A possible correlation between the presence of mesophilic aeromonads and the number of faecal coliforms present in three fresh-water habitats subject to differing levels of faecal pollution was investigated. Concentration of Aeromonas spp. between 10(2)-10(9) cfu/100 ml and faecal coliforms of between 9-10(7) cfu/100 ml were found in the waters. In water free from faecal pollution there was no correlation but in polluted waters there was a significant relationship between the numbers of aeromonads, faecal coliform and the concentration of organic matter measured by biological oxygen demand.  相似文献

The effect of releasing untreated sewage from Victoria Falls Town into the Zambezi river was determined by bacteriological examination of water samples collected upstream of Victoria Falls and for 22 km downstream. Most probable numbers of faecal coliforms and Escherichia coli were estimated. Water upstream of the falls, on the Zimbabwe side of the river, contained between seven and 130 E. coli per 100 ml. This section of the river was free from major sources of faecal pollution. Below the falls, but before the Victoria Falls Town sewage outfall, numbers of E. coli were between 1.8 × 102 and 1.4 × 104/100 ml, indicating the existence of a sewage discharge other than that from Victoria Falls Town. The river was also highly polluted from the Victoria Falls Town sewage outfall to a point 18.6 km downstream. The highest E. coli count was 3.3 × 104/100 ml and declined slowly to 1.4 × 103/100 ml 18.6 km downstream of the outfall.  相似文献

The microbiological quality of the River Riato (Spain) was evaluated. The influence of cattle that roam free in the warm season was marked. The degree of faecal pollution in the river was higher than predicted from the river basin geographical characteristics. The counts of faecal indicators greatly increased when the cattle were allowed to roam free. Counts of enterobacteria and faecal coliforms ranged from 103 to 106/100 ml. Faecal streptococci counts were smaller (< 10/100 ml). Escherichia coli and Pseudomonas aeruginosa were isolated from all samples. Streptococcus bovis was also isolated but not Strep. faecalis .  相似文献

The sludge from hospital waste treatment facilities is a potential source of infectious organisms. The average numbers of micro-organisms in the sludge of hospital wastewater in Taiwan were as follows: total count 8·1 × 107 cfu g−1 (dry weight of sludge), and 1·4 × 106, 3·6 × 105, 1·6 × 105, 2·2 × 105 and 5·5 × 104 cfu g−1 (dry weight of sludge) for total coliforms, faecal coliforms, faecal streptococci, Pseudomonas aeruginosa and Salmonella spp., respectively . Salmonella spp. were detected in 37% (10 of 27) of the sludges from hospital wastewaters. Therefore, the treatment of such sludge to reduce pathogenic micro-organisms should be considered.  相似文献

Thirty-nine samples of freshly drawn ewes' milk collected at 13 farms, and 120 samples of raw ewes' milk collected on arrival at a dairy were examined. Farm samples had geometric mean counts of 4.4 × 102 Enterobacteriaceae/ml, 3.9 × 102 coliforms/ml and 2.0 × 102 faecal coliforms/ml, whereas the respective mean counts were 6.2 × 103/ml, 5.4 × 103/ml and 1.3 × 103/ml for dairy samples. Salmonellas were not detected by enrichment procedures in any of the 159 samples examined. Escherichia coli (47.5% strains), Enterobacter cloacae (17.7%), Ent. agglomerans (11.3%), Hafnia alvei (6.5%) and Klebsiella oxytoca (6.0%) were the predominant species in 434 Enterobacteriaceae strains isolated from farm samples. Levels and species of Enterobacteriaceae found in the present work in raw ewes' milk imply a considerable risk of early blowing in cheese-making from unpasteurized milk.  相似文献

The efficacy of high-temperature, short-time (HTST) pasteurization (72 °C/15 s) when low numbers (≤ 103 cfu ml −1 ) of Mycobacterium paratuberculosis are present in milk was investigated. Raw cows' milk spiked with Myco. paratuberculosis (103 cfu ml−1, 102 cfu ml−1, 10 cfu ml−1, and 10 cfu 50 ml−1) was subjected to HTST pasteurization using laboratory pasteurizing units. Ten bovine strains of Myco. paratuberculosis were tested in triplicate. Culture in BACTEC Middlebrook 12B radiometric medium detected acid-fast survivors in 14·8% and 10% of HTST-pasteurized milk samples at the 103 and 102 cfu ml−1 inoculum levels, respectively, whereas conventional culture on Herrold's egg yolk medium containing mycobactin J detected acid-fast survivors in only 3·7% and 6·7% of the same milk samples. IS900-based PCR confirmed that these acid-fast survivors were Myco. paratuberculosis . No viable Myco. paratuberculosis were isolated from HTST-pasteurized milk initially containing either 10 cfu ml−1 or 10 cfu 50 ml−1.  相似文献

Abstract A method was developed for direct extraction, purification and amplification of DNA from forest soil. Eighty-two % of the DNA in Pseudomonas aeruginosa UG2Lr introduced into soil was recovered. The detection limit for the strain was approximately 800 cfu g−1 of dry soil based on the polymerase chain reaction (PCR). Survival of κ-carrageenan-encapsulated and unencapsulated UG2Lr was monitored by antibiotic selective and bioluminescence-based nonselective plating and PCR-amplification of a tnsA fragment. After freeze-thaw treatment of soil samples, the unencapsulated UG2Lr declined from an initial population density of 1 × 109 cfu g−1 of dry soil to below the detection threshold of both selective (14 cfu g−1 of dry soil) and nonselective (1 × 103 cfu g−1 of dry soil) plating. However, presence of nonculturable UG2Lr cells in the soil was revealed by PCR and resuscitation of the bacteria. Population density of the encapsulated UG2Lr increased from 2.7 × 106 to 2.9 × 108 cfu g−1 of dry soil after a 3-week incubation at 22°C and declined to 6.3 × 106 cfu g−1 of dry soil after the freeze-thaw treatment.  相似文献

C.M. DAVIES and S.C. APTE.2000. A 1-h fluorimetric assay of β- d -galactosidase activity was evaluated for determining thermotolerant coliforms (TTC) in sewage samples. Above TTC concentrations of 2·3 × 103 colony-forming units (cfu) 100 ml−1, the assay response was related to TTC concentration. However, below this concentration, a large background signal was observed which was independent of TTC concentration. A separation scheme involving various filtration treatments and additions of a β- d -galactosidase inhibitor was devised and used to quantify the sources of this anomalous assay response. The interferences encountered were largely due to the presence in sewage of non-specific cell-free enzymes or other cell-free substances that were capable of hydrolysing the fluorogenic substrate. Despite this apparent limitation, the fluorimetric enzyme assay has potential as an 'early warning' indicator of treatment process failure and gross sewage contamination and leakage in situations where TTC concentrations exceed 2·3 × 103 cfu 100 ml−1  相似文献

Bacteria grew to viable populations of 108–109 cfu/g during the fermentation of soybeans into tempe with the fungus, Rhizopus oligosporus. Bacillus pumilus and B. brevis were the predominant bacterial species, reaching populations of approximately 108 cfu/g during the 48 h fermentation. Species of Streptococcus faecium, Lactobacillus casei, Klebsiella pneumoniae and Enterobacter cloacae also contributed to the fermentation and achieved populations of 106–107 cfu/g. and accepted 25 May 1989  相似文献

A.O. EJIOFOR AND N. OKAFOR. 1991. Bacillus thuringiensis serotype H-14 spores and crystals, produced in 5 1 fermenters, were centrifuged and resuspended in emulsified palm olein to give 3.2 × 1011 colony forming units (cfu)/ml. The suspension was mixed with a cassava—molasses—palm olein—charcoal (CMPC-2) mixture which served as the carrier, adhesive, dispersant and protectant. The final concentration of the formulation was 3.2 × 109 cfu/ml. The lethal concentrations capable of killing 50% of the test population (LC50) of CMPC-2 during 0, 1 and 2 years of storage at 32 ± 4.C were 0.056, 0.058 and 0.058 mg/ml respectively as against 0.054, 0.051 and 0.054 mg/ml for the Institut Pasteur Standard—1978 (IPS-78) during the corresponding period. The2 tests showed that the results were homogeneous at P = 0.05. The relative potencies of the preparation were 964.3, 879.3 and 931 International toxic units (ITU) Aedes aegypti as compared with the 1000 ITU assigned to IPS-78. At 95% confidence limits there was no significant difference between the potencies of CMPC-2 and IPS-78. Field tests showed that CMPC-2 provided between 87.5 and 100% control of natural populations of Aedes spp. and Cutex spp. Sedimentation tests showed that CMPC-2 settled markedly during storage. This, therefore, required that the product be thoroughly shaken before use.  相似文献

Salad vegetables are often consumed fresh and thus can act as effective media for the transmission of associated pathogens. Fresh samples of cucumber, carrot and lettuce were collected from different markets in Dhaka metropolitan city, Bangladesh. Bacterial loads were found to be 7·1 times 104 to 6·34 times 108 colony forming unit (cfu)/100 g. Escherichia coli, Klebsiella, Enterobacter , and Serratia were amongst the coliforms (lactose fermenters) while Pseudomonas, Shigella and Acinetobacter were non-lactose fermenters associated with the samples.  相似文献

Methane-producing, cellulolytic, feather-degrading, and total anaerobic microbial populations were enumerated in four laboratory-scale (l l) thermophilic (50°C) poultry waste digesters over a 40d period. Four different operation conditions were: 5 d retention time (RT), 6% volatile solids (VS); 5 d RT, 3% VS; 10 d RT, 6% VS; and 10 d RT, 3% VS. Laying hen manure was the sole source of substrate and micro-organisms. At theoretical steady state (day 40) the biogas volumetric rate was near 3.0 l/l digester volume (l/l/d) in all but the 10 d RT, 3% VS digester which was 2 l/l/d. The total viable anaerobic population was > 106 cfu/ml digester fluid at the first sampling and stabilized at 107–108 cfu/ml between days 20 and 40 in all digesters. Methane-producing bacteria increased from ≤ 10/ml early in the sampling period to 105/ml at steady state in all but the 5 d RT, 3% VS digester which was highest at 107/ml. Cellulolytic micro-organisms were low throughout the 40 d, generally less than 10/ml. Feather-degrading micro-organisms ranged from near 102–105 at steady state and were decreasing in number near day 40 in all but the 10 d RT, 6% VS digester which maintained 105/ml after day 20. A feather-degrading culture was enriched from this digester and subsequently adapted to grow in a medium with feather as the sole source of carbon. Results of this study provide information regarding potential biological upgrading of poultry waste digesters for increased operational efficiency and potential industrial application of a feather-hydrolytic micro-organism.  相似文献

The relationships between total coliform, faecal coliform, enterococci and salmonella concentrations were investigated at Berry and Howard Pools in the Australian wet/dry tropics. Both pools have catchments with minimal human activity and no major point source of faecal pollution. Forty-five indicator and salmonella enumerations were made from each pool over a 1 year period. Salmonellas were isolated from 69% and 96% of samples collected from Howard and Berry Pools respectively, the maximum (MPN) concentration was 110/100 ml. Native fauna were the primary salmonella source. Spearman rank correlations between indicator organisms and salmonella at Howard Pool were significant at the 5% level and approximated 0.6. At Berry Pool, total coliform and enterococci Spearman rank correlations with salmonella were also statistically significant, approximating 0.3; faecal coliforms and salmonella rankings, however, were unrelated. The higher correlation coefficients at Howard Pool were attributed to its small catchment (4 km2) and the more recent nature of faecal contamination compared with Berry Pool which has a catchment of 130 km2. The results highlight the spatial variability of the indicator/pathogen numerical relationship. Total coliform and enterococci counts, as indicators of faecal pollution, were similar and more consistent than faecal coliforms.  相似文献

The microbiological quality and heterotrophic bacterial populations of 26 thermal mineral water springs in Spain were studied. In most of the springs the number of viable aerobes was less than 103 cfu ml-1 and the number of sporulated bacteria less than 102 cfu ml-1. No significant differences were foundin the counts obtained with Plate Count Agar (PCA) and PCA diluted 1 : 10 and incubated at 22°, 37° and 45°C. Total coliforms were found in 14 springs, faecal streptococci in three, spores of sulphite-reducing Clostridium and Pseudomonas aeruginosa in seven. Neither Escherichia coli nor Staphylococcus aureus were found. A total of 665 strains were isolated and 85·4% of these identified; 329 were Gram-positive and 239 were Gram-negative. The genera most prevalent present in the springs were Pseudomonas (in 92.3%), Bacillus (65.4%), Enterobacter, Micrococcus and Staphylococcus (50%), Acinetobacter (42.3%), Arthrobacter (38.4%), Clostridium (27%) and Xanthomonas (23%). Gram-negative bacteria predominated in the mesothermal springs and Gram-positive bacteria in the hyper- and hypothermal springs. The most common Gram-negative rod species isolated were Ps. fluorescens, Ps. aeruginosa, Ps. putida, Ent. agglomerans, Ent. sakazakii, Ac. calcoaceticus and Ent. amnigenus.  相似文献

In a 29 month study of bacterial populations at three sites on the Welsh River Dee, the aerobic heterotrophic bacteria increased from an average basal level of about 1.2 times 104 colony-forming units (cfu)/ml near the source of the river, to 2 times 105 cfu/ml in the lower reaches. The ratio, total bacterial cell count: viable count, decreased from 70 in the upland reaches to 10 in the lower parts of the river. There was no apparent seasonal variation in bacterial numbers but on occasions the bacterial populations in both upland and lowland reaches of the river increased simultaneously. Fluctuations in bacterial numbers over a 50-fold range were observed in this study. Bacterial isolates from both upland and lowland sites were predominated by two groupings of bacteria, the Pseudomonas—Agrobacterium—Alcaligenes group and the Flavobacterium—Cytophaga—Flexibacter group. Results suggested that the latter group may have been part of the autochthonous population. Seasonal variation in heterotrophic potential ( V max) for acetate uptake was shown to occur over a 30-fold range in the lowland reaches of the River Dee. Peaks in activity at the lowland site occurred during the summer months, the range of V max values for acetate ranged from 0.2 to 30 μg/1/h. Fluctuations in V max values from the upland site were not seasonal but were instead linked to faecal pollution, V max values from this site range from 0.04 to 3 μg/1/h.  相似文献

Burgos cheese was manufactured from pasteurized ewes milk inoculated with Staphylococcus aureus strains FRI 137 and FRI 361, at levels of ca 103 and 105 cfu/ml and stored at 4°, 10° and 15°C and at room temperature (10°-15°C). Populations of Staph. aureus and mesophilic aerobes, pH, and production of thermonuclease and enterotoxins C1 and C2 were investigated. Aerobic counts increased during cheese-making and storage. With both test strains, important growth was observed only during the storage period, the larger levels corresponding to the higher temperatures. Although Staph. aureus strains attained populations of over 108 cfu/g, no enterotoxin was detected. Strain FRI 361 reached 107 cfu/g without production of a detectable amount of thermonuclease. With strain FRI 137, the minimal population associated with enzyme activity was influenced by the inoculum size. Staphylococcus aureus counts are better indicators of staphylococcal growth in Burgos cheese than the thermonuclease test.  相似文献

The in vitro antimicrobial activity of geraniol and citronellol towards seven strains of Erwinia amylovora , the causal agent of 'fire blight'of Rosaceous plants, was assessed in tube cultures. All of the strains tested at 1 × 105 cfu/ml were inhibited for 24 h by geraniol in the range 600–1500 mg/1, whereas its minimum bactericidal concentration was 800–1700 mg/1. Citronellol was less effective, being bactericidal for only two of seven strains. RIF-NY, isolated from apple orchards, was relatively resistant to geraniol; 1700 mg/1 of the chemical only reduced the growth of an inoculum of 1 × 107 cfu/ml. In general, such terpenoids commenced exerting a bactericidal effect 6 h after addition to the suspensions, even if geraniol added at 1700 mg/1 to 1 × 103 cfu/ml of five strains, commenced its bactericidal activity earlier than 6 h.  相似文献

Four media were tested for their ability to detect the soft rot potato pathogens Erwinia chrysanthemi (Ech) and Erwinia carotovora ssp. atroseptica (Eca) in potato tubers by means of automated conductance measurements. The specificity of the conductimetric assays was determined by testing a set of different Erwinia spp. and potato-associated saprophytes, including the genera Pseudomonas, Bacillus, Enterobacter and Flavobacterium. All bacteria tested produced conductance responses in Special Peptone Yeast Extract, whereas in minimal medium with L-asparagine only Erwinia spp. and Pseudomonas spp. were able to generate large conductance responses. In minimal medium supplemented with glucose and trimethylamine- N -oxide only Enterobacteriaceae, Erwinia spp. included, generated conductance responses, while with pectate as sole carbon source only Erwinia spp. produced distinct conductance responses. The pectate medium proved to be particularly useful for specific automated conductimetric detection of Erwinia spp. in potato peel extracts. Within 48 h, the detection threshold of the conductimetric assay for Eca varied between 102 and 103 cfu per ml peel extract at both incubation temperatures of 20° and 26°C. Ech was detected at concentrations of 104–105 or 103–104 cfu ml-1 at 20° and 26°C, respectively. To eliminate 'false'-positive reactions in conductimetry caused by Erwinia carotovora ssp. carotovora , results of the conductance measurements have to be confirmed by other techniques, like serology or DNA assays.  相似文献

Abstract The fate of a Bacillus amyloliquefaciens with the recombinant plasmid pSB20 sprayed on the phyllosphere of grass, and of a Tn 5 marked Pseudomonas syringae sprayed on the phyllosphere of bush beans was studied in planted soil microcosms. B. amyloliquefaciens showed a decline from 1.5×108 to 3.1×102 cfu g−1 on the phylloplane of grass in the course of the experiment. B. amyloliquefaciens was easy to follow by selective cultivation due to the complete absence of bacterial background growth. Southern blot hybridization of Hin dIII digested genomic DNA showed plasmid restriction patterns identical with pSB20 indicating high plasmid stability. In total DNA extracts from phyllosphere bacteria the recombinant plasmid was detectable by Southern blot hybridization up to 6×104 cfu g−1 (wet weight). Counts of hybridizing colonies showed that P. syringae established on the phyllosphere of bush beans at between 5×103 and 4×106 cfu g−1 fresh weight. During senescence of the bean plants the strain was no longer detectable by selective cultivation and subsequent colony hybridization. In contrast, Tn5 marked DNA was detected after PCR amplification over the whole period of the experiment.  相似文献

Abstract The distribution of heterotrophic bacteria in polluted coastal and unpolluted pelagic seawaters was studied using a 14C-MPN method with either five of seven kinds of 14C-organic compounds as substrates. The total number of heterotrophic bacteria in pelagic waters ranged from 9.2 × 103 to 5.4. ¢ 104 cell/ml and more than 85% of the heterotrophic bacteria were represented by obligate oligotrophs. In coastal waters, the number of heterotrophs was one order of magnitude higher (av. 3.5 ¢ 105 cells/ml), and eutrophic and facultatively oligotrophic bacteria were predominant. Oligotrophs in pelagic waters had a high specificity for the utilization of amino acids, especially glycine, and acetate-utilizing bacteria were scarce. The in situ maximum uptake rates of glutamate and glycine were much higher than those of glycolate and acetate. Acetate uptake rates were extremely low or not detectable in pelagic waters. The specificity of uptake kinetics is assumed to depend on the existence of obligate oligotrophs as dominant bacteria in pelagic seawater.  相似文献
