目的探讨n-3、n-6多不饱和脂肪酸(PUFA)高脂饮食对SD大鼠肝脏Srebp2及Hmgcr表达的影响。方法 SD大鼠随机分为3组,分别为对照组(NC)、n-3 PUFA组(TUF)及n-6 PUFA组(SUF),8周后测量血清总胆固醇(TCH)水平,实时定量PCR检测Srebp2及Hmgcr基因表达,Western blotting检测SREBP2及HMGCR蛋白表达。结果 TUF组、SUF组血清TCH含量均显著降低(P<0.001),TUF组最低。TUF组、SUF组Srebp2及Hmgcr基因表达水平均显著增高(P=0.007,P=0.012),SUF组Srebp2最高,TUF组Hmgcr最高。TUF组HMGCR蛋白表达显著高于NC组及SUF组,SUF组与NC组差异无显著性;三组SREBP2蛋白表达。结论 n-3、n-6PUFA降低血清胆固醇不是通过抑制Srebp2及Hmgcr表达实现的。  相似文献

四个罗非鱼选育品种抗链球菌病能力差异研究   总被引:1,自引:0,他引:1  
为筛选出抗病力优良的罗非鱼品种, 以奥利亚罗非鱼“夏奥1号”、尼罗罗非鱼“99”埃及品系、吉富罗非鱼“中威1号”和奥尼罗非鱼为研究对象, 33℃水温暂养7d后分别进行无乳链球菌人工感染实验, 连续7d统计累计死亡率, 并于人工感染后0、24h、48h和72h采集血液和组织样本, 研究这4个罗非鱼选育品种抗链球菌病能力的差异。结果显示: 感染7d后奥尼罗非鱼的累计死亡率最低; 奥尼罗非鱼的谷草转氨酶(AST)感染前后始终都低于其余3个品种罗非鱼(P<0.05); 埃及尼罗和奥尼在感染72h后球蛋白(GLO)分别显著升高1.13倍和1.41倍; 奥尼罗非鱼白蛋白/球蛋白(A/G)在感染前后没有显著性变化(P>0.05), 而其余3个品种罗非鱼A/G比值在感染后都显著性降低(P<0.05); 埃及尼罗的碱性磷酸酶(AKP)在感染72h后显著降低(P<0.05), 奥利亚和吉富的AKP表现为先上升后下降, 奥尼的AKP感染前后没有显著性变化(P>0.05); 各品种罗非鱼血清中的乳酸脱氢酶(LDH)感染后都显著升高(P<0.05); 奥利亚、吉富和奥尼罗非鱼的超氧化歧化酶(SOD)感染48h时都显著升高(P<0.05); 奥尼罗非鱼在感染前后溶菌酶(LZM)活性都显著高于其余3个品种罗非鱼(P<0.05)。组织病理学结果显示:吉富和奥尼肝细胞水肿变性, 而奥利亚和埃及尼罗出现大面积肝细胞脂肪变性; 每个品种罗非鱼均呈现严重的脾炎, 奥利亚、埃及尼罗和吉富的脾脏中大量铁血黄素沉积; 每种罗非鱼呈现不同程度的肾小球萎缩, 肾小管上皮细胞变性、坏死。研究表明奥尼罗非鱼抗链球菌病能力最强, 感染后血清中AST水平与肝受损程度呈一定的正相关, LZM水平和罗非鱼抗链球菌病能力呈一定的正相关。  相似文献

为了探索亚东鲑(Salmon trutta)多元化养殖模式,以初始体质量(100.05±1.12) g幼鱼作为研究对象,开展循环水和开放流水模式养殖比较,试验周期180d,分析比较其生长性能、血清生化指标和肌肉营养成分差异。结果表明:在开放流水模式下增重率、肥满度、肝体指数、特定生长率和饲料转化率均显著高于循环水模式(P<0.05),存活率二者差异不显著(P>0.05);在循环水模式下补体C4、总蛋白和生长激素均显著低于开放流水模式(P<0.05),但溶菌酶、谷丙转氨酶和谷草转氨酶相反(P<0.05),补体C3、碱性磷酸酶、酸性磷酸酶和球蛋白在两种模式之间差异均不显著(P>0.05);肌肉主要营养成分粗蛋白、粗脂肪含量开放流水模式高于循环水模式,氨基酸组成二者差异不显著(P>0.05),脂肪酸中油酸、α-亚麻酸、C20:1、C20:2和MUFA含量均显著低于开放流水(P<0.05),而EPA+DHA、SAFA和PUFA含量相反,二者之间差异性显著(P<0.05),矿物质镁含量循环水模式显著高于开...  相似文献

试验旨在研究低磷饲料中添加α-酮戊二酸(α-ketoglutarate, AKG)对松浦镜鲤(Cyprinus carpio Songpu)生长性能、体成分和血清生化指标的影响。选取平均体重为(6.67±0.14) g的松浦镜鲤630尾, 随机分成7组, 每组3个重复。分别投喂有效磷0.73%的正常磷饲料和有效磷0.47%的低磷饲料, 在低磷饲料中分别添加0、0.4%、0.8%、1.2%、1.6%、2.0% (干物质含量)的AKG, 配制成等氮等能饲料, 试验期为8周。结果表明: 低磷组饲料系数(FCR)显著高于正常磷组(P<0.05), 但增重率(WGR)、特定生长率(SGR)、蛋白质效率(PER)、脏体指数(VSI)及肥满度(CF)差异不显著(P>0.05); 与低磷组比, 0.4%AKG组FCR显著降低(P<0.05), 1.6%AKG组血清磷、白蛋白含量(ALB)显著增加(P<0.05); 0.8%AKG组谷草转氨酶(AST)活性和1.20%AKG组谷丙转氨酶(ALT)活性显著高于正常磷组(P<0.05); 添加不同水平AKG对血清其他指标无显著影响(P>0.05)。与正常磷组相比, 低磷组粗灰分含量、骨钙含量和钙磷沉积率显著降低(P<0.05); 与低磷组相比, 添加不同水平AKG磷沉积率显著增加(P<0.05), 0.4%AKG组, 骨钙含量显著增加(P<0.05), 0.8%AKG组, 骨磷含量显著增加(P<0.05); 低磷饲料添加AKG对其他体成分指标无显著影响(P>0.05)。由此得出, 低磷饲料中添加适量的AKG可以降低FCR, 提高松浦镜鲤脊椎骨钙磷含量。  相似文献

试验旨在研究低鱼粉高脂饲料中添加大豆卵磷脂对黄鳝(Monopterus albus)生长、血清生化指标及肠道菌群的影响,为降低黄鳝鱼粉使用量提供依据。以初始体重(20.03±0.01) g黄鳝为研究对象,以含有42%鱼粉、22%豆粕、6%粗脂肪的饲料为正常鱼粉组,另以含有22%鱼粉、52%豆粕和9%粗脂肪的饲料为低鱼粉高脂组,分别在低鱼粉高脂组中添加1%和2%的大豆卵磷脂,进行为期56d的养殖试验,于室外池塘进行网箱养殖,日投喂1次,喂量为黄鳝体重3%—5%。结果表明:(1)与对照组相比,低鱼粉高脂组黄鳝增重率显著降低(P<0.05),饵料系数与肝体比均显著增加(P<0.05),但添加2%大豆卵磷脂后黄鳝增重率显著提高(P<0.05),而饵料系数与肝体比显著降低(P<0.05);(2)对比正常鱼粉组,低鱼粉高脂饲料组黄鳝血清甘油三酯、总胆固醇、高密度脂蛋白、低密度脂蛋白、血氨、尿素氮、谷草转氨酶、谷丙转氨酶、酸性磷酸酶与免疫球蛋白M含量显著增加(P<0.05),且碱性磷酸酶活性显著降低(P<0.05),而添加2%大豆卵磷脂使黄鳝血清的高密度脂蛋白含量...  相似文献

为研究育肥饲料中混合植物油替代鱼油对中华绒螯蟹(Eriocheir sinensis)成体雄蟹常规成分和脂肪酸组成的影响,采用豆油和菜籽油混合物替代鱼油制成5种不同鱼油替代水平(0%、25%、50%、75%和100%)的等氮等脂育肥饲料(分别记为1#~5#饲料组)饲喂雄蟹,测定5组雄蟹肝胰腺、肌肉和性腺中的常规成分和脂肪酸组成,并对实验数据进行方差分析。结果显示:(1)饲料1#组性腺灰分含量显著高于饲料4#和5#组(P0.05),但各组性腺中的水分、总脂和蛋白含量均无显著差异(P0.05);饲料1#组肝胰腺中的水分和灰分最高,但其总脂含量低于其他组,各组肝胰腺的蛋白含量无显著差异(P0.05);除1#饲料组外,肌肉中的总脂和灰分含量随鱼油替代水平的升高而显著上升(P0.05),而水分和蛋白含量均无显著差异(P0.05)。(2)各饲料组精巢中总饱和脂肪酸(∑SFA)、总多不饱和脂肪酸(∑PUFA)和总高度不饱和脂肪酸(∑HUFA)含量无显著差异(P0.05),其总n-6多不饱和脂肪酸(∑n-6PUFA)含量随鱼油替代水平升高而升高,而总n-3多不饱和脂肪酸(∑n-3PUFA)含量和∑n-3PUFA/∑n-6PUFA比值呈显著下降趋势(P0.05)。(3)肝胰腺中∑n-3PUFA和∑HUFA含量具有显著的组间差异,且均以饲料3#组最高。但各组的∑PUFA和∑n-6PUFA含量差异并不显著(P0.05)。(4)肌肉中大部分脂肪酸组成无显著差异,仅∑n-6PUFA含量随鱼油替代水平升高而升高。综上,中华绒螯蟹育肥饲料中植物油(豆油与菜籽油含量为1︰1)替代鱼油对成体雄蟹可食组织中水分和蛋白含量并无显著影响,但会对其脂肪酸组成造成显著的影响,50%的鱼油替代水平有利于雄蟹肝胰腺和肌肉中的脂肪沉积。  相似文献

促性腺激素释放激素(GnRH)的主要作用是刺激脑垂体促性激素(GtH)的释放, 亦可促进鱼类生长激素(GH)的释放。促黄体素释放激素类似物(LHRH-A)是哺乳类GnRH的类似物, 为了分析LHRH-A对尼罗罗非鱼生长调节的作用, 设计了长期和短期2个实验, 采用腹腔注射(剂量0.1 μg/g体重)方法, 分析LHRH-A对尼罗罗非鱼绝对生长率、特定生长率、肝体系数和肥满度的影响, 并应用荧光实时定量PCR方法检测在注射LHRH-A后不同时段(6h、12h、24h、2周)尼罗罗非鱼垂体GH、肝脏GHR和肝脏IGF-I基因的表达变化。结果表明, LHRH-A组尼罗罗非鱼的绝对生长率、特定生长率、肝体系数、肥满度均显著高于对照组(P<0.05); 注射LHRH-A后12h、24h垂体GH mRNA表达水平均显著升高(P<0.05), 2周后恢复到对照组水平; 注射LHRH-A后24h和2周肝脏GHR mRNA表达水平显著上升(P<0.05); 注射LHRH-A后6h肝脏IGF-I mRNA表达水平显著升高(P<0.05), 12h、24h和2周恢复到对照组水平。以上结果提示, LHRH-A可显著上调尼罗罗非鱼生长轴相关基因的表达, 从而促进鱼类的生长。  相似文献

为了研究亚麻籽油(LO)和豆油(SO)完全替代鱼油(FO)对大黄鱼(Larimichthys crocea)肝脏和肌肉脂肪酸组成及Δ6Fad (Δ6 Fatty acid desaturase)基因表达的影响。实验用豆油和亚麻籽油替代鱼油制备了3种等氮等脂的精制饲料, 在海水浮动网箱中进行了为期10周的养殖实验。实验结果表明: (1)鱼油组的增重率、饲料效率和特定生长率均显著高于亚麻籽油组和豆油组(P<0.05), 但对成活率, 肝体指数和脏体指数没有显著影响(P>0.05); (2)亚麻籽油和豆油完全替代鱼油显著改变了鱼体肝脏和肌肉的脂肪酸组成, 降低了肝脏和肌肉LC-PUFA (Long chain-polyunsaturated fatty acid)的相对含量(P<0.05), 在亚麻籽油组(LO)和豆油组(SO)中没有显著差异(P>0.05), 在各处理组中, 肌肉的n-3LC-PUFA的相对含量显著高于肝脏(P<0.05); (3)亚麻籽油和豆油显著上调了肌肉和肝脏中Δ6Fad基因的表达量(P<0.05), 其表达量在肝脏中分别升高了7.6和6.5倍, 在肌肉中分别上升了2.2和2.8倍。结果表明, 在实验条件下亚麻籽油和豆油完全替代鱼油对大黄鱼生长具有不利影响, 亚麻籽油和豆油替代鱼油降低了肝脏和肌肉中LC-PUFA的含量, 提高了Δ6Fad基因的表达量。  相似文献

为提高紫黑翼蚌(Potamilus alatus)繁殖效率,探讨寄生对淡水石首鱼(Aplodinotus grunniens)营养指标的影响,实验通过比较钩介幼虫脱落数量,测试了不同规格寄主鱼的寄生效果,分析了寄生对寄主鱼血清生化指标及肌肉中氨基酸、脂肪酸含量的变化。结果显示:大规格组每尾鱼脱落数显著高于小规格组(P<0.05),小规格组每千克寄生鱼平均脱落稚贝数显著高于大规格组(P<0.05);寄生导致肌肉中肌酐显著升高(P<0.05),谷丙转氨酶、谷草转氨酶和乳酸脱氢酶显著降低(P<0.05),血糖、总蛋白、白蛋白、甘油三酯、总胆固醇、高密度脂蛋白和低密度脂蛋白与对照组无显著性差异(P>0.05);寄生组灰分与对照组无显著性差异(P>0.05),寄生组肌肉水分和粗脂肪含量显著高于对照组(P<0.05),粗蛋白显著低于对照组(P<0.05);寄生组天冬氨酸、丙氨酸、谷氨酸、酪氨酸、甘氨酸和精氨酸含量显著低于对照组(P<0.05),非必需氨基酸含量显著低于对照组(P<0.05),其他氨基酸之间无显著性差异(P>0.05)...  相似文献

3种鲳属鱼不同组织脂肪酸组成的比较研究   总被引:1,自引:0,他引:1  
3种野生鲳属鱼银鲳(Pampus argenteus)、灰鲳(Pampus cinereus)和中国鲳(Pampus sinensis)样本于2010年3—4月采集于浙江东海海域,用GC/MS法分别对其肝脏和肌肉的总脂和脂肪酸组成进行了分析和评价,同时比较了野生和养殖银鲳的脂肪酸组成。结果表明:3种鲳鱼肝脏总脂平均值(19.95%)显著高于肌肉总脂平均值(11.69%)(P<0.05),灰鲳肝脏和肌肉中的总脂含量显著高于银鲳和中国鲳(P中国鲳>灰鲳;肝脏的脂肪酸种类组成较丰富,MUFA含量中国鲳>银鲳>灰鲳(P<0.05),多不饱和脂肪酸(PUFA)含量灰鲳>银鲳>中国鲳(P<0.05);肌肉中MUFA含量银鲳>灰鲳>中国鲳(P<0.05),PUFA含量3种鲳鱼无显著差异(P>0.05)。EPA+DHA含量在3种鲳鱼肌肉中无显著差异(P>0.05),但在中国鲳肝脏未检测到EPA,在灰鲳肝脏和中国鲳肝脏和肌肉中未检测到DHA,而在银鲳肝脏和肌肉组织中EPA和DHA均有分布,DHA含量分别达到13.99%和15.95%;野生银鲳的油酸、EPA和DHA含量显著高于养殖银鲳,PUFA含量是养殖银鲳的1.53倍。由实验结果及与其它经济鱼类相比可知,3种鲳属鱼主要是以不饱和脂肪酸为主,在2种组织中平均占53.89%,EPA+DHA含量平均占PUFA的82.54%,这3种鲳鱼具有很高的营养价值并且银鲳更胜一筹,同时野生银鲳品质优于养殖银鲳,但不失为一种优良的养殖品种。  相似文献

以网箱(W)、工厂化循环水(G)和流水池塘(C)养殖4 kg左右的三倍体虹鳟为研究对象, 通过检测表观、肉质、气味及营养价值相关111个品质指标来评价不同养殖模式下三倍体虹鳟鱼肉品质的差异。结果表明, 在3种养殖模式下三倍体虹鳟鱼肉品质存在明显差异。其中, 网箱养殖三倍体虹鳟鱼肉色调角、硬度、内聚性、弹性、咀嚼性、汁液流失率和失水率均显著高于其他养殖模式(P<0.05), 而挥发性气味物质中1-庚醇、庚醛、辛醛、壬醛和气味活度总值显著低于其他养殖模式(P<0.05); 工厂化循环水养殖三倍体虹鳟肥满度和鱼肉的红色值、色度值、盐溶性蛋白含量、总脂肪酸含量和n-3脂肪酸含量显著高于其他养殖模式(P<0.05); 流水池塘养殖三倍体虹鳟肥满度、鱼肉的黄色值、色度值、水分、脂肪含量、n-3脂肪酸含量和多不饱和脂肪酸含量最低(P<0.05), 出肉率、黏附性、肌糖原含量及挥发性风味物质中1-辛烯-3-醇、2,3-辛二酮、戊醛、庚醛、辛醛、(E)-2-癸烯醛和气味活度总值最高(P<0.05)。总之, 三种养殖模式养殖的三倍体虹鳟品质各具特色: 网箱养殖三倍体虹鳟鱼肉质地坚实富有弹性, 挥发性风味物质含量较低; 工厂化循环水养殖三倍体虹鳟具有较红的肉色并富含不饱和脂肪酸; 流水池塘养殖三倍体虹鳟形体优美且出肉率相对较高, 挥发性风味物质含量丰富。  相似文献

文章旨在探索开放流水养殖模式下,暂养对黄颡鱼(Pelteobagrus fulvidraco)肌肉品质和营养价值的影响。试验选用处于快速生长期的大规格鱼种(15.69±2.28) g黄颡鱼为试验对象,随机分为4组,分别暂养0(对照组)、20d、30d和40d,每组3个重复。试验期间投喂黄颡鱼配合饲料,在养殖过程中定期测量水质。在试验结束后对其生长性能、血清生化指标、肌肉氨基酸、脂肪酸组成及质构特性进行比较。结果显示:(1)暂养期间试验池的水质氨态氮含量在0.03—0.05 mg/L,亚硝态氮含量均在0.01 mg/L,溶氧水平均超过9.0 mg/L。(2)暂养时间延长,黄颡鱼末体重和增重率呈上升趋势,特定生长率呈下降趋势,各组间差异显著(P<0.05);暂养20d、30d和40d组的成活率、肥满度、肝体比及脏体比组间无显著性差异(P>0.05)。(3)暂养30d和40d组血清中的白蛋白和总蛋白含量显著高于对照组和暂养20d组(P<0.05),但其γ-谷氨酰基转移酶和碱性磷酸酶活性、总胆固醇和总胆汁酸含量显著低于对照组和暂养20d组(P<0.05);对照组、暂养2...  相似文献

为探究水体中Cu2+对罗非鱼(GIFT tilapia, Oreochromis niloticus)生长性能、肝胰脏脂代谢和脾脏免疫功能的影响, 实验设置水体Cu2+浓度分别为0(对照组)、0.2、0.4和0.8 mg/L, 每个浓度处理组设置3个实验重复, 选择初始体重为(0.45±0.02) g的健康罗非鱼幼体, 随机分配至12个养殖缸中, 进行4周的养殖实验。结果显示: (1) 在水体Cu2+胁迫下, 罗非鱼的存活率、增重率和特定生长率均显著下降; 0.4和0.8 mg/L Cu2+浓度组罗非鱼的肝体比显著高于对照组和0.2 mg/L处理组。(2) 0.4和0.8 mg/L处理组罗非鱼肝胰脏中甘油三酯含量显著高于对照组, 但与0.2 mg/L处理组之间无显著差异; 肝胰脏中总胆固醇含量与3-羟基-3-甲基戊二酸单酰辅酶A还原酶活力在各组间无显著差异。(3) 在水体Cu2+胁迫下, 罗非鱼血清低密度脂蛋白胆固醇、总胆固醇含量和谷草转氨酶活力均显著上升; 血清甘油三酯含量呈先下降后上升的趋势, 且0.8 mg/L处理组含量显著高于0.2 mg/L处理组; 0.8 mg/L Cu2+处理组罗非鱼血清谷丙转氨酶活力显著高于0.2和0.4 mg/L处理组, 但与对照组无显著差异。(4) 组织学结果表明, 水体过量Cu2+使罗非鱼肝胰脏组织空泡化现象严重, 脂滴含量显著高于对照组; 脾脏组织出现较大面积的巨噬细胞中心, 且脂褐素含量显著增加。研究表明, 水体Cu2+暴露显著降低了罗非鱼生长性能, 并导致肝胰脏和血清脂肪沉积, 造成肝胰脏、脾脏损伤。研究为明确重金属环境下养殖鱼类腹腔脂肪蓄积及脾脏损伤机制提供了数据参考。  相似文献

The combined effect of dietary carbohydrate type and n-3 PUFA (EPA+DHA) on pre- and post-challenge haemato-immunological responses in Labeo rohita juveniles was studied. Fish were fed for 67days with six different test diets containing either gelatinised (G) or non-gelatinised (NG) corn (43%) with three levels of n-3 PUFA (0.5%, 1.0% and 2.0%). During the pre-challenge period, significantly higher (P<0.05) NBT, serum lysozyme activity, total protein and globulin content was recorded in the NG carbohydrate fed groups. Highest NBT value was recorded in the groups fed with 1.0% n-3 PUFA, whereas the highest serum lysozyme activity (P<0.05) was recorded at either 0.5% or 2.0% n-3 PUFA fed groups in both the pre- and post-challenge period. Feeding of NG corn significantly increased the total leucocyte count, lysozyme activity, A/G ratio and decreased the total erythrocyte count, haemoglobin, serum total protein and globulin content of L. rohita juveniles during the post-challenge period. Similarly, feeding of n-3 PUFA at any level significantly increased the immunological parameters like lysozyme activity or A/G ratio, whereas total leukocyte count increased due to feeding of either 0.5% or 1.0% n-3 PUFA. The NBT and albumin values remained similar in both the pre- and post-challenge period. After challenge with Aeromonas hydrophila, the highest survival was recorded in the NG carbohydrate fed groups, whereas the lowest survival was recorded in the highest level of n-3 PUFA fed group irrespective of dietary carbohydrate type. Thus, a high level of G carbohydrate as well as n-3 PUFA is found to be immunosuppressive in L. rohita juveniles. NG carbohydrate supplemented with 1.0% n-3 PUFA is found to be optimum to enhance the immunity in L. rohita juveniles.  相似文献

Juvenile tench (initial weight of about 57 g) were fed feed supplemented with fish oil (group FO), linseed oil (group LO), peanut oil (group PO), or rapeseed oil (group RO) containing 47% protein and 12% fat for 55 days. The inclusion of the tested oils was 50 g kg−1 (42% total crude lipids in diets). No significant differences were noted in the fish growth performance. The proximate composition of the whole fish bodies and the viscera (water, protein, fat, ash) was similar in all the dietary treatments (P > 0.05). Differences were noted only with regard to the ash content of the fillets (P < 0.05). The analysis of the fatty acids profiles of tench (whole fish) indicated there were significant differences in the total content of monoenoic and polyenoic (PUFA) acids. Significant differences were also noted with regard to n-3 PUFA and n-6 PUFA. Consequently, the ratio of n-3/n-6 acids ranged from 1.6 (group PO) to 2.08 (group LO; P < 0.05). The feed applied was not confirmed to have had an impact on the fatty acids profile of the tench fillets. There was a statistically significant intergroup difference in the content of saturated fatty acids (SFA) in tench viscera. In the fish fed vegetable oils supplemented diets, the level of SFA was lower (P < 0.05).  相似文献

High saturated fatty acid (SFA) diets contribute to the development of insulin resistance, whereas fish oil-derived n-3 polyunsaturated fatty acids (PUFA) increase the secretion of adiponectin (Ad), an adipocyte-derived protein that stimulates fatty acid oxidation (FAO) and improves skeletal muscle insulin response. We sought to determine whether fish oil could prevent and/or restore high SFA diet-induced impairments in Ad and insulin response in soleus muscle. Sprague-Dawley rats were fed 1) a low-fat control diet (CON group), 2) high-SFA diet (SFA group), or 3) high SFA with n-3 PUFA diet (SFA/n-3 PUFA group). At 4 wk, CON and SFA/n-3 PUFA animals were terminated, and SFA animals were either terminated or fed SFA or SFA/n-3 PUFA for an additional 2 or 4 wk. The effect of diet on Ad-stimulated FAO, insulin-stimulated glucose transport, and expression of Ad, insulin and inflammatory signaling proteins was determined in the soleus muscle. Ad stimulated FAO in CON and 4 wk SFA/n-3 PUFA (+36%, +39%, respectively P ≤ 0.05) only. Insulin increased glucose transport in CON, 4 wk SFA/n-3 PUFA, and 4 wk SFA + 4 wk SFA/n-3 PUFA (+82%, +33%, +25%, respectively P ≤ 0.05); this effect was lost in all other groups. TLR4 expression was increased with 4 wk of SFA feeding (+24%, P ≤ 0.05), and this was prevented in 4 wk SFA/n-3 PUFA. Suppressor of cytokine signaling-3 expression was increased in SFA and SFA/n-3 PUFA (+33 and +18%, respectively, P ≤ 0.05). Our results demonstrate that fish oil can prevent high SFA diet-induced impairments in both Ad and insulin response in soleus muscle.  相似文献

Catches of the economically important Nile tilapia (Oreochromis niloticus L.) from two eutrophic tropical lakes in Uganda, Lake Mburo and Murchison Bay, Lake Victoria, were examined to determine the presence of microcystins (MCs) in gut, liver and muscle of the fish. Analysis for MCs (RR, LR and YR) in both fish and water samples was by liquid chromatography coupled with mass spectroscopy (LC-MS) method. Physico-chemical parameters were also measured to establish the status of both lakes. MC-RR was the most prominent MC detected in Lake Mburo and Murchison Bay samples, there was no evidence of significant seasonal variation in the concentration of MCs in fish tissue. MCs were detected in all water samples from both study lakes. The mean concentration of MCs in water was found higher in dry times for Lake Mburo (P < 0.05) and higher in wet times for Murchison Bay (P < 0.05). MC concentrations in the fish guts were positively related with MC concentrations in water samples from Murchison bay (P < 0.05), no such correlation was found in Lake Mburo. In eutrophic tropical lakes, fish seem to have a high tolerance to the toxicity of cyanotoxins including MCs. However, there is a possibility of accumulating these toxins in their tissue with the threat of transferring them higher up in the food chain. Due to a low sample size and short sampling period, the results can only serve to highlight the potential risk of MC accumulation in Nile tilapia in these lakes. Further studies are needed for the purpose of risk assessment.  相似文献

Polyunsaturated fatty acids of n-3 series (n-3 PUFA) were shown to increase basal fat oxidation in humans. The aim of the study was to compare the effect of n-3 PUFA added to a very low calorie diet (VLCD), with VLCD only during three-week inpatient weight reduction. Twenty severely obese women were randomly assigned to VLCD with n-3 PUFA or with placebo. Fatty acids in serum lipid fractions were quantified by gas chromatography. Differences between the groups were determined using ANOVA. Higher weight (7.55+/-1.77 vs. 6.07+/-2.16 kg, NS), BMI (2.82+/-0.62 vs. 2.22+/-0.74, p<0.05) and hip circumference losses (4.8+/-1.81 vs. 2.5+/-2.51 cm, p<0.05) were found in the n-3 group as compared to the control group. Significantly higher increase in beta-hydroxybutyrate was found in the n-3 group showing higher ketogenesis and possible higher fatty acid oxidation. The increase in beta-hydroxybutyrate significantly correlated with the increase in serum phospholipid arachidonic acid (20:4n-6; r = 0.91, p<0.001). In the n-3 group significantly higher increase was found in n-3 PUFA (eicosapentaenoic acid, 20:5n-3, docosahexaenoic acid, 22:6n-3) in triglycerides and phospholipids. The significant decrease of palmitoleic acid (16:1n-7) and vaccenic acid (18:1n-7) in triglycerides probably reflected lower lipogenesis. A significant negative correlation between BMI change and phospholipid docosahexaenoic acid change was found (r = -0.595, p<0.008). The results suggest that long chain n-3 PUFA enhance weight loss in obese females treated by VLCD. Docosahexaenoate (22:6n-3) seems to be the active component.  相似文献

N-3 long-chain polyunsaturated fatty acids (n-3 LCPUFA) are subject of growing interest as they are of particular relevance for meat quality and human health. However, their content in the muscles of cattle is generally low probably as the complex result of their biosynthesis from dietary n-3 PUFA in the muscle and/or in other tissues/organs and of their subsequent uptake by the muscle. In view of this, this study aimed at understanding whether the changes in the muscle n-3 LCPUFA content, depending on the diet (maize silage v. grass) or the muscle type (Rectus abdominis, RA v. Semitendinosus, ST) in 12 Charolais steers, were related to variations in the gene expression of proteins involved in n-3 LCPUFA biosynthesis or cellular uptake. Tissue fatty acid composition was analysed by gas-liquid chromatography and mRNA abundance of proteins by quantitative real-time PCR. The grass-based diet resulted in a 2.3-fold (P < 0.0002) increase in both RA and ST n-3 LCPUFA content compared with the maize silage-based diet, whereas no difference in the expression of genes involved in n-3 LCPUFA biosynthesis and uptake was observed between diets. ST exhibited a 1.5-fold higher n-3 LCPUFA content than RA (P < 0.003), whereas the gene expression of proteins involved in n-3 LCPUFA biosynthesis and uptake was 1.3- to 18-fold higher in RA than in ST (P < 0.05). In conclusion, diet- or muscle type-dependent changes in the muscle n-3 LCPUFA content of Charolais steers did not seem to be mediated by the gene expression regulation of proteins involved in the biosynthesis or uptake of these fatty acids.  相似文献
