Corynebacterium acetoacidophilum RYU3161 was cultivated in al-histidine-limited fed-batch culture. To investigate the effect of cell growth on thel-proline production, 5l fed-batch culture was performed using an exponential feeding rate to obtain the specific growth rates (μ) of 0.04, 0.06, 0.08, and 0.1 h−1. The results show that the highest production ofl-proline was obtained at μ=0.04 h−1. The specificl-proline production rate (Qp) increased proportionally as a function of the specific growth rate, but decreased after it revealed the maximum value at μ=0.08 h−1. Thus, the highest productivity ofl-proline was 1.66 g L−1 h−1 at μ=0.08 h−1. The results show that the production of L-proline inC. acetoacidophilum RYU3161 has mixed growth-associated characteristics.  相似文献

A thermostable lipase was produced in continuous cultivation of a newly isolated thermophilic Bacillus sp. strain IHI-91 growing optimally at 65 °C. Lipase activity decreased with increasing dilution rate while lipase productivity showed a maximum of 340 U l−1 h−1 at a dilution rate of 0.4 h−1. Lipase productivity was increased by 50% compared to data from batch fermentations. Up to 70% of the total lipase activity measured was associated to cells and by-products or residual substrate. Kinetic and stoichiometric parameters for the utilisation of olive oil were determined. The maximal biomass output method led to a saturation constant K S of 0.88 g/l. Both batch growth data and a washout experiment yielded a maximal specific growth rate, μmax, of 1.0 h−1. Oxygen uptake rates of up to 2.9 g l−1h−1 were calculated and the yield coefficient, Y X/O, was determined to be 0.29 g dry cell weight/g O2. From an overall material balance the yield coefficient, Y X/S, was estimated to be 0.60 g dry cell weight/g olive oil. Pseudomonas putida KT2440 grew on glucose at a specific rate of 0.48 h−1 but accumulated almost no poly-3-hydroxyalkanoates (PHA). Subsequent nitrogen limitation on nonanoic acid resulted in the accumulation of only 27% medium-chain-length PHA (MCL-PHA). In contrast, exponential nonanoic acid-limited growth (μ = 0.15 h−1) produced 70 g l−1 biomass containing 75% PHA. At a higher exponential feed rate (μ = 0.25 h−1), the overall productivity was increased but less biomass (56 g l−1) was produced due to higher oxygen demand, and the biomass contained less PHA (67%). It was concluded that carbon-limited exponential feeding of nonanoic acid or related substrates to cultures of P. putida KT2440 is a simple and highly effective method of producing MCL-PHA. Nitrogen limitation is unnecessary.  相似文献

Heterotrophic bacterial and phytoplankton biomass, production, specific growth rates and growth efficiencies were studied in July 2001 and January 2002 during both spring and neap tides, along a tidal cycle, at three sites in a subtropical estuary. Major freshwater inputs located in the Northern region led to differences in both phytoplankton and bacterioplankton biomass and activity along the estuary. While in the Northern region phytoplankton is light-limited, with mean phytoplankton production (PP) between 1.1 and 1.9 μg C l−1 h−1 and mean specific growth rates (PSG) between 0.14 and 0.16 d−1, the Southern region registered values as high as 24.7 μg C l−1 h−1 for PP and 2.45 d−1 (mean PP between 3.4 and 7.3 μg C l−1 h−1; mean PSG between 0.28 and 0.57 d−1). On the other hand, maximum bacterial production (BP: 63.8 μg C l−1 h−1) and specific growth rate (BSG: 32.26 d−1) were observed in the Northern region (mean BP between 3.4 and 12.8 μg C l−1 h−1; mean BSG between 1.98 and 6.67 day−1). These bacterial activity rates are among the highest recorded rates in estuarine and coastal waters, indicating that this system can be highly heterotrophic, due to high loads of allochthonous carbon (mainly derived from mangrove forest). Our results also showed that, despite that BP rates usually exceeded PP, in the Southern region BP may be partially supported (∼45%) by PP, since a significant regression was observed between BP and PP (r = 0.455, P < 0.001). The aim of this research was to test whether NH4 + and NO3 affect the growth, P demand, cell composition and N2 fixation of Cylindrospermopsis raciborskii under P limitation. Experiments were carried out in P-limited (200 μg l−1 PO4-P) chemostat cultures of C. raciborskii using an inflowing medium containing either 4,000 μg l−1 NH4-N, 4,000 μg l−1 NO3-N or no combined N. The results showed the cellular N:P and C:P ratios of C. raciborskii decreased towards the Redfield ratio with increasing dilution rate (D) due to the alleviation of P limitation. The cellular C:N and carotenoids:chlorophyll-a ratios also decreased with D, predominantly as a result of an increase in the chlorophyll-a and N content. The NH4 + and NO3 supply reduced the P maintenance cell quota of C. raciborskii. Consequently, the biomass yield of the N2-grown culture was significantly lower. The maximum specific growth rate of N2-grown culture was also the lowest observed. It is suggested that these differences in growth parameters were caused by the P and energy requirement for heterocyte formation, nitrogenase synthesis and N2 fixation. N2 fixation was partially inhibited by NO3 and completely inhibited by NH4 +. It was probably repressed through the high N content of cells at high dissolved N concentrations. These results indicate that C. raciborskii is able to grow faster and maintain a higher biomass under P limitation where a sufficient supply of NH4 + or NO3 is maintained. Information gained about the species-specific nutrient and pigment stoichiometry of C. raciborskii could help to access the degree of nutrient limitation in water bodies. Candida cylindracea NRRL Y-17506 was grown to produce extracellular lipase from oleic acid as a carbon source. Through flask cultures, it was found that the optimum initial oleic acid concentration for cell growth was 20 g l−1. However, high initial concentrations of oleic acid up to 50 g l−1 were not inhibitory. The highest extracellular lipase activity obtained in flask culture was 3.0 U ml−1 after 48 h with 5 g l−1 of initial oleic acid concentration. Fed-batch cultures (intermittent and stepwise feeding) were carried out to improve cell concentration and lipase activity. For the intermittent feeding fed-batch culture, the final cell concentration was 52 g l−1 and the extracellular lipase activity was 6.3 U ml−1 at 138.5 h. Stepwise feeding fed-batch cultures were carried out to simulate an exponential feeding and to investigate the effects of specific growth rate (0.02, 0.04 and 0.08 h−1) on cell growth and lipase production. The highest final cell concentration obtained was 90 g l−1 when the set point of specific growth rate (μset) was 0.02 h−1. High specific growth rate (0.04 and 0.08 h−1) decreased extracellular lipase production in the later part of fed-batch cultures due to build-up of the oleic acid oversupplied. The highest extracellular lipase activity was 23.7 U ml−1 when μset was 0.02 h−1, while the highest lipase productivity was 0.31 U ml−1 h−1 at μset of 0.08 h−1.  相似文献

The biokinetics of glucose metabolism were evaluated in Aeromonas hydrophila during growth in an anaerobic biosystem. After approx 34 h growth, A. hydrophila metabolized 5,000 mg glucose l−1 into the end-products ethanol, acetate, succinate and formate. The maximum growth rate, μ m, half saturation coefficients, K s, microbial yield coefficient, Y, cell mass decay rate coefficient, k d, and substrate inhibition coefficient, K si were 0.25 ± 0.03 h−1, 118 ± 31 mg glucose l−1, 0.12 μg DNA mg glucose−1, 0.01 h−1, and 3,108 ± 1,152 mg glucose l−1, respectively. These data were used to predict the performance of a continuous growth system with an influent glucose concentration of 5,000 mg l−1. Results of the analysis suggest that A. hydrophila will metabolize glucose at greater than 95% efficiency when hydraulic retention times (HRTs) exceed 7 h, whereas the culture is at risk of washing out at an HRT of 6.7 h.  相似文献

The growth performance of malolactic fermenting bacteria Oenococcus oeni NCIMB 11648 and Lactobacillus brevis X2 was assessed in continuous culture. O. oeni grew at a dilution rate range of 0.007 to 0.052 h−1 in a mixture of 5:6 (g l−1) of glucose/fructose at an optimal pH of 4.5, and L. brevis X2 grew at 0.010 to 0.089 h−1 in 10 g l−1 glucose at an optimal pH of 5.5 in a simple and safe medium. The cell dry weight, substrate uptake and product formation were monitored, as well as growth kinetics, yield parameters and fermentation balances were also evaluated under pH control conditions. A comparison of growth characteristics of two strains was made, and this showed significantly different performance. O. oeni has lower maximum specific growth rate (μmax=0.073 h−1), lower maximum cell productivity (Q x max=17.6 mg cell l−1 h−1), lower maximum biomass yield (Y x/s max=7.93 g cell mol−1 sugar) and higher maintenance coefficient (m s=0.45 mmol−1 sugar g−1 cell h−1) as compared with L. brevis X2max=0.110 h−1; Q x max=93.2 g−1 cell mol−1 glucose; Y x/s max=22.3 g cell mol−1 glucose; m s=0.21 mmol−1 glucose g−1 cell h−1). These data suggest a possible more productive strategy for their combined use in maturation of cider and wine.  相似文献

Propionibacterium shermanii CDB 10014 is able to grow even at high oxygen transfer rates (24.0 mmol O2 l−1 h−1), in contrast to reports in the specialised literature, where all Propionibacteria are considered oxygen-sensitive microorganisms. Propionic acid is the main product in anaerobiosis. The presence of oxygen in the system leads to an inhibition of propionic acid production while acetic acid formation is enhanced. At high oxygen supply rates no propionic acid is produced and acetic acid is the main product. Lactic acid is also produced in reasonable quantities (2.7 g l−1). The growth rate (μmax) is higher in anaerobiosis (0.19 h−1) than in aerobiosis (0.12–0.15 h−1). The cell yield is higher in aerobiosis (0.18–0.22 g g−1) than in anaerobiosis (0.14 g g−1) suggesting the oxidative metabolism of glucose by Propionibacterium shermanii CDB 10014. The production of recombinant glycoproteins in Dictyostelium discoideum by conventional cell culture methods was limited by low cell density as well as low growth rate. In this work, cotton towel with a good adsorption capability for D. discoideum cells was used as the immobilization matrix in an external fibrous bed bioreactor (FBB) system. With batch cultures in the FBB, the concentration of immobilized cells in the cotton fiber carrier increased to 1.37 × 108 cells per milliliter after 110-h cultivation, which was about tenfold higher than the maximal cell density in the conventional free-cell culture. Correspondingly, a high concentration of soluble human Fas ligand (hFasL; 173.7 μg l−1) was achieved with a high productivity (23 μg l−1 h−1). The FBB system also maintained a high density of viable cells for hFasL production during repeated-batch cultures, achieving a productivity of 9∼10 μg l−1 h−1 in all three batches studied during 15 days. The repeated-batch culture using immobilized cells of D. discoideum in the FBB system thus provides a good method for long-term and high-level production of hFasL.  相似文献

In this study we manipulated both nitrogen and phosphorus concentrations in stream mesocosms to develop quantitative relationships between periphytic algal growth rates and peak biomass with inorganic N and P concentrations. Stream water from Harts Run, a 2nd order stream in a pristine catchment, was constantly added to 36 stream-side stream mesocosms in low volumes and then recirculated to reduce nutrient concentrations. Clay tiles were colonized with periphyton in the mesocosms. Nutrients were added to create P and N concentrations ranging from less than Harts Run concentrations to 128 μg SRP l−1 and 1024 μg NO3-N l−1. Algae and water were sampled every 3 days during colonization until periphyton communities reached peak biomass and then sloughed. Nutrient depletion was substantial in the mesocosms. Algae accumulated in all streams, even streams in which no nutrients were added. Nutrient limitation of algal growth and peak biomass accrual was observed in both low P and low N conditions. The Monod model best explained relationships between P and N concentrations and algal growth and peak biomass. Algal growth was 90% of maximum rates or higher in nutrient concentrations 16 μg SRP l−1 and 86 μg DIN l−1. These saturating concentrations for growth rates were 3–5 times lower than concentrations needed to produce maximum biomass. Modified Monod models using both DIN and SRP were developed to explain algal growth rates and peak biomass, which respectively explained 44 and 70% of the variance in algal response.  相似文献

Kinetic parameters describing growth and decay of mesophilic (30°C) and thermophilic (55°C) aerobic biomass were determined in continuous and batch experiments by using oxygen uptake rate measurements. Biomass was cultivated on a single soluble substrate (acetate) in a mineral medium. The intrinsic maximum growth rate (μ max) at 55°C was 0.71±0.09 h−1, which is 1.5 times higher than the μ max at 30°C (0.48±0.11 h−1). The biomass decay rates increased from 0.004 h−1 at 30°C to 0.017 h−1 at 55°C. Monod constants were very low for both types of biomass: 9±2 mg chemical oxygen demand (COD) l−1at 30°C and 3±2 mg COD l−1at 55°C. Theoretical biomass yields were similar at 30 and 55°C: 0.5 g biomass COD (g acetate COD)−1. The observed biomass yields decreased under both temperature conditions as a function of the cell residence time. Under thermophilic conditions, this effect was more pronounced due to the higher decay rates, resulting in lower biomass production at 55°C compared to 30°C. Electronic Publication  相似文献

Four automatic substrate feeding strategies were developed and investigated in this study to obtain rapid, repeatable, and reliable high cell densities of Pseudomonas putida KT2440 from glucose. Growth yield data of the key nutrients, Y X/Glucose, Y X/NH4, Y X/PO4, Y X/Mg, and Y CO2/Glucose, were determined to be 0.41, 5.44, 13.70, 236, and 0.65 g g−1, respectively. Although standard exponential feeding strategy worked well when the predetermined μ was set at 0.25 h−1, an exponential glucose feeding strategy with online μ max estimation resulted in a higher average biomass productivity (3.4 vs 2.8 g l−1 h−1). A CO2 production rate based pulse glucose feeding strategy also resulted in good overall productivity (3.0 g l−1 h−1) and can be used as an alternative to pH-stat or DO-stat feeding. A cumulative CO2 production based continuous feed with real-time cumulative glucose consumption estimation resulted in much higher biomass productivity (4.3 g l−1 h−1) and appears to be an excellent and reliable approach to fully automating high-cell-density fed-batch cultivation of P. putida.  相似文献

The vertical distribution of a bloom-forming Microcystis population was studied based on the relevant limnological parameters obtained from the lower Nakdong River (Mulgum) during the summer of 1994. Over three months (late June to late September), a high abundance of Microcystis population (mean ± SD, 2.9 ± 8.4 × 105 cells ml−1, n = 40) and algal biomass (mean ± SD, chlorophyll a, 131 ± 346 μg l−1, n = 31) was persistent throughout the entire water column (0–5 m, n = 11). The vertical distribution of carbon content was uneven, with a high concentration near the surface zone (mean ± SD, total, 7.9 ± 7.8; Microcystis, 5.2 ± 8.3 μg C ml−1, n = 15). Incorporating limnological and meteorological factors, a diel study of the vertical distribution of Microcystis showed that the chlorophyll a concentration was highest near the surface zone on a calm night (wind velocity, <2 m s−1, 2300–700) but was evenly distributed on a windy day (>4 m s−1, 1100–1900). Among many possible factors, wind velocity may have played an important role in controlling the vertical distribution of Microcystis in the lower Nakdong River. Castor bean (Ricinus communis L.) plants were grown for 5–7 weeks in a controlled environment at 350 μl l−1 or 700 μl l−1 CO2. Carbon assimilation, assimilate deposition, dark respiration and assimilate mobilization were measured in leaves 2, 3 and 4 (counted from the base of the plant), and a balance sheet of carbon input and export was elaborated for both CO2 concentrations. Carbon dioxide assimilation was nearly constant over the illumination period, with only a slight depression occurring at the end of the day in mature source leaves, not in young source leaves. Assimilation was ca. 40% higher at 700 μl l−1 than at 350 μl l−1 CO2. The source leaves increased steadily in weight per unit area during the first 3 weeks, more at 700 μl l−1 than at 350 μl l−1 CO2. On top of an irreversible weight increase, there was a large gain in dry weight during the day, which was reversed during the night. This reversible weight gain was constant over the life time of the leaf and ca. 80% higher at 700 μl l−1 than at 350 μl l−1. Most of it was due to carbohydrates. The carbon content (as a percentage) was not altered by the CO2 treatment. Respiration was 25% higher in high-CO2 plants when based on leaf area, but the same when based on dry weight. The rate of carbon export via the phloem was the same during the daytime in plants grown at 350 μl l−1 and 700 μl l−1 CO2. During the night the low-CO2 plants had only 50% of the daytime export rate, in contrast to the high-CO2 plants which maintained the high export rate. It was concluded that the phloem loading system is saturated during the daytime in both CO2 regimes, whereas during the night the assimilate supply is reduced in plants in the normal CO2 concentration. Two-thirds of the carbon exported from the leaves was permanently incorporated as plant dry matter in the residual plant parts. This “assimilation efficiency” was the same for both CO2 regimes. It is speculated that under 350 μl l−1 CO2 the growing Ricinus plant operates at sink limitation during the day and at source limitation during the night. This paper focuses on modelling the growth rate and exopolysaccharides production of Anabaena sp. ATCC 33047, to be used in carbon dioxide removal and biofuels production. For this, the influence of dilution rate, irradiance and aeration rate on the biomass and exopolysaccharides productivity, as well as on the CO2 fixation rate, have been studied. The productivity of the cultures was maximum at the highest irradiance and dilution rate assayed, resulting to 0.5 gbio l−1 day−1 and 0.2 geps l−1 day−1, and the CO2 fixation rate measured was 1.0 gCO2 l−1 day−1. The results showed that although Anabaena sp. was partially photo-inhibited at irradiances higher than 1,300 μE m−2 s−1, its growth rate increases hyperbolically with the average irradiance inside the culture, and so does the specific exopolysaccharides production rate. The latter, on the other hand, decreases under high external irradiances, indicating that the exopolysaccharides metabolism hindered by photo-damage. Mathematical models that consider these phenomena have been proposed. Regarding aeration, the yield of the cultures decreased at rates over 0.5 v/v/min or when shear rates were higher than 60 s−1, demonstrating the existence of thus existence of stress damage by aeration. The behaviour of the cultures has been verified outdoors in a pilot-scale airlift tubular photobioreactor. From this study it is concluded that Anabaena sp. is highly recommended to transform CO2 into valuable products as has been proved capable of metabolizing carbon dioxide at rates of 1.2 gCO2 l−1 day−1 outdoors. The adequacy of the proposed equations is demonstrated, resulting to a useful tool in the design and operation of photobioreactors using this strain.  相似文献

The influence of (NH4)2SO4 concentration and dilution rate (D) on actual and potential H2 photoproduction has been studied in ammonium-limited chemostat cultures of Rhodobacter capsulatus B10. The actual H2 production in a photobioreactor was maximal (approx. 80 ml h−1 l−1) at D = 0.06 h−1 and 4 mM (NH4)2SO4. However, it was lower than the potential H2 evolution (calculated from hydrogen evolution rates in incubation vials), which amounted to 100–120 ml h−1 l−1 at D = 0.03–0.08 h−1. Taking into account the fact that H2 production in the photobioreactor under these conditions was not limited by light or lactate, another limiting (inhibiting) factor should be sought. One possibility is an inhibition of H2 production by the H2 accumulated in the gas phase. This is apparent from the non-linear kinetics of H2 evolution in the vials or from its inhibition by the addition of H2; initial rates were restored in both cases after the vials had been refilled with argon. The actual H2 production in the photobioreactor at D = 0.06 h−1 was shown to increase from approximately 80 ml h−1 l−1 to approximately 100 ml h−1 l−1 under an argon flow at 100 ml min−1. Under maximal H2 production rates in the photobioreactor, up to 30% of the lactate feedstock was utilised for H2 production and 50% for biomass synthesis. The uptake of soluble phosphate by the green sulfur bacterium Chlorobium limicola UdG6040 was studied in batch culture and in continuous cultures operating at dilution rates of 0.042 or 0.064 h–1. At higher dilution rates, washout occurred at phosphate concentrations below 7.1 μM. This concentration was reduced to 5.1 μM when lower dilution rates were used. The saturation constant for growth on phosphate (K μ) was between 2.8 and 3.7 μM. The specific rates of phosphate uptake in continuous culture were fitted to a hyperbolic saturation model and yielded a maximum rate (Va max) of 66 nmol P (mg protein)–1 h–1 and a saturation constant for transport (K t) of 1.6 μM. In batch cultures specific rates of phosphate uptake up to 144 nmol P (mg protein)–1 h–1 were measured. This indicates a difference between the potential transport of cells and the utilization of soluble phosphate for growth, which results in a significant change in the specific phosphorus content. The phosphorus accumulated within the cells ranged from 0.4 to 1.1 μmol P (mg protein)–1 depending on the growth conditions and the availability of external phosphate. Transport rates of phosphate increased in response to sudden increases in soluble phosphate, even in exponentially growing cultures. We investigated the impact of different nitrogen (N) and phosphorus (P) compounds and concentrations on the growth of Gyrodinium instriatum Freudenthal et Lee in laboratory experiments, and possible links to blooms of this species at Hakozaki Fishing Port, Fukuoka, Japan. G. instriatum utilized only inorganic N compounds as N sources for growth. In contrast, G. instriatum utilized many inorganic and organic phosphorus compounds. We used the Monod equation to describe the growth rate of G. instriatum in N- or P-limited batch cultures as a function of ambient nutrient concentrations. Kinetic growth parameters for maximum specific growth rate (μmax) and half-saturation nutrient concentration (K S) were 0.57 divisions d−1 and 14.2 μmol l−1, respectively, under N-limitation and 0.65 divisions d−1 and 1.75 μmol l−1, respectively, under P-limitation. Compared with these K S values, all in situ average dissolved inorganic nitrogen (DIN) concentrations in Hakozaki Fishing Port were higher than K S for N, but all in situ average dissolved inorganic phosphorus (DIP) concentrations were lower than K S for P, whether a red tide occurred or not bloom. Moreover, average DIP concentration in April (a month critical to red-tide genesis) of 2004 (a non-red-tide year) was less than half those in 2002 and 2003 (red-tide years). Thus, differences in DIP concentrations may be an important factor controlling blooms of G. instriatum in Hakozaki Fishing Port.  相似文献

Abstract The investigation of the bacterial community in the Kühw?rter Wasser, a macrophyte-dominated arm of the River Danube backwater system near Vienna, revealed that variation in microbial densities and biomass could be related to a characteristic sequence in morphotype composition over the seasons. Maximal bacterial cell numbers and biomass occured in early summer, with values of up to 9 × 109 cells l−1 and 122 μg C l−1, respectively, caused by a massive increase of vibrio-shaped cells. On the other hand, in early spring, filamentous bacteria were responsible for a marked increase in bacterial biomass, making up 40% of the total bacterial biomass. Over the year, rod-shaped cells were the dominating morphotype, while the biomass of cocci was rather negligible. In winter, cell numbers and biomass showed minimal values with 2.0 × 109 cells l−1 and 28 μg C l−1, respectively, and bacteria were considered to be substrate and temperature limited during this period. Saturation values of the incorporation of 3H-thymidine into DNA, for the estimation of bacterial secondary production, varied seasonally, ranging from 5 nm to 40 nm. Thus, saturation experiments needed to be conducted on a regular basis. Also, the amount of labeled thymidine in the DNA, as a percentage of labeled thymidine in the TCA precipitate, varied over the year. Minimum values of 45% were recorded during the cold season, while maximum values of 75–80% at the beginning of June coincided with high chlorophyll a values and minimal K m-values derived from saturation experiments. The potential role of the nitrogen-rich nucleoside thymidine as a readily utilizable substrate for bacteria during labeling experiments, under varying conditions of substrate availability, is discussed. Bacterial secondary production rates ranged from 0.3 μg C l−1 h−1 in winter to values of 10 μg C l−1 h−1 in August, where phytoplanktonic biomass reached the summer maximum, and bacterial biomass was calculated to be renewed 3 times per day. An estimation of the bacterial carbon demand showed that for the major part of the year, with the exception of early spring, the bacterioplankton community in the Kühw?rter Wasser was dependent on carbon sources other than phytoplanktonic primary production.
