Desert hedgehog (Dhh) is a cell-signaling molecule that was first discovered in DROSOPHILA: A unique testicular phenotype has been described in neonatal and adult Dhh-null animals that includes anastomotic seminiferous tubules, pertitubular cell abnormalities, and absence of adult-type Leydig cells. In the present study, we addressed the developmental basis for the abnormalities previously described for the adult Dhh-null phenotype. The source of Dhh is the Sertoli cell, and receptors are localized on peritubular cells and possibly Leydig cells. The development of testes from Dhh-null mouse embryos was studied using light and electron microscopy at 11.5, 12.5, 13.5, and 16.5 days postcoitum (dpc) and was compared with that in control Dhh heterozygous and wild-type embryos. Dhh-null and control testes were generally similar during the period of early cord formation (11.5-12.5 dpc). By 13.5 dpc, the basal lamina delimiting the cords was lacking in some regions and disorganized in Dhh-null testes, and occasional germ cells were seen outside cords. At 16.5 dpc, these defects were more prominent and cord organization was less well defined than in controls. In addition, there were numerous extracordal germ cells, some of which were partially enclosed by a somatic cell of unknown identity. Numerous fibroblast-like cells, apparently secreting collagen and basal lamina, characterized the interstitium of the Dhh-null testis. These defects likely stem from abnormal peritubular stimulation due to the lack of Dhh, leading to the abnormalities seen in the developmental stages studied here and in the adult testis.  相似文献

Elevated water temperature has been found to cause gonadal degeneration in fish, including the partial or complete loss of germinal elements, and might impair fertility and reproductive performance. Germ cell-deficient and even completely sterile pejerrey Odontesthes bonariensis have been found in two lagoons in Argentina, and exposure to warm water is one of the possible causes of these abnormalities. This experiment was conducted (a) to compare the histological characteristics of the abnormal gonads from wild pejerrey with those of animals exposed to heat in the laboratory and (b) to examine whether short-term pulses of heat similar to diurnal temperature variations in natural environments during summer can trigger gonadal cell degeneration in adult pejerrey. Wild fish with gonadal abnormalities were obtained from the San Miguel del Monte and Lacombe Lagoons (Buenos Aires Province, Argentina). Laboratory specimens were obtained by exposure of adult pejerrey to five thermal regimes (constant 24 degrees and 29 degrees C and 12-h cycles of 27 degrees -31 degrees, 28 degrees -30 degrees, and 28 degrees -31 degrees C) for up to 16 wk. Germ cell-deficient specimens for histological comparison with wild animals were also obtained by exposing larvae and juveniles for 8-12 wk to 29 degrees C and rearing until they became adults. The histological characteristics of the abnormal gonads of wild pejerrey closely resembled those of fish partially or completely sterilized by high water temperature in the laboratory. The results indicate that fluctuating (high) thermal regimes triggered germ cell disappearance in a manner comparable to a constant temperature of 29 degrees C. These results support the notion that high temperature during unusually warm summers might trigger germ cell degeneration and could be the cause of the observed gonadal abnormalities in wild pejerrey.  相似文献

Mice (CBAxC57BL) F of both sexes were exposed within the 10 km zone of Chernobyl nuclear power station. Genetic damage of phone chronic effect of increased radiation in exposed adult mice and in the course of embryogenesis was studied. The total absorbed radiation doses in testes varied from 1.85 to 0.42 Gy in embryos and from 3.4 to 2.7 Gy in adult males. Increase of dominant lethal mutations (DLM) and abnormal sperm heads (ASH) was only observed right after the end of exposure of adult males. The yield of reciprocal translocations (RT) in these males was relatively low. Among the males exposed at the stage of early embryogenesis, 4 heterozygotes for RT were revealed. Pregnancy exposure to di(n-butyl) phthalate (DBP) in rats induces a testicular dysgenesislike syndrome (TDS) in male offspring. Earlier studies suggested altered Sertoli cell development/maturation may result, especially in testes that become cryptorchid. This study quantitatively assessed Sertoli cell numerical and functional development in DBP-exposed rats and compared (unilaterally) cryptorchid and scrotal testes. Pregnant rats were gavaged with 500 mg/kg/day DBP or corn oil from embryonic (E) Days 13.5 to 21.5. Male offspring were sampled on E21.5 or Postnatal Day 6, 10, 15, 25, or 90. Sertoli cell number in DBP-exposed males was reduced by approximately 50% at E21.5 but recovered to normal by Days 25-90, accompanied by significant changes in plasma inhibin B and testosterone levels. Sertoli cell maturational development in DBP-exposed males, assessed using five protein markers (anti-müllerian hormone, cytokeratin, androgen receptor, CDKN1B, and Nestin), was largely normal, with some evidence of delayed maturation. However, in adulthood, Sertoli cells (SC) in areas lacking germ cells (Sertoli cell-only [SCO] tubules) often exhibited immature features, especially in cryptorchid testes. Sertoli cells in DBP-exposed animals supported fewer germ cells during puberty, but this normalized in scrotal testes by adulthood. Scrotal and especially cryptorchid testes from DBP-exposed animals exhibited abnormalities (SCO tubules, focal dysgenetic areas) at all postnatal ages. Cryptorchid testes from DBP-exposed animals exhibited more Sertoli cell abnormalities at Day 25 compared with scrotal testes, perhaps indicating more severe underlying Sertoli cell malfunction in these testes. Our findings support the concept of altered Sertoli cell development in TDS, especially in cryptorchid testes, but show that maturational defects in Sertoli cells in adulthood most commonly reflect secondary dedifferentiation in absence of germ cells.  相似文献

The values of gonad index calculated by the allometric relationships of dry gonad weight over dry body weight of the black scallop, Chlamys varia (L.), gave peak values for May and September during 1976 and 1977. Histological studies on gonads from natural populations and periodic biopsies of gonads from marked animals in experimental cages showed that, in addition to intermittent spawnings between May and September, two peak spawning periods were noteworthy. These were a partial spawning in May–June and a complete spawning in September–October. The energy spent as gametes for these two spawnings has been calculated for individual males and females in each age group of the population and the cost of spawning by the whole population has been estimated. The population spent more energy for spawning in 1977 (144.49 kcal/100 m2) than in 1976 (83.70 kcal/100 m2) and the reproductive effort of an average adult was 0.04 in 1976 and 0.10 in 1977.  相似文献

The requirement for a functional retinoblastoma gene, Rb-1, in murine development around days 12-15 of gestation precludes monitoring the effect of loss of Rb-1 function on later stages of development and on tumorigenesis in adult mice. Here we describe the developmental rescue of embryonic stem cells carrying two inactive Rb-1 alleles in chimeric mice. Rb-1- cells contributed substantially to most tissues in adult chimeras, including blood, liver and central nervous system, which were severely affected in pure Rb-1- embryos. The adult chimeric erythroid compartment appeared completely normal, but an increased number of nucleated red cells was observed during fetal liver erythropoiesis in highly chimeric embryos. No ostensive abnormalities were seen in the developing and adult CNS. However, the developing retina of chimeric Rb-1- embryos showed ectopic mitoses and substantial cell degeneration, while the contribution of Rb-1- cells to the adult retina was much reduced. Moreover, the formation of lens fibre cells was severely disturbed. No retinoblastomas developed in any of these mice. Instead, nearly all animals died of pituitary gland tumours which were exclusively derived from Rb-1- cells.  相似文献

Expression of the diphtheria toxin A-chain gene was directed to the male germ line by fusion to 1 kilobase of the 5'-flanking DNA of the rat histone H1t gene. Two independent lines of mice were established that expressed the toxic transgene. Female carriers were fertile; males were sterile although otherwise apparently normal. Adult transgenic males had very small testes that were virtually devoid of germ cells. A developmental study showed that germ cells survived until late fetal life but that testes of 3-day-old transgenic mice were severely depleted of prospermatogonia. During postnatal development of transgenic animals, remaining germ cells progressed to the pachytene stage of meiosis in 10% to 30% of tubular cross sections but degenerated before the completion of meiosis. By 3 mo of age the residual germ cells had almost completely disappeared. These transgenic lines demonstrate the complete tissue specificity of the H1t promoter and reveal a period of its activity just prior to formation of the definitive adult spermatogonial stem cell population. Whereas full expression of H1t occurs only in mid to late pachytene spermatocytes, one or more of the factors that impart tissue specificity to its expression must be transiently activated in the neonatal germ line. This report discusses the possibility that this genetic technique for eliminating germ cells may have practical application in making recipients for spermatogonial stem cell transplantation.  相似文献

Purkinje cell degeneration (pcd) is a neurological mutation in the mouse that causes male sterility, but not female sterility. In order to assess the effects of this mutation on spermiogenesis, the structure of the testis and of epididymal spermatozoa was examined by transmission and scanning electron microscopy. In the mutant males, the sperm count was reduced, sperm were nonmotile, and 93% of the sperm were characterized by structural abnormalities of the head, the tail, or both. In the testes of mutant mice, Sertoli cell structure was normal, as were also the early stages of spermiogenesis. However, the elongating and maturing spermatids were characterized by abnormally shaped heads and tails with extraneous and ectopic outer dense fibers. These defects were common in the testes of the mutant mice and rare in the testes of the littermate control mice. It was concluded that the structural abnormalities of the pcd sperm occurred during spermiogenesis and were not due to degeneration of the sperm in the epididymis. These structural abnormalities are similar to those found in all other reported male sterile mutants of the mouse; therefore, although they are caused by the expression of the pcd gene, they are not unique to the expression of this gene.  相似文献

Chimeric animals were produced by aggregation of 8-cell-stage embryos from two strains of hamsters (LVG and Bio 1.5). Two series of experiments were performed. In the first series, embryo pairs in contact with each other were classified as aggregates even if 2 distinct embryos could still be distinguished. Of 88 aggregates transferred, 2 chimeras were obtained. Pregnancy rate was 25%, and embryo survival was 35%. In the second set of experiments, only embryo pairs that had coalesced to form a single giant blastocyst were classified as aggregates. Of 56 aggregates transferred, 6 chimeras were obtained. Pregnancy rate was 83%, and embryo survival was 30%. Of the 8 chimeras, 6 were phenotypic males, and 2 were phenotypic females. Both females were germ line chimeras. Of the 6 males, 4 reproduced normally, 1 had abnormal external genitalia but normal spermatogenesis, and 1 was sterile and had atrophic testes. Each of the fertile males transmitted only a single component, either the LVG or the Bio 1.5. Examination of the testes from the sterile chimera revealed that in excess of 80% of the seminiferous cords were devoid of germ cells. These results demonstrate that hamster chimeras can be obtained by aggregation of 8-cell-stage embryos.  相似文献

The brown hare, Lepus europaeus, has a mating season which extends from January to September. Adult males exhibit pronounced seasonal changes in the reproductive tract which are associated with changes in LH secretion. Maximum plasma levels of immunoreactive LH occur between March and June and minimal levels in the autumn non-mating period from September to December; this seasonal cycle in gonadotrophin output is reflected by the appropriate changes in the secretion of testosterone from the testes and in the activity of the accessory sex glands. Juvenile animals reach puberty only during the adult mating season, and the age of puberty thus varies with the date of birth. Males born before May reach puberty and become fertile at 3 months of age, while those born from May to July grow to a mature body size during the autumn non-mating season but puberty is delayed for several months. Since some animals experiencing delayed puberty were found to have elevated plasma levels of LH and testosterone, it is concluded that puberty is not completely suppresed by the environmental effects of the autumn, but that the developmental process is prolonged, resulting in the juveniles being synchronized with the adults in their reproductive activity.  相似文献

Homozygous Purkinje cell degeneration (pcd) mutant males exhibit abnormal sperm development. Microscopic examination of the testes from pcd(3J)-/- mice at postnatal days 12, 15, 18 and 60 revealed histological differences, in comparison to wild-type mice, which were evident by day 18. Greatly reduced numbers of spermatocytes and spermatids were found in the adult testes, and apoptotic cells were identified among the differentiating germ cells after day 15. Our immunohistological analysis using an antihuman AGTPBP1 antibody showed that AGTPBP1 was expressed in spermatogenic cells between late stage primary spermatocytes and round spermatids. A global gene expression analysis from the testes of pcd(3J)-/- mice showed that expression of cyclin B3 and de-ubiquitinating enzymes USP2 and USP9y was altered by >1.5-fold compared to the expression levels in the wild-type. Our results suggest that the pcd mutant mice have defects in spermatogenesis that begin with the pachytene spermatocyte stage and continue through subsequent stages. Thus, Agtpbp1, the gene responsible for the pcd phenotype, plays an important role in spermatogenesis and is important for survival of germ cells at spermatocytes stage onward.  相似文献

Testes of Heliothis virescens were examined in vitro for the ability to produce ecdysteroids. Production was detected in testes removed from larvae at mid and late periods in the last instar, in testes from pupae after the third day of purpal development, and in testes taken from young adult males. The testes in the isolated abdomens do not fuse during later stages of spermatogenesis as they do in intact insects. Spermatocysts in testes of larval abdomens isolated from the upper body early in the last instar developed to mature sperm despite low haemolyph and testis ecdysteroid titres. Gonadectomized animals developed and moulted normally, indicating that ecdysteroids from the testes are not essential to general development.  相似文献

Some males of a mutant strain of King-Holtzman rats exhibit an anomalous heritable defect manifested as either unilateral or bilateral ectopic testes. In the adult, these testes contain seemingly immature Sertoli and Leydig cells, seminiferous tubules greatly reduced in diameter, and exhibit arrested spermatogenesis. Thus, the affected testis is essentially sterile. An inability to produce normal amounts of testosterone and androstenedione by these gonads is probably a reflection of changes that have been effected in their Leydig cells. Thus, this study suggests that abnormal function of the Leydig and Sertoli cells and seminiferous tubule failure in these mutant animals result from the physiologically cryptorchid condition.  相似文献

Females of the cotton leafworm (Spodoptera littoralis) reared in long day conditions (LD 16:8 h) and mated to males kept throughout the whole period of development in continuous light (LL) oviposit very small numbers of mostly sterile eggs. It was found that in control males reared from the first larval instar in long day conditions there was a large accumulation of euperene sperm bundles in their testes on day 1 after imaginal moult. On day 10 of adult life the number of the sperm bundles was very small. In males kept from the first instar in continuous light there was also high number of sperm bundles on day 1 after imaginal moult but it did not decrease significantly on day 10 as was observed in controls. Transfer of different developmental stages of S. littoralis from long day conditions to continuous light resulted in a big difference in the density of eupyrene sperm bundles in their testes. In control insects reared through the whole of their development in long day conditions there was a significant decrease in the density of eupyrene sperm bundles on day 10 of adult life. By contrast, in males in continuous light, regardless of their developmental stage when transferred, there was either no change in density of sperm bundles in day 10 adults or there was a significant increase in comparison with day 1 adults. The highest density of eupyrene sperm bundles was observed in day 10 adults when they were transferred to continuous light shortly before moulting to the last instar (as day 4 larvae in the last stadium or day 1 pupae). Generally, the density of eupyrene sperm bundles on day 10 of adult development was about 2–2.5 times higher in males in continuous light then those under long day conditions. The results presented here indicate that the last larval instar and the pupa are the stages most sensitive to constant light treatment, which greatly reduces the amount of eupyrene sperm bundles released from the testes.  相似文献

In several primate taxa there is evidence that the social and physical environment can exert a significant effect on reproductive behavior and biology. In this paper we examine social and physiological factors influencing group composition and reproduction in free-ranging moustached tamarin monkeys (Saguinus mystax mystax). This species is characterized by cooperative care of the young and a breeding system that includes both polyandrous and polygyandrous matings. Body measurements collected on adult males residing in multimale groups indicate marked within-group differences in testes volume. In 12 of 17 groups examined, testes volume of at least two resident adult males differed by 21–174%. Among these males, testes volume was not correlated with either body weight or adult age class. We also examine whether factors such as time of year had an effect on reproductive condition. An analysis of body measurements of 128 adult male and 127 adult female moustached tamarins, wild-trapped and released in northeastern Peru, indicates cyclic changes in genital size. For males, mean monthly testes volume in July (712 mm3) was twice that recorded in June (351 mm3). Females exhibited a similar pattern. Although endocrine information on intra- and intersexual social effects on fertility are unavailable for S. mystax, given the high degree of social cooperation and lack of overt aggression among adult male group members, we offer the possibility that resident male moustached tamarins compete for access to the groups' lone breeding female through socially induced reproductive suppression and sperm competition. © 1996 Wiley-Liss, Inc.  相似文献

Knockout (KO) mice lacking the orphan nuclear receptor steroidogenic factor 1 (SF-1, officially designated Nr5a1) have a compound endocrine phenotype that includes adrenal and gonadal agenesis, impaired expression of pituitary gonadotropins, and structural abnormalities of the ventromedial hypothalamic nucleus. To inactivate a conditional SF-1 allele in the gonads, we targeted the expression of Cre recombinase with a knock-in allele of the anti-Müllerian hormone type 2 receptor locus. In testes, Cre was expressed in Leydig cells. The testes of adult gonad-specific SF-1 KO mice remained at the level of the bladder and were markedly hypoplastic, due at least partly to impaired spermatogenesis. Histological abnormalities of the testes were seen from early developmental stages and were associated with markedly decreased Leydig cell expression of two essential components of testosterone biosynthesis, Cyp11a and the steroidogenic acute regulatory protein. In females, the anti-Müllerian hormone type 2 receptor-Cre allele directed Cre expression to granulosa cells. Although wild-type and SF-1 KO ovaries were indistinguishable during embryogenesis and at birth, adult females were sterile and their ovaries lacked corpora lutea and contained hemorrhagic cysts resembling those in estrogen receptor alpha and aromatase KO mice. Collectively, these studies establish definitively that SF-1 expression in the gonads is essential for normal reproductive development and function.  相似文献

Skeletal and dental pathology of free-ranging mountain gorillas   总被引:1,自引:0,他引:1  
The mountain gorillas of the central Virungas have been the subject of field study for the last 30 years; however, our understanding of morbidity and mortality in these apes is limited. This paper describes pathological conditions of the skeleton and dentition of these animals and evaluates lesions in relation to behavioral and environmental data. The skeletal remains of 31 mountain gorillas from the Karisoke Research Center were examined for enamel wear, carious lesions, abscesses, periodontal disease, antemortem tooth loss, trauma, inflammation, arthritis, neoplasia, and developmental anomalies. Two infants, three juveniles, 13 adult males, and 13 adult females form the sample. Enamel wear in the permanent posterior dentition is moderate. Six periapical abscesses were seen; three are associated with antemortem tooth breakage. No carious lesions were observed. Pronounced calculus buildup and alveolar resorption are the most notable pathological conditions of the dentition and affect all adult animals. The primary affliction of the skeleton is arthritis, which affects 14 animals. Vertebral degenerative disease predominates, but there is also temporomandibular joint involvement. Fractures occur at seven locations in the postcranium. In addition, there are five cranial injuries, including a fractured sagittal crest, and a penetrating wound to the vault, which is believed to result from a bite. Also thought to result from a bite is a case of cranial osteomyelitis. The only other inflammatory responses are two cases of idiopathic periostitis and one idiopathic lytic lesion. Button osteomas affect two animals and are the only neoplastic conditions observed. Two animals are afflicted by developmental abnormalities: one animal by idiopathic vertebral fusion and the other by spinal scoliosis.  相似文献

The wild bonobos (Pan paniscus) of Wamba, Zaire have an extremely high frequency of external abnormal traits. Out of 96 animals in a sample comprising all age and sex classes from two different natural groups, 46 individuals had at least one limb defect. The frequency of abnormalities was greater in males than in females, and increased directly with age. Almost all infants and juveniles had completely normal limbs, but virtually all males and over half the females which had reached their prime were found to have one or more limb defects. These findings indicate that congenital factors contribute little to the frequency of abnormalities in this sample. Survival rate is the same in individuals with defects as in those without defects. It appears that individual social status is far more important in procuring foods than locomotor ability. Thus, the operation of such abnormalities as a selective factor in intraspecific competition is minimized in the network of their social relations.  相似文献

A recessive lethal mutation in axolotls that involves the kidneys, the Mullerian ducts and the limbs was described by Humphrey (1967). In the present experiments, we have examined the structural defects that lead to kidney malfunction and subsequent death in homozygous mutants and compared the defects with those observed in other axolotls lacking this mutant gene. The ultrastructure of the mesonephric kidney was studied in homozygous s/s short-toes axolotls with ascites and/or edema and hemorrhages (group 1a); in s/s short-toes axolotls not yet expressing kidney malfunction symptoms (group 1b); in normal siblings, either +/+ or +/s, without affected limbs but possibly heterozygous for the 's' gene (group 1c); in +/+ animals with malfunctioning kidneys that lack the short-toes gene (group 2a) and in normal +/+ animals from different gene pools (group 2b). In all of the short-toes animals that expressed pathological phenotypes, the proximal tubule showed abnormal morphology. There was no morphological evidence of kidney abnormality in the siblings having normal limbs or in the group 1b axolotls examined. However, mesonephroi from animals (group 2a) of other gene pools that had ascites exhibited a different and more pronounced pathology. On the basis of the dramatically distinct proximal tubule pathology of the sporadic floaters, we conclude that this phenotype is more likely caused by infection than to a variant of the short-toes gene.  相似文献
