摘要:目的 通过分离鉴定鲍曼不动杆菌噬菌体并进行遗传信息分析,为今后噬菌体用于治疗鲍曼不动杆菌引起的感染提供依据。方法 以鲍曼不动杆菌临床分离株为宿主菌,从医院污水中分离鲍曼不动杆菌噬菌体并进行纯化、电镜观察形态特征、提取噬菌体DNA,进行全基因组测序,分析全基因组的结构特征,比较基因组分析其进化关系。结果 分离到鲍曼不动杆菌裂解性噬菌体LZ35,电镜观察显示,该噬菌体属于有尾噬菌体目肌尾病毒科。基因组全长44 885 bp,G+C含量为37.95%,含有83个开放阅读框,其中22个编码序列可预测其功能,61个编码序列为未知基因。噬菌体LZ35的基因组与鲍曼不动杆菌噬菌体IME-AB2和YMC-13-01-C62具有很高的同源性(分别为97%和99%),与鲍曼不动杆菌噬菌体YMC11/12/R1215的进化关系最近。结论 以鲍曼不动杆菌临床分离株为宿主菌,分离到鲍曼不动杆菌裂解性噬菌体LZ35,明确了其形态和基因组特征,为防治噬菌体疗法奠定基础。  相似文献

从污水中分离鉴定鲍曼不动杆菌噬菌体,并对其生物学特性进行分析,为开发针对细菌感染的噬茵体生物制剂提供前期工作.采用双层琼脂法分离可裂解鲍曼不动杆菌的噬菌体,通过负染法电镜观察噬茵体的大小和形态,将噬菌体和宿主菌以不同比例混合,测定噬菌体的最佳感染复数并观察一步生长曲线,提取噬菌体核酸进行酶切电泳分析,通过SDS-PAGE分析噬菌体的结构蛋白和非结构蛋白.成功分离3株可裂解鲍曼不动杆菌的噬菌体(分别命名为ΦAb-1、ΦAb-2和ΦAb-3),电镜显示噬菌体的头部呈二十面体,直径约50nm,有一短尾.噬菌体ΦAb-1的最佳感染复数为10-2,一步生长曲线表明噬菌体在裂解宿主菌时,潜伏期为20 min,爆发期为30 min,裂解量为190 PFU/cell.限制性酶切电泳显示噬菌体ΦAb-1的基因组大小约40 kb左右,为双股环状DNA.SDS-PAGE的噬菌体蛋白电泳包括7种蛋白,分子量在29 ~ 116 ku.噬菌体ΦAb-1、ΦAb-2和ΦAb-3对鲍曼不动杆菌具有很强的裂解毒性.根据其形态、结构和噬菌体分类法,鲍曼不动杆菌噬菌体属于有尾病毒目,足尾病毒科.  相似文献

鲍曼不动杆菌的分离及耐药分析   总被引:1,自引:1,他引:1  
目的:监测近年临床标本鲍曼不动杆菌的分离、分布特点及耐药情况,以有助于临床合理用药。方法:从临床标本中分离出64株鲍曼不动杆菌,采用K—B方法,严格按照美国临床实验室标准化委员会(NCCLS)2001制定的最新规则及标准进行结果判读。结果:在统计年限内,我院共分离出不动杆菌84株,鲍曼不动杆菌64株,占不动杆菌的76.1%,其中痰标本分离出49株,占76.5%;尿标本分离出3株,占4.6%;分泌物标本分离出12株,占18.7%。鲍曼不动杆菌对β-内酰胺类抗生素交叉耐药严重,除亚胺培南外,其耐药率在10.0~84.8%之间。结论:鲍曼不动杆菌在临床的分离率逐年增加,耐药性逐渐增强,应引起重视。  相似文献

【目的】预测并分析82株全基因组测序的鲍曼不动杆菌前噬菌体的携带情况,鉴定前噬菌体编码的抗生素耐药基因和毒力因子。【方法】利用PHASTER (Phage Search Tool Enhanced Release)软件预测鲍曼不动杆菌携带的前噬菌体,采用CARD (The Comprehensive Antibiotic Research Database)和VFDB(VirulenceFactorsDatabase)在线分析软件预测前噬菌体编码的抗生素耐药基因和毒力因子。【结果】预测到472条鲍曼不动杆菌前噬菌体,其中完整型前噬菌体201条,疑似型前噬菌体91条,缺陷型前噬菌体180条。平均每株鲍曼不动杆菌基因组中可携带至少2条完整型前噬菌体。每株鲍曼不动杆菌所携带的全部前噬菌体占其基因组比例约为4%–6%。29条前噬菌体携带77个耐药基因,耐药表型共有14种,分别来自15个不同的家族,涵盖6种抗生素耐药的作用机制。132条前噬菌体编码毒力基因,归类为38种毒力基因和34种毒力因子。不同类型的前噬菌体普遍携带1–2种毒力因子,少数前噬菌体携带3种及以上毒力因子。分析毒力因子可能的宿主...  相似文献

【背景】噬菌体解聚酶是噬菌体在裂解细菌过程中产生的一种抗菌蛋白,关于鲍曼不动杆菌荚膜分型及常见型别噬菌体解聚酶的研究报道较少。【目的】以KL2型鲍曼不动杆菌为研究对象,从噬菌体IME-AB2中克隆解聚酶,在大肠杆菌中进行可溶性表达并研究其体外抗菌活性。【方法】应用二代测序及生物信息学方法鉴定鲍曼不动杆菌荚膜型,分析IME-AB2全基因组。应用分子克隆技术克隆ORF76假定的尾丝蛋白(Putative Tail Fiber)基因,构建重组表达载体pEASY-Blunt-E1-gp76,在大肠杆菌BL21(DE3)中诱导表达,通过Ni-NTA亲和层析纯化解聚酶,研究解聚酶体外抗菌活性。【结果】构建了pEASY-Blunt-E1-gp76解聚酶重组表达质粒,该重组质粒在大肠杆菌中得到可溶性表达;体外活性分析显示,该重组蛋白在体外能够对所有的KL2型鲍曼不动杆菌具有较好的抗菌活性,解聚酶联合人和狗的血清具有很好的杀菌活性。【结论】鉴定解聚酶并提高其抗菌谱具有重要意义,也是噬菌体及解聚酶用于治疗耐药菌研究领域急需解决的重要问题之一。  相似文献

目的:从医院废水中快速分离多株不同的铜绿假单胞杆菌噬菌体,研究其生物学特性,为建立铜绿假单胞杆菌噬菌体库做准备。方法:利用噬菌斑法从未经处理的医院污水中分离和鉴定铜绿假单胞杆菌噬菌体,根据感染谱的不同确定它们为不同的铜绿假单胞杆菌噬菌体;重点研究其中一株宿主谱较广的噬菌体的生物学特性,采用负染法电镜观察噬菌体的形态和大小,提取该噬菌体的基因组并进行酶切电泳分析,测定噬菌体感染复数并观察其一步生长曲线。结果:通过噬菌斑法分离出90株铜绿假单胞杆菌噬菌体。电镜观察显示,噬菌体Pa27P1头部呈立体对称,有一长尾;酶切结果显示,噬菌体Pa27P1的基因组为双链DNA;生长曲线表明噬菌体Pa27P1感染宿主菌的潜伏期为25 min,爆发时间为25 min,裂解量为514。结论:90株铜绿假单胞杆菌噬菌体中有5株具有较广的噬菌谱,其组合能裂解所有18株铜绿假单胞杆菌,为深入研究铜绿假单胞杆菌噬菌体的生物学特性及其功能提供了依据。  相似文献

以上海某些医院临床分离到的多重耐药肺炎克雷伯菌为宿主菌,从不同环境的污水中分离获得1株肺炎克雷伯菌噬菌体KP002。电子显微镜显示其为有尾噬菌体,头部直径约70nm,尾长约80nm,尾宽约20nm。对其生物学特性进行研究,结果显示此株噬菌体在pH 3~9及4~50℃的环境中具有较高活性;6min吸附率达95%以上;潜伏期为10min,爆发期为50min;裂解量为172pfu/cell。结果表明,该噬菌体对pH值和温度适应范围较宽。对其全基因组进行测序分析,结果显示其基因组为环状双链DNA,全长47 173bp,GC含量为48%。本研究筛选获得1株对pH值和温度适应范围较宽的耐药肺炎克雷伯菌烈性噬菌体KP002,为建立耐药肺炎克雷伯菌的噬菌体库以用于治疗临床多重耐药菌感染提供了新的思路。  相似文献

目的 通过对重庆医科大学附属第二医院近4年临床分离的103株鲍曼不动杆菌的耐药性和同源性分析,了解该院鲍曼不动杆菌的耐药性特点及院内感染流行状况.方法 2008年至2011年间该院临床科室分离的103株鲍曼不动杆菌,进行药敏试验并对耐药性分析;提取细菌基因组DNA,以随机扩增DNA多态性(RAPD)方法进行基因分型.结果 药敏试验结果显示分离出的103株菌呈现出高耐药率及多重耐药率的特点,其中对左氧氟沙星和亚胺培南的耐药性最低,分别为69.9%和57.3%.103株多重耐药鲍曼不动杆菌用RAPD分型共分为16种基因型:A~P,其中A型84株,为主要流行型别;B型3株;C型、G型各2株;其余12型各1株.结论 该院鲍曼不动杆菌具有多重耐药及高耐药率,可能存在以A型鲍曼不动杆菌克隆株传播方式的院内流行,临床上应加强对A型鲍曼不动杆菌的监控,采取有效的措施以预防院内感染的爆发流行.  相似文献

目的:利用临床耐药粪肠球菌分离裂解性噬菌体,为应用噬菌体治疗耐药粪肠球菌感染提供基础。方法:利用噬菌斑实验分离噬菌体并观察噬菌斑形态;双层平板培养法测定噬菌体效价、最佳感染复数及一步生长曲线;负染法电镜观察噬菌体形态;蛋白酶K/SDS法提取噬菌体基因组,酶切处理后琼脂糖凝胶电泳分析。结果:分离出一株噬菌体IME-EF1,该噬菌体能裂解多株临床分离的粪肠球菌;电镜观察呈蝌蚪形,最佳感染复数为1;通过绘制一步生长曲线,证明该噬菌体感染后的潜伏期为25 min,爆发期为35 min,裂解量为60 pfu。结论:研究结果表明利用临床分离的耐药粪肠球菌分离裂解性噬菌体是可行的,有望为耐药粪肠球菌的抗生素替代疗法奠定基础。  相似文献

目的 了解医院鲍曼不动杆菌的耐药性变迁和临床分布特点,为合理应用抗菌药物和预防控制医院感染提供依据.方法 回顾性分析2001年1月到2011年12月临床标本中分离的l 613株鲍曼不动杆菌资料;用WHONET 5.4分析鲍曼不动杆菌的分离率和耐药率变迁.结果 鲍曼不动杆菌感染主要发生在ICU和呼吸内科,分别占43.89%和24.49%;痰标本所占比例最高(70.30%).鲍曼不动杆菌、多药耐药鲍曼不动杆菌的分离率及对临床常用的15种抗菌药物耐药率总体呈上升趋势;并且2009年呈飞跃式上升且2010和2011年居高不下,201 1年分离出泛耐药鲍曼不动杆菌,分离率高达57.55%.临床常用药氨苄西林/舒巴坦、头孢哌酮/舒巴坦和米诺环素近三年的耐药率和中介率也明显升高.结论 鲍曼不动杆菌的耐药形势严峻,多药耐药鲍曼不动杆菌和泛耐药鲍曼不动杆菌比例明显增加,重症监护病房和呼吸科是预防控制的重点科室.  相似文献

A lytic bacteriophage isolated from sewage was found to attack strains of Aerobacter aerogenes, Escherichia coli, and Klebsiella pneumoniae, but not members of the genera Salmonella, Proteus, and Serratia. The phage, designated phimp, contained deoxyribonucleic acid with a 50% guanine plus cytosine ratio and a molecular weight of 23.1 x 10(6) daltons. Single-step growth experiments of phimp plated at 37 C on A. aerogenes A2 gave a mean latent period of 20 min, an average burst size of 103 plaque-forming units/infected cell, and an average adsorption rate constant of 3 x 10(-10) ml/min. Electron microscopy of phimp revealed a phage with a flexible tail (165 nm long and 6 nm wide). The phage head had a hexagonal outline (62 nm in diameter).  相似文献

1. An anti-Escherichia coli phage has been isolated and its behavior studied. 2. A plaque counting method for this phage is described, and shown to give a number of plaques which is proportional to the phage concentration. The number of plaques is shown to be independent of agar concentration, temperature of plate incubation, and concentration of the suspension of plating bacteria. 3. The efficiency of plating, i.e. the probability of plaque formation by a phage particle, depends somewhat on the culture of bacteria used for plating, and averages around 0.4. 4. Methods are described to avoid the inactivation of phage by substances in the fresh lysates. 5. The growth of phage can be divided into three periods: adsorption of the phage on the bacterium, growth upon or within the bacterium (latent period), and the release of the phage (burst). 6. The rate of adsorption of phage was found to be proportional to the concentration of phage and to the concentration of bacteria. The rate constant ka is 1.2 x 10–9 cm.8/min. at 15°C. and 1.9 x 10–9 cm.8/min. at 25°. 7. The average latent period varies with the temperature in the same way as the division period of the bacteria. 8. The latent period before a burst of individual infected bacteria varies under constant conditions between a minimal value and about twice this value. 9. The average latent period and the average burst size are neither increased nor decreased by a fourfold infection of the bacteria with phage. 10. The average burst size is independent of the temperature, and is about 60 phage particles per bacterium. 11. The individual bursts vary in size from a few particles to about 200. The same variability is found when the early bursts are measured separately, and when all the bursts are measured at a late time.  相似文献

【目的】自然界中噬菌体种类繁多,其裂菌功能在针对细菌耐药方面具有潜在应用价值。不同噬菌体也呈现出显著的基因多样性及宿主特异性。从上海某猪场仔猪肠内容物样品中分离、纯化大肠杆菌的裂解性噬菌体,分析其生物学特性和病毒学特征,为探索应用噬菌体治疗细菌性感染提供研究材料。【方法】采用双层琼脂平板法分离、纯化噬菌体,观察噬菌斑特征,通过电镜观察噬菌体形态特征,测定其裂菌谱、最佳感染复数、一步生长曲线和生物学特性,进行噬菌体全基因组测序和遗传进化分析。【结果】分离、纯化获得一株能高效裂解大肠杆菌K-12菌株的噬菌体,命名为v B_Eco S_SH2(SH2),噬菌斑呈圆形、大而透明、边缘整齐。电镜观察SH2的头部呈二十面体立体对称,尾部较长。噬菌体的潜伏期为10 min,暴发期为60 min,裂解量高达121 PFU/感染细胞,其最佳感染复数为0.1。基因组测序和比对结果表明,SH2的核酸类型为ds DNA,基因组全长为49 088 bp,G+C%含量为45%,Gen Bank登录号为KY985004,结合电镜观察及BLASTp分析,确定其属于有尾噬菌体目长尾噬菌体科成员。同源性及进化分析表明,该噬菌体为大肠杆菌T1样噬菌体的新成员。【结论】分离鉴定了一株裂解效率极高的大肠杆菌T1样噬菌体,并确认其为T1样噬菌体新成员,为研究大肠杆菌噬菌体及其抗菌应用提供了新的实验材料。  相似文献

Phage H22 was isolated from sewage using Pseudomonas aeruginosa NCTC 8505 (serotype 0:3) as the host. Although not O-specific, this phage was found to have lipopolysaccharide (LPS) as a receptor. The broad host-range and lack of O-specificity of the phage suggested that its receptor site was in the core region of the LPS. Phage H22 had a Bradley type A structure. It was unaffected by chloroform and diethyl ether, and was stable between pH 5 and 8 and in the temperature range 0 to 60 degrees C. The adsorption rate constant was 14.6 X 10(-9) ml min-1. The phage had a latent period of 43 min, with a rise time of 18 min and a burst size of 6. The adsorption of phage to whole cells and LPS occurred over a broad pH range. Maximum adsorption occurred at 50 degrees C and pH 7.5 in the presence of 0.001 M Ca2+.  相似文献

A DNA-containing bacteriophage, designated RS1, infecting Rhodopseudomonas spheroides 2.4.1, has been isolated from sewage. The buoyant density of RS1 in CsCl equilibrium centrifugation is 1.50 g/cm(3), and the buoyant density of RS1 DNA is 1.706. The phage possesses a polyhedral head, approximately 65 nm in diameter, and a tail 60 nm long. When grown on aerobic cells, RS1 has a latent period of 120 min and an average burst size of 20. When grown on anaerobic cells, RS1 has a latent period of 150 min, and a burst size similar to that observed during aerobic infection. The adsorption rate constant of RS1 to aerobic cells is 1.2 x 10(-9) ml/min, and 0.58 x 10(-9) ml/min to anaerobic cells. Adsorption of RS1 to R. spheroides requires the presence of divalent cations.  相似文献

A virulent bacteriophage which causes a decrease in acid production during fermentation of a lactic acid beverage named Yakult with Lactobacillus casei was isolated from the abnormal fermentation tank and named PL-1. L. casei S strain was the exclusive host cell among 18 lactic acid bacteria tested. The plaque was round with an average diameter of about 0.5 mm. It exhibited serological cross-reaction with previously isolated J1 phage. Under an electron microscope, the phage had a spermatozoon shape, with an icosahedral head (63 nm) and a long tail (12.5 by 275 nm) with about 55 striae. The free phage particles were stable at pH 5 to 8. The phage was quite sensitive to ultraviolet irradiation or to heating (60 C, 5 min), and the host was more sensitive than the phage to these treatments. Many kinds of antimicrobial chemicals were also phagocidal. Calcium ion (5 mm) was specifically essential for the phage growth cycle. A one-step growth experiment under optimum conditions (37 C and pH 6.0) showed that the eclipse period was about 75 min, that the latent period was 100 min after the phage infection, and that the average burst size was about 200. The possibility of arresting phage development in lactic acid fermentation is discussed.  相似文献

The temperate bacteriophage phiLC3, isolated from Lactococcus lactis subsp. cremoris, has an isometric head and a flexible tail containing 1 major protein and 8 minor proteins. Infection of a permissive L. lactis host strain yields a burst of about 50 phages per infected cell with a latent period of 60 min. A detailed restriction map of the phage chromosome was constructed by using 12 different restriction enzymes. The phage chromosome is a 33-kb linear double-stranded DNA molecule with unique cohesive ends and with a G + C content of 36.5%. Chemical sequencing of the DNA ends revealed 13-base 3' extended complementary single strands with a relatively high percentage of G + C. Pulsed-field gel electrophoretic analysis of DNA from a strain lysogenized with phiLC3 was used to localize the prophage to a 320-kb BamHI restriction endonuclease fragment from the host chromosomal DNA. This result indicates that lysogeny involves integration of the phage into the host chromosome. A spontaneous phiLC3 clear plaque mutant that was unable to give rise to lysogens was isolated.  相似文献

To search for candidate control agents against Aeromonas salmonicida subsp. salmonicida infections in aquaculture, one bacteriophage (phage), designated as PAS-1, was isolated from the sediment samples of the rainbow trout (Oncorhynchus mykiss) culture farm in Korea. The PAS-1 was morphologically classified as Myoviridae and possessed approximately 48 kb of double-strand genomic DNA. The phage showed broad host ranges to other subspecies of A. salmonicida as well as A. salmonicida subsp. salmonicida including antibiotic-resistant strains. Its latent period and burst size were estimated to be approximately 40 min and 116.7 PFU/cell, respectively. Furthermore, genomic and structural proteomic analysis of PAS-1 revealed that the phage was closely related to other Myoviridae phages infecting enterobacteria or Aeromonas species. The bacteriolytic activity of phage PAS-1 was evaluated using three subspecies of A. salmonicida strain at different doses of multiplicity of infection, and the results proved to be efficient for the reduction of bacterial growth. Based on these results, PAS-1 could be considered as a novel Aeromonas phage and might have potentiality to reduce the impacts of A. salmonicida infections in aquaculture.  相似文献
