利用改良的SMA培养基富集培养从植物的落花上分离筛选出的产β-胡萝卜素的菌株SC-2。该菌株在以棉籽饼粉为主要原料的培养基上生长良好。最高生物量达54mg干菌体/ml培养液。胡萝卜素含量1.85mg/g干菌体。HPLC测定β-胡萝卜素占总胡萝卜素的90.5%。  相似文献

微生物生产弹性蛋白酶的研究   总被引:3,自引:0,他引:3  
从10个不同省区采集的500多个土样中,分离得到了多株产弹性蛋白酶的菌株,主要集中在黄杆菌属和色杆菌属中。经过细胞工程技术处理后,黄杆菌属中的F-122菌在摇瓶发酵中的产酶能力达到240u/ml,比出发菌株提高了1.5倍。该酶在Tris—HCl缓冲液中最适pH为8.0,最适酶解温度43℃。在pH6.0—9.5范围内稳定。CuSO4、ZnSO4和FeSO4强烈抑制酶的活性,NaCl、KCl对酶活影响微弱。60℃,30min酶活完全丧失。  相似文献

本文建立了检测人C蛋白的双抗夹心ELISA方法。该方法的最佳实验条件为:-抗浓度5-8.5μg/ml;二抗密度为1:200-1:1000倍稀释。制作了C蛋白浓度与OD值的关系的工作曲线,指出了用内插法测定C蛋白的最佳浓度范围为0-100mg/ml之间。  相似文献

茶儿茶素氧化产物体外清除·OH自由基作用的研究   总被引:6,自引:1,他引:5  
本文采用2-脱氧-D-核糖(DR)法产生.OH,测定了不同浓度的茶儿素氧化产物A及C对.OH的清除作用。结果表明,在一定的浓度范围内,氧化产物A及C均有很强的清除.OH的作用且最佳清除浓度为200μg/ml。在茶多酚、氧化产物A及C三种物质中,以产物C的清除作用最强,其IC50值为7.3μg/ml,其次是A,IC50为10.1μg/ml,最后是茶多酚,IC50值为70μg/ml。  相似文献

本文用化学发光法检测HMME的’O2和ROS产量,并与HpD相比较,同时观察HMME光敏效应与浓度、激光照射功率密度、照射时间、激光波长的关系。结果显示:HMME浓度在2.5-40μg/ml范围内,其光敏效应随浓度的增加而呈线形上升,以后便略下降;578.2nm激光以10mW/cm^2照射2.5μg/ml,HMME的’O2和ROS产量是HpD的8倍;HMME光敏效应与激光照射的功率密度密切相关。各  相似文献

用化学发光法研究了天竺葵花粗担液对超氧阴离子自由基(O2)^=和羟自由基(OH)的清除作用,用分光不镀法研究了它对1,1-二苯基-2-苦肼基自由基(DPPH0的清除作用。结果表明,天竺葵花具有很强的清除自由基活性。其粗提物清除O2^-分别为3.5μg/ml(红花),3.0μg/ml(粉花),1.6μg/ml(浅粉花),与茶多酚(IC50=1.1μg/ml)相近,清除OH的IC50分别为5.7μg/  相似文献

通过对发酵基质和发酵关键参数的优化,确定了发酵培养基的磷酸盐浓度为0.15M,甘油浓度为1.2ml/L,补料中甘油浓度为20ml/L,发酵过程中溶解氧控制在30%~60%,pH控制在6.85左右。在5L在NBS-Bioflo3000型自动控制发酵罐中采取加速补料的补料分批培养,重组大肠杆菌YK537/pSB-TK经10h30°C培养和5h42°C诱导培养,最终密度达到60OD600,rhTNFα-DK2的表达量占菌体总蛋白的50%以上,每升发酵液纯化可得到近2g的rhTNFα-DK2。  相似文献

对采自全国各地的50份上样进行筛选,获得一株产酶活性较高的菌株B-9545,通过摇瓶培养确定了产酶的最适培养基Ⅱ(g/100ml):复合蛋白胨1.0;玉米淀粉1.5;玉米浆1.5;NaC10.5;K2HpO40.1;MgSO40.02;CaCO30.5;Tween800.1;pHg—10,在该条件下,30℃荡培养48h,酶活力可达596u/ml。对菌株B—9545的产酶特性研究表明:该酶对高pH环境有较强的适应性,因此,可用于碱性洗涤用酶。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. We also discuss the development and practicality of various cell culture and animal model systems to study KSHV replication and pathogenesis.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
