The nematode Steinernema carpocapsae (All) strain was significantly more effective against peachtree borer larvae (Synanthedon exitiosa [Lepidoptera: Sesiidae]) than Steinernema riobrave (7-12) strain in field and laboratory experiments. Eighty-eight percent control of peachtree borer larvae was obtained with S. carpocapsae in the field trial when applied at 3 x 10(5) infective juveniles per tree, and 92% mortality was obtained in the lab assay using 50 infective juveniles per larva.

Fruit bins infested with diapausing larvae of codling moth larvae, Cydia pomonella (L.), are a source of reinfestation of orchards and may jeopardize the success of mating disruption programs and other control strategies. Bins are not routinely treated for control of overwintering codling moth before placing them in orchards. Entomopathogenic nematodes provide a noninsecticidal alternative to methyl bromide that could be applied at the time bins are submerged in dump tanks at the packing house for flotation of fruit. Diapausing codling moth larvae in miniature fruit bins were highly susceptible to infective juveniles of Steinernema carpocapsae (Weiser). Immersion of bins in suspensions of S. carpocapsae ranging from 5 to 100 infective juveniles per milliliter of water resulted in 68-100% mortality. Immersion times of 1 or 5 min in suspensions with 5 infective juveniles of S. carpocapsae per milliliter of water, with and without Tween 80 (0.01%), yielded essentially the same mortality of codling moth larvae. Highest mortalities in codling moth larvae (88%) after treatment of bins in suspensions of 5 infective juveniles of S. carpocapsae per milliliter of water were observed after incubation for 24 h at 25 degrees C and 70% RH. Lowest mortalities (37%) were observed after incubation at 15 degrees C and 35% RH. Comparative tests conducted with Heterorhabditis marelatus Liu & Berry, Steinernema kraussei (Steiner), and S. carpocapsae with 5 infective juveniles per milliliter of water resulted in 21.7, 53.9, and 68.7% mortality, respectively. The use of miniature fruit bins as described in this article provides an effective means of assessing nematode efficacy without the cumbersome size of commercial bins.

The objective of this study was to determine the susceptibility of the two primary direct insect pests of hazelnuts in Oregon to three species of entomopathogenic nematodes. The entomopathogenic nematodes (Heterorhabditis marelatus Pt. Reyes, Steinernema carpocapsae All and Steinernema kraussei L137) were used in laboratory soil bioassays to determine their virulence against filbertworm, Cydia latiferreana (Walsingham) (Lepidoptera: Tortricidae) and filbert weevil, Curculio occidentalis (Casey) (Coleoptera: Curculionidae). All three nematode species were infective in laboratory bioassays. Infectivity ranged from 73-100% and 23-85% for filbertworm and filbert weevil, respectively. Field results were similar to those found in the laboratory with filbertworm larvae being more susceptible to nematode infection.

Multiple-species natural enemy approach for the biological control of the alfalfa snout beetle, Otiorhynchus ligustici (L.) (Coleoptera: Curculionidae), was compared with using single-species of natural enemies in the alfalfa ecosystem by using entomopathogenic nematodes with different dispersal and foraging behaviors. Steinernema carpocapsae NY001 (ambush nematode), Heterorhabditis bacteriophora Oswego (cruiser nematode), and Steinernema feltiae Valko (intermediate nematode) were applied in single-species, two-species combinations, and one three-species combination treatments at 2.5 x 10(9) infective juveniles per hectare. All nematode species persisted for a full year (357 d). S. carpocapsae NY001 protected the plants from root-feeding damage better than H. bacteriophora Oswego but allowed for higher larval survival than all other nematode treatments. S. feltiae Valko protected the plants better than H. bacteriophora Oswego and controlled alfalfa snout beetle larvae better than S. carpocapsae NY001. H. bacteriophora Oswego allowed for similar root damage compared with control plots but reduced larval populations better than S. carpocapsae NY001. The combination of S. carpocapsae NY001 and H. bacteriophora Oswego provided significantly better protection for the plants than the control (unlike H. bacteriophora Oswego alone) and reduced host larva survival more than S. carpocapsae NY001 alone. The combination S. feltiae Valko and H. bacteriophora Oswego could not be statistically separated from the performance of S. feltiae Valko applied alone.

Entomopathogenic nematode species available in Europe were screened for their efficacy against both the root-feeding larvae and silk-feeding adults of the western corn rootworm, Diabrotica virgifera virgifera LeConte. Laboratory screening tests were aimed at the selection of candidate biological control agents for the management of this invasive alien pest in Europe. Steinernema glaseri, S. arenarium, S. abassi, S. bicornutum, S. feltiae, S. kraussei, S. carpocapsae and Heterorhabditis bacteriophora were studied to determine their virulence against third instar larvae and adults of D. v. virgifera in small-volume arenas (using nematode concentrations of 0.5, 0.8, 7.9 and 15.9 infective juveniles cm-2). All nematode species were able to invade and propagate in D. v. virgifera larvae, but adults were rarely infected. At concentrations of 7.9 and 15.9 cm-2, S. glaseri, S. arenarium, S. abassi and H. bacteriophora caused the highest larval mortality of up to 77%. Steinernema bicornutum, S. abassi, S. carpocapsae and H. bacteriophora appeared to have a high propagation level, producing 5970+/-779, 5595+/-811, 5341+/-1177 and 4039+/-1025 infective juveniles per larva, respectively. Steinernema glaseri, S. arenarium, S. feltiae, S. kraussei and H. bacteriophora were further screened at a concentration of 16.7 nematodes cm-2 against third instar larvae in medium-volume arenas (sand-filled trays with maize plants). Heterorhabditis bacteriophora, S. arenarium and S. feltiae caused the highest larval mortality with 77+/-16.6%, 67+/-3.5%, and 57+/-17.1%, respectively. In a next step, criteria for rating the entomopathogenic nematode species were applied based on results obtained for virulence and propagation, and for current production costs and availability in Europe. These criteria were then rated to determine the potential of the nematodes for further field testing. Results showed the highest potential in H. bacteriophora, followed by S. arenarium and S. feltiae, for further testing as candidate biological control agents.

The effects of different application technologies were evaluated on the concentration, viability, and efficiency of infective juveniles of the nematodes Heterorhabditis indica Poinar, Karunakar & David and Steinernema sp. (IBCB-n6) to control Spodoptera frugiperda Smith on corn plants. Two hundred and eighty infective juveniles of Steinernema sp. were required to kill 100% third-instar fall armyworms in petri dishes, as compared to 400 infective juveniles of the H. indica nematode to obtain 75% fall armyworm control. It is possible to spray entomopathogenic nematodes without significant loss in their concentration and viability, with equipment that produces electrical charges to the spraying mix, and with those using hydraulic and rotary nozzle tips. The concentrations of infective juveniles of H. indica and Steinernema sp. nematodes were reduced by 28% and 53%, respectively, when hydraulic spraying nozzles that require 100-mesh filtrating elements were used. Tensoactive agents of the organosilicone and ethoxylate groups did not affect the viability of infective juveniles of Steinernema sp. juveniles. Spraying corn plants (V6 growth stage) with up to 288 million infective juveniles of Steinernema sp. per hectare, diluted in the spraying mix up to 800 L ha-1, with 0.01% ethoxylate tensoactive agent, or at the same volume followed by artificial rain (6 mm water depth) was not sufficient to control S. frugiperda in a controlled environment.

We studied the influence of entomopathogenic nematodes , Steinernema carpocapsae and S. riobravis, on natural populations of plant - parasitic nematodes (PPNs) infesting turfgrass in Georgia and South Carolina . S. riobravis applied at 6 109 infective juveniles (IJs) / acre provided up to 95 - 100% control of the root - knot , Meloidogyne sp ., sting , Belonolaimus longicaudatus, and ring nematode , Criconemella sp ., in Georgia , but S. carpocapsae had no effect . S. riobravis was as effective as the chemical nematicide , Fenamiphos (Nemacur 10G) at 4 weeks after treatment and more effective at 8 weeks after treatment . In South Carolina , both S. riobravis and S. carpocapsae applied at 1 109 IJs / acre provided up to 86 - 100 % control of the root - knot , sting and ring nematodes . Application of 6 109 IJs / acre increased control by only 4 - 14 % over the 1 109 dosage . Possible causes of differences in efficacy of S. carpocapsae at the two sites are discussed . It is concluded that S. riobravis may provide effective , predictable and economical control of PPNs in turfgrass .

The susceptibility of codling moth diapausing larvae to three entomopathogenic nematode species was assessed in the laboratory using a bioassay system that employed cocooned larvae within cardboard strips. The LC50values forSteinernema carpocapsae, S. riobrave, andHeterorhabditis bacteriophorawere 4.7, 4.8, and 6.0 infective juveniles/cm2, respectively. When a discriminating concentration of 10 infective juveniles/cm2of each of the three nematode species was evaluated at 15, 20, 25, and 30°C,S. carpocapsaewas the most effective nematode with mortalities ranging from 66 to 90%. Mortalities produced byS. riobraveandH. bacteriophoraat the four temperatures were 2–94 and 25–69%, respectively. Studies were also conducted to test infectivity at 10, 35, and 40°C. No mortality was produced by any of the nematode species at 10°C.S. riobravewas the most infective nematode at 35°C producing 68% mortality which was more than twice that observed forS. carpocapsaeorH. bacteriophora.Codling moth larvae treated with 10 infective juveniles/cm2ofS. carpocapsaeand kept in 95+% RH at 25°C for 0–24 h followed by incubation at 25–35% RH indicated that more than 3 h in high humidity was needed to attain 50% mortality. Trials ofS. carpocapsae, S. riobrave, andH. bacteriophoraat 50 infective juveniles/cm2against cocooned larvae on pear and apple logs resulted in reductions of codling moth adult emergence of 83, 31, and 43%, respectively, relative to control emergence. Trials of the three entomopathogenic nematodes at 50 infective juveniles/cm2against cocooned larvae in leaf litter resulted in 99 (S. carpocapsae), 80 (S. riobrave), and 83% (H. bacteriophora) mortality, respectively. Our results indicate good potential of entomopathogenic nematodes, especiallyS. carpocapsae, for codling moth control under a variety of environmental conditions.

Four species of entomopathogenic nematodes, Steinernema carpocapsae , Heterorhabditis bacteriophora , H. indica and H. marelatus , were tested for their ability to kill and reproduce in larvae of the Asian longhorn beetle, Anoplophora glabripennis (Motchulsky). The larvae were permissive to all four species but mortality was higher and production of infective juveniles was greater for S. carpocapsae and H. marelatus . The lethal dosage of H. marelatus was determined to be 19 infective juveniles for second and third instar larvae and 347 infective juveniles for fourth and fifth instar larvae. H. marelatus infective juveniles, applied via sponges to oviposition sites on cut logs, located and killed host larvae within 30 cm galleries and reproduced successfully in several of the larvae.

The infectivity and reproductive potential of the entomopathogenic nematode Heterorhabditis bacteriophora (Oswego strain), at different concentrations, was studied. Seventy to 80.0% mortality to late instar larvae of the clover root curculio, Sitona hispidulus, and 40.0-76.0% mortality to pupae, was observed at concentrations of 15-100 infective juveniles. There were no significant differences in mortality among nematode concentrations. LC(50) levels of 4.0 and 21.4 nematodes were determined for clover root curculio larvae and pupae, respectively. Nematodes did not cause significant mortality to adult or first instar clover root curculio. H. bacteriophora was able to complete its development and reproduce in 74.0-95.0% of clover root curculio late instar larvae and pupae. Reproductive potential in curculio larvae and pupae ranged from 0 to 7040 infective juveniles per host. Larvae exposed to 100 nematodes had a reproductive potential significantly higher than in those larvae exposed to 15 and 50 nematodes. Reproductive potential in pupae decreased with an increased nematode dose, indicating potential crowding effects. Host larval and pupal mass were positively correlated with nematode progeny production.

Sugar beet, Beta vulgaris L. is a strategic crop of sugar industry in Egypt. It is threatened by several insect pests among most important of them is the beet fly Pegomyia mixta. This work deals with the biological control of this insect using four entomopathogenic nematodes (EPNs). The nematodes included Steinernema carpocapsae S2, Steinernema feltiae, Heterorhabditis bacteriophora (HB1-3) and Heterorhabditis bacteriophora S1. Daily mortality of larvae and pupae of P. mixta were recorded after treatment with serial concentrations (500, 1000, 2000 and 4000 infective juveniles (IJs)/ml) of each of four studied EPNs. In the laboratory all tested nematodes killed the larvae inside their mines in the sugar beet leaves and developed in their bodies in different extends. They also killed the insect pupae in the soil and developed in their bodies. Young larvae were more susceptible than old ones. New pupae were more susceptible than old ones. In the field a single spray of S. feltiae or H. bacteriophora caused 81.3 or 75.9% reduction in the larval population of the in sugar beet leaves.

Anhydrobiosis is considered to be an important means of achieving storage stability of entomopathogenic nematodes that are used in biological control. This study explored the effects of anhydrobiosis on longevity and infectivity of infective juveniles (IJs) of three species of entomopathogenic nematodes Steinernema carpocapsae, Steinernema feltiae, and Steinernema riobrave at 5 and 25 degrees C. Anhydrobiosis was induced in water-dispersible granules (WG) at 0.966-0.971 water activity and 25 degrees C following a 7-day preconditioning of IJs at 5 degrees C in tap water. Survival and infectivity of the desiccated (anhydrobiotic) IJs was compared with non-desiccated IJs stored in water for different periods. Anhydrobiosis increased longevity of S. carpocapsae IJs by 3 months and of S. riobrave by 1 month in WG at 25 degrees C as compared with IJs stored in water. However, desiccation decreased S. feltiae longevity at 25 degrees C and of all three species at 5 degrees C. These results demonstrate a shelf-life of 5 months for S. carpocapsae at 25 degrees C and 9 months at 5 degrees C in WG with over 90% IJ survival. For S. feltiae, over 90% survival occurred only for 2 months at 25 degrees C and 5 months at 5 degrees C in WG. Steinernema riobrave had over 90% survival only for 1 month at 25 degrees C and the survival dropped below 85% within 1 month at 5 degrees C. Induction of anhydrobiosis in WG resulted in 85, 79 and 76% reduction in oxygen consumption by S. carpocapsae, S. feltiae, and S. riobrave IJs, respectively. Differences in IJ longevity among three species in water at 25 degrees C were related both to the initial lipid content and the rate of lipid utilisation, but not at 5 degrees C. The one-on-one infection bioassays indicated that desiccation had no negative effect on the infectivity of any of the nematode species suggesting no harmful effect on the IJs and/or their symbiotic bacteria. The species differences in IJ longevity and desiccation survival at different temperatures are discussed in relation to their foraging strategy and temperature adaptation.

Fruit bins infested with diapausing codling moth larvae, Cydia pomonella (L.), are a potential source of reinfestation of orchards and may jeopardize the success of mating disruption programs and other control strategies. Entomopathogenic nematodes (EPNs) were tested as a potential means of control that could be applied at the time bins are submerged in dump tanks. Diapausing cocooned codling moth larvae in miniature fruit bins were highly susceptible to infective juveniles (IJs) of Steinernema carpocapsae (Weiser) and Steinernema feltiae (Filipjev) in a series of experiments. Cocooned larvae are significantly more susceptible to infection than are pupae. Experimental treatment of bins in suspensions of laboratory produced S. feltiae ranging from 10 to 100 IJs/ml of water with wetting agent (Silwet L77) resulted in 51-92% mortality. The use of adjuvants to increase penetration of hibernacula and retard desiccation of S. feltiae in fruit bins resulted in improved efficacy. The combination of a wetting agent (Silwet L77) and humectant (Stockosorb) with 10 S. feltiae IJs/ml in low and high humidity resulted in 92-95% mortality of cocooned codling moth larvae versus 46-57% mortality at the same IJ concentration without adjuvants. Immersion of infested bins in suspensions of commercially produced nematodes ranging from 10 to 50 IJs/ml water with wetting agent in an experimental packing line resulted in mortality in cocooned codling moth larvae of 45-87 and 56 - 85% for S. feltiae and S. carpocapsae, respectively. Our results indicate that EPNs provide an alternative nonchemical means of control that could be applied at the time bins are submerged in dump tanks at the packing house for flotation of fruit.

Simultaneous use of parasitoids and entomopathogenic nematodes for codling moth (CM) control could produce an antagonistic interaction between the two groups resulting in death of the parasitoid larvae. Two ectoparasitic ichneumonid species, Mastrus ridibundus and Liotryphon caudatus, imported for classical biological control of cocooned CM larvae were studied regarding their interactions with Steinernema carpocapsae. Exposure of M. ridibundus and L. caudatus developing larvae to infective juveniles (IJs) of S. carpocapsae (10 IJs/cm2; approximately LC(80-90) for CM larvae) within CM cocoons resulted in 70.7 and 85.2% mortality, respectively. However, diapausing full grown parasitoid larvae were almost completely protected from nematode penetration within their own tightly woven cocoons. M. ridibundus and L. caudatus females were able to detect and avoid ovipositing on nematode-infected cocooned CM moth larvae as early as 12h after treatment of the host with IJs. When given the choice between cardboard substrates containing untreated cocooned CM larvae and those treated with an approximate LC95 of S. carpocapsae IJs (25 IJs/cm2) 12, 24, or 48h earlier, ovipositing parasitoids demonstrated a significant preference for untreated larvae. The ability of these parasitoids to avoid nematode-treated larvae and to seek out and kill cocooned CM larvae that survive nematode treatments enhances the complementarity of entomopathogenic nematodes and M. ridibundus and L. caudatus.

Eight laboratory-reared ant species were fed baits of house fly, Musca domestica L., pupae treated with hydramethylnon. Two fire ant species, Solenopsis invicta Buren and Solenopsis geminata (F.), and Pheidole morrissi (Forel) were killed; average percentage of mortality of the five other species was less than 20%. In contrast, all species that were fed the commercial fire ant bait containing hydramethylnon (Amdro) died or were adversely affected. In the field, applications of house fly pupae and eye gnat, Hippelates pusio Loew, pupae dipped in acetone solutions of fenoxycarb significantly reduced population indices of the red imported fire ant, S. invicta, compared with commercial formulations of fenoxycarb (Logic) and hydramethylnon (Amdro). Field observations showed that the pupae of either species can be carried or moved by one or two worker ants. The smooth, hard cuticle of the pupae make them easy to handle and apply with application equipment. The current cost of house fly pupae is considerably more than the cost of a granular carrier, pregel defatted corn grits. However, if mass-production methods reduce this price differential, fly pupae could become an effective and more species-specific fire ant bait carrier.

The objective of this study was to determine the susceptibility of the strawberry crown moth, Synanthedon bibionipennis (Boisduval) (Lepidoptera: Sesiidae) larvae to two species of entomopathogenic nematodes. The entomopathogenic nematodes Steinernema carpocapsae (Weiser) strain Agriotos and Heterorhabditis bacteriophora (Steiner) strain Oswego were evaluated in laboratory soil bioassays and the field. Both nematode species were highly infective in the laboratory bioassays. Last instars were extremely susceptible to nematode infection in the laboratory, even in the protected environment inside the strawberry (Fragaria x ananassa Duch.) crown. Infectivity in the laboratory was 96 and 94% for S. carpocapsae and H. bacteriophora, respectively. Field applications in late fall (October) were less effective with S. carpocapsae and H. bacteriophora, resulting in 51 and 33% infection, respectively. Larval mortality in the field from both nematode treatments was significantly greater than the control, but treatments were substantially less efficacious than in the laboratory. Soil temperature after nematode applications in the field (11 degrees C mean daily temperature) was below minimum establishment temperatures for both nematode species for a majority of the post-application period. It is clear from laboratory data that strawberry crown moth larvae are extremely susceptible to nematode infection. Improved control in the field is likely if nematode applications are made in late summer to early fall when larvae are present in the soil and soil temperatures are more favorable for nematode infection.

Abstract: The strains Steinernema feltiae Otio and A54, Steinernema ceratophorum D43 and Steinernema carpocapsae BJ were tested for their infectivity to the larvae and pupae of beetle ( Luperomorpha suturalis Chen) at 25 ± 0.5°C and 15 ± 0.5°C in laboratory conditions. The results, based on comparison of the insect mortalities and nematode penetration rates among four nematode strains, showed that S. feltiae Otio was a potential biocontrol agent of the larvae and pupae of L. suturalis . The mortalities of the larvae and pupae exposed to S. feltiae Otio strain were 95.8 and 97.1% at 25 ± 0.5°C and 78.0 and 83.0% at 15 ± 0.5°C, respectively. The nematode penetration rates of S. feltiae Otio of the larvae and pupae were 15.6 and 19.0% at 25 ± 0.5°C, 2.6 and 6.3% at 15 ± 0.5°C, respectively. Field efficacy of S. feltiae Otio strain was examined against beetle larvae in Hebei province, northern China. The population reduction of insect larvae was 77.8 and 13.9% at doses of 30 and 15 infective juveniles (IJs)/cm2 of S. feltiae Otio after 38 days of treatment and 90.2 and 92.4% after 100 days of treatment. However, the population of the insect larvae was reduced only to 15.5 and 15.7% when treated with pesticide after 38 and 100 days, respectively. The efficiency between the two nematode doses was not significantly different but it was remarkably higher than that of the pesticide after 100 days of application. The results suggest that S. feltiae Otio strain could be an alternative to pesticide for beetle control.

A recently introduced fungus gnat, Bradysia agrestis, has caused serious problems in Korean propagation houses where vegetable seedlings are produced for transplant into the fields. Although chemical insecticides are available against this pest, alternate control measures are needed. A Korean isolate of the entomopathogenic nematode, Steinernema carpocapsae Pocheon strain, was tested against this insect in the laboratory and propagation house. In the laboratory, S. carpocapsae affected oviposition, with the untreated females laying an average of 121±25 eggs, whereas the treated females averaged 7±2 eggs. The infectivity of S. carpocapsae to the fungus gnat was affected by the developmental stage and temperature, with highest mortality observed with the third and fourth instars and pupal stage. Nematode mortality in the second instar fungus gnat ranged between 23 and 35%, but showed no significant differences among the temperatures tested. The egg and first instar were not infected by the nematode. In nematode dispersal studies, adult female fungus gnats alone dispersed S. carpocapsae from the nematode-treated area to the control area at a higher rate than male and female gnats or male gnats alone. In the propagation house experiments with watermelon seedlings, no significant difference was observed in fungus gnat larval reduction at S. carpocapsae concentrations of 5, 10, or 20 infective juveniles (IJs)/g of soil at 7, 14, and 21 days after treatment. In comparison with the control, the S. carpocapsae treatments significantly reduced B. agrestis larval numbers. When the watermelon seed was treated with S. carpocapsae at sowing, the larval density of B. agrestis was significantly reduced, that is, the number of B. agrestis larvae ranged from four to eight and from five to eight in the nematode-treated plots compared with 26 and 30 in the control plots on the 17th and 34th day post-treatment, respectively. In the chemical insecticide treatments, diflubenzuron and chlorpyrifos were significantly more effective than S. carpocapsae and diazinon in reducing larval populations of the fungus gnat. Our data show that, although some of the chemical insecticides were more effective than S. carpocapsae Pocheon strain, the nematode was still an effective tool for management of the fungus gnat larvae and in protecting seedlings from damage in propagation houses.
