Studies were conducted to determine if in vivo exposure to dinitrotoluenes (DNT), which is associated with circulatory disorders of atherosclerotic etiology in humans, is associated with alterations of vascular smooth muscle cells (SMC) consistent with the atherogenic process. Sprague-Dawley rats (150-180 g) were injected IP for 5 days/week for 8 weeks with 2,4- or 2,6-DNT (0.5, 5, or 10 mg/kg) or medium chain triglyceride (MCT) oil. Histopathologic evaluation of aortae from animals exposed to either isomer showed dysplasia and rearrangement of SMC at all doses tested. Reduced 3H-thymidine incorporation was observed in primary cultures of aortic SMC from DNT-exposed animals relative to vehicle controls. This inhibitory response was maintained for up to two passages in culture after which a significant increase in thymidine incorporation was observed. Exposure of SMC from naive animals to DNT in vitro (1–100 µM) did not alter the extent of thymidine incorporation in cycling or growth-arrested cultures. In contrast, exposure to 2,4- or 2,6-diaminotoluene (DAT) (1–100 µM), carcinogens which share toxic metabolic intermediates in common with DNT, inhibited replicative DNA synthesis and stimulated unscheduled DNA synthesis in cycling and growth-arrested cultures of SMC, respectively. Cadmium-2-acetylaminofluorene interaction in isolated rat hepatocytes   总被引:1,自引:0,他引:1  
Cadmium (Cd) is a non-essential, highly toxic heavy metal and a ubiquitous environmental contaminant. Evidence exists that Cd can affect parameters which are of great importance in the response towards xenobiotics. However, there is a lack of information about the mechanisms that take place at the cellular and molecular levels upon dual exposure to Cd and other toxins. The purpose of the present work was therefore to examine the biochemical interactions between Cd and a well-known genotoxic hepatocarcinogen, 2-acetylaminofluorene (AAF) in isolated rat hepatocytes. The cells were incubated for 10 hr with a sub-cytotoxic concentration (0.22 M) of 109Cd. This was followed by a 10 hr exposure to 1 M [3H]AAF. Cellular distribution of Cd and 3H was determined. Sephadex G-75 elution profiles of the cytosol showed that Cd was almost entirely associated with the intermediate molecular weight (IMW) fractions containing metallothionein (MT) (>80%), and with high molecular weight proteins. In parallel, the highest proportion of 3H was found in the low molecular weight components. Further analysis of IMW fractions by DEAE A-25 anion-exchange chromatography revealed that, in addition to Cd, there was some 3H which coeluted along with MT-I and MT-II isoforms, but preferentially with MT-I. Moreover, Cd pretreatment caused a 1.6-fold increase in MT level, as measured by the silver-saturation assay. Under these conditions, there was a 17% lower binding of 3H to the DNA. This reduced binding was neither accompanied by diminished AAF uptake nor by inhibition of cytochrome P-450 activity. Taken together, these results suggest that Cd exposure has a protective effect against the genotoxicity of AAF. Zinc gets into the brain parenchyma across the blood-brain and the blood-cerebrospinal fluid barriers, while cadmium hardly gets into the brain parenchyma. Because histidine may be involved in zinc transport across the brain barrier systems, the binding to histidine was compared between zinc and cadmium to understand the difference in brain permeability to both metals. Sephadex G-10 gel filtration indicated that 109Cd, unlike 65Zn, does not bind to histidine. When the plasma incubated with 65Zn or 109Cd was dialyzed in physiological saline containing histidine (0-10 mM), 65Zn concentration in the dialysate was increased with the increase of the histidine concentration, suggesting the transfer of zinc from plasma proteins to histidine. The low affinity of zinc to plasma proteins may be important for brain permeability to this metal. On the other hand, 109Cd was not detected in the dialysate in the presence of 0.1 mM histidine, which is equal to the concentration in the plasma, suggesting no transfer of cadmium from plasma proteins to histidine. These results suggest that the avid binding of cadmium to plasma proteins is related to brain impermeability to this metal.  相似文献

The repair activity of a human transformed cell line, RSa, which was found to be highly sensitive to the lethal effects of 254 nm far-ultraviolet radiation, was compared with that of HeLa cells by evaluating the range of UV-induced incorporation of [methyl-3H]thymidine ([3H]dThd) or 5-[6-3H]bromodeoxyuridine ([3H]BrdUrd) into deoxyribonucleic acid. Direct scintillation counting was used for measuring the extent of unscheduled DNA synthesis (UDS) in UV-irradiated cells, which were treated with hydroxyurea or with arginine deprivation. More quantitative measurements were made by using the density labeling and equilibrium centrifugation method for assaying repair replication. All the amounts of UDS and repair replication in RSa cells were markedly below those in HeLa cells. The possible relationships of the low repair activity to abnormally high UV sensitivity in RSa cells are discussed.  相似文献

The incorporation of [3H]thymidine into DNA due to unscheduled DNA synthesis (UDS) induced by N-OH-2-acetylaminofluorene (N-OH-AAF), aflatoxin B1 (AFB1), ethyl methanesulfonate (EMS) and ultra-violet light was quantitated by autoradiography and by scintillation spectrometry on acid precipitable macromolecules or DNA insolated by isopycnic banding in cesium chloride (CsCl). Dose-dependent increases in UDS due to N-OH-AAF and AFB1 treatment were found. Only 2-fold increases at the highest dose levels were found, however, when incorporated [3H]thymidine was quantitated by scintillation spectrometry. Seven, 11, and 25-fold increases in UDS induced by AFB1, N-OH-AAF and ultra-violet light, respectively, were found when incorporated [3H]thymidine was quantitated by autoradiography, indicating a high sensitivity for detecting ‘long patch’ repair by this technique. Scintillation spectrometry was completely ineffective in detecting EMS-induced UDS, whereas autoradiography demonstrated a small, but significant induction in [3H]thymidine incorporation at high dose levels. The non-proliferative nature of the primary hepatocyte prohibits the uniform radioactive prelabeling of DNA, necessary in other techniques, for the detection of ‘short patch’ repair induced by compounds such as EMS. Therefore, the sensitivity of the primary cultured rat hepatocyte in conjunction with UDS for detecting DNA damage caused by mutagens and carcinogens which induce ‘short patch’ repair may be limited to the autoradiographic analysis of the unscheduled incorporation of [3H]thymidine.  相似文献

Addition of zinc lowers the toxicity level of cadmium in cultured CHO cells. Cell survival and protein synthesis were used to measure the cellular toxicity of cadmium.109Cd was used to measure cadmium uptake by the cells. The results suggest that these class IIB transition metals, zinc and cadmium, share a common transport mechanism. Thus, the antagonism appears to involve a reduction in the influx of cadmium due to the presence of zinc.  相似文献

We have examined the relative sensitivity of Aedes albopictus C7-10 mosquito cells to irradiation with ultraviolet light from a germicidal lamp. On the basis of plating efficiency, C7-10 cells were approximately two times more resistant to UV light than human 293 leukemia cells. Recovery after UV irradiation was accompanied by an increase in unscheduled DNA synthesis (UDS), which was measured by incorporation of 3H-thymidine into acid-precipitable DNA in the presence of hydroxyurea. Under standardized conditions, UDS was maximal after a 10 min exposure (120 J/m2), and declined after longer exposures. In addition, UV treatment is associated with a small but reproducible increase in repair of plasmid DNA in transiently transfected cells. We anticipate that analysis of DNA repair activities in mosquito cells will identify molecular targets that might control longevity in transgenic mosquitoes.  相似文献

The biological activity of natural and synthetic mineral fibers has been examined. Natural attapulgite [(Mg, Al)2Si4O10(OH).4H20], synthetic xonotlite [Ca3Si3O8(OH)2] and natural sepiolite [Mg2Si3O8.2H2O] were selected. Genotoxic effects were investigated by means of a well established cellular model based upon the measurement of unscheduled DNA synthesis (UDS) in rat hepatocytes in primary culture. The intrinsic capacity of the fibers (1 and 10 µ/ml) to induce UDS was first tested. None of the fiber types showed detectable UDS-eliciting activity. Also, the possible modulation of the cellular response to genotoxic agents by the materials was examined by exposing the cells to mixtures of 2-acetylaminofluorene (AAF) (0.05 and 0.25 µg/ml) and fibers (1 and 10 µg/ml). In these experiments, the UDS response was significantly diminished in the presence of xonotlite. This phenomenon may reflect changes in the uptake and/or metabolism of AAF or may result from an inhibition of DNA repair processes, the latter suggesting a possible cocarcinogenic potential for this synthetic silicate. The effect of regucalcin, isolated from rat liver cytosol, on neutral proteolytic activity in the hepatic cytosol was investigated. The Ca2+-requiring proteinase required 5–10 µM Ca2+ for maximal activity in the presence of a protein substrate (globin). The proteinase activity was markedly elevated by the addition of regucalcin (0.25–2.0 µM) in the absence or presence of Ca2+ (5.0 µM) added. The effect of regucalcin, however, was the greater in the absence of Ca2+ than that in the presence. The pronounced effect of regucalcin on the proteinase activity was also seen in the presence of 1.0 mM EGTA with or without Ca2+ (5.0 µM). In the absence of Ca2+, the regucalcin-increased proteinase activity was clearly inhibited by the presence of anti-regucalcin antiserum (diluted to 240-fold), leupeptin (20 and 200 µg/ml), and heavy metals (25 µM cadmium or 25 µM zinc), although the inhibition was not complete at the concentration used. The present findings suggest that regucalcin increases proteolytic activity in rat liver cytosol, and that regucalcin may activate Ca2+-independent neutral cysteinyl-proteinase.  相似文献

In primary culture of normal adult rat hepatocytes, human serum heated at 56°C for 30 min stimulated dose-dependently [3H]thymidine incorporation into trichloroacetic acid insoluble fraction of the cells, most of which was solubilized into hot trichloroacetic acid solution. The solubilized fraction was reduced when hydroxyurea was added to the culture. The heated serum also increased dose-dependently protein synthesis and cell viability determined from morphological findings. These results suggest that human serum has heat-stable factors stimulating DNA synthesis and maintaining cell viability of cultured rat hepatocytes.  相似文献

Speciation of carrier-free109Cd, added in cationic form to prefiltered extracts obtained by leaching forest soil samples with distilled water, was analyzed using electrophoresis, dialysis, centrifugation, and ultrafiltration. Rapid establishment of isotopic equilibrium between the added109Cd and stable cadmium present in the extracts was observed. All the data obtained indicated that109Cd and also stable cadmium were present in the analyzed extracts in the form of neutral or negatively charged organic complexes or small colloids. The results of electrophoresis enabled the characterization, at least semiquantitative, of the abundance and electrophoretic mobility of the forms present. The incomplete dialysis of109Cd from the soil extracts through cellophane membrane against water proved the presence of organic associates with molecular weights higher than 104. Dialysis against the same, but unlabeled extract was always complete, indicating the reversible (labile) nature of the organic forms of cadmium. Assessment of the stability constants of the organic forms using a simple discrete two-site model suggested that humate and/or fulvate complexes of cadmium were formed.  相似文献

Soybean (Glycine max L.) plants exposed to 109Cd readily absorb the element. Differential centrifugation of leaf, stem, and root homogenates followed by radioassay showed that Cd was associated primarily with the 105,000g supernatant. Separation of this fraction by gel chromatography and subsequent analysis by radioassays revealed that 109Cd was bound to macromolecules of >50,000, 13,800, and 2,280 molecular weights. The >50,000 and 2,280 molecular weight fractions probably are nonspecific binding of Cd to normal cell components. The 13,800 molecular weight 109Cd-bound component was found to be inducible by cadmium. It had a high ultraviolet absorbance at 254 nm and a low absorbance at 280 nm at pH 8.6.  相似文献

In this study, a hydroponics experiment was conducted to investigate the characteristics of Cd tolerance and accumulation of Elsholtzia argyi natively growing on the soil with high levels of heavy metals in a Zn/Pb mining site. Seedlings of E. argyi grown for 4 weeks and then were treated with 0(CK), 5, 10, 15, 20, 25, 30, 40, 50, 100 μM Cd for 21days. Each treatment had three replications. No visual toxic symptoms on shoots of E. argyi were observed at Cd level ≤50 μM. The results indicated that the dry biomass of each tissue and the whole plants of the treatments with ≤40 μM cadmium were similar to that of the control, implying that E. argyi was a cadmium tolerant plant. The results also showed that the shoot Cd concentration significantly (P < 0.05) increased with the increase in the Cd level in nutrient solution. The shoot Cd concentration of the treatment with 40 μM Cd was as high as 237.9 mg kg–1, which was higher than 100 mg kg–1, normally used as the threshold concentration for identifying the Cd hyperaccumulating plant. It could be concluded that E. argyi was a Cd tolerant and accumulating plant species.  相似文献

A field growth chamber study was conducted to determine the effects of ozone and simulated acid rain (SAR) on soil heavy metals. Loblolly pine (Pinus taeda L.), grown in open-top chambers, was exposed to three concentrations of ozone (charcoal filtered air with 0.026 µL O3 L-1, and two non-filtered treatments in which ozone concentrations were 0.074 µL L-1 and 0.147 µL L-1, respectively) and two levels of SAR (pH 3.5 and 5.2). Ozone was applied for 12 h d-1 for 9 months and acid rain deposition was 125 mm event-1. After 9 months exposure, soil pH, organic matter and DTPA-extractable heavy metals (Cd, Pb, Zn, Mn, Fe, Cu) were determined on soil samples collected from exposed chambers at two depths (0–15 cm and 15–30 cm). Simulated acid rain decreased the original soil pH. The concentrations of Cd, Pb and Mn at SAR pH 3.5 were significantly higher than at SAR pH 5.2. Ozone did not affect Zn, Fe and Cu, but a significant interaction between pH and O3 on Mn, Pb and Cd was observed. Due to the poor drainage capacity of this soil, leaching of heavy metals was not observed.  相似文献

Summary Rice plants are grown in nutrient solutions (Hoagland-Arnon 1) containing different concentrations of cadmium (0.005; 0.010 and 0.100 ppm) and one concentration of zinc (0.070 ppm) traced respectively with109Cd and65Zn. Distribution and localization of both elements are studied at plant and cellular levels. Contrary to zinc, cadmium remains fixed by radicular sites and its translocation index is very low. Centrifugation of roots homogenates showed that % Cd recovered in the residue increases with Cd concentration and emphasize the high capacity of Cd fixation by walls when Cd external concentration is high. The relative distributions of Cd and Zn present at the same concentration in the growing medium are quite identical. In the 100,000×g supernatant fraction of roots, both elements are associated with compounds of low molecular weight, Cd being bound to higher MW ligands than zinc (1000 to 3000 for zinc). The fact that Cd was recovered in fractions in which most of35S-cysteine was present suggests that in plants, like in animals, peptides rich in — SH residues may play a role in the fixation of heavy metals. The effects of Ca and La on Cd transport across apical and basolateral gill surfaces were examined in isolated perfused Carcinus gills. In the gills exposed to diluted sea water enriched with calcium by addition of CaCl2 to 11.9 and 15.9 mmol·l-1 the Cd influxes decreased by 55% and 85%, respectively, compared to those observed in diluted sea water (6.9 mmol Ca·l-1). When Ca was added to the perfusate to increase its concentration at the basolaterally oriented gill surfaces, Cd influx inhibition was less pronounced than when Ca was added apically. The effect of Ca on 109Cd influxes appears, therefore, to be exerted at the apical gill surface. Additionally, the effects of various concentrations of the non-specific Ca channel blocker La (added to diluted sea water) on 109Cd influxes in isolated perfused Carcinus gills were studied. 109Cd influxes are shown to be lanthanum concentration dependent. The half-maximum inhibition of cadmium influxes by La occurred at 1.4·10-6 mol·l-1. The present report demonstrates differential DNA-repair activity among 14 strains of immature (20 ± 2 days old) male mice (inbred strains: C57BL/6J, RF/J, Nude homo/nu, RIII/2J, Pl/J, AKR/J, Nude hetro/nude, C3H/HeJ, SWR/J, SM/J, ST/J, LP/J, BALB/cJ and random-bred strain: CD-1). The prespermiogenic cells were isolated and enriched by collagenase-trypsin digestion of seminiferous tubules and subsequent 3% albumin-gradient centrifugation. Enriched prespermiogenic cells demonstrated a viabiilty greater than 95% by trypan blue exclusion criteria. For in vitro unscheduled DNA synthesis (UDS) determination, prespermiogenic cells (106 cells/ml) were incubated with methyl methanesulfonate (0.4 mM) in the presence of 20 mM hydroxyurea (HU). At 20 mM HU concentration, 90% of S-phase DNA activity in prespermiogenic cells was inhibited and thus, the net UDS activity following MMS exposure was readily determined. MMS-induced UDS activity in the CD-1 mouse strain was both linear up to 4 h of incubation and dose-dependent at 4 h incubation. The apparent Km for MMS-induced UDS activity in prespermiogenic cells was approx. 1.8 × 10?4 M. Of the 14 mice strains tested, C57BL/6J and RF/J exhibited the highest DNA-repair activity, while BALB/cJ, LP/J, and ST/J showed the lowest. A maximal difference in UDS activity fo 3.5-fold was observed between C57BL/6J and BALB/cJ. Furthermore, a 2.5-fold difference was also noted between RF/J and LP/J mouse strains. Thus, wide variations in DNA-repair activity among 14 mouse strans were clearly demonstrated. Whether genetically select mouse strains with the lowest DNA-repair activity should have greater sensitivity toward environmental mutagens needs to be tested.  相似文献

Genotypic variation in uptake and distribution of cadmium (Cd) was studied in 19 inbred lines of maize (Zea mays L.). The inbred lines were grown for 27 days on an in situ Cd-contaminated sandy soil or for 20 days on nutrient solution culture with 10 µg Cd L-1. The Cd concentrations in the shoots showed large genotypic variation, ranging from 0.9 to 9.9 µg g-1 dry wt. for the Cd-contaminated soil and from 2.5 to 56.9 µg g-1 dry wt. for the nutrient solution culture. The inbred lines showed a similar ranking for the Cd concentrations in the shoots for both growth media (r2=0.89). Two main groups of inbreds were distinguished: a group with low shoot, but high root Cd concentrations (shoot: 7.4±5.3 µg g-1 dry wt.; root: 206.0±71.2 µg g-1 dry wt.; shoot Cd excluder) and a group with similar shoot and root Cd concentrations (shoot: 54.2±3.4 µg g-1 dry wt.; root: 75.6±11.2 µg g-1 dry wt.; non-shoot Cd excluder). The classification of the maize inbred lines and the near equal whole-plant Cd uptake between the two groups demonstrates that internal distribution rather than uptake is causing the genotypic differences in shoot Cd concentration of maize inbred lines. Zinc (Zn), a micronutrient chemically related to Cd, showed an almost similar distribution pattern for all maize inbred lines. The discrepancy in the internal distribution between Cd and Zn emphasizes the specificity of the Cd distribution in maize inbred lines.  相似文献

Phytolacca americana L. can accumulate large amounts of heavy metals in its aerial tissues, especially cadmium (Cd) and manganese (Mn). It has great potential for use in phytoextraction of metals from multi-metal-contaminated soils. This study was conducted to further investigate the Cd- and Mn-tolerance strategies of this plant. Concentrations of non-protein thiols (NPTs) and phytochelatins (PCs) in leaves and roots increased significantly as the concentration of Cd in solution increased. The molar ratios of PCs:soluble Cd ranged from 1.8 to 3.6 in roots and 8.1 to 31.6 in leaves, suggesting that the cellular response involving PC synthesis was sufficient to complex Cd ions in the cytosol, especially that of leaves. In contrast, excess Mn treatments did not result in a significant increase in NPT or PC concentrations in leaves or roots. Oxalic acid concentrations in leaves of plants exposed to 2 or 20 mM Mn reached 69.4 to 89.3 mg (0.771 to 0.992 mmol) g–1 dry weight, respectively, which was approximately 3.7- to 8.6-fold higher than the Mn level in the 0.6 M HCl extract. Thus, oxalic acid may play an important role in the detoxification of Mn.  相似文献

Cd++ inhibits EGF-induced 3H-thymidine incorporation in serum deprived NRK-49F cells in a dose dependent pattern. The underlying mechanisms for this inhibition are largely unknown. EGF-induced myc mRNA accumulation in NRK-49F cells and the effects of Cd++ on this response were examined under conditions that result in partial or complete inhibition of EGF-induced DNA synthesis. It was found that doses of Cd++ that inhibit EGF-induced DNA synthesis do not inhibit EGF-induced protein synthesis and myc mRNA accumulation. Cd++ doses of 0.5 µM and 1 µM were found actually to increase EGF-induced myc mRNA accumulation and amino acid incorporation. These results show that the effect of Cd++ on EGF-induced DNA synthesis is not due to inhibition of entrance into G1, but rather that Cd++ acts on events subsequent to myc accumulation; that is, events associated with either G1 progression, entry into S or DNA synthesis.Abbreviations EGF Epidermal Growth Factor - 3HTdr Tritium thymidine - MeAIB Methylaminoisobutyrate  相似文献
