Ecologically, sea urchins are an important species in marine habitats around the world. Economically, sea urchins are prized for their gonads (uni). With fisheries declining worldwide, intensive sea urchin culture has been proposed. For urchins in commercial culture, any factor that affects survivorship negatively should be addressed to maximize cost benefit. One potential obstacle to optimizing culture of sea urchins is cannibalism. Approximately 2000 adult and juvenile Lytechinus variegatus (1 g-45 g) were collected from Port Saint Joseph Peninsula State Park, FL between June and September 2009. Urchins were held in recirculating tanks at different sizes, densities, and feeding regimes for 4 weeks. Starvation and high density contributed to the highest level of cannibalism among small (12-21 g) urchins (percent cannibalism = 18.8%), whereas fed, high density conditions contributed to the highest level of cannibalism among large (32-37 g) urchins (percent cannibalism = 18.4%). These results suggest that (1) small urchins cannibalize at higher rates than large urchins, and (2) increased density is an important contributing factor leading to cannibalism. We quantified stress, defined as a decrease in production as a result of environmental conditions, by evaluating weight gain within each treatment and suggest that weight loss or minimal weight gain is an indicator of stress. We hypothesize increased stress caused by competitive interference can lead to increased cannibalism and decreased growth rates, even when food is not limiting. Ecologically, there are no reports of cannibalism of urchins in wild populations. Consequently, the role of cannibalism in regulating sea urchin community structure is not known. However, factors affecting cannibalism of L. variegatus in the laboratory may provide insight into the conditions that could result in cannibalism in wild populations. From an aquaculture perspective, it is important to determine those factors that contribute to the incidence of cannibalism in sea urchins so that the appropriate culture conditions can be maintained to reduce the incidence of cannibalism.  相似文献

Biological differences between cell types and developmental processes are characterised by differences in gene expression profiles. Gene-distal enhancers are key components of the regulatory networks that specify the tissue-specific expression patterns driving embryonic development and cell fate decisions, and variations in their sequences are a major contributor to genetic disease and disease susceptibility. Despite advances in the methods for discovery of putative cis-regulatory sequences, characterisation of their spatio-temporal enhancer activities in a mammalian model system remains a major bottle-neck. We employed a strategy that combines gnathostome sequence conservation with transgenic mouse and zebrafish reporter assays to survey the genomic locus of the developmental control gene PAX6 for the presence of novel cis-regulatory elements. Sequence comparison between human and the cartilaginous elephant shark (Callorhinchus milii) revealed several ancient gnathostome conserved non-coding elements (agCNEs) dispersed widely throughout the PAX6 locus, extending the range of the known PAX6 cis-regulatory landscape to contain the full upstream PAX6-RCN1 intergenic region. Our data indicates that ancient conserved regulatory sequences can be tested effectively in transgenic zebrafish even when not conserved in zebrafish themselves. The strategy also allows efficient dissection of compound regulatory regions previously assessed in transgenic mice. Remarkable overlap in expression patterns driven by sets of agCNEs indicates that PAX6 resides in a landscape of multiple tissue-specific regulatory archipelagos.  相似文献

Cyclin D genes regulate the cell cycle, growth and differentiation in response to intercellular signaling. While the promoters of vertebrate cyclin D genes have been analyzed, the cis-regulatory sequences across an entire cyclin D locus have not. Doing so would increase understanding of how cyclin D genes respond to the regulatory states established by developmental gene regulatory networks, linking cell cycle and growth control to the ontogenetic program. Therefore, we conducted a cis-regulatory analysis on the cyclin D gene, SpcycD, of the sea urchin, Strongylocentrotus purpuratus, during embryogenesis, identifying upstream and intronic sequences, located within six defined regions bearing one or more cis-regulatory modules each.  相似文献

Previously starved urchins, Lytechinus variegatus, (36.0 ± 0.8 (SE) mm test diameter) were held in replicated (3) 10-L aquaria with artificial seawater at 22 ± 2  °C and 32‰ salinity and fed three diet treatments. Urchins were fed diets containing 9 : 35, 20 : 23 or 31 : 12% dry protein: % dry carbohydrate (P : C) ad libitum for a 65-day period. Gonads from urchins fed the 9 : 35 P : C diet had similar organic, lower ash, and lower water content than urchins fed the 31 : 12 P : C protein diet. Water content varied with both diet and nutritional history; consequently, water content may have limited value as a predictor of gonad nutritional status. Protein and carbohydrate concentrations in the gonad were directly related to the dietary composition of these nutrients; gonad lipids did not vary with diet. Excess carbohydrates are frequently stored as fats in fish and mammals but this does not appear to be the case in L. variegatus. Test carbohydrate storage and gut protein storage also reflected dietary composition. Image analysis of ovaries indicated decreased nutritive phagocyte volume, increased germinal epithelium volume and larger oocyte diameters in urchins fed high protein, low carbohydrate diets. Analysis of testes also indicated decreased nutritive phagocyte volume and increased gamete volume with urchins fed high protein, low carbohydrate diets, but differences among treatments were less obvious than in ovaries. This study suggests that high protein, low carbohydrate diets promote gamete growth and development. In addition, the biochemical and gametic composition of gonads can be altered by manipulating dietary composition. This could affect the quality and value of sea urchin roe for human consumption.  相似文献

Questions surround the mechanism of mutually exclusive expression by which Plasmodium falciparum mediates activation and silencing of var genes. These encode PfEMP1 proteins, which function as cytoadherent and immunomodulatory molecules at the surface of parasitised erythrocytes. Current evidence suggests that promoter silencing by var introns might play a key role in var gene regulation. To evaluate the impact of cis-acting regulatory regions on var silencing, we generated P. falciparum lines in which luciferase was placed under the control of an UpsA var promoter. By utilising the Bxb1 integrase system, these reporter cassettes were targeted to a genomic region that was not in apposition to var subtelomeric domains. This eliminated possible effects from surrounding telomeric elements and removed the variability inherent in episomal systems. Studies with highly synchronised parasites revealed that the UpsA element possessed minimal activity in comparison with a heterologous (hrp3) promoter. This may result from the integrated UpsA promoter being largely silenced by the neighbouring cg6 promoter. Our analyses also revealed that the DownsA 3′ untranslated region further decreased the luciferase activity from both cassettes, whereas the var A intron repressed the UpsA promoter specifically. By applying multivariate analysis over the entire cell cycle, we confirmed the significance of these cis-elements and found the parasite stage to be the major factor regulating UpsA-promoter activity. Additionally, we observed that the UpsA promoter was capable of nucleating reversible silencing that spread to a downstream promoter. We believe these studies are the first to analyse promoter activity of Group A var genes, which have been implicated in severe malaria, and support the model that var introns can further suppress var expression. These data also suggest an important suppressive role for the DownsA terminator. Our findings imply the existence of multiple levels of var gene regulation in addition to intrinsic promoter-dependent silencing.  相似文献
