The adsorption of the hydrophobic anion [W(CO)(5)CN](-) to human lymphoid Jurkat cells gave rise to an additional anti-field peak in the rotational spectra of single cells, indicating that the cell membrane displayed a strong dielectric dispersion in the kilohertz to megahertz frequency range. The surface concentration of the adsorbed anion and its translocation rate constant between the two membrane boundaries could be evaluated from the rotation spectra of cells by applying the previously proposed mobile charge model. Similar single-cell electrorotation experiments were performed to examine the effect of phloretin, a dipolar molecule known to influence the dipole potential of membranes, on the transport of [W(CO)(5)CN](-) across the plasma membrane of mammalian cells. The adsorption of [W(CO)(5)CN](-) was significantly reduced by phloretin, which is in reasonable agreement with the known phloretin-induced effects on artificial and biological membranes. The IC(50) for the effect of phloretin on the transport parameters of the lipophilic ion was approximately 10 microM. The results of this study are consistent with the assumption that the binding of phloretin reduces the intrinsic dipole potential of the plasma membrane. The experimental approach developed here allows the quantification of intrinsic dipole potential changes within the plasma membrane of living cells.  相似文献

The charge-pulse relaxation spectrum of nonperfused and perfused (turgescent) cells of the giant marine alga Ventricaria ventricosa showed two main exponential decays with time constants of approximately 0.1 msec and 10 msec, respectively, when the cells were bathed in artificial sea water (pH 8). Variation of the external pH did not change the relaxation pattern (in contrast to other giant marine algae). Addition of nystatin (a membrane-impermeable and pore-forming antibiotic) to the vacuolar perfusion solution resulted in the disappearance of the slow exponential, whereas external nystatin decreased dramatically the time constant of the fast one. This indicated (by analogy to corresponding experiments with Valonia utricularis, J. Wang, I. Spiess, C. Ryser, U. Zimmermann, J. Membrane Biol. 157: 311-321, 1997) that the fast relaxation must be assigned to the RC-properties of the plasmalemma and the slow one to those of the tonoplast. Consistent with this, external variation of [K+]o or of [Cl-]o as well as external addition of K+- or Cl--channel/carrier inhibitors (TEA, Ba2+, DIDS) affected only the fast relaxation, but not the slow one. In contrast, addition of these inhibitors to the vacuolar perfusion solution had no measurable effect on the charge-pulse relaxation spectrum. The analysis of the data in terms of the "two membrane model" showed that K+- and (to a smaller extent) Cl--conducting elements dominated the plasmalemma conductance. The analysis of the charge-pulse relaxation spectra also yielded the following area-specific data for the capacitance and the conductance for the plasmalemma and tonoplast (by assuming that both membranes have a planar surface): (plasmalemma) Cp = 0.82 * 10(-2) F m-2, Rp = 1.69 * 10(-2) Omega m2, Gp = 5.9 * 10(4) mS m-2, (tonoplast) Ct = 7. 1 * 10(-2) F m-2, Rt = 14.9 * 10(-2) Omega m2 and Gt = 0.67 * 10(4) mS m-2. The electrical data for the tonoplast show that (in contrast to the literature) the area-specific membrane resistance of the tonoplast of these marine giant algal cells is apparently very high as reported already for V. utricularis. The exceptionally high value of the area-specific capacitance could be explained - among other interpretations - by assuming a 9-fold enlargement of the tonoplast surface. The hypothesis of a multifolded tonoplast was supported by transmission electronmicroscopy of cells fixed under maintenance of turgor pressure and of the electrical parameters of the membranes. This finding indicates that the tonoplast of this species exhibited a sponge-like appearance. Taking this result into account, it can be easily shown that the tonoplast exhibits a high-resistance (1.1 Omega m2). Vacuolar membrane potential measurements (performed in parallel with charge-pulse relaxation studies) showed that the potential difference across the plasmalemma was mainly controlled by the external K+-concentration which suggested that the resting membrane potential of the plasmalemma is largely a K+-diffusion potential. After permeabilization of the tonoplast with nystatin the potential of the intact membrane barrier dropped from about slightly negative or positive (-5.1 to +18 mV, n = 13) to negative values (-15 up to -68 mV; n = 8). This indicated that the cytoplasm of V. ventricosa was apparently negatively charged relative to the external medium. Permeabilization of the plasmalemma by addition of external nystatin resulted generally in an increase in the potential to slightly more positive values (-0.8 to +4.3 mV; n = 5), indicating that the vacuole is positively charged relative to the cytoplasm. These findings apparently end the long-term debate about the electrical properties of V. ventricosa. The results presented here support the findings of Davis (Plant Physiol. 67: 825-831, 1981), but are contrary to the results of Lainson and Field (J. Membrane Biol. 29: 81-94, 1976).  相似文献

Reaction of the allylidene tungsten complex [W(CPhCHCHMe)Br2(CO)2(4-picoline)] (1) with the dithiocarbamates MS2CNR2 (a: M=Na, R=Et; b: M=Na, R=Me; c: M=Li, R=Ph) in THF at 50 °C affords the vinylketene tungsten complexes [W(S2CNR2)2(OCCPhCHCHMe)(CO)] (2a–c). At lower temperatures, four reaction intermediates (3–6) may be discerned. Spectroscopic studies indicate that these compounds contain η4-allyldithiocarbamate ligands which are generated by addition of dithiocarbamate across the metal-carbon double bond of the allylidene-tungsten unit in 1. The structures of [W(S2CNEt2)2(OCCPhCHCHMe)(CO)] (2a) and of one intermediate, [W(η4-Et2NCS2CPhCHCHMe)(S2CNEt2)(CO)2] (5a) were elucidated by X-ray crystallography.  相似文献

Mo(CO)4(LL) complexes, where LL = polypyridyl ligands such as 2,2′-bipyridine and 1,10-phenanthroline, undergo quasi-reversible, one-electron oxidations in methylene chloride yielding the corresponding radical cations, [Mo(CO)4(LL)]+. These electrogenerated species undergo rapid ligand substitution in the presence of acetonitrile, yielding [Mo(CO)3(LL)(CH3CN)]+; rate constants for these substitutions were measured using chronocoulometry and were found to be influenced by the steric and electronic properties of the polypyridyl ligands. [Mo(CO)3(LL)(CH3CN)]+ radical cations, which could also be generated by reversible oxidation of Mo(CO)3(LL)(CH3CN) in acetonitrile, can be irreversibly oxidized yielding [Mo(CO)3(LL)(CH3CN)2]2+ after coordination by an additional acetonitrile. Infrared spectroelectrochemical experiments indicate the radical cations undergo ligand-induced net disproportionations that follow first-order kinetics in acetonitrile, ultimately yielding the corresponding Mo(CO)4(LL) and [Mo(CO)2(LL)(CH3CN)3]2+ species. Rate constants for the net disproportionation of [Mo(CO)3(LL)(CH3CN)]+ and the carbonyl substitution reaction of [Mo(CO)3(LL)(CH3CN)2]2+ were measured. Thin-layer bulk oxidation studies also provided infrared characterization data of [Mo(CO)4(ncp)]+ (ncp = neocuproine), [Mo(CO)3(LL)(CH3CN)]+, [Mo(CO)3(LL)(CH3CN)2]2+ and [Mo(CO)2(LL)(CH3CN)3]2+ complexes.  相似文献

N-[(2-Pyridyl)methyliden]-6-coumarin (L) is synthesized by the condensation of 6-aminocoumarin and pyridine-2-carboxaldehyde. Group-6 tetracarbonyl complexes, [M(CO)4(L)] (M = Cr, Mo, and W) are synthesized and characterized by mass spectrometry and NMR, FT-IR and UV-visible spectroscopy. X-ray crystal structure of [Cr(CO)4(L)] shows N(pyridine), N(imine) chelation to chromium(0). A supramolecular chain is formed by C-H?O and π?π interactions. The ligand and the complexes are fluorescent. Cyclic voltammetry of the complexes exhibit quasireversible M(I)/M(0) redox couple. The complexes exhibit potential antioxidant property both in cell free and in-vitro studies and highest activity is observed to [W(CO)4(L)]. Density functional theory (DFT) computation has been performed to correlate with the electronic configuration, composition of wave functions with the UV-visible spectra and redox properties.  相似文献

We have analyzed the structure of cytochrome c (cyt c) bound in a variety of complexes in which negatively charged molecular groups interact with the positively charged binding domain around the heme crevice of cyt c. Using resonance Raman spectroscopy, we could demonstrate that these interactions induce the same conformational changes as they were observed in the surface-enhanced resonance Raman experiments of cyt c adsorbed on the Ag electrode [Hildebrandt & Stockburger (1989) Biochemistry (preceding paper in this issue)]. When cyt c is bound to (As4W40O140)27-, state II is stabilized, whereas in complexes with phosvitin and cytochrome b5 state I is formed. The complexes with phospholipid vesicles and inverted micelles reveal a mixture of both states. It is suggested that these systems as well as cyt c adsorbed on the Ag electrode may be regarded as model systems for the physiological complexes of cyt c with cytochrome oxidase and cytochrome reductase. On the basis of our findings it is proposed that the biological electron-transfer reactions are controlled by electric field induced conformational transitions of cyt c upon complex formation with its physiological redox partners.  相似文献

Various Cr, Mo and W carbonyl complexes of a tridentate ligand containing N and P as donor atoms, bis(2-(diphenylphosphino)ethyl)benzylamine (DPBA), have been synthesized. Reaction of M(CO)6 (M = Cr, Mo and W) with DPBA in a 1:1 mole ratio in toluene or xylene, resulted in the formation of facial and meridional complexes of the type [M(CO)3(DPBA)] (M = Cr, Mo and W). Interaction of Cr(CO)6 or Mo(CO)6 with DPBA and PPh3 in toluene yielded complexes of the composition [Cr(CO)3(DPBA)(PPh3)] and [Mo(CO)2(DPBA)(PPh3)], respectively. However reaction of W(CO)6 with DPBA and PPh3 yielded only [W(CO)3(DPBA)]. Reaction of Cr(CO)6 with DPBA and 1,2-bis(diphenylphosphino)ethane(diphos) in toluene for 24 h resulted in the formation of a mixed ligand complex, [Cr(CO)4(DPBA)(diphos)] where both the ligands coordinate to the metal atom through only one of their donor atoms. A unique binuclear complex of the composition [Mo(CO)2(DPBA)(diphos)]2 resulted, with the tridentate ligand DPBA acting as a bidentate bridging ligand, by the reaction of Mo(CO)6 with DPBA and diphos in refluxing xylene for 24 h. All the complexes are characterized by elemental analysis and infrared spectra. The 31P{1H} and 1H NMR spectral data of the complexes gave valuable information in elucidating the structures of the complexes. The ligand DPBA has found to behave in a triligate monometallic, biligate monometallic, monoligate monometallic and biligate bimetallic manner.  相似文献

Interactions of structurally dissimilar anionic compounds with the plasma membrane of HEK293 cells were analyzed by patch clamp and electrorotation. The combined approach provides complementary information on the lipophilicity, preferential affinity of the anions to the inner/outer membrane leaflet, adsorption depth and transmembrane mobility. The anionic species studied here included the well-known lipophilic anions dipicrylamine (DPA), tetraphenylborate (TPB) and [W2(CO)10(S2CH)], the putative lipophilic anion and three new heterocyclic W(CO)5 derivatives. All tested anions partitioned strongly into the cell membrane, as indicated by the capacitance increase in patch-clamped cells. The capacitance increment exhibited a bell-shaped dependence on membrane voltage. The midpoint potentials of the maximum capacitance increment were negative, indicating the exclusion of lipophilic anions from the outer membrane leaflet. The adsorption depth of the large organic anions DPA, TPB and increased and that of W(CO)5 derivatives decreased with increasing concentration of mobile charges. In agreement with the patch-clamp data, electrorotation of cells treated with DPA and W(CO)5 derivatives revealed a large dispersion of membrane capacitance in the kilohertz to megahertz range due to the translocation of mobile charges. In contrast, in the presence of TPB and no mobile charges could be detected by electrorotation, despite their strong membrane adsorption. Our data suggest that the presence of oxygen atoms in the outer molecular shell is an important factor for the fast translocation ability of lipophilic anions.  相似文献

The seven-coordinate complexes [MI2(CO)3(NCMe)2] (M = Mo and W) react with one equivalent of BiPh3 in CH2Cl2 at room temperature to give the monoacetonitrile complexes [MI2(CO)3(NCMe)(BiPh3)]. The molybdenum complex [MoI2(CO)3(NCMe)(BiPh3)] after stirring in CH2Cl2 at room temperature for 5 h affords the iodide-bridged dimer [Mo(μ-I)I(CO)3(BiPh3)]2, whereas the tungsten complex [WI2(CO)3(NCMe)(BiPh3)] does not appear to dimerise even after stirring for 48 h in CH2Cl2 at room temperature. Reaction of [MI2(CO)3(NCMe)2] with two equivalents of BiPh3 gives the bistriphenylbismuth compounds [MI2(CO)3(BiPh3)2] in good yield. The new mixed ligand complexes [MI2(CO)3L(BiPh3)] were prepared either by reaction of [MI2(CO)3(NCMe)(BiPh3)]in situ with one equivalent of L(L = P(OPh)3), or an in situ reaction of [MI2(CO)3(NCMe)L] (L = PPh3 and SbPh3; and L = AsPh3 and PPh2Cy (for M = Mo only) with an equimolar quantity of BiPh3. Reaction of [MoI2(CO)3(NCMe)(BiPh3)] with one equivalent of 2,2′-bipyridyl (bipy) in CH2Cl2 at room temperature afforded the cationic complexes [MoI(CO)3(bipy)(BiPh3)]I in good yield. The complex [WI2(CO)3(NCMe)(BiPh3)] (prepared in situ) reacts with two equivalents of NaS2CNMe2·2H2O to eventually give the non-triphenylbismuth containing product [W(CO)3(S2CNMe2)2] in high yield.  相似文献

Charge-pulse experiments were performed on cells of the giant marine algaHalicystis parvula. At normal pH (8.2), the voltage decay following a charge-pulse of 500 ns duration fed to the vacuole could be described by summing two exponential relaxations. The amplitudes and time constants of these relaxations were widely separated. The parameters of the two relaxation processes were found to be pH-dependent. Reduction of the external pH value from pH 8.2 to 5 resulted in a complete change of the two relaxation processes within a few minutes. Only one relaxation process could be observed at pH 5, within the time resolution of our instrumentation. The experimental data could not be explained by a two-membrane model with reasonable values for the specific capacitances of tonoplast and plasmalemma. The results of the charge-pulse relaxations were found to be consistent with the assumption that both membranes have very similar electrical properties and that both contain mobile charges with a total surface concentration of about 30 nmol·m-2 and a translocation-rate constant of about 500·s-1. The mobile charges became neutralized at pH 5 hhich led to a decrease of the apparent specific capacitance of the algal cells. Fatty acid binding proteins (FABPs) can facilitate the transfer of long-chain fatty acids between intracellular membranes across considerable distances. The transfer process involves fatty acids, their donor membrane and acceptor membrane, and FABPs, implying that potential protein-membrane interactions exist. Despite intensive studies on FABP-membrane interactions, the interaction mode remains elusive, and the protein-membrane association and dissociation rates are inconsistent. In this study, we used nanodiscs (NDs) as mimetic membranes to investigate FABP-membrane interactions. Our NMR experiments showed that human intestinal FABP interacts weakly with both negatively charged and neutral membranes, but it prefers the negatively charged one. Through simultaneous analysis of NMR relaxation in the rotating-frame (R1ρ), relaxation dispersion, chemical exchange saturation transfer, and dark-state exchange saturation transfer data, we estimated the affinity of the protein to negatively charged NDs, the dissociation rate, and apparent association rate. We further showed that the protein in the ND-bound state adopts a conformation different from the native structure and the second helix is very likely involved in interactions with NDs. We also found a membrane-induced FABP conformational state that exists only in the presence of NDs. This state is native-like, different from other conformational states in structure, unbound to NDs, and in dynamic equilibrium with the ND-bound state.  相似文献

The complex cis-dicyanobis(2,2'-bipyridine)ruthenium(II) forms various bimetallic complexes with mercury(II)chloride, such as [(NC)Ru(bpy)2(CN)-HgCl2], [Cl2Hg-(NC)Ru(bpy)2(CN)-HgCl2-(NC)Ru(bpy)2(CN)-HgCl2] and [Cl2Hg-(NC)Ru(bpy)2(CN)-(HgCl2)] in CH3CN. These polynuclear complexes of the equilibrium system have been identified and characterized by their formation constants and absorption spectra. Excitation of bimetallic complexes produces the MLCT state localized on [Ru(bpy)2(CN)2] ligand, resulting in the cleavage of the bond formed between the nitrogen atom of the coordinated cyanide ligand and the Hg(II) central atom in ground state. Unlike many photoinduced metal ligand dissociations, the dissociated fragment remains in a luminescent excited state.  相似文献

The mitochondrial precursor protein, apocytochrome c, binds to model membranes containing negatively charged phospholipids (Rietveld, A., Sijens, R., Verkleij, A.J. and Kruijff, B. (1983) EMBO J. 2, 907-913). In the present paper the effect of apocytochrome c on the lipid distribution in model membranes, consisting of neutral and acidic phospholipids, is examined. Both ESR and fluorescence energy transfer experiments show that the protein preferentially interacts with the negatively charged phospholipid in the mixed model membranes. Semi-quantitative analysis of the fluorescence energy transfer from the single tryptophan in apocytochrome c to the parinaric acid in phosphatidylserine or phosphatidylcholine in mixed bovine brain phosphatidylserine/egg phosphatidylcholine vesicles reveals and average donor-acceptor distance of 22-26 A and 26-30 A for phosphatidylserine and phosphatidylcholine, respectively. In addition, these experiments demonstrate that this preferential interaction does not induce the separation of large domains enriched in complexes of apocytochrome c with negatively charged phospholipids and domains enriched in neutral lipids.  相似文献

Use and application of Schiff bases are extended to many different fields of technology. (ISE)M(CO)5 complex [M?=?Cr (1), Mo (2), W (3), and where ISE is 3[4-ethyl(phenly)imino][indoline-2-one]; and (ISB)M(CO)5 [M?=?Cr (4), Mo (5), W (6)], where ISB is 3[4-butly(phenly)imino][indoline-2-one] were investigated by computational methods. Computations were carried out using density functional theory (DFT) with B3LYP and CAM-B3LYP functionals, in conjunction with LanL2DZ basis set for metals and cc-PVTZ basis set for other atoms. Time-dependent density functional theory (TDDFT) was used at the same level to obtain the electronic transitions. Molecular orbital energies, UV-Vis spectra, and total electron densities of investigated molecules were shown in the gas phase and in THF. Metal complexes showed higher absorption coefficients compared to ISE and ISB in the visible region. Additionally, they displayed absorption peaks at longer wavelengths and full MLCT character in solution, and W complexes required less energy compared to the complexes of other investigated metal ions. Among the investigated systems, (ISE)W(CO)5 and (ISB)W(CO)5 complexes with lowest HOMO-LUMO gaps are found to be the best candidates for photosensitive material production.

A bioorganometallic approach to malaria therapy led to the discovery of ferroquine (FQ, SSR97193). To assess the importance of the electronic properties of the ferrocenyl group, cyclopentadienyltricarbonylrhenium analogues related to FQ, were synthesized. The reaction of [N-(7-chloro-4-quinolinyl)-1,2-ethanodiamine] with the cyrhetrenylaldehyde complexes (η(5)-C(5)H(4)CHO)Re(CO)(3) and [η(5)-1,2-C(5)H(3)(CH(2)OH)(CHO)]Re(CO)(3) produces the corresponding imine derivatives [η(5)-1,2-C(5)H(3)(R)(CHN-CH(2)CH(2)NH-QN)]Re(CO)(3) R=H 3a; R=CH(2)OH 3b; QN=N-(7-Cl-4-quinolinyl). Reduction of 3a and 3b with sodium borohydride in methanol yields quantitatively the amine complexes [η(5)-1,2-C(5)H(3)(R)(CH(2)-NH-CH(2)CH(2)NH-QN)]Re(CO)(3) R=H 4a; R=CH(2)OH 4b. To establish the role of the cyrethrenyl moiety in the antimalarial activity of this series, purely organic parent compounds were also synthesized and tested. Evaluation of antimalarial activity measured in vitro against the CQ-resistant strains (W2) and the CQ-susceptible strain (3D7) of Plasmodium falciparum indicates that these cyrhetrene conjugates are less active compared to their ferrocene and organic analogues. These data suggest an original mode-of-action of FQ and ferrocenyl analogues in relationship with the redox pharmacophore.  相似文献

Certain amphiphilic copolymers form lipid-bilayer nanodiscs from artificial and natural membranes, thereby rendering incorporated membrane proteins optimal for structural analysis. Recent studies have shown that the amphiphilicity of a copolymer strongly determines its solubilization efficiency. This is especially true for highly negatively charged membranes, which experience pronounced Coulombic repulsion with polyanionic polymers. Here, we present a systematic study on the solubilization of artificial multicomponent lipid vesicles that mimic inner mitochondrial membranes, which harbor essential membrane-protein complexes. In particular, we compared the lipid-solubilization efficiencies of established anionic with less densely charged or zwitterionic and even cationic copolymers in low- and high-salt concentrations. The nanodiscs formed under these conditions were characterized by dynamic light scattering and negative-stain electron microscopy, pointing to a bimodal distribution of nanodisc diameters with a considerable fraction of nanodiscs engaging in side-by-side interactions through their polymer rims. Overall, our results show that some recent, zwitterionic copolymers are best suited to solubilize negatively charged membranes at high ionic strengths even at low polymer/lipid ratios.  相似文献

The dielectric properties of suspensions of intact cells of Methylophilus methylotrophus, Paracoccus denitrificans and Bacillus subtilis have been measured in the frequency range 1 kHz to 13 MHz. All possess a pronounced dispersion corresponding in magnitude and relaxation time to the "beta-dispersion" in a terminology defined by Schwan [Adv. Biol. Med. Phys. 5:147-209 (1957)]. The latter two strains, but not M. methylotrophus, also possess a substantial alpha-dispersion. The relaxation time of the beta-dispersion of B. subtilis is significantly lower than that of the other two strains, due to the higher internal K+ content of this Gram-positive organism. Treatment of P. denitrificans or B. subtilis with lysozyme greatly reduces the magnitude of the alpha-dispersion; in the latter case it is virtually abolished. The magnitude of both the alpha- and beta-dispersions of protoplasts of these organisms is significantly decreased by treatment with the cross-linking reagent glutaraldehyde, indicating that diffusional motions of the lipids and/or proteins in the protoplast membranes contribute to the dielectric relaxations observed in this frequency range. Such motions cannot be unrestricted, as in the "fluid mosaic" model, since the relaxation times of the lipids and proteins, if restricted by hydrodynamic forces alone, should then correspond, in protoplasts of this radius (0.4-0.5 micron), to approximately 10 Hz. Even after treatment of the (spherical) protoplasts with glutaraldehyde, the breadth of the remaining beta-dispersion is still significantly greater than (a) that of a pure Debye dispersion and (b) that to be expected solely from a classical Maxwell-Wagner-type mechanism. It is recognised that the surfaces of the protein complexes in such membranes extend significantly beyond the membrane surface as delineated by the phospholipid head-groups; such molecular granularity can in principle account for the broadened dielectric relaxations in the frequency range above 1 kHz, in terms of the impediment to genuinely tangential counterion relaxation caused by the protruding proteins themselves. The relaxation time of a previously observed, novel, low-frequency, glutaraldehyde-sensitive (mu-) dispersion in bacterial chromatophore suspensions, as well as that of their alpha-dispersion, is significantly increased by increasing the aqueous viscosity with glycerol. This finding is consistent with the view that, from a dielectric standpoint, the motions of charged proteins (and lipids) in biological membranes are rather tightly coupled to those of the adjacent ions and dipoles in the electric double layer.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献

The photochemical reaction of (eta5-C5H5)Mo(CO)3I with maleimide in the presence of diisopropylamine yielded complex (eta5-C5H5)Mo(CO)3(eta1-N-maleimidato) 4 in 52% yield. The single-crystal X-ray structure of this complex was determined and shows unusual interactions between oxygen atoms of the maleimidato ligand and carbon atoms of the cis-CO ligands. The tungsten analogue of 4, (eta5-C5H5)W(CO)3(eta1-N-maleimidato) 5, was synthesized in 37% yield by the reaction of (eta5-C5H5)W(CO)3I with the thallium(I) salt of maleimide. Complexes 4 and 5 reacted with cysteine ethyl ester and glutathione to afford products of the addition of the sulfhydryl group to the ethylenic bond of the maleimidato ligand. The reaction of 4 and 5 with glutathione proceeded faster than the reaction of the analogous complex (eta5-C5H5)Fe(CO)2(eta1-N-maleimidato) (3). However, all these complexes react with glutathione more slowly than N-ethylmaleimide. Complexes 4 and 5 were used for labeling of bovine serum albumin (BSA), enriched in thiol groups by reaction with Traut's reagent. Reaction of thiolated BSA containing 7.4 SH groups with 4 and 5 gave bioconjugates bearing 6.9 and 6.4 metallocarbonyl moieties, respectively. Under the same conditions, reaction with 3 afforded a BSA conjugate containing 7.6 metallocarbonyl moieties. Labeling was presumed to be site-specific, as the number of metallocarbonyl entities matched very well with the initial number of SH groups measured for the thiolated BSA sample. IR spectra of BSA labeled with 4 and 5 show intense nu(C[triple bond]O)) bands (2042 and 1948 cm(-1) in the latter case), enabling sensitive detection of the bioconjugates in biological samples. Complexes 4 and 5 (especially the latter) should be of interest as heavy atom phasing reagents for protein X-ray crystallography.  相似文献

We find that curvature-driven growth of pores in electrically charged membranes correctly reproduces charge-pulse experiments. Our model, consisting of a Langevin equation for the time dependence of the pore radius coupled to an ordinary differential equation for the number of pores, captures the statistics of the pore population and its effect on the membrane conductance. The calculated pore radius is a linear, and not an exponential, function of time, as observed experimentally. Two other important features of charge-pulse experiments are recovered: pores reseal for low and high voltages but grow irreversibly for intermediate values of the voltage. Our set of coupled ordinary differential equations is equivalent to the partial differential equation used previously to study pore dynamics, but permits the study of longer timescales necessary for the simulations of voltage-clamp experiments. An effective phase diagram for such experiments is obtained.  相似文献

Carr-Purcell-Meiboom-Gill (CPMG) relaxation dispersion nuclear magnetic resonance (NMR) spectroscopy has emerged as a powerful method for quantifying chemical shifts of excited protein states. For many applications of the technique that involve the measurement of relaxation rates of carbon magnetization it is necessary to prepare samples with isolated (13)C spins so that experiments do not suffer from magnetization transfer between coupled carbon spins that would otherwise occur during the CPMG pulse train. In the case of (13)CO experiments however the large separation between (13)CO and (13)C(alpha) chemical shifts offers hope that robust (13)CO dispersion profiles can be recorded on uniformly (13)C labeled samples, leading to the extraction of accurate (13)CO chemical shifts of the invisible, excited state. Here we compare such chemical shifts recorded on samples that are selectively labeled, prepared using [1-(13)C]-pyruvate and NaH(13)CO(3,) or uniformly labeled, generated from (13)C-glucose. Very similar (13)CO chemical shifts are obtained from analysis of CPMG experiments recorded on both samples, and comparison with chemical shifts measured using a second approach establishes that the shifts measured from relaxation dispersion are very accurate.  相似文献
