Insect-vectored plant viruses can induce changes in plant phenotypes,thus influencing plant-vector interactions in a way that may promote their dispersal according to their mode of transmission (i.e.,circulative vs.noncirculative).This indirect vector manipulation requires host-virus-vector coevolution and would thus be effective solely in very specific plant-virus-vector species associations.Some studies suggest this manipulation may depend on multiple factors relative to various intrinsic characteristics of vectors such as transmission efficiency.In anintegrative study,we tested the effects of infection of the Brassicaceae Camelina sativa with the noncirculative Cauliflower mosaic virus (CaMV)or the circulative Turnip yellows virus (TuYV)on the host-plant colonization of two aphid species differing in their virus transmission efficiency:the polyphagous Myzus persicae,efficient vector of both viruses,and the Brassicaceae specialist Brevicoryne brassicae,poor vector of TuYV and efficient vector of CaMV.Results confirmed the important role of virus mode of transmission as plant-mediated effects of CaMV on the two aphid species induced negative alterations of feeding behavior (i.e.,decreased phloem sap ingestion)and performance that were both conducive for virus fitness by promoting dispersion after a rapid acquisition.In addition,virus transmission efficiency may also play a role in vector manipulation by viruses as only the responses of the efficient vector to plant-mediated effects of TuYV,that is,enhanced feeding behavior and performances,were favorable to their acquisition and further dispersal.Altogether,this work demonstrated that vector transmission efficiency also has to be considered when studying the mechanisms underlying vector manipulation by viruses.Our results also re- inforce the idea that vector manipulation requires coevolution between plant,virus and vector.  相似文献

In recent years,various serious diseases caused by Zika virus (ZIKV) have made it impossible to be ignored.Confirmed existence of ZIKV in semen and sexually transmission of ZIKV suggested that it can break the blood–testis barrier (BTB),or Sertoli cell barrier (SCB).However,little is known about the underlying mechanism.In this study,interaction between actin,an important component of the SCB,and ZIKV envelope (E) protein domain Ⅲ (EDⅢ) was inferred from coimmunoprecipitation (Co-IP) liquid chromatography–tandem mass spectrometry (LC–MS/MS) analysis.Confocal microscopy confirmed the role of actin filaments (F-actin) in ZIKV infection,during which part of the stress fibers,the bundles that constituted by paralleled actin filaments,were disrupted and presented in the cell periphery.Colocalization of E and reorganized actin filaments in the cell periphery of transfected Sertoli cells suggests a participation of ZIKV E protein in ZIKV-induced F-actin rearrangement.Perturbation of F-actin by cytochalasin D (CytoD) or Jasplakinolide (Jas)enhanced the infection of ZIKV.More importantly,the transepithelial electrical resistance (TEER) of an in vitro mouse SCB (mSCB) model declined with the progression of ZIKV infection or overexpression of E protein.Co-IP and confocal microscopy analyses revealed that the interaction between F-actin and tight junction protein ZO-1 was reduced after ZIKV infection or E protein overexpression,highlighting the role of E protein in ZIKV-induced disruption of the BTB.We conclude that the interaction between ZIKV E and F-actin leads to the reorganization of F-actin network,thereby compromising BTB integrity.  相似文献

Vanilloid receptors 1 (VRs1) expressed in a subpopulation of sensory neurons and responsible for processing of chemical and thermal noxious stimuli were also shown to be expressed in several cerebral structures and to be involved in the regulation of glutamatergic synaptic transmission. In this study, we started to investigate the possibility that VRs1 are also involved in the regulation of GABA-ergic synaptic transmission. For this purpose, the effect of a VR1 agonist, capsaicin, on spontaneous GABA-ergic inhibitory postsynaptic currents (IPSCs) was studied in hippocampal cell cultures using a patch-clamp technique. It was found that capsaicin (10 μM) decreased both the frequency and amplitude of spontaneous IPSCs. This finding suggests the involvement of VRs1 in the regulation of neuronal firing in some GABA-ergic interneurons and in the modulation of the efficacy of GABA-ergic synaptic transmission. However, considering the direction of the effect (a decrease in the IPSC frequency) and lack of its desensitization, the involvement of other receptor(s) also cannot currently be ruled out. A mathematical model for the dynamical transmission of polio is considered,with the aim of investigating the impact of environment contamination.The model captures two infection pathways through both direct human-to-human transmission and indirect human-to-environment-to-human transmission by incorporating the environment as a transition and/or reservoir of viruses.We derive the basic reproduction number R0^env We show that the disease free equilibrium is globally asymptotically stable(GAS)if R0^env<1,while if R0^env>1,there exists a unique endemic equilibrium which is locally asymptotically stable(LAS).Similar results hold for environmental contamination free sub-model(without the incorporation of the indirect transmission).At the endemic level,we show that the number of infected individuals for the model with the environ mental-related contagion is greater than the corresponding number for the environmental contamination free sub-model.In conjunction with the inequality R0相似文献

Noxious stimuli cause pain by activating cutaneous nociceptors.The Aδ-and C-fibers of dorsal root ganglion(DRG) neurons convey the nociceptive signals to the laminae Ⅰ—Ⅱ of spinal cord.In the dorsal horn of spinal cord,the excitatory afferent synaptic transmission is regulated by the inhibitory neurotransmitter γ-aminobutyric acid and modulators such as opioid peptides released from the spinal interneurons,and by serotonin,norepinepherine and dopamine from the descending inhibitory system.In contrast to the accumulated evidence for these central inhibitors and their neural circuits in the dorsal spinal cord,the knowledge about the endogenous suppressive mechanisms in nociceptive DRG neurons remains very limited.In this review,we summarize our recent findings of the presynaptic suppressive mechanisms in nociceptive neurons,the BNP/NPR-A/PKG/BK_(Ca) channel pathway,the FSTL1/α1Na~+-K~+ ATPase pathway and the activin C/ERK pathway.These endogenous suppressive systems in the mechanoheat nociceptors may also contribute differentially to the mechanisms of nerve injury-induced neuropathic pain or inflammation-induced pain.  相似文献

Human immunodeficiency virus (HIV) is the infectious agent causing acquired immu-nodeficiency syndrome (AIDS),a deadliest scourge of human society. Hepatitis C virus (HCV) is a major causative agent of chronic liver disease and infects an estimated 170 million people worldwide,resulting in a serious public health burden. Due to shared routes of transmission,co-infection with HIV and HCV has become common among individuals who had high risks of blood exposures. Among hemophiliacs the co-infection rate accounts for 85%; while among injection drug users (IDU) the rate can be as high as 90%. HIV can accelerate the progression of HCV-related liver disease,particularly when immunodeficiency has developed. Although the effect of HCV on HIV infection is controversial,most studies showed an increase in mortality due to liver disease. HCV may act as a direct cofactor to fasten the progression of AIDS and decrease the tolerance of highly active antiretroviral therapy (HARRT). Conversely,HAART-related hepatotoxicity may enhance the progression of liver fibrosis. Due to above complications,co-infection with HCV and HIV-1 has imposed a critical challenge in the management of these patients. In this review,we focus on the epidemiology and transmission of HIV and HCV,the impact of the two viruses on each other,and their treatment.  相似文献

In animal cells, Golgi apparatus is located near the microtubule organizing center (MTOC) and its position is determined partly by 58K protein. By sodium dodecyl-sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and immuno-blotting methods, a 58K-like protein has been found in pollen grains and pollen tubes of Lilium davidii. Its molecular weight is very similar to that of the 58K protein of animal cells. By immunofluorescence labeling, under a confocal laser scanning microscope (CLSM), the animal 58K antibody revealed a punctate staining in pollen grains and pollen tubes, which is consistent with the distribution of Golgi apparatus in plant cells. In addition, immuno-gold labeling and transmission electron microscopy showed that the 58K-like protein bound mainly to the membrane of vesicles-like structure near Golgi apparatus. This is the first demonstration of the 58K-like protein in plant cells.  相似文献

Duck circovirus (DuCV),a potential immunosuppressive virus,was investigated in Southern China from March 2006 to December 2009 by using a polymerase chain reaction (PCR) based method.In this study,a total of 138 sick or dead duck samples from 18 different farms were examined with an average DuCV infection rate of ~35%.It was found that ducks between the ages of 40~60 days were more susceptible to DuCV.There was no evidence showing that the DuCV virus was capable of vertical transmission.Farms with positive ...  相似文献

Prior to 2003, there were only about 150 HIV/AIDS patients were treated with HAART in some clinical trials and about 100 HIV/AIDS patients were treated by private sources.Results of those treatments are the scientific basis for development of the therapeutic strategies in China. In March of 2003, the Chinese government initiated China CARES program. In November of 2003, the Chinese Ministry of Health announced a national policy of free ARV treatment to all HIV Chinese citizens who were in poverty and required ARV therapy. There are total of 19,456 HIV/AIDS patients received free ARV drugs to date in 159 regions and 441 towns.Current challenges are how to follow-up and evaluate those patients in the clinical settings. The longer the therapy is postponed, the more side effects and the higher probability of drug resistance are going to occur. The skin is the largest organ of the body and is a potential route of exposure to sunscreens and cosmetics containing nanoparticles;however,the permeability of the skin to these nanoparticles is currently unknown.In this paper,we studied the transdermal delivery capacity through mouse skin of water-soluble CdSeS quantum dots(QDs) and the deposition of these QDs in the body.QD solution was coated onto the dorsal hairless skin of male ICR mice.Fluorescence microscopy and transmission electron microscopy(TEM) were used to observe the distribution of QDs in the skin and organs,and inductively coupled plasma-mass spectrometry(ICP-MS) was used to measure the 111Cd content to indicate the concentration of QDs in plasma and organs.Experimental results indicate that QDs can penetrate into the dermal layer and are limited to the uppermost stratum corneum layers and the hair follicles.Through blood circulation,QDs deposit mostly in liver and kidney and are difficult to clear.111Cd concentration was greater than 14 ng g-1 in kidney after 120 h after 0.32 nmol QDs was applied to a mouse.These results suggest that QDs have in vivo transdermal delivery capacity through mouse skin and are harmful to the liver and kidney.  相似文献

In this study, the effects of acute SO_2 derivatives and chronic lead exposure together on sodium cur-rents (INa) were investigated in acutely isolated rat hippocampal neurons by using the whole-cell patch clamp techniques. We found that chronic lead exposure hardly reduced the amplitudes of INa. In the normal condition, sodium current started to appear at around ?70 mV, and reached the peak current at around ?40 mV. After chronic lead exposure, the data changed to ?70 and ?30 mV. After adding SO2 derivatives, the data changed to ?80 and ?40 mV, respectively. SO_2 derivatives caused a significant in-crease of INa in hippocampal chronic-lead exposed neurons. Chronic lead exposure induced a right shift of the activation curve and a left shift of the inactivation curve of sodium channels. SO_2 derivatives caused negative shifts of the activation and inactivation curves of INa in hippocampal chronic-lead ex-posed neurons. Lead exposure put off the time reaching the peak of INa activation. SO_2 derivatives in-creased the time constants of inactivation after lead exposure. The interaction of lead and SO_2 deriva-tives with voltage-dependent sodium channels may lead to changes in electrical activity and contribute to worsening the neurotoxicological damage.  相似文献

Prevention of mother-to-child transmission (PMTCT) of HIV with highly active antiretroviral therapy (HARRT) allows the HIV~+ pregnant mothers to have vaginal delivery and breastfeed.Here we investigated the maternal plasma immunoglobulin,cytokine secretion and the outcome of the exposed infants among the HIV~+ HAART treated pregnan women in Nigeria.In this study,different plasma immunoglobulins and cytokines were measured in the HIV~+ HAART treated pregnant mothers.Pooled culture supernatants of B and T lymphocytes showed lower levels of IFN-γ,IL-10 and IL-4.There were lower IFN-γ and IL-10 secretions at 1st trimester;however,IL-10 continued to be lower throughout 2nd and 3rd trimesters.TNF-α secretion significantly decreased as pregnancy progressed to term.There were high plasma IgG and low IgM in the HIV~+ HAART treated pregnant women.Plasma IgG was high during 1st and 3rd trimesters.After one year of follow up,all the exposed children were seronegative for HIV-1 and HIV-2.Vaginal delivery and breastfeeding among HIV~+ HAART treated mothers have shown to be safe.The use of HAART by the infected mothers and the use of septrin and niverapin by the exposed infants prevented mother to-child transmission of HIV.  相似文献

A set of trisomics of Chinese cabbage was used for determining the n-I-1 gamete transmission rate and locating the gene controlling 2n gamete formation on the corresponding chromosome. The results showed that the transmission rates of extra chromosomes in different trisomics varied from 0% to 15.38% by male gametes and from 0% to 17.39% by female gametes. Of the nine F2 populations derived from the hybridizations between each trisomic and Bp058 (2n gamete material), only Tri- 4xBp058 showed that the segregation ratio of plants without 2n gamete formation to plants with 2n gamete formation was 10.38:1, which fitted the expected segregation ratio of the trisomics (AAa) based on the 7.37% of n+l gamete transmission through female and 5.88% through male. In other populations the segregation ratios varied from 2.48:1 to 3.72:1, which fitted the expected 3:1 segregation ratio of the bisomics (Aa). These results suggested that the gene controlling 2n gamete formation in Chinese cabbage Bp058 was located on chromosome 4. Further trisomic analysis based on the chromosome segregation and the incomplete stochastic chromatid segregation indicated that the gene locus was tightly linked to the centromere.  相似文献

Swarming behavior facilitates pair formation,and therefore mating,in many eusocial termites.However,the physiological adjustments and morphological transformations of the flight muscles involved in flying and flightless insect forms are still unclear.Here,we found that the dispersal flight of the eusocial termite Reticulitermes chinensis Snyder led to a gradual decrease in adenosine triphosphate supply from oxidative phospho?rylation,as well as a reduction in the activities of critical mitochondrial respiratory enzymes from preflight to dealation.Correspondingly,using three-dimensional reconstruction and transmission electron microscopy(TEM),the flight muscles were found to be gradually deteriorated during this process.In particular,two tergo-pleural muscles(IItpm5 and IIItpm5)necessary to adjust the rotation of wings for wing shedding behavior were present only in flying alates.These findings suggest that flight muscle systems vary in function and morphology to facilitate the swarming flight procedure,which sheds light on the important role of swarming in successful extension and fecundity of eusocial termites.  相似文献

In this work,we have introduced an eco-epidemiological model of an infected predator prey system.Incorporation of prey refuge gives that a fraction of the infected prey is available to the predator for consumption.Moreover,to make the model more realistic to the environment,we have introduced strong Allee effect in the susceptible population.Boundedness and positivity of the solution have been established.Local stability conditions of the equilibrium points have been found with the help of Routh-Hurwitz criterion and it has been observed that if a prey population is infected with a lethal disease,then both the prey(susceptible and infected)and predator cannot survive simultaneously in the system for any parametric values.The disease transmission rate and the attack rate on the susceptible have an important role to control the system dynamics.For different values of these two key parameters,we have got only healthy or disease-free or predation-free or a fluctuating disease-free or even a fluctuating predator-free system with some certain parametric conditions.  相似文献

In this paper,we study a deterministic model with non-autonomous system for mixed cultivars to assess the effect of cultivar susceptibility and seasonal variation on banana Xanthomonas wilt(BXW)disease dynamics.A special case of two cultivars classified as highly susceptible for inflorescence infection(ABB)and less susceptible(AAA)cultivar is considered.The basic reproduction number corresponding to the non-autonomous sys­tem is derived and numerically computed to determine disease dynamics.Results showed that the disease dies out whenever the periodic basic reproduction number is less than unity and a periodic solution is obtained when it is greater than one.Results further showed that for both cultivars,the basic reproduction number increases with increas­ing values of the transmission rates and declines exponentially with increasing values of roguing rates.The critical roguing rate of ABB-genome cultivar was higher than that of AAA-genome cultivars.The peaks in disease prevalence indicate the importance of effective implementation of controls during the rainy season.We conclude that highly susceptible cultivars play an important role in the spread of BXW and control measures should be effectively implemented during the rainy season if BXW is to be eradicated.  相似文献
