The growth and survival of juvenile Haliotis rubra, when fed with the diatom Navicula sp. cultured in f/2 medium containing combined nitrogen at 24.71 mg NO3-N L–1 (high), 12.35 mg NO3-N L–1 (standard) or 2.47 mg NO3-N L–1 (low), were compared in a 33-day trial. The alga in the low nitrogen medium contained 37% less total amino acid than that in the high and standard nitrogen media. There was a slightly greater reduction in essential amino acids (40%) compared to non-essential amino acids (35%). Juvenile abalone feeding on Navicula grown in medium with low nitrate and lower total amino acid content grew more slowly than when fed on the same species grown in standard or higher nitrogen medium with a higher amino acid content. The growth rate of juveniles was highest (43 m d–1) in the high nitrate treatment followed (40 m d–1) by the standard nitrate treatment and lowest (31 m d–1) in the low nitrate treatment. The survival of the juveniles was also effected by the diet. Survival was better in the high and standard nitrogen media (88%) than the low nitrogen medium (75%). The results suggest that in order to achieve uniformity in nutritional quality of diatoms and good growth of abalone juveniles in commercial abalone nurseries, the nitrogen concentration in tanks should be monitored and additional nitrate added to provide an optimum concentration of between 2 and 12 mg NO3-N L–1.  相似文献

Microsatellite loci were isolated in Haliotis fulgens using a (CT)n enriched‐genomic library. From 33 sequenced clones, 21 microsatellites regions were identified, 15 with the expected (CT)n. Eight microsatellite loci were amplified, six of which were polymorphic with a range of three to 20 alleles, and five cross‐amplified in two other species (Haliotis rufescens and Haliotis corrugata). These microsatellites will be useful as population genetic markers in the three species.  相似文献

A traditional Chinese medicine (TCM) preparation was formulated from orange peel (Pericarpium Citri Reticulatae), hawthorn (Crataegus pinnatifida), astragalus (Astragalus membranaceus (Fisch.) Bunge), pilose asiabell root (Radix codonopsis), indigowoad root (Radix isatidis), taraxacum (Herba taraxaci) and malt (Fructus Hordei Germinatus) at a weight ratio of 1:1:1.5:1.5:1.5:1.5:2. A feeding experiment was conducted to determine the effects of TCM on innate immunity of abalone, Haliotis discus hannai Ino. Artificial diets containing 1%, 3%, 5% TCM preparation, 1% hawthorn or 1% astragalus, respectively, were fed to juvenile abalone (initial weight 10.38+/-2.51 g; initial shell length 44.15+/-4.15 mm) for 80 days. A TCM-free diet was used as a control. Each diet was fed to three replicate groups of abalone using a randomized design. The results indicated that phagocytic activity was significantly higher in abalone fed 3%, 5% TCM preparation, 1% astragalus or 1% hawthorn (P<0.05). Respiratory burst activity was significantly higher in abalone fed 1%, 3%, 5% TCM preparation, 1% astragalus or 1% hawthorn (P<0.05). Agglutination titre was significantly higher in abalone fed 5% TCM preparation (P<0.05). Weight gain ratio (WGR), daily increment in shell length (DISL), total haemocyte count (THC), plasma protein concentration, and the activity of acid phosphatase (ACP) were not significantly affected by the TCM preparation (P>0.05). These results indicate that TCM preparation can modulate the immunity of H. discus hannai, and it is very possible that TCM might be used as immunostimulants in abalone farming.  相似文献

Lipid composition of abalone was examined over a one-year interval. A feeding trial was designed to cover a full reproductive cycle in young adult green abalone, Haliotis fulgens, consisting of five diet treatments: the macrophytic algal phaeophyte Egregia menziesii, rhodophyte Chondracanthus canaliculatus, chlorophyte Ulva lobata, a composite of the three algae and a starvation control. The lipid class, fatty acid, sterol and 1-O-alkyl glyceryl ether profiles were determined for foot, hepatopancreas/gonad tissues and larvae. The major fatty acids were 16:0, 18:0, 18:1(n-7)c, 18:1(n-9)c, 20:4(n-6), 20:5(n-3) and 22:5(n-3), as well as 14:0 for abalone fed brown and red algae. 4,8,12-Trimethyltridecanoic acid, derived from algae, was detected for the first time in H. fulgens (hepatopancreas complex, 1.2–13.9%; larvae, 0.5% of total fatty acids). Diacylglyceryl ethers were present in larvae (0.6% of total lipid). The major 1-O-alkyl glycerols were 16:0, 16:1 and 18:0. Additionally, 18:1(n-9) was a major component in hepatopancreas/gonad and larvae. The major sterol was cholesterol (96–100% of total sterols). Highest growth rates were linked to temperature and occurred in abalone fed the phaeophyte E. menziesii (43 μm·day–1, 56 mg·day–1 yearly mean), an alga containing the highest levels of C20 polyunsaturated fatty acids and the highest ratio of 20:4(n-6) to 20:5(n-3). This study provides evidence of the influence of diet and temperature on seasonal changes in abalone lipid profiles, where diet is most strongly related to body mass and temperature to shell length. The allocation of lipids to specific tissues in green abalone clarifies their lipid metabolism. These results provide a basis for improving nutrition of abalone in mariculture through formulation of artificial feeds.  相似文献

An epidemic of mass mortality of abalone (Haliotis diversicolor supertexta) postlarvae aged 40 days or less has existed across south coast of China since the second half of 2002. Among 20 bacterial strains isolated from diseased abalone postlarvae on 2216E marine agar plates during an outbreak of postlarval disease in August 2005, a predominant strain (designated strain 20) was demonstrated to be virulent to postlarvae with an LD(50) value of 1.0x10(5) colony forming units (CFUml(-1)) on day 4, while the other 19 strains were either avirulent (16 strains) or weakly virulent (3 strains). The same bacterium could be re-isolated from postlarvae after bacterial challenge using 2216E marine agar plates. Preliminary toxicity tests of ECPs of strain 20 revealed that at 2.77mgproteinml(-1), crude ECPs completely liquefied postlarvae within 24h, leaving only shells. API 20E analysis identified strain 20 as Klebsiella oxytoca. 16S and ITS rDNA sequencing and phylogenetic analyses further confirmed this identification. Antibiotic susceptibility tests showed that strain 20 exhibited 94% of susceptibility to 16 various antibiotics tested and only showed resistance to streptomycin. Results of this work demonstrated that K. oxytoca is also linked to this epidemic in Fujian, China. This is considered to be the first report regarding K. oxytoca involved in the mass mortality of postlarval abalone in south China and the world.  相似文献

Beach-cast kelp (the most widely used feed for commercially grown South African abalone) is plentiful during winter months when periodic storms cause kelp to wash ashore. During summer, however, this resource is not always readily available and farmed abalone are often starved for short periods. The aim of this research was to assess how periodic kelp starvation influences growth of the commercially grown abalone, Haliotis midae Linnaeus. Growth of grow-out abalone was monitored on a commercial abalone farm over a period of six months and consisted of 3 treatments with 2 replicates (n = ±250 abalone per replicate). The treatments were: Control (abalone always given more kelp than what they typically needed); Treatment 1 (abalone fed their weekly ration only once a week); Treatment 2 (abalone fed half their weekly ration every 3 and then 4 days respectively). While the data at first suggest that the control animals outperform the treatment animals, after undergoing an initial adjustment period to the new feeding regime, the treatment animals perform better. Weight gain and feed conversion efficiencies show that the treatment animals perform better overall. The control animals generally required much more feed to produce comparable increases in both length and weight compared to the treatment animals. This study has shown that periodic bouts of starvation is beneficial to Haliotis midae, allowing variable growth spurts when returned to full feed rations. Biochemical changes in the body components during gonad maturation of the tropical abalone Haliotis varia were investigated using wild collected specimens from the Gulf of Mannar, on the southeast coast of India. The gonadosomatic index (GSI) and the hepatosomatic index (HSI) showed negative correlations throughout the study period as well as during the progression of gonad maturation stages. The highest GSI for both the sexes were in the ripe stages followed by late maturing stages. The HSI ranged from 2.97 to 6.71 in females, and 3.55 to 5.09 in males. Among the biochemical components analysed, lipid and carbohydrate contents showed significant variations in the different tissues of H. varia during the progress of gonad maturation. The highest protein content was in the foot muscle and the lowest was in the digestive gland. Total lipids in the ovary were always higher compared with that of the testis and the values ranged from 12.60 to 26.49%, registering the highest value in the ripe ovary. Gonad carbohydrate content was lower when the lipid content was higher, suggesting the conversion of carbohydrate to lipids. The present study demonstrates the role of nutrient translocation between body parts as an essential part of the reproductive physiology of abalone.  相似文献

The settlement, early growth and survival of the larval abalone Haliotis discus hannai in response to eight monospecific benthic diatoms were examined in the laboratory. Postlarvae showed active settling and feeding behaviour in all diatom species and in naturally occurring diatoms. Larval settlement rates differed significantly between experimental substrata after 24 h and 48 h. Nitzschia sp. (96.67 %), Hantzschia amphioxys var. leptocephala (95.00 %) and Navicula seminulum (90.00 %) strongly induced larval settlement of H. discus hannai. Postlarvae could feed on benthic diatoms (< 36 μm in shell length) with both weak and strong adhesion on the 4th day after settlement. Greatest growth (shell length) occurred on Nitzschia sp. (786.84 μm ± 2.50 SE and 773.09 μm ± 2.09 SE). Survival of postlarvae was also greatest on Nitzschia sp. (95.33 % ± 1.45 SE). These results indicate the effectiveness of Nitzschia sp., H. amphioxys var. leptocephala, N. seminulum, Rhaphoneis surirella and Navicula corymbosa as single species over natural diatoms in larval settlement and postlarvae growth of H. discus hannai. Thus, Nitzschia sp. has the best potential diet for larval settlement and postlarvae growth of H. discus hannai. H. amphioxys var. leptocephala and N. seminulum can be used as cues to induce larvae settlement, and R. surirella and N. corymbosa can be used as food for growing postlarval.  相似文献

Postlarvae of Macrobrachium lanchesteri were stocked in confined waters. The larvae were fed on an ad libitum food of Tubifex tubifex worms for 30 days and their rearing has been established. 1 Kg of initially stocked postlarvae need 4.5 Kg of live T. tubifex to produce 0.537 Kg of new flesh. Withering syndrome (WS) is a disease of wild and cultured abalone caused by a Rickettsiales-like prokaryote (WS-RLP). This study compared the pathologic changes that occur during the progression of WS in red abalone to those caused by environmental stresses consisting of elevated temperature and food limitation and determined the impact of these stressors on WS prevalence and intensity. Farmed red abalone were administered a feed-based oxytetracycline therapeutic treatment to assure WS-RLP-free status prior to initiation of the experiment. Groups were then held in each of eight combinations of exposed vs. unexposed to WS-RLP, elevated vs. ambient temperature, and high vs. low food supply, for 447 days. Mortality was associated with starvation and disease but not elevated temperature alone. Elevated temperature significantly affected WS-RLP transmission: only 1.7% of WS-RLP- exposed abalone held at ambient temperature (12.3 degrees C) became infected compared to at least 72% of those held at elevated temperature (18.7 degrees C). Among exposed abalone at elevated temperature, fed animals exhibited greater infection prevalence but not greater infection intensity or digestive gland changes than starved animals, suggesting that abalone acquire infections by ingesting contaminated food. Food, temperature, WS-RLP exposure, and most of their interactions had significant effects on body condition and foot atrophy. Immunohistochemical detection of cell proliferation and apoptosis revealed no differences between normal digestive gland and that infected with WS-RLP. Body mass shrinkage, foot atrophy, elevated mortality, and decreased foot and digestive gland glycogen were observed in both WS-affected and starved, unexposed abalone, with the WS-RLP-exposed, starved group held at elevated temperature faring worst. Among exposed and unexposed animals, food supply but not temperature affected body mass and growth. These data demonstrate that the high morbidity and mortality exhibited by WS-RLP-infected abalone is a consequence of disease and not direct thermal stress. Drug residue analysis indicated oxytetracycline concentrations of up to 600 ppm in the digestive gland at 38 days post-treatment, an unusual degree of tissue retention of this therapeutant.  相似文献

Toxic algal blooms are common world-wide and pose a serious problem to the aquaculture and fishing industries. Dinoflagellate species such as Karenia brevis, Karenia mikimotoi, Heterosigma akashiwo and Chatonella cf. antiqua are recognised toxic species implicated in various faunal mortalities. Toxic blooms of Karenia cristata were observed on the south coast of South Africa for the first time in 1988 and were responsible for mortalities of wild and farmed abalone. K. cristata and various other dinoflagellate species common along the South African coast, as well as K. mikimotoi (Isolation site: Norway, Univ. of Copenhagen) and K. brevis (Isolation site: Florida, BIGELOW), were tested for toxicity by means of a bioassay involving Artemia larvae as well as abalone larvae and spat. K. cristata, like K. brevis, contains an aerosol toxin; however, the toxin present in K. cristata has not yet been isolated and remains unknown. K. brevis was, therefore, used to determine which developmental phase of the bloom would affect abalone farms most, and whether ozone could be used as an effective mitigating agent. Of the 17 dinoflagellate species tested, K. cristata, Akashiwo sanguinea, K. mikimotoi and K. brevis pose the greatest threat to the abalone mariculture industry. K. brevis was most toxic during its exponential and stationary phases. Results suggest that ozone is an effective mitigation agent but its economic viability for use on abalone farms must still be investigated.  相似文献

The red abalone, Haliotis rufescens (Swanson 1822), is an important species for commercial aquaculture in Mexico, with annual production levels around 68 t. Not surprisingly, there is great interest in increasing production and its cultivation success. In order to have a better understanding of the genes involved in the sexual differentiation, this study analyzed the early differential expression of eight sex-related genes (VCP 2.2, VERL, VTGI, DMRT1, FP, LYS, SARIP, TEKT A1) by RT-qPCR in abalones from 5 to 45 mm of shell length. Sex identification was evaluated using VERL and LYS genes expression levels. These genes differentiated females and males from 16 mm of shell length and above. All eight sex-specific genes showed expression in all organisms. However, their level was higher in the sex group to which they correspond. In contrast, differential expression levels occurred in individuals with a shell length of 26–35 mm and 36–45 mm. These results suggest that sex gene expression is not entirely sex specific, and sexual differentiation in red abalone occurs between 16 and 25 mm of shell length. More studies are needed to determine the function of these genes in the sex that they are not associated with.  相似文献

The mitochondrial enzyme manganese superoxide dismutase (mitMn-SOD) is one of the antioxidant enzymes involved in cellular defense against oxidative stress and catalyzes the conversion of O2 into the stabler H2O2. In this study, a putative gene encoding Mn-SOD from disk abalone (Haliotis discus discus, aMn-SOD) was cloned, sequenced, expressed in Escherichia coli K12(TB1) and the protein was purified using pMAL protein purification system. Sequencing resulted ORF of 681 bp, which corresponded to 226 amino acids. The protein was expressed in soluble form with molecular weight of 68 kDa including maltose binding protein and pI value of 6.5. The fusion protein had 2781 U/mg activity. The optimum temperature of the enzyme was 37 °C and it was active in a range of acidic pH (from 3.5 to 6.5). The enzyme activity was reduced to 50% at 50 °C and completely heat inactivated at 80 °C. The alignment of aMn-SOD amino acid sequence with Mn-SODs available in NCBI revealed that the enzyme is conserved among animals with higher than 30% identity. In comparison with human mitMn-SOD, all manganese-binding sites are also conserved in aMn-SOD (H28, H100, D185 and H189). aMn-SOD amino acid sequence was closer to that of Biomphalaria glabrata in phylogenetic analysis.  相似文献

Summary Four intraspecific slave raids involving 3 colonies of the European amazon antPolyergus rufescens Latr. occurred during hot and sunny afternoons in July 1992 near Parma, Italy. Generally, no fighting between the resident ants and the invaders was recorded, and pillage of the brood lasted just a few minutes. The brood captured during 3 raids was collected and transported ot the laboratory where slavemaker callow workers eclosed and were accepted by both the raiders and hosts present in artificial mixed colonies. This is the first case of functional intraspecific slavery recorded in the field for the obligatory dulotic formicine antP. rufescens.  相似文献

Summary

This study was conducted to examine the nutritional value of eight algal diets for two species of abalone, Haliotis tuberculata and Haliotis discus hannai, by measuring biochemical composition of the algae and relating this to feeding rate, growth and biochemical composition of the animals. Nutritional value of algal diets can be divided into three categories for each species of abalone. For H. tuberculata the best performance was on the mixed diet and Palmaria palmata intermediate was Alaria esculenta, Ulva lactuca and Laminaria digitata, and lowest growth was on Laminaria saccharina and Chondrus crispus. For H. discus hannai, best performance was on A. esculenta, P. palmata and the mixed diet; intermediate was on L. saccharina and L. digitata and lowest was on U. lactuca. It is generally accepted that high “balanced” levels of protein (>15%), lipid (3–5%) and carbohydrate (20–30%), with no detrimental substances in natural algae are essential for optimal growth performance of these abalone. The fact that A. esculenta, L. saccharina and U. lactuca had different dietary values for the two abalone species indicates specific nutritive requirements and/or digestive physiology. Overall, H. tuberculata grew faster, had higher food conversion efficiencies and muscle yield than H. discus hannai. Generally abalone fed on the highest category diets, had higher muscle yields and levels of protein, visceral lipids and muscle carbohydrate. Viscera and foot muscle are reservoirs for lipid and carbohydrate, respectively. The effect of algal diet on sexual maturation is similar to that on somatic growth.  相似文献

Cryopreservation could provide stock quantities of embryos for transgenic research. This study aimed to determine the least toxic cryoprotective agent for Haliotis midae embryos. They were exposed for 30 min to concentrations varying from 5% to 20% of the following cryopreservatives: methanol (MET), polyethylene glycol (PEG), dimethyl sulfoxide (ME2SO) and glycerol (GLY). In contrast to cryopreservation studies done in other molluscs, PEG showed the least toxicity to H. midae embryos in concentrations ranging from 5% to 15%. MET was also less toxic than ME2SO and GLY at correlating concentrations. GLY showed the most toxic effects with most embryos dead or abnormal at concentrations above 15%.  相似文献
