At present there are a large number of people capable of conducting the task of surface and area radiation monitoring including external monitoring of personnel. Once the extent and the intensity of radioactivity in an area is determined, good use of personnel can be made without too much risk. This is fortunate for the medical profession whose personnel can devote their talents to casualty care during or following nuclear warfare. Most individuals who know how to detect and measure the extent of radioactive contamination are also capable of conducting personnel decontamination operations and would do so if necessary. Consequently the spread of contamination can be minimized by adequate decontamination and the medical personnel can treat casualties who are relatively free of external radioactive contamination. The appropriate use of trained manpower and radiation detection equipment which are available in California combined with sufficient rehearsals prior to a nuclear war will greatly reduce any casualty damage due to radioactive fallout. The chances of survival of individuals can be greatly improved with a little knowledge of protection from radioactive contamination and of salvage of food and water.  相似文献

Finite element modelling of contracting skeletal muscle   总被引:2,自引:0,他引:2
To describe the mechanical behaviour of biological tissues and transport processes in biological tissues, conservation laws such as conservation of mass, momentum and energy play a central role. Mathematically these are cast into the form of partial differential equations. Because of nonlinear material behaviour, inhomogeneous properties and usually a complex geometry, it is impossible to find closed-form analytical solutions for these sets of equations. The objective of the finite element method is to find approximate solutions for these problems. The concepts of the finite element method are explained on a finite element continuum model of skeletal muscle. In this case, the momentum equations have to be solved with an extra constraint, because the material behaves as nearly incompressible. The material behaviour consists of a highly nonlinear passive part and an active part. The latter is described with a two-state Huxley model. This means that an extra nonlinear partial differential equation has to be solved. The problems and solutions involved with this procedure are explained. The model is used to describe the mechanical behaviour of a tibialis anterior of a rat. The results have been compared with experimentally determined strains at the surface of the muscle. Qualitatively there is good agreement between measured and calculated strains, but the measured strains were higher.  相似文献

Background

Systematic approaches for identifying proteins involved in different types of cancer are needed. Experimental techniques such as microarrays are being used to characterize cancer, but validating their results can be a laborious task. Computational approaches are used to prioritize between genes putatively involved in cancer, usually based on further analyzing experimental data.  相似文献

Complete sequencing of the oligosaccharide units of glycoproteins can be performed by conventional physical techniques if nanomole quantities of pure molecules are available. However, isolation of sufficient quantities of a glycoprotein may not be technically feasible (e.g., the analysis of biosynthetic intermediates, or rare molecules). Alternatively, partial structural analyses may answer the biological question at hand. In both instances, radioactive sugars can be used to metabolically label the oligosaccharide units of a glycoprotein, permitting substantial structural characterization. Several aspects of this approach are discussed in this overview, including selection of the labeled precursor, maximization of uptake and incorporation, determinants of the specificity of labeling, and general principles for the release and structural analysis of labeled oligosaccharides. Particular advantages include simplicity, ease of use without sophisticated instrumentation, and the fact that purification to radiometric homogeneity is sufficient. Radioactive tracer techniques cannot replace conventional approaches to sequencing oligosaccharides. However, they do provide a rapid, relatively simple approach to obtaining considerable information from limited amounts of material. For molecules such as short-lived biosynthetic intermediates, there is no substitute for these techniques. This approach has been responsible for the initial identification and characterization of many novel oligosaccharides of biological interest.  相似文献

Postoperative bronchopleural fistulas may be demonstrated quickly and safely by inhalation of nebulized 99mTc-albumin, or other similar radiopharmaceuticals. Posterior scintiphotographs of the distribution of the inhaled radioactive material are obtained with the gammaray scintillation camera while the patient is in the upright and lateral decubitus positions. The observation of radioactive material in the pleural cavity indicates an abnormal communication. The radiation-absorbed doses for lung and total body are well within acceptable limits when a 99mTc tag is used. About 15 minutes are required to complete this relatively safe diagnostic test.  相似文献

A portable meter has been developed for measuring low frequency currents that flow in the human body. Although the present version of the meter was specifically designed to measure 50/60 Hz "contact currents," the principles involved can be used with other low frequency body currents. Contact currents flow when the human body provides a conductive path between objects in the environment with different electrical potentials. The range of currents the meter detects is approximately 0.4-800 microA. This provides measurements of currents from the threshold of human perception (approximately 500 microA(RMS)) down to single microampere levels. The meter has a unique design, which utilizes the human subject's body impedance as the sensing element. Some of the advantages of this approach are high sensitivity, the ability to measure current flow in the majority of the body, and relative insensitivity to the current path connection points. Current measurement accuracy varies with the accuracy of the body impedance (resistance) measurement and different techniques can be used to obtain a desired level of accuracy. Techniques are available to achieve an estimated +/-20% accuracy.  相似文献

Depending on the microbiological quality, it may be necessary to reduce the number of microorganisms on the packing material before use with food products. Treatment with hydrogen peroxide at elevated temperatures is most commonly used. Residues of hydrogen peroxide, however, are undesirable. Systems not using chemicals obviously would be preferable. For products which do not allow the growth of bacteria, such as many acid products and products with a low water activity, often it is sufficient to inactivate moulds and yeasts. Moulds and yeasts can be inactivated by temperatures below 100°C provided the water activity is high enough. At low humidities at the same temperatures hardly any reduction in viable count is obtained. A prototype machine was built to investigate the inactivation of microorganisms on the surface of packing material, using moist heat for a short time, similar to the time needed for decontamination by peroxide. The number of viable dry spores of Penicillium roqueforti can be reduced by a factor of >1000 within 3 s at 90°C and 100% humidity. Moist heat decontamination is a promising method which could help manufacturers pack food in a microbiologically safe manner, without the use of chemicals. Further work is needed, however, to determine the inactivation of other relevant microorganisms. Received 09 September 1996/ Accepted in revised form 31 January 1997  相似文献

Quantification of the mechanical behavior of hyperelastic membranes in their service configuration, particularly biological tissues, is often challenging because of the complicated geometry, material heterogeneity, and nonlinear behavior under finite strains. Parameter estimation thus requires sophisticated techniques like the inverse finite element method. These techniques can also become difficult to apply, however, if the domain and boundary conditions are complex (e.g. a non-axisymmetric aneurysm). Quantification can alternatively be achieved by applying the inverse finite element method over sub-domains rather than the entire domain. The advantage of this technique, which is consistent with standard experimental practice, is that one can assume homogeneity of the material behavior as well as of the local stress and strain fields. In this paper, we develop a sub-domain inverse finite element method for characterizing the material properties of inflated hyperelastic membranes, including soft tissues. We illustrate the performance of this method for three different classes of materials: neo-Hookean, Mooney Rivlin, and Fung-exponential.  相似文献

The selectivity of chitosan has been modified through metal ion imprinting technique for its potential application in nuclear industry. Considerable reduction in radioactive waste volume, generated during the chemical decontamination of nuclear power plants, can be achieved through the selective removal of the radionuclides. In this context, a Co(II) imprinted chitosan was synthesized using epichlorohydrin as the crosslinker. The selective removal of Co(II) in presence of Fe(II), which is the major non-radioactive ion present in excess during decontamination, was studied. The imprinted chitosan showed selective sorption of Co(II) over Fe(II), while the raw chitosan was selective to Fe(II) over Co(II). The imprinted chitosan was found to retain the enhanced selectivity towards Co(II) under various solution conditions, including typical nuclear reactor decontamination formulations containing strong complexants. The highest uptake by the imprinted chitosan, with maximum selectivity for Co(II) over Fe(II), was obtained in citrate medium at pH 4.8.  相似文献

Interest in the study of yeast biology has increased dramatically in the past few years. Since these organisms are eukaryotic, some phenomena observed in yeast may provide a useful model for similar phenomena in multicellular organisms. Yeast has several advantages as an experimental organism and many methods used for bacteria can be adapted to them. Yeast is simple to grow, cultures are easily maintained, and classical and molecular genetic techniques can be used. The ability to approach problems genetically and biochemically has lead to substantive progess with this group of organisms in areas such as cell biology (1) and gene expression (2). This review is intended to introduce investigators to practical techniques for the growth and radioactive labeling of yeast, primarily of Saccharomyces cerevisiae. For genetic techniques, readers are referred to a recent laboratory manual (3) and reviews (4,5).  相似文献

In Plodia interpunctella, radioactive labelling techniques indicate that male-derived substances, transferred to the female during copulation, enter unfertilized eggs. These substances are proteinaceous and the cumulative amount of material entering the unfertilized eggs increases with time since mating. Following courtship, smaller males complete coupling with females at a lower frequency than larger males. Smaller males also transfer smaller (by weight) ejaculates than larger males. Ejaculate weight is about 4% of male body weight in P. interpunctella. However, neither fecundity nor the number of deposited eggs are a function of ejaculate weight. Consequently, ejaculate materials cannot be designated a form of paternal investment, despite the observed transfer to unfertilized eggs.  相似文献

Uranium has been defined in material research engineering field as one of the most energetic radioactive elements in the entire Mendeleev periodic table. The manipulation of uranium needs higher theories and sophisticated apparatus even in nuclear energy extraction or in many other chemical applications. Above the nuclear exploitation level, the chemical conventional approaches used, require a higher temperature and pressure to control the destination of ionic form. However, it has been discovered later that at biological scale, the manipulation of this actinide is possible under friendly conditions. The review summarizes the relevant properties of uranium element and a brief characterization of nanoparticles, based on some structural techniques. These techniques reveal the common link between chemical approaches and biological assistance in nanoparticles. Also, those biological entities have been able to get it after reduction. Uranium is known for its ability to destroy ductile materials. So, if biological cell can really reduce uranium, then how does it work?  相似文献

Dissimilatory metal reducing bacteria (DMRB) widely exist in the subsurface environment and are involved in various contaminant degradation and element geochemical cycling processes. Recent studies suggest that DMRB can biosynthesize metal nanoparticles during metal reduction, but it is unclear yet how such biogenic nanomaterials would affect their decontamination behaviors. In this study, we found that the dechlorination rates of carbon tetrachloride (CT) by Shewanella putrefaciens CN32 was significantly increased by 8 times with the formation of biogenic ferrous sulfide (FeS) nanoparticles. The pasteurized biogenic FeS enabled 5 times faster dechlorination than abiotic FeS that had larger sizes and irregular structure, confirming a significant contribution of the biogenic FeS to CT bioreduction resulting from its good dispersion and relatively high dechlorination activity. This study highlights a potentially important role of biosynthesized nanoparticles in environmental bioremediation.  相似文献

6-(2,3,4-trihydroxy-3-methylbutylamino) purine (trihydroxyzeatin) applied to soybean callus is metabolised slowly. After 48 h only one peak of biological activity which co-eluted with the applied cytokinin was detected. When the callus was incubated on a medium which contained 10–5 M trihydroxyzeatin, spiked with 8 {14C} trihydroxyzeatin, for 28 days, three peaks of biological activity and three peaks of radioactivity were detected. One of the biologically active and radioactive peaks co-eluted with zeatin. Another of the radioactive peaks co-eluted with N-(purin-6-yl) glycine. From the data obtained it apears that trihydroxyzeatin can be both oxidized and reduced by soybean callus. The potential to be converted to zeatin may explain why trihydroxyzeatin and its parent compound, which is usually rapidly metabolised by living material, are equally active in the soybean callus bioassay. From the radioactive data obtained it appears that trihydroxyzeatin is susceptible to oxidation to form N-(purin-6-yl) glycine.  相似文献

Evidence to inform decontamination practices at Ebola holding units (EHUs) and treatment centres is lacking. We conducted an audit of decontamination procedures inside Connaught Hospital EHU in Freetown, Sierra Leone, by assessing environmental swab specimens for evidence of contamination with Ebola virus by RT-PCR. Swabs were collected following discharge of Ebola Virus Disease (EVD) patients before and after routine decontamination. Prior to decontamination, Ebola virus RNA was detected within a limited area at all bedside sites tested, but not at any sites distant to the bedside. Following decontamination, few areas contained detectable Ebola virus RNA. In areas beneath the bed there was evidence of transfer of Ebola virus material during cleaning. Retraining of cleaning staff reduced evidence of environmental contamination after decontamination. Current decontamination procedures appear to be effective in eradicating persistence of viral RNA. This study supports the use of viral swabs to assess Ebola viral contamination within the clinical setting. We recommend that regular refresher training of cleaning staff and audit of environmental contamination become standard practice at all Ebola care facilities during EVD outbreaks.  相似文献

Approximately 10-15% of couples experience infertility and male factors contribute to half of these cases. It was usually thought that infertility cannot be transmitted, but accumulating evidence indicates that many cases are indeed caused by genetic defects, some inherited. The use of single nucleotide polymorphisms (SNP) arrays allowing to genotype the totality of the genome recently led to identify several genes which, when mutated, generate specific infertility phenotypes. With the tremendous progresses in high throughput sequencing techniques, we can expect many more new genes involved in fertility to be identified in the next years. For the patients concerned, these findings mean the possibility of an accurate diagnosis and improved prognosis. Furthermore, these data will lead to a better understanding of the molecular mechanisms underlying spermatogenesis and thus should contribute to identify and offer new therapeutic strategies for the treatment of infertility.  相似文献

The concepts of radiation protection in survival operations are explained, and procedures devised to control radiation hazards for the protection of the population and maintenance of the operating efficiency of survival operations personnel are presented.Radiation protection is a command function. The medical responsibility is to provide advice on the probable effects of radiation exposure in the light of existing knowledge of these effects in man.The major hazard is that of external exposure to penetrating gamma radiation. Radiation exposure guides indicate that persons may be exposed to not more than 100 r whole body radiation in a six-week period, or 200 r whole body radiation in a period in excess of six weeks, without loss of operational efficiency. Beta radiation from fallout deposited on skin or clothing may produce burns, but these injuries will not be incapacitating and can be controlled by simple procedures. The internal hazard is mainly from ingestion of food or water contaminated with radioactive material. For protection, only canned or packaged foods and water from covered or deep wells are consumed during the early days after a nuclear attack.  相似文献

An incident in the course of the shipment of radioactive material could have serious consequences for affected states, whether or not a release of radiation actually resulted from the incident. In particular, small island states could potentially suffer serious damage to their reputations and to their economies. Pacific Island states are taking an initiative in response to their exposure to this risk. Assurances that they will not be left unsupported in such circumstances are being sought through a process of direct discussions presently taking place with source states and operators involved in the shipment of radioactive material through the Pacific.

This article examines the legal background to these discussions, investigating the legal uncertainties in the situation of states affected by an incident in the course of shipment of radioactive material that arise from limitations in the nuclear civil liability conventions and other aspects of applicable international law.  相似文献

Magnetic chitosan composite particles with 40 μm average size and 24 emu/g saturation magnetization obtained by an in situ procedure were evaluated as a new low-cost adsorbent for radioactive wastewater decontamination. Sorbent characterization by SEM, EDX, FTIR and magnetization measurements proved that the target ions were bound and their surface distribution was uniform. The 18 emu/g magnetization of the metal loaded particles was high enough to ensure their easy magnetic field separation and recovery. The parameters influencing the sorption process were optimized with respect to sorbent mass, target ion concentration and contact time. The material under study had superior adsorption capacity both for uranyl (666.67 mg/g) and thorium (312.50 mg/g) ions when compared to other low-cost adsorbents reported in literature. The adsorption process is spontaneous and endothermic. The material may be regenerated and re-used.  相似文献

Previous work has shown that the principal alpha-tubulin within Chlamydomonas reinhardtii flagellar axonemes differs from the major alpha-tubulin in the cell body. These two variants of alpha-tubulin are related to one another since posttranslational modification of the cell body form converts it to the axonemal form. When flagella are induced to assemble in the absence of de novo protein synthesis, tritiated acetate can be used to posttranslationally label alpha-tubulin in vivo, and under these conditions, no other flagellar polypeptides exhibit detectable labeling [L'Hernault, S. W., & Rosenbaum, J. L. (1983) J. Cell Biol. 97, 258-263]. In the present report, this labeling method has been used to provide material for chemical analysis of the tritiated moiety that is posttranslationally added to flagellar alpha-tubulin. This radioactivity was volatile after acid hydrolysis, suggesting that the posttranslational modification is the addition of neither an amino acid nor carbohydrate. Treatment of posttranslationally 3H-labeled alpha-tubulin with hydrazine yields radioactive acetylhydrazine, indicating that the moiety involved in posttranslational modification is an acetyl group. Analysis of complete proteolytic digests by thin-layer chromatography has revealed that this acetyl group is located on the epsilon-amino group of a flagellar alpha-tubulin lysine residue.  相似文献
