Coupons of stainless steel type AISI-304 were exposed to the industrial cooling system of a petrochemical plant fed by seawater from the Guanabara Bay, Rio de Janeiro, Brazil, in order to study thein situ formation of biofilms. Bacteria, microalgae and fungi were detected on the coupons as soon as 48 h after exposure. Their respective numbers were determined at times 48, 96 and 192 h and over the following 8 weeks. Aerobic, anaerobic and sulfate-reducing bacteria were quantified according to the technique of the most probable number, and fungi by the pour plate technique. The number of microorganisms present in the forming biofilm varied over the experimental period, reaching maximal levels of 14×1011 cells cm–2, 30×1013 cells cm–2, 38×1011 cells cm–2 and 63×105 cells cm–2, respectively, for aerobic bacteria, anaerobic bacteria, sulfate-reducing bacteria and fungi, and the dynamics of this variation depended on the group of microorganisms.Bacillus sp,Escherichia coli, Serratia sp andPseudomonas putrefaciens were identified among the aerobic bacteria isolated. Additionally, microalgae and bacteria of the genusGallionella were also detected. Nonetheless, no evidence of corrosion was found on the stainless steel type AISI-304 coupons over the experimental period.  相似文献

Two families of tandemly repeated satellite DNAs were isolated from the neotropical primate Callimico goeldii (Goeldi's marmoset). One satellite, CgoA, is over 70% A+T and has a monomer length of 338 bp. The other satellite, CgoB, is 50% A+T and has a monomer length of 916 bp. Both CgoA and CgoB hybridize strongly with Callimico DNA, but not with the DNA of other new and old world primates. Based upon a neutral substitution rate of 1.5×10–9/site per year for primates, sequence data from 15 CgoA monomers indicate that the tandem array is at least 30 million years old. Since no other neotropical primate has amplified CgoA sequences, the data suggest that the ancestor of Callimico separated from the other neotropical primates at least 30 million years ago. This value is about fourfold larger than the value of 7–9 million years derived from immunological data by Sarich and Cronin (1980). Possible reasons for this discrepancy are discussed. In microbiological studies in streams, pebble samples have until now been taken out of the water following the conventional method. However, this allows the loss of microorganisms as a result of the removal of overlying water. In the present study, to minimize the loss of microorganisms, we have developed a new sampling method, called the submerged method, for collecting microorganisms living on pebbles in streams. The abundance of microorganisms on natural pebbles and artificial clay tiles, both of which had biofilms developing on their surfaces, was measured using both the conventional and submerged methods and the results from the two methods were compared. The cell densities of bacteria (0.10–14.00 × 108 cells cm–2), heterotrophic nanoflagellates (0.36–50.30 × 104 cells cm–2), and ciliates (0.071–88.27 × 102 cells cm–2) measured by the submerged method tended to be higher than those obtained by the conventional method, although there were only a few cases in which a significant difference existed between microbial abundances determined by the two methods. Also supported by microscopic observation, these results suggest the presence of planktonic and/or weakly attached microorganisms on substrate materials in streams. Significant correlations between the concentration of chlorophyll a and the cell densities of heterotrophic microorganisms and significant correlations among heterotrophic microorganisms suggest the presence of active microbial food webs in streams.  相似文献

Thermophilic Bacillus species readily attached and grew on stainless steel surfaces, forming mature biofilms of >106.0 cells/cm2 in 6 h on a surface inoculated with the bacteria. Clean stainless steel exposed only to pasteurized skim milk at 55 °C developed a mature biofilm of >106.0 cells/cm2 within 18 h. When bacilli were inoculated onto the steel coupons, 18-h biofilms were 30 m thick. Biofilm growth followed a repeatable pattern, with a reduction in the numbers of bacteria on the surface occurring after 30 h, followed by a recovery. This reduction in numbers was associated with the production of a substance that inhibited the growth of the bacteria. Variations in the environment, including pH and molarity, affected the viability of the cells. Chemicals that attack the polysaccharide matrix of the biofilm were particularly effective in killing and removing cells from the biofilm, demonstrating the importance of polysaccharides in the persistence of these biofilms. Treatment of either the biofilm or a clean stainless steel surface with lysozyme killed biofilm cells and prevented the attachment of any bacteria exposed to the surface. This suggests that lysozyme may have potential as an alternative control method for biofilms of these bacteria.  相似文献

A novel yeast species within the Metschnikowiaceae is described based on a strain from the sugarcane (Saccharum sp.) rhizoplane of an organically managed farm in Rio de Janeiro, Brazil. The D1/D2 domain of the large subunit ribosomal RNA gene sequence analysis showed that the closest related species were Candida tsuchiyae with 86.2% and Candida thailandica with 86.7% of sequence identity. All three are anamorphs in the Clavispora opuntiae clade. The name Candida middelhoveniana sp. nov. is proposed to accommodate this highly divergent organism with the type strain Instituto de Microbiologia, Universidade Federal do Rio de Janeiro (IMUFRJ) 51965T (=Centraalbureau voor Schimmelcultures (CBS) 12306T, Universidade Federal de Minas Gerais (UFMG)-70T, DBVPG 8031T) and the GenBank/EMBL/DDBJ accession number for the D1/D2 domain LSU rDNA sequence is FN428871. The identification and characterisation of Cryptosporidiumgenotypes and subtypes are fundamental to the study of cryptosporidiosis epidemiology, aiding in prevention and control strategies. The objective was to determine the genetic diversity ofCryptosporidium in samples obtained from hospitals of Rio de Janeiro, Brazil, and Buenos Aires, Argentina. Samples were analysed by microscopy and TaqMan polymerase chain reaction (PCR) assays forCryptosporidium detection, genotyped by nested-PCR-restriction fragment length polymorphism (RFLP) analysis of the 18S rRNA gene and subtyped by DNA sequencing of the gp60 gene. Among the 89 samples from Rio de Janeiro, Cryptosporidium spp were detected in 26 by microscopy/TaqMan PCR. In samples from Buenos Aires,Cryptosporidium was diagnosed in 15 patients of the 132 studied. The TaqMan PCR and the nested-PCR-RFLP detected Cryptosporidium parvum, Cryptosporidium hominis, and co-infections of both species. In Brazilian samples, the subtypes IbA10G2 and IIcA5G3 were observed. The subtypes found in Argentinean samples were IbA10G2, IaA10G1R4, IaA11G1R4, and IeA11G3T3, and mixed subtypes of Ia and IIa families were detected in the co-infections. C. hominis was the species more frequently detected, and subtype family Ib was reported in both countries. Subtype diversity was higher in Buenos Aires than in Rio de Janeiro and two new subtypes were described for the first time.  相似文献

Biofilms on biocathodes can catalyze the cathodic oxygen reduction and accordingly guarantee high cathode redox potentials. The present research assessed the use of biocathodes in full-sediment microbial fuel cells. Carbon felt-based biocathodes were evaluated in freshwater systems, and an extension of their application to brackish systems and/or stainless steel webs as base material was considered. Efficient biocathodes could be developed within days through inoculation with active microorganisms. Carbon felt was found most suited for the biocathodes in freshwater with increased performance at salinities around 80–250 mM. Maximum long-term performance reached 12.3 μW cm−2 cathode. The relative benefit of stainless steel seemed to increase with increasing salinity. A combination of stainless steel cathodes with biofilms could, however, also result in decreased electrical performance. In an efficiently catalyzing cathodic biofilm, an enrichment with an uncultured Proteobacterium—previously correlated with steel waste—was observed.  相似文献

AISI-1020 carbon steel coupons were fixed onto a water circulation loop in order to study the effect of varying NaCl concentrations on formation of biofilms by natural populations of microorganisms. Overall, we observed a reduction in the number of bacteria attached to the metal surfaces as NaCl levels increased. At 12.85 and 80 g/l NaCl, the respective bacterial counts were: 1.7×109 CFU/cm2 and 7.5×102 CFU/cm2 for aerobic species; 1.3×104 CFU/cm2 and 2.1×10 CFU/cm2 for anaerobic species; and 1.8×103 CFU/cm2 and 4.6×10 CFU/cm2 for sulfate-reducing species. However, the opposite trend was observed for the numbers of iron-reducing bacteria: 4.1×106 CFU/cm2 at 12.85 g/l NaCl and 7.5 108 CFU/cm2 at 80 g/l NaCl, respectively. Fungal counts remained constant throughout the experimental period. The salt concentration at which the maximum corrosion rate was observed was 35 g/l. In view of the marked loss of metal mass recorded at this salinity, AISI-1020 carbon steel proved to belong to the group of alloys less resistant to corrosion. A culture-independent molecular phylogenetic analysis was carried out to study for the first time the diversity of bacterial ammonia monooxygenase subunit A (amoA) and nitrogenase reductase subunit H (nifH) genes from Urca inlet at Guanabara Bay in Rio de Janeiro, Brazil. Most bacterial amoA and nifH sequences exhibited identities of less than 95% to those in the GenBank database revealing that novel ammonia-oxidizing bacteria and nitrogen-fixing microorganisms may exist in this tropical marine environment. The observation of a large number of clones related to uncultured bacteria also indicates the necessity to describe these microorganisms and to develop new cultivation methodologies.  相似文献

Summary The corrosion of AISI C1020 carbon steel in an anoxic, marine, sulphide-containing environment was examined as a function of bacterial physiology and consortial complexity. The carbon steel was exposed to three organism;Eubacterium limosum, Desulfovibrio sp. andDesulfobacter sp. which were provided with H2/CO2, butanol, glucose, and acetate as carbon and electron sources. A consortium of these bacteria utilizing hydrogen gave rise to relatively high corrosion rates (5.7×10–4 mhos cm–2) with respect to corrosion resulting from bacteria supplied with organic electron sources (0.6–1.6×10–4 mhos cm–2). Disproportionation of electrons between sulphate reduction and fermentation had a significant effect on the corrosion rate in the case ofDesulfovibrio. Surface examination using scanning electron microscopy coupled with electrochemical impedance spectroscopy supported the hypothesis that the corrosion rate was controlled by the relative intactness of a ferrous sulphide film in which the bacteria were embedded.  相似文献

Examination of 248 adult specimens of whitemouth croaker Micropogonias furnieri from five localities along the Brazilian coast revealed 8735 parasites belonging to 41 metazoan species. Samples from Ceará to Bahia and Rio de Janeiro to Santa Catarina showed a high level of correct assignation (92 and 87%, respectively) and cross assignation (i.e. almost all specimens misidentified in Ceará were assigned to Bahia and almost all specimens misidentified in Bahia were classified as Ceará), so samples were pooled in the northern and south‐eastern samples, and Rio Grande do Sul was considered a southern area. Eight parasite species were characteristic of the northern localities, five species were found just in the area associated with south‐eastern localities and two species were characteristic of the southern area providing first evidence of stock discreteness. The multivariate discriminant analysis successfully discriminated three groups of localities associated with three stocks of M. furnieri in Brazil: a northern stock associated with Ceará and Bahia, a south‐eastern stock related to Rio de Janeiro and Santa Catarina and a southern stock in the area of Rio Grande do Sul, which could be considered as the northern limit of the stock associated with the Common Fishing Zone of Uruguay and Argentina.  相似文献

The brown seaweed Sargassum vulgare is abundant along the coast of Rio de Janeiro state. An investigation of the spatial variation of antifouling activity was conducted, in which algae were collected at five locations along the coast of Rio de Janeiro during October 2008. Hexane, dichloromethane, methanol and acetone/water extracts were prepared and screened for their bioactivities against the growth of five strains of marine fouling bacteria, five biofilm-associated microphytobenthic strains and attachment of the mussel Perna perna. The most active were the hexane extracts from Bananal algae that inhibited the growth of all microalgae tested; the methanol and dichloromethane extracts from Mar do Norte, which inhibited Vibrio aestuarianus and Pseudoalteromonas elyakovii and the polyphenol extracts from Ilha de Itacuruçá and Bananal that inhibited mussel attachment, respectively, by 64% and 71% compared to controls.  相似文献

Laboratory model systems were developed for studying Shewanella putrefaciens adhesion and biofilm formation under batch and flow conditions. S. putrefaciens plays a major role in food spoilage and may cause microbially induced corrosion on steel surfaces. S. putrefaciens bacteria suspended in buffer adhered readily to stainless steel surfaces. Maximum numbers of adherent bacteria per square centimeter were reached in 8 h at 25°C and reflected the cell density in suspension. Numbers of adhering bacteria from a suspension containing 108 CFU/ml were much lower in a laminar flow system (modified Robbins device) (reaching 102 CFU/cm2) than in a batch system (reaching 107 CFU/cm2), and maximum numbers were reached after 24 h. When nutrients were supplied, S. putrefaciens grew in biofilms with layers of bacteria. The rate of biofilm formation and the thickness of the film were not dependent on the availability of carbohydrate (lactate or glucose) or on iron starvation. The number of S. putrefaciens bacteria on the surface was partly influenced by the presence of other bacteria (Pseudomonas fluorescens) which reduced the numbers of S. putrefaciens bacteria in the biofilm. Numbers of bacteria on the surface must be quantified to evaluate the influence of environmental factors on adhesion and biofilm formation. We used a combination of fluorescence microscopy (4′,6′-diamidino-2-phenylindole staining and in situ hybridization, for mixed-culture studies), ultrasonic removal of bacteria from surfaces, and indirect conductometry and found this combination sufficient to quantify bacteria on surfaces.  相似文献

Biofilm formation on metallic surfaces in marine and freshwater environments often precedes corrosion and other biofouling conditions. Attachment is mediated by such environmental factors as the presence of surface conditioning films, fluid dynamics, bulk-phase nutrient levels, and surface chemistry. In this study, we utilized a Fowler Cell Adhesion Measurement Module to demonstrate that the changes in cellular concentration and composition of a monoculture ofPseudomonas atlantica biofilms on stainless steel were a function of the applied shear force. At shear forces in the range of 3–10 dynes cm–2 (1.0 liter min–1), attachment as measured by acridine orange direct microscopic counts was greatest at the higher shear forces.14C-Acetate uptake activity on the stainless steel surfaces increased with shear stress. Acetate incorporation ranged from 1×10–5 to 19×10–5 mol cm–2 between 0.15 and 30 dynes cm–2 for 30 min uptake periods. On a per cell basis, however, activity decreased with shear, indicating a shift in metabolism. Fourier transform infrared spectroscopy revealed that protein and carbohydrate concentrations also increased with the applied shear. Increased biofilm CN ratios and total fatty acids were associated with the higher shear stresses. Neither radius of interaction nor biofilm age appeared to significantly influence the relationship between fluid shear and attachment and cellular composition ofP. atlantica biofilms in the range of 1–10 dynes cm–2.  相似文献

Although Rio de Janeiro is one of the best sampled states of Brazil for bats, there are still some poorly studied islands on its coast. Hence, we inventoried the bat species of an island on the southwestern coast of Rio de Janeiro, compared five sites sampled within this island, and compared its bat community with seven other sites in the same region. We carried out sampling in July 1995 and from September 2006 to August 2010, comprising 58 samplings. We captured a total of 1502 individuals of 25 bat species with mist nets set far from roosts, or close to or inside roosts. The most abundant species were Artibeus lituratus, Molossus molossus, and Carollia perspicillata. Total sampling effort was 89,400 m2 h. The present study was one out of five inventories carried out in the southwestern coast of Rio de Janeiro that registered more than 1000 bat individuals from more than 24 bat species. Long-term inventories on islands are still needed, because they can result in a large number of species and captures as observed in the present study.  相似文献

Callimico goeldii (Goeldi's marmoset) is a neotropical primate with 2n=47,X1X2Y in the male, and 2n=48,X1X1X2X2 in the female, due to a Y-autosome translocation. Karyological comparisons of Callimico, Callithrix jacchus and Cebus apella suggest that Callimico is a member of the Callitrichidae. Isozyme data and restriction mapping of LINE-1 repetitive elements in these species and in a variety of other neotropical primates confirm these findings and supply strong evidence for including Callimico in the Callitrichidae. Pulsed light is a nonthermal processing technology recognized by the FDA for killing microorganisms on food surfaces, with cumulative fluences up to 12 J cm−2. In this study, we investigated its efficacy for inactivating murine norovirus 1 (MNV-1) as a human norovirus surrogate in phosphate-buffered saline, hard water, mineral water, turbid water, and sewage treatment effluent and on food contact surfaces, including high-density polyethylene, polyvinyl chloride, and stainless steel, free or in an alginate matrix. The pulsed-light device emitted a broadband spectrum (200 to 1,000 nm) at a fluence of 0.67 J cm−2 per pulse, with 2% UV at 8 cm beneath the lamp. Reductions in viral infectivity exceeded 3 log10 in less than 3 s (5 pulses; 3.45 J cm−2) in clear suspensions and on clean surfaces, even in the presence of alginate, and in 6 s (11 pulses; 7.60 J cm−2) on fouled surfaces except for stainless steel (2.6 log10). The presence of protein or bentonite interfered with viral inactivation. Analysis of the morphology, the viral proteins, and the RNA integrity of treated MNV-1 allowed us to elucidate the mechanisms involved in the antiviral activity of pulsed light. Pulsed light appeared to disrupt MNV-1 structure and degrade viral protein and RNA. The results suggest that pulsed-light technology could provide an effective alternative means of inactivating noroviruses in wastewaters, in clear beverages, in drinking water, or on food-handling surfaces in the presence or absence of biofilms.  相似文献

Biofouling and microbiologically influenced corrosion are processes of material deterioration that originate from the attachment of microorganisms as quickly as the material is immersed in a nonsterile environment. Stainless steels, despite their wide use in different industries and as appliances and implant materials, do not possess inherent antimicrobial properties. Changes in hygiene legislation and increased public awareness of product quality makes it necessary to devise control methods that inhibit biofilm formation or to act at an early stage of the biofouling process and provide the release of antimicrobial compounds on a sustainable basis and at effective level. These antibacterial stainless steels may find a wide range of applications in fields, such as kitchen appliances, medical equipment, home electronics, and tools and hardware. The purpose of this study was to obtain antibacterial stainless steel and thus mitigate the microbial colonization and bacterial infection. Copper is known as an antibacterial agent; in contrast, niobium has been demonstrated to improve the antimicrobial effect of copper by stimulating the formation of precipitated copper particles and its distribution in the matrix of the stainless steel. Thus, we obtained slides of 3.8% copper and 0.1% niobium alloyed stainless steel; subjected them to three different heat treatment protocols (550°C, 700°C, and 800°C for 100, 200, 300, and 400 hours); and determined their antimicrobial activities by using different initial bacterial cell densities and suspending solutions to apply the bacteria to the stainless steels. The bacterial strain used in these experiments was Escherichia coli CCM 4517. The best antimicrobial effects were observed in the slides of stainless steel treated at 700°C and 800°C using an initial cell density of approximately 105 cells ml−1 and phosphate-buffered saline as the solution in which the bacteria came into contact with copper and niobium–containing steel.  相似文献

Short-term spatial and temporal heterogeneity of oxygen dynamics and net primary production were studied in a tree day diurnal variation at a tidal tropical salt flat in the estuarine system of Sepetiba/Guaratiba coastal plain, Rio de Janeiro, Brazil. Oxygen concentrations were measured in situ with high temporal and spatial resolution oxygen microsensors. The results showed a remarkable heterogeneity of both oxygen penetration depth (from 0.18 to 0.85 cm) and net primary production (from −0.085 to 0.115 μmol O2 cm−2 s−1) at different stations and sampling periods. Fast variations in abiotic factors like salinity and light due to the variable rainy weather were possibly the drivers of the high heterogeneity. In conclusion, short-term temporal changes could have a remarkable influence in sediment microalgae primary production. Not considering these changes can lead to wrong conclusions concerning the role and importance of sediment microalgae on tidal salt flats.  相似文献

Culture and molecular-based techniques were used to characterize bacterial diversity in the cooling water system of a fast breeder test reactor (FBTR). Techniques were selected for special emphasis on sulphate-reducing bacteria (SRB). Water samples from different locations of the FBTR cooling water system, in addition to biofilm scrapings from carbon steel coupons and a control SRB sample were characterized. Whole genome extraction of the water samples and SRB diversity by group specific primers were analysed using nested PCR and denaturing gradient gel electrophoresis (DGGE). The results of the bacterial assay in the cooling water showed that the total culturable bacteria (TCB) ranged from 103 to 105 cfu ml?1; iron-reducing bacteria, 103 to 105 cfu ml?1; iron oxidizing bacteria, 102 to 103 cfu ml?1 and SRB, 2–29 cfu ml?1. However, the counts of the various bacterial types in the biofilm sample were 2–3 orders of magnitude higher. SRB diversity by the nested PCR-DGGE approach showed the presence of groups 1, 5 and 6 in the FBTR cooling water system; however, groups 2, 3 and 4 were not detected. The study demonstrated that the PCR protocol influenced the results of the diversity analysis. The paper further discusses the microbiota of the cooling water system and its relevance in biofouling.  相似文献
