The separate package and spillover models of photosynthetic enhancement are generalized and extended to light saturation assuming hyperbolic or exponential rate versus intensity curves. The available data are more consistent with the exponential model at high light intensities. Two significant conclusions follow: (a) the decreases in enhancement observed at high light intensities are accounted for by the curvatures of the light intensity curves for photosynthesis, and (b) nonlinearity of rate versus intensity curves may be sufficient to reduce enhancement greatly even when the intensity curves appear linear.  相似文献

1. When copper chloride is allowed to act on Aspergillus niger there is at first a period during which there is no change in the rate of the production of carbon dioxide, following which the rate of respiration falls. The interval of no change is called the latent period. 2. When the copper is removed from the external solution before the end of the latent period this interval is prolonged. The rate of respiration then falls to a new level below the normal level. 3. Experiments on Nitella and on Valonia indicate that the copper penetrates the cell almost immediately. 4. The length of the latent period varies inversely as a constant power of the concentration of the copper. 5. These results are explained by assuming that the copper is made active in the respiration system by means of a reversible reaction. By using appropriate velocity constants the experimental curves can be duplicated by calculated curves.  相似文献

Water deficit is one of the major limiting factors in vegetation recovery and restoration in loess, hilly-gully regions of China. The light responses of photosynthesis in leaves of two-year old Prunus sibirica L., Hippophae rhamnoides L., and Pinus tabulaeformis Carr. under various soil water contents were studied using the CIRAS-2 portable photosynthesis system. Light-response curves and photosynthetic parameters were analyzed and fitted using the rectangular hyperbola model, the exponential model, the nonrectangular hyperbola model, and the modified rectangular hyperbola model. Under high light, photosynthetic rate (P N) and stomatal conductance (g s) were steady and photoinhibition was not significant, when the relative soil water content (RWC) varied from 56.3–80.9%, 47.9–82.9%, and 33.4–92.6% for P. sibirica, H. rhamnoides, and P. tabulaeformis, respectively. The light-response curves of P N, the light compensation point (LCP), and the dark respiration rate (R D) were well fitted using the above four models. The nonrectangular hyperbola was the best model in fitting the data; the modified rectangular hyperbola model was the second, and the rectangular hyperbola model was the poorest one. When RWC was higher or lower than the optimal range, the obvious photoinhibition and significant decrease in P N with increasing photosynthetic photon flux density (PPFD) were observed in all three species under high light. The light saturation point (LSP) and apparent quantum yield also decreased significantly, when the upper limit of PPFD was 200 μmol m?2 s?1. Under these circumstances, only the modified rectangular hyperbola model was able to fit well the curves of the light response, LCP, LSP, R D, and light-saturated P N.  相似文献

Growth curves, calculated for field populations of B. glabrata, were not materially affected by habitat, altitude or season. A mean growth curve was therefore used to estimate the age-frequency distribution of snails in successive field samples. These data permitted the construction of ecological life tables and the estimation of r, the intrinsic rate of natural increase (or decrease) of the different populations. The calculated values of r were inserted in a simple model of unlimited population growth but the resultant curves poorly represented the observed data. A model for unlimited growth was more satisfactory for pond and marsh populations but, apparently, immigration made it less satisfactory for stream and banana drain populations. Nevertheless, r may still be of value in predicting repopulation rates in certain habitats after a mollusciciding which does not kill the entire snail population.  相似文献

The growth and survival of coniferous and broad-leaved trees were followed over a 5-yr period in a temperate old-growth mixed forest in Japan, and dynamic features of the forest were studied in relation to the life history of the dominants, the coniferous Abies homolepis and the broad-leaved Fagus crenata. During this period, the gap formation rate was 31m2 ha?1yr?1, the mortality of trees > 2m high was 1.7%/yr, and the rate of loss in basal area 1.4%/yr. These values were much higher than the recruitment, 0.3%/yr, and the total growth of surviving and new trees, 0.6%/yr, owing to the inhibition of regeneration by understorey dwarf bamboo (Sasa borealis). A transition matrix model based on DBH size classes predicts that the basal area of the forest will decrease by 14% in 50 yr, but that the DBH distribution of trees > 10 cm diameter will change little. Equilibrium DBH distributions assuming recruitment being equal to mortality, were quite different between broad-leaved and coniferous trees, reflecting different survivorship curves of the two dominants. The composition and structure of the forest may change depending on the pattern and frequency of disturbances, or episodic events, notably the synchronous death of Sasa borealis.  相似文献

Potassium conductance-voltage curves have been determined for a squid axon in high external potassium solution for a wide range of divalent cation concentrations. A decrease in divalent ion concentration shifts the conductance-voltage curve along the voltage axis in the direction of more hyperpolarized voltages by as much as 9 mv for an e-fold change in concentration. When the divalent ion concentration is less than about 5 mM, a further decrease does not cause a significant shift of the conductance-voltage curve. These results can be explained by assuming that on the outer surface of the membrane there is a negative fixed charge which can bind calcium ions, and that the axon is sensitive to the resulting double-layer potential. From our data, the best value for charge density was found to be one electronic charge per 120 square angstroms, and a lower limit to be one electronic charge per 280 square angstroms.  相似文献

Caldicellulosiruptor saccharolyticus displays superior H2 yields on a wide range of carbon sources provided that lactate formation is avoided. Nevertheless, a low lactate flux is initiated as the growth rate declined in the transition to the stationary phase, which coincides with a drastic decrease in the glucose consumption and acetate production fluxes. In addition, the decrease in growth rate was accompanied by a sudden increase and then decrease in NADH levels. The V′MAX of the lactate dehydrogenase (LDH) doubled when the cells entered the stationary phase. Kinetic analysis revealed that at the metabolic level LDH activity is regulated through (i) competitive inhibition by pyrophosphate (PPi, ki=1.7 mM) and NAD (ki=0.43 mM) and (ii) allosteric activation by FBP (300%), ATP (160%) and ADP (140%). From these data a MWC-based model was derived. Simulations with this model could explain the observed lactate shift by displaying how the sensitivity of LDH activity to NADH/NAD ratio varied with different PPi concentrations. Moreover, the activation of LDH by ATP indicates that C. saccharolyticus uses LDH as a means to adjusts its flux of ATP and NADH production. To our knowledge, this is the first time PPi is observed as an effector of LDH.  相似文献

Relative frequencies of large and small genome rearrangements (inversions and transpositions) in the evolution of prokaryotic genomes can be evaluated using the ratio between the index S (the ratio of the number of identical pairs of neighboring genes in two genomes to the total number of genes in the sample of interest) and 1–6L/n, where L is the mean difference in intergenic distances and n is the number of genes in the sample. The S value uniformly decreases with the fixation of genome rearrangements, while the decrease rate of 1–6L/n is determined by the rearrangement size. Specifically, large inversions and transpositions lead to a dramatic decrease in the index value, while small rearrangements result in an insignificant decrease. The ratio between these indices was computed for twenty pairs of closely related species belonging to different groups of bacteria and archaea. The pairs examined strongly differed in the relative frequency of large and small rearrangements. However, computer simulation showed that the total variation can be reproduced with the same input parameters of the model. This means that the differences observed can be stochastic and can be interpreted without assuming different mechanisms and factors of genome rearrangements for different groups of prokaryotes. Relative frequencies of large and small rearrangements displayed no noticeable correlations with taxonomic position, total rate of rearrangement fixation, habitation conditions, and the abundance of transposons and repetitive sequences. It is suggested that, in some cases, phage activity increases the frequency of large genome rearrangements.  相似文献

Analyses of survival data of a mixture of Salmonella spp. at fixed temperatures between 55°C (131°F) and 71.1°C (160°F) in ground beef matrices containing concentrations of salt between 0 and 4.5%, concentrations of sodium pyrophosphate (SPP) between 0 and 0.5%, and concentrations of sodium lactate (NaL) between 0 and 4.5% indicated that heat resistance of Salmonella increases with increasing levels of SPP and salt, except that, for salt, for larger lethalities close to 6.5, the effect of salt was evident only at low temperatures (<64°C). NaL did not seem to affect the heat resistance of Salmonella as much as the effects induced by the other variables studied. An omnibus model for predicting the lethality for given times and temperatures for ground beef matrices within the range studied was developed that reflects the convex survival curves that were observed. However, the standard errors of the predicted lethalities from this models are large, so consequently, a model, specific for predicting the times needed to obtained a lethality of 6.5 log10, was developed, using estimated results of times derived from the individual survival curves. For the latter model, the coefficient of variation (CV) of predicted times range from about 6 to 25%. For example, at 60°C, when increasing the concentration of salt from 0 to 4.5%, and assuming that the concentration of SPP is 0%, the time to reach a 6.5-log10 relative reduction is predicted to increase from 20 min (CV = 11%) to 48 min (CV = 15%), a 2.4 factor (CV = 19%). At 71.1°C (160°F) the model predicts that more than 0.5 min is needed to achieve a 6.5-log10 relative reduction.  相似文献

The role of nitrate as a promoter of germination of Sisymbrium officinale seeds was examined in optimal light conditions. It was shown that the requirement for nitrate was absolute. This was true for all seed lots used. The probit of germination in water was log-linearly related to the level of endogenous nitrate. Preincubation at 15°C resulted in an immediate decrease in germination, whereas in 25 millimolar KNO3 the decrease was delayed. The decline of germination in water was strongly correlated with the rate at which nitrate leached from the seeds. The germination response to a range of KNO3 concentrations was followed during preincubation at 24-hour intervals. During the entire 264-hour preincubation period increasingly higher nitrate concentrations were required to maintain a response. This resulted in a right-hand shift of the dose-response curve parallel to the x axis. After 120 hours the high maximum germination level started to decline. The dose-response curves could be simulated by an equation from the receptor-occupancy theory. It is proposed that induction of secondary dormancy is a result of a decrease of the number of nitrate receptors. After 24 and 48 hours of preincubation, the nitrate-response curves were biphasic. The biphasic character could be related to the level of endogenous nitrate and to a differential requirement for nitrate of two fractions of the seed population. Similarities with the behavior of fluence-response curves after prolonged dark incubation led to the hypothesis that phytochrome and nitrate share the same site of action.  相似文献

C3 photosynthesis at high light is often modeled by assuming limitation by the maximum capacity of Rubisco carboxylation (VCmax) at low CO2 concentrations, by electron transport capacity (Jmax) at higher CO2 concentrations, and sometimes by triose-phosphate utilization rate at the highest CO2 concentrations. Net photosynthetic rate (PN) at lower light is often modeled simply by assuming that it becomes limited by electron transport (J). However, it is known that Rubisco can become deactivated at less than saturating light, and it is possible that PN at low light could be limited by the rate of Rubisco carboxylation (VC) rather than J. This could have important consequences for responses of PN to CO2 and temperature at low light. In this work, PN responses to CO2 concentration of common bean, quinoa, and soybean leaves measured over a wide range of temperatures and PPFDs were compared with rates modeled assuming either VC or J limitation at limiting light. In all cases, observed rates of PN were better predicted by assuming limitation by VC rather than J at limiting light both below and above the current ambient CO2. One manifestation of this plant response was that the relative stimulation of PN with increasing the ambient CO2 concentration from 380 to 570 µmol mol–1 did not decrease at less than saturating PPFDs. The ratio of VC to VCmax at each lower PPFD varied linearly with the ratio of PN at low PPFD to PN at high PPFD measured at 380 µmol(CO2) mol–1 in all cases. This modification of the standard C3 biochemical model was much better at reproducing observed responses of light-limited PN to CO2 concentrations from pre-industrial to projected future atmospheric concentrations.  相似文献

Freeman-Sheldon syndrome (FSS) is characterized by congenital contractures resulting from dominant point mutations in the embryonic isoform of muscle myosin. To investigate its disease mechanism, we used Drosophila models expressing FSS myosin mutations Y583S or T178I in their flight and jump muscles. We isolated these muscles from heterozygous mutant Drosophila and performed skinned fiber mechanics. The most striking mechanical alteration was an increase in active muscle stiffness. Y583S/+ and T178I/+ fibers’ elastic moduli increased 70 and 77%, respectively. Increased stiffness contributed to decreased power generation, 49 and 66%, as a result of increased work absorbed during the lengthening portion of the contractile cycle. Slower muscle kinetics also contributed to the mutant phenotype, as shown by 17 and 32% decreases in optimal frequency for power generation, and 27 and 41% slower muscle apparent rate constant 2πb. Combined with previous measurements of slower in vitro actin motility, our results suggest a rate reduction of at least one strongly bound cross-bridge cycle transition that increases the time myosin spends strongly bound to actin, ton. Increased ton was further supported by decreased ATP affinity and a 16% slowing of jump muscle relaxation rate in T178I heterozygotes. Impaired muscle function caused diminished flight and jump ability of Y583S/+ and T178I/+ Drosophila. Based on our results, assuming that our model system mimics human skeletal muscle, we propose that one mechanism driving FSS is elevated muscle stiffness arising from prolonged ton in developing muscle fibers.  相似文献

A few smell deficient mutants of Drosophila have been characterized recently. In a series of experiments, we have compared the slow electrical response of the antenna (EAG) to calibrated concentrations of butanol and aliphatic n-acetates of olf C mutants with those of wild flies. This mutation was found to affect the dose-response curves specifically. Our data can be explained by assuming that this mutation affects the peripheral olfactory membrane at several levels.  相似文献

Edwardsiella tarda is one of the leading fish pathogens for the aquaculture industry. To realize efficient disease control of edwardsiellosis, a predictive model for E. tarda in seawater was developed. The modified logistic model was used to regress the growth curves of E. tarda JN at five different temperatures (range from 10 to 30 °C) and four organic nutrient concentrations (range from 5 to 40 mg l?1 measured by chemical oxygen demand (COD)). The modeling effects of temperature and COD on the specific growth rate (μ) were developed by square-root model and saturation-growth rate model, respectively. The growth model was validated in turbot aquaculture tanks by estimating the dynamics of inoculated E. tarda. The accurate feeding of probiotic Bacillus pumilus strain H2 was calculated based on the estimation of E. tarda. Results showed that the logistic model produced a good fit to the growth curves of E. tarda JN (average R2?=?0.962). The overall predictions based on above models agreed well with the growth curve of E. tarda JN observed by plate counting in the validation tests (average Af?=?1.16; average Bf?=?1.32). The use of predicted amount of B. pumilus (5.66 log CFU ml?1) successfully prevent the deterioration of disease for turbot with 13.3% mortality rate in a recirculating aquaculture system (RAS), while the feeding of 0 and 3.0 log CFU ml?1 of B. pumilus resulted in 53.7 and 75.3% of turbot mortality rate, respectively. In conclusion, accurate estimation of E. tarda realized the precise feeding of probiotics, which successfully prevent the rapid progression of the edwardsiellosis.  相似文献

1. It had been shown in previous publications that the osmotic pressure of a 1 per cent solution of a protein-acid salt varies in a characteristic way with the hydrogen ion concentration of the solution, the osmotic pressure having a minimum at the isoelectric point, rising steeply with a decrease in pH until a maximum is reached at pH of 3.4 or 3.5 (in the case of gelatin and crystalline egg albumin), this maximum being followed by a steep drop in the osmotic pressure with a further decrease in the pH of the gelatin or albumin solution. In this paper it is shown that (aside from two minor discrepancies) we can calculate this effect of the pH on the osmotic pressure of a protein-acid salt by assuming that the pH effect is due to that unequal distribution of crystalloidal ions (in particular free acid) on both sides of the membrane which Donnan''s theory of membrane equilibrium demands. 2. It had been shown in preceding papers that only the valency but not the nature of the ion (aside from its valency) with which a protein is in combination has any effect upon the osmotic pressure of the solution of the protein; and that the osmotic pressure of a gelatin-acid salt with a monovalent anion (e.g. Cl, NO3, acetate, H2PO4, HC2O4, etc.) is about twice or perhaps a trifle more than twice as high as the osmotic pressure of gelatin sulfate where the anion is bivalent; assuming that the pH and gelatin concentrations of all the solutions are the same. It is shown in this paper that we can calculate with a fair degree of accuracy this valency effect on the assumption that it is due to the influence of the valency of the anion of a gelatin-acid salt on that relative distribution of the free acid on both sides of the membrane which Donnan''s theory of membrane equilibrium demands. 3. The curves of the observed values of the osmotic pressure show two constant minor deviations from the curves of the calculated osmotic pressure. One of these deviations consists in the fact that the values of the ascending branch of the calculated curves are lower than the corresponding values in the curves for the observed osmotic pressure, and the other deviation consists in the fact that the drop in the curves of calculated values occurs at a lower pH than the drop in the curves of the observed values.  相似文献

The lattice-model of Changeux, Thiery, Tung & Kittel (1966) was extended in order to examine the co-operative response of chemically excitable membrane and the exact mathematical correspondence to the Ising model was shown. In this model, two conformational states S and R with different affinities for the ligand are assumed to be accessible to each protomer, which is interacting with the nearest-neighbor protomers. The model is applicable to any kind of symmetrically interacting system consisting of oligomers and lattices and is an extension of previously proposed models of allosteric protein. It includes the model of Monod, Wyman, & Changeux (1965) and that of Koshland, Némethy & Filmer (1966) as the extreme cases of the oligomer. By assuming that a state-transition from S to R in a protomer is accompanied by a unit increase in conductance, the characteristics of dose-response curves of chemically excitable membrane are examined. The Hill's coefficient nH of dose-response curve, the measure of the co-operativity, is shown to be proportional to the square of the mean fluctuation of the state function, the fraction of protomers in R state.  相似文献

Carbon fiber microelectrodes and carbon fiber composite minielectrodes (CFM/CFCM) have been generally used for measurements of nitric oxide (NO) concentration in chemical and biological systems. The response time of a CFM/CFCM is usually from milliseconds to seconds depending on the electrode size, the thickness of coating layers on the electrode, and NO diffusion coefficients of the coating layers. As a result, the time course of recoded current changes (It curves) by the CFM/CFCM may be different from the actual time course of NO concentration changes (ct curves) if the half-life of NO decay is close to or shorter than the response time of the electrode used. This adds complexity to the process for determining rate constants of NO decay kinetics from the recorded current curves (It curves). By computer simulations based on a mathematical model, an approximation method was developed for determining rate constants of NO decay from the recorded current curves. This method was first tested and valuated using a commercial CFCM in several simple reaction systems with known rate constants. The response time of the CFCM was measured as 4.7 ± 0.7 s (n = 5). The determined rate constants of NO volatilization and NO autoxidation in our measurement system at 37 °C are (1.9 ± 0.1) × 10?3 s?1 (n = 4) and (2.0 ± 0.3) × 103 M?1 s?1 (n = 7), which are close to the reported rate constants. The method was then applied to determine the rate of NO decay in blood samples from control and smoking exposed mice. It was observed that the NO decay rate in the smoking group is >20% higher than that in control group, and the increased NO decay rate in the smoking group was reversed by 10 μM diphenyleneiodonium chloride (DPI), an inhibitor of flavin enzymes such as leukocyte NADPH oxidase.  相似文献

The main objetive of this work was to evaluate and model the biofilm growth of the Saccharomyces cerevisiae (beticus ssp.) yeast during the biological aging of some types of wines. Thus, we have study how the biofilm growth, the glycerine is consumed and the acetaldehyde is produced, and how this phenomena are affected by the media ethanol concentration (0–17%, v/v), under experimental conditions similar to the industrial ones. In consequence, the growth of the S. cerevisiae (beticus ssp.) biofilm on the surface of the liquid was studied and kinetically modelled. Growth curves were fitted by using general kinetic models that include biomass and substrate inhibition factors. The alcohol content of the medium for the fastest growth rate of biofilm was found to be 4.3%, v/v. The proposed kinetic models for biomass growth, glycerine consumption and acetaldehyde formation fit well with the experimental data.The growth kinetics of S. cerevisiae beticus ssp. in biofilm phase presents a typical discontinuous microbial growth profile (with lag, exponential and stationary phases). The glycerine consumption is directly related to the substrate concentrations (ethanol and glycerine). Finally, the rate of acetaldehyde formation suggests a model associated with the rate of microbial growth, which is modified by a substrate-dependent factor. The suggested model can be used for optimization and control processes of biological aging of wines.  相似文献

Concentration-time courses measured by dynamic contrast-enhanced (DCE) imaging can be described by a convolution of the arterial input with an impulse response function, QT(t), characterizing tissue microcirculation. Data analysis is based on two different approaches: computation of QT(t) by algebraic deconvolution (AD) and subsequent evaluation according to the indicator dilution theory (IDT) or parameterization of QT(t) by analytical expressions derived by compartmental modeling. Pitfalls of both strategies will be addressed in this study.Tissue data acquired by DCE-CT in patients with head-and-neck cancer and simulated by a reference model (MMID4) were analyzed by a two-compartment model (TCM), a permeability-limited two-compartment model (PL-TCM) and AD. Additionally, MMID4 was used to compute the ‘true’ response function that corresponds to the simulated tumor data.TCM and AD yielded accurate fits, whereas PL-TCM performed worse. Nevertheless, the corresponding response functions diverge markedly. The response curves obtained by TCM decrease exponentially in the early perfusion phase and overestimate the tissue perfusion, QT(0). AD also resulted in response curves starting with a negative slope and not – as the ‘true’ response function in accordance with the IDT – with a horizontal plateau. They are thus not valid responses in the sense of the IDT that can be used unconditionally for parameter estimation.Response functions differing considerably in shape can result in virtually identical tissue curves. This non-uniqueness makes a strong argument not to use algebraic but rather analytical deconvolution to reduce the class of solutions to representatives that are in accordance with a-priori knowledge. To avoid misinterpretations and systematic errors, users must be aware of the pitfalls inherent to the different concepts.  相似文献

Photosynthetic response of Microcystis aeruginosa PCC7806 to different concentrations of phosphorus supply was studied so as to elucidate if the declining process of Microcystis bloom under freshwater ecosystem is related to soluble reactive phosphorus (SRP) decrease in water volume. Growth rate of M. aeruginosa PCC7806 was significantly reduced under P-deficient conditions, and its photosynthetic activity in terms of rETRmax (maximum electron transport rate) decreased significantly after 48 h growth, while it kept elevating and reached to a relative stable value when supplied with rich phosphorus of 0.6 mg/L. With the increasing actinic irradiance along the rapid light curves of M. aeruginosa PCC7806 cultured under low-phosphorus level, qP (photochemical quenching) and rETR (relative electron transport rate) decreased greatly, and the increase in qN (non-photochemical quenching) and ΦPS (actual photochemical efficiency of PSII) was obviously inhibited. The affinity of M. aeruginosa PCC7806 to inorganic carbon was reduced evidently in 0.02 mg/L P compared with in 0.6 mg/L P. When P was reduced from 0.6 to 0.02 mg/L, the decreasing rate of rETRmax (77%) was significantly greater than that of photosynthetic carbon assimilation (22%), which indicated that down-regulation of CO2 affinity caused by P-deficiency was, but not the only reason that resulted in the decline of photosynthetic efficiency. Instantaneous low-temperature significantly limited rETRmax under rich-P condition but had no effect on it when P was insufficient, and 1% ethanol could enhance rETRmax at low-P level but did not influence it at rich-P level. These two results proved that the decrease in thylakoid membrane fluidity caused by P-deficiency was another important reason that results in the decline of photosynthetic efficiency of M. aruginosa PCC7806.  相似文献
