The influence of site fertility on soil microbial biomass and activity is not well understood but is likely to be complex because of interactions with plant responses to nutrient availability. We examined the effects of long-term (8 yr) fertilization and litter removal on forest floor microbial biomass and N and C transformations to test the hypothesis that higher soil resource availability stimulates microbial activity. Microbial biomass and respiration decreased by 20–30 % in response to fertilization. Microbial C averaged 3.8 mg C/g soil in fertilized, 5.8 mg C/g in control, and 5.5 mg C/g in litter removal plots. Microbial respiration was 200 µg CO2-C g–1 d–1 in fertilized plots, compared to 270 µg CO2-C g–1 d–1 in controls. Gross N mineralization and N immobilization did not differ among treatments, despite higher litter nutrient concentrations in fertilized plots and the removal of substantial quantities of C and N in litter removal plots. Net N mineralization was significantly reduced by fertilization. Gross nitrification and NO3 immobilization both were increased by fertilization. Nitrate thus became a more important part of microbial N cycling in fertilized plots even though NH4 + availability was not stimulated by fertilization.Soil microorganisms did not mineralize more C or N in response to fertilization and higher litter quality; instead, results suggest a difference in the physiological status of microbial biomass in fertilized plots that influenced N transformations. Respiration quotients (qCO2, respiration per unit biomass) were higher in fertilized plots (56 µg CO2-C mg C–1 d–1) than control (48 µg CO2-C mg C–1 d –1) or litter removal (45 µg CO2-C mg C–1 d–1), corresponding to higher microbial growth efficiency, higher proportions of gross mineralization immobilized, and lower net N mineralization in fertilized plots. While microbial biomass is an important labile nutrient pool, patterns of microbial growth and turnover were distinct from this pool and were more important to microbial function in nitrogen cycling.

The effects of drought on soil dynamics after fire are poorly known, particularly its long‐term (i.e., years) legacy effects once rainfall returns to normal. Understanding this is particularly important for nutrient‐poor soils in semi‐arid regions affected by fire, in which rainfall is projected to decrease with climate change. Here, we studied the effects of post‐fire drought and its legacy on soil microbial community structure and functionality in a CistusErica shrubland (Spain). Rainfall total and patterns were experimentally modified to produce an unburned control (natural rainfall) and four burned treatments: control (natural rainfall), historical control (long‐term average rainfall), moderate drought (percentile 8 historical rainfall, 5 months of drought per year), and severe drought (percentile 2, 7 months of drought). Soil nutrients and microbial community composition (ester‐linked fatty acid approach) and functionality (enzyme activities and C mineralization rate) were monitored during the first 4 years after fire under rainfall treatments, plus two additional ones without them (six post‐fire years). We found that the recovery of burned soils was lower under drought. Post‐fire drought increased nitrate in the short term and reduced available phosphorus, exchangeable potassium, soil organic matter, enzyme activities, and carbon mineralization rate. Moreover, drought decreased soil total microbial biomass and fungi, with bacteria becoming relatively more abundant. Two years after discontinuing the drought treatments, the drought legacy was significant for available phosphorus and enzyme activities. Although microbial biomass did not show any drought legacy effect, the proportion of fungi and bacteria (mainly gram‐positive) did, being lower and higher, respectively, in former drought‐treated plots. We show that drought has an important impact on soil processes, and that some of its effects persist for at least 2 years after the drought ended. Therefore, drought and its legacy effects can be important for modeling biogeochemical processes in burned soils under future climate change.

Long- and short-term effects of fire on nitrogen cycling in tallgrass prairie
Fires in the tallgrass prairie are frequent and significantly alter nutrient cycling processes. We evaluated the short-term changes in plant production and microbial activity due to fire and the long-term consequences of annual burning on soil organic matter (SOM), plant production, and nutrient cycling using a combination of field, laboratory, and modeling studies. In the short-term, fire in the tallgrass prairie enhances microbial activity, increases both above-and belowground plant production, and increases nitrogen use efficiency (NUE). However, repeated annual burning results in greater inputs of lower quality plant residues causing a significant reduction in soil organic N, lower microbial biomass, lower N availability, and higher C:N ratios in SOM. Changes in amount and quality of below-ground inputs increased N immobilization and resulted in no net increases in N availability with burning. This response occurred rapidly (e.g., within two years) and persisted during 50 years of annual burning. Plant production at a long-term burned site was not adversely affected due to shifts in plant NUE and carbon allocation. Modeling results indicate that the tallgrass ecosystem responds to the combined changes in plant resource allocation and NUE. No single factor dominates the impact of fire on tallgrass plant production.

Exotic Earthworm Invasion and Microbial Biomass in Temperate Forest Soils
Invasion of north temperate forest soils by exotic earthworms has the potential to alter microbial biomass and activity over large areas of North America. We measured the distribution and activity of microbial biomass in forest stands invaded by earthworms and in adjacent stands lacking earthworms in sugar maple-dominated forests in two locations in New York State, USA: one with a history of cultivation and thin organic surface soil horizons (forest floors) and the other with no history of cultivation and a thick (3–5 cm) forest floor. Earthworm invasion greatly reduced pools of microbial biomass in the forest floor and increased pools in the mineral soil. Enrichment of the mineral soil was much more marked at the site with thick forest floors. The increase in microbial biomass carbon (C) and nitrogen (N) in the mineral soil at this site was larger than the decrease in the forest floor, resulting in a net increase in total soil profile microbial biomass in the invaded plots. There was an increase in respiration in the mineral soil at both sites, which is consistent with a movement of organic matter and microbial biomass into the mineral soil. However, N-cycle processes (mineralization and nitrification) did not increase along with respiration. It is likely that the earthworm-induced input of C into the mineral soil created a microbial sink for N, preventing an increase in net mineralization and nitrification and conserving N in the soil profile.

Grass species and soil type effects on microbial biomass and activity
We evaluated plant versus soil type controls on microbial biomass and activity by comparing microbial biomass C, soil respiration, denitrification potential, potential net N mineralization and nitrification in different soils supporting four grass species, and by growing a group of 10 different grass species on the same soil, in two experiments respectively. In the first experiment, none of the microbial variables showed significant variation with grass species while all variables showed significant variation with soil type, likely due to variation in soil texture. In the second experiment, there were few significant differences in microbial biomass C among the 10 grasses but there were significant relationships between variation in microbial biomass C and potential net N mineralization (negative), soil respiration (positive) and denitrification (positive). There was no relationship between microbial biomass C and either plant yield or plant N concentration. The results suggest that 1) soil type is a more important controller of microbial biomass and activity than grass species, 2) that different grass species can create significant, but small and infrequent, differences in microbial biomass and activity in soil, and 3) that plant-induced variation in microbial biomass and activity is caused by variation in labile C input to soil.

The recovery of soil biochemical properties under grazed, grass-clover pasture, after simulated lignite mining, was studied over a 5-year period in a mesic Typic Dystrochrept soil at Waimumu, Southland, New Zealand. The restoration procedures involved four replacement treatments, after A, B, and C horizon materials had been separately removed, from all except the control, and stockpiled for 2–3 weeks. In each replacement treatment, the effects of ripping to 1.8 m depth, mole drainage, and the use of fertilizer nitrogen were also investigated.Replacement treatment markedly influenced the recovery of herbage production and soil organic C and total N contents, N mineralization, microbial biomass (as indicated by mineral-N flush) and invertase and sulphatase activities. The effectiveness of replacement treatments decreased in the order: 1. control (no stripping or replacement). 2 A, B, and C horizon materials replaced in the same order. 3. A, B, and C horizon materials each mixed with an equal amount of siltstone overburden and replaced in order, 4. A and B horizon materials mixed before replacing over C horizon material.Ripping increased herbage production, net N mineralization, and to some extent microbial biomass. Drainage had little, if any, effect.Fertilizer N also stimulated herbage production, but depressed clover growth. Over 2.5 years, it had little detectable effect on the soil properties.Increases in soil invertase and, to a lesser extent, sulphatase activity during the trial were closely related to changes in herbage production. Microbial biomass increased more rapidly than did soil organic C in the early stages of the trial.Rates of net N mineralization strongly suggest that N availability would have limited pasture growth, especially in the treatments with mixed soil materials.

In order to investigate the effects of savanna fires on nutrient cycling a field experiment was carried out in an open woodland savanna of southwest Ethiopia. This involved manipulations of fire, fuel load and ash fertilisation in a fully factorial design, and recording of responses in plants, soil inorganic and microbial nutrient pools up to 1 year after the disturbances. As plant biomass nitrogen (N) was only 3.5% of that in topsoil the N loss in a single fire event was relatively small. The microbial N pool size in the topsoil was similar to the N pool size in the aboveground part of the plants. Soil microbial biomass carbon increased slightly 12 days after the low severity fire, but the effect was transient and was not accompanied by an increase in microbial N. Instead, the soil inorganic N concentration increased strongly 1 day after the fire, remained higher up to 3 months after the fire and probably caused the 40% higher grass biomass in burned than unburned plots, and the similar sized increase in grass nitrogen, phosphorus and potassium pools in the following rainy season. In contrast, broad-leaved herbs showed less strong increments in biomass and nutrient pool sizes. Fire interacted with fuel load, as burning of plots with double plant biomass led to reduced microbial biomass, plant nutrient pools and herb (but not grass) biomass. Low-severity-fire nutrient losses appear to be moderate and may be replenished from natural sources. However, in areas with frequent fires and high grass biomass (fuel) loads, or with late fires, nutrient losses could be much larger and non-sustainable to the persistence of the woodland savanna ecosystem.

In this study we characterized spatial heterogeneity of soil carbon and nitrogen pools, soil moisture, and soil pH of the first 15?cm of the soil profile; depth of the organic horizon; forest floor covers; and understory vegetation abundances in three sites (1999, 1987 and 1920 wildfires) of a boreal forest chronosequence of interior Alaska. We also investigated the cross-dependence between understory vegetation distribution and soil characteristics. Our results showed higher microbial respiration rates and microbial biomass in the oldest site and greater net N mineralization rates in the mid-successional site. Although spatial heterogeneity was absent at the scale studied for the majority of soil variables (60%), understory vegetation abundances and forest floor cover, spatial heterogeneity decreased with time after fire for the depth of organic horizon, soil microbial biomass, N mineralization rates and feathermoss cover. Our results also showed that increasing time after fire decreased the number of correlations between understory vegetation and soil characteristics while it increased between forest floor covers and soil characteristics. Overall, our study suggest that fire initially creates a patchy mosaic of forest floor cover, from fire hot spots, where high intensity burning exposes mineral soil, to practically unburned areas with intact mosses and lichens. As time since fire passes, forest floor cover and soil characteristics tend to become more uniform as understory species fill in severely burned areas.

Factors influencing rates of C and N mineralization of soil and plant materials, and the reliability of different procedures for estimating microbial biomass, were measured in a soil (Typic Dystrochrept) that had been restored under grazed pasture in a temperate environment for 10–11 years after 20 cm of the original topsoil had been removed by stripping. Rates of net N mineralization were appreciably lower, but CO2-C production higher, in the stripped than in the unstripped soil. These activities were not influenced directly by levels of soil mineral-N, but they were influenced by differences in plant composition. Herbage and litter, and roots, from the stripped plots were generally mineralized more readily to CO2-C, but more slowly to net mineral-N, than were the corresponding materials from the unstripped plots. Rates of mineralization of herbage and litter, or roots, were mainly indistinguishable in stripped and unstripped soil, whereas rates of mineralization of all standing dead material were lower in stripped soil. Measurements of extractable-C flush, and of CO2-C flush (using a fumigated soil control) and mineral-N flush by fumigation-incubation procedures, indicated that microbial biomass in stripped soil had recovered to at least 88 percent of the levels in unstripped soil. Substrate-induced respiration also generally indicated high levels of recovery of microbial biomass. The fumigation-incubation procedure appeared to under-estimate microbial biomass markedly in stripped soil when unfumigated soil controls were used; the used of a large soil inoculum (20 percent w/w) only sometimes overcame this problem. Possible reasons for apparent anomalies in estimation of microbial C are discussed.

Understanding the responses of terrestrial ecosystems to global change remains a major challenge of ecological research. We exploited a natural elevation gradient in a northern hardwood forest to determine how reductions in snow accumulation, expected with climate change, directly affect dynamics of soil winter frost, and indirectly soil microbial biomass and activity during the growing season. Soils from lower elevation plots, which accumulated less snow and experienced more soil temperature variability during the winter (and likely more freeze/thaw events), had less extractable inorganic nitrogen (N), lower rates of microbial N production via potential net N mineralization and nitrification, and higher potential microbial respiration during the growing season. Potential nitrate production rates during the growing season were particularly sensitive to changes in winter snow pack accumulation and winter soil temperature variability, especially in spring. Effects of elevation and winter conditions on N transformation rates differed from those on potential microbial respiration, suggesting that N‐related processes might respond differently to winter climate change in northern hardwood forests than C‐related processes.

Fire is a fundamental reorganizing force in chaparral and other Mediterranean-type ecosystems. Postfire nutrient redistribution and cycling are frequently invoked as drivers of ecosystem recovery. The extent to which N is transported from slopes to streams following fire is a function of the balance between the rate at which soil microbes retain and metabolize N into forms that readily dissolve or leach, and how rapidly recovering plants sequester this mobilized N. To better understand how fire impacts this balance, we sampled soil and plant N dynamics in 17 plots distributed across two burned, chaparral-dominated watersheds in Santa Barbara County, California. We measured a variety of ecosystem properties in both burned and unburned plots on a periodic basis for 2 years, including soil water content, pH, soil and plant carbon and nitrogen, extractable inorganic nitrogen, dissolved organic nitrogen, and microbial biomass. In burned plots, nitrification was significantly enhanced relative to rates measured in unburned plots. Ephemeral herbs established quickly following the first postfire rain events. Aboveground plant biomass assimilated N commensurate with soil net mineralization, implying tight N cycling during the early stages of recovery. Microbial biomass N, on the other hand, remained low throughout the study. These findings highlight the importance of herbaceous species in conserving ecosystem nutrients as shrubs gradually recover.

Four biochar types, produced by slow pyrolysis of poultry litter (PL) and pine chips (P) at 400 or 500 °C, were added to two adjacent soils with contrasting soil organic matter (SOM) content (8.9 vs. 16.1 g C kg?1). The N mineralization rate was determined during 14‐week incubations and assessments were made of the microbial biomass C, dehydrogenase activity, and the microbial community structure (PLFA‐extraction). The addition of PL biochars increased the net N mineralization (i.e., compared to the control treatment) in both soils, while for treatments with P biochars net N immobilization was observed in both soils. Increasing the pyrolysis temperature of both feedstock types led to a decrease in net N mineralization. The ratio of Bacterial to Fungal PLFA biomarkers also increased with addition of biochars, and particularly in the case of the 500 °C biochars. Next to feedstock type and pyrolysis temperature, SOM content clearly affected the assessed soil biological parameters, viz. net N mineralization or immobilization, MBC and dehydrogenase activity were all greater in the H soil. This might be explained by an increased chance of physical contact between the microbial community activated by SOM mineralization upon incubation and discrete biochar particles. However, when considering the H soil's double C and N content, these responses were disproportionally small, which may be partly due to the L soil's, somewhat more labile SOM. Nonetheless, increasing SOM content and microbial biomass and activity generally appears to result in greater mineralization of biochar. Additionally, higher N mineralization after PL addition to the H soil with lower pH than the L soil can be due to the liming effect of the PL biochars.

Prescribed fire has become a common tool of natural area managers for removal of non‐indigenous invasive species and maintenance of barrens plant communities. Certain non‐native species, such as tall fescue (Festuca arundinacea), tolerate fire and may require additional removal treatments. We studied changes in soil N and C dynamics after prescribed fire and herbicide application in remnant barrens in west central Kentucky. The effects of a single spring burn post‐emergence herbicide, combined fire and herbicide treatments, and an unburned no‐herbicide control were compared on five replicate blocks. In fire‐plus‐herbicide plots, fescue averaged 8% at the end of the growing season compared with 46% fescue cover in control plots. The extent of bare soil increased from near 0 in control to 11% in burned plots and 25% in fire‐plus‐herbicide plots. Over the course of the growing season, fire had little effect on soil N pools or processes. Fire caused a decline in soil CO2 flux in parallel to decreased soil moisture. When applied alone, herbicide increased plant‐available soil N slightly but had no effect on soil respiration, moisture, or temperature. Fire‐plus‐herbicide significantly increased plant‐available soil N and net N transformation rates; soil respiration declined by 33%. Removal of non‐native plants modified the chemical, physical, and biological soil conditions that control availability of plant nutrients and influence plant species performance and community composition.

The effect of climate change on biologically mediated soil processes is likely to be complex and difficult to predict. The direct effect of changes in soil moisture and temperature may be obscured or even reversed by changes in the return of organic substrates to the soil. This paper is the first in a series reporting the results from a medium-term investigation into the effect of simulated climate change on soil N mineralization in a seminatural calcareous grassland in southern England. Gross rates of N mineralization were determined by isotope dilution in plots subjected to winter warming (3 °C above ambient), enhanced or decreased summer rainfall and combinations of these treatments. The results from the control treatment, reported here, show a strong seasonality of gross N mineralization with rates highest in the Spring and Autumn and lowest in Summer. They indicate that water availability is the main restraint on microbial processes and plant growth and that, in the short term, enhanced summer rainfall is likely to be the main factor influencing nutrient turnover. Further papers will report the results from the imposed treatments.

Given the same amount of irrigation volume, applying alternate partial root-zone irrigation (PRI) has improved crop N nutrition as compared to deficit irrigation (DI), yet the mechanisms underlying this effect remain unknown. Therefore, the objective of this study was to investigate whether PRI induced soil dry/wet cycles facilitate soil organic N mineralization hereby contributing to the improvement of N nutrition in tomatoes. The plants were grown in split-root pots in a climate-controlled glasshouse and were subjected to PRI and DI treatments during early fruiting stage. 15N-labeled maize residues were incorporated into the soils. Results showed that PRI resulted in 25% higher net 15N mineralization than did DI, indicating that the enhanced mineralization of soil organic N alone could account for the 16% increase of N accumulation in the PRI than in the DI plants. The higher net N mineralization under PRI was coincided with an intensified soil microbial activity. In addition, even though soil chloroform fumigation labile carbon (CFL-C, as an index of microbial biomass) was similar for the two irrigation treatments, a significant increase of chloroform fumigation labile nitrogen (CFL-N) was found in the PRI wetting soil. Consequently, the C:N ratio of the chloroform fumigation labile pool was remarkably modified by the PRI treatment, which might indicate physiological changes of soil microbes or changes in labiality of soil organic C and N due to the dry/wet cycles of soils, altering conditions for net N mineralization. Moreover, in both soil compartments PRI caused significantly less extractable organic carbon (EOC) as compared with DI; whilst in the PRI wetting soil significantly higher extractable organic nitrogen (EON) was observed. A low EOC:EON ratio in the PRI wetting soil may indicate an increasing net mineralization of the organic N as a result of microbial metabolism. Conclusively, PRI induced greater microbial activity and higher microbial substrates availability are seemingly responsible for the enhanced net N mineralization and improved N nutrition in tomato plants.

Assessing the status of soil nutrients with their corresponding microbial communities provides important information about degraded soils during the restoration of coastal wet pine forests. Net nitrogen mineralization, nitrogen‐oxidizing bacteria (NOB), and soil microbial biomass were compared with patch‐derived volume along a 110‐year longleaf pine (Pinus palustris Mill.) chronosequence for identifying a trajectory and ecological benchmark during forest restoration. Net nitrogen mineralization rates decreased significantly in the maturing‐aged, pine patches, driven by a larger drop in net nitrification. Net nitrification and abundance of NOB were higher in young pine patches compared to soils from the maturing (86–110 years) pine patches. Gross nitrate fluxes followed the nonfungal portion of the soil microbial biomass along the chronosequence, declining in 64‐year‐old pine patches. Microbial biomass peaked in patches 17–34 years of age, but significantly declined in the older patches. Fungal biomass leveled off without decline. Ammonium was the major source of nitrogen within the maturing pine patches as well as the wetland patches, indicating that ammonium maintains longleaf pine during growth‐limiting conditions. Nitrate dominated during rapid tree growth, optimally in mesic conditions. The relative amounts of available ammonium to nitrate can be used to model nitrogen cycling in facultative‐wetland pine forests of the coastal United States as soils alternate between wet and mesic conditions. A key restoration benchmark occurred after 86 years of pine development when pine patch growth rates slowed, with lower numbers of NOB, when the nonfungal biomass leveled off, and net nitrification rates are at a minimum, during pine maturation.

The aim of this study was to clarify the effects of clear-cutting and burning (CCB) on soil fertility in a Pinus massoniana (Masson pine) plantation after CCB in Fujian Province, China. We investigated changes in nitrogen (N) mineralization potential (N0), N mineralization rate constant (k) and the apparent activation energy (Ea) of the soil, with a mathematical analysis using a kinetics model based on the results of in vitro incubation. Changes in the amount of microbial biomass nitrogen (MBN), as well as the number of heterotrophic and nitrite-oxidizing bacteria, were also investigated. The N0, MBN and the number of fungi and actinomycetes in forest soil was reduced for at least 18 months after CCB. The number of heterotrophic and nitrite-oxidizing bacteria increased, and k, Ea and N mineralization became greater after 6 months of CCB, compared with the control plots. Because there were few young trees planted, which would have taken up mineralized N in the post-CCB site, it is probable that a high proportion of the mineralized N that accumulated in the soil may have been lost during the summer rainy season. Therefore, it is suggested that CCB led to a deficiency in available N during short rotations, which resulted in soil degradation.
