Resistance to primary and secondary infections with H. contortus was compared in St. Croix, 3/4 St. Croix, Florida Native, Barbados Blackbelly and domestic crossbred lambs. On the basis of worm counts, fecal egg counts and packed cell volumes, St. Croix and 3/4 St. Croix lambs were the most resistant to secondary infections, Florida Native and Barbados lambs were intermediate in resistance, and the domestic lambs were the least resistant. Breed differences were highly variable during primary infection. Breed differences were most pronounced prior to puberty, with domestic lambs highly susceptible to secondary infections. After puberty, domestic lambs were better able to resist secondary infections. Age apparently had little or no effect on the parasite resistance of exotic breeds. Sex differences were significant only after puberty when ewe lambs were more resistant to secondary infection than ram lambs. In one of three experiments lambs of hemoglobin genotype AB were more resistant to secondary infection than lambs of genotype BB in terms of fecal egg counts, packed cell volumes and the proportion of ‘responder’ and ‘nonresponder’ lambs but not in terms of worm counts, although subtle differences may not have been detected because of small numbers of lambs. 相似文献

Ewes of three exotic breeds (Florida Native, Barbados Blackbelly and St. Croix) showed no periparturient rise in fecal egg counts (PPR) when housed from late fall through lambing and weaning. Domestic breed ewes (Rambouillet and Finn-Dorset × Rambouillet) showed a pronounced PPR 6–7 weeks post-lambing while St. Croix × domestic ewes showed an intermediate PPR under similar conditions. There was no PPR in non-lambing ewes.In a second experiment, breed differences in the PPR were not as pronounced when ewes were allowed to graze a contaminated pasture after lambing. Florida Native, St. Croix and St. Croix × domestic showed lower PPRs than the remaining breeds (Barbados Blackbelly, F1 hybrids of exotic and domestic breeds and domestic ewes), but the magnitude of the PPR was generally small in all breeds. No relationship was observed between hemoglobin genotype and PPR in either experiment.It was concluded that a low or absent PPR is an important manifestation of breed resistance in exotic ewes and that this may be useful as a marker for breeding parasite-resistant sheep. 相似文献

Abstract Nematode parasites have developed resistance to anthelmintics. Biological control of gastrointestinal nematodes (GIN) in sheep is a promising non-chemical control method. Two experiments were done using Merino sheep. In each experiment, gender, initial egg count per gram (EPG) and initial body weight (BW) aided in placing animals into four groups, each of which was randomly fed with one of four treatments. Experiment 1 evaluated anthelmintic effects of Bacillus thuringiensis Berliner (Bt), Clonostachys rosea f. rosea Schroers (C. rosea) and diatomaceous earth (DE) in sheep. Bacillus thuringiensis and C. rosea were fed to sheep at a rate of 1 g kg?1 BW, and DE was fed at 2% of sheep diet. Relative to the control, treatments had no effect (P>0.05) on EPG, but reduced (P<0.001) larvae per gram (LPG) in faecal culture. Efficacy varied with time (P<0.001). On Day 7, Bt, C. rosea and DE had efficacies of 76, 87 and 61%, respectively. In experiment 2, efficacy of feeding 1 g kg?1 BW of C. rosea chlamydospores to sheep every day, every second day and every third day was tested. Daily feeding of fungal chlamydospores had no effect on EPG (P>0.05), but reduced (P<0.001) LPG (12±1.67) more than every second day (39±0.77) or third day (58±1.77). On Day 12, feeding daily, every second day and every third day had efficacies of 90, 63 and 49%, respectively. These studies suggest that each of Bt, C. rosea isolates and DE products has a potential to affect nematode larvae, and daily use of C. rosea had the highest effect as a biological control of nematodes in this study. 相似文献

The fungus Clonostachys rosea f. rosea has the potential to control gastrointestinal nematodes (GIN) of sheep. A study was conducted using adult Merino sheep. Gender, egg count and initial body weights (BWs) were used to categorise animals into four groups which were randomly assigned to one of four C. rosea dietary treatments. Treatments were mixed with a complete diet and fed to sheep once daily for 10 weeks. There was no effect of treatment on eggs per gram of faeces. However, treatments reduced the number of larvae per gram in faecal cultures (P < 0.001) and larval development (LD) time (P < 0.001). Efficacy of treatments increased (P < 0.001) with time. On Day 70, treatments of 0.25, 0.5 and 1.0 g of C. rosea chlamydospores per kilogram BW reduced LD time by 33, 72 and 89%, respectively, whereas in the control, LD was reduced by only 2.6%. C. rosea significantly (P < 0.001) reduced numbers of third-stage larvae (L3) found in pastures. Our findings suggest that daily feeding with C. rosea has a potential to reduce larval stages of GIN in pastures. 相似文献

The high prevalence of anthelmintic-resistant gastrointestinal nematodes (GINs) throughout the world has led to the need for alternative worm control strategies. One of the possible substitutes to reduce the problems of drug resistance and residue is the evaluation/breeding of small ruminants for greater resistance to the GINs (organically produced), which in turn would be a helpful tool to predict the performance of an animal. At present, the existing diversity in the genetic potential to resist/tolerate GINs infection both within and between breeds has been validated. Successful selection of animals to define the genotype and identified resistance is related to the employed markers. A number of phenotypic traits such as faecal egg count (FEC), worm burden, serum antibodies, peripheral eosinophilia, packed cell volume, live weight, serum protein and albumin concentrations have been used for this purpose both in natural and artificial infections. Relatively resistant/tolerant animals have also been found to have mastocytosis, globule leucocytes, high levels of histamine and immunoglobulin (Ig) A and IgE concentrations. Of these traits, the principal and most practical measurement used to assess resistance status in animals undergoing similar parasite challenges is FEC. FEC has a positive/negative correlation with other biochemical, cellular and immunological parameters; however, the reliability of individual trial is often questioned and valuable information regarding the genetic makeup can be obtained from pooled data of a large number of trials and parameters. Florida Native sheep is among the sheep breeds best adapted to humid and hot climatic conditions such as those of Florida, USA, and have shown a superior ability to regulate nematode burdens. This is one of the oldest sheep breeds in North America and is an endangered species. To ensure genetic diversity and long-term survival of the breed, protection of the current genetic stock is critical and conservation efforts are required to promote its breeding and production. The objective of the present study was to investigate the importance of additive and non-additive genetic effects on resistance to natural Haemonchus contortus infections in Florida Native sheep using a whole genome scan. A total of 200 sheep were evaluated in the present study. Phenotypic records included faecal egg count (FEC, eggs/gram), FAMACHA® score, packed cell volume (PCV, %), body condition score and average daily gain (ADG, kg). Sheep were genotyped using the GGP Ovine 50K SNP chip and 45.2 k single nucleotide polymorphism (SNP) markers spanning the entire genome were available for quality control procedures. Mixed models were used to analyse the response variables and included the identity by state matrix to control for population structure. Bonferroni correction was used to control for multiple testing and a second arbitrary threshold (0.1 × 10?3) was used. Fifteen SNPs with additive and non-additive genetic effects and located in Ovis aries chromosome OAR1, 2, 3, 6, 8, 10, 11, 12, 13 and 21 were associated with FEC, FAMACHA® score, PCV and ADG. The present study was conducted to assess the comparative physiological responses of stress in sheep selected for resistance or susceptibility to Haemonchus contortus in the semi-arid region during different seasons. A total of 20 adult Malpura sheep from resistant and susceptibility line were selected for study. The intensity of strongyle infection, blood biochemical and cortisol levels were estimated monthly basis while meteorological data were collected everyday throughout the year. The temperature-humidity index was the highest during dry-hot season. The faecal egg count increased significantly (p < 0.05) during June–August (rainfall season) in susceptible sheep as compared to resistant sheep. The plasma cortisol level increased significantly (p < 0.05) in both the groups during dry hot (March–May) season and it remained higher in the susceptible line till June–August. The total plasma protein reduced significantly during hot-humid with scantly rainfall (September–November) season as compared to other season in susceptible line. It is pertinent to conclude that though sheep remained in stress during dry hot (March–May) season, but effects of stress on the intensity of strongyle infection and blood biochemical were better tolerated by animals of resistant line during hot-humid season compared to susceptible line under semi-arid region. 相似文献

Wild-caught rainbowfish Melanotaenia spp. originating from three isolated populations were infected with a quantified dosage of parasites Ichthyophthirius multifiliis in a controlled environment. The Melanotaenia eachamensis from Dirran Creek were much more susceptible to ichthyophthiriasis than were M. splendida from the Lake Tinaroo or Bluewater Creek populations. When the highly susceptible Dirran Creek rainbowfish were crossed with rainbowfish from a fourth population, Lake Eacham M. eachamensis , they produced hybrids with significantly higher resistance than pure-bred Dirran Creek, but not higher than pure-bred Lake Eacham fish. Hence, intraspecific hybridization increased resistance to I. multifiliis infection in M. eachamensis. Hosts from all three populations were much less susceptible to infection on their second exposure to the parasite. However, the Bluewater Creek population was better able to acquire immunity to I. multifiliis than either the Dirran Creek or Lake Tinaroo populations. It is tentatively suggested that there may be a link between the heterozygosity of populations of rainbowfish and their initial ability to resist infection by Ichthyophthirius multifiliis. 相似文献

Resistance to parasites is believed to have a widespread influence on demographic and adaptive processes. In systems where parasites impose a fitness cost on their host, heterozygotes may be selected because they are more resistant to parasites than homozygotes. Our objective was to assess the relationships between genomewide individual heterozygosity and abomasal nematode burdens in female Svalbard reindeer (Rangifer tarandus platyrhynchus) after the effects of host age, locality, season, and year had been accounted for. Samples were obtained from 306 female reindeer that were culled and genotyped at nine microsatellite loci. Reindeer in our study populations are mainly parasitized by the gastrointestinal nematodes Ostertagia gruehneri and Marshallagia marshalli. The infection intensity of each parasite differed between subpopulations, and among host age classes, seasons and years. We found no significant relationships between abomasal worm burdens, or lumen and mucosa larvae, of either O. gruehneri or M. marshalli and individual heterozygosity (or mean d(2)) alone or in interactions with host age, locality, and year. Although we analysed one of the largest data set available to date on gastrointestinal nematodes of a wild ruminant, we used a typical data set of nine genetic neutral markers that may have had low power to detect heterozygosity-fitness correlations. We conclude that the proportion of the variance in parasite resistance explained by individual heterozygosity for neutral genetic markers is low in Svalbard reindeer and in vertebrates in general, and we suggest that the candidate-gene approach might be more fruitful for further research on gene-fitness correlations. 相似文献

In dairy sheep flocks from Mediterranean countries, replacement and adult ewes are the animals most affected by gastrointestinal nematode (GIN) infections. In this study, we have exploited the information derived from an RNA-Seq experiment with the aim of identifying potential causal mutations related to GIN resistance in sheep. Considering the RNA-Seq samples from 12 ewes previously classified as six resistant and six susceptible animals to experimental infection by Teladorsagia circumcincta, we performed a variant calling analysis pipeline using two different types of software, gatk version 3.7 and Samtools version 1.4. The variants commonly identified by the two packages (high-quality variants) within two types of target regions – (i) QTL regions previously reported in sheep for parasite resistance based on SNP-chip or sequencing technology studies and (ii) functional candidate genes selected from gene expression studies related to GIN resistance in sheep – were further characterised to identify mutations with a potential functional impact. Among the genes harbouring these potential functional variants (930 and 553 respectively for the two types of regions), we identified 111 immune-related genes in the QTL regions and 132 immune-related genes from the initially selected candidate genes. For these immune-related genes harbouring potential functional variants, the enrichment analyses performed highlighted significant GO terms related to apoptosis, adhesion and inflammatory response, in relation to the QTL related variants, and significant disease-related terms such as inflammation, adhesion and necrosis, in relation to the initial candidate gene list. Overall, the study provides a valuable list of potential causal mutations that could be considered as candidate causal mutations in relation to GIN resistance in sheep. Future studies should assess the role of these suggested mutations with the aim of identifying genetic markers that could be directly implemented in sheep breeding programmes considering not only production traits, but also functional traits such as resistance to GIN infections. 相似文献

Abstract An investigation was conducted in pots to access the susceptibility of three lepidopteran pests, namely, gram pod borer, Helicoverpa armigera, greater wax moth, Galleria mellonella, and rice moth, Corcyra cephalonica, to two recently described species, Steinernema masoodi, S. seemae, and three indigenous S. carpocapsae, S. glaseri and S. thermophilum entomopathogenic nematodes (EPN). The suitability of these lepidopterans for the in vivo mass production of the nematodes was also estimated. Among the five species of EPN, S. masoodi, S. seemae and S. carpocapsae were found most pathogenic to C. cephalonica, bringing about mortality within 24 h, followed by H. armigera (36, 38 and 48 h, respectively) and G. mellonella (30, 36 and 48 h, respectively). The other species of EPN, viz., S. glaseri and S. thermophilum was the least pathogenic, which killed the larvae of C. cephalonica in 29 and 36 h, respectively, G. mellonella in 48 h, and H. armigera in 38 and 56 h, respectively. Galleria mellonella was found the most suitable host for the mass production of infective juveniles (IJs) of S. seemae, which yielded higher IJs than S. carpocapsae. Helicoverpa armigera was the next best suitable alternate host, which produced maximum IJs in case of S. seemae followed by S. masoodi, S. carpocapsae, S. glaseri and S. thermophilum. Rice moth, Corcyra cephalonica was the least suitable host. The susceptibility of H. armigera to five tested EPN species and susceptibility of G. mellonella and C. cephalonica to S. masoodi and S. seemae are new records. 相似文献

Dietary carotenoids have been shown to confer immunological benefits to some species of animals in which males also use these pigments to attract mates. Thus, the potential exists for an allocation trade-off between the sexual and immunological functions of carotenoids. Food availability may also influence immune system function. The present study examined the effects of carotenoid and food availability on the resistance of male guppies ( Poecilia reticulata Peters) from four wild populations to the parasite Gyrodactylus turnbulli Harris. Intermediate levels of carotenoid ingestion resulted in the lowest parasite loads, which suggests that carotenoids strengthen parasite resistance at low levels but either benefit parasites or suppress host immunity at high levels. Males raised on the high-food level initially had fewer parasites, suggesting heightened innate immunity relative to males raised on the low-food level. Over the course of the experiment, however, the high-food males supported higher parasite population growth rates than the low-food males. The results obtained emphasize the importance of evaluating the effects of diet on multiple aspects of immune system function, and caution against assuming that positive effects of carotenoids on immunity in one context will automatically translate to other contexts. © 2006 The Linnean Society of London, Biological Journal of the Linnean Society , 2006, 89 , 301–309. 相似文献

Hookworms (genera Ancylostoma and Necator) are amongst the most prevalent and important parasites of humans globally. These intestinal parasites ingest blood, resulting in anemia, growth stunting, malnutrition, and adverse pregnancy outcomes. They are also critical parasites of dogs and other animals. In addition, hookworms and hookworm products are being explored for their use in treatment of autoimmune and inflammatory diseases. There is thus a significant and growing interest in these mammalian host-obligate parasites. Laboratory research is hampered by the lack of good means of cryopreservation and recovery of parasites. Here, we describe a robust method for long-term (≥3 year) cryopreservation and recovery of both Ancylostoma and Necator hookworms that is also applicable to two other intestinal parasites that passage through the infective L3 stage, Strongyloides ratti and Heligmosomoides polygyrus bakeri. The key is a revised recovery method, in which cryopreserved L1s are thawed and raised to the infective L3 stage using activated charcoal mixed with uninfected feces from a permissive host. This technique will greatly facilitate research on and availability of gastrointestinal parasitic nematodes with great importance to global health, companion animal health, and autoimmune/inflammatory disease therapies. 相似文献

Significant breed differences were detected in fecal egg counts and fluke counts following experimental infection of five breeds of sheep with metacercariae of F. hepatica. Two experiments compared the responses of Barbados Blackbelly, St. Croix, Florida Native, Finn X Rambouillet and Targhee sheep to primary and challenge infections (Expt. 1) and to multiple infections (Expt. 2). Barbados Blackbelly sheep were more susceptible to infection than the other breeds in both experiments. St. Croix and Florida Native sheep were the most resistant breeds in Expt. 1, while Targhee and Florida Native were the most resistant in Expt. 2. A genome‐wide scan was performed to detect quantitative trait loci (QTL) for resistance to the gastrointestinal nematode Haemonchus contortus in a double backcross population of Red Maasai and Dorper sheep. The mapping population comprised six sire families, with 1026 lambs in total. The lambs were artificially challenged with H. contortus at about 6.5 months of age, and nine phenotypes were measured: fecal egg count, packed cell volume decline, two weight traits and five worm traits. A subset of the population (342 lambs) was selectively genotyped for 172 microsatellite loci covering 25 of the 26 autosomes. QTL mapping was performed for models which assumed that the QTL alleles were either fixed or segregating within each breed, combined with models with only an additive QTL effect fitted or both additive and dominance QTL effects fitted. Overall, QTL significant at the 1% chromosome‐wide level were identified for 22 combinations of trait and chromosome. Of particular interest are a region of chromosome 26 with putative QTL for all nine traits and a region of chromosome 2 with putative QTL for three traits. Favorable QTL alleles for disease resistance originated in both the Red Maasai and Dorper breeds, were not always fixed within breed and had significant dominance effects in some cases. We anticipate that this study, in combination with follow‐up work and other relevant studies, will help elucidate the biology of disease resistance. 相似文献

It is generally assumed that resistance to parasitism entails costs. Consequently, hosts evolving in the absence of parasites are predicted to invest less in costly resistance mechanisms than hosts consistently exposed to parasites. This prediction has, however, rarely been tested in natural populations. We studied the susceptibility of three naïve, three parasitized and one recently isolated Asellus aquaticus isopod populations to an acanthocephalan parasite. We found that parasitized populations, with the exception of the isopod population sympatric with the parasite strain used, were less susceptible to the parasite than the naïve populations. Exposed but uninfected (resistant) isopods from naïve populations, but not from parasitized populations, exhibited greater mortality than controls, implying that resistance entails survival costs primarily for naïve isopods. These results suggest that parasites can drive the evolution of host resistance in the wild, and that co‐existence with parasites may increase the cost‐effectiveness of defence mechanisms. 相似文献

Summary The main method used for the control of gastrointestinal nematodes in sheep production is the application of chemotherapeutic agents, which often lead to the selection of parasites resistant to given active principles. Biological control can be considered a promising alternative, contributing to an increase in the efficacy of verminous control. We determined the in vitro activity and in situ survival of the predatory fungi Arthrobotrys musiformis and Arthrobotrys conoides during passage through the gastrointestinal tract of sheep after oral administration of conidia in microencapsulated form and as a liquid in natura. Initial in vitro tests showed that both fungi were efficient in the predation of trichostrongylid L3 larvae present in the faeces of sheep naturally infected with gastrointestinal nematodes. The fungi presented high nematophagous activity, which was 99.3% for A. conoides and 73.7% for A. musiformis. A. conoides did not survive passage through the gastrointestinal tract under the conditions of the present experiment. On the other hand, A. musiformis was reisolated after administration in either microencapsulated or liquid form, suggesting that this species is a promising alternative for the control of nematodes in sheep since it survives without any protection (in natura). 相似文献

Radopholus similis and Meloidogyne spp. are the main nematode parasites of banana plants grown under plastic shelters in Morocco. A test was made in pots to evaluate the resistance of four genotypes of banana to these nematodes. Infection by Meloidogyne spp. brought about an increase in root weight in all banana plants tested because of gall formation. The inoculation of R. similis produced a reduction in length and diameter of the pseudo-trunk as well as in root and aerial mass in all genotypes. Pisang jari buaya showed the significantly lowest number of Meloidogyne nematodes per 10 g of roots, whereas for R. similis, the significantly smallest numbers were obtained in Pisang berlin and Pisang jari buaya. Therefore, Pisang jari buaya was the only banana genotype studied to show some degree of resistance to both nematodes. 相似文献

A genome‐wide scan for quantitative trait loci (QTL) affecting gastrointestinal nematode resistance in sheep was completed using a double backcross population derived from Red Maasai and Dorper ewes bred to F1 rams. This design provided an opportunity to map potentially unique genetic variation associated with a parasite‐tolerant breed like Red Maasai, a breed developed to survive East African grazing conditions. Parasite indicator phenotypes (blood packed cell volume – PCV and faecal egg count – FEC) were collected on a weekly basis from 1064 lambs during a single 3‐month post‐weaning grazing challenge on infected pastures. The averages of last measurements for FEC (AVFEC) and PCV (AVPCV), along with decline in PCV from challenge start to end (PCVD), were used to select lambs (N = 371) for genotyping that represented the tails (10% threshold) of the phenotypic distributions. Marker genotypes for 172 microsatellite loci covering 25 of 26 autosomes (1560.7 cm ) were scored and corrected by Genoprob prior to qxpak analysis that included Box–Cox transformed AVFEC and arcsine transformed PCV statistics. Significant QTL for AVFEC and AVPCV were detected on four chromosomes, and this included a novel AVFEC QTL on chromosome 6 that would have remained undetected without Box–Cox transformation methods. The most significant P‐values for AVFEC, AVPCV and PCVD overlapped the same marker interval on chromosome 22, suggesting the potential for a single causative mutation, which remains unknown. In all cases, the favourable QTL allele was always contributed from Red Maasai, providing support for the idea that future marker‐assisted selection for genetic improvement of production in East Africa will rely on markers in linkage disequilibrium with these QTL. 相似文献
