Bacterial population in Russian space station "Mir"   总被引:3,自引:0,他引:3  
We had the opportunity to investigate the bacterial population in air samples, condensation water, and inner wall swabs from the Russian space station Mir. From the first and second air samples during the mission, 29 and 7 bacterial colonies were collected, respectively. The values were equivalent to 16.8 and 4.0 cfu/100 liter air, respectively. Condensation water was collected from three different sites. The total viable bacterial counts were 2.1 x 10(6), 5.2 x 10(2), and 3.0 x 10(1) cfu/ml. The phylogenetic position of each isolate was determined by total 16S rDNA sequencing. Bacteria from air samples were mainly Gram-positive (35/36 colonies), and staphylococci occupied dominant specifically (23/36 colonies). On the other hand, Gram-negative bacteria were mainly isolated from condensation water samples. Most strains were thought to be opportunistic pathogens or environmental bacteria (such as those that inhabit soil, water, or air) found on earth. However, 6 of 23 isolates were suspected to be new species according to phylogenetic analysis and quantitative DNA-DNA hybridization data. The isolation of the other levels 3 and 2 bacteria, using specific selective media, was unsuccessful because all samples were heavily contaminated with fungi. To overcome this situation, PCR methods were applied to survey most levels 3 and 2 pathogenic bacteria in the condensation water samples. Up to 380 different primers for bacterial pathogens were used in this study. Only Mycobacterium avium 16S DNA sequences, however, could be amplified from the three water samples. The average bacteria count was estimated to be about 10(4) organisms/ml water.  相似文献

To provide a behavior-based estimate of odor similarity in larval Drosophila, we use 4 recognition-type experiments: 1) We train larvae to associate an odor with food and then test whether they would regard another odor as the same as the trained one. 2) We train larvae to associate an odor with food and test whether they prefer the trained odor against a novel nontrained one. 3) We train larvae differentially to associate one odor with food, but not the other one, and test whether they prefer the rewarded against the nonrewarded odor. 4) In an experiment like (3), we test the larvae after a 30-min break. This yields a combined task-independent estimate of perceived difference between odor pairs. Comparing these perceived differences to published measures of physicochemical difference reveals a weak correlation. A notable exception are 3-octanol and benzaldehyde, which are distinct in published accounts of chemical similarity and in terms of their published sensory representation but nevertheless are consistently regarded as the most similar of the 10 odor pairs employed. It thus appears as if at least some aspects of olfactory perception are "computed" in postreceptor circuits on the basis of sensory signals rather than being immediately given by them.  相似文献

随机选用121对分布于水稻12条染色体上的微卫星引物对经卫星搭载的水稻品种秋光突变后代(SP10)7个株系进行分子检测,结果发现突变系与亲本秋光之间存在着不同程度的多态性,多态性位点的多少随突变系与原种间差异大小而变化;多数突变系和原种相比都存在多个位点突变。实验结果表明空间诱变可使植物DNA分子多个区段内发生重复或缺失等结构性变异,其诱变机理和一般的诱变因素导致DNA少数碱基发生点突变不同。  相似文献

Membrane proteins comprise up to one-third of prokaryotic and eukaryotic genomes, but only a very small number of membrane protein structures are known. Membrane proteins are challenging targets for structural biology, primarily due to the difficulty in producing and purifying milligram quantities of these proteins. We are evaluating different methods to produce and purify large numbers of prokaryotic membrane proteins for subsequent structural and functional analysis. Here, we present the comparative expression data for 37 target proteins, all of them secondary transporters, from the mesophilic organism Salmonella typhimurium and the two hyperthermophilic organisms Aquifex aeolicus and Pyrococcus furiosus in three different Escherichia coli expression vectors. In addition, we study the use of Lactococcus lactis as a host for integral membrane protein expression. Overall, 78% of the targets were successfully produced under at least one set of conditions. Analysis of these results allows us to assess the role of different variables in increasing "expression space" coverage for our set of targets. This analysis implies that to maximize the number of nonhomologous targets that are expressed, orthologous targets should be chosen and tested in two vectors with different types of promoters, using C-terminal tags. In addition, E. coli is shown to be a robust host for the expression of prokaryotic transporters, and is superior to L. lactis. These results therefore suggest appropriate strategies for high-throughput heterologous overproduction of membrane proteins.  相似文献

To ensure safety of cosmonauts impacted by +Gx loads during insertion into orbit of a Soyuz-type space vehicle, it is critical to conduct appropriate medical selection of candidates to space flights, to give them proper complete physical and specialized centrifuge training, and to make it sure that at the moment of actual injection the cosmonaut's posture is optimal relative to the g-vector and that the body is tight fitting the custom-molded couch. No AGS is needed. For the purposes of protection of cosmonauts descending in the Soyuz vehicles, vital are countermeasures applied in the course of flight, water-salt supplements on the descent day, and the optimal posture, an anti-g suit, and a custom-molded couch with an additional body-restraint system and dampers used on the phase of reentry.  相似文献

Researchers report the results of experiments conducted onboard MIR in 1990, 1995, and 1996 in raising edible crops. In the 1990 experiment, radishes and Chinese cabbage were grown successfully, though the experimental plants were up to four times smaller than controls at harvest. The 1995 experiment in growing wheat through a complete life cycle was not completed. The 1996 experiment was successful in growing wheat through a complete life cycle to the seed stage. No seeds developed on any of the 279 ears harvested in that experiment. Reasons for the seedless development are explored.  相似文献

Previously, we demonstrated that mitochondrial NAD(P)H is the primary target of singlet oxygen (1O(2)) generated by photoactivation of mitochondria-selective rhodamine derivatives. Hence, local NAD(P)H oxidation/fluorescence decrease may be used to reveal the site of intracellular 1O(2) generation. Therefore, in addition to the previously used tetramethylrhodamine methylester (TMRM), 2('),4('),5('),7(')-tetrabromorhodamine 123 bromide (TBRB) and rhodamine 123 (Rho 123), we tested here whether mitochondrial NAD(P)H of cultured hepatocytes is directly oxidized upon irradiation of different "mitochondrial" photosensitizers (Photofrin; protoporphyrin IX; Al(III) phthalocyanine chloride tetrasulfonic acid; meso-tetra(4-sulfonatophenyl)porphine dihydrochloride; Visudyne). In contrast to TMRM and Rho 123, which directly oxidized NAD(P)H upon irradiation, irradiation of intracellular TBRB and the photochemical drugs only indirectly affected mitochondrial NAD(P)H due to loss of mitochondrial integrity. In line with this result only TMRM and Rho 123 exclusively localized within the mitochondrial matrix. Due to these results it is doubtful whether real mitochondrial photosensitizers actually exist among the photochemical drugs applicable/used for photodynamic therapy.  相似文献

A major goal of research in ecology and evolution is to explain why species richness varies across habitats, regions, and clades. Recent reviews have argued that species richness patterns among regions and clades may be explained by "ecological limits" on diversity over time, which are said to offer an alternative explanation to those invoking speciation and extinction (diversification) and time. Further, it has been proposed that this hypothesis is best supported by failure to find a positive relationship between time (e.g., clade age) and species richness. Here, I critically review the evidence for these claims, and propose how we might better study the ecological and evolutionary origins of species richness patterns. In fact, ecological limits can only influence species richness in clades by influencing speciation and extinction, and so this new "alternative paradigm" is simply one facet of the traditional idea that ecology influences diversification. The only direct evidence for strict ecological limits on richness (i.e., constant diversity over time) is from the fossil record, but many studies cited as supporting this pattern do not, and there is evidence for increasing richness over time. Negative evidence for a relationship between clade age and richness among extant clades is not positive evidence for constant diversity over time, and many recent analyses finding no age-diversity relationship were biased to reach this conclusion. More comprehensive analyses strongly support a positive age-richness relationship. There is abundant evidence that both time and ecological influences on diversification rates are important drivers of both large-scale and small-scale species richness patterns. The major challenge for future studies is to understand the ecological and evolutionary mechanisms underpinning the relationships between time, dispersal, diversification, and species richness patterns.  相似文献

The efficacy of countermeasure exercise for diminishing disturbances induced by microgravity in motor system and its visceral supply during different stages of long-duration flight was evaluated. The results of both bicycle and locomotor testing indicate that physical fitness of cosmonaut does not become worse in the course of the long-duration flight. On the contrary, the lowest fitness was recorded at the first stage of mission, just after one month of flight. The "dead period" at the beginning of space flight seems to be a manifestation of the acute decrease in physical condition on transition from 1 G to microgravity, when none of the regular countermeasure regimes is sufficiently effective and acute increase of volume and intensity of training is impossible under the conditions of space flight.  相似文献

Experiment on STS 51-C in January 1985, carried out on blood samples obtained from patients with heart disease, diabetes, hyperlipidaemia and cancer showed that, under zero gravity, the morphology of red cell aggregates aggregates was normal, in contradistinction to the parallel and simultaneous observations under 1 g, which showed large and unorientated clumps of red cells. As such clumps could be considered of disadvantage in the microcirculation and tissue perfusion, the zero gravity observations were significant in a number of ways. In particular, a preliminary deduction (subject to further zero g experimentation) was that cell-cell interaction and adhesion are affected by zero gravity, and that most likely the microarchitecture of the cell membrane is modified; and that probably the receptors, their position and/or activity, are affected by zero gravity. Of particular interest could be a possible change in the properties of the discrete surface areas which respond preferentially to specific macromolecules (or ligands). There is a dissonance between these in vitro results and theoretical deductions on flow in the microcirculations by Oka, and as well of deductions on space sickness by Dintenfass, both assuming a disabling effect of zero g on the in vivo microcirculation. This dissonance should be explored, as effect of zero g might be different on blood flow in vivo and in vitro. However, the data available from the in vitro experiment suggest that studies in immunology and oncology might be enriched by zero gravity findings; and that studies under zero gravity might open a new avenue of research in these important fields.  相似文献

Experiments have been performed to measure the cooling rates of air space and equatorial regions of the incubated egg. The air space has a low specific heat so that it cools faster than the rest of the egg when an incubating bird leaves the nest. This rapid cooling of the air space reduces the water loss that would occur when a bird leaves the nest by 50% when compared with the water loss from an equivalent area of the rest of the shell. The overall influence of the air space is probably to reduce water losses during incubation by 10–15%.  相似文献

A project on "Aggregation of Red Cells" has been accepted by NASA in 1977. An automated slit-capillary photo-viscometer has been designed during 1979-1984, and its last version met NASA's space hazards requirements. The 'heart' of instrument is a set of two highly polished glass plates, spaced by a gap of 12.5 micrometers. An original drum-like infusion pump allows utilization of up to eight blood samples. During a sequential process, blood flows through the slit, and then stops to allow formation of aggregates. Micro- and macro-photography is carried out, and 500 photographs are obtained. Blood from normal donors and patients with history of ischaemic heart disease, colon cancer, juvenile-onset diabetes, hyperlipidaemia, etc., is anticoagulated and adjusted to haematocrit of 0.30 using native plasma. Samples are divided, and infused into the 'flight' and 'ground' instruments. Prior to experiment temp. is 5 degrees C; temp. during experiment is 25 degrees C. Experiments took place on 24-25 January 1985, on the middeck of space shuttle 'Discovery'. Subsequent results showed that red blood cells do not change shape under zero gravity; that aggregation of red cells does take place; that aggregates in pathologic blood show morphology of normal rouleaux under zero gravity, while identical blood shows clumps of red cells on the ground. The latter observation suggests that zero gravity might affect cell-to-cell interaction, and perhaps microstructure of the cell membrane. These aspects must remain however tentative till a confirmation by subsequent experiments can be obtained.  相似文献

The present experiments were carried out to determine the effects of energy deposition from energetic iron (56Fe particles, an important component of cosmic rays) on motor behavioral performance and to determine if the observed deficits were caused by alterations in the neostriatum (an important motor control area). Neostriatal function was assessed with two correlated parameters, i.e., motor behavioral performance (wire suspension task), and oxotremorine-enhanced K(+)-evoked release of dopamine from perifused striatal slices. Rats were exposed to one of several doses of 56Fe-particle irradiation (0.10-1.0 Gy) and tested on a wire suspension task at 3-180 days postirradiation. Results indicated that profound decrements occurred in both of these indices. The effects on K(+)-evoked release of dopamine were evident for as long as 180 days after irradiation, and a subsequent experiment indicated that these effects appeared as early as 12 h postirradiation. Since similar findings have been observed in aged rats, the results are discussed in terms of these particles producing a possible accelerated striatal aging effect.  相似文献

We employed an associative learning paradigm to test the hypothesis that exercise hyperpnea in humans arises from learned responses forged by prior experience. Twelve subjects undertook a "conditioning" and a "nonconditioning" session on separate days, with order of performance counterbalanced among subjects. In both sessions, subjects performed repeated bouts of 6 min of treadmill exercise, each separated by 5 min of rest. The only difference between sessions was that all the second-to-penultimate runs of the conditioning session were performed with added dead space in the breathing circuit. Cardiorespiratory responses during the first and last runs (the "control" and "test" runs) were compared for each session. Steady-state exercise end-tidal PCO(2) was significantly lower (P = 0.003) during test than during control runs for both sessions (dropping by 1.8 +/- 2 and 1.4 +/- 3 Torr during conditioning and nonconditioning sessions, respectively). This and all other test-control run differences tended to be greater during the first session performed regardless of session type. Our data provide no support for the hypothesis implicating associative learning processes in the ventilatory response to exercise in humans.  相似文献
