The long-term survival of Escherichia coli in river water   总被引:11,自引:0,他引:11  
Escherichia coli introduced into autoclaved filtered river water survived for up to 260 d at temperatures from 4 degrees to 25 degrees C with no loss of viability. Survival times were less in water which was only filtered through either a Whatman filter paper or a 0.45 micron Millipore filter or in untreated water, suggesting that competition with the natural microbial flora of the water was the primary factor in the disappearance of the introduced bacteria. Survival was also dependent upon temperature with survival at 4 degrees C greater than 15 degrees C greater than 25 degrees C greater than 37 degrees C for any water sample. Direct counts showed that bacterial cells did not disappear as the viable count decreased. The possession of the antibiotic resistance plasmids, R1drd-19 or R144-3, did not enhance survival nor cause a faster rate of decay, indicating that the metabolic burden imposed by a plasmid was not a factor in survival under starvation conditions. There was no evidence of transfer of either plasmid at 15 degrees C or of loss of plasmid function during starvation.  相似文献

Cattle and sheep are major reservoirs of Escherichia coli O157 and consequently these and certain other farm animals can pass out large numbers of this organism in their faeces. Thus the ability of the organism to survive in faeces, on pastureland and in associated water systems has important implications for its spread to crops by direct application of manure, by irrigation with infected water or directly to man by contact with animals or contaminated soil. Model systems were used to determine the persistence of the organism in river water, cattle faeces, soil cores and on stainless steel work surfaces. Survival of the organism was found to be greatest in soil cores containing rooted grass. Under these conditions viable numbers were shown to decline from approximately 10(8) g(-1) soil to between 10(6) and 10(7) g(-1) soil after 130 d. When the organism was inoculated into cattle faeces it remained detectable at high levels for more than 50 d. In contrast the organism survived much less readily in cattle slurry and river water where it fell in numbers from more than 10(6) ml(-1) to undetectable levels in 10 and 27 d, respectively. The survival of E. coli O157 was also investigated on stainless steel surfaces, where as air-dried deposits, it was shown to survive for periods in excess of 60 d. It was most stable at chill temperatures (4 degrees C) and viability was only partially reduced at 18 degrees C. In addition to stainless steel, the organism was shown to survive for extended periods on domestic (plastic) cutting boards, both at room and chill temperatures. Sanitizing agents, such as hypochlorites and a compound comprising both cationic and anionic-based active ingredients were found to be effective in killing various VTEC on stainless steel surfaces.  相似文献

Gravid females of Capnia bifrons (Newman) from Windermere (English Lake District) were almost completely ovoviviparous, the eggs hatching within 15 min after oviposition in the water. When kept in the laboratory at constant temperatures between 3.8 and 19.8°C, few females survived to lay eggs at temperatures above 12.1°C. The relationship between air temperature (T°C) and the egg incubation period (Y days between fertilisation and oviposition) was given by the regression equation: Y = 316.4 T−0.9996 (r2= 0.957, p < 0.001). This equation successfully predicted egg incubation periods for gravid females kept in cages in the field.
Comparisons with similar studies on four non-ovoviviparous species of Plecoptera showed that egg development was rarely more rapid in C. bifrons . It was also shown that the hypothesis of ovoviviparity being an adaptation to combat low water temperatures could be rejected for C. bifrons from Windermere.  相似文献

SYNOPSIS. Trypanosoma conorhini has been isolated from infected Triatoma rubrofasciata in Singapore. Parasites derived from the triatomid gut were inoculated into NNN medium and some cultures maintained at 25° to 27°C while others were kept at 37°. In cultures kept at lower temperatures crithidial forms predominated; however, at 37° blood-stream trypanosome forms developed and survived for 10 days or more.  相似文献

In order to clarify the source of reovirus pollution in river water, comparative surveys have been carried out between reovirus isolates from river water and those from sewage, human or animal, by making use of the analysis of genomic RNA-migration pattern of reovirus in polyacrylamide gel electrophoresis (electropherotype). The strains of reovirus serotype 1 and 2 isolated from river water were classified into 3 and 9 electropherotypes, respectively, and 8 out of these 12 types were also found among strains isolated from sewage or human. When the monthly distribution of the river isolates classified by electropherotypes was compared with that of the sewage isolates, there were cases in which strains of the same electropherotype were simultaneously isolated from both sources. The electropherotypes of 3 isolates from pig and field rodents were different from those of the other isolates. The electropherotype of an oyster isolate coincided with that of some of the isolates from humans and river water. These results indicate that the major sources of reoviruses polluting river water may be the human excretion.  相似文献

1. An investigation of the influence of previous thermal and nutritional experience on body temperatures and metabolic rate has been carried out with growing piglets. Littermates were kept, from shortly after birth, at either 10 or 35 degrees C and fed either a high (H) or a low (L) energy intake. At 8 weeks of age the animals were exposed to a series of environmental temperatures of 10, 20, 27 and 35 degrees C for 1.5 hr and their rates of oxygen consumption were determined over the last 45 min. At the end of the session body temperatures were measured. 2. Rectal temperatures measured 24 hr after the start of the last meal were higher at each test temperature in piglets which had been living at 35 degrees C than in those at 10 degrees C. Also, rectal temperatures were higher in those on the H intake for animals which had been living in either the hot or the cold environment. 3. Skin temperature on the back was similar in all groups at any given test temperature although there was a tendency for those on an H intake to have the higher temperatures. Skin temperatures of the legs and ears were higher in the 10H and 10L groups than in the 35H or 35L groups at all the test environmental temperatures; energy intake had little effect. 4. Metabolic rate was greater for the animals on the H than the L intake, for those which had been living at either 10 or 35 degrees C at all the test environmental temperatures. The analysis did not reveal any significant difference related to the overall effect of living temperature, which was independent of energy intake. 5. At thermal neutrality (27 degrees C) there was a significant interaction, between energy intake and normal living temperature, on metabolic rate. Living temperature was found to modify the effect of intake: the difference between the two intakes was greater in those from the cold environment than from the hot.  相似文献

Decontamination of drinking water by direct heating in solar panels   总被引:2,自引:2,他引:0  
A device was developed for direct heating of water by solar radiation in a flow-through system of copper pipes. An adjustable thermostat valve prevents water below the chosen temperature from being withdrawn. The results show that it is possible to eliminate coliform and thermotolerant coliform bacteria from naturally contaminated river water by heating to temperatures of 65 °C or above. Artificial additions of Salmonella typhimurium , Streptococcus faecalis and Escherichia coli to contaminated river water were also inactivated after heating to 65 °C and above. The total viable count could be reduced by a factor of 1000. The heat-resistant bacteria isolated from the Mlalakuva River (Tanzania) were spore-forming bacteria which exhibited greater heat resistance than commonly used test bacteria originating from countries with colder climates. To provide a good safety margin it is recommended that an outlet water temperature of 75 °C be used. At that temperature the daily production was about 50 l of decontaminated water per m2 of solar panel, an amount that could be doubled by using a heat exchanger to recycle the heat.  相似文献

We examined seasonal changes in freeze tolerance and the susceptibility of larvae of the gall fly, Eurosta solidaginis to inoculative freezing within the goldenrod gall (Solidago sp.). In late September, when the water content of the galls was high (approximately 55%), more than half of the larvae froze within their galls when held at -2.5 degrees C for 24 h, and nearly all larvae froze at -4 or -6 degrees C. At this time, most larvae survived freezing at > or = -4 degrees C. By October plants had senesced, and their water content had decreased to 33%. Correspondingly, the number of larvae that froze by inoculation at -4 and -6 degrees C also decreased, however the proportion of larvae that survived freezing increased markedly. Gall water content reached its lowest value (10%) in November, when few larvae froze during exposure to subzero temperatures > or = -6 degrees C. In winter, rain and melting snow transiently increased gall water content to values as high as 64% causing many larvae to freeze when exposed to temperatures as high as -4 degrees C. However, in the absence of precipitation, gall tissues dried and, as before, larvae were not likely to freeze by inoculation. Consequently, in nature larvae freeze earlier in the autumn and/or at higher temperatures than would be predicted based on the temperature of crystallization (T(c)) of isolated larvae. However, even in early September when environmental temperatures are relatively high, larvae exhibited limited levels of freezing tolerance sufficient to protect them if they did freeze.  相似文献

Batch microcosms containing various water types (de-ionized and river water with or without sediment), incubated at a range of temperatures (5-37 degrees C), were used to facilitate a comparative evaluation of the significance of such variables and their interactions upon the collective and individual survival of four species of thermophilic Campylobacter. All variables significantly influenced (P < = 0.031) population decay rates. Minimal decay for the group was identified at low temperatures (5 degrees C) in river water, i.e. nutrient-containing microcosms. Collective decay rates within river water microcosms were significantly decreased (P = 0.03) from those observed in de-ionized water, particularly at environmental temperatures (5 and 15 degrees C). However, the increased nutrient levels observed in sediment-containing microcosms did not significantly (P = 0.41) reduce population decay rates. Overall, Camp. jejuni populations demonstrated the most resilience to the environmental stressors evaluated, with the exception of 15 degrees C where Camp. lari was the most persistent. Campylobacter coli and Camp. upsaliensis demonstrated comparable survival characteristics but were less resilient than Camp. jejuni and Camp. lari. These observations identify the suitability of water systems as a reservoir and medium for Campylobacter infection, and potentially identifies Camp. jejuni and Camp. lari as the main protagonists of water-mediated campylobacteriosis.  相似文献

A strain of Legionella pneumophila serogroup 1 isolated from the environment had a decimal reduction time in water at 50 degrees C (D50) of 111 min, a D54 of 27 min and a D58 of 6 min. There was little loss of viability at 46 degrees C. Other environmental organisms, a Pseudomonas sp., a Micrococcus sp. and a coliform survived less well at these temperatures. A species of Sarcina had a survival time greater than the L. pneumophila at all the temperatures tested. Other strains of legionellas were tested at 50 degrees C and decimal reduction times calculated. These ranged from 80 min for another strain of L. pneumophia serogroup 1 to 216 min for L. bozemannii . Legionella micdadei did not survive well at 50 degrees C.  相似文献

Bacteriol. 84:1173-1180. 1962.-The ld(50) dose for mice of Salmonella typhimurium, strain RIA, is 4.1 x 10(5) for animals individually housed without bedding and maintained at 25 C. It is 3.8 x 10(3) for animals similarly housed but kept at 5 C. An intravenous injection of 0.1 ml of Proferrin 2 hr prior to infection with RIA lowers the ld(50) to 4.9 x 10(3) and to 4.0 x 10(1) for mice kept, respectively, at 25 and at 5 C. Low environmental temperature and "blockade" of the reticuloendothelial system (RES) lower the resistance of mice to about the same degree, but low temperature and RES impairment together lower resistance as if each were acting independently. No effect of cold could be detected in mice infected with the highly virulent SR-11 strain of S. typhimurium, since all animals died after infection with only a few cells. Mice that were natural carriers of salmonellae, as judged by fecal discharge, were highly resistant to challenge and responded to cold in a manner similar to normal mice infected with RIA. Strain RIA could be isolated from the tissues of infected animals with greater frequency and persisted longer in mice maintained at 5 C than those at 25 C. Staphylococci were isolated from livers of animals that survived salmonella infection for 14 days at 5 C, and the incidence of staphylococci was proportional to the number of salmonellae injected. At 25 C, only a small percentage of mice had staphylococci in tissues and these occurred independent of the infectious dose of salmonellae. These observations were made on normal mice infected with RIA and on carrier mice infected with SR-11.  相似文献

This paper reports the first year results of field experiments to determine the survival times of pathogens in livestock manures during storage and following land application, using viable count methods. E. coli O157, Salmonella and Campylobacter survived in stored slurries and dirty water for up to three months, with Listeria surviving for up to three months. In contrast, all these pathogens survived for less than one month in solid manure heaps where temperatures greater than 55 degrees C were obtained. Following manure spreading to land, E. coli O157, Salmonella and Campylobacter generally survived in the soil for up to one month after application to both the sandy arable and clay loam grassland soils, whereas Listeria commonly survived for more than one month. These data are being used to develop guidelines on the management of manures to minimize the risks of pathogen transfer from animal manures to the human food chain.  相似文献

Erwinia chrysanthemi was isolated over three summers by filtration, with or without enrichment, of water samples from the headwaters of the Murray and Murrum-bidgee Rivers and from several locations downstream. The bacterium could not be detected in streams rising adjacent to headwater catchments nor could it be detected in freshly voided faeces of native and domestic animals in the vicinity of the Mur-rumbidgee headwaters. Although it survived passage through the intestinal tract of a guinea pig and fowl, it did not persist in either animal. The bacterium survived for at least 211 days at 16C in sterile distilled water. The specific localities from which the bacterium was isolated and the lack of external sources of inoculum indicate that E. chrysanthemi is most likely a constituent of the sessile bacterial population on stream weeds and sediments and is not a contaminant of this alpine environment.  相似文献

The thermoregulatory behavior of Hemigrapsus nudus, the amphibious purple shore crab, was examined in both aquatic and aerial environments. Crabs warmed and cooled more rapidly in water than in air. Acclimation in water of 16 degrees C (summer temperatures) raised the critical thermal maximum temperature (CTMax); acclimation in water of 10 degrees C (winter temperatures) lowered the critical thermal minimum temperature (CTMin). The changes occurred in both water and air. However, these survival regimes did not reflect the thermal preferences of the animals. In water, the thermal preference of crabs acclimated to 16 degrees C was 14.6 degrees C, and they avoided water warmer than 25.5 degrees C. These values were significantly lower than those of the crabs acclimated to 10 degrees C; these animals demonstrated temperature preferences for water that was 17 degrees C, and they avoided water that was warmer than 26.9 degrees C. This temperature preference was also exhibited in air, where 10 degrees C acclimated crabs exited from under rocks at a temperature that was 3.2 degrees C higher than that at which the 16 degrees C acclimated animals responded. This behavioral pattern was possibly due to a decreased thermal tolerance of 16 degrees C acclimated crabs, related with the molting process. H. nudus was better able to survive prolonged exposure to cold temperatures than to warm temperatures, and there was a trend towards lower exit temperatures with the lower acclimation (10 degrees C) temperature. Using a complex series of behaviors, the crabs were able to precisely control body temperature independent of the medium, by shuttling between air and water. The time spent in either air or water was influenced more strongly by the temperature than by the medium. In the field, this species may experience ranges in temperatures of up to 20 degrees C; however, it is able to utilize thermal microhabitats underneath rocks to maintain its body temperature within fairly narrow limits.  相似文献

The nutritional versatility and growth kinetics of Aeromonas hydrophila were studied to determine the nature and the growth-promoting properties of organic compounds which may serve as substrates for the growth of this organism in drinking water during treatment and distribution. As an initial screening, a total of 69 different organic compounds were tested at a concentration of 2.5 g/liter as growth substrates for 10 A. hydrophila strains. Of these strains, strain M800 attained the highest maximum colony counts in various types of drinking water and river water and was therefore used in further measurements of growth at low substrate concentrations. A mixture of 21 amino acids and a mixture of 10 long-chain fatty acids, when added to drinking water, promoted growth of strain M800 at individual compound concentrations as low as 0.1 microgram of C per liter. Mixtures of 18 carbohydrates and 18 carboxylic acids clearly enhanced growth of the organism at individual compound concentrations above 1 microgram of C per liter. Growth measurements with 63 individual substrates at a concentration of 10 micrograms of C per liter gave growth rates of greater than or equal to 0.1/h with two amino acids, nine carbohydrates, and six long-chain fatty acids. Ks values were determined for arginine (less than or equal to 0.3 micrograms of C per liter), glucose (15.9 micrograms of C per liter), acetate (11.1 micrograms of C per liter), and oleate (2.1 micrograms of C per liter). The data obtained indicate that biomass components, such as amino acids and long-chain fatty acids, can promote multiplication of aeromonads in drinking water distribution systems at concentrations as low as a few micrograms per liter.  相似文献

Total plate counts were determined on boneless cooked, cubed chicken meat obtained from a commercial processor. Survival of the natural flora was determined after the meat was freeze-dehydrated and rehydrated at room temperature for 30 min and 50, 85, and 100 C for 10 min. Total counts of bacteria in the rehydrated samples were determined during storage of the meat at 4, 22, and 37 C until spoilage odor was detectable. Meat samples were inoculated with Staphylococcus aureus, then dried, rehydrated, and stored at the same temperatures. Numbers of surviving organisms in the inoculated samples were determined with use of both selective and nonselective media. Representative genera surviving the various rehydration treatments were determined. Approximately 32% of the bacteria in the meat survived during dehydration and rehydration at room temperature. Many numbers and types of vegetative bacteria also survived rehydration at 50 C. When meat was rehydrated at 85 or 100 C, the initial count was less than one per gram. The only organisms isolated from samples rehydrated at 85 or 100 C were of the genus Bacillus. S. aureus in inoculated samples survived dehydration and rehydration at 60 C. Storage of all rehydrated samples at 4 C gave a good shelf life (18 or more days). The study indicates that freeze-dehydrated meat should be produced with adequate microbiological control and that such meat should be rehydrated in very hot water.  相似文献

Female CD-1 mice were injected with an LD50 dose of Streptococcus pneumoniae and then exposed to 2.45 GHz (CW) microwave radiation at an incident power density of 10 mW/cm2 (SAR = 6.8 W/kg), 4 h/d for 5 d at ambient temperatures of 19 °C, 22 °C, 25 °C, 28 °C, 31 °C, 34 °C, 37 °C and 40 °C. Four groups of 25 animals were exposed at each temperature with an equal number of animals concurrently sham-exposed. Survival was observed for a 10-d period after infection. Survival of the sham-exposed animals increased as ambient temperature increased from 19 °C–34 °C. At ambient temperatures at or above 37 °C the heat induced in the body exceeded the thermoregulatory capacity of the animals and deaths from hyperthermia occurred. Survival of the microwave-exposed animals was significantly greater than the shams (~20%) at each ambient temperature below 34 °C. Based on an analysis of the data it appears that the hyperthermia induced by microwave exposure may be more effective in increasing survival in infected mice than hyperthermia produced by conventional methods (ie, high ambient temperature). Microwave radiation may be beneficial to infected animals at low and moderate ambient temperatures, but it is detrimental when combined with high ambient temperatures.  相似文献

AIMS: To determine whether diurnal and seasonal variations in campylobacters in surface waters result from the effects of temperature and u.v. radiation, and whether natural populations of Campylobacter lari and urease-positive thermophilic campylobacters (UPTC) from birds survive better in surface waters than Camp. jejuni from sewage. METHODS AND RESULTS: Natural populations of Camp. lari and UPTC in sea water, and Camp. jejuni in river water, were exposed to artificial sunlight (equivalent to a sunny day in June). Both populations became non-culturable within 30 min, with T90s of 15 min and 25 min, respectively. Cultures of Camp. jejuni became non-culturable within 40 min and those of Camp. coli, Camp. lari and UPTC, within 60 min. In darkness, survival was temperature-dependent. Natural populations took 12 h at 37 degrees C and 5 days at 4 degrees C to become non-culturable in sea water, and slightly less in river water. Cultures of Camp. lari and UPTCs survived for significantly longer than Camp. jejuni and Camp. coli. Loss of culturability for all isolates was most rapid at 37 degrees C and slowest at 4 degrees C. Newly isolated strains from sea water and river water behaved in an almost identical manner to NCTC strains. CONCLUSION: Campylobacter lari and UPTCs survive for longer in surface waters than Camp. jejuni and Camp. coli, particularly in the dark. Low Campylobacter numbers in coastal waters in the summer, especially in the afternoon, are due to the combined effects of higher temperatures and higher levels of u.v. radiation. SIGNIFICANCE AND IMPACT OF THE STUDY: Campylobacter lari and UPTCs from birds predominate in bathing waters in Morecambe Bay because they are better able to survive; they also originate from closer to the shore than Camp. jejuni and Camp. coli in sewage effluent, which survive poorly and die before the incoming tide reaches the shore. The predominance of Camp. jejuni in river water results from its dominance of the inputs and not from its ability to survive.  相似文献

Supercooling point (SCP) values and cold-hardiness were measured in individual ontogenetic stages of Delia radicum (Diptera:Anthomyiidae) in various physiological states (winter diapause, summer quiescence, and normal development). Winter diapause-destined mature third-instar larvae had a lower SCP (-9.9 degrees C) than their nondiapause counterparts (-5.2 degrees C), and more of them survived exposure to -10 degrees C for 5 h to pupariation and adult emergence. Values of SCPs were equal in both diapause and nondiapause states of prepupal and pupal stages. The lowest SCP (ca. -20 degrees C) was found in the stage of phanerocephalic pupa (PCP) regardless of the physiological state. The cold-hardiness of PCP corresponded with a low SCP value only in diapausing pupae stored for 80 days at 3 degrees C and in pupae which had terminated their diapause and whose further development was inhibited by storage at low temperatures (3 degrees C). Such pupae survived exposure to temperatures close to their SCP (14 days at -17 degrees C). However, this high cold-hardiness was only acquired after some time and/or exposure to 3 degrees C, as the PCP at the beginning of diapause showed significantly impaired cold-hardiness despite the fact that their SCP was low. The cold-hardiness of nondiapausing PCP did not correspond at all to that of low SCP, as no pupa survived the exposure to -17 degrees C for 1 day; survival rates at temperatures of -13.5 and -10 degrees C were also remarkably lower than those in diapausing pupae. Cold-hardiness in D. radicum was closely connected with the diapause syndrome but the changes in SCP value corresponded rather with the ontogeny of this insect.
