The contamination of beef carcasses was studied together with the capacity of meat surfaces to retain bacteria along the processing line in the slaughter hall. The results showed that the contamination varied along the processing line, but that this pattern was essentially dependent on the contamination at the dressing station. It decreased or remained unchanged during the first 12 min and then increased, even without additional contamination. The contamination varied according to carcasses and micro-organisms studied and was not greatly affected by spray cleaning. The number of bacteria retained changed at a rate similar to that of the contaminants. The attachment was instantaneous. The results are discussed and compared with the various hypotheses about contamination and bacterial attachment processes.  相似文献

Bone and tissue allografts are widely used in transplantation. The increasing demand for safe allografts must be met, while minimizing disease transmission. We analysed the incidence and potential risk factors of allograft contamination and the effectiveness of disinfection, by reviewing 22 years of tissue bank activity and 474 donor procurements. We also compared different disinfection procedures used over the 22 years. The overall contamination rate was 10.1%. Risk factors were related to the donor or procurement method. Immediate culture at the tissue recovery site diminished the rate of false positives by reducing later sample manipulation. High-virulence allograft contamination was mainly related to donor factors, while low-virulence contamination was related to procurement methods. Analysis of donor-related risk factors showed no statistical differences for age, sex, or cause of death. An intensive care unit stay was associated with less contamination with high-virulence microbes. Procurement in a setting other than an operating theatre was associated with higher contamination rate. Team experience reduced contamination. Pelvic and tendon allografts were most frequently contaminated. Proper disinfection considerably reduced the contamination rate to 3.6%. We conclude that procurement must be performed under aseptic conditions, with short delays, and by trained personnel. Grafts should be disinfected and packed as soon as possible.  相似文献

Cross contamination of foodborne pathogens in the retail environment is a significant public health issue contributing to an increased risk for foodborne illness. Ready-to-eat (RTE) processed foods such as deli meats, cheese, and in some cases fresh produce, have been involved in foodborne disease outbreaks due to contamination with pathogens such as Listeria monocytogenes. With respect to L. monocytogenes, deli slicers are often the main source of cross contamination. The goal of this study was to use a fluorescent compound to simulate bacterial contamination and track this contamination in a retail setting. A mock deli kitchen was designed to simulate the retail environment. Deli meat was inoculated with the fluorescent compound and volunteers were recruited to complete a set of tasks similar to those expected of a food retail employee. The volunteers were instructed to slice, package, and store the meat in a deli refrigerator. The potential cross contamination was tracked in the mock retail environment by swabbing specific areas and measuring the optical density of the swabbed area with a spectrophotometer. The results indicated that the refrigerator (i.e. deli case) grip and various areas on the slicer had the highest risk for cross contamination. The results of this study may be used to develop more focused training material for retail employees. In addition, similar methodologies could also be used to track microbial contamination in food production environments (e.g. small farms), hospitals, nursing homes, cruise ships, and hotels.  相似文献

We investigated the influence of inter-electrode spacing on the degree of crosstalk contamination in surface electromyographic (sEMG) signals in the tibialis anterior (target muscle), generated by the triceps surae (crosstalk muscle), using bar and disk electrode arrays. The degree of crosstalk contamination was assessed for voluntary constant-force isometric contractions and for dynamic contractions during walking. Single-differential signals were acquired with inter-electrode spacing ranging from 5 mm to 40 mm. Additionally, double differential signals were acquired at 10 mm spacing using the bar electrode array. Crosstalk contamination at the target muscle was expressed as the ratio of the detected crosstalk signal to that of the target muscle signal. The crosstalk contamination ratio approached a mean of 50% for the 40 mm spacing for triceps surae muscle contractions at 80% MVC and tibialis anterior muscle contractions at 10% MVC. For single differential recordings, the minimum crosstalk contamination was obtained from the 10 mm spacing. The results showed no significant differences between the bar and disk electrode arrays. During walking, the crosstalk contamination on the tibialis anterior muscle reached levels of 23% for a commonly used 22 mm spacing single-differential disk sensor, 17% for a 10 mm spacing single-differential bar sensor, and 8% for a 10 mm double-differential bar sensor. For both studies the effect of electrode spacing on crosstalk contamination was statistically significant. Crosstalk contamination and inter-electrode spacing should therefore be a serious concern in gait studies when the sEMG signal is collected with single differential sensors. The contamination can distort the target muscle signal and mislead the interpretation of its activation timing and force magnitude.  相似文献

Lethally irradiated conventional mice were killed at daily intervals after irradiation. A high correlation was found between the occurrence of bacterial over-growth in the digestive tract and invasion into the regional lymph nodes the spleen and the blood. By oral contamination of mice with quite high doses of either an exogenous or an endogenous biotype of an Enterobacteriaceae species at day 4 after irradiation, it was attempted to induce such condition of intestinal over-growth. In all animals such an abnormal colonization of the contaminant was indeed achieved in this way and the mice died from an infection caused by the biotype used for the contamination.The interval between contamination and death was different when different biotypes were used for contamination and occurred sooner after irradiation when an endogenous biotype was used for (re)contamination. It is concluded that oral contamination with exogenous as well as with endogenous potentially pathogenic bacteria during a period of severely decreased resistance to infections must be avoided. The colonization resistance of the digestive tract of these animals should be maintained as high as possible.  相似文献

A survey was made over 5 years of the incidence of contamination by Phoma foveata and Polyscytalum pustulans on 3-year-old virus-tested stem cutting (VTSC) stocks assessed on three occasions during the storage season, and on their daughter tubers at five farms. The contamination occurring on plants grown in small plots from stem-cutting/microplant tubers was also assessed at the farms.
Fungicide treatment soon after harvest reduced the incidence of both pathogens, especially P. pustulans . Tuber contamination was similar for samples collected after fungicide treatment and before planting. The incidence of contamination on daughter tubers, relative to that on the untreated mother tubers, was less with P. foveata but was similar or greater with P. pustulans . Contamination by both pathogens varied between farms and was reflected in the degree of contamination occurring on daughter tubers from stem-cutting/microplant tubers.
The incidence of P. foveata on stems, soil and tubers of seed stocks classified at Foundation Stock grade from various regions of Scotland was assessed over 2 years. The incidence of tuber contamination was correlated ( P < 0.05) with the amount of tuber-borne soil contamination for all comparisons, but was correlated with stem inoculum in only 7 out of 13 comparisons. The largest amounts of stem inoculum of P. foveata were found in stocks in Aberdeenshire.
These results are discussed in relation to the spread of both pathogens to healthy VTSC stocks.  相似文献

AIMS: A comparison of Enterobacteriaceae, coliform and Escherichia coli counts in chicken carcasses with and without visible faecal contamination was conducted to evaluate the role of contamination as a vehicle for generic E. coli, coliform and other enterobacteria contaminating broiler chicken carcasses when processed under routine commercial operations. METHODS AND RESULTS: Samples were removed from the processing line immediately after evisceration, inside-outside shower and chilling for microbiological analysis. After evisceration, mean counts were significantly different only for E. coli (P < or = 0.05) in chicken carcasses with and without visible faecal contamination. While the spray wash practice was not efficient enough for complete removal of the visible contamination from carcasses, leading to microbiological reduction percentages lower than expected, 25 ppm chlorinated water chilling did reduce the contamination level considerably in all samples. CONCLUSIONS: Carcasses with and without visible faecal contamination harboured E. coli and other potentially hazardous enterobacteria. E. coli was the predominant strain isolated in all samples, Enterobacter cloacae being next most frequent. SIGNIFICANCE AND IMPACT OF THE STUDY: The zero tolerance of visible faecal contamination requirement alone is not sufficient to assure safety and to improve the microbial quality of carcasses.  相似文献

To examine the level of contamination of Eshghabad region, Neyshabur, the concentration of heavy metals in soil was measured. For this purpose, 37 samples were taken from surface soil and the total concentration of heavy elements was measured by inductively coupled plasma (ICP). The mean concentration of Pb, Ni, Cu, Zn, and Fe was obtained to be 195, 87.3, 22.8, 274.8, and 2.5%, respectively. Using five valid indicators, the intensity of metal contamination in soil was calculated and compared. Furthermore, using statistical analyses, the relationships between the elements, their origins, and the spatial distribution of metals across various stations were investigated. The results indicated that the mean value of all of the studied metals (except for iron and copper) is greater than the mean concentration of metals in the Earth's crust. The indicators showed very high contamination for lead, while low to medium contamination for other elements of interest in the farming soils of the region. Statistical analyses indicated that there is a relatively similar contamination intensity across all of the studied stations. Considering the quality standards, the soils of this region are threatened by contamination of lead.  相似文献

通过易地栽种实验,研究了经历不同重金属污染时期的玉米种群在正常条件下的平均相对生长率、不同时间的开花百分率及其整株重、株高、穗重、50 粒种子重、有效穗长和无效穗长等数量性状在不同玉米种群间的差异.结果表明,经历较长时期重金属污染适应的玉米种群在正常条件下栽种,具有较低的平均相对生长率,生活史缩短,植株矮化,产量降低.可以看出,玉米对重金属污染的适应导致了生态分化和品种退化,并讨论了玉米对重金属污染适应的耐性代价.  相似文献

Studies using archived scales and otoliths to examine ancient fish populations have become increasingly common, despite many methodological challenges in ancient DNA research. Here, we describe a case of DNA contamination in both modern and historical samples of Pacific herring (Clupea pallasii), where the source of the contamination is likely from milt spillage during collection. We describe a series of experiments to remove contamination using pre-extraction wash treatments. Though contamination was easily removed from contemporary fin clippings, no method was successful at removing contamination from historical scales. We discuss the implications of our findings to the genetic analysis of archived samples.  相似文献

The study, carried out in two regions of the USSR and aimed at estimation of the contamination of products supplied by industrial poultry complexes (IPC), revealed that the contamination of these products was closely related to the Campylobacter contamination of the personnel of IPC. The causes of high Campylobacter contamination of the products of IPC at all technological stages of their production are described. The species, serovars and biovars of Campylobacter strains isolated from different sources were determined, which made it possible to carry out the specific and intraspecific differentiation of these strains.  相似文献

Sensitive molecular methods, such as the PCR, can detect low-level contamination, and careful technique is required to reduce the impact of contaminants. Yet, some assays that are designed to detect high copy-number target sequences appear to be impossible to perform without contamination, and frequently, personnel or laboratory environment are held responsible as the source. This complicates diagnostic and research analysis when using molecular methods. To investigate the air specifically as a source of contamination, which might occur during PCR setup, we exposed tubes of water to the air of a laboratory and clean hood for up to 24 h. To increase the chances of contamination, we also investigated a busy open-plan office in the same way. All of the experiments showed the presence of human and rodent DNA contamination. However, there was no accumulation of the contamination in any of the environments investigated, suggesting that the air was not the source of contamination. Even the air from a busy open-plan office was a poor source of contamination for all of the DNA sequences investigated (human, bacterial, fungal, and rodent). This demonstrates that the personnel and immediate laboratory environment are not necessarily to blame for the observed contamination.  相似文献

We examined the effects of urban-derived fluvial contamination on aquatic bryophytes with the aim of improving use of these organisms for active biomonitoring of this type of contamination. Samples of the aquatic moss Fontinalis antipyretica were transplanted to five sites of a river affected by different levels of urban contamination. The aquatic contamination was characterized by a series of physical and chemical parameters, which were then related to the physiological responses determined in transplants of the bryophyte. Comparison of the data revealed the physiological parameters that best differentiated between levels of contamination. The active biomonitoring technique proved to be a clear and rapid method of determining the existence of a contamination gradient on the basis of the degree of pigmentary and photosynthetic stress in the moss transplants. Of the parameters considered, the concentration of phaeopigments and the net photosynthetic rate provided the clearest responses.  相似文献

AIMS: The mechanisms of Salmonella contamination in an oilmeal plant were investigated and the basic data were collected in order to achieve control of Salmonella in oilmeal. METHODS AND RESULTS: Salmonella was detected in all contamination vectors and environmental factors investigated, namely: operators, processing floor, dust in the air and rodents. In particular, high concentrations of Salmonella were detected on the processing floor of the manufacturing area, which has high oil content. Steam was the most effective disinfection method used for the processing floor, as the effects of heat sterilization and disinfection may work in tandem. In addition, restricting the movement of operators of the production chain remarkably reduced Salmonella contamination, even in areas of otherwise high contamination. CONCLUSIONS: Within the oilmeal plant, high Salmonella contamination rates for the processing floor represent the greatest risk of contamination of oilmeal via operators, dust in the air and rodents. Therefore, control of the processing floor is the most important means for reducing the oilmeal contamination rate. SIGNIFICANCE AND IMPACT OF THE STUDY: Specific Salmonella control methods for oilmeal plants have been established.  相似文献

BackgroundSplash basins are used in orthopaedic surgery cases to wash and hold instrumentsintraoperatively. This systematic review aims to summarize information on contamination of splash basins intraoperatively.MethodsA systematic review was conducted using the following search terms: “splash basin” or “splash bucket.” Two authors independently reviewed the literature. Studies were included if they reported on intraoperative splash basin contamination rates. Studies were excluded if they were not relevant to orthopaedic surgery, non-English articles, or repeat studies yielded by different online databases.ResultsThere were seven studies included in this review. The median contamination rate of sterile water or physiologic saline splash basins was 23.9% [range: 2%-74%]. The addition of surgical antiseptics to sterile water splash basins was associated with 0% contamination rates in two studies. The most frequent splash basin contaminants identified in bacterial culture were coagulase negative staphylococcus (50%) and staphylococcus aureus (10%).ConclusionThe splash basin appears to be a frequent source of contamination in the operating room. Many studies suggest abandoning splash basin use altogether, although the efficacy of alternative methods such as cleaning instruments with lap pads in avoiding contamination of the sterile field has not been studied. Further investigation into surgical teams’ use of the splash basin and the contents of the splash basin as they relate to contamination rates may help advance our understanding of optimal use of this surgical tool. The bacterial diversity of a chronically oil-polluted retention basin sediment located in the Berre lagoon (Etang-de-Berre, France) was investigated. This study combines chemical and molecular approaches in order to define how the in situ petroleum hydrocarbon contamination level affects the bacterial community structure of a subsurface sediment. Hydrocarbon content analysis clearly revealed a gradient of hydrocarbon contamination in both the water and the sediment following the basin periphery from the pollution input to the lagoon water. The nC17 and pristane concentrations suggested alkane biodegradation in the sediments. These results, combined with those of terminal-restriction fragment length polymorphism analysis of the 16S rRNA genes, indicated that bacterial community structure was obviously associated with the gradient of oil contamination. The analysis of bacterial community composition revealed dominance of bacteria related to the Proteobacteria phylum (Gamma-, Delta-, Alpha-, Epsilon- and Betaproteobacteria), Bacteroidetes and Verrucomicrobium groups and Spirochaetes, Actinobacteria and Cyanobacteria phyla. The adaptation of the bacterial community to oil contamination was not characterized by dominance of known oil-degrading bacteria, because a predominance of populations associated to the sulphur cycle was observed. The input station presented particular bacterial community composition associated with a low oil concentration in the sediment, indicating the adaptation of this community to the oil contamination.  相似文献

The purpose of the study was to investigate the association between the hygienic practice of slaughterhouse workers and the microbiological contamination level of carcasses. In 5 Finnish slaughterhouses the workers’ hygienic practice was observed and the carcasses were sampled by the swabbing method. The overall means (log10 cfu cm−2) of the aerobic plate count in pork and beef carcasses were 3.23 and 2.65, respectively. Hygienic practice was found to be associated with the carcass contamination level, especially the disinfection frequency. In those slaughterhouses, where the disinfection frequency was low, the contamination level of carcasses was high. Designing slaughtering lines so as to make hygienic working possible is evidently very important. However, the enforcement of hygienic practice, such as the regular disinfection of working tools, is also important in reducing the microbiological contamination of carcasses.  相似文献

A mycological survey was carried out on durum wheat (Triticum durum) samples from the main production area of Argentina. The isolation frequency and relative density of species of dematiaceous fungi, and genus Fusarium were calculated. Alternaria alternata and Fusarium graminearum were the predominant fungal species. An analysis of deoxynivalenol (DON) natural contamination was also performed on a limited number of samples (60). DON contamination levels in positive samples ranged from 26 to 6400 μg/kg. The non-parametric techniques applied showed that there is a positive relationship between DON contamination and F. graminearum relative densities and a negative relationship between DON contamination and A. alternata relative densities. Molecular typing was applied to an in-plant lactic acid bacterium (LAB) contamination analysis of a vacuum-packaged sliced cooked whole-meat product. A total of 982 LAB isolates from the raw mass, product, and the environment at different production stages were screened by restriction endonuclease (EcoRI and HindIII) analysis. rRNA gene restriction patterns were further determined for different strains obtained from each source. These patterns were used for recognizing the spoilage-causing LAB strains from the product on the sell-by day and tracing the sources and sites of spoilage LAB contamination during the manufacture. LAB typing resulted in 71 different ribotypes, of which 27 were associated with contamination routes. Raw material was distinguished as the source of the major spoilage strains. Contamination of the product surfaces after cooking was shown to be airborne. The removal of the product from the cooking forms was localized as a major site of airborne LAB contamination. Food handlers and some surfaces in contact with the product during the manufacture were also contaminated with the spoilage strains. Some LAB strains were also able to resist cooking in the core of the product bar. These strains may have an effect on the product shelf life by contaminating the slicing machine. The air in the slicing department and adjacent cold room contained very few LAB. Surface-mediated contamination was detected during the slicing and packaging stages. Food handlers also carried strains later found in the packaged product. Molecular typing provided useful information revealing the LAB contamination sources and sites of this product. The production line will be reorganized in accordance with these results to reduce spoilage LAB contamination.  相似文献
