In leaves of field-grown grapevine, the contents of chlorophyll, carotenoids, and soluble proteins and the activities of ribulose-1,5-bisphosphate carboxylase (RuBPC) and nitrate (NR) and nitrite (NiR) reductases were decreased in phytoplasma-infected leaves, but the contents of soluble sugars and total saccharides were markedly increased. In isolated thylakoids, phytoplasma caused marked inhibition of whole chain and photosystem 2 (PS2) activities. The artificial exogenous electron donor, diphenyl carbazide, significantly restored the loss of PS2 activity in infected leaves.  相似文献

Chlorophyllin (Chlin), the Mg-chlorin obtained from chlorophyll (Chl) was employed as substrate of Mg-dechelatase. The release of Mg2+ was associated with a shift of absorption from 644 to 687 nm. Changes of absorption at 687 nm were taken as a measure of Mg-dechelatase activity present in preparations of oilseed rape thylakoids. Absorption changes were correlated linearly with enzyme dose. The pH optimum of Mg release from Chlin was ca 9 with a broad flank down to pH 7. The reaction showed saturation kinetics with an apparent Km value of ca 17 n M . The activity was inhibited in the presence of cysteamine or reduced glutathion. There was no effect of the thiol reagent N-ethyl maleimide. The bulk of dechelatase activity was associated with the chloroplast membranes. The enzyme is partially latent and the appearance of full activity requires the solubilization of thylakoids with detergent. The highest activities were detected in mature green rape cotyledons. During dark-induced senescence the activity declined at roughly the same rate as Chl was lost in the leaf tissue.  相似文献

Three minor Chl a proteins were detected in electrophoretic profiles from wild-type maize thylakoids. The spectral characteristics of these Chl proteins and the apparent molecular weights of their constituent apoproteins suggested that they were associated with the Photosystem-II reaction center. One of these Chl a-proteins, CPa-1, was present in wild-type thylakoids and a photochemically active Photosystem-II particle, but was missing from thylakoids of a mutant-lacking Photosystem-II reaction center. CPa-2, on the other hand, was enriched in mutant thylakoids but was completely missing from the Photosystem-II particles. We conclude that CPa-1 is most likely to contain the photoactive chlorophyll of Photosystem II, while CPa-2 is not required for Photosystem-II activity. The apparent molecular weights of the major CPa-1 and CPa-2 apoproteins were 48 000 and 42 000, respectively. The third minor Chl protein seems most likely to be an electrophoretic variant of CPa-1 and has been designated CPa-11. Seven other Chl proteins were detected in wild-type profiles. Many of these Chl proteins appeared to be oligomers or highly order complexes of LHCP and CP-1.  相似文献

In leaves of four tomato (Lycopersicon esculentum Mill.) cultivars (Red Cloud, Floradade, Peto 95, and Scorpio) the contents of chlorophyll (Chl) (a+b), Chl a, and -carotene decreased due to 100 mM NaCl treatment as compared with those of controls. The contents of soluble sugars and total saccharides were significantly increased in leaves of NaCl-treated plants, but the starch content was not significantly affected. Transmission electron microscopy indicated that in leaves of NaCl-treated plants, the chloroplasts were aggregated, the cell membranes were distorted and wrinkled, and there was no sign of grana and thylakoid structures in chloroplasts.  相似文献

The functional organization of competent photosynthetic units in developing thylakoids from intermittent-light grown pea as well as in the unstacked, stacked and phosphorylated stacked thylakoids from its mature chloroplasts was characterized by polarographic measurements of action spectra, reaction centre contents and optical cross-sections for PS I-mediated O2 uptake and PS II-mediated O2 evolution. The minimum antenna sizes of 60 and 37 chlorophyll a molecules for PS I and PS II, respectively, were determined in developing thylakoids with a ratio of Chl a/Chl b>50. In mature chloroplasts, the embedded light-harvesting chlorophyll a/b-binding (LHC) protein complexes increased the PS I and PS II effective antenna sizes by 3–6 times depending on the thylakoid membrane organization. In unstacked thylakoids, a randomization of PS I, PS II and LHC II led to the most uniform spectral distribution of light harvesting between the two photosystems but caused the maximal difference of their antenna sizes to be 370 and 100 Chls for the competent PS I and PS II units, respectively. Following the Mg2+-induced stacking of thylakoids, opposite complementary changes of the action spectra, antenna sizes and Chl a/Chl b ratios indicated a redistribution of a LHC II pool of 100 Chl ( a + b) molecules from PS I to PS II. Unlike to the stroma-exposed PS II in unstacked thylakoids, the granal PS II units of 200 Chls demonstrated an additional 2-fold increase of the effective antenna size due to energy transfer within PS II dimers under strong background illumination, which closed >90% of reaction centres. Protein phosphorylation of the stacked thylakoids induced a significant inactivation of the O2-evolving PS II centres but did not cause complementary changes of the action spectra and antenna sizes of the competent PS I and PS II. In this case, light harvesting parameters of the O2-evolving PS II units were nearly unaffected, whereas the obvious relative increase of the PS I activity at 650 nm and its decrease at >700 nm both in the action spectrum and optical cross-section measurements might suggest a substitution of PS I units in the O2-reducing fraction by another distinct fraction of -type which in turn is not the same to PS I units in unstacked thylakoids.  相似文献

Changes in contents of pigments, chlorophyll-protein complex, and photosynthetic activities were investigated in field grown apple (Malus pumila Mill.) leaves infected by Apple Proliferation phytoplasma. The contents of chlorophyll a+b (Chl) and carotenoids (Car) markedly decreased in infected leaves. Similar results were also observed for content of total soluble proteins and ribulose-1,5-bisphosphate carboxylase activity. When various photosynthetic activities were followed in isolated thylakoids, phytoplasma infection caused a marked inhibition of whole chain and photosystem 2 (PS2) activity. Smaller inhibition of photosystem 1 (PS1) activity was observed even in severely infected leaves. The artificial exogenous electron donors, MnCl2 diphenyl carbazide, and NH2OH, did not restore the loss of PS2 activity in both mildly and severely infected leaves. Similar results were obtained by Chl fluorescence measurements. The marked loss of PS2 activity in infected leaves was due to the reduction of contents of chlorophyll and light-harvesting chlorophyll-protein 2 complexes. A variety of unicellular algae, thylakoids from higher plants in different stages of maturity and isolated pigment-protein complexes were oriented in stretched polyvinyl alcohol films. Low temperature linear dichroism (LD) spectra of Chlorella pyrenoidosa and higher plant thylakoids in the films were very similar to those obtained after orientation of similar samples using magnetic or electric fields. Positive LD bands corresponding to Chl a (670) and (682) and negative bands due to Chl a (658) and Chl b(648) were resolved in spectra of the light harvesting Chl a/b protein. Chl b (648) and Chl a (658) and (670) were not seen in the LD spectrum of thylakoids from plants grown in intermittent light, the Chl b-less mutant of barley, Euglena gracilis or the cyanobacteria, Phormidium luridum and Anacystis nidulans, but did appear upon chloroplast maturation in Romaine lettuce and during the greening of etiolated and intermittent light plants. The highly oriented long wavelength Chl a (682) in the light-harvesting complex may represent residual PS II whose peak dichroism is centered at 681 nm. The PS I preparation had a Chl a/b ratio of approx. 6 and the LD spectrum was positive with a maximum at 690-694 nm and a band of lower amplitude at 652 nm. The minor LD band was not observed in PS I preparations from organisms that lack chl b such as the cyanobacteria, intermittent light plants and the Chl b-less mutant of barley. We suggest that the 652 nm band is due to Chl b molecules associated with the antenna of PS I and are distinct from those on the light harvesting complex whose orientation is different. We also conclude that all the Chl a forms are oriented and that the long geometric axes of the pigment-protein complexes, as deduced from the configuration they assume in the stretched films, are axes that normally lie parallel to the plane of the native thylakoid.  相似文献

In order to obtain information on the organization of the pigment molecules in chlorophyll (Chl) a/b/c-containing organisms, we have carried out circular dichroism (CD), linear dichroism (LD) and absorption spectroscopic measurements on intact cells, isolated thylakoids and purified light-harvesting complexes (LHCs) of the prasinophycean alga Mantoniella squamata. The CD spectra of the intact cells and isolated thylakoids were predominated by the excitonic bands of the Chl a/b/c LHC. However, some anomalous bands indicated the existence of chiral macrodomains, which could be correlated with the multilayered membrane system in the intact cells. In the red, the thylakoid membranes and the LHC exhibited a well-discernible CD band originating from Chl c, but otherwise the CD spectra were similar to that of non-aggregated LHC II, the main Chl a/b LHC in higher plants. In the Soret region, however, an unusually intense (+) 441 nm band was observed, which was accompanied by negative bands between 465 and 510 nm. It is proposed that these bands originate from intense excitonic interactions between Chl a and carotenoid molecules. LD measurements revealed that the Q(Y) dipoles of Chl a in Mantoniella thylakoids are preferentially oriented in the plane of the membrane, with orientation angles tilting out more at shorter than at longer wavelengths (9 degrees at 677 nm, 20 degrees at 670 nm and 26 degrees at 662 nm); the Q(Y) dipole of Chl c was found to be oriented at 29 degrees with respect to the membrane plane. These data and the LD spectrum of the LHC, apart from the presence of Chl c, suggest an orientation pattern of dipoles similar to those of higher plant thylakoids and LHC II. However, the tendency of the Q(Y) dipoles of Chl b to lie preferentially in the plane of the membrane (23 degrees at 653 nm and 30 degrees at 646 nm) is markedly different from the orientation pattern in higher plant membranes and LHC II. Hence, our CD and LD data show that the molecular organization of the Chl a/b/c LHC, despite evident similarities, differs significantly from that of LHC II.  相似文献

Wheat (Triticum aestivum L. Sakha 61) grains were exposed to microwave radiation of a wavelength 2.85 cm and frequency 10.525 GHz for 15, 45, or 75 min. The exposed grains were germinated and then harvested after 7 and 14 d. While 15-and 45-min exposure of the grains stimulated seedling shoot length and fresh and dry masses, the exposure for 75 min had no pronounced effects. 15 and 75-min irradiation increased succulence and pigment contents in 7-and 14-d-old seedlings. While the ratios chlorophyll (Chl) a/b, carotenoids (Car)/Chl were higher in 7-d-old seedlings than in the control ones for all doses, they decreased in 14-d-old seedlings. The microwave radiation increased protein and amino acid contents, but decreased the contents of saccharides, nucleic acids, and phenolic compounds. Low dose (15 min) stimulated proline synthesis, whereas the other doses showed negative effect on its production.  相似文献

Ribulose-1,5-bisphosphate carboxylase (RuBPC) activity and contents of soluble proteins were also significantly lower in shade needles. In isolated thylakoids, a marked lower rate of whole chain and photosystem (PS) 2 activities were observed in shade needles. Smaller lower rate of PS1 activity was also observed in shade needles. The artificial exogenous electron donors, diphenyl carbazide (DPC) and NH2OH, significantly restored the loss of PS2 activity in shade needles. Similar results were obtained when Fv/Fm was evaluated by Chl fluorescence measurements. The marked lower rate of PS2 activity in shade needles was due to the lower contents of 47, 33, 28–25, 23, and 17 kDa polypeptides. This conclusion was confirmed by immunological studies showing that the content of the 33 kDa protein of the watersplitting complex was diminished significantly in shade needles.  相似文献

In grapevine (Vitis vinifera L.) leaf chlorophyll (Chl) a and Chl b and carotenoid contents were higher in plants grown at low photon flux densities (PFD) than in those grown at medium and high PFD. The highest Chl a variable to maximum fluorescence ratio Fv/Fm was observed in plants grown at medium PFD while the minimum fluorescence F0 was highest in those at high PFD. In isolated thylakoids, both high and low PFD caused marked inhibition of whole chain and photosystem 2 (PS2) activities. Cadmium (Cd) and kinetin (Kin) significantly affected the growth and contents of chlorophyll (Chl) and of soluble and reserve (hydrolysable) saccharides in sunflower plants. Cd-treated plants had lower contents of Chl and soluble saccharides and produced less dry matter than control (Cd-untreated) plants. Chla stability to heat (CSI) increased at all Cd concentrations. The same was true for Chlb stability (0–10 μM Cd). Spraying sunflower shoots with Kin solutions counteracted the deleterious effects of Cd. Kin application enhanced the Chla andb contents, Chla/b ratio, content of soluble saccharides and dry matter, and to less extent Chl stability. The relative role of Kin in affecting the parameters tested (as indicated by η2 values) was predominant while that of Cd was subsidiary except for Chla stability. The role of Cd×Kin interaction was dominant for hydrolysable saccharides. Hence spraying shoots of plants grown under increasing Cd concentration with Kin can partially alleviate inhibitory effects of cadmium.  相似文献

The effect of iron deficiency on photosynthetic pigments, ribulose-1,5-bisphosphate carboxylase (RuBPC), and photosystem activities were investigated in field grown grapevine (Vitis vinifera L. cv. Pinot noir) leaves. The contents of chlorophyll (Chl) (a+b) and carotenoids per unit fresh mass showed a progressive decrease upon increase in iron deficiency. Similar results were also observed in content of total soluble proteins and RuBPC activity. The marked loss of large (55 kDa) and small (15 kDa) subunits of RuBPC was also observed in severely chlorotic leaves. However, when various photosynthetic electron transport activities were analysed in isolated thylakoids, a major decrease in the rate of whole chain (H2O methyl viologen) electron transport was observed in iron deficient leaves. Such reduction was mainly due to the loss of photosystem 2 (PS2) activity. The same results were obtained when Fv/Fm was evaluated by Chl fluorescence measurements in leaves. Smaller inhibition of photosystem 1 (PS1) activity was also observed in both mild and severely chlorotic leaves. The artificial electron donors, diphenyl carbazide and NH2OH, markedly restored the loss of PS2 activity in severely chlorotic leaves. The marked loss of PS2 activity was evidently due to the loss of 33, 23, 28-25, and 17 kDa polypeptides in iron deficient leaves.  相似文献

The grapevine (Vitis vinifera L. cv. Riesling) plants subjected to water deficit were studied for changes in relative water content (RWC), leaf dry mass, contents of chlorophyll (Chl), total leaf proteins, free amino acids, and proline, and activities of ribulose-1,5-bisphosphate carboxylase (RuBPC), nitrate reductase (NR), and protease. In water-stressed plants RWC, leaf dry matter, Chl content, net photosynthetic rate (P N), and RuBPC and NR activities were significantly decreased. The total leaf protein content also declined with increase in the accumulation of free amino acids. Concurrently, the protease activity in the tissues was also increased. A significant two-fold increase in proline content was recorded.  相似文献

The impacts of solar UV (280–400 nm) radiation on photosynthetic activities and polypeptide composition of thylakoids of cluster bean (Cyamopsis tetragonoloba L, UV-B sensitive) and black gram (Vigna mungo L., UV-B resistant) plants were compared. The activity of photosystem 1 and especially photosystem 2 increased in cluster bean while decreased in black gram, when either UV-B or UV-B + UV-A radiation was removed as compared to control plants. Exclusion of UV-B radiation caused changes in the protein composition of the thylakoids particularly in the 33, 23 and 17 kDa proteins of photosystem 2.  相似文献

Carbonic anhydrase (CA) activity of pea thylakoids, thylakoid membranes enriched with photosystem I (PSI-membranes), or photosystem II (PSII-membranes) as well as both supernatant and pellet after precipitation of thylakoids treated with detergent Triton X-100 were studied. CA activity of thylakoids in the presence of varying concentrations of Triton X-100 had two maxima, at Triton/chlorophyll (triton/Chl) ratios of 0.3 and 1.0. CA activities of PSI-membranes and PSII-membranes had only one maximum each, at Triton/Chl ratio 0.3 or 1.0, respectively. Two CAs with characteristics of the membrane-bound proteins and one CA with characteristics of the soluble proteins were found in the medium after thylakoids were incubated with Triton. One of the first two CAs had mobility in PAAG after native electrophoresis the same as that of CA residing in PSI-membranes, and the other CA had mobility the same as the mobility of CA residing in PSII-membranes, but the latter was different from CA situated in PSII core-complex (Ignatova et al. 2006 Biochemistry (Moscow) 71:525–532). The properties of the “soluble” CA removed from thylakoids were different from the properties of the known soluble CAs of plant cell: apparent molecular mass was about 262 kD and it was three orders more sensitive to the specific CA inhibitor, ethoxyzolamide, than soluble stromal CA. The data are discussed as indicating the presence of, at least, four CAs in pea thylakoids.  相似文献

As compared with the swamp reed (SR) ecotype of Phragmites communis growing in the desert region of northwest China, plants of the dune reed (DR) ecotype from the same region possessed lower chlorophyll (Chl) content in leaves, and less thylakoids and grana stacks in chloroplasts. Tube gel electrophoresis without stain showed that the contents of Chl-protein (Pro) components related to photosystem 2 (PS2) were markedly lower in the DR thylakoid membranes than in the SR thylakoid membranes, while the contents of Chl-Pro components associated with PS1 were almost the same in both types. SDS-PAGE analysis indicated that the content of polypeptides of the light-harvesting Chl a/b complex of PS2 (LHC2) was lower in the DR thylakoids. Besides, the conformation of LHC2 within the DR thylakoid membranes was also altered as indicated by circular dichroism spectra. Hence in the DR, reduced energy harvesting by declining the size of LHC2 might be responsible for the down-regulated PS2 activity. Chl fluorescence parameters. Fv/Fm and quantum efficiency of PS2 (PS2), were lower in the DR leaves than in the SR ones. However, non-photochemical quenching coefficient (qN) was greater in DR than that in SR, implying other energy dissipation way exists in the DR photosynthetic membranes.  相似文献

Thylakoids isolated from winter rye (Secale cereale L. cv Puma) grown at 20°C (nonhardened rye, RNH) or 5°C (cold-hardened rye, RH) were characterized using chlorophyll (Chl) fluorescence. Low temperature fluorescence emission spectra of RH thylakoids contained emission bands at 680 and 695 nanometers not present in RNH thylakoids which were interpreted as changes in the association of light-harvesting Chl a/b proteins and photosystem II (PSII) reaction centers. RH thylakoids also exhibited a decrease in the emission ratio of 742/685 nanometers relative to RNH thylakoids.

Room temperature fluorescence induction revealed that a larger proportion of Chl in RH thylakoids was inactive in transferring energy to PSII reaction centers when compared with RNH thylakoids. Fluorescence induction kinetics at 20°C indicated that RNH and RH thylakoids contained the same proportions of fast (α) and slow (β) components of the biphasic induction curve. In RH thylakoids, however, the rate constant for α components increased and the rate constant for β components decreased relative to RNH thylakoids. Thus, energy was transferred more quickly within a PSII reaction center complex in RH thylakoids. In addition, PSII reaction centers in RH thylakoids were less connected, thus reducing energy transfers between reaction center complexes. We concluded that both PSII reaction centers and light-harvesting Chl a/b proteins had been modified during development of rye chloroplasts at 5°C.

Recent studies have shown that coleoptile chloroplasts operate the xanthophyll cycle, and that their zeaxanthin concentration co-varies with their sensitivity to blue light. The present study characterized the distribution of photosynthetic pigments in thylakoid pigment–protein complexes from dark-adapted and light-treated coleoptile and mesophyll chloroplasts, the low temperature fluorescence emission spectra, and the rates of PS I and PS II electron transport in both types of chloroplasts from 5-day-old corn seedlings. Pigments were extracted from isolated PS I holocomplex, LHC IIb trimeric and LHC II monomeric complexes and analyzed by HPLC. Chlorophyll distribution in coleoptile thylakoids showed 31% of the total collected Chl in PS I and 65% in the light harvesting complexes of PS II. In mesophyll thylakoids, the values were 44% and 54%, respectively. Mesophyll and coleoptile PS I holocomplexes differed in their Chl t a/Chl t b ratios (8.1 and 6.1, respectively) and -carotene content. In contrast, mesophyll and coleoptile LHC IIb trimers and LHC II monomers had similar Chl t a/Chl t b ratios and -carotene content. The three analyzed pigment–protein complexes from dark-adapted coleoptile chloroplasts contained zeaxanthin, whereas there was no detectable zeaxanthin in the complexes from dark-adapted mesophyll chloroplasts. In both chloroplast types, zeaxanthin and antheraxanthin increased markedly in the three pigment–protein complexes upon illumination, while violaxanthin decreased. In mesophyll thylakoids, zeaxanthin distribution as a percentage of the xanthophyll cycle pool was: LHC II monomers > LHC IIb trimers > PS I holocomplex, and in coleoptile thylakoids, it was: LHC IIb trimers > LHC II monomers = PS I holocomplex. Low temperature (77 K) fluorescence emission spectra showed that the 686 nm emission of coleoptile chloroplasts was approximately 50% larger than that of mesophyll chloroplasts when normalized at 734 nm. The pigment and fluorescence analysis data suggest that there is relatively more PS II per PS I and more LHC I per CC I in coleoptile chloroplasts than in mesophyll chloroplasts. Measurements of t in vitro uncoupled photosynthetic electron transport showed approximately 60% higher rates of electron flow through PS II in coleoptile chloroplasts than in mesophyll chloroplasts. Electron transport rates through PS I were similar in both chloroplast types. Thus, when compared to mesophyll chloroplasts, coleoptile chloroplasts have a distinct PS I pigment composition, a distinct chlorophyll distribution between PS I and PS II, a distinct zeaxanthin percentage distribution among thylakoid pigment–protein complexes, a higher PS II-related fluorescence emission, and higher PS II electron transport capacity.
