invasive pest of coconut (Cocos nucifera) in Southeast Asia. We examined three artificial diets that were identical except for their agar content. The survival rate from hatching to adult emergence was 26.0% when beetles were reared on a ‘soft diet’ (20 g/l agar), 16.0% on a ‘hard diet’ (40 g/l agar), and 41.0% on a ‘mixed diet’, in which the hard diet was used for the first instar and the soft diet for later instars. Females raised as larvae on the mixed diet and then as adults on the soft diet laid few eggs, which did not hatch. However, females reared on the mixed diet as larvae and then on fresh leaves as adults consistently laid eggs that hatched. We then examined the suitability of B. longissima larvae reared on the mixed diet as hosts for the larval parasitoid Asecodes hispinarum, a specialist parasitoid of this beetle. Of the oviposited hosts, 75.0% became mummified and 41.7% produced adult wasps. These results indicate that the mixed diet may be useful for rearing B. longissima larvae as hosts for the rearing of A. hispinarum.  相似文献

The oak platypodid, Platypus quercivorus (Murayama), is the most destructive pest of Quercus and related fagaceous species in Japan. To find an effective biocontrol agent, we determined the effectiveness of entomopathogenic microorganisms against P. quercivorus. P. quercivorus-infested Oak (Quercus serrata, Q. crispula and Q. acutissima) logs were obtained from various regions in Japan, and 28 isolates of microorganisms were isolated from P. quercivorus larvae from those logs. Using molecular methods, the microbes were then classified into 18 species. After a bioassay, we found that three fungi, Lecanicillium sp., Isaria tenuipes, Beauveria bassiana, and the bacterium Serratia marcescens had the greatest potential as biological control agents against P. quercivorus. Another bioassay was then performed to determine the effect of spore or cell densities on the pathogenicity of the four microorganisms, and optimum spore or cell densities for the microbes were determined to be 106 and 107/ml. Based on the results and other criteria, we tentatively selected B. bassiana as the best candidate for future use against P. quercivorus larvae.  相似文献

Dietary exposure studies are initial steps in environmental risk assessments of genetically engineered plants on non‐target organisms. These studies are conducted in the laboratory where surrogate species are exposed to purified and biologically active insecticidal compounds at higher concentrations than those expected to occur in transgenic crops foliage. Thus, dietary exposure (early tier) tests provide robust data needed to make general conclusions about the susceptibility of the surrogate species to the test substance. For this, we developed suitable artificial diet and used it to establish a dietary exposure test for assessing the toxicity of midgut‐active insecticidal compounds to the larvae of the Asian ladybird beetle Harmonia axyridis (Pallas) (Coleoptera: Coccinellidae). Using boric acid as a model compound, we validated the bioassay established for H. axyridis larvae. An artificial diet containing boric acid which negatively affected survival, development and adult weights was offered to larvae and indicated that the bioassay was able to detect toxic effects of insecticidal substances incorporated in diets. Using this dietary exposure test, environmental risk assessment of Cry1Ac, Cry2Ab, Cry1Ca, Cry1F and the non‐Cry protein Vip3Aa was evaluated by analysing pupation rates, adult emergence rates, 7‐day larval weights, and freshly emerged male and female weights among the toxin treatments and a pure artificial diet. These life‐table parameters did not vary among artificial diets containing 200 μg/g Bt proteins or pure artificial diet. In contrast, boric acid adversely affected all life‐table parameters. Thus on these bases, we concluded H. axyridis larvae are not sensitive to these Bt proteins expressed in genetically engineered crops.  相似文献

The mirid bug Macrolophus caliginosus is commercially reared on eggs of Ephestia kuehniella, constituting an effective but expensive factitious food. Artificial diets can decrease the rearing costs of this natural enemy, but developing and evaluating an artificial diet is a very time-consuming activity. In the current study, development and reproduction of M. caliginosus on two artificial diets based on egg yolk were investigated. The artificial diets resulted in longer development and lower adult weights, but survival was comparable with that of control insects fed E. kuehniella eggs. Reproductive potential of the predator reared on factitious and artificial foods was assessed using a dissection method. The influence of nymphal food on fecundity was less important than that of adult food. Adults fed E. kuehniella eggs had a preoviposition period of about 4 days, whereas adults offered only plant material started laying eggs about 7 days after emergence. Ovarian scores at day 7 were higher for females fed E. kuehniella eggs than for those given access only to a tobacco leaf. Ovarian scores were not significantly affected by mating status. In a final test, a parallel comparison of two methods for assessing reproductive response to diet was made. Here, adult couples were offered one of four diets: E. kuehniella eggs, one of two artificial diets or no food. Half of the females were dissected and the other half was held for determining lifetime oviposition. Females fed E. kuehniella eggs had superior ovarian scores and laid more eggs than those fed either artificial diet or those given no extra food. A good correlation (r = 0.97) was obtained between ovarian scores and oviposition data, indicating that dissecting females after 1 week provides a reliable estimate of fecundity as affected by diet quality. Rapid reproductive assessments as used in the current study will help to increase the rate of development of artificial diets and may contribute to more cost effective production methods for augmentative biological control agents.  相似文献

Development, survivorship, pupal weight, oviposition, and life table parameters of the oriental tobacco budworm, Helicoverpa assulta Guenée, were evaluated in the laboratory on an artificial diet, pepper (Capsicum frutescens L.), and tobacco (Nicotiana tobacum L.). We found that the average developmental time of immature stages was longest on tobacco (36.2 d), intermediate on pepper (34.4 d), and shortest on artificial diet (33.5 d). Immature survival from egg to pupa varied from 31% on tobacco, 43% on pepper, and 74% on artificial diet. Pupal weight ranged from 197.4 mg/pupa on tobacco, 233.1 mg/pupa on pepper and 253.4 mg/pupa on artificial diet. The average numbers of eggs laid by adults reared as larvae on the artificial diet, pepper, or tobacco were 614, 421 and 334 eggs/female, respectively. Numbers of remaining eggs in ovaries of the adult females reared as larvae on the artificial diet, pepper, or tobacco were 16, 26, and 42 eggs/female, respectively. The longevity of adult females developed from larvae reared on the three diets was not significantly different, whereas the longevity of male adults from the larvae reared on artificial diet was longer (16.8 d) than that for males reared on tobacco (13.8 d) and pepper (13.3 d). The intrinsic, finite, gross, and net rates of increase were highest for females reared as larvae on artificial diet, lowest for females emerging from larvae reared on tobacco, and intermediate for females emerging from larvae reared on pepper. Generation times and doubling time of H. assulta were shortest for larvae fed artificial diet, intermediate from larvae reared on pepper, and longest from larvae reared on tobacco. We concluded that the artificial diet was the most suitable larval diet of H. assulta followed by pepper, and tobacco.  相似文献

Nutritional indices, development rates, percent dry weights and total lipids were determined in gypsy moth larvae (Lymantria dispar L.) reared on a high wheat germ (HWG) diet or diets prepared from lyophilized, ball-milled oak or pine foliage as the only source of dietary nitrogen (N). With regard to both total and proteinaceous N content, HWG diet>oak diet>pine diet. All nutritional indices measured were significantly lower in second instars fed pine diet vs. oak diet. Protein supplementation of pine diet with either casein or ovalbumin to bring total N up to the level present in oak diet resulted in small increased in approximate digestibility (AD) and effciency of conversion of ingested food (ECI), but relative growth rate (RGR) remained unaffected. The low RGR of larvae fed pine diet (unsupplemented or protein supplemented), as compared to those fed HWG or oak diet, was accompanied by significantly lower larval percent dry weight and percent total lipid. In contrast, RGR, larval percent dry weight and total lipid values were comparable in second instars fed HWG or oak diet. Insects reared from the first through the final instar on oak diet exhibited lower pupal weights compared to those reared on HWG. Casein addition to oak diet generally resulted in even more extended larval development times and further reduced pupal weights, but wheat germ addition to oak diet did not alter development rates and caused an increase in pupal weights.  相似文献

Artificial diets have been developed for Lucilia sericata (Meigen) blowfly larvae; however, diets for adults have not yet been developed. An adult diet that excludes animal tissues and animal‐derived ingredients and promotes not only ovarian development, but also oviposition, would aid in basic research and maggot debridement therapy. We have successfully developed artificial diets that exclude animal tissues and animal‐derived ingredients for L. sericata adults. The outcomes of the diets were comparable with those of a beef liver diet in terms of oviposition, adult survival and number of offspring.  相似文献

Abstract: Three questions regarding possible benefits of mixed diets for the specialist aphid predator, Coccinella septempunctata larvae were investigated. (1) Do aphids species from different host plants complement each other nutritionally? (2) Is a mixed diet of high‐quality aphids beneficial? (3) How does the quality of mixed diets depend on the quality of constituent species? All mix‐combinations of aphid species of high (Metopolophium dirhodum), intermediate (Myzus persicae), and poor food quality (Aphis sambuci), and the three single‐species diets were compared. A mixed diet of two high‐quality species (Sitobion avenae and M. dirhodum) was also compared with single‐species diets. Larvae that were given pure A. sambuci and a mixed diet of A. sambuci + M. persicae died within 18 days and none of the larvae developed to fourth instar. Metopolophium dirhodum was generally of higher quality as food than M. persicae, whereas the mixed diet of M. dirhodum + M. persicae was intermediate. Sitobion avenae and M. dirhodum were found to have approximately the same food value. Coccinella septempunctatam larvae that were offered a mixed diet of these two high‐quality aphids gained no extra advantage. Overall, no benefit from mixing of aphid species was found. The quality of mixed diets depended on the quality of the constituent species.  相似文献

Little is known about the larviculture of the chub, Leuciscus cephalus (L.), an endangered cyprinid species endemic to European flowing waters. The use of decapsulated Artemia cysts as food for chub larviculture was investigated. After 3‐day feeding with the rotifer Brachionus calyciflorus, the larvae were fed on different diets: (i) dried decapsulated Artemia cysts, (ii) Artemia nauplii, (iii) rotifers for seven more days and then Daphnia collected from a pond, and (iv) an artificial diet. After a 24‐day rearing period, the highest survival rate was obtained with the larvae receiving decapsulated Artemia cysts. Feeding of the larvae with an artificial diet resulted in a significantly lower survival rate compared with the other groups. At the end of the experiment, the larvae fed on Artemia nauplii yielded a significantly higher mean length compared with the other groups. Feeding an artificial diet resulted in a significantly lower average weight and mean length gain compared with the other groups.  相似文献

The ladybird Hippodamia variegata Goeze is a widespread species found in many parts of the world. It is an efficient predator which feeds on a wide range of pests, specially aphids and other sap feeders. Mass rearing of this predator, in order to use in biological control programmes, entails a great deal of difficulties, among which are the aspects associated with nutrition. Applying artificial diets is of a great importance in mass rearing of biological control agents. We investigated the use of 15 artificial diets that included three separated experiments, in order to rear larval stages of this ladybird. The survival rate and developmental time were assessed on each treatment. The survival rate of the larvae on best diet was 53.30, 93.33, and 93.33% in different experiments. The developmental time in the experiment 1, 2 and 3 for the larva fed on the Aphis fabae was 15.59, 15.51 and 15.94, respectively, but when nourished by the best artificial diets, this factor was 26.59, 26.60 and 20.59, respectively. Developmental time for the larvae fed on A. fabae was significantly shorter than developmental time of larvae on artificial diets. Our results showed that artificial diets have the capacity to support the larval development to adulthood.  相似文献

This study examined larval food habits of two syntopic, related, carabids Amara chalcites Dejean and A. congrua Morawitz using a laboratory-rearing experiment, and the results were compared with those of a previous stable-isotope analyses of the same species. Larvae were reared on six different diets (seeds of Capsella bursa-pastoris (Linnaeus) Medikus, Stellaria media (Linnaeus) Villars, Taraxacum officinale Weber ex Wiggers; mixed seeds; Tenebrio molitor Linnaeus larvae; and Tenebrio larvae + mixed seeds). For both species, beetles fed seed diets exhibited moderate to high survival rates, with the exception of those fed Stellaria seeds, on which A. congrua showed low survival rates. The pure-animal diet resulted in high survival rates of A. chalcites but low survival rates of A. congrua. These results were consistent with those of the isotope analysis, in which A. chalcites larvae were more carnivorous than were A. congrua larvae, and the larvae of the two species appeared to prefer different types of seeds. In contrast, the effect of diet on adult weight differed between the two methods. In the laboratory experiments, supplementing seeds with animal food during the larval stage positively affected adult weight for both species, whereas in the isotope analysis, the positive effect only occurred in A. chalcites. Possible explanations for the discrepancy between the two methods and suggestions for future research are provided.  相似文献

Effects of dietary nicotine and macronutrient ratio on M. sexta larvae were examined. Larvae were fed a carbohydrate-biased, protein-biased or diet having equal amounts of casein and sucrose, with and without nicotine. Without nicotine, larvae displayed compensatory feeding on the low protein diet, but despite consuming more, grew least on this diet. Nicotine at 0.5% had no effect on nutrient consumption. Nicotine at 1.0 and 2.0% reduced overall consumption and thereby also reduced nicotine consumption. Larvae parasitized by C congregata displayed reduced nutrient intake and growth on all diets. Parasitized larvae responded to 1% nicotine similarly to unparasitized larvae. At 0.5% nicotine, they displayed reduced consumption on all diets, possibly due to altered chemoreceptor sensitivity to nicotine. When offered a choice of two diets having different macronutrient ratios, one with and the other without 0.1% nicotine, all larvae preferred the diet lacking nicotine and failed to regulate nutrient intake such that the nutrient intake target, a ratio of nutrients supporting optimal growth, was achieved. Parasitized larvae consumed less nicotine on a fresh weight basis than unparasitized insects, suggesting that the feeding response of parasitized larvae to nicotine minimizes the exposure of nicotine to developing parasites.  相似文献
