Periphyton colonization of natural rock surfaces and granite tiles was followed experimentally in the Matamek River, an acidic (pH 5.5) sixth order boreal stream in northeast Quebec, Canada. Accumulation of chlorophyll a and freshweight algal biomass was logarithmic over a 25 day colonization period. The major colonizers were Tabellaria flocculosa (Roth) Kütz., T. fenestrata (Lyngb.) Kütz., Mougeotia sp., and Eunotia pectinalis (Kütz) Rabh., and its varieties. The microcolonization sequence on granite tiles, followed over 27 days with scanning electron microscopy, showed an initial accumulation of algal cells on the upstream and downstream margins and in depressions, followed by a gradual filling-in of the flat surfaces. It is hypothesized that the observed slow rate of colonization was due to the high surface tension of the granite substratum and the absence of preconditioning by an organic film. It is further hypothesized that the increase in cellular carbon fixation rates of T. flocculosa measured over a 23 day period using nuclear track autoradiography parallels the development of an algal-detrital microcosm on the granite tile, and is evidence for the establishment of localized tight nutrient spiralling. It is suggested that the periphyton community may be regarded as a nutrient recycling system at a microenvironmental level which may be especially important in oligotrophic systems.  相似文献

Navicula mutica (Kütz.) var. mutica was isolated from the air, cloned on agar, cultured in soil-water bottle, and studied with transmission and scanning electron micros-ropy. The frustules were lanceolate to ovoid with rounded apices, with the apical axis 8.5 ± 3.2 μ and the trans-apical and the transapical axis 3.6 ± 0.6 μm. Striae were composed of two or three puncta, and the mantle bore a single row of puncta aligned with the striae. The ends of the raphe turned away from an isolated punctual in the central area of the valve. The mantle puncta and one or two of the valve-face puncta in each stria opened into a series of transapical grooves in the interior of the valve, the grooves contributing to the appearance of striae in the light microscope. The interior of the mantle also possessed a pair of longitudinal grooves, discontinuous at the apices of the valves. An undulate advalvar margin of the valvocopula likely articulates along the interior longitudinal groove of the mantle. The projections of the undulate margin are perhaps positioned between the transapical grooves and along the longitudinal groove between the dentiform structures formed by the intersection of the double-grooved system. The girdle bands each had two (occasionally three) rows of pores. The pleurae margins were straight and not undulate.  相似文献

Rhoicosphenia Grun. is a relatively isolated genus among the biraphid diatoms. Morphological changes in an isopolar member of the genus, Rh. genuflexa (Kütz.) Medlin, were investigated using light and scanning electron microscopy. The fully raphid valve showed changes in its flexure that could be correlated with size reduction during its life history from the initial cells to the smallest cells found in the population. Bands showed changes in number (from three to one) related to size reduction. Rh. genuflexa is morphologically similar to Rh. abbreviata (C. Ag.) Lange-Bert. (=Rh. curvata (Kütz.) Grun.), although the two are distinct taxa. These observations support previous contentions that Rhoicosphenia is a natural taxonomic grouping.  相似文献

Epilithic and epiphytic diatoms were collected monthly from 18 June 1979 through 19 May 1980 from both shaded and unshaded sites of Sessums Creek, a shallow, oligotrophic and strongly alkaline stream in northeastern Mississippi. The dominant macroalgae were filamentous green algae belonging to the non-mucilage producing genera Cladophora and Rhizoclonium. Ninety taxa were identified with Navicula minima Grun. and Nitzschia dissipata (Kütz.) Grun. accounting for 50% of all valves counted. Other important taxa were Navicula menisculus var. upsaliensis (Grun.) Grun., N. cryptocephala var. veneta (Kütz.) Rabh., Achnanthes minutissima Kütz. and Cymbella turgida (Greg.) Cl. With few exceptions, the more dominant taxa were equally abundant in the shaded and unshaded sites and also in the epilithon and epiphyton. Species diversity (H') and the number of taxa in a sample in all four habitats showed an identical pattern, being lowest in winter and highest in fall. No one pair of habitats was more structurally similar than any-other pair when compared by a similarity index (SIMI). Apparently, differences in light intensity and substrate type had little effect on diatom distribution in Sessums Creek and it was concluded that the locality sampled supported a single, nearly homogeneous diatom community.  相似文献

The attachment of the psammophytic alga Caulerpa mexicana Sond. ex Kütz., a coenocytic green alga, to crushed CaCO3 particles was examined utilizing the scanning electron microscope and fluorescently tagged antivitronectin antibodies. Plants attached to the substrate through morphologically variable tubular rhizoidal extensions that grew from the stolon. In this study, we describe two means of attachment: (i) the rhizoid attachment to limestone gravel by thigmoconstriction, where tubular extensions of the rhizoid wrapped tightly around the substrate and changed morphology to fit tightly into crevices in the limestone, and (ii) through adhesion pads that formed in contact with the limestone granules. Flattened rhizoidal pads were observed to secrete a fibrillar material that contained vitronectin‐like proteins identified through immunolocialization and that facilitated binding of the rhizoid to the substrate.  相似文献

报道了2004年对黑龙江省七星河湿地国家级自然保护区的硅藻门植物研究的结果,共发现155个分类单位,包括113种39变种3变型,分别隶属于2纲6目9科26属,其中中国新记录2种2变种1变型共计5个分类单位,初步分析了七星河湿地硅藻植物群落的种类组成及生态类型,大部分的硅藻属于普生种类,同时出现了一些喜酸种类如:Eunotia pectinalis var. ventralis(Ehr.)Hust., Tabellaria flocculosa(Roth) Kütz.,Gomphonema augur Ehr.,Navicula subtilissima Cl.等,还有少量的嗜碱性种类出现。  相似文献

Navicula ulvacea (Berkeley ex Kütz.) P. T. Cleve, a marine colonial blade-forming diatom originally discovered in Scotland was collected at North Rustico, Prince Edward Island, during winter 1971. It formed dense stands of Porphyra-like fronds, ranging up to 60 mm long × 20 mm wide. These occurred under ice-covered water on rocks 1–2 m below zero tide. Comparisons made at the light and scanning electron microscope level compared favorably with original Aberdeen material. Clonal cultures established from single cells at first also formed blades, but rarely more than 10 mm long. In numerous serial subcultures made since 1971 no further blades have been formed. Instead, growth consisted of well-separated cells or short filaments scattered over the bottom and sides of the vessels. Older cultures developed prostrate sheets of filaments, embedded in a mucilagenous matrix and occasional bubble-like eruptions. Results of the cultures suggest that N. ulvacea probably occurs in a prostrate form in nature, perhaps more commonly than in the blade form. Literature records since the original collection in 1844 are brought together but are considered doubtful. A discussion on nomenclature is included.  相似文献

于2016年5月对浙江省仙居国家公园进行藻类多样性调查时,发现了中国淡水硅藻的1个新记录种——偏肿内丝藻兴凯变种[Encyonema ventricosum var.hankensis(Skvortzov) RodionovaPomazkina]。该文采用光镜和扫描电镜对偏肿内丝藻兴凯变种浙江种群的形态特征进行了观察和描述,并将其与该种的其他地理种群及内丝藻属其他相似物种的形态特征进行比较,同时讨论了该物种的生境特征。结果表明:(1)偏肿内丝藻兴凯变种壳面均具明显背腹之分,背缘明显拱起,腹缘轻微波曲;端部呈头状至喙状。(2)偏肿内丝藻兴凯变种浙江地理种群在背部中央区域具一孤点,而其兴凯种群对孤点这一特征未有描述。(3)偏肿内丝藻兴凯变种在俄罗斯的兴凯湖和贝加尔湖等地有分布,该种在中国是首次报道。  相似文献

Isolates from four sites were identified as varieties of the diatom Gomphonema parvulum Kütz. The material was examined by light and electron microscopical techniques. A detailed account of the morphology of this species is given.

No fundamental differences between the varieties were found apart from cell shape. It is considered that cell size is of no importance to classification below the species level since reduction in size occurred in all the isolates during normal growth.  相似文献

为探明杭白芷(Angelica dahurica var.formosana)根中分泌道发生方式、分布及其挥发油转运积累特征,该研究利用光镜及透射电子显微镜技术观察分泌道发生过程及挥发油转运特征,结合组织化学定位确定挥发油的主要积累部位。结果表明:杭白芷根中分泌道由中柱鞘细胞最先发生,次生结构中分泌道主要分布在韧皮部和皮层中;挥发油的合成不仅与分泌细胞中质体及细胞质有关,而且还与周围细胞关系密切;分泌细胞内高尔基体和内质网丰富,可能先通过形成小泡参与转运,再经由细胞壁向腔道内转移;相邻分泌细胞靠近角隅处的细胞壁分泌活动活跃,腔道内积累大量电子致密物质;成熟分泌道中分泌细胞及其腔道内积累大量油滴,因此挥发油主要积累场所为分泌细胞及其腔道。该研究明确了杭白芷根中分泌道的发生方式、分布及其挥发油积累部位,揭示了分泌道发育过程中挥发油的转运积累特征,为进一步阐明分泌组织生长发育与有效成分积累关系提供了理论依据。  相似文献

采用毛细管复洗技术建立海链藻属(Thalassiosira)物种的单克隆培养株系,结合形态学和分子系统学研究,对小字海链藻原变种(Thalassiosira minuscula var.minuscula Krasske)的形态特征进行观察,并报道了1个新变种:小字海链藻双卫变种(T.minuscula var.bicustodis X.H.Guo,Y.Q.GuoY.Li)。双卫变种与原变种的形态特征基本一致,区别仅在于唇形突的伴生支持突数目不同,双卫变种具有2个,原变种只有1个。基于对多个株系生活史的连续观察,确认其伴生支持突的数目稳定,具有分类学意义。  相似文献

The morphological and molecular differentiation of the Micrasterias truncata (Corda) ex Bréb species complex was investigated. In total, 17 strains traditionally assigned to M. truncata were isolated from different European localities (Czech Republic, southwest France, Ireland), and obtained from public culture collections. In addition, strains of the morphologically similar species, M. decemdentata (Nägeli) W. Archer and M. zeylanica F. E. Fritsch, were also included. Molecular phylogenetic analysis based on trnGucc intron sequences revealed five well supported clades. Two Australian strains assigned to M. truncata var. pusilla G. S. West formed a lineage sister to M. zeylanica. This was evident from a concatenated phylogeny based on small subunit rDNA and trnGucc intron sequences. The isolated position of these strains was also illustrated by parallel landmark‐based geometric morphometric analysis of cell shapes. The strains NIES 783 and NIES 784 probably represent a separate species. Particular analysis, including additional strains, is needed to resolve the relationship inside this lineage. The second phylogenetic lineage, containing two strains of M. truncata var. semiradiata (Kützing) Wolle, was also different from other strains on the basis of morphometric data. We suggest recognizing this variety as a separate species, Micrasterias semiradiata L.A. Brébisson ex F. T. Kützing. The remaining three clades formed a firmly supported group of the ‘core’M. truncata recognized by both molecular markers. However, neither any morphological, morphometric, nor geographical pattern was detected among members of these three clades. This pattern could be caused by a relatively recent origin of these lineages that may represent a sympatric, truly cryptic species. Strains attributable to traditional morphologically defined variety M. truncata var. neodamensis were nested within the ‘core’M. truncata.  相似文献

Several strains of Microsporum audouini, M. langeroni and M. rivalieri were observed by light and scanning electron microscopies. The macroaleuriospore shape and ornamentation were illustrated and compared for the 3 species as the pectinate hyphae for M. langeroni and M. rivalieri and the chlamydospores and particular hyphae for M. audouini and M. langeroni. The 3 species are morphologically very close but they are distinguishable from each other on the basis of morphology, geographic distribution and pathogenicity.  相似文献

A Japanese clinical isolate (KU-A-0094) which was identified by de Hoog et al. as Exophiala jeanselmei var. lecanii-corni with difficulty, was compared with 5 strains including the type cultures of E. jeanselmei var. lecanii-corni, var. jeanselmei and E. castellanii using RFLP (restriction fragment length polymorphism) patterns of mtDNA (mitochondrial DNA). RFLP patterns of KUA-0094 were identical with those of E. jeanselmei var. lecanii-corni and different from those of E. castellanii with restriction enzymes of HaeIII, MspI and hindIII. Therefore, de Hoog et al.'s identification of KU-A-0094 was confirmed. Additionally, mtDNA-RFLP patterns of E. jeanselmei var. lecanii-corni and E. jeanselmei var. jeanselmei were also different from each other. Consequently E. jeanselmei var. lecanii-corni seem to be a species in its own right rather than a variant of E. jeanselmei. A case of polymorphism in the desmid Micrasterias pinnatifida (Kütz.) Ralfs is described. It is based on material collected from the overflow channel of a dam in Caeté, Minas Gerais State. After careful examination of nearly 500 specimens, 26 different morphological expressions of the alga were found which led the authors to make the following deductions: (1) for delimitation of species and infraspecific taxa in desmids it is absolutely necessary to analyze sample populations; (2) the morphological characteristics presently used, for delimination of some varieties of M. Pinnatifida (Kütz.) Ralfs seem to have no taxonomical value if only isolated individuals are examined; (3) the varieties granulata, inflata, and tridentata must be considered with some caution, until further studies are carried out, because they can be merely distinct morphological expressions of M. pinnatifida (Kütz.) Models of the diatom associations found in upper Florida Bay and adjoining sounds have been constructed utilizing Q modal factor-vector analysis and ecologic diversity indices (Shannon index, number of species, evenness). Four distinct associations were defined using Q-mode factor-vector analysis. Two associations were epiphytic, occurring on Thalassia testudinum König—Association I was characterized by Cocconeis placentula Ehr., Association III by Cylindrotheca closterium (Ehr.) Reim. & Lewin and Cocconeis placentula Ehr. The other two associations were epipelic, occurring on the carbonate mud substratum—Association II was characterized by Cyclotella striata (Kütz.) Grun., Rhopalodia gibberula (Ehr.) O. Müller and Surirella fastuosa (Ehr.) Kütz., Association IV by Fragilaria crotonensis Kitton and Cyclotella striata (Kütz.) Grun. The majority of the 161 species identified were present in both the epiphytic and epipelic assemblages. Only 33 species were restricted to the epiphyton, and 18 species restricted to the epipelon. The epipelic assemblage was significantly more diverse than was the epiphytic assemblage. A general trend of increased diversity away from terrestrial environs toward more open areas of water in both the epipelon and epiphyton was also found.  相似文献

Spirulina major Kütz. was observed with the electron microscope and compared at the ultrastructural level with a species of Arthrospira. The presence, of crosswalls which develop by progressive ingrowth from peripheral walls was demonstrated in S. major. Since cellular septation is not consistent with the generally accepted characters of the genus Spirulina, a taxonomic shift may be indicated.  相似文献

The diatom Eunotia pectinalis (O. F. Müll.?) Rabh. exhibited considerable morphological variability in samples collected over a 13-month period from a softwater Rhode Island stream. All observed morphotypes were classified into three main complexes based upon their valve configuration. These groupings were confirmed by discriminant analysis using quantitative valve characteristics and were named in accordance with their associated varieties: minor (Kütz.) Rabh., pectinalis and ventricosa Grun. Clonal cultures were subjected to a defined series of temperatures, agitation rates and light intensities in order to assess inherent morphological variability. Similar trends were evident for all complexes. Over time, valve length decreased, valve breadth enlarged, and striae number per 10 μm at valve center and apex increased. Morphological plasticity was evident in culture and appeared to be more a function of the clone genotype than the environmental conditions to which it was subjected. One clone originally identified as E. pectinalis var. minor assumed certain morphological features of E. vanheurckii Patr. after four months in culture.  相似文献

Pathogenic and genetic variability among seven populations of Phytophthora parasitica var. nicotianae from individual tobacco fields (Yunnan, Shandong, Henan, Heilongjiang, Shanxi, Fujian and Sichuan provinces) were investigated using pathogenicity and randomly amplified polymorphic DNA (RAPD) analyses; 63 strains were isolated from different fields of seven tobacco growing regions, using tobacco cv. Hongda as a baiting host. Pathogenic variability was evaluated in greenhouse studies using five tobacco cultivars that have different levels of resistance to tobacco black shank; 75 and 73% of the strains were pathogenic on M3 and M4, 29 and 33% on M1 and M2, and 94% were pathogenic on M5, respectively. Disease severity incited by different strains varied significantly on individual tobacco cultivars. The percentage of strains pathogenic on different cultivars varied among locations. Genotypic variation among 63 strains was evaluated by RAPD analysis. Ten primers detected 89 polymorphic bands. Cluster and principal coordinates analysed cluster groups. the minor group contained 26 strains, and major group contained 37 strains. Estimates of genetic diversity based on RAPD analysis ranged from 0.24 to 0.34 within populations to 0.36 among all strains from all populations. Phytophthora parasitica var. nicotianae populations were genotypically and phenotypically variable, but no distinct genotypic differences were identified among populations from the seven locations.  相似文献

Limnological gradients of small, oligotrophic, and low conductance lakes in northern New England were defined by principal components analysis; relationships of sedimented diatom species to the gradients were investigated by correlation analysis. Diatom distributions were most strongly related to the gradient of pH and alkalinity and the covarying variables, conductance, Mg, Ca, total Al, and exchangeable Al. Weaker relationships to lake morphology, dissolved organic carbon and water color, altitude and marine aerosol inputs, and the distinctive water chemistry of some New Hampshire lakes were also present. Results for 16 taxa of importance in our studies of lake acidity are given in detail and are compared to results from other regions of eastern North America. Planktonic taxa were absent below pH 5.5, with the exception of the long form of Asterionella ralfsii var. americana Korn. The two forms of this taxon differed ecologically: the long form (>45μm) had an abundance weighted mean (AWM) pH 4.90 and occurred mostly in lakes that were deep relative to transparency; the short form (<45μm)had an AWM pH and occurred on lakes that were shallow relative to transparency. The ecological advantage of a “splitter” approach to diatom taxonomy was demonstrated by examination of other taxa as well, including Tabellaria flocculosa (Roth) Kütz. These results have important implications for paleolimnological interpretations.  相似文献
