本文探讨胶原蛋白肽对高脂日粮小鼠抗氧化能力及血脂代谢的影响。40只小鼠随机分为5组(n=8):分别饲喂正常日粮,高脂日粮,添加0.5%和1%胶原蛋白肽及0.1%硫辛酸的高脂日粮。42 d后测定小鼠全血(血清)和组织活性氧(Reactive oxygen species,ROS)水平、丙二醛(Malondialdehyde,MDA)含量、抗氧化酶活力及血脂水平。结果表明:0.5%和1%胶原蛋白肽都可显著降低机体ROS水平及MDA含量(P<0.05),增强抗氧化能力(P<0.05),改善血脂水平(P<0.05)。1%胶原蛋白肽能使机体抗氧化能力及血脂水平恢复到正常水平,0.5%的添加量效果不显著。适量的胶原蛋白肽可有效提高机体抗氧化能力,缓解高脂膳食造成的氧化应激,改善血脂代谢。  相似文献

目的:氧化应激和炎症反应是NASH进展的关键因素,同时二者之间存在着密切关系,而转录因子Nrf2和NF-kB分别是氧化应激和炎症信号通路的关键调控靶点,因此,研究Nrf2对高脂饮食诱导小鼠肝脏NF-kB信号通路的影响,对探讨NASH进展具有重要的意义。方法:雄性野生型(WT)和Nrf2基因敲除(Nrf2-/-)ICR小鼠各10只,随机分为WT对照组(Control)、Nrf2-/-对照组(KO)、WT高脂饮食组(HFD)和Nrf2-/-高脂饮食组(KOHFD)(n=5)。喂养8周后,观察肝脏光镜下改变,检测肝脏GSH、MDA、TNFα和IL-6水平。Western-Blot检测肝脏NF-kB蛋白表达水平,观察敲除Nrf2对肝脏NF-kB活性作用的影响。结果:1.光镜下观察,Control组与KO组小鼠肝脏结构无明显变化,HFD组小鼠肝脏呈现大片脂肪沉积和炎症细胞浸润,KOHFD组小鼠肝脏则呈现明显的大泡性变性,且炎症细胞浸润较HFD组明显加重;2.与Control组相比,KO组小鼠肝脏MDA轻度升高,GSH轻度降低,但无明显差异,而HFD组和KOHFD组小鼠肝脏MDA显著升高(P〈0.05),GSH显著降低(P〈0.05),且KOHFD组MDA明显高于HFD组(P〈0.05),GSH明显低于HFD组(P〈0.05)。3.ELISA结果显示,与Control组相比,KO组小鼠肝脏TNFα和IL-6分泌轻度增加,而HFD组和KOHFD组小鼠肝脏TNFα与IL-6水平显著升高(P〈0.05),且KOHFD组小鼠肝脏TNFα与IL-6显著高于HFD组(P〈0.05);4.Western-Blot结果显示,Control组和KO组之间无明显差异,而KOHFD组和HFD组小鼠肝脏胞核NF-kB蛋白表达水平显著升高,且KOHFD组高于HFD组。结论:敲除Nrf2可以显著加重高脂饮食诱导的小鼠肝脏氧化应激水平,进而促进NF-kB的活化,从而为通过以Nrf2为靶点治疗NASH提供重要的实验依据。  相似文献

为考察铁皮石斛多糖对高脂饮食小鼠肠黏膜屏障的影响,采用水提醇沉法提取铁皮石斛多糖,联合高脂饲料给予小鼠8周后观察肠黏膜结构及肠黏膜菌群的变化。结果显示高脂饮食显著破坏了肠黏膜结构,表现为肠黏膜萎缩,上皮细胞脱落并伴有炎性渗出,Corynebacterium_1及Staphylococcus等与感染及炎症相关的菌属大量增殖。铁皮石斛多糖对肠黏膜结构有较好的保护作用,并可减少Corynebacterium_1的丰度,同时提高肠黏膜共生菌Candidatus_Arthromitus的丰度,促进了Muribaculaceae、Bacteroides、Lachnospiraceae_NK4A136_group等碳水化合物代谢、短链脂肪酸产生相关菌的增殖。研究表明铁皮石斛多糖对肠黏膜屏障的保护作用或与其维持肠黏膜结构完整,调节肠黏膜菌群组成及促进碳水化合物代谢,生成短链脂肪酸有关。  相似文献

观察高脂饮食联合链脲佐菌素(STZ)对小鼠糖脂代谢及胰岛功能损伤的影响。将体质量为18~20 g的SPF级雄性C57BL/6J小鼠40只,随机分为对照组和模型组,每组20只。对照组小鼠饲喂对照饲料,模型组小鼠饲喂高脂饲料。1周后,禁食16 h,测量小鼠体质量与空腹血糖;尾静脉采血,分离血清;每周1次,连续4周。4周后,对照组小鼠腹腔注射柠檬酸缓冲液,模型组小鼠腹腔注射STZ,剂量为40 mg·kg~(-1),每天1次,连续3 d。继续饲养2周后,测量随机血糖,以小鼠血糖≥16.7 mmol·L~(-1)者即判定为2型糖尿病。对照组及2型糖尿病小鼠经腹腔注射葡萄糖以进行糖耐量试验。与对照组同一时间的体质量和血糖进行比较,模型组小鼠体质量、血糖均升高,除第1周血糖外,差别均有统计学意义(P0.05)。采用重复测量方差分析,发现喂养时间对体质量(F=200.831)和血糖(F=7.025)均有影响,差异有统计学意义(P0.05);不同喂养时间对低密度脂蛋白(F=30.793)、甘油三酯(F=34.027)和高密度脂蛋白(F=30.793)均有影响,差异有统计学意义(P0.05)。不同饲料喂养与不同喂养时间对体质量和甘油三酯存在交互作用(P0.05)。比较糖耐量曲线发现,成模小鼠较对照组小鼠糖耐量降低。采用ELISA试剂盒检测小鼠血清中TNF-α含量,成模小鼠的TNF-α含量高于对照组,差异有统计学意义(P0.05)。高脂饮食可导致C57BL/6J小鼠糖脂代谢紊乱,给予STZ后引起了小鼠糖耐量降低及炎症损伤,高脂饮食联合STZ可提高小鼠2型糖尿病模型的成模率。  相似文献

本研究旨在探索白藜芦醇(RSV)对不同程度肥胖小鼠脂肪氧化应激状态和血脂的影响。高脂日粮(HFD)处理12周的昆明小鼠分为3类:肥胖抵抗(DIO-R)、中体重(Med)和肥胖(DIO),分别饲喂HFD、HFD+0.3 g/kg RSV和HFD+0.6 g/kg RSV日粮18周,并以正常日粮小鼠为对照。结果表明,0.6 g/kg RSV处理可显著降低DIO小鼠体重、腹脂率,显著提高脂肪组织抗氧化能力,改善血脂。0.3 g/kg RSV处理对DIO-R小鼠也有类似趋势,但0.6 g/kg RSV处理引起DIO-R小鼠脂肪组织抗氧化能力下降、血脂紊乱。总之,RSV在不同程度肥胖小鼠具有剂量特异性的氧化应激调控作用。  相似文献

The effect of a freshwater clam (Corbicula fluminea) extract (FCE) on cholesterol metabolism in rats fed on a high-cholesterol diet was investigated. When rats were fed various amounts of FCE in addition to the high-cholesterol diet for 2 wk, the serum and hepatic cholesterol levels were gradually reduced in a dose-dependent manner, as compared with the control group. The excretion of neutral sterols and bile acids into the feces was increased by feeding FCE. Several phytosterols were detected in the feces of rats fed on the FCE-containing diet. In addition, substantial amounts of phytosterols were found in FCE. Cholesterol 7α-hydroxylase (CYP7A1) mRNA in the liver of the rats fed on the FCE-containing diets was higher than that of rats fed on the high-cholesterol diets without FCE. These results may suggest that enhanced cholesterol degradation and the excretion of neutral sterols and bile acids contributed to the hypocholesterolemic effect of FCE observed in the hypercholesterolemic rats fed on the high-cholesterol diet.  相似文献

Feeding of a threonine-deficient diet to rats weighing approximately 53 g or 99 g caused a significant rise in liver lipids compared to the control diet containing 7% amino acid mixture. Whereas, when rats weighing approximately 155 g were fed either the control diet or the threonine-deficient diet, liver lipid content was essentially the same for both groups. Therefore, in the present paper, young rats were used to clarify the mechanism of liver lipid accumulation in threonine-deficiency. The increase in dietary fat content of the threonine-deficient diet did not prevent the lipid accumulation in rat liver. The rates of in vivo incorporation from radioactive acetate into liver lipids, body lipids and respiratory CO2 of rats fed either the control diet or the threonine-deficient diet were measured. The threonine-deficient group tended to be lower in total activity of both the liver lipids and body lipids than those of the control group. Thus, these results suggest that the development of this type of fatty liver might not be due to the stimulation of lipid synthesis in the liver. In the serum of rats fed the threonine-deficient diet, the protein content of β-lipoproteins was significantly lower and free fatty acid level tended to be lower than the values of the control animals, respectively. From these results, decreased trasport of lipids from the liver may thus be considered a potential major factor responsible for the excessive lipid accumulation in the liver of rats fed the threonine-deficient diet.  相似文献

We investigated the effects of various dietary fibers or their likenesses on the apparent fat digestibility by rats fed on a high-fat diet. Each of 23 different fibers was added at 5% (w/w) to a purified diet containing 20% (w/w) corn oil. The rats were fed these diets for 2 weeks, and the feces were collected from each animal during the last 3 days. When compared with cellulose (control), 10 of the tested fibers significantly increased the fecal lipid excretion. Among these fibers, chitosan markedly increased the fecal lipid excretion and reduced the apparent fat digestibility to about a half relative to the control. The apparent protein digestibility was not greatly affected by chitosan. The fatty acid composition of the fecal lipids closely reflected that of the dietary fat. These results suggest that chitosan has potency for interfering with fat digestion and absorption in the intestinal tract, and for facilitating the excretion of dietary fat into the feces.  相似文献

Direct gas chromatographic determination of tobacco smoke was developed. Tobacco smoke condensate was collected on a glass fiber filter and the components were converted into their trimethylsilyl derivatives and then subjected to glass capillary column gas chromatography. By this method, all tobacco smoke components, including unstable phenolic substances and water-soluble polyhydroxy compounds, were simultaneously determined. Compositional differences between lamina and midrib smoke of flue-cured tobacco leaves were also clarified by the method. The results indicate that there are quantitative differences in nicotine, phenols, levoglucosan, quinic acid γ-lactone and the other major components between lamina and midrib smoke.  相似文献

The mechanisms underlying the progression of simple steatosis to steatohepatitis are yet to be elucidated. To identify the proteins involved in the development of liver tissue inflammation, we performed comparative proteomic analysis of non-alcoholic steatohepatitis (NASH). Mice fed a methionine and choline deficient diet (MCD) developed hepatic steatosis characterized by increased free fatty acid (FFA) and triglyceride levels as well as alpha-SMA. Two-dimensional proteomic analysis revealed that the change from the normal diet to the MCD diet affected the expressions of 50 proteins. The most-pronounced changes were observed in the expression of proteins involved in Met metabolism and oxidative stress, most of which were significantly downregulated in NASH model animals. Peroxiredoxin (Prx) is the most interesting among the modulated proteins identified in this study. In particular, cross-regulated Prx1 and Prx6 are likely to participate in cellular defense against the development of hepatitis. Thus, these Prx isoforms may be a useful new marker for early stage steatohepatitis. Moreover, curcumin treatment results in alleviation of the severity of hepatic inflammation in steatohepatitis. Notably, curcumin administration in MCD-fed mice dramatically reduced CYP2E1 as well as Prx1 expression, while upregulating Prx6 expression. These findings suggest that curcumin may have a protective role against MCD fed-induced oxidative stress.  相似文献

l-Methionine γ-lyase (EC 4.4.1.11) catalyzes α,β-elimination of l-2-amino-3-(N-methylamino)propionic acid and l-2-amino-3-(N-hydroxyethylamino)propionic acid to yield pyruvate, ammonia, and the corresponding amines. These amino acids also undergo the enzymatic β-replacement reaction with thiols to produce the corresponding S-substituted cysteines. Thus, l-methionine γ-lyase cleaves a C-N bond in addition to C-S, C-Se, and C-O bonds at the β position of amino acids by elimination and replacement reactions. A linear relationship between the reactivity, (log(Vmax/Km) and the pKa value of the conjugated acid of the leaving group has been found for Se-methyl-l-selenocysteine, S-methyl-l-cysteine, and O-methyl-l-serine. However, l-2-amino-3-(N-methylamino)propionic acid has shown lower reactivity than that expected from the pKa value of methylammonium ions.  相似文献

The effects of the addition of individual amino acids on methionine-induced hypercholesterolemia (experiment 1), and the interacting effects of dietary protein level and sulfur-containing amino acids and glycine on plasma cholesterol concentration (experiment 2) were studied in growing rats fed on a high cholesterol diet. In experiment 1, rats were fed on a 25% casein-0.75% methionine (25CM) diet containing 2.5% of individual amino acids for 2 weeks. Methionine-induced hypercholesterolemia was prevented by the concurrent addition of glycine or serine, but the other amino acids tested (alanine, threonine, leucine, phenylalanine, lysine, arginine, and glutamic acid) had no effect. Histidine rather enhanced the hypercholesterolemia. In experiment 2, rats were fed on a 10%, 25%, or 50% casein diet containing 0.75% methionine, 0.60% cystine, 0.63% taurine, 2.5% glycine, or 0.75% methionine +2.5% glycine for 3 weeks. Dietary addition of 0.75% methionine increased the plasma cholesterol concentration for the 25% and 50% casein diets, but it decreased the plasma cholesterol for the 10% casein diet. When the addition level of methionine was doubled in the 10% casein diet, the plasma cholesterol concentration was significantly higher for the 1.5% methionine-added diet than for the 0.75% methionine-added diet. Cystine and taurine lowered plasma cholesterol for all dietary casein levels. Methionine-induced hypercholesterolemia with 25% and 50% casein diets was prevented by the glycine supplementation. These data suggest that sulfur-containing amino acids and glycine are important in plasma cholesterol regulation.  相似文献

The objective of the present study was to investigate the cholesterol-reducing effect of medium-chain fatty acids (MCFAs) completed by elevated excretion of fecal neutral steroids and/or bile acids. Blood and liver lipid profiles, fecal neutral steroids, bile acids, and mRNA and protein expression of the genes relevant to cholesterol homeostasis were measured and analyzed in C57BL/6J mice fed a cholesterol-rich diet with 2% caprylic acid or capric acid for 12 weeks. Blood total cholesterol and low-density lipoprotein cholesterol (LDL-c) levels were reduced significantly as compared to diet with palmitic acid or stearic acid. Caprylic acid promoted the excretion of fecal neutral steroids, especially cholesterol. The excretion of fecal bile acids, mainly in the form of cholic acid was enhanced and accompanied by elevated expression of mRNA and the protein of hepatic cholesterol 7α-hydroxylase (CYP7A1). These results indicate that MCFAs can reduce blood cholesterol by promoting the excretion of fecal cholesterol and cholic acid.  相似文献

本文研究了丹酚酸B(Salvianolic acid B,Sal B)对饲喂高脂日粮小鼠抗氧化及淋巴细胞亚群免疫功能调节的影响。选取4w龄雄性C57BL/6小鼠40只,随机平均分成4组:即正常对照组(正常日粮),高脂日粮组(高脂日粮),Sal B低剂量组(高脂日粮+0.002%Sal B)和Sal B高剂量组(高脂日粮+0.004%Sal B)。八周后测定血脂、全血和脾脏活性氧(reactive oxygen species,ROS)水平、抗氧化酶活性及MDA含量;采用FACSAria流式细胞仪分析小鼠外周血及脾脏淋巴细胞亚群。结果表明:Sal B可显著降低高脂日粮小鼠血脂水平,全血及脾脏ROS水平,提高抗氧化能力,提高CD4+/CD8+T细胞比例及CD19+细胞百分率(P0.05)。低剂量Sal B组抗氧化与免疫调节功能好于高剂量Sal B组。  相似文献

An increasing number of studies indicate that dairy products, including whey protein, alleviate several disorders of the metabolic syndrome. Here, we investigated the effects of whey protein isolate (whey) in mice fed a high-fat diet hypothesising that the metabolic effects of whey would be associated with changes in the gut microbiota composition. Five-week-old male C57BL/6 mice were fed a high-fat diet ad libitum for 14 weeks with the protein source being either whey or casein. Faeces were collected at week 0, 7, and 13 and the fecal microbiota was analysed by denaturing gradient gel electrophoresis analyses of PCR-derived 16S rRNA gene (V3-region) amplicons. At the end of the study, plasma samples were collected and assayed for glucose, insulin and lipids. Whey significantly reduced body weight gain during the first four weeks of the study compared with casein (P<0.001–0.05). Hereafter weight gain was similar resulting in a 15% lower final body weight in the whey group relative to casein (34.0±1.0 g vs. 40.2±1.3 g, P<0.001). Food intake was unaffected by protein source throughout the study period. Fasting insulin was lower in the whey group (P<0.01) and glucose clearance was improved after an oral glucose challenge (P<0.05). Plasma cholesterol was lowered by whey compared to casein (P<0.001). The composition of the fecal microbiota differed between high- and low-fat groups at 13 weeks (P<0.05) whereas no difference was seen between whey and casein. In conclusion, whey initially reduced weight gain in young C57BL/6 mice fed a high-fat diet compared to casein. Although the effect on weight gain ceased, whey alleviated glucose intolerance, improved insulin sensitivity and reduced plasma cholesterol. These findings could not be explained by changes in food intake or gut microbiota composition. Further studies are needed to clarify the mechanisms behind the metabolic effects of whey.  相似文献

We showed previously that UBXD8 plays a key role in proteasomal degradation of lipidated ApoB in hepatocarcinoma cell lines. In the present study, we aimed to investigate the functions of UBXD8 in liver in vivo. For this purpose, hepatocyte-specific UBXD8 knockout (UBXD8-LKO) mice were generated. They were fed with a normal or high-fat diet, and the phenotypes were compared with those of littermate control mice. Hepatocytes obtained from UBXD8-LKO and control mice were analyzed in culture. After 26 wk of a high-fat diet, UBXD8-LKO mice exhibited macrovesicular steatosis in the periportal area and microvesicular steatosis in the perivenular area, whereas control mice exhibited steatosis only in the perivenular area. Furthermore, UBXD8-LKO mice on a high-fat diet had significantly lower concentrations of serum triglyceride and VLDL than control mice. A Triton WR-1339 injection study revealed that VLDL secretion from hepatocytes was reduced in UBXD8-LKO mice. The decrease of ApoB secretion upon UBXD8 depletion was recapitulated in cultured primary hepatocytes. Accumulation of lipidated ApoB in lipid droplets was observed only in UBXD8-null hepatocytes. The results showed that depletion of UBXD8 in hepatocytes suppresses VLDL secretion, and could lead to periportal steatosis when mice are fed a high-fat diet. This is the first demonstration that an abnormality in the intracellular ApoB degradation mechanism can cause steatosis, and provides a useful model for periportal steatosis, which occurs in several human diseases.  相似文献

We investigated the effects of non-pungent pepper powder fermented by Bacillus licheniformis SK1230 on the fat accumulation in mice. Four weeks of feeding a high-fat diet with fermented pepper powder resulted in a significantly decreased hepatic total-lipid level and increased serum HDL-cholesterol, and tended to lower the fat weight. These results suggest that fermented pepper powder inhibited fat accumulation and improved lipid metabolism in mice fed the high-fat diet.  相似文献
