Native southern bean mosaic virus contains a significant number of Mg2+ and Ca2+ ions. These can be removed by treatment with EDTA causing the virus to swell by 7% in radius at alkaline pH values. The swollen virions are susceptible to protease and nuclease digestion. They are likely to be an intermediate during assembly and disassembly. Crystals of the metal-free virus have been grown and were found to be approximately isomorphous with the orthorhombic type III southern bean mosaic virus crystals (Akimoto, T., Wagner, M.A., Johnson, J.E., and Rossmann, M.G. (1975) J. Ultrastruct. Res. 53, 306-318), although the cell dimensions are longer by 2%. Native rhombohedral type II crystals disintegrate on changing the pH or increasing the ionic strength of the mother liquor. Damage can be prevented by addition of ethylene glycol. At alkaline pH values, these crystals also show a 2% increase in their cell dimensions as well as a significant alteration in their diffraction patterns. In the type II and III crystals, the viruses pack with only their 5-fold axes in contact. Thus, the difference of the apparent swelling in solution and in the crystals may be one of differential swelling over the virus surface.  相似文献

RNA isolated from southern bean mosaic virions contains, in small amount, a subgenomic RNA (molecular weight, 0.38 × 106) that serves in vitro as an mRNA for southern bean mosaic virus coat protein. The RNA has a 5′-linked protein indistinguishable from the protein linked to the 5′ end of full-length genomic RNA. Its base sequence, determined to 91 bases from the 3′ end, is identical to the 3′-terminal sequence of the genomic RNA. The results suggest that the coat protein messenger sequence exists as a “silent” cistron near the 3′ end of the genomic RNA.  相似文献

The T = 3 capsid of southern bean mosaic virus is analyzed in detail. The beta-sheets of the beta-barrel folding motif that form the subunits show a high degree of twist, generated by several beta-bulges. Only 34 water molecules were identified in association with the three quasi-equivalent subunits, most of them on the external viral surface. Subunit contacts related by quasi-3-fold axes are similar, are dominated by polar interactions and have almost identical calcium binding sites. There is no metal ion on the quasi-3-fold axis, as previously reported. Subunits related by quasi-2-fold and icosahedral 2-fold axes have different contacts but nevertheless display almost identical interactions between the antiparallel helices alpha A. A dipole-dipole type interaction between these helices may produce an energetically stable hinge that allows two types of dimers in a T = 3 assembly. The temperature factor distribution, the hydrogen-bonding pattern, and the contacts across the icosahedral 2-fold axes suggest that one of the dimer types is present in the intact virion and probably also in solution; the other is produced only during capsid assembly. Interactions along the 5-fold axes are mainly polar and possibly form an ion channel. The beta-sheet structures of the three subunits can be superimposed with considerable precision. Significant relative distortions between quasi-equivalent subunits occur mainly in helices and loops. The two dimeric forms and the subunit distortions are the consequence of the non-equivalent subunit environments in the capsid.  相似文献

Purified preparations of an isolate of black raspberry latent virus (BRLV) contained quasispherical particles with a mean diameter of 28·5 nm; these particles were resolved into three sedimenting components (s20, w= 82S, 95S and 104S), but when centrifuged to equilibrium in caesium chloride solution they formed a single infective band (σ= 1·35 g/cm3). During electrophoresis in polyacrylamide gels, virus particles separated into three classes, and virus RNA was resolved into three major (mol. wt 1·35, 1·10 and 0·85 × 106) and one minor (mol. wt 0·4 × 106) component. The protein from virus particles had an estimated mol. wt of 28000. Isolates of BRLV were found to be serologically related but not identical to some strains of tobacco streak virus. No symptoms developed in black raspberry seedlings infected with BRLV by mechanical inoculation, nor in eight red raspberry cultivars infected by graft inoculation. However, graft inoculation of BRLV to Rubus henryi, R. phoenicolasius and Himalaya blackberry induced symptoms typical of necrotic shock disease.  相似文献

Summary Virus distribution patterns and ultrastructural changes in soybean callus cells after infection with the type, or bean strain, of southern bean mosaic virus (SBMV) were observed. Calli grown in liquid Linsmaier and Skoog (LS) medium were inoculated with SBMV and incubated in fresh LS medium. Calli were sampled at 5, 10, 15, and 20 d after inoculation and examined by transmission electron microscopy. Five days after inoculation, viruslike particles (VLP) measuring 22 to 27 nm in d were observed in the cytoplasm. The particles formed loose aggregates with some tendency to associate in regular patterns. By the 10th d after infection, particles were observed in the vacuoles in similar loose arrangements. Viruslike particles were readily identified in vacuoles because of the absence of ribosomes. Crystalline aggregations of VLP were found from Day 10 to Day 20 in the cytoplasm only. Five days after inoculation particles similar to the VLP observed in the cytoplasm also were present in nuclei. Other cytopathic effects were noted, particularly several types of inclusion bodies. These observations differ considerably from reports of the type strain in intact bean plant tissues in the frequent occurrence of VLP in vacuoles and virus crystals in cytoplasm of soybean callus infected with the same strain of virus. The effect on serological activity of methods of storing antisera to southern bean mosaic virus and tobacco mosaic virus was tested. Aliquots (0.5 ml) were (1) stored at —70, —20, +4, +26 or +37 °C, (2) repeatedly frozen and thawed, (3) stored in 5% peptone, 0.02% sodium azide or 50% glycerol, (4) diluted before storage, or (5) freeze-dried. All samples were titrated against purified virus after 1, 6 and 12 months. Most of the treatments had little effect on the precipitin titres of the antisera, but the titres were decreased by storage at 37 °C. Southern bean mosaic virus RNA was translated in wheat embryo extracts and in nuclease-treated rabbit reticulocyte lysates. Four principal products were synthesized: two related products with molecular weights of 105,000 and 75,000, a product with a molecular weight of 29,000 that closely resembled coat protein, and a product with a molecular weight of 14,000. Studies on the southern bean mosaic virus coat protein have established the molecular weight of this protein, its amino acid composition, the nature of its C-terminal amino acid, and the blockage of the N-terminal residue by an acetyl group. After hydrolysis of the protein by trypsin, the hydrolysate was fractionated by ion-exchange chromatography. Among the purified tryptic peptides were isolated the N- and the C-terminal peptides where sequences were determined, principally by mass spectrometry.  相似文献

The effect of temperature on the number of lesions and the time of their appearance was studied by inoculating French bean leaves (Phaseolus vulgaris L. cv. Perli?ka) with lucerne mosaic virus either 24 or 48 h before or, 24 or 48 h after they were exposed to various temperatures. The temperatures tested were 23, 25, 27, 30, 33 and 36° C. Before and after such exposures the plants were kept in a constant temperature of 25° C. By increasing the temperature before inoculation the number of lesions increased in comparison with the control. The optimal temperature for the maximum number of lesions is between 27° and 30° C. There is no significant difference between those experiments when the exposure time was 24 h or 48 h before inoculation. The same temperatures applied for 24 or 48 h after inoculation have a decreasing effect upon the number of lesions formed by LMV on French bean leaves. The decrease is 30 to 75%. In this case the first necrotic local lesions appeared 42 h after inoculation when exposed to higher temperatures above 27° C for 24 h, and 60 h after inoculation when exposed to these temperatures for 48 h. The shape of lesions varied a little in both cases as the pictures show.  相似文献

After evaluation of the responses of bean and broad bean common cultivars against an isolate of Cucumber mosaic virus (CMV-K) and Bean yellow mosaic virus (BYMV-K), interaction of isolates was statistically studied on co-infected plants of bean cv. Bountiful and broad bean cv. Lahijan at two trials. Based on viral relative concentration determined by quantitative enzyme-linked immunosorbent assay, BYMV interacts synergistically with CMV in bean at 14 days post inoculation, while in co-infection with BYMV, CMV interacts antagonistically in both host plants at least in one of the two trials. This suggests that CMV/BYMV interaction is dependent on host species and developmental stage of plant. Co-infection like single infection with CMV in bean plants led to significantly decrease in plants’ height and fresh weight than BYMV singly infected and healthy plants, while viral infection of broad bean plants did not significantly affect growth parameters. Decline effect of viral infection (especially co-infection) on chlorophyll and carotenoids value of bean plants was greater than those of broad bean. Viral infection (singly or doubly) caused irregular changes in nutrient elements values of both hosts compared with healthy ones.  相似文献

The effects of vertical and horizontal clinostating with container velocity 2 rpm and platform velocity 1 rpm on Apogee wheat resistance to wheat streak mosaic virus (WSMV) were studied. For the first time the yield of grain was obtained from both, healthy and the WSMV-infected wheat plants under conditions of simulated microgravity (clinorotation) which is accounted for by the inclusion of various mechanisms in the process of plant adaptation to the factors of stresses. For the first time the stages of viral infection development were elucidated under the conditions of prolonged clinostating and the presence of the WSMV was detected in wheat plants on the 18th day after inoculation employing the method of polymerase chain reaction (PCR). In the test variant with vertical clinostating (R=1,0) the least favorable conditions for viral reproduction were identified. In the same variant the increases of the total nitrogen content in the ears were observed which may be an evidence of interferon-like protein synthesis in plant similar to the interferon system functioning in animals.  相似文献

Bean golden mosaic virus (BGMV) DNA 1 and 2 have little sequence homology with maize streak virus (MSV), wheat dwarf virus (WDV), and chloris striate mosaic virus (CSMV) DNAs. BGMV DNA 1 and beet curly top virus (BCTV) DNA are closely related, whereas BGMV DNA 2 and BCTV DNA are not related. Direct amino acid homologies of predicted proteins between BGMV ORFs and MSV ORFs, WDV ORFs or CSMV ORFs were 40-50%. BGMV 1L1 and BCTV L1, and BGMV IL3 and BCTV L4 were highly conserved. The sequence TAATATTAC was detected in the loops of hairpin structures of 5 gemini-viruses.  相似文献

Southern bean mosaic virus (SBMV) is a spherical plant virus. It possesses an intrinsic (structural) anisotropy due to the orientation of valence bonds of the amino acid and/or nucleic acid residues. According to the light scattering theory of Rayleigh and Gans for optically anisotropic spheres, the degree of linear polarization of the scattered light depends on the intrinsic anisotropy, the relative refractive index of the sphere to the solvent, the scattering angle, and the inclination angle of linearly polarized incident light to the scattering plane. By using the theoretical expression of the degree of linear polarization, the intrinsic anisotropy parameter of southern bean mosaic virus in the compact spherical state was calculated with the appropriate experimental values. This novel method should be useful in elucidating the internal structure of spherical viruses and other spherical complexes of macromolecules. © 1993 John Wiley & Sons, Inc.  相似文献
