Members of seven genra of bacteria, pathogens and nonpathogens of plants, adhered to young leaves when leaves were suspended in cell suspensions for 10 min. With Pseudomonas lachrymans, the adherence rate (cells applied vs. cells adhering) to host (cucumber) and nonhost (chrysanthemum) leaves was a straight-line, log-log function, as was the adherence of Escherichia coli and Serratia marcescens to cucumber leaves. Adhering cells of these three bacteria were washed with water from cucumber leaves at a straight-line, log-log rate. Adhered cells of P. lachrymans were most commonly found near veins on cucumber leaves. There appeared to be a polymeric surface layer on this bacterium on the cucumber leaf when leaves bearing bacteria were stained with ruthenium red and viewed in thin section.  相似文献

True fluorescence spectra of leaves are obtained combining experimental emission and reflectance data. The model used is validated by measuring the system fluorescence in the absence of reabsorption processes (thin layer of chloroplasts).  相似文献

Leaf shape is an important factor in optimal plant growth, because leaves are the main photosynthetic organs. Plants exhibit plasticity in leaf shape and structure, allowing them to optimize photosynthetic efficiency. In Arabidopsis thaliana(L.) Heynh., several types of leaves develop differentially, according to light intensity and quality. When shaded, the expansion of leaf lamina is inhibited, while the petiole elongation is enhanced. This phenomenon is part of the so-called shade-avoidance syndrome. Under low light, A. thaliana develops shade leaves with only one layer of palisade tissue, whereas under high light, it develops sun leaves that have nearly two complete layers of palisade tissue. Although the molecular mechanisms of these photomorphogenic phenomena in leaves are not well understood, recent studies of A. thaliana have provided some insight. For example, some cytochrome P450s may be involved in the specific control of the petiole length during photomorphogenesis. On the other hand, switching between sun and shade leaves is regulated by long-distance signaling from mature leaves in Chenopodium album. Here we provide an overview of the mechanisms of photomorphogenesis in leaves based on recent findings.  相似文献

Summary In order to study the foliar endophytes from teak (Tectona grandis L.) and rain tree (Samanea saman Merr.) growing in the campus of Chulalongkorn University, healthy leaves were collected at two-monthly intervals during January to December. The number of genera and species, together with their colonization frequency (CF%) in mature teak and rain tree leaves were greater than those in the young leaves. More endophytic isolates in the leaves of both trees were recovered during the rainy season. The fungal genera found in both young and mature teak leaves were Alternaria, Colletotrichum, Nigrospora, Phomopsis and mycelia sterilia. Phomopsis was the dominant genus in both young (newly emerged) and mature leaves. Fusarium, Penicillium, Schizophyllum commune and members of the Xylariaceae were found only in mature leaves. For the rain tree leaves, species of Phomopsis and mycelia sterilia were found in both young newly emerged and mature leaves. Colletotrichum and Penicillium were found only in mature leaves, whereas Nigrospora was found only in young newly emerged leaves. In this study, Phomopsis was the dominant genus in the leaves of both tree species. A total of 37 isolates of endophytic fungi isolated from teak and rain tree leaves were tested for the production of antimicrobial activities. Out of these, 18 isolates could produce inhibitory substances effective against Bacillus subtilis, Staphylococcus aureus and Escherichia coli and 3 isolates inhibited growth of Candida albicans in vitro.  相似文献

Cysteine synthase [O-Acetyl-L-serine acetate-lyase (adding hydrogen-sulfide) EC 4.2.99.8] has been highly purified from the extract of rape, Brassica chinensis var. Komatsuna. The purified preparation appeared to be homogeneous on Sephadex G-100 gel filtration and dodecylsulfate-polyacrylamide gel electrophoresis, showing a molecular weight of about 62,000. The latter method also suggested that this enzyme was composed of two identical subunits. The enzyme contained 2 moles of pyridoxal phosphate per mole of enzyme.  相似文献

Photosynthetic and respiratory activities have been measured in leaves of Hordeum vulgare L. var. Manchuria (barley) after infection with Erysiphe graminis var. hordei (powdery mildew). Two isogenic lines, one resistant to infection and the other highly susceptible, were examined.

These isogenic lines showed very different physiological responses following infection. Photosynthesis and the chlorophyll content of resistant leaves was unaffected by infection. Respiration increased slightly and this was accompanied by small increases in activities of enzymes of glycolysis, the pentose-P pathway and the tricarboxylic acid cycle.

The infection of susceptible leaves resulted in a slight increase in photosynthesis 48 hours after inoculation, but subsequently there was a progressive decrease in the photosynthesis of these leaves compared with that of noninfected leaves. The capacity of infected leaves for partial reactions of photosynthesis such as the Hill reaction and the photoreduction of nicotinamide adenine dinucleotide phosphate (NADP1) decreased during the later stages of infection. The levels of chlorophyll, NADPH-diaphorase and aldolase also declined. There was no detectable difference in the respiration of infected and noninfected leaves until 48 hours after inoculation. After this time, the infected leaves showed a higher respiration, the maximum difference occurring about 144 hours after inoculation. The respiratory increase was not accompanied by significant changes in the levels of enzymes of glycolysis and the tricarboxylic acid cycle with the exception of malate dehydrogenase which was lower in infected leaves. In contrast, the activities of glucose-6-P dehydrogenase and 6-P-gluconate dehydrogenase showed changes similar to that observed for respiration.

The respiration and the activities of glucose-6-P dehydrogenase and 6-P-gluconate dehydrogenase did not increase in infected leaves of etiolated plants, even when excellent growth of the fungus was established by growing the plants in White's basal medium supplemented with sucrose. The respiration of a susceptible mutant barley (the yellow-green virescent mutant of the variety Himalaya) when grown in the light at 11° was not changed by infection although the characteristic respiratory rise occurred in plants grown at 15°. At the lower temperature chloroplasts fail to develop in this mutant, although development is normal at 15°.

It is suggested that the pathogen is not directly responsible for the increase in respiration in green leaves, rather that this is a response in the host cells to a loss of photosynthetic capacity.

Potamogeton crispus L. grows as a winter producing annual in the shallow lakes of the Pongolo Floodplain, South Africa. Colonization of leaves by algal and bacterial epiphytes, as seen by scanning electron microscopy, followed the established pattern of increasing diversity and density with leaf age. It was apparent from the micrographs that the primary and subsequent colonizers were present even after death of the host leaf. Cross sections of leaves, viewed by transmission electron microscopy, illustrated that bacterial attachment did not damage the surface of young leaves. There was, however, extensive inward swelling and disorganization of the epidermal walls, characteristic of a reaction to invasion by pathogens. In older leaves the swelling was also present in mesophyll cells, while bacteria had invaded and degraded the epidermal cell wall. The bacterial invasion was concomitant with signs of senescence, and in dead leaves the organisms had penetrated and degraded the epidermis and mesophyll cell walls. The epiphyton/ host relationship may therefore be considered necrotrophic with important consequences for the transfer of energy from producers to consumers during decomposition.  相似文献

The topical anti-inflammatory properties of Bauhinia tarapotensis Benth. (Leguminosae) leaves have been studied by the inhibition of the croton oil-induced ear edema in mice. A bioassay-guided fractionation showed an interesting anti-inflammatory activity of the chloroform extract, that justifies the activity of the whole herbal drug. The main anti-inflammatory principles of B. tarapotensis leaves are triterpenic acids of ursane and oleanane series. The antiphlogistic activity of mixtures constituted of two ursane and oleanane isomers with different hydroxylation pattern, in the ratio 2:1, is comparable to that of indomethacin (ID50 ranging from 95 to 147 microg/cm2 and 93 microg/cm2, respectively).  相似文献
