It has been proposed that Fe stress may be used in the study of limiting factors in photosynthesis as an experimental means of varying photochemical capacity in vivo (Plant Physiol 1980 65: 114-120). In this paper the effect of Fe stress on photosynthetic unit number, size, and composition was investigated by measuring P700, cytochrome (Cyt) f, chlorophyll (Chl) a, and Chl b in sugar beet leaves. The results show that when Fe stress reduced Chl per unit area by 80% (from 60 to 12 micrograms per square centimeter), it decreased the number of P700 molecules per unit area by 88% and Cyt f per unit area by 86%; over the same range the Chl to P700 ratio increased by 37% but there was no significant change in the Chl to Cyt f ratio. These data suggest that Fe stress decreases photochemical capacity and Chl per unit area by diminishing the number of photosynthetic units per unit leaf area.  相似文献

At present, chlorophyll meters are widely used for a quick and nondestructive estimate of chlorophyll (Chl) contents in plant leaves. Chl meters allow to estimate the Chl content in relative units - the Chl index (CI). However, using such meters, one can face a problem of converting CI into absolute values of the pigment content and comparing data acquired with different devices and for different plant species. Many Chl meters (SPAD-502, CL-01, CCM-200) demonstrated a high degree of correlation between the CI and the absolute pigment content. A number of formulas have been deduced for different plant species to convert the CI into the absolute value of the photosynthetic pigment content. However, such data have not been yet acquired for the atLEAF+ Chl meter. The purpose of the present study was to assess the applicability of the atLEAF+ Chl meter for estimating the Chl content. A significant species-specific exponential relationships between the atLEAF value (corresponding to CI) and extractable Chl a, Chl b, Chl (a+b) for Calamus dioicus and Cleistanthus sp. were shown. The correlations between the atLEAF values and the content of Chl a, Chl b, and Chl (a+b) per unit of leaf area was stronger than that per unit of dry leaf mass. The atLEAF value- Chl b correlation was weaker than that of atLEAF value-Chl a and atLEAF value-Chl (a+b) correlations. The influence of light conditions (Chl a/b ratio) on the atLEAF value has been also shown. The obtained results indicated that the atLEAF+ Chl meter is a cheap and convenient tool for a quick nondestructive estimate of the Chl content, if properly calibrated, and can be used for this purpose along with other Chl meters.  相似文献

The interactive effects of shade and drought on the morphological and physiological traits of Catalpa bungei plantlets were assessed. Seedling growth, biomass, biomass allocation, leaf morphology, chlorophyll (Chl) content and gas-exchange parameters were measured in plants raised for 3 months under three light levels [80% (HI), 50% (MI), 30% (LI)] and two water levels [moisture (M) and drought (D)]. The results showed that shade greatly decreased growth, biomass, leaf area (LA) and Chl a/b; increased specific leaf area (SLA) and Chl content; and reduced photosynthetic rate (P n). Drought reduced the growth, biomass, LA, SLA, Chl a/b, P n, stomatal conductance (G s), transpiration rate (T r) and intercellular carbon dioxide concentration (C i) and increased the Chl content. Stomatal closure was an early physiological response to water stress. Light, water and their interaction significantly affected plant traits and their bivariate relationships. The phenotypic plasticity index of light (0.47) was much higher than that of water (0.21), indicating that light was the main driver of the variations observed. Under drought stress, growth, biomass, leaf and stem biomass allocation significantly decreased in the HI and MI environments, whereas no significant difference was observed in growth or biomass parameters under the LI condition. Furthermore, no significant difference was observed in P n, G s, or T r under the LI condition under water stress. Our results showed that shade did not alter the negative effects caused by drought stress in MI but did alleviate the negative effects of the LI condition. In summary, the effect of drought on C. bungei plantlets depends on the irradiance conditions.  相似文献

Using iron stress to reduce the total amount of light-harvesting and electron transport components per unit leaf area, the influence of light-harvesting and electron transport capacity on photosynthesis in sugar beet (Beta vulgaris L. cv F58-554H1) leaves was explored by monitoring net CO2 exchange rate (P) in relation to changes in the content of Chl.

In most light/CO2 environments, and especially those with high light (≥1000 microeinsteins photosynthetically active radiation per square meter per second) and high CO2 (≥300 microliters CO2 per liter air), P per area was positively correlated with changes in Chl (a + b) content (used here as an index of the total amount of light-harvesting and electron transport components). This positive correlation of P per area with Chl per area was obtained not only with Fe-deficient plants, but also over the normal range of variation in Chl contents found in healthy, Fe-sufficient plants. For example, light-saturated P per area at an ambient CO2 concentration close to normal atmospheric levels (300 microliters CO2 per liter air) increased by 36% with increase in Chl over the normal range, i.e. from 40 to 65 micrograms Chl per square centimeter. Iron deficiency-mediated changes in Chl content did not affect dark respiration rate or the CO2 compensation point. The results suggest that P per area of sugar beet may be colimited by light-harvesting and electron transport capacity (per leaf area) even when CO2 is limiting photosynthesis as occurs under field conditions.

Sexual dimorphisms of dioecious plants are important in controlling and maintaining sex ratios under changing climate environments. Yet, little is known about sex-specific responses to elevated CO2 with soil nitrogen (N) deposition. To investigate sex-related physiological and biochemical responses to elevated CO2 with N deposition, Populus cathayana Rehd. was employed as a model species. The cuttings were subjected to two CO2 regimes (350 and 700???mol?mol?1) with two N levels (0 and 5?g?N?m?2?year?1). Our results showed that elevated CO2 and N deposition separately increased the total number of leaves, leaf area (LA), leaf mass, net photosynthetic rate (P n), light saturated photosynthetic rate (P max), chlorophyll a (Chl a), and chlorophyll a to chlorophyll b ratio (Chl a/b) in both males and females of P. cathayana. However, the effects on LA, leaf mass, P n, P max, Chl a and Chl a/b were weakened under the combined treatment of elevated CO2 and N deposition. Males had higher leaf mass, P n, P max, apparent quantum yield (??), carboxylation efficiency (CE), Chl a, Chl a/b, leaf N, and root carbon to N ratio (C/N) than did females under elevated CO2 with N deposition. In contrast to males, females had significantly higher levels of soluble sugars in leaves and greater starch accumulation in roots and stems under the same condition. The results of the present work imply that P. cathayana females are more responsive and suffer from greater negative effects on growth and photosynthetic capacity than do males when grown under elevated CO2 with soil N deposition.  相似文献

The photosynthetic characterization of the oriental lily (Lilium) cv. Sorbonne and its response to increasing water stress were analyzed based on the net photosynthetic rate (P n), stomatal conductance (g s), intercellular CO2 concentration (Ci), transpiration rate (E), water use efficiency (WUE), and stomatal limitation (Ls) in the Horqin Sandy Land of western China. A photosynthesis-PAR response curve was constructed to obtain light-compensation and light-saturation points (LCP and LSP), the maximum photosynthetic rates (P max) and dark respiration rates (R D). The growth of lilies in the pots was analyzed after anthesis. Various intensities of water stress (5, 10, and 20 days without water, and an unstressed control) were applied. The results indicated that drought stress not only significantly decreased P n, E, g s, photosynthetic pigment content (Chl a, Chl b, and Chl (a + b)) and increased intrinsic water use efficiency (WUE), but also altered the diurnal pattern of gas exchange. Drought stress also affected the photosynthesis (P n)-PAR response curve. Drought stress increased LCP and R D and decreased LSP and P max. There were both stomatal and nonstomatal limitations to photosynthesis. Stomatal limitation dominated in the morning, whereas nonstomatal limitation dominated in the afternoon. Thus, drought stress decreased potential photosynthetic capacity and affected the diurnal pattern of gas exchange and P n-PAR response curves, thereby reducing plant quality (lower plant height, flower length, flower diameter, and leaf area). Water stress is likely the main limitation to primary photosynthetic process in the lily. Appropriate watering is recommended to improve photosynthetic efficiency and alleviate photodamage, which will increase the commercial value of the lily in the Horqin Sandy Land.  相似文献

To understand the ecophysiological adaptation mechanisms of Calligonum roborovskii to altitude variation, this study analyzed chlorophyll a (Chl a), chlorophyll b (Chl b), Chl (a + b), carotenoid (Car), malondialdehyde (MDA), ascorbate (AsA), proline (Pro), membrane permeability (MP), reactive oxygen species (ROS), specific leaf area (SLA), leaf mass per area (LMA), leaf nitrogen content based on mass (Nmass), and the activities of peroxidase (POD), catalase (CAT), superoxide dismutase (SOD), and ascorbate peroxidase (APX) in leaves of plants inhabiting different altitudes (A1: 2100 m, A2: 2350 m, A3: 2600 m) on the northern slope of the Kunlun Mountains. The results showed that Chl a, Chl b, Chl (a + b), SLA, Nmass, and the activity of CAT increased with increasing altitude. LMA, MP, MDA, Car, Pro, AsA, O2, H2O2 and the activities of SOD, POD, and APX decreased with increasing altitude. The test results also showed that, changes in venvironmental factors along an altitudinal gradient are not obvious. Soil water content is the main ecological factor. With increasing altitude, soil water content increased significantly. More non-enzymatic and enzymatic antioxidants played an important role in eliminating intracellular ROS. They kept the cell membrane in a stable state and ensured the normal growth of C. roborovskii.  相似文献

Patchouli (Pogostemon cablin Benth.) plants grown under shade (LI) showed an increased height due to internodal elongation, leaf area, leaf area index, and chlorophyll (Chl)b, Chl (a+b) and carotenoid (Car) contents compared to the plants grown in sunlight (HI). The number of branches and green leaves decreased under LI treatment, with a marginal variation in the patchouli oil yield. A comparison between the relative contents of photosynthetic pigments indicated that Chlb and Car accumuled preferentially over Chla in the LI grown plants.  相似文献

Agronomic traits, photosynthetic pigments, gas exchange, and chlorophyll (Chl) fluorescence parameters of red stem buckwheat (Fagopyrum dibotrys Hara) mutants induced by γ-radiation were compared with green control at seedling stage. Plant height, number of first-class branches, and rhizome biomass were inhibited significantly (p<0.01). Chl a, Chl b, and Chl a+b contents decreased with elevated dose of γ-rays, while increasing carotenoid content indicated that buckwheat was capable of adjusting to the radiation damage. Decrease in net photosynthetic rate was the result of both stomatal and non-stomatal limitations. Fluorescence parameters, such as F0, Fm, Fv/Fm, Fv/F0, ΦPS2, electron transport rate, and photochemical quenching declined significantly (p<0.01) as compared with control due to photoinhibition, while non-photochemical quenching increased to enhance thermal dissipation. Lower parameters implied that leaf tissue was damaged significantly by high dose of γ-radiation and therefore leaf senescence was accelerated.  相似文献

In order to investigate the effect of day/night temperature difference (DIF) on photosynthetic characteristics of tomato plants (Solanum lycopersicum, cv. Jinguan 5) at fruit stage, an experiment was carried out in climate chambers. Five day/night temperature regimes (16/34, 19/31, 25/25, 31/19, and 34/16°C) with respective DIFs of -18, -12, 0, +12, and +18 were used and measured at mean daily temperature of 25°C. The results showed that chlorophyll (Chl) a, Chl b, net photosynthetic rate (PN), stomatal conductance (gs), maximum quantum yield of PSII photochemistry (Fv/Fm), effective quantum yield of PSII photochemistry (?PSII), and photochemical quenching (qp) significantly increased under positive DIF, while they decreased with negative DIF. In contrast, the Chl a/b ratio and nonphotochemical quenching (NPQ) decreased under positive DIF, while increased with negative DIF. Chl a, Chl b, PN, gs, Fv/Fm, ?PSII, and qp were larger under +12 DIF than those at +18 DIF, while Chl a/b and NPQ showed an opposite trend.  相似文献

The effect of supplementary UV-B radiation on Korean pine (Pinus koraiensis Sieb. et Zucc) was investigated. Compared with the control, the T1, T2, and T3 UV-B treatments increased by 1.40, 2.81, and 4.22 kJ m?2 d?1, respectively. Gas-exchange parameters, photosynthetic pigment concentrations, contents of secondary metabolites, epicuticular wax, free radical, malondialdehyde (MDA), and the activities of antioxidant enzymes were determined after 40 d of exposure. The concentrations of chlorophyll (Chl) a, Chl b, total Chl, carotenoid (Car), and the ratio Chl a/b in the pine needles were in the following order: T1 > T2 > T3. Compared with the control, the contents of flavonoids and epicuticular wax significantly decreased in all levels of supplementary UV-B radiations (p<0.05). Moreover, the contents of hydrogen peroxide (H2O2) and MDA significantly increased with the enhanced UV-B radiations (p<0.05). Korean pine can increase the catalase, ascorbate peroxidase, and superoxide dismutase activities to prevent oxidative stress by supplementary UV-B radiation. However, its defence mechanism is not efficient enough to prevent UV-Binduced damage.  相似文献

The faldh gene encodes the Brevibacillus brevis glutathione-dependent formaldehyde dehydrogenase (FALDH), an enzyme involved in formaldehyde metabolism. In the present work, we have investigated the physiological characteristics of transgenic faldh tobacco under formaldehyde stress. Overexpression of B. brevis FALDH confers tobacco tolerance to high HCHO concentrations. The transgenic tobacco lines had the higher biomass, produced the higher content of total proteins and soluble sugars, the lower levels of MDA, protein carbonyl (PC), and H2O2 as compared with the wild-type tobacco under HCHO stress. The contents of chlorophyll (Chl), including Chl a, Chl b, and the ratio of Chl a/b, and the content of anthocyanidin in transgenic plants under HCHO stress were also higher than that in wild-type tobacco. These results show that high HCHO tolerance and changes of physiological characteristics related to stress tolerance were due to the overexpressing of FALDH in tobacco.  相似文献

Effects of plant hormones indole-3-yl-acetic acid (IAA), gibberellic acid (GA), benzylaminopurine (BAP), abscisic acid (ABA) and ethrel (ETH) in 5 M concentration on gas exchange, ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO, EC 4.1.1.39) activity, pigment content and yield in cotton (Gossypium hirsutum L. cv. H-777) under drought were studied. At reproductive stage (55 – 60 d after sowing) these hormones were sprayed on shoots one day prior to stress imposition by withholding irrigation. The soil moisture of control plants was kept at field capacity. Net photosynthetic rate (PN), stomatal conductance (gs), transpiration rate (E), carboxylation efficiency (CE), water use efficiency (WUE), RuBPCO activity, boll number per plant, seed number per plant and lint mass per plant significantly decreased at drought while chlorophyll (Chl) b content and flower number per plant increased. ABA and ETH significantly reduced gas exchange parameters, Chl a and Chl b content. Detrimental drought effect on PN, gs, E, CE, RuBPCO and lint mass per plant was significantly alleviated by BAP and also its effect on seed number and lint mass per plant was significantly alleviated with the ABA treatment.  相似文献

We investigated the photosynthetic characteristics of Chorispora bungeana under conditions of drought stress caused by different concentrations of polyethylene glycol-6000 (PEG; 0, 5, 20, and 40%) and various concentrations of exogenous glycine (0, 5, 10, and 20 mM) with 20% PEG. We showed that moderate and severe drought stress of PEG reduced the chlorophyll (Chl) content (both Chl a and b), maximal quantum yield of PSII photochemistry (Fv/Fm), actual photochemical efficiency of PSII in light (YII), and quantum yield of regulated energy dissipation (YNPQ), while Chl a/b and quantum yield of nonregulated energy dissipation (YNO) increased. The low and moderate drought stress increased Mg2+ and Fe3+ contents, while a decrease in Mg2+ and Fe3+ was found under severe drought stress. Compared to sole PEG stress, the addition of exogenous 10 mM glycine increased Chl, Mg2+ and Fe3+ contents, Fv/Fm, YII, and YNPQ, and reduced YNO. On the contrary, 20 mM glycine showed an opposite effect, except for YNO. Our results proved that Chl contents and fluorescence parameters are reliable indicators for drought tolerance of C. bungeana. We suggest that a proper glycine content can relieve the effect of drought stress on C. bungeana.  相似文献

We found inconsistencies in the commonly used data for chlorophyll analysis in 80% acetone. Recently developed extinction coefficients for chlorophyll b in N,N-dimethylformamide (DMF) based on values from 80% acetone are low as a result of these inconsistencies. We determined extinction coefficients of chlorophyll a (Chl a) and chlorophyll b (Chl b) in DMF for wavelengths of 618 to 665 nanometers. The simultaneous equations necessary for quantifying Chl a, Chl b, or total Chl in DMF in the absence of other chlorophyllous pigments are: Chl a = 12.70A664.5 - 2.79A647; Chl b = 20.70 A647 - 4.62A664.5; total Chl = 17.90A647 + 8.08A664.5, where A = absorbance in 1.00 centimeter cuvettes and Chl = milligrams per liter.

N,N-Dimethylformamide is a very convenient solvent for Chl extraction since it is effective on intact plant parts and Chl is quite stable in DMF. There was no difference in the amount of Chl extracted when plant tissue was stored for 1 or 3 days at three temperatures, with or without solvent added.

The size of the Emerson photosynthetic unit was measured in Chlorella pyrenoidosa strain no. 252 grown at light intensities between 50 and 1000 foot candles. The Emerson photosynthetic unit changed from a minimum size of 1970 molecules chlorophyll a + b/O2 per flash in cells grown at 1000 foot candles to a maximum size of 3150 molecules chlorophyll a + b/O2 per flash for cells grown at 50 foot candles. The size changes were interpreted as a partial adaptation where the trapping center antenna responded to changes in incident light intensity. Light-induced changes in chlorophyll content and size of the Emerson photosynthetic unit were directly related.  相似文献

The finding of unique Chl d- and Chl f-containing cyanobacteria in the last decade was a discovery in the area of biology of oxygenic photosynthetic organisms. Chl b, Chl c, and Chl f are considered to be accessory pigments found in antennae systems of photosynthetic organisms. They absorb energy and transfer it to the photosynthetic reaction center (RC), but do not participate in electron transport by the photosynthetic electron transport chain. However, Chl d as well as Chl a can operate not only in the light-harvesting complex, but also in the photosynthetic RC. The long-wavelength (Qy) Chl d and Chl f absorption band is shifted to longer wavelength (to 750 nm) compared to Chl a, which suggests the possibility for oxygenic photosynthesis in this spectral range. Such expansion of the photosynthetically active light range is important for the survival of cyanobacteria when the intensity of light not exceeding 700 nm is attenuated due to absorption by Chl a and other pigments. At the same time, energy storage efficiency in photosystem 2 for cyanobacteria containing Chl d and Chl f is not lower than that of cyanobacteria containing Chl a. Despite great interest in these unique chlorophylls, many questions related to functioning of such pigments in primary photosynthetic processes are still not elucidated. This review describes the latest advances in the field of Chl d and Chl f research and their role in primary photosynthetic processes of cyanobacteria.  相似文献

The variation of light intensity has obvious effects on leaf external morphology, internal anatomy, and physiological characteristics; it even induces changes in secondary metabolite production. The effects of different irradiance levels on biomass, gas exchange parameters, and photosynthetic pigment contents in Mahonia bodinieri (Gagnep.) Laferr. were analyzed here. Combined analyses of physiology, cytology, and HPLC were used to study the differences in leaf morphology, structure, physiological characters, and alkaloid content in response to different irradiances. The results indicated that the highest foliar biomass was observed under I 50 (50 % of full sunlight) followed by I 30 (30 % of full sunlight), the highest net photosynthetic rate, stomatal conductance, transpiration rate values were observed under I 30 followed by I 50, and lower values occurred in I 10 (10 % of full sunlight) and I 100 (full sunlight). With increased light intensity, total leaf area and the contents of chlorophyll a (Chl a), chlorophyll b (Chl b), and chlorophyll (Chl a+b) per unit leaf area were clearly reduced, whereas leaf mass per area, carotenoid content, leaf thickness, thickness of palisade and spongy parenchyma, and stomatal density were all significantly increased. Electron microscopic observation revealed that the number of grana, stroma lamellae and the number of starch grains in chloroplasts were decreased, the number of plastoglobuli was increased when irradiance levels increased. The estimated total yield of alkaloids in a single plant was higher under I 30 and I 50 than under I 10 or I 100 as a result of the higher biomass of the plants. Therefore, I 30 and I 50 were not only beneficial to increase biomass, but also suitable for the synthesis and accumulation of the major secondary metabolites (alkaloids). Our findings provide valuable data for the determination and regulation of irradiance levels during artificial cultivation of M. bodinieri.  相似文献

Cell suspension cultures were established from the callus proliferation of leaf explants of 10- to 12-day-old seedlings of the peanut (Arachis hypogaea L. var. TMV-3). The cells could be cultivated in both agitated and still media, the latter promoting more of chlorophyll (Chl) synthesis. High Chl content (210-240 micrograms Chl per gram fresh weight), yield of free and pipetable cells, presence of all the pigments in the same ratio as that of the leaf tissue, and high rates of O2 evolution (140-170 micromoles O2 per milligram Chl per hour) were some of the desirable features of the still-grown cell cultures. However, considerable variations with regard to the above characters were observed between the cell cultures of different varieties of the peanut.

O2 evolution by the cultured cells was dependent on exogenous supply of HCO3. A well-developed photosynthetic apparatus as evidenced from photosystem I and photosystem II activities of the isolated chloroplasts and variable fluorescence measurements with the cell cultures was further documented by electron microscopic evidence of distinct granal stackings in chloroplasts and sodium dodecyl sulfate-polyacrylamide gel separation of thylakoid membranes into P700 Chl a protein complex and light-harvesting Chl a/b complex. Evidence is presented for the relative increase in the Chl associated with P700 Chl a protein complex in contrast to the light-harvesting Chl a/b complex in the cultured cells as compared to intact leaf.

Caragana korshinskii Kom. is a perennial xerophytic shrub, well known for its ability to resist drought. In order to study ecophysiological responses of C. korshinskii under extreme drought stress and subsequent rehydration, diurnal patterns of gas exchange and chlorophyll (Chl) fluorescence parameters of photosystem II as well as Chl content were analyzed. Plant responses to extreme drought included (1) leaf abscission and using stem for photosynthesis, (2) improved instantaneous water-use efficiency, (3) decreased photosynthetic rate and partly closed stomata owing to leaf abscission and low water status, (4) decreased maximum photochemical efficiency of photosystem II (PSII) (variable to maximum fluorescence ratio, Fv/Fm), quantum efficiency of noncyclic electron transport of PSII, and Chl a and Chl b. Four days after rehydration, new leaves budded from stems. In the rewatered plants, the chloroplast function was restored, the gas exchange and Chl fluorescence returned to a similar level as control plant. The above result indicated that maintaining an active stem system after leaf abscission during extreme drought stress may be the foundation which engenders these mechanisms rapid regrowth for C. korshinskii in arid environment.  相似文献
