The segment polarity gene engrailed is involved in the determination of segment posterior identity in Drosophila. engrailed has been largely used for comparative developmental studies due to its evolutionary conservation from nematodes to humans. By in situ hybridization of an engrailed cDNA probe from Drosophila to polytene chromosomes of fourth instar larvae of Rhynchosciara americana we have shown that engrailed-like sequences must be localized in band 6 of chromosome A in this species. The pattern of engrailed protein expression during R. americana embryo development is diffuse at first evolving into a nuclear striped pattern after quite a length of time. In addition, our results suggest a possible developmentally regulated molecular modification of engrailed protein in R. americana embryos.  相似文献

Taxonomy of the genus Cristulariella is revised, retaining Cristulariella (Crist.) depraedans as the type. Two new species, Crist. cercidiphylli and Crist. corni, are additionally described under the genus. The new anamorphic genus Hinomyces is erected to accommodate Botrytis (Cristulariella) moricola and Cristulariella pruni. A new genus and species, Nervostroma depraedans, is erected in the Sclerotiniaceae to accommodate the teleomorph of Crist. depraedans, with an additional species, Nervostroma cercidiphylli.  相似文献

A number of species of the genus Erythrina are rich in secondary metabolites, particularly phenolics and alkaloids that exhibit interesting anti-inflammatory, anti-plasmodial, bactericidal, curariform and fungicidal activities. Unfortunately, the isolation of these compounds through the extraction of organs and seeds of whole plants is becoming more difficult since the natural growing areas of many of the species, and particularly of Erythrina americana, are being urbanised. Plant tissue culture not only constitutes a viable method through which to preserve the species, but may also represent a constant and stable source of target alkaloids. Currently, however, in vitro systems, and especially cell suspension cultures, only accumulate low levels of the desired products. A number of strategies for increasing production have been proposed, the most successful of which involves elicitation of suspension cells with plant growth regulators. In this context, excellent results have been reported following elicitation of cell cultures derived from different species and genera with jasmonic acid and methyl jasmonate. The present paper provides a brief review of the novel approaches to the use of Erythrina alkaloids that have recently been described, and of the advances that have been made in the formation of tissue cultures of E. americana and their subsequent elicitation in attempts to augment alkaloid accumulation.  相似文献

European pine martens (Martes martes)were once distributed across much ofwestern Europe. A combination of factors, suchas persecution, trapping, and habitat loss haveled to sharp declines in the species' numbersand range and, as such, local populations havebecome more vulnerable to extinction. Toevaluate the influence of these factors on boththe level of genetic variation and populationstructure, we genotyped pine martens fromacross much of their current distribution. Continental M. martes populations werefound to have a higher level of geneticstructure and lower genetic variation thantheir North American sibling species, M.americana, sampled throughout Canada. Thedifferences among mainland populations of thesespecies may lie in greater levels of habitatfragmentation and persecution experienced byEuropean martens, though it is difficult toexclude more ancient processes such as theinfluence of glaciations. Among islandpopulations of the two species, the Scottishpopulation revealed a similar level ofstructure and variation to the M. a.atrata population of Newfoundland, howeverIreland was more differentiated with lessgenetic variation. Our work usingmicrosatellites also extends previous mtDNAevidence for the presence of M. americanahaplotypes in England, raising the possibilityof hybridization with M. martes. Thesefindings may influence current discussions onthe status of English martens and theappropriateness of proposed re-introductions byrevealing that some indigenous martens persistin England, despite the presence of somepotential hybrids in the region.  相似文献

Polytretophora macrospora is introduced based on specimens from two species ofPandanus in Seychelles. The new species is compared with currently accepted species. A key and a comparative synopsis toPolytetophora species are provided. Numerous specimens ofP. calcarata, collected on members of the Pandanaceae from Australia, Fiji, Hong Kong, Malaysia, New Caledonia, Palau, Philippines, Seychelles and Solomon Islands are also reported, along withP. dendroidea onPandanus sp. from Malaysia.  相似文献

As part of a study carried out for detecting Arcobacter spp. in shellfish, three mussel isolates that were Gram-negative slightly curved rods, non-spore forming, showed a new 16S rDNA-RFLP pattern with a specific identification method for the species of this genus. Sequences of the 16S rRNA gene and those of the housekeeping genes rpoB, gyrB and hsp60 provided evidence that these mussel strains belonged to an unknown genetic lineage within the genus Arcobacter. The similarity between the 16S rRNA gene sequence of the representative strain (F79-6T) and type strains of the other Arcobacter species ranged between 94.1% with A. halophilus and 99.1% with the recently proposed species A. defluvii (CECT 7697T). DDH results between strain F79-6T and the type strain of the latter species were below 70% (53 ± 3.0%). Phenotypic characteristics together with MALDITOF mass spectra differentiated the new mussel strains from all other Arcobacter species. All the results indicate that these strains represent a new species, for which the name Arcobacter ellisii sp. nov. with the type strain F79-6T (=CECT 7837T = LMG 26155T) is proposed.  相似文献

Four novel yeast species are described, two from decaying mushrooms, viz. Candida cretensis and Candida vadensis, and two from rotten wood, viz. Blastobotrys robertii and Candida scorzettiae. Accession numbers for the CBS and ARS Culture Collections, and GenBank accession numbers for the D1/D2 domains of the large subunit of ribosomal DNA are: B. robertii CBS 10106T, NRRL Y-27775, DQ839395; C. cretensis CBS 9453T, NRRL Y-27777, AY4998861 and DQ839393; C. scorzettiae CBS 10107T, NRRL Y-27665, DQ839394; C. vadensis CBS 9454T, NRRL Y-27778, AY498863 and DQ839396. The GenBank accession number for the ITS region of C. cretensis is AY498862 and that for C. vadensis is AY498864. C. cretensis was the only species of the four that displayed fermentative activity. All four type strains grew on n-hexadecane. C. scorzettiae is the only one of the new species that assimilates some phenolic compounds, viz. 3-hydroxy derivatives of benzoic, phenylacetic and cinnamic acids, but not the corresponding 4-hydroxy acids. In this study two actinomycete strains were isolated in Cape Town (South Africa), one from a compost heap (strain 202GMOT) and the other from within the fynbos-rich area surrounded by the horseracing track at Kenilworth Racecourse (strain C2). Based on 16S rRNA gene sequence BLAST analysis, the strains were identified as members of the genus Nocardia. Phylogenetic analysis showed that the strains clustered together and are most closely related to Nocardia flavorosea NRRL B-16176T, Nocardia testacea JCM 12235T, Nocardia sienata IFM 10088T and Nocardia carnea DSM 43397T. This association was also supported by gyrB based phylogenetic analysis. The results of DNA–DNA hybridization and physiological tests allowed genotypic and phenotypic differentiation of both strains 202GMOT and C2 from related species. However, their high DNA relatedness showed that they belong to the same species. Strain 202GMOT was selected as the type strain to represent this novel species, for which the name Nocardia rhamnosiphila is proposed (=DSM 45147T = NRRL B-24637T).  相似文献

Summary In the American cockroach, Periplaneta americana, and the Australian field cricket, Teleogryllus commodus, the two nerves supplying the bases of the cerci are joined by a branch that crosses behind the last abdominal ganglion. This commissural ring nerve is restricted to females, and it contains many axons filled with granular and agranular vesicles. The axons stem from somata located within the ganglion. There are one (Periplaneta) or two (Teleogryllus) groups of median neurons with bilaterally symmetrical bifurcations, and a group of postero-ventral neurons on each side. In T. commodus, these neurons are distinct from others associated with the cerci. In the two species, the ring nerve neurons contribute to a neuropile near the root of each cereal nerve. The bifurcating median neurons arborize on both sides before entering the ring nerve, while the postero-ventral ones branch more extensively ipsilateral to their somata. The possibilities are discussed that the bifurcating neurons may be homologous to dorsal unpaired median neurons, and that the ring nerve may be a neurohemal area.  相似文献

A locally severe crown disease of exotic plantation Eucalyptus trees has been recorded periodically in New Zealand since 1986. Symptoms include leaf spots, petiole infection and twig and small branch lesions. Outbreaks of disease are episodic and individual trees may show marked variation in crown symptoms ranging from unaffected to total defoliation. Two previously unknown species of Phytophthora are associated with the disease. These are described and formally designated here as P. captiosa, from Eucalyptus botryoides and E. saligna; and P. fallax, from E. delegatensis, E. fastigata, E. nitens and E. regnans. Both P. captiosa and P. fallax have non-papillate, non-caducous sporangia and both are self-fertile. Phylogenetic analysis on the basis of ITS rDNA sequence data indicates they are closely related to each other but evolutionarily distant from the majority of described Phytophthora taxa. They share a common ancestor with another assemblage of Phytophthora lineages that includes P. insolita, P. macrochlamydospora and P. richardiae. Sporulation of P. captiosa and P. fallax has not been observed in the field. The mode of infection and spread of these non-caducous Phytophthora species in the eucalypt tree canopy remains unknown. This issue, and the possible geographic origins of these two Phytophthora species are discussed.  相似文献

Three new species of Candida and a new species of Trigonopsis are described based on their recognition from phylogenetic analysis of gene sequences from large subunit ribosomal RNA, ITS1/ITS2 rRNA, mitochondrial small subunit rRNA and cytochrome oxidase II. Candida infanticola sp. nov. (type strain NRRL Y-17858, CBS 7922) was isolated from the ear of an infant in Germany and is closely related to Candida sorbophila. Candida polysorbophila sp. nov. (type strain NRRL Y-27161, CBS 7317) is a member of the Zygoascus clade and was isolated in South Africa as a contaminant from an emulsion of white oil and polysorbate. Candida transvaalensis sp. nov. (type strain NRRL Y-27140, CBS 6663) was obtained from forest litter, the Transvaal, South Africa, and forms an isolated clade with Candida santjacobensis. Trigonopsis californica sp. nov. (type strain NRRL Y-27307, CBS 10351) represents a contaminant from wine in California, and forms a well-supported clade with Trigonopsis cantarellii, Trigonopsis variabilis and Trigonopsis vinaria.  相似文献

Summary Induction of flowering by photoperiod was studied in the parental, F1, F2, and reciprocal backcross generations of crosses between three photoperiod-responsive Aeschynomene americana L. lines. Generation means appeared additive. Analysis with Mather and Jinks' scaling tests showed little or no epistasis and indicated that an additive-dominance model was adequate. Partitioning components of variation revealed that nearly all variation was additive genetic with dominance and environmental variation negligible. An additive genetic model with two loci, each with two alleles and all alleles having equal net effect, was tested using Power's partitioning method. Results demonstrated that the model fit the data and that there is a major additive genetic system controlling flowering in these crosses, with minor genetic and environmental influences present. Additional tests of native North American Cirsium species, Saussurea americana, and modern safflower cultivars (Carthamus tinctorius) were requested by regulators and specific interest groups during the risk assessment of foreign isolates of Puccinia jaceae var. solstitialis for biological control of yellow starthistle (YST, Centaurea solstitialis) in the United States. These tests supplement an earlier, extensive host range determination that established P. jaceae from YST as generally host specific and potentially useful for biological control. The additional research was in response to potential hazards identified in an earlier study, changes in safflower cultivars, and concern that P. jaceae might cause a safflower seedling disease similar to hypocotyl infections from infestation by Puccinia carthami teliospores. S. americana, a close relative of yellow starthistle, had not been tested previously. All tests were conducted in a containment greenhouse. Foliage of 19 Cirsium species, 11 safflower cultivars, and S. americana was inoculated with urediniospores and subjected to a 16-h dew period at 18–20 °C. Neither the Cirsium species nor S. americana became infected after foliar inoculations. Compared to foliar infections by P. carthami from safflower in California, only minor infections developed from inoculations with P. jaceae. These were similar to infections observed in earlier studies, and it was not possible to maintain P. jaceae under optimal greenhouse conditions on safflower foliage. Quantitative teliospore inoculations with P. jaceae did not cause infection on safflower hypocotyls, even though large cankers occurred on plants inoculated with P. carthami teliospores. Clear microscopic evidence of infection also was observed in hypocotyls inoculated with P. carthami. These data suggest that native (including rare, threatened, or endangered) Cirsium spp., modern safflower cultivars, and S. americana are not likely to be adversely affected by the use of P. jaceae for biological control of YST. Results from these studies substantiate previous findings and were incorporated in a proposal for permission to use P. jaceae for YST control in California.  相似文献

Summary The secretion present at the lumen of the salivary glands of spinning larvae ofRhynchosciara americana was studied cytochemically and with microspectrophotometry and fluorescence and quantitative polarization microscopy. It was found that structural proteins, including glycoproteins and lipoproteins, occur in this secretion. Findings involving spectral absorption profiles after xylidine ponceau staining, patterns of birefringence and dispersion of birefringence, and lack of dichroism after xylidine ponceau staining and of blue fluorescence after ANS staining are highly suggestive that the secretion ofR. americana differs from classical silks not only in terms of composition but also of macromolecular array. The silk secretion ofR. americana also appears to differ from that of another sciarid,Bradysia spatitergum. Part of the glycoproteins present at the glandular lumen is assumed to be extruded from cells of the posterior zone of the glands, whereas other glycoproteins (or their glycidic radicals) are probably removed from fat body cells via cells of the anterior zone of the glands. The salivary secretion of the spinning larvae ofR. americana contains calcium and is devoid of acid glycosaminoglycans.  相似文献

Morphological and molecular phylogenetic analyses were conducted on 12 strains ofFusarium, deposited in MAFF asF. subglutinans (≡F. moniliforme var.subglutinans≡F. sacchari var.subglutinans) orFusarium sp. because they formed aerial conidia in false heads in the dark. These strains were resolved as three distinct species within theGibberella fujikuroi species complex. A new species,F. fractiflexum, and two species new to Japan,F. circinatum andF. concentricum, are described and illustrated and their morphological features are discussed.Fusarium fractiflexum, isolated from diseased yellow leaf spots ofCymbidium spp., is differentiated from other fusaria based on its yellowish colonies and aerial conidia formed in false heads in the dark and in zigzag-like conidial chains under black light. Japanese strains ofF. circinatum also formed elongate, coiled sterile hyphae. Phialidic aerial conidia with a pointed apex and a wedgeshaped base were found inF. concentricum cultured under black light and represent a new diagnostic character of the species, in addition to colonies with alternating concentric rings when cultured on PDA. Based on DNA sequences of the β-tubulin gene and two other loci, strains ofF. fractiflexum were resolved phylogenetically as members of the Asian clade of theG. fujikuroi species complex. In addition, Japanese strains ofF. circinatum andF. concentricum were phylogenetically identical to the ex-type strains.  相似文献

The taxonomic affiliation was determined for four Xenorhabdus strains isolated from four Steinernema hosts from different countries. As compared to the five validly described Xenorhabdus species, i.e., X. nematophila, X. japonica, X. beddingii, X. bovienii and X. poinarii, these isolates represented novel species on the basis of 16S rRNA gene sequences and riboprint patterns, as well as by physiological and metabolic properties. They were named Xenorhabdus budapestensis sp. nov., type strain DSM 16342T, isolated from Steinernema bicornutum; Xenorhabdus ehlersii sp. nov., type strain DSM 16337T, isolated from Steinernema serratum; Xenorhabdus innexi sp. nov., type strain DSM 16336T isolated from Steinernema scapterisci; and Xenorhabdus szentirmaii sp. nov., type strain DSM 16338T, isolated from Steinernema rarum.  相似文献

Eurytemora americana and Acartia tonsa are the two most important copepods from the Bahía Blanca estuary plankton. For this study, 30 females were selected from recently preserved samples. Prosome length (PL), width (PW) and height (PH), urosome length (UL) and width (UW) measured in each female and subsequently the following ratios PL/PW, PL/PH, PW/PH and PL/UL were determined. Individual volumes for E. americana and A. tonsa females were estimated following the morphometric method in order to obtain individual biomass values. Regression tests were conducted for both species between individual volume and prosome length or width as independent variables, fitting data to a power model. Regression lines were compared through covariance analysis and single pair comparisons in order to evaluate the temporal behavior of the relationships. PL/UL was the best ratio for separating one species from the other. Average individual volume and std. error estimated for E. americana and A. tonsa were 0.0749 (±0.0097) mm3 and 0.0399 (±0.0049) mm3, respectively. Strong differences between both copepods morphometric variable and rate values were observed. A size and volume decrease was detected associated with higher temperatures and less phytoplankton available for food. According to R 2 values of regression lines prosome width and prosome length were good volume predictors on different dates for E. americana, however, prosome width was the best predictor on the majority of dates for A. tonsa. Differences in slopes and mean variances found between regression lines demonstrated seasonal variability. General regression equations between volume and PL or PW for each species could be derived.  相似文献

Intercellular signaling molecules of the transforming growth factor- (TGF-) superfamily are required for pattern formation in many multicellular organisms. The decapentaplegic (dpp) gene of Drosophila melanogaster has several developmental roles. To improve our understanding of the evolutionary diversification of this large family we identified dpp in the grasshopper Schistocerca americana. S. americana diverged from D. melanogaster approximately 350 million years ago, utilizes a distinct developmental program, and has a 60-fold-larger genome than D. melanogaster. Our analyses indicate a single dpp locus in D. melanogaster and S. americana, suggesting that dpp copy number does not correlate with increasing genome size. Another TGF- superfamily member, the D. melanogaster gene 60A, is also present in only one copy in each species. Comparison of homologous sequences from D. melanogaster, S. americana, and H. sapiens, representing roughly 900 million years of evolutionary distance, reveals significant constraint on sequence divergence for both dpp and 60A. In the signaling portion of the dpp protein, the amino acid identity between these species exceeds 74%. Our results for the TGF- superfamily are consistent with current hypotheses describing gene duplication and diversification as a frequent response to high levels of selective pressure on individual family members.  相似文献

A new species, Strobilomyces verruculosus, is described and illustrated. It is morphologically distinct from other species of Strobilomyces by having a verruculose pileus with small subpyramidal scales, a long and thick stipe with small warty to appressed scales, subdecurrent tubes, and incompletely reticulate basidiospores. Molecular phylogenetic analyses using the mitochondrial cytochrome oxidase subunit 3 (cox3) gene support that it belongs to the genus Strobilomyces and is highly differentiated from the other members of this genus found in Japan.  相似文献

A new wood-decaying polypore, Antrodiella chinensis, is described and illustrated from China based on morphological characteristics and phylogenetic analysis of rDNA ITS sequences. The new species belongs to the Antrodiella americana complex, and is characterized by an annual habit, resupinate basidiocarps, cream to straw coloured pore surface with larger and regular pores, and oblong-ellipsoid basidiospores. Discriminating characteristics between the new species and the closely related species in the complex are discussed.  相似文献
