The phospholipid composition of 8 Escherichia coli strains differing in their capacity for survival in the air with a relative humidity of 30% has been studied. The study has revealed that, irrespective of the phase of growth and the nature of the culture medium, the capacity of E. coli cells for survival in the air is related to their phospholipid composition, this capacity being the higher, the greater the content of total phospholipids and cardiolipin and the lower the concentration of phosphatidyl glycerine.  相似文献

Al toxicity in cultured tobacco cells (Nicotiana tabacum L. cv Samsun; nonchlorophyllic cell line SL) has been investigated in nutrient medium. In this system, Al and Fe(II) (ferrous ion) in the medium synergistically result in the accumulation of both Al and Fe, the peroxidation of lipids, and eventually death in cells at the logarithmic phase of growth (+P cells). A lipophilic antioxidant, N,N[prime]-diphenyl-p-phenylenediamine, protected +P cells from the peroxidation of lipids and from cell death, suggesting that a relationship exists between the two. Compared with +P cells, cells that had been starved of Pi (-P cells) were more tolerant to Al, accumulated 30 to 40% less Al and 70 to 90% less Fe, and did not show any evidence of the peroxidation of lipids during Al treatment. These results suggest that -P cells exhibit Al tolerance because their plasma membranes are protected from the peroxidation of lipids caused by the combination of Al and Fe(II). It seems likely that the exclusion of Fe from -P cells might suppress directly Fe-mediated peroxidation of lipids. Furthermore, since -P cells accumulated [beta]-carotene, it is proposed that this carotenoid pigment might function as a radical-trapping antioxidant in the plasma membrane of cells starved of Pi.  相似文献

A new approach to the analysis of the oxidative processes participated in regulation of metabolism in norm and the assessment of the biological consequences under the action of damaging factors of different nature and intensity for animal groups is suggested using the analysis of interrelations between the different parameters of the physicochemical regulatory system of the lipid peroxidation in tissues of the laboratory mice (the phospholipid composition, amounts of the oxidation products in lipids) by changes of the scale and direction of interrelations between the reciprocal parameters of the lipid peroxidation system in norm.  相似文献

The effects of a mixture of unsaturated fatty acids (UFA) on changes in the lipid oxidation substrate, a system of natural antioxidants and the functioning of a system controlling lipid peroxidation were studied. It was shown that arachidonate induces the incorporation of fatty acids into individual lipid fractions and the dilution of lipids by UFA esters, as a result of which the relative amount of natural antioxidants diminishes. Under these conditions, the oxidative capacity of lipids decreases, which manifests itself in a reduction of the rate of natural antioxidants utilization and an increase in their number. It was shown that purposeful modification of the lipid substrate does not interfere with the interaction between individual parameters of the system controlling lipid peroxidation.  相似文献

It has been established experimentally that the character and the scale of interrelations between various parameters of the phospholipid composition and their group-average values depend significantly on the characteristics of lipids (the antioxidative activity, quantity of peroxides, and antiperoxide activity) of animal liver and blood erythrocytes. It was shown that a single injection of low-toxicity Tween 80 and acetone at low doses modifies the intensity of lipid peroxidation in the liver and changes the physicochemical properties of lipids, and the interrelations between the parameters that are coordinated in norm, thereby causing the disturbance of biochemical functions of the liver.  相似文献

The metabolites 2-octaprenyl-1,4-hydroquinone (1) and 2-(24-hydroxy)-octaprenyl-1,4-hydroquinone (2), isolated from the sponge Ircinia spinosula, along with a series of synthetic derivatives, were evaluated for their antioxidant capacity, in order to establish a potential relationship between structural characteristics and antioxidant activity. The antioxidant potential of both natural and synthesised compounds was evaluated in vitro by their ability: (1) to interact with the stable free 1,1-diphenyl-2-picrylhydrazyl radical (DPPH) and (2) to inhibit the peroxidation, induced by the Fe(++)/ascorbate system, of heat inactivated hepatic microsomal membrane lipids. Metabolite 1 presented a strong interaction with DPPH and had a moderate effect on lipid peroxidation, while metabolite 2 interacted extensively with DPPH and exhibited a significant effect against lipid peroxidation. All derivatives retaining the free 1,4-hydroquinone system maintained fully or partly the free radical scavenging capacity.  相似文献

Erythrocytes isolated from 131 cases of Non-Insulin Dependent Diabetes Mellitus (NIDDM) were studied for lipid peroxidation, antioxidant defences, and the maximum peroxidisable substrate in the cell membrane. Antioxidant defences are lowered in NIDDM, followed by significant rise in lipid peroxidation products. However, in the erythrocyte membrane, the total polyunsaturated peroxidisable lipids are lower than in normal erythrocytes which may be a causative factor affecting the survival of the cells.  相似文献

The effect of diamide on the physicochemical state of proteins and lipids of human erythrocyte membrane was studied. It was found that diamide at a concentration of 1 mM decreases the content of the SH-groups of membrane proteins by approximately 50%, resulting in enhanced vesiculation of erythrocytes upon metabolic exhaustion of cells. It was shown using fluorescein isothiocyanate-labeled concanavalin A and 4,4'-diisothiocyano-2,2'-stilbene disulfonate that diamide changes the structural state of the main integral protein of erythrocyte membranes, the band 3 protein. Changes in the microviscosity of the membrane lipid bilayer depending on diamide concentration were determined from the changes in the fluorescence parameters of the lipophilic probes (pyrene and 1,6-diphenyl-3,5-hexatriene). The level of lipid peroxidation products in membranes remained unchanged. It follows from these data that the SH-oxidizing agent diamide does not directly interact with the lipid bilayer of membrane and produces changes in the physicochemical state of lipids presumably by disrupting protein-lipid interactions that take place upon oxidation of the SH-groups and cross-linking of membrane proteins.  相似文献

Mouse fibroblast LM cells have been modified with respect to their phospholipid composition in all subcellular fractions, including the nuclear membrane. The content of polyunsaturated fatty acids (PUFA) was significantly increased but no difference in cell survival after X-irradiation could be observed between the normal and PUFA enriched cells. It is concluded that the radiosensitive PUFAs in the membranes are well protected against radiation damage. This protection of the PUFA cells could not be caused by vitamin E, because this membrane protector was not present in these fibroblasts. The content of glutathione (GSH) was about the same in the normal and the modified cells. Reduction of the cellular GSH content to less than 5 per cent of that for non-treated cells did not alter cellular survival after radiation of either normal or PUFA enriched cells under oxic or anoxic conditions. The radiosensitive lipids present in the membranes of the PUFA enriched cells proved to be vulnerable to radiation-induced lipid peroxidation when extracted from the cells and reconstituted into liposomes, indicating that the fatty acids per se are peroxidizable. It is concluded that the lipids in the membranes of mammalian cells are not the principal target in radiation-induced reproductive death, and that no generalization is permitted with respect to glutathione, as being the major hydrogen donating species in mammalian cells responsible for the repair of those target molecules responsible for cell survival after radiation.  相似文献

Lipid droplets (LDs) are ubiquitous and physiologically active organelles regulating storage and mobilization of lipids in response to metabolic demands. Among the constituent LD neutral lipids, such as triacylglycerols, cholesterol esters, and free fatty acids, oxidizable polyunsaturated molecular species may be quite abundant, yet the structural and functional roles of their oxidation products have not been studied. Our previous work documented the presence of these peroxidized species in LDs. Assuming that hydrophilic oxygen-containing functionalities may markedly change the hydrophobic/hydrophilic molecular balance, here we utilized computational modeling to test the hypothesis that lipid peroxidation causes redistribution of lipids between the highly hydrophobic core and the polar surface (phospho)lipid monolayer—the area enriched with integrated enzymatic machinery. Using quantitative liquid chromatography/mass spectrometry, we characterized molecular speciation of oxTAGs in LDs of dendritic cells in cancer and hypoxic trophoblasts cells as two cellular models associated with dyslipidemia. Among the many types of oxidized lipids identified, we found that oxidatively truncated forms and hydroxyl derivatives of TAGs were the prevailing oxidized lipid species in LDs in both cell types. Using coarse-grained molecular dynamics (CG-MD) simulations we established that lipid oxidation changed their partitioning whereby oxidized lipids migrated into the outer monolayer of the LD, where they can affect essential metabolic pathways and undergo conversions, possibly leading to the formation of oxygenated lipid mediators.  相似文献

In nearly every living organism, metabolites derived from lipid peroxidation, the so-called oxylipins, are involved in regulating developmental processes as well as environmental responses. Among these bioactive lipids, the mammalian and plant oxylipins are the best characterized, and much information about their physiological role and biosynthetic pathways has accumulated during recent years. Although the occurrence of oxylipins and enzymes involved in their biosynthesis has been studied for nearly three decades, knowledge about fungal oxylipins is still scarce as compared with the situation in plants and mammals. However, the research performed so far has shown that the structural diversity of oxylipins produced by fungi is high and, furthermore, that the enzymes involved in oxylipin metabolism are diverse and often exhibit unusual catalytic activities. The aim of this review is to present a synopsis of the oxylipins identified so far in fungi and the enzymes involved in their biosynthesis.  相似文献

The effect of various fatty acids on lipid peroxidation of liver microsomes induced by different methods in vitro was studied using oxygen uptake and malonaldehyde (MDA) production. It was observed that fatty acids with a single double bond are effective inhibitors of peroxidation. Stereo and positional isomers of oleic acid were equally effective as oleic acid. There was an absolute requirement for a free carboxyl group, since methyl esters of fatty acids and long-chain saturated and unsaturated hydrocarbons could not inhibit peroxidation. Saturated fatty acids with a chain length of 12-16 carbon atoms showed inhibition, whereas more than 18 carbon atoms reduced the inhibitory capacity. Fatty acids of lower chain length such as capric and caprylic acids did not show inhibition. Fatty acid inhibition was partially reversed by increasing the concentration of iron in the system. Peroxidation induced by methods which were independent of iron was not inhibited by fatty acids. It was observed that intestinal microsomes which were resistant to peroxidation due to the presence of nonesterified fatty acids in their membrane lipids were able to peroxidise by methods which do not require iron. These results suggest that certain fatty acids inhibit peroxidation by chelating available free iron. In addition, they may also be involved in competing with the esterified fatty acids in the membrane lipids which are the substrates for peroxidation.  相似文献

Membrane lipids play important roles in the regulation of cell fate, including the execution of ferroptosis. Ferroptosis is a non-apoptotic cell death mechanism defined by iron-dependent membrane lipid peroxidation. Phospholipids containing polyunsaturated fatty acids (PUFAs) are highly vulnerable to peroxidation and are essential for ferroptosis execution. By contrast, the incorporation of less oxidizable monounsaturated fatty acids (MUFAs) in membrane phospholipids protects cells from ferroptosis. The enzymes and pathways that govern PUFA and MUFA metabolism therefore play a critical role in determining cellular sensitivity to ferroptosis. Here, we review three lipid metabolic processes—fatty acid biosynthesis, ether lipid biosynthesis, and phospholipid remodeling—that can govern ferroptosis sensitivity by regulating the balance of PUFAs and MUFAs in membrane phospholipids.  相似文献

The highly conserved self-degradation pathway known as autophagy plays important roles in regulating T lymphocyte homeostasis. Recently, we found that T lymphocytes lacking the autophagy-related gene Atg5 or Atg7 have defective survival and contain expanded mitochondria and endoplasmic reticulum (ER); however, whether these defects are caused by impaired autophagy or by defects in their autophagy-independent signaling capacity of Atg5 or Atg7 in T lymphocytes remains unknown. Furthermore, the function of the microtubule-associated protein L chain 3 (LC3) conjugation system in T lymphocytes remains unclear. To address these questions, we generated conditional knockout mice with specific deletion of Atg3, a ubiquitin enzyme E2-like molecule involved in the LC3 conjugation system, in T lymphocytes. Atg3-deficient T lymphocytes displayed a phenotype similar to those of Atg7- and Atg5-deficient T cells. The survival of Atg3-deficient naive CD4(+) and CD8(+) T cells was defective. Furthermore, the mitochondria and ER were expanded in Atg3-deficient T cells. Interestingly, mitochondrial and ER content did not change instantly upon inducible deletion of Atg3 in mature T lymphocytes in vitro. Instead, it began to expand 10 d after inducible deletion of Atg3 in mature T lymphocytes, and mitochondrial content continued to increase on day 18. Cell death began to increase 24 d after inducible deletion of Atg3. These data show that the LC3 conjugation system is essential for autophagy in T lymphocytes. Our data suggest that autophagy promotes T lymphocyte survival by regulating organelle homeostasis and that the decreased survival of autophagy-deficient T cells is due to the temporal accumulation of these autophagy-related defects.  相似文献

PURPOSE OF REVIEW: Cellular uptake of plasma lipids is to a large extent mediated by specific membrane-associated proteins that recognize lipid-protein complexes. In the kidney, the apical surface of proximal tubules has a high capacity for receptor-mediated uptake of filtered lipid-binding plasma proteins. We describe the renal receptor system and its role in lipid metabolism in health and disease, and discuss the general effect of the diseased kidney on lipid metabolism. RECENT FINDINGS: Megalin and cubilin are receptors in the proximal tubules. An accumulating number of lipid-binding and regulating proteins (e.g. albumin, apolipoprotein A-I and leptin) have been identified as ligands, suggesting that their receptors may directly take up lipids in the proximal tubules and indirectly affect plasma and tissue lipid metabolism. Recently, the amnionless protein was shown to be essential for the membrane association and trafficking of cubilin. SUMMARY: The kidney has a high capacity for uptake of lipid-binding proteins and lipid-regulating hormones via the megalin and cubilin/amnionless protein receptors. Although the glomerular filtration barrier prevents access of the large lipoprotein particles to the proximal tubules, the receptors may be exposed to lipids bound to filtered lipid-binding proteins not associated to lipoprotein particles. Renal filtration and receptor-mediated uptake of lipid-binding and lipid-regulating proteins may therefore influence overall lipid metabolism. The pathological mechanisms causing the pronounced atherosclerosis-promoting effect of uremia may involve impairment of this clearance pathway.  相似文献

Lipid peroxidation in rat uterus has been studied using NADPH- and ascorbate-induced systems. Lipid peroxidation in rat uterus is low as compared to rat liver. Uterus is more sensitive to ascorbate-induced lipid peroxidation than that induced by NADPH. Uterus contains lower amounts of phospholipids and has a lesser degree of unsaturation in lipids. Co-factor studies show that Fe2+ is more important for ascorbate-induced lipid peroxidation. Endometrium is more sensitive to ascorbate-induced lipid peroxidation than myometrium. It also contains more total lipids and phospholipids besides having a higher degree of unsaturation in the lipids as compared to myometrium. Among the subcellular fractions, mitochondria are more prone to ascorbate-induced lipid peroxidation, whereas microsomes are more sensitive to NADPH-induced lipid peroxidation. Uteri from old rats (24 months) and pregnant rats are more resistant to lipid peroxidation than those from 3-month-old control rats. Uterus of pregnant rats contains more factors which inhibit lipid peroxidation and also has a lesser degree of unsaturation in lipids compared with uterus of control rats. The possible consequences of the resistance of uterus to lipid peroxidation, especially during pregnancy and senescence, are discussed.  相似文献

The isolated hepatocytes were incubated in the medium, containing cadmium chloride or hydrogen peroxide. Influence of the latter on the intensity of lipid peroxidation and contents of some lipids fractions, as well as viability of hepatocytes in these conditions has been studied. It is shown that under such cultivation conditions the activation of lipid peroxidation in the hepatocytes takes place. Its activation in presence of cadmium chloride was one of the factors of the membranes damage. The changes in the content of some fractions of lipids were similar both under the incubations of the cells with cadmium chloride and hydrogen peroxide. This allows one to suppose that cadmium chloride causes changes in the lipid composition of membranes as a result of intensification of lipid peroxidation.  相似文献

Plant exhibits various patterns of survival under salinity and their growth and development depend on their capacity to overcome the stress. Present investigation was focused on the response and regulation of the antioxidant defense system and the level of lipid peroxidation in Panicum miliacium and Panicum sumatrense under salt treatments. NaCl stress was imposed for 20 days after sowing of two Panicum species. The changes in the antioxidant enzyme activity like superoxide dismutase, catalase, peroxidase, and ascorbate peroxidase and the rate of lipid peroxidation level in terms of malondialdehyde (MDA) were recorded in both Panicum species. A great correlation exists between the antioxidant enzymes and lipid peroxidation. The defense mechanism activated in Panicum species studied was confirmed by the increased antioxidant enzyme activities under progressive NaCl stress. MDA content remained close to control at moderate NaCl concentrations and increased at higher salinities. Although lipid peroxidation increased in both Panicum species under salt stress the percent of increase was low in P. sumatrense indicating its salt-tolerant nature. Another possible conclusion is that improved tolerance to salt stress may be accomplished by increased capacity of antioxidative system.  相似文献

Metabolic processes and environmental conditions cause the constant formation of oxidizing species over the lifetime of cells and organisms. This leads to a continuous oxidation of intracellular components, including lipids, DNA and proteins. During the extensively studied process of lipid peroxidation, several reactive low-molecular weight products are formed, including reactive aldehydes as 4-hydroxynonenal (HNE). These aldehydic lipid peroxidation products in turn are able to modify proteins. The degradation of oxidized and oxidatively modified proteins is an essential part of the oxidant defenses of cells. The major proteolytic system responsible for the removal of oxidized cytosolic and nuclear proteins is the proteasomal system. The proteasomal system by itself is a multicomponent system responsible for the degradation of the majority of intracellular proteins. It has been shown that some, mildly cross-linked, HNE-modified proteins are preferentially degraded by the proteasome, but extensive modification with this cross-linking aldehyde leads to the formation of protein aggregates, that can actually inhibit the proteasome. This review summarizes our knowledge of the interactions between lipid peroxidation products, proteins, and the proteasomal system.  相似文献

We generalize the results of multiyear studies of the level of antioxidative activity of lipids isolated from tissues of laboratory rodents of different species and lines. A classification of lipids according to their ability to inhibit thermal autooxidation of methyl oleate is proposed. The involvement of lipids in low-temperature autooxidation reactions at the radical initiation and chain propagation stages was proved using the model developed. In addition to antioxidative activity, the initial content of peroxides in lipids, determined by their degree of unsaturation, and the antiperoxide activity of lipids are proposed for quantitative estimation of the kinetic characteristics of lipids of mammalian tissues. The dependence of effects on the rate of radical initiation in the system is shown, which is determined by the influence of the physicochemical properties of lipids on the coordination of relationships and balance of biochemical functions in biological objects differing in the intensity of oxidation processes.  相似文献
