A factorial experiment shows highly significant effects of temperature(12 5–22.5°C) and salinity (17.8–34 S) on thegrowth rate of Gyrodinium aureolum, with a significant temperature-salinityinteraction. The maximum growth rate of G aureolum is measuredto 0.61 div. day–1 at 20°C and 22.3 S. Gyrodiniumaureolum does not grow at temperatures :10 °C or 25°Cand at salinities 12 S. The cellular content of carbon (C) andnitrogen (N) and the elemental ratios N/C, P/C and N/P are significantlyaffected by the temperature The cellular content of phosphorus(P) and the elemental ratios P/C and N/P vary significantlywith salinity Significant temperature-salinity interactionsare found for the cellular content of carbon, nitrogen and phosphorus.Variations in the N/P ratio indicate that G.aureolum has a largestorage capacity for phosphorus It is suggested that temperatureis one important limiting factor in the initiation of bloomsof G.aureolum in north European waters.  相似文献

Water extracts of the red-tide dinoflagellate Alexandrium excavatumgrown at ‘high’ light intensity (200 µE m–2s–1) show a broad absorbance maximum in the UV regionof the spectrum (310–360 nm). Using TLC and reverse-phaseHPLC a series of mycosporine-like amino acids have been characterized:mycosporine-glycine (max = 310 nm), palythine (max = 320 nm),asterina-330 (max = 330 nm), shinorine (max = 334 nm), porphyra-334(max= 334 nm), palythenic acid (max = 337 nm) and the isomericmixture of usujirene and palythene (max = 359 nm). From theobserved spectral changes during transference from ‘low’(20 µE m–2 s–1) to ‘high’ (200µE m–2 s–1) light intensities and vice versa,the series of compounds are supposed to be biogenically relatedto one another. The presence of these compounds in A.excavatumis discussed in relation to their possible role in the photoprotectionto deleterious UV radiation.  相似文献

Wilting of leaves of Vicia faba L., which occurs when the pressurepotential (p) is zero, and the leaf-water potential () at wiltingboth depend entirely upon the solute potential at incipientplasmolysis (so) and not on soil-water status. Wilting in V.faba is acropetal; this is consistent with the hypothesis thatthere is a gradient of decreasing so up the plant and that wateris transferred from the lower to the upper leaves, hasteningthe overall water loss from the lower leaves to the point whenp is zero. The gradient in so up the plant is of the order of3–8 bar. It is proposed that wilting when p>0 (i.e. > so) shouldbe ‘apparent wilting’ and that when p0 (i.e. so),‘true wilting’.  相似文献

Changes in the major alcohol-soluble, low molecular weight carbohydratesof P. purpurea, O--D-galactopyranosyl-(1-2)-glycerol (‘floridoside’)and O--D-galactopyranosyl-(1-1)-glycerol (‘isofloridoside’),have been examined in response to salinity variation. ‘Floridoside’is shown to vary in absolute amount, increasing in hypersalineand decreasing in hyposaline media. ‘Isofloridoside’content per cell does not change in a similar manner. Responsesare almost identical under light or dark conditions, ‘floridoside’changes being complete within 24 h. Decreasing the externalwater potential using ionic and non-ionic solutes has the sameeffect upon galactosyl-glycerol content. The amount of ‘floridoside’synthesized, and degraded under hypersaline and hyposaline conditionsrespectively is shown to be insufficient to restore cell volumeto its original value. It is therefore suggested that the primaryfunction of ‘floridoside’ increases in concentratedsea-waters is that of a compatible solute, serving to protectthe cell during periods when the external salt content is increaseddramatically.  相似文献

Stable carbon isotopes (13C) were determined for phytoplanktonand dissolved inorganic carbon (DIC) from Lake Apopka, a shallow,polymictic and hypereutrophic lake in Florida, USA. Bulk planktondominated by pico- and nanqanobacteria were enriched in 13(–13.1± 1.1%) as a result of assimilation of extremely 13C-richDIC (13C = 9.6 ± 3.0%). Diatoms (Aulacoseira spp.) hada 13C of –14.3 ± 0.6% that was slightly more negativethan that of small cyanobacteria. Meroplanktonic diatoms hada 13C (–13.6 ± 1.8%), similar to their planktoniccounterparts. The 13C of a colonial cyanobacterium (Microcystisincerta) was exceptionally heavy (–3.0 ± 1.0%)and attributed to localized carbon limitation. Seasonal variationin 13C of bulk plankton was small (4%) relative to reports forother lacustrine systems No difference in the 13C of bulk planktonhorn surface water between stratified and non-stratified periodswas found. No measurable changes in 13C of bulk plankton wereindicated in light and dark incubation experiments Frequentwind mixing of the water column, high DIC concentration, andconsistently high lake productivity were used to explain thetemporal and spatial isotope consistency of phytoplankton inthis lake.  相似文献

We describe the occurrence of phycocyanin and phycoerythrocyanin-likepigment in natural Microcystis colonies collected in Lake Kasumigaura.For differentiating pigments, the combination of simultaneousdetermination of molecular weights of and ß and subunitsby electrospray ionization mass spectrometry with HPLC separationand fluorescence measurements was used. It was suggested thatthe natural Microcystis colonies may contain the phycoerythrocyanin-likepigment, judged from the excitation and emission spectra. Measuredpigments consisted of different molecular weights of and ßsubunits, but the molecular weights of and ß subunitsin both phycocyanin and phycoerythrocyanin-like pigments werenot always identical.  相似文献

The stable carbon isotope composition of particulate organicmatter expressed as 13C was measured in cultures of 13 speciesof marine microalgae in different phylogenetic groups. The effectsof salinity variations and changes in photoperiod were alsoassayed for three of them (i.e. Skeletonema costatum, Amphidiniumopercularum and Isochrysis galbana); the effect of nature ofnitrogen supply (nitrate. ammonium) was studied for one (S.costatum).These environmental parameters were chosen because of theirvariability in the ocean and their possible effects on 13C valuesof phytoplankton organic carbon. Batch culture conditions andsampling time after inoculum were strongly controlled in orderto provide cells in good physiological state which were comparablefrom one culture to the other. In the same way, sampling waslimited to the first 2 days of exponential growth, in orderto avoid a possible dissolved inorganic carbon (DIC) limitation.Carboxylase activities [of the enzyme ribulose 1,5-bisphosphatecarboxylase oxygenase (Rubisco), and the three ß carboxylases:phosphoenolpyruvate carboxylase (PEPC), phosphoenolpyruvatecarboxykinase (PEPCK) and pyruvate carboxylase (PC)] and totalchlorophyll a concentrations were assayed simultaneously. The13C values observed were between –30.2 and –12.7i.e. comparable to those observed in the world's oceans. Theisotopic composition of phytoplankton organic carbon was shownto be under the influence of the parameters tested but 13C variationsare specific to the species considered. The nature of ßcarboxylase found in each species, or systematic position, couldnot be linked to the isotopic composition of organic carbon.No linear or single correlation between 13C variations and environmentalmodifications were observed and there is no evidence for a simpleand universal relation between 13C of phytoplankters and theirenvironment. In monospecific cultures as in the field, 13C fractionationby Rubisco (and eventually by PEPCK) may be counterbalancedby other mechanisms.  相似文献

Zooplankton contribution to particulate phosphorus and nitrogen in lakes   总被引:1,自引:0,他引:1  
Based on the recognition of rather constant species-specificelement to dry weight ratios in freshwater zooplankton, poolsof metazoan zooplankton P and N were calculated for 45 Norwegianlakes of varying trophy. On the average zooplankton constituted20.4±12.3% and 4.6±4.3% of particulate P and Nrespectively. The fraction was considerably higher in many oligotrophiclakes, at most >50 and 20% respectively, but declined to15% (P) and 5% (N) in meso- to eutrophic lakes. In general,phytoplankton contribution to particulate P was <50%, leavinga large share of particulate P to bacteria and microzooplankton.The zooplankton proportion of particulate P was only weaklyinfluenced by the predation pressure in terms of fish communitystructure. Zooplankton P is important in the overall lake metabolism,and knowledge of this pool may be used to estimate zooplankton-mediatedloss and regeneration of P and improve total nutrient elementbudgets in lakes.  相似文献

The N-linked sugar chains were liberated as oligosaccha-ridesfrom octopus rhodopsin by hydrazinolysis. Most of the oligosaccharideswere neutral, and separated into two major components by columnchromatography using immobilized lectins and Bio-Gel P-4. The fibre length L and average helical angle of microfibrilorientation in fibres from successive growth rings in both normaland ‘rubbery’ wood of stems of apple var. Lord Lambournehave been measured. It has been shown that the normal relationship,L = A+B cot , does not hold for ‘rubbery wood’.There is a tendency for the helix to remain flat, the angle fairly constant independently of fibre length. This, togetherwith abnormalities in the lignin chemistry, causes the rubberywood to have low tensile strength and high extensibility.  相似文献

In nature, large concentrations of the toxic bloom-forming dinoflagellate,Gonyaulax tamarensis, are frequently observed in the vicinityof the pycnocline. In the absence of a pycnocline the organismis usually recorded near the surface, where light levels aremore advantageous for photosynthesis. In this paper we examinethe swimming behaviour of G.tamarensis when exposed to varyingdegrees of stratification and investigate whether the maintenanceof a subsurface (pycnocline) population is the result of retentionof the algae by a physical barrier or active accumulation ofthe organisms at a density interface. The study indicates thatG.tamarensis cells presented with a halocline of S<{smalltilde}6–7 (occurring over a few centimeters) cross thissalinity barrier and accumulate at the highest available photonflux density ({small tilde}100 µmol m–2 s–1).Cells exposed to a gradient of S>{small tilde}7remain atthe halocline (pfd={small tilde}40 µmol m–2 s–1).However, when light above the pycnocline is attenuated by theaddition of food colour to the medium, the cells cross a haloclineof S=10 and accumulate at the highest available photon fluxdensity. In the absence of added nutrients (inorganic N andP) the organism fails to exhibit a phototactic response. Thus,the presence of a strong halocline does not represent an inpenetrablephysical barrier for G.tamarensis and the development of pycnoclinepopulations of this organism is a function of density, lightand nutrient climate.  相似文献

Larqué-Saavedra, A., Rodriguez, M. T., Trejo, C. andNava, T. 1985. Abscisic acid accumulation and water relationsof four cultivars of Phaseolus vulgaris L. under drought.—J.exp. Bot 36: 1787–1792. Plants of four cultivars of Phaseolus vulgaris L. differingin drought resistance were grown in pots under greenhouse conditionsand prior to flowering water was withheld from the pots untilthe mid-day transpiration rate reached values below 1.0 µgH2O cm–2 s–1 (designated the ‘drought’stage). At this point leaves were harvested on 3 or 4 occasionsover 24 h to determine the abscisic acid (ABA) concentration,total water potential (), solute potential (1) and turgor potential(p). Results showed that values of , 1, and p differed between cultivarswhen they reached the ‘drought’ stage. The stomatalsensitivity to changes in and p, was as follows: Michoacán12A3 > Negro 150 Cacahuate 72 > Flor de Mayo. These datacorrelated well with the pattern of drought resistance reportedfor the cultivars. ABA accumulation at the ‘drought’ stage differedbetween cultivars at each sampling time, but overall differencesin ABA level between cultivars were not significant. ABA levelsdid not, therefore, correlate with the drought resistance propertiesreported for the cultivars. Xyloglucan is a polysaccharide found in the primary cell wallsof all higher plants examined. Its cellulose-like backbone,which is about 0.15 to 1.5 µm long, consists of 300 to3 000 ß-(14)-linked D-glucopyranose residues. About60–75% (or, in grasses, about 30–40%) of the glucoseresidues have side-chains attached to position 6. The majorside-chains are: D-xylopyranosyl--1 -, D-galactopyranosyl-ß-(12)-D-xylopyranosyl--I , L-arabinofuranosyl-(1 -2)-D-xylopyranosyl--1-, and (except in grasses) L.-fucopyranosyl--(1 -2)-D-galactopyranosyl-ß-(1-2)-D-xylopyranosyl--1-. There is some regularity in the distribution of these side-chainsalong the backbone. Xyloglucan plays two very different r?les in the control ofcell growth: (a) as a major building material of the wall [concentrationof xyloglucan in the wall in vivo 10% (w/v)] it probably directlydictates wall extensibility and, therefore, the rate of cellexpansion and (b) it can be broken down to a fucose-containingoligosaccharide which [at a concentration of 0.0000001% (w/v)]exerts a hormone-like anti-auxin effect on growth. In addition,xyloglucan lacking fucose is used by certain dicotyledonousseeds as a food reserve which is mobilized after germination.Xyloglucan is, therefore, the subject of considerable currentinterest in several apparently disparate areas of botany. Colonies of Trichodesmium spp. are conspicuous, macroscopiccomponents of the life in tropical and subtropical oceans. Thelarge size and the morphology of the colony raise questionsregarding the mechanism of carbon supply for photosynthesis.Constraints on these mechanisms may be indicated by the stablecarbon isotopic composition (13C) that reflects the balancebetween carbon supply and speciation, as well as the growthrate and colony size. The 13C of Trichodesmium off Bermuda measuredhere revealed a strong correlation between size of individualcolonies and season. The smallest colonies, 2–7 µgC colony–1, showed the lightest 13C composition (–19),increasing to asymptotic values of –12 above 7 µgC colony–1. The average 13C of the colonies was lightestimmediately after the onset of stratification in the SargassoSea, gradually increasing by 4 to heavier values during thesummer. We propose that the mass effect is due to increaseduse of HCO3 by the larger colonies, whereas the seasonalinfluence may be related to changes in irradiance and pCO2 affectingthe internal carbon cycling.  相似文献

Resting egg production and oviducal cycling were investigatedfor the calanoid copepods Arctodiaptomus alpinus and Acanthodiaptomusdenticornis both in the laboratory and in a small karstic alpinelake by making a census of the number of eggs produced and theproportion of females in each of four morphologically distinguishedreproductive conditions each day in the laboratory or duringa 2–3 week period in lake enclosures. In the laboratory,individuals were maintained on a mixed diet of natural phytoplanktonat constant temperatures of 4, 10, 15 and 20C, respectively.Both species differed considerably in their temperature requirementsfor reproduction. Lifetime fecundity was highest at 10C in A.alpinusand at 20C in A.denticornis, with up to 327 eggs –1 spawnedin the former and up to 582 eggs –1 in the latter species.Unfavorable temperatures were further reflected in an increasein egg mortality and the allocation of time spent in a post-reproductivephase, as well as in a decrease of longevity. Increasing temperaturesenhanced egg production rates due to decreasing clutch productionperiods, although clutch size was negatively correlated withtemperature. Maximum rates reached 5.88 and 7.98 eggs –1day–1 in the laboratory, and 0.73 and 0.55 eggs –1day–1 in enclosures in A.alpinus and A.denticornis, respectively.Egg production rates and clutch size were clearly governed bynutritional conditions in the lake, but were less affected byfood supply in the laboratory. Here, rates of egg productionwere adapted to improving food supply by increasing the frequencyof spawning events, rather than the number of eggs per clutch.No correlation was found between female body size and reproductiveparameters in the laboratory. A very low proportion of totalclutch production resulted in clutches composed of subitaneouseggs, i.e. 0.14% in A.denticornis and 1.20% in A.alpinus. Oviducalphase duration allocations indicate that there exists a temperatureoptimum for gamete maturation.  相似文献

Avena coleoptile hemicelluloses were fractionated into water-solublehemicelluloses I and IIB and water-insoluble hemicellulose IIA.These hemicelluloses were then subjected to glycosidic linkageanalysis by methylation technique, which revealed that hemicelluloseI was predominantly composed of arabinoxylans and ß-(l4)glucans and hemicellulose IIB was composed of arabinoxylans,ß-(l4) : (l3)-mixed linked glucans, ß-(l4)-glucansand xyloglucans. Hemicellulose IIA was mainly composed of xyloglucansand probably ß-(l4)-glucans. Methylation analysisof hemicelluloses extracted from Avena coleoptile segments treatedwith auxin in the presence of mannitol (0.15 M) indicated thatauxin apparently had no effect on the structure of arabinoxylanand caused a specific decrease in the amount of ß-(l4): (l3)-mixed linked glucan. Previous study on the binding properties of a lectin isolatedfrom Codium fragile subspecies tomentosoides (CFT) indicatesthat this lectin recognizes the GalNAc1 sequence at both reducingand nonreducing ends. In this study, the carbohydrate specificityof CFT was further characterized by quantitative precipitin(QPA) and inhibition of lectin-enzyme binding assays. Of theglycoforms tested for QPA, all asialo-GalNAc1 containing glyco-proteinsreacted well with the lectin. Asialo hamster and ovine submandibularglycoproteins, which contain almost exclusively Tn (GalNAclSer/Thr)residues as carbohydrate side chains, and Streptococcus typeC polysaccharide completely precipitated the lectin added, whilethe GalNAcβcontaining Tamm-Horsfall Sd(a+) glycopro-teinand its asialo product were inactive. Among the oligo-saccharidestested for inhibiting lectin-glycoprotein interaction, GalNAc13GalNAcβ13Gal14Galβ14GIc(Fp)and Galβ13GalNAc1benzyl (T) were the best, and about 125-foldmore active than GalNAc They were about 3.3, 6.6, and 43 timesmore active than Tn containing glycopeptides, GalNAc13(LFuc12)Gal(Ah) and Galβ13GalNAc(T), respectively. From the presentand previous results, it is concluded that the combining siteof CFT is probably of a groove type that recognizes from GalNAclto pentasaccharide(Fp). Light-limited photosynthetic carbon incorportion is expectedto be directly proportional to the scalar quantum irradiance.The proportionality constant is , where mis the maximum quantum yield (mol C Einstein–1 absorbed)and $$\stackrel{\&macr;}{{\hbox{ k }}_{\hbox{ c }}}$$ isthe mean spectral absorption coefficient (m2 mg–1 chla). Recent efforts to evaluate of in situphytoplankton photosynthesis are variously flawed. Lack of evidenceof proportionality and lack of correction of cosine to scalarirradiance are common deficiencies. Most data, as we interpretthem, indicate values in the range 0.0003 – 0.0006 mol C m2 Einstein1 abs mg1 chl a. New determinationsin lrondequoit Bay, New York, lie in this range. Most estimatesof at depth have been about 0.010 m2 mg–1chl a. Similar values are being obtained for total particulatesfrom lrondequoit Bay; whether detritus contributes significantlyis not yet known. Published data, in our view, all point tovalues of m in situ in the range 0.03–0.07 mol C Einstein–1abs. Information on the biosynthesis of the D-arabinans of the cellwall of Mycobacterium tuberculosis is rapidly emerging, withthe promise of new targets for drug development against tuberculosis.Accordingly, arabinosyl transferase assays were developed utilizingsynthesized [1–14C]-β-D-arabinofuranosyl-1-monophosphoryldecaprenolas donor and a variety of O- and S-alkyl arabinosides as acceptors.These were: -D-Araf-(15)--D-Araf-O- and -S-alkyl di-arabinosidesand -D-Araf-(15)--D-Araf-(15)--D-Araf-O- and -S-alkyl triarabinosides.Whereas the O- and S-alkyl monosaccharide acceptors were inactive,the O- and S-alkyl disaccharide and the O- and S-alkyl trisaccharideacceptors (<C12) possessed considerable acceptor activity,and the trisaccharide acceptors were more potent than the correspondingdisaccharides. The O-alkyl disaccharide acceptors with a C8alkyl chain were more active than those containing the C6 orC10 analogs. Chemical analysis of the enzymatically synthesizedproducts of the reactions demonstrated that β-D-arabinofuranosyl-1-monophosphoryldecaprenolwas an effective donor for two of the three potential arabinosyltransferases: β-D-arabinofuranosyl-1-monophosphoryldecaprenol:arabinan (15) arabinosyl transferase and β-D-arabinofuranosyl-1-monophosphoryl-decaprenol:arabinan β(12) arabinosyl transferase. The β(12) arabinosyltransferase activity was more in evidence in the presence ofthe O-alkyl disaccharide acceptor, whereas both transferaseswere about equivalent in the presence of the S-alkyl trisaccharideacceptor.
