Newly synthesized histones have been extracted from Rana pipiens oocytes or cleaving embryos previously injected with [3H]lysine or [3H]arginine. The radioactive proteins were fractionated by cation-exchange chromatography and electrophoresis on acid/urea or SDS-polyacrylamide gels; histones were identified by coelectrophoresis with authentic markers. From percentage total incorporation in the putative histones, and absolute rates of lysine or arginine incorporation, rates of histone synthesis were estimated. Rates of histone synthesis in two-cell embryos were at least 10-fold higher than in maturing oocytes. Between the two-cell and blastula stages, the rate increased an additional threefold, from about 1200 pg hr?1 per embryo to about 4500 pg hr?1 per embryo. While all histone classes are synthesized during cleavage, synthesis of the various classes is not coordinated; histones are not synthesized in the same relative proportions at which they are found in blastula chromatin. The synthesis of histone H4 in particular is barely detectable during cleavage. This, and other observations, suggested the existence of cytoplasmic histone pools. In approaching the possible existence of histone pools, the amount of H4 present in oocytes was determined. Oocytes contain about 74 ng of H4, an amount sufficient to allow development to the blastula stage. The synthesis and activity of tyrosinase during development of the frog Rana pipiens 总被引:2,自引:0,他引:2
We have established by radioimmunoprecipitation that tyrosine-DOPA oxidase (TDO, tyrosinase) [EC 1.14.18.1] is first synthesized by frog embryos at the early neurula stage soon after embryonic induction of the neural plate by the underlying chordamesoderm. The DOPA moiety of the enzyme, at the time of its first appearance, is almost inactive enzymatically and can be activated by mild proteolysis (with trypsin). A very large increase in the amount of active DOPA oxidizing enzyme (without trypsinization) is observed at hatching (stage 21), and this is accompanied by melanin deposition in pigment cells. The tyrosine moiety of the enzyme is also partially inactive at the time of first synthesis, but the ratio of active to inactive enzyme remains approximately constant throughout early development. DOPA decarboxylase enzymatic activity is first detected at neurula stage, and this activity is accompanied by the first appearance of catechol amines. 相似文献

The cells that line the lumen of rabbit aorta have been obtained in a quantity sufficient for mass culture. Autoradiography of the cells incubated with 35S-sulfuric acid and electrophoretic analysis of 35S mucopolysaccharides extracted from the cells and from the medium indicate that these cells synthesize and secrete various species of sulfated mucopolysaccharides. In the steady state, only one major and one minor component are found inside the cell. The major intracellular component is resistant to degradation by chondroitinase ABC. Two major fractions and one minor fraction are found in the culture medium at the end of a 24 h labelling period. Both of the major components extracted from the medium are not present in the cell in an appreciable amount. 相似文献

Changes in the activity of the tRNA methyltransferases have been found in all differentiating systems studied. Activity was examined in extracts of Rana pipiens embryos and in larval and adult liver by in vitro assay using S-adenosyl-l-[methyl-14C]methionine as the methyl donor. Specific activities of tRNA methyltransferases decreased, beginning with the time of feeding, when using high concentrations of the crude liver enzyme. A new methyltransferase activity, glycine N-methyltransferase, appeared at the time of feeding. Apparently, the glycine methyltransferase is active before the onset of any of the characteristic metamorphic changes of other liver enzymes. Using partially purified enzyme from adult liver, the Km of glycine methyltransferase for S-adenosylmethionine is 0.3 mM and the Ki for S-adenosylhomocysteine, a competitive inhibitor, is 0.08 mM. 相似文献

This investigation is concerned with the role played by irradiated sperm in the subsequent embryonic development of the frog Rana pipiens. The ultraviolet irradiation of the sperm was followed by the fertilization of untreated eggs. Determinations included mitotic index, nuclear diameter, chromosomal constitution, and percentage survival. Results show that the low-dose embryos were the most severely affected ones in spite of their diploid constitution. Embryos resulting from higher dosages of sperm irradiation were all haploid but showed less severe growth and cytological abnormalities. It is conjectured that radiation-induced damage to the sperm also involves its protamine moiety since considerable damage can occur without any alteration in the chromosomal constitution of the low-dose embryos. 相似文献

Summary In early diplotene frog oocytes incubated to illustrate thiamine pyrophosphatase (TPPase) activity, reaction product is uniformly distributed within the compartments of the endoplasmic reticulum and nuclear envelope as well as within the saccules and small vesicles comprising the dictyosomes. With continued oocyte development the reaction product becomes concentrated in localized regions of the dictyosome saccules. Eventually, the enzyme is no longer apparent within the endoplasmic reticulum, but is concentrated in the cisternae of the inner dictyosome saccules. The variations noted suggest that the enzyme is synthesized early in diplotene by the endoplasmic reticulum and is subsequently transported to the Golgi apparatus where it is consistently observed at later developmental stages. TPPase activity is also present in the Golgi apparatus of follicle and theca cells as well as in ovarian epithelial cells. The enzyme is also detected in micropinocytotic vesicles contained within the cells comprising the follicle envelope and in intercellular spaces of the follicle. Horseradish peroxidase injected into the coelomic cavity is transported via micropinocytotic vesicles into and through the cells comprising the follicle envelope and in intercellular spaces. The exogenous protein is not found even after a prolonged time period in early diplotene oocytes. The protein is, however, present in large spherical and tubular vesicles in the cortex of vitellogenic oocytes approximately 500 microns in diameter. DNA-dependent RNA polymerase was extracted from oocytes of the frog, Rana pipiens. The bulk of the enzyme activity was present in the germinal vesicle and the amounts of each major form of such activity did not significantly change during oocyte maturation. Therefore, either nuclear polymerase activity is conserved after breakdown of the oocyte nucleus during maturation or, alternatively, de novo synthesis of the enzymes must occur during oocyte maturation concomitant with degradation. We have measured rates of protein synthesis in oocytes and determined a maximum rate of synthesis for RNA polymerases. Our kinetic studies show that no more than 20, 10, and 5% of RNA polymerases type I, IIa, and IIb, respectively, could be synthesized during steroid-induced oocyte maturation. These results thus show that the bulk of RNA polymerase accumulates in the germinal vesicle during oogenesis, is dispersed into the cytoplasm during maturation, and, since only limited synthesis seems to be occurring, the polymerase is available during embryogenesis. 相似文献
