Seeds (nutlets) of Rumex obtusifolius L. fail to germinate in darkness at 25° C, but are stimulated by short exposure to red light (R) the effectiveness of which can be negated by a subsequent short exposure to far red light (F) indicating phytochrome control. Short periods of elevated temperature treatment (e.g. 5 min at 35° C) can induce complete germination in darkness. Although short F cannot revert the effect of 35° C treatment, cycling the phytochrome pool by exposure to short R before short F results in reversion of at least 50% of the population. Prolonged or intermittent F can also revert the germination induced by 35° C treatment. The effect of elevated temperature treatment is interpreted on the basis of two possible models; (i) that it increases the sensitivity of the seeds to a low level of pre-existing active form of phytochrome (Pfr) (ii) that it induces the appearance of Pfr in the dark. Summary Plants of a biennial strain ofHyoscyamus niger were either vernalized at a temperature of +2–3°C. for (F) 17, (G) 31 or (A) 48 days, or exposed to alternating temperatures (E: daily 16 hours at +2–3° and 8 hours at +20°; D: daily 8 hours at +2–3° and 16 hours at +20°) for a period of 48 days. The vernalizing effect, as revealed by the date of shooting, was greatest after treatment A; after treatment E it was even smaller than after treatment G, and very little after treatments F and D (majority of plants strictly vegetative at the end of the experiment). This result does not give any hint towards a connection between vernalization and thermoperiodism. — In contrast to the date of shooting, the date of opening the first flower depends, among others, upon the size of the plants at the start of the cold treatment.  相似文献

Interstrain differences in thermoregulation of rats are important in biomedical research because subtleties in thermoregulatory sensitivities may greatly affect data collected. Little is known regarding how individual rodent strains differentially utilize behavioral thermal preference to regulate core temperature (Tc). Sprague-Dawley (SD) and Fischer 344 (F344) rats are known to have differences in thermoregulation including heat tolerance and are useful models to study interstrain differences in thermoregulation. Adult male SD and F344 rats of similar body size were implanted with radiotelemetry thermoprobes (DSI) to measure Tc and MA and housed in either a longitudinal temperature gradient with an ambient temperature (Ta) range of ∼15–40 °C to measure selected Ta (STa) or control environment maintained at a Ta of 23 °C. When continuously monitored for 48 h, Tc and MA increased at night, while STa decreased, according to their normal circadian cycle in both strains. SD rats were more active than F344 rats throughout the circadian cycle (SD gradient: day=12.9±1.2 m/h, night=32.1±2.4 m/h; F344 gradient: day=4.1±0.6 m/h, night=16.8±1.8 m/h; p<0.05 interstrain and circadian effects). The STa of each strain was greater during the daytime (SD: 26.4±0.2 °C; F344: 27.8±0.3 °C) than at night (SD: 24.7±0.3 °C; F344: 25.7±0.3 °C) confirming past studies that thermopreference during the day and night is greater than standard room temperature (∼23 °C). Correlations between MA and Tc suggest that MA has a greater effect on Tc in the F344 but not the SD strain when housed in a temperature gradient. There were significant strain differences in Tc depending on whether rats were housed in a temperature gradient. That is, the control F344 rats had a lower Tc during the transition from dark to light compared to rats housed in a gradient. Tc of the SD strain was unaffected by housing in the gradient. Rats are typically housed at a standard room temperature of 23 °C. However, the results demonstrate that when given the opportunity to behaviorally thermoregulate in a temperature gradient, the F344 strain selects a warmer environment that affects the regulation of Tc. This may be important in the experimenters' choice of ambient temperatures to house and study rats and other rodents.  相似文献

Deletion of phenylalanine 508 (ΔF508) is the most prevalent disease-causing mutation resulting in retention of the immature CFTR in the endoplasmic reticulum. The most common strategy to induce the delivery of ΔF508-CFTR to the surface of cells is by reducing the incubation temperature (≈28 °C). Cell surface biotinylation of HEK293T cells grown at 37 °C for 48 h, confirmed the presence of mature wild-type CFTR, but not ΔF508-CFTR at the cell surface. On the other hand, cells incubated at 28 °C for 16 h showed both mature and immature ΔF508-CFTR at their surface. The trafficking of immature ΔF508-CFTR, but not mature ΔF508-CFTR, to the cell surface occurred at low temperature even upon addition of BFA, suggesting the involvement of a Golgi-independent pathway. These results suggest that low temperature induces the appearance of a mix population of mature and immature CFTR molecules at the plasma membrane through distinct pathways.  相似文献

Summary The interrelationship between the growth ofCoprinus lagopus andFusarium udum on pigeon pea substrates in soil was studied at 22-2°C and 30±2°C in relation to their competitive saprophytic ability and antagonism.C. lagopus was observed to be a potent and frequent colonizer of pigeon pea substrates in soil along withF. udum. Saprophytic colonization of pigeon pea substrate byF. udum precolonized byC. lagopus was inhibited in all inoculum soil mixtures. Saprophytic colonization of the substrate byF. udum was better at 22°C than at 30°C and that byC. lagopus better at 30°C than at 22°C. The colonization of substrate by each fungus was suppressed by the other fungus depending upon the temperature used. Hyphal parasitism and colony interactions between these fungi and also with a number of other saprophytic fungi were investigated. No antagonistic reaction was observed betweenF. udum andC. lagopus. However, these fungi were observed to be potent antagonists against other microfungi tested. The formation of fruiting bodies ofC. lagopus was also observed in the laboratory on nutrient media but more frequently on pigeon pea substrates.  相似文献

Summary A computer model predicted the minimum stem surface temperature, which generally occurred at the apex, for various species of cacti under a particular set of environmental conditions. Based on stem mass, spines, and apical pubescence for the four Ferocactus species found in the southwestern United States, F. acanthodes had the highest minimum apical temperatures and hence was predicted to range the furthest north, F. wizlizenii next, then F. covillei, and finally F. viridescens, in agreement with field observations. Direct measurement of apical temperatures at night showed that F. viridescens was about 2°C colder than a side-by-side F. acanthodes, in agreement with the model. The simulated apical temperature of Trichocereus chilensis increased about 0.3°C for each 50 cm increase in height up to 2 m; observations at a high elevation site in central Chile showed that the freezing damage progressively halved over this sequence of height intervals. The upper elevational limit of Eriosyce ceratistes and T. chilensis at different latitudes from 29°S to 35°S indicated that the populations were responsive to changes of only 0.1°C. Such temperature sensitivities underscore the importance of morphological differences in establishing the low temperature limits on the ranges of cacti in particular and plants in general.  相似文献

Summary Extreme temperatures near the soil surface, which can reach 70°C at the main study site in the northwestern Sonoran Desert, markedly affect seedling survival. Computer simulations indicated that for the rather spherical barrel cactus Ferocactus acanthodes (Lem.) Britt. & Rose the maximum surface temperature decreased 8°C and the minimum temperature increased 3°C as the seedling height was increased from 1 mm up to 50 mm. Simulated changes in shortwave and longwave irradiation alone showed that shading could decrease the maximum temperature by about 5°C for the common desert agave, Agave deserti Engelm., and raise the minimum 1°C. Actual field measurements on seedlings of both species, where shading would affect local air temperatures and wind speeds in addition to irradiation, indicated that shading decreased the average maximum surface temperature by 11°C in the summer and raised the minimum temperature by 3°C in winter.Seedlings grown at day/iight air temperatures of 30°C/20°C tolerated low temperatures of about -7°C and high temperatures of about 56°C, as measured by the temperature where stain uptake by chlorenchyma cells was reduced 50%. Seedling tolerance to high temperatures increased slightly with age, and F. acanthodes was more tolerant than A. deserti. Even taking the acclimation of high temperature tolerance into account (2.7°C increase per 10°C increase in temperature), seedlings of A. deserti would not be expected to withstand the high temperatures at exposed sites, consistent with previous observations that these seedlings occur only in protected microhabitats. Based primarily on greater high temperature acclimation (4.3°C per 10°C), seedlings of F. acanthodes have a greater high temperature tolerance and can just barely survive in exposed sites. Wide ranges in photoperiod had little effect on the thermal sensitivities of either species. When drought increased the chlorenchyma osmotic pressure from about 0.5 MPa to 1.3 MPa, seedlings of both species became about 2°C less tolerant of high temperatures, which would be nonadaptive in a desert environment, and 2°C more tolerant of low temperatures, which also occurs for other species.In conclusion, seedlings of A. deserti and F. acanthodes could tolerate tissue temperatures over 60°C when acclimated to high temperatures and below -8°C when acclimated to low temperatures. However, the extreme environment adjacent to desert soil requires sheltered microhabitats to protect the plants from high temperature damage and also to protect them from low temperature damage at their upper elevational limits.  相似文献

We investigated the behavioural responses of two gobiid fish species to temperature to determine if differences in behaviour and ventilation rate might explain any apparent vertical zonation. A survey of the shore at Manly, Moreton Bay revealed Favonigobius exquisitus to dominate the lower shore and Pseudogobius sp.4 the upper shore. These species were exposed to a range of temperatures (15–40°C) in aquaria for up to 6 h. At 20 °C F. exquisitus exhibited a mean gill ventilation rate of 26 ± 1.4 bpm (beats per minute) differing significantly from Pseudogobius, which ventilated at a fivefold greater rate of 143 ± 6 bpm. The ventilation rate in F. exquisitus underwent a fivefold increase from normal local water temperature (20 °C) to high temperature (35 °C) conditions, whereas that of Pseudogobius did not even double, suggesting that Pseudogobius sp. is a better thermal regulator than F. exquisitus.While both species emerged from the water at high temperatures (>30 °C) the behaviours they exhibited while immersed at high temperature were quite different. F. exquisitus undertook vertical displacement movements we interpret as an avoidance response, whereas Pseudogobius sp. appeared to use a coping strategy involving movements that might renew the water mass adjacent to its body. The thermal tolerances and behaviours of F. exquisitus and Pseudogobius sp. are in broad agreement with their vertical distribution on the shore.  相似文献

A technique has been developed that enables most humans to establish rapid self-regulatory control of their own skin temperature when provided with immediate visual feedback information concerning variations in local skin temperature. Training took place during 15-min periods within 45-min sessions. Clear evidence of learning was usually manifested within 4 sessions. After acquisition occurred, mean change per session was approximately 2.2° F, ranging up to 6.5° F. Training was continued with some subjects who were taught to alter temperature in opposite directions during successive periods on the same day. After practice, these subjects routinely displayed ranges of 9–14° F within 15 min.Portions of this paper were presented at meetings of the Biofeedback Research Society, St. Louis, October 1971, and Boston, November 1972, and the Eastern Psychological Association, New York, April 1972.  相似文献

Skin temperature from three recording sites (web dorsum and two digital sites) on one hand were compared over a 30-minute period during which room temperature was raised from 66° to 80°F causing skin temperatures to range from 78.5° to 92.3°F. The web dorsum remained significantly warmer than either digital site at the lowest skin temperatures; no significant inter-site differences were seen as skin temperature gradually increased. These findings imply that none of the three sites is more advantageous than the others when recording hand temperature values around or above 85°F. However, at the lower levels of skin temperature, there is a floor below which the web dorsum is no longer a sensitive indicator. The production of type A trichothecene mycotoxins by 19 Fusaria, including 12Fusarium sporotrichioides, 4F. chlamydosporum and 3F. graminearum at 15°C and 25°C over a 35-day period was analyzed by ELISA using antibodies cross-reactive with most type A trichothecenes after conversion to T-2 tetraol tetraacetate. The toxin production peaked at 20–25 days of incubation with maximum yield between 4–6 mg type A trichothecene/ml of culture medium for 5F. sporotrichioides cultures and between 1 to 2 mg/ml for 6F. sporotrichioides cultures. OneF. sporotrichioides produced 700 µg type A trichothecenes/ml of culture medium. Detectable type A trichothecene was also found in the culture extracts ofF. chlamydosporum andF. graminearum, but the yield was very low (less than 100 µg/ml). Quantitative determination of individual trichothecenes was achieved by separation of different toxin in HPLC and followed by ELISA analysis. Eight to 10 immunoreactive peaks, corresponding to various type A trichothecenes, were detected in all the fungal extracts. T-2 tetraol (T-2-4ol), 4-acetyl-T-2 tetraol (4-Ac-T-2-4ol), neosolaniol (NEOS), diacetoxyscirpenol (DAS), HT-2 and T-2 toxin accounted for more than 85% of the total toxins. In general, low temperature was preferred for total type A trichothecene production. More T-2-4ol, 4-Ac-T-2-4ol, HT-2 and DAS were produced at 25°C. In contrast, more T-2 toxin and NEOS were produced at 15°C. Transformation of T-2 toxin and NEOS to polar metabolites such as T-2-4ol, 4-acetyl-T-2-4ol and HT-2 by various strains were observed at both temperatures after 25 days incubation.  相似文献

Mortality from acute myocardial infarction (MI) over the 5 year period 1982–1987 in Brown County, Wisconsin, was analyzed to assess the relationship with environmental temperature. Deaths occurrring on the day of and the day following a significant snowfall as well as deaths occuring in health care facilities were eliminated from consideration because the focus was upon temperature, not snowfall or events within a hospital. These criteria resulted in the inclusion of 1,802 days and 926 cases of acute MI. The mean temperature on the day of death was obtained from climatological data and were grouped into six categories covering a range of temperatures from<–17.8°C (0°F) to 16.1°C (61°F). The number of deaths in each category was tabulated. The effect of temperature, sex, and age were analyzed by regression analysis. The results indicated a linear increase in mortality as mean daily temperature decreased over the temperature range. The inverse temperature effect was most pronounced in males over the age of 60. These results indicate that cold temperatures appear to be associated with an increased mortality from myocardial infarction.  相似文献

The ribosome inactivating proteins (RIPs) of type 1 are plant toxins that eliminate adenine base selectively from the single stranded loop of rRNA. We report six crystal structures, type 1 RIP from Momordica balsamina (A), three in complexed states with ribose (B), guanine (C) and adenine (D) and two structures of MbRIP-1 when crystallized with adenosine triphosphate (ATP) (E) and 2′-deoxyadenosine triphosphate (2′-dATP) (F). These were determined at 1.67 Å, 1.60 Å, 2.20 Å, 1.70 Å, 2.07 Å and 1.90 Å resolutions respectively. The structures contained, (A) unbound protein molecule, (B) one protein molecule and one ribose sugar, (C) one protein molecule and one guanine base, (D) one protein molecule and one adenine base, (E) one protein molecule and one ATP-product adenine molecule and (F) one protein molecule and one 2′-dATP-product adenine molecule. Three distinct conformations of the side chain of Tyr70 were observed with (i) χ1 = − 66°and χ2 = 165° in structures (A) and (B); (ii) χ1 = − 95° and χ2 = 70° in structures (C), (D) and (E); and (iii) χ1 = − 163° and χ2 = 87° in structure (F). The conformation of Tyr70 in (F) corresponds to the structure of a conformational intermediate. This is the first structure which demonstrates that the slow conversion of DNA substrates by RIPs can be trapped during crystallization.  相似文献

Inhibition and recovery of net CO2 uptake and three photosynthetic electron transport reactions as well as plant survival following high-temperature treatments were investigated for Opuntia ficus-indica. For plants maintained at 30°C/20°C day/night air temperatures, treatment at 60°C for 1 h irreversibly inhibited net CO2 uptake and photosynthetic electron transport, resulting in plant death in about 60 days. When a plant maintained at 30°C/20°C was treated at 55°C for 1 h, net CO2 uptake was completely inhibited 1 d after the treatment but fully recovered in 60 d. Differential inactivation of photosystem (PS) I, PSII, and whole chain electron transport activities occurred; PSI was the most tolerant of 55°C and took the least time (45 d) for total recovery. All 30°C/20°C plants survived a 1-h treatment at 55°C, although some pale green areas were observed on the cladode surfaces. In contrast to growing at 30°C/20°C, plants acclimated to 45°C/35°C survived 60°C for 1 h without showing any necrotic or pale green areas on the cladode surfaces. When such a plant was transferred to 30°C/20°C following the high-temperature treatment, recovery in net CO2 uptake began in 1 d and progressed to complete recovery by 30 d. The effects of growth temperature and winter duration on the leaf phenology of Erythronium japonicum were examined in two experiments. Bulbs wintered in the field were cultivated at 10 and 20° C and the bulbs were cultivated at 15° C after chilling treatment at 3° C for 60 and 120 days and without chilling in winter. The plants cultivated at 20° C showed significantly earlier leaf emergence, a more rapid rate of leaf extension and shorter leaf longevity than those cultivated at 10° C. The decrease in the leaf longevity at 20° C resulted from the decreases in the durations of all of the developmental, mature, and senescent phases. The bulbs without chilling treatment did not sprout leaves and those with chilling treatment sprouted leaves. The increase in the length of chilling treatment from 60 to 120 days affected leaf phenology in same manner as the increase in the growth temperature from 10 to 20° C.  相似文献

Summary Comparison of different post-plating temperature regimes with a control treatment (27° C) revealed that a short-term cold (8/14°C:2/2 days or 14°C:4 days) as well as a heat treatment (30°C:14 days) increased the production of embryro-like-structures (ELS) from cultured maize anthers. The beneficial effects of short-term cold treatments were magnified 2–3 times when L-proline (PROL) was added to the induction medium (125–500 mg/L). In the best treatment (14°C:4 days, 125 mg/L L-proline) one genotype produced 143.5 ELS/100 anthers. Anthers subjected to high temperature (30°C:4 days, 30°C:7 days, 30°C:14 days) generally showed a lower response than did cold treated anthers, although genotypic differences were observed. The cell-bound polysaccharides (PSs) of Bifidobacterium adolescentis 94 BIM were stripped from the cell surface with 2% sodium dodecyl sulfate (SDS), 1.5% Cetavlon, and 1% Triton X-100 and purified by precipitation with 5 volumes of ethanol. According to the extraction conditions used, the polysaccharide preparations were designated as PS-SDS-6°C, PS-SDS-100°C, PS-Cet, and PS-Trit. The gel-permeation chromatography of the first two preparations with the use of a Bio-Gel P-10 column and 1% acetic acid as the eluant yielded two peaks, F1 and F2, which contained carbohydrates and no phosphorus. All polysaccharides were primarily composed of glucose and galactose. The polysaccharides PS-Cet and PS-Trit were found to be branched and contain glucose residues at the terminal position, position 4, and position 6, and galactose residue at position 3. The effects of temperature acclimation and acute temperature change were investigated in postprandial green shore crabs, Carcinus maenas. Oxygen uptake, gut contractions and transit rates and digestive efficiencies were measured for crabs acclimated to either 10 °C or 20 °C and subsequently exposed to treatment temperatures of 5, 15, or 25 °C. Temperature acclimation resulted in a partial metabolic compensation in unfed crabs, with higher oxygen uptake rates measured for the 10 °C acclimated group exposed to acute test temperatures. The Q10 values were higher than normal, probably because the acute temperature change prevented crabs from fully adjusting to the new temperature. Both the acclimation and treatment temperature altered the characteristics of the specific dynamic action (SDA). The duration of the response was longer for 20 °C acclimated crabs and was inversely related to the treatment temperature. The scope (peak oxygen consumption) was also higher for 20 °C acclimated crabs with a trend towards an inverse relationship with treatment temperature. Since the overall SDA (energy expenditure) is a function of both duration and scope, it was also higher for 20 °C acclimated crabs, with the highest value measured at the treatment temperature of 15 °C. The decline in total SDA after acute exposure to 5 and 25 °C suggests that both cold stress and limitations to oxygen supply at the temperature extremes could be affecting the SDA response. The contractions of the pyloric sac of the foregut region function to propel digesta through the gut, and contraction rates increased with increasing treatment temperature. This translated into faster transit rates with increasing treatment temperatures. Although pyloric sac contractions were higher for 20 °C acclimated crabs, temperature acclimation had no effect on transit rates. This suggests that a threshold level in pyloric sac contraction rates needs to be reached before it manifests itself on transit rates. Although there was a correlation between faster transit times and the shorter duration of the SDA response with increasing treatment temperature, transit rates do not make a good proxy for calculating the SDA characteristics. The digestive efficiency showed a trend towards a decreasing efficiency with increasing treatment temperature; the slower transit rates at the lower treatment temperatures allowing for more efficient nutrient absorption. Even though metabolic rates of 10 °C acclimated crabs were higher, there was no effect of acclimation temperature on digestive efficiency. This probably occurred because intracellular enzymes and digestive enzymes are modulated through different control pathways. These results give an insight into the metabolic and digestive physiology of Carcinus maenas as it makes feeding excursions between the subtidal and intertidal zones.  相似文献

The ability to germinate under a variety of environmental conditions is essential for plant species inhabiting a wide range of altitudes and latitudes. Phacelia secunda J. F. Gmel. (Hydrophyllaceae) is a perennial herb with wide latitudinal and altitudinal distributional ranges. In the central Chilean Andes (33 °S) P. secunda can be found from 1600 m sealevel up to the vegetation limit at 3400 m. It has been suggested that seeds from populations encountering long periods with snow cover and adverse winter conditions would require longer periods of cold stratification for germination than those from populations exposed to milder winters. Given that the snow-free period decreases with elevation, seeds from high elevation populations could require longer period of cold stratification to germinate. Moreover, it has been shown that seeds from arctic and higher elevations environments are adapted to germinate better under high temperature conditions. Germination response with increasing periods of cold stratification (0–6 mo.) and under two contrasting thermoperiods (20 °/1O °C; 10 °/5 °C; 12 h day/night), were studied for 4 populations of P. secunda located at 1600, 2100, 2900 and 3400 m a.s.l. Initiation of germination required increasingly longer periods of stratification with elevation, and proportionately fewer seeds germinated for any one stratification treatment at the higher elevations. Seeds from higher elevations germinated to a higher percentage under the high than the low temperature thermoperiods. These results illustrates a significant variation in germination characteristics over a spatially short environmental gradient.  相似文献

A study was made of the effect of high temperature on the growth response of Brussels sprout filaments to ethylene. Filaments with or without the anthers attached were incubated continuously at 25 °C or 35 °C for 7 days or for 2 days at 35 °C followed by 5 days at 25 °C. Growth was reduced during both 35 °C treatments compared to that of filaments at continuous 25 °C. Ethylene had little effect on filament growth at continuous 25 °C, whereas with treatment for either 2 or 7 days at 35 °C ethylene promoted filament growth considerably. Thus ethylene effectively overcame the growth inhibition induced by the 35 °C treatment.High temperature treatments reduced ethylene production from filaments alone, and from filaments with anthers attached. The ethylene biosynthesis inhibitor aminoethoxyvinylglycine (AVG) and the ethylene action inhibitor AgNO3 enhanced filament growth at 25 °C but had little or no effect at 35 °C. The relevance of temperature to ethylene sensitivity is discussed in relation to filament growth and to other plant processes in general.  相似文献
