Wild and solid-state cultures (SSC) of Taiwanofungus camphoratus (aka Antrodia camphorata and Chang-chih [CC]) were sequentially extracted with cold water, methanol, and hot water to get cold-water-soluble (CWS), methanol-soluble (MS), and hot-water-soluble (HWS) extracts, respectively. Only the MS extract exhibited angiotensin-converting enzyme (ACE) inhibitory activities. The antihypertensive effects of the MS extract (10 mg/kg BW) were measured in spontaneously hypertensive rats (SHR) and Wistar Kyoto (WKY) rats. MS extract of the SSC type was able to effectively lower the systolic blood pressure (SBP) and diastolic blood pressure (DBP) of SHR, but not of WKY rats, the results being significantly different from those for distilled water only (the blank). However, wild CC and its MS extract were not as effective as the SSC type in reducing SHR blood pressure and had no effect on WKY rats. SSC-type CC might be developed into a health food with the ability to regulate blood pressure.  相似文献

This study was initiated to screen the antioxidant activities, tyrosinase inhibitory effects on the fruiting bodies of Pleurotus ferulae extracted with acetone, methanol and hot water. The antioxidant activities were performed on β-carotene–linoleic acid, reducing power, DPPH, ferrous ions chelating abilities, and xanthine oxidase. In addition to this, phenolic compounds were also analyzed. The methanolic extract showed the strongest β-carotene–linoleic acid inhibition and high reducing power as compared to other extracts. The scavenging effects on DPPH radicals, the acetonic and methanolic extracts were more effective than hot water extracts. The strongest chelating effect was obtained from the methanolic extract as compared to the tested synthetic antioxidant. Gallic acid, protocatechuic acid, caffeic acid, vanillin, ferulic acid, naringin, resveratrol, naringenin, hesperetin, formononetin and biochanin-A were detected from acetonitrile and hydrochloric acid (5:1) solvent extract. Xanthine oxidase and tyrosinase inhibitory activities of acetonic, methanolic, and hot water extracts of P. ferulae increased with increasing concentration. The results suggested that consumption of P. ferulae might be beneficial to the antioxidant, xanthine oxidase, and tyrosinase protection system of the human body against oxidative damage and others complications.  相似文献

Animal model studies suggest that diets containing Laminaria angustata, a brown seaweed commonly eaten in Japan, inhibit breast carcinogenesis. In order to identify the compound(s) in the seaweed responsible for tumor-inhibiting activity, we used Ames/mammalian microsome assay system to determine the antimutagenic (or anticarcinogenic) effect of various solvents and water extracts of Laminaria angustata. The antimutagenic effects of acetone, ether, chloroform, chloroform + methanol, hot water and cold water extracts on the mutagenicity induced by 7,12-dimethylbenz[a]anthracene (DMBA), a breast carcinogen, and 3,2'-dimethyl-4-aminobiphenyl (DMAB), a colon and breast carcinogen, was studied using the Salmonella typhimurium strains TA98 and TA100. All extracts were nonmutagenic in both bacterial tester strains. The addition of 10-100 mg solvent extracts of seaweed/plate greatly inhibited DMAB-induced mutagenicity in both tester strains (80-96% inhibition) and DMBA-induced mutagenicity in TA100 (about 82%), whereas hot and cold water extracts produced a moderate inhibition in a dose-related manner in both strains.  相似文献

The sensitivity of two Gram positive (Staphylococcus aureus and Bacillus subtilis) and two Gram negative (Escherichia coli and Pseudomonas aeruginosa) pathogenic multi-drug resistant bacteria was tested against the crude extracts (cold aqueous, hot aqueous, and methanol extracts) of leaves and seeds of Argemone mexicana L. (Papaveraceae) by agar well diffusion method. Though all the extracts were found effective, yet the methanol extract showed maximum inhibition against the test microorganisms followed by hot aqueous extract and cold aqueous extract.  相似文献

We have screened a number of plants from the Indian soil for potential antioxidant properties out of which fifteen extracts were found to be positive. Leaves/bulk from the plants were crushed and extracted with organic solvents by three different ways. The first group of plants were extracted with CHCL3:CH3OH (2:1), evaporated, partitioned between petroleum ether and methanol (9:1), aqueous methanolic part re-partitioned between methanol: H2O (4:1) and dichloromethane. Methanol was evaporated from the aqueous methanolic part and extracted with n-butanol. The second group of plants were extracted with methanol followed by partitioning between petroleum ether and CH3OH. The rest of the extraction procedure was the same as above. A third extraction procedure was used for Ocimum sanctum which after extraction with CHCL3:CH3OH (2:l), partitioned between CCL4 and CH3OH:H2O (9:1). Aqueous methanolic part was repartitioned between CH3OH:H2O (4:1) and CHCI3 and CHCI3 soluble part was used for the study. Free radical scavenging activities of the plant extracts were examined by chemiluminescence method. Peroxyl radical was generated from 2,2'-azobis(2-amidinopropane) dihydrochloride (AAPH), superoxide radical (O2--) from xanthine/xanthine oxidase (XO) and hydroxyl radical (OH) from Xanthine/XO/ FeClJ EDTA. In addition, O2-- and OH. scavenging activities were also determined by cytochrome C reduction and deoxyribose oxidation methods, respectively. The results of this study demonstrate that these plant extracts possess potent antioxidant activities.  相似文献

Previous experiments indicated that water extracts of lambsquarters (Chenopodium album) and green foxtail (Setaria viridis), among others, stimulated growth of Bradyrhizobium japonicum in broth culture. Water extracts of lambsquarters shoots collected before or after anthesis were equally stimulatory. The stimulatory effect of extracts of lambsquarters when heated to 100°C for 30 min or autoclaved for 15 min was reduced by about 20% compared to untreated extracts. Extracts of green foxtail were less affected by higher temperature under similar conditions. Extracts of both green foxtail and lambsquarters completely lost their stimulatory effect following exposure to aerial microflora for 120 h. Water extract of lambsquarters shoots was more stimulatory than methanol extract, and neither ether nor butanol extracts resulted in stimulation. Both shoots and roots of lambsquarters and green foxtail were sequentially extracted first by water followed by methanol and vice-versa. The bioassay of these extracts indicated that there could be two components of the growth factor-one, larger component is soluble in water, the other, smaller component is soluble in methanol. After fractionation of the crude aqueous extract of lambsquarters shoots by four organic solvents, the residual aqueous extract retained the growth factor. Dialysis of the residual aqueous extract of lambsquarters shoots through a membrane (MWCO 1000) indicated that the molecular weight of the growth factor was less than 1000. The fraction having molecular weight <1000 was separated by paper chromatography using 6% acetic acid as developer. The fraction with Rf 0.91 showed the highest stimulation of the bacterium.  相似文献

The whole seed (W), endosperm (E) and hull (H) of five cultivars of Job’s tears (Coix lachryma-jobi Linn. var. ma-yuen Stapf) including Thai Black Phayao, Thai Black Loei, Laos Black Loei, Laos White Loei and Laos Black Luang Phra Bang were processed before solvent extraction by non-cooking, roasting, boiling and steaming Each part of the Job’s tears was extracted by the cold and hot process by refluxing with methanol and hexane. The total of 330 extracts included 150 methanol extracts and 180 hexane extracts were investigated for anti-proliferative activity on human colon adenocarcinoma cell line (HT-29) by the sulforhodamine B (SRB) assay. The extracts which gave high anti-proliferative activity were tested for apoptotic activity by acridine orange and ethidium bromide double staining and anti-oxidative activities including free radical scavenging and lipid peroxidation inhibition activities. The extract from the hull of Thai Black Loei roasted before extracting by hot methanol (M-HTBL-R2) showed the highest anti-proliferative activity on HT-29 with the IC50 values of 11.61 ± 0.95 μg/ml, while the extract from the non-cooked hull of Thai Black Loei by cold methanol extraction (M-HTBL-N1) gave the highest apoptosis (8.17 ± 1.18%) with no necrosis. In addition, M-HTBL-R2 and M-HTBL-N1 indicated free radical scavenging activity at the SC50 values of 0.48 ± 0.12 and 2.47 ± 1.15 mg/ml, respectively. This study has demonstrated the anti-colorectal cancer potential of the M-HTBL-R2 and M-HTBL-N1 extracts.  相似文献

Polyphenols content (as catechin equivalents) and tocopherol content were determined in borage defatted meal and borage oil, respectively. In addition, antioxidant activity of extracts obtained from borage defatted meal was evaluated. A cold pressing process was used for the extraction of Borago officinalis oil, resulting in a defatted meal (by-product). Polyphenols from this defatted borage meal were extracted using several solvents. An extract containing highly soluble solids and phenolic compounds with antioxidant activity (as free radical-scavenging, DPPH) was obtained when methanol was used. The tocopherol content was higher in oil extracted by cold pressing than in oil extracted with petroleum ether as organic solvent. An enzymatic treatment was applied (45 °C, 20% moisture, 0.25% E/S ratio, 1:1 Olivex:Celluclast enzymatic mixture) previously to borage oil extraction, which improved the antioxidant content in the borage defatted meal by three-folds, as compared to the values obtained by a nonenzyme-aided process.  相似文献

This is the first report on the phenolic composition and biological activities of endemic species Salvia jurisicii Ko?anin (Lamiaceae) originating from Macedonia. Aerial parts of S. jurisicii were extracted with dichloromethane, ethyl acetate, methanol, ethanol and water. All extracts were tested spectrophotometrically for total phenolic and flavonoid contents, while their phenolic composition was analyzed using HPLC-DAD. The antioxidant activity of extracts was studied using 2,2-dyphenyl-1-picrylhydrazyl, 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid), ferric reducing ability of plasma and ß-carotene bleaching assays. Being the most frequently used solvents for extraction of bioactive ingredients from medicinal plants, water and ethanol extracts were chosen for further testing of antimicrobial, cytotoxic and antineurodegenerative activities. More polar solvent extracts showed higher total phenolic content and lower flavonoid content. Ethanol and methanol extracts, followed by water extract exhibited the strongest antioxidant activity. Extracts showed antimicrobial activity against certain bacteria and micromycetes. The cytotoxicity assay showed slight toxicity to HCT-116 cell line. In the antineurodegenerative assays, the extracts performed lower inhibition of acetylcholinesterase and tyrosinase than standards. According to the results of this study, it can be inferred that more polar extracts of S. jurisicii were quantitatively richer in total phenolics, showing stronger antioxidant activity. Ethanol extract showed stronger biological activities comparing to water extract.  相似文献

Two cultivars of alfalfa (Medicago sativa L.), cold-tolerant Vernal and cold-sensitive Sonora, were grown under summer, winter, and dehardening conditions to determine the solubility characteristics and relationships of several dehydrogenases to cold tolerance.Soluble enzymatic proteins, extracted with three extractants, from lyophilized crown and root tissues, were separated by polyacrylamide disc gel electrophoresis.Gels assayed for glutamate, NAD-malate, NADP-malate, isocitrate, lactate, 6-phosphogluconate, and glucose-6-phosphate dehydrogenases showed quantitative differences in isoenzymes that were influenced by cultivar, extractant, and environmental differences.For both cultivars, enzyme activity was lowest during summer, increased in winter, and decreased during dehardening. Dehydrogenase activity, therefore, was closely associated with the fluctuations in soluble protein concentration, which were related to environmental changes and cold tolerance.Additional isoenzymes of isocitrate, lactate, and glucose-6-phosphate dehydrogenases were detected in the winter samples of both cultivars; however, most of the qualitative differences observed were generally due to the differential solubilities of isoenzymes in the three extractants.Comparison of data obtained from the use of frozen and unfrozen extracts indicated differential stabilities of the dehydrogenases to freezing in the different extractants. Glutamate, NAD-malate, and NADP-malate dehydrogenases were fairly stable to freezing whereas isocitrate, lactate, 6-phosphogluconate, and glucose-6-phosphate dehydrogenases were labile. Detectable levels of the latter dehydrogenases in frozen extracts were evident only in certain extracts of winter samples, indicating the importance of the nature of the extraction medium in protecting against enzyme denaturation.Since both cultivars showed similar changes in dehydrogenase activities at most times, the increased enzyme levels during winter coincided with increased levels of soluble protein and soluble sugars, which are indicative of the broad spectrum of metabolic changes involved in the attainment of the cold-tolerant state.  相似文献

Abstract— Glycogen was prepared from rapidly frozen rat brain by the usual techniques and found to contain considerable amounts of non-glycogen carbohydrate. The crude glycogen was partially purified by extraction with hot or cold water and reprecipitation. Enzymic estimation showed that the carbohydrate extracted into hot water contained only 50 per cent of glucose after hydrolysis; of the hot water insoluble material, namely some 30 per cent of the total carbohydrate present in the crude glycogen, less than half of the carbohydrate was released by hydrolysis in 1 M-HC1. The glycogen soluble in hot water incorporated 14C from [14C]glucose at considerably higher rates than the residual material and also decreased more rapidly during post-mortem autolysis. Glycogen extracted into cold water was of higher purity than that extracted by hot water; although the material behaved as glycogen during precipitation and re-extraction it contained only 75 per cent of its carbohydrate as glucose. Contaminants included fucose, galactose and hexuronic acid. The rates of metabolism of the partially purified glycogen are compared with published rates; it is suggested that the observed rates are inaccurate due to the impurities present in brain glycogen prepared by classical techniques.  相似文献

The antioxidant activities of methanol and ethyl ether extracts obtained from Thymus zygis, collected during the flowering or non-flowering period, were evaluated and compared. To investigate this potential, extracts were tested on their capacity to react with diphenyl-picrylhydrazyl (DPPH) in a homogeneous medium, and to inhibit Fe2+/ascorbate-induced membrane lipid peroxidation, as estimated by the formation of thiobar-bituric acid-reactive substances (TBARS). Although methanol extracts reduce DPPH radicals more efficiently than ethyl ether extracts, suggesting a potent radical scavenger activity, the ethyl ether extracts were found to be most active in inhibiting lipid peroxidation in sarcoplasmic reticulum (SR) membranes. In addition, both extracts present peroxyl and superoxide radical scavenging activities. Peroxyl radicals were generated by the water soluble 2, 2A-azobis(2-amidinopropane) dihydrochloride (AAPH) azoinitiator, and the scavenging activities of the extracts were measured by the inhibition of cis-parinaric acid (PnA) fluorescence decay in SR. Superoxide radicals were generated either by an enzymatic or a non-enzymatic system, and the scavenger ability was evaluated by the inhibition of nitrob-lue tetrazolium reduction. Methanolic extracts are more potent as scavengers of peroxyl and super oxide radicals than the ethyl ether extracts. Apparently, there is a relationship between antioxidant potency and the total phenolic groups content in each extract.  相似文献

Analyses on biological activities of Gnetum gnemon were done to determine the total phenolic and antioxidants of the plant. Four parts of G. gnemon were used in this study, which were leaf, bark, twig, and seeds of the plant. All parts were extracted in methanol, ethanol, hexane, chloroform and hot water using reflux. The total phenolic content of the plant extracts were determined by using Folin-Ciocalteu method. The results demonstrated that the bark from hot water extract showed the highest total phenolic at 10.71?±?0.01 mg GAE/ FDW, while the lowest was chloroform extract of seed at 2.15?±?0.01 mg GAE/ FDW. The antioxidant activity of the plant extracts were determined by using DPPH and FRAP assays, respectively. The DPPH results showed that all plant extracts demonstrated weak free radical scavenging activity tested at the final concentration of 300 μg/ml. In contrast, the methanolic twig extract showed strong reducing power activity (FRAP) at 83.55?±?1.05%, while the hot water seed extract showed the least activity at 41.86?±?4.22% tested at the final concentration of 300 μg/ml. However, there were no correlation between total phenolics and both antioxidant assays tested.  相似文献

Locust bean gum extracted from two carob flours from eastern and western Mediterranean sources were fractionated on the basis of their solubility in water. Weight-average molecular weights determined by sedimentation equilibrium were about 300 000 for both the hot water and cold water soluble fractions, whereas a commercial sample of guar gum had a molecular weight of 700 000. Their values were lower than would be predicted from Mark-Houwink relationships where molecular weights were originally determined by light scattering.

The hot water soluble fraction from the eastern Mediterranean flour showed unexpected rheological behaviour. It had an extremely high Huggins' constant and a different relationship between the coil overlap parameter and the zero shear rate viscosity compared with previously reported results for galactomannans. Both effects may be explained by the anomalously low intrinsic viscosity of this fraction when determined by a Huggins' extrapolation. The use of the Kraemer extrapolation gave significantly higher intrinsic viscosities for this particular sample. Gels formed from the two hot water soluble fractions with κ-carrageenan had similar rheological properties.  相似文献

Three underutilized leafy vegetables Sarcochlamys pulcherrima (Roxb.) Gaudich (SP), Ipomoea aquatica Forssk. (IA) and Zanthoxylum rhetsa (Roxb.) DC (ZR) were extracted with different solvents viz. 95 % ethyl alcohol, methanol and hot water. The extracts were evaluated for their antioxidant potential via DPPH, ABTS and FRAP assay along with electroanalytical studies using cyclic voltammetry. The antidiabetic potential was determined by recording their α-amylase and α-glucosidase inhibitory assay. The total phenolic content (TPC), total flavonoid content (TFC) and the liquid chromatography-mass spectrometry (LC/MS) based phytochemical profiles of the extracts were also determined. All three extracts of SP exhibited significant antioxidant capacity. The antidiabetic potential of the IA and ZR extracts was found to be higher than or at par with that of standard acarbose. LC/MS studies reveal the presence of hitherto reported antioxidant and antidiabetic compounds like gamma-aminobutyric acid, cinnamic acid, caffeic acid, α-viniferin, piperlonguminine, niacin, kaempferol, etc., in the extracts.  相似文献

BackgroundMedicinal plants are important source of drugs with pharmacological activities. Therefore, there is always rising demands to discover more therapeutic agents from various species. Orthosiphon stamineus, Gynura procumbens and Ficus deltoidea are high valued medicinal plants of Malaysia contain rich source of phenolic and flavonoid compounds. The aims of the present study were to evaluate anti-oxidant, antimicrobial and anti-proliferative effects on A549, HePG2 and MCF7 cell lines of four different extracts of Orthosiphon stamineus, Gynura procumbens and Ficus deltoidea.MethodologyThe leaves of all selected plants were extracted with methanol, chloroform, ethyl acetate and butanol separately with simple cold maceration. Antioxidant activity of all crude extracts were quantitatively measured against DPPH and Ferric Reducing Assay. Antimicrobial evaluation was done by Microdilution and MTT assay and antipoliferative activity of all extracts of selected plant were evaluated against A549, HePG2 and MCF7 cell lines.ResultsResults showed that methanol extract exhibited highest percentage free radical scavenging activity of almost all extracts of selected plants. Antimicrobials results showed chloroform and methanol extracts of O. stamineus extract were the two most active extracts against resistant MRSA but not S. aureus. Only methanol extract of G. procumbens showed antimicrobial activity against the tested pathogens. Chloroform and methanol extracts of F. deltoidea elicited antimicrobial activity against S. aureus but not MRSA. Antiproliferative activity against three tested cell lines results showed that ethyl acetate extract of O. stamineus showed good effect whereas methanol extract of F. deltoidea and G. procumbens exhibited good antiproliferative activity.ConclusionsThe results of the present investigation demonstrated significant variations in the antioxidant, antimicrobial and antiproliferative effects of different solvent extracts. These data could be helpful in isolation of pure potent compounds with good biological activities from the extracts of plants.  相似文献

Flax bast tissue was sequentially extracted using hexane, propanol, methanol and water as solvents and extracts were analyzed using reverse phase HPLC and 13C NMR. Results indicated a large variety of aromatic constituents including flavonoids and hydroxy-methoxy cinnamic acids linked to oligosaccharides and hydroxy acids through glycosidic linkages. The extracts inhibited cellulase and pectinase activities and can thus influence retting.  相似文献
