种蛋开窗技术及其对鸡胚发育影响的研究   总被引:2,自引:1,他引:1  
种蛋开窗对禽胚实施操作的技术在转基因禽类、利用多能细胞保存禽类种质资源以及禽胚实验模型等领域的研究具有重要价值。以家鸡新产蛋(含有禽胚胎干细胞)为材料,研究出了种蛋开窗切块自动复位技术,并在此基础上以接近禽胚实施操作为目的,进行了不同封口方法、不同开窗位置和添加液体排除因开窗形成的空气泡等对种蛋孵化率影响的实验。结果表明:种蛋赤道面开窗不同封口方式对孵化率影响的差异不显著,最高孵化率可达到65%;赤道面开窗,通过添加液体排除空气泡的影响不会提高孵化率,空气泡的大小与孵化率没有明显相关性;种蛋气室端开窗(针刺透内壳膜操作胚胎)方式,比赤道面开窗方式的孵化率明显提高,两种方法能见胚率均在90%以上;采用以上技术对种蛋开窗、找胚和封口,制作效率在每人每小时30个以上。本文系统地研究和报道了种蛋开窗操作禽胚简单而实用的技术,有助于解决目前种蛋开窗技术复杂且孵化率低的困境。  相似文献

为了解低强度氦氖激光对心肌细胞超微结构、超氧化物歧化酶(SOD)和丙二醛(MDA)的影响,采用2.87J/cm^2,7.17J/cm^2和28.66J/cm^2三种剂量氦氖激光照射大鼠心前区,电镜观察心肌超微结构,测定心肌组织SOD和MDA的含量。结果发现心肌线粒体有轻微损伤,心肌组织SOD含量在7.17J/cm^2剂量显著下降(P<0.05),而在28.66J/cm^2剂量显著升高(P<0.05),心肌组织MDA含量在7.17J/cm^2和28.66J/cm^2两种剂量下降有显著升高(P<0.01),在2.87J/cm^2剂量,心肌组织SOD和MDA含量均无显著变化(P<0.05)。结果提示心肌SOD含量和脂质过氧化随不同的激光剂量而变化。  相似文献

本试验采用氦氖激光(632.8nm)以不同的功率和不同功率密度,或者不同的辐照方式,辐照孔雀幼鱼(Poecilia reticulata)、尼罗罗非鱼(Oreochromis niloticus)和淡水白鲳(Ephippus orbis)。实验结果表明,He-Ne激光辐照对三种幼鱼的生长均有促进的作用,并且不同的功率和功率密度的影响是不同的,适当的激光参量会产生较佳的影响效果。  相似文献

Abstract: Expanding populations of resident Canada geese (Branta canadensis) are resulting in increased conflicts with humans. Nonlethal and humane means are needed for managing Canada goose flocks at a variety of sites, including golf courses, industrial parks, government sites, and city parks. Decreased egg production and hatching are side effects of nicarbazin, a veterinary drug used to treat coccidiosis in chickens. Capitalizing on these effects, we developed nicarbazin as a reproductive inhibitor for Canada geese and conducted a field efficacy study. We recruited study sites in 2002 and 2003. Following laboratory testing, we conducted a field efficacy trial of nicarbazin for reducing the hatchability of Canada goose eggs in spring 2004 in Oregon, USA. The study began in February 2004 at 10 sites in Oregon, with 2 control and 3 treated sites on each side of the Cascades. We fed bait daily to resident Canada geese for approximately 6 weeks. We located and monitored nests until hatching or ≥5 days beyond the expected hatching date to determine hatchability. We completed data collection in May 2004. Geese consumed 8,000 kg of bait, with 5,100 kg of OvoControl G® (Innolytics, LLC, Rancho Santa Fe, CA) 2,500-ppm nicarbazin bait consumed among 6 treated sites and 2,900 kg of untreated bait consumed among 4 control sites. We monitored 63 nests at treated sites and 46 nests at control sites to determine hatching success of eggs. There was a 62% reduction in the percentage of nests with 100% hatchability at treated sites as compared to controls. There was a 93% increase in the percentage of nests at treated sites with 0% hatchability as compared to nests with no eggs hatching at control sites. Hatchability from treated sites versus control sites was reduced 36% (F = 5.72, P = 0.0622). We submitted results from this study to support Environmental Protection Agency registration of nicarbazin as a reproductive inhibitor for use in Canada geese. We have shown that treatment of resident Canada geese with OvoControl G 2,500-ppm nicarbazin bait by licensed, trained applicators immediately prior to and during the breeding season can reduce hatchability of eggs laid by treated geese, thereby reducing recruitment of goslings into problem resident Canada goose populations.  相似文献

为探讨NO对He-Ne激光和增强UV-B辐射小麦(Triticum aestivuml)气孔运动的作用机理,采用低剂量(5 mW.mm-2)He-Ne激光和增强(10.08 kJ.m-2.d-1)UV-B辐射并结合药理学实验和激光共聚焦显微技术,对ML7113小麦的叶片及表皮条进行不同的处理,结果显示:(1)UV-B辐射既可诱导小麦叶片气孔关闭,又能够明显增加气孔保卫细胞和叶片的NO水平,且NO清除剂明显抑制了UV-B辐射诱导的小麦叶片气孔关闭,同时气孔保卫细胞和叶片内的NO含量明显减少。(2)一氧化氮合酶(NOS)抑制剂L-NAME对经UV-B辐射诱导的小麦幼苗气孔开度及保卫细胞和叶片内NO含量的抑制程度明显大于硝酸还原酶(NR)抑制剂NaN3对其的抑制程度,说明一氧化氮合酶(NOS)合成途径是小麦叶片经UV-B辐射后NO的主要产生途径。(3)就气孔开度而言,L〉CK〉BL〉B。就小麦叶片及保卫细胞内NO含量而言,B〉BL〉CK〉L。就硝酸还原酶(NR)和一氧化氮合酶(NOS)的活性而言,B组NR活性最低,NOS活性最高,L组NR活性最高,NOS活性最低。表明经He-Ne激光和增强UV-B辐射诱导的小麦气孔开度的变化确实与保卫细胞及叶片中NO含量的多少有关,气孔开度的减小及增大对应于NO含量的增多或减少,同时进一步证实了小麦叶片经He-Ne激光单独辐照后,NO的主要合成途径也来源于NOS途径。  相似文献

Summary A combined method has been developed for selective cytolysis in vitro as well as in vivo using a photosensitizer haematoporphyrin-protein conjugate as the targeting molecule and low-power He-Ne laser (632.8 nm) irradiation in order to activate the sensitizer to its excited, toxic triplet energy state. The specificity of the procedure was demonstrated in vitro by purging a mixed cell population from one component, and in vivo in an animal (nude mice) xenograft tumour model, where human cancer cells were destroyed by the immunotargeting method using monoclonal-antibody-haematoporphyrin (mAb-HP) conjugate (a-PNAr-I mAbs, which bind to the cell surface antigens of gastric cancer cells) and soft laser irradiation. The cell destruction was dependent on the doses of both mAb-HP and He-Ne laser light energy, and occurred only in target cell populations.  相似文献

本文以离体培养的Raji细胞为材料,采用ELLMAN方法检测了不同剂量的He-Ne激光对Raji细胞膜表面SH含量的影响。发现0.5J/cm^2He-Ne激光能量可明显增加膜表面SH含量(P〈0.05)。大于或小于此剂量的He-Ne激光对膜表面SH含量的影响均不明显(P〉0.05)。提示0.5J/cm^2的激光能量对膜有刺激作用。  相似文献

在水温(23±0.5)℃、pH 7.0~8.0、溶解氧>5 mg· L-1的条件下,研究孵化过程中不同盐度对拟目乌贼胚胎发育和孵化率的影响.结果表明,拟目乌贼卵透明略带奶油色,近椭圆形,短径为0.596~1.758 cm,长径为1.452~3.136 cm,刚产出的卵重约2g;孵化完成时,卵膜内的重量达到4.5g.在盐度为28‰时,经过28~30 d孵化,卵的短径和长径平均分别增加了0.601、0.615 cm,孵化率为57.0%;盐度为23‰时,卵的长径和短径也相应增加,增幅分别为0.567、1.263 cm,孵化率为13.5%;盐度为33‰时,经过28~30 d孵化,卵短径平均增长0.150 cm,而长径却缩短了0.141 cm,孵化率为9.5%;在盐度为13‰、18‰、38‰和43‰条件下,胚胎停止发育,孵化率为0.结果表明,拟目乌贼属于狭盐性种类,盐度变化对拟目乌贼胚胎发育和孵化有直接影响.  相似文献

采用低剂量(5 mW.mm-2)He-Ne激光辐照经增强UV-B辐射处理(10.08 KJ.m-2.d-1)后的‘ML7113’小麦幼苗,待小麦幼苗长7 d后,对各处理组小麦幼苗的核蛋白进行提取,通过紫外吸收法测定各个处理组幼苗核蛋白的含量。采用SDS聚丙烯酰胺凝胶电泳法(SDS-PAGE)进行电泳。结果显示:小麦幼苗经UV-B辐射后核蛋白含量明显增高;单独激光处理后,小麦幼苗核蛋白含量低于对照组;经He-Ne激光和增强UV-B辐射复合处理后,小麦幼苗核蛋白含量低于UV-B辐射处理组,而高于对照组,推测一定剂量的He-Ne激光在一定程度上抑制了增强UV-B辐射的促进作用。  相似文献

采用He-Ne激光(5 mW.mm-2)和增强UV-B(10.08 kJ.m-2.d-1)辐照‘晋麦8号’小麦幼苗,5 d后测定各处理组小麦叶片的蛋白酶、转氨酶活性以及可溶性蛋白质含量与组成的变化。结果显示,增强UV-B辐射使小麦叶片蛋白酶活性极显著升高,转氨酶活性降低,可溶性蛋白质含量极显著下降,蛋白质谱带增加;单独He-Ne激光处理使蛋白酶活性下降,转氨酶活性升高,可溶性蛋白质含量增加,对蛋白质条带影响不明显;与单独UV-B辐射相比,经He-Ne激光辐照和UV-B辐射复合处理后,蛋白酶的活性明显降低,转氨酶的活性增加,可溶性蛋白质含量增加,并且使UV-B辐射诱导出的新带减弱或消失。研究发现,增强UV-B辐射能减弱小麦幼苗蛋白代谢中正常基因的表达,但又激活了一些抗性基因的表达而诱导产生新的胁迫蛋白;一定剂量的He-Ne激光辐照可解除UV-B对小麦幼苗正常基因表达的抑制而使其蛋白质代谢加强。  相似文献

Fertile transgenic barley by particle bombardment of immature embryos   总被引:5,自引:0,他引:5  
Transgenic, fertile barley (Hordeum vulgare L.) from the Finnish elite cultivar Kymppi was obtained by particle bombardment of immature embryos. Immature embryos were bombarded to the embryonic axis side and grown to plants without selection. Neomycin phosphotransferase II (NPTII) activity was screened in small plantlets. One out of a total of 227 plants expressed the transferred nptII gene. This plant has until now produced 98 fertile spikes (T0), and four of the 90 T0 spikes analyzed to date contained the nptII gene. These shoots were further analyzed and they expressed the transferred gene. From green grains, embryos were isolated and grown to plantlets (T1). The four transgenic shoots of Toivo (the T0 plant) produced 25 plantlets as T1 progeny. Altogether fifteen of these T1 plants carried the transferred nptII gene as detected with the PCR technique, fourteen of which expressed the nptII gene. The integration and inheritance of the transferred nptII gene was confirmed by Southern blot hybridization. Although present as several copies, the transferred gene was inherited as a single Mendelian locus into the T2 progeny.  相似文献

分别采用5 mW·mm-2 He-Ne激光辐照、10.08 kJ·m-2·d-1 增强UV-B辐射及二者组合对"晋麦8号"小麦幼苗进行处理,测定各处理幼苗叶片可溶性糖、还原性糖、蔗糖合成酶(SS)、蔗糖磷酸合成酶(SPS) 以及α,β-淀粉酶的变化.研究分析He-Ne激光对增强UV-B辐射引起小麦损伤的修复效应.结果表明:He-Ne激光辐照可使UV-B辐射后小麦幼苗可溶性糖含量升高,还原性糖含量降低,SS,SPS活力降低,α,β-淀粉酶活力升高.该结果同时表明可溶性糖、还原性糖、SS、SPS、α,β-淀粉酶的变化同小麦幼苗损伤修复的能力相关,一定剂量的He-Ne激光辐照可部分修复增强UV-B对小麦幼苗糖代谢的辐射损伤.  相似文献
