Summary 1. Investigations were carried out on monthly voyages to the Schlei, a fjord of the western Baltic Sea, from 1972 to 1974.2. The Schlei is characterized by two successive water blooms — one in the spring caused byChlorella sp., and a Cyanophyceae bloom in late summer withMicrocystis aeruginosa as the dominant species.3. The blooms are accompanied by different bacteria populations. It was noted, e. g., that the proportion of red pigmented bacteria is greatest during the spring, whereas in late summer the numbers of pigmented bacteria decrease and white or colorless forms dominate.4. In order to determine the relationship of the bacteria to the phytoplankton blooms, the predominant algae and bacteria species in the Schlei were isolated, and laboratory experiments with these microorganisms were carried out.5. The results of these experiments showed that the growth of the red bacteria was clearly enhanced when cultivated together withChlorella sp.6. The enhancement takes place immediately with actively growingChlorella cultures. Apparently these algae secrete organic substances into the culture medium during their exponential phase of growth, which are utilized by the red bacteria. It is likely that a similar process occurs in the Schlei. The rapid increase in red bacteria has been shown to take place parallel to the development of theChlorella water bloom in the spring. The proportion of pigmented bacteria decreases in the late summer with the breakdown of theChlorella bloom, and a new bacterial population becomes dominant.7. Other experiments withChlorella sp. showed thatEscherichia coli is strongly inhibited by the presence of these algae. The inhibition takes place only afterChlorella has reached the end of the exponential phase of growth, however. The inhibitory substances are probably autolysis or degradation products which accumulate in the culture medium during the stationary phase of algal growth.8. None of the bacteria tested had any influence on the growth rate ofChlorella sp., nor were the algae cells colonized by bacteria at any time.9. With the development of theMicrocystis bloom in late summer, white or colorless bacteria are more frequently found in the waters of the Schlei. At the same time, there is generally a decrease in the saprophyte counts including pigmented bacteria.10. Two strains of white bacteria commonly associated withMicrocystis were able to suppress the growth of many other bacteria on agar plates, especially red, yellow and brown pigmented species.11. Sterile filtrates prepared from the culture solutions of the white bacteria had no effect on the strains tested, however. The suppression on agar plates is therefore more likely due to competition for nutrients. There is no evidence that antibiotic substances are involved.12. Filtrates prepared from non-axenic cultures ofMicrocystis aeruginosa during the exponential as well as stationary phase of growth also did not inhibit the growth of the bacteria tested.13. The decrease in the numbers of saprophytic bacteria in the Schlei during the summer may be due in part to the presence of the large white colonies on agar plates which prevent other bacteria from developing. These white bacteria are apparently associated withMicrocystis water blooms; possibly a symbiotic or mutualistic relationship exists between these microorganisms.
Untersuchungen über die Beziehungen zwischen Algenblüten und Bakterienpopulationen in der Schlei (westliche Ostsee)
Kurzfassung Die Wechselbeziehungen zwischen Algenblüten und Bakterien wurden in der Schlei, einer Förde der westlichen Ostsee, untersucht. Die Wasserblüten, im Frühjahr vonChlorella spec. und im Spätsommer vonMicrocystis aeruginosa gebildet, werden von Änderungen in den Bakterienpopulationen begleitet. Laborversuche wurden durchgeführt mit den hauptblütenbildenden Algenarten und den dominanten Bakterienarten. Die Ergebnisse zeigen, daß das Wachstum von einigen rotpigmentierten Bakterien(Flavobacterium), die die Wasserblüte vonChlorella begleiten, durch Stoffe, die von den Algen in der logarithmischen Wachstumsphase in das Medium abgegeben werden, gefördert wird.Microcystis aeruginosa wird von mindestens zwei chitinabbauenden Bakterienarten (darunterPseudomonas spec.) begleitet. Die ökologischen Zusammenhänge werden diskutiert.

Summary The green parameciumParamecium bursaria has many endosymbiotic algae in its cytoplasm. Here, we cloned and characterized endosymbiotic algae fromP. bursaria and examined in detail the interaction between the cloned algae and algae-free paramecia. Homogenates ofP. bursaria were cultured on agar plates containing various kinds of media to establish clones of the endosymbiotic algae. Many algal colonies were obtained from poorly nutritious medium (CA medium) after one month in culture. Algae were picked up from these colonies and inoculations were repeated 9 times on agar plates containing CA medium. On enriched media including bacto-peptone, glucose, proteose-peptone and/or yeast extract, however, bacteria and mold grew rapidly and no algal colonies were formed. When the cloned algae were cultured in liquid CA medium, they grew faster than on agar plates and the numbers stayed constant at 1 × 107 algae/ml after 7 days in culture. They revealed high infectivity to algae-free paramecia, and an incubation period of 24 h and at least 1 × 103 algae/paramecium were required to achieve successful infection (80–90%). The growth and infection rate did not change through 74 repeated inoculations of algae in liquid CA medium. Optical microscopic observations revealed marked morphological similarity between endosymbiotic algae and free-livingChlorella, but the latter showed no infectivity to algae-free paramecia. The cloned endosymbiotic algae presented here will provide an excellent opportunity to examine the mechanism of symbiont-host interaction.  相似文献

The antibacterial properties of the indigenous microflora of rainbow trout ( Oncorhynchus mykiss Walbaum) and the potential use of inhibitory bacteria as fish probiotics were investigated. A total of 1018 bacteria and yeasts were isolated on tryptone soy agar (TSA) from skin, gills and intestine. Forty-five of these inhibited growth of the fish pathogenic bacterium Vibrio anguillarum in a well diffusion assay. The antagonism was most prominent among Pseudomonas spp., as 28 (66%) of the antagonistic bacteria belonged to this genus, despite constituting only 15% of the total tested flora. As pseudomonads are typically siderophore producers, chrome azurol S (CAS) agar was used as a semi-selective medium for isolation of antagonistic bacteria. On this medium, 75% of the iron-chelating strains were inhibitory to V. anguillarum . Eight strains out of a subset of 11 antagonists caused a 3–6 log unit reduction in the density of V. anguillarum [measured by polymerase chain reaction (PCR) detection in a most probable number (MPN) regimen] in a broth co-culture assay. Survival of rainbow trout infected with vibriosis was improved 13–43% by six out of nine antagonistic strains tested in vivo. All disease-protecting strains were pseudomonads, isolated from CAS plates, whereas two Carnobacterium spp. that were antagonistic in in vitro well diffusion assays did not alter the accumulated mortality of rainbow trout. The addition of live bacterial cultures to fish-rearing water may thus improve survival of the fish; however, in vitro antagonism could not completely predict an in vivo effect. Further studies on the underlying mechanism of activity are required to design appropriate selection criteria for fish probiotic bacteria.  相似文献

ABSTRACT. Twenty-six species of bacteria and seven species of yeasts were aseptically presented separately as potential food sources to the estuarine heterotrich ciliate, Fabrea salina, under standardized conditions of cell number, medium, and temperature. The bacteria and yeasts were classified according to their effect on the intrinsic growth rate of the ciliate: Nutritious bacteria: Photobacterium fisherii, Vibrio neresis, V. natriegens, Flavobacterium sp. strain ASN16, V. harveyi, Xanthomonas sp. strain ASN22; Maintainer bacteria: V. vulnificus, Vibrio sp. strain V344, V. alginolyticus, V. anguillarum, Bacillus sp. strain ST332B2, Vibrio sp. strain V415, Acinetobacter sp. strain BHT8, Flectobacillus marinus, and Enterobacter aerogenes; Maintainer yeasts: Candida albicans and Cryptococcus marcerans strain 2; Nonnutriuous bacteria: V. parahaemolyticus, Planococcus sp. strain ASN13, P. mandapapanensis, Hyphomicrobium vulgari, Pseudomonas sp. strain CNS1, an unidentified gram-negative, yellow-pigmented, motile bacillus strain IG9A2, Thiobacillus thioparus, Escherichia coli, Corynebacterium sp. strain BR 17, Achromobacter sp. strain 23030, and Oceanospirillum beijerinckii; Nonnutriuous yeasts: C. marcerans strain 1, Saccharomyces cerevisiae, Debaryomyces hansenii, an unidentified black yeast, and C. laurentii. Of more than 400 bacteria isolated from turbot (Scophthalmus maximus), 89 have previously been shown to inhibit the in vitro growth of the fish pathogen Vibrio anguillarum. The aim of the present study was to investigate the potential of seven of these strains, as well as of intestinal isolates (four strains) from a closely related fish, dab (Limanda limanda), for colonizing farmed turbot as a means of protecting the host from infection by V. anguillarum. In addition, the inhibitory effect of these strains on the pathogen was further studied. Colonization potential was measured by the capacity of the strains to adhere to and grow in turbot intestinal mucus. These parameters were also used to investigate the potential of V. anguillarum to amplify in the turbot intestinal tract. Because of the observed rapid growth of V. anguillarum in intestinal mucus, it can be proposed that the intestinal tract is a site for V. anguillarum multiplication. Strains isolated from the intestine showed greater capacity for adhesion to and growth in fish intestinal mucus than did the pathogen and the skin mucus isolates. All of the isolates released metabolites into the culture medium that had inhibitory effects against V. anguillarum. The results are discussed with emphasis on administering bacteria of host origin to farmed turbot in order to control V. anguillarum-induced disease.  相似文献

海洋环境中,细菌和微藻之间的物质交换是生源要素在自然界中迁移转化的重要方式。为进一步了解生源要素的生物地球化学循环,在实验室模拟条件下,研究了共培养体系中营养盐和有机物在细菌和微藻之间的转换。通过纯培养中肋骨条藻(Skeletonema costatum)、东海原甲藻(Prorocentrum donghaiense)、天然海水中的细菌以及藻菌混合培养,分析了营养盐和有机物随藻菌生物量的变化情况,并计算了溶解有机碳(DOC)和溶解有机氮(DON)的浓度比值[(DOC/DON)a]。结果发现,在共培养体系中,细菌对中肋骨条藻的生长有抑制作用,对东海原甲藻影响不明显;中肋骨条藻有利于细菌生长,东海原甲藻抑制细菌生长,这种不同可能与微藻的粒径有关。海洋细菌在2种藻的指数生长均期均会促进微藻吸收氨氮(NH_4-N),但在生长末期NH_4-N以释放为主。硝氮(NO_3-N)的浓度与藻的生长呈负相关,但在衰亡期NO_3-N略有增加,表明NO_3-N再生所需时间较长。细菌对硝氮的吸收量较少,但对其再生有贡献。细菌和中肋骨条藻对磷酸盐(PO_4-P)的吸收存在竞争,但与东海原甲藻的竞争关系不明显。不同培养体系中DOC浓度变化不同,在藻菌共培养体系中增加较快,纯藻培养体系中增加缓慢,在纯菌培养体系中缓慢减少。通过对DOC与DON浓度比值的分析,发现用判断颗粒有机碳(POC)来源的方法可以分析DOC的来源。  相似文献

Anabaena species are commonly colonized by bacteria, especially during N2-fixing blooms. Generally these associations do not represent bacterial attack on algal hosts. Instead, the algal N2-fixing capabilities are increased in the presence of the bacteria. Possible mechanisms promoting the mutual growth of algae and attached bacteria were investigated by observing specific sites of bacterial attachment, by noting reduced microzones created by the bacteria, and by locating sites of bacterial uptake of organics representative of algal excretion products.Attached bacteria show preference for typical algal excretion products and their growth is enhanced by such products. In return, enhancement of algal nitrogenase activity occurs when bacteria create O2-consuming microzones around the nitrogenase-bearing heterocysts.  相似文献

Cells of Chlorella pyrenoidosa, derived from vanadium free agar slants, respond with great sensitivity to microamounts of vanadium, added as NH4VO3 to autotrophic liquid cultures. Between 0.01 and 1 g V per litre nutrient medium (2·10-10-2·10-8g-at/l), the algae respond with a continuous increase in dry weight. At higher V-concentrations, further enhancement in biomass is accompanied by a additional increase in chlorophyll content. Maximum V-effect on both parameters was found to be at 500g V/l (10-5 g-at/l). Dry weight as well as chlorophyll content of Chlorella are decreased by concentrations above 25 mg V/l; 100 mg V/l (2·10-3 g-at/l) stop growth and cause death of the cells. The toxic threshold for the V-content in the algae was determined to be at 150–200 g V/g (3–4·10-6 g-at/g) dry weight.Two different pH-optima for a positive vanadium action on dry weight and chlorophyll biosynthesis were established, the first at pH 7, the other in the range pH 7.5–8. Contrary to an expectation from the size-efficiency hypothesis, small herbivore zooplankton such as Ceriodaphnia often competitively predominate against large species such as Daphnia. However, little is known about critical feeding conditions favoring Ceriodaphnia over Daphnia. To elucidate these conditions, a series of growth experiments was performed with various types of foods in terms of phosphorus (P) contents and composition (algae and bacteria). An experiment with P-rich algae showed that the threshold food level, at which an individual’s growth rate equals zero, was not significantly different between the two species. However, the food P:C ratio, at which the growth rate becomes zero, was lower for Daphnia than for Ceriodaphnia, suggesting that the latter species is rather disfavored by P-poor algae. Ceriodaphnia showed a higher growth rate than Daphnia only when a substantial amount of bacteria was supplied together with a low amount of P-poor algae as food. These results suggest that an abundance of bacteria relative to algae plays a crucial role in favoring Ceriodaphnia over Daphnia because these are an important food resource for the former species but not for the latter.  相似文献

Bacteria inhibitory to fish larval pathogenic bacteria were isolated from two turbot larva rearing farms over a 1-year period. Samples were taken from the rearing site, e.g., tank walls, water, and feed for larvae, and bacteria with antagonistic activity against Vibrio anguillarum were isolated using a replica plating assay. Approximately 19,000 colonies were replica plated from marine agar plates, and 341 strains were isolated from colonies causing clearing zones in a layer of V. anguillarum. When tested in a well diffusion agar assay, 173 strains retained the antibacterial activity against V. anguillarum and Vibrio splendidus. Biochemical tests identified 132 strains as Roseobacter spp. and 31 as Vibrionaceae strains. Partial sequencing of the 16S rRNA gene of three strains confirmed the identification as Roseobacter gallaeciensis. Roseobacter spp. were especially isolated in the spring and early summer months. Subtyping of the 132 Roseobacter spp. strains by randomly amplified polymorphic DNA with two primers revealed that the strains formed a very homogeneous group. Hence, it appears that the same subtype was present at both fish farms and persisted during the 1-year survey. This indicates either a common, regular source of the subtype or the possibility that a particular subtype has established itself in some areas of the fish farm. Thirty-one antagonists were identified as Vibrio spp., and 18 of these were V. anguillarum but not serotype O1 or O2. Roseobacter spp. strains were, in particular, isolated from the larval tank walls, and it may be possible to establish an antagonistic, beneficial microflora in the rearing environment of turbot larvae and thereby limit survival of pathogenic bacteria.  相似文献

为改善采矿废弃物上植被生长状况,提高植物成活率,该研究采用盆栽试验法,以高丹草为材料,选用摩西球囊霉(Glomus mosseae,G.m)和地表球囊霉(G.versiforme,G.v)两种AM真菌,分别研究单接种和混合接种对粉煤灰(S1)、煤矸石(S2)和粉煤灰与煤矸石混合物(S3)三种煤矿废弃物基质上高丹草(Sorghum bicolor×S.sudanense)生长及叶绿素荧光的影响,并以正常沙土(S4)作为对照。结果表明:(1)4种基质上,3种接种处理均获得较高侵染率,在基质S1、S3和S4上均为接种摩西球囊霉对高丹草根系侵染率最高,分别为49.04%、57.40%、43.34%,在基质S2上,混合接种处理对高丹草根系侵染效果最好,达49.33%。(2)3种煤矿废弃物基质上高丹草根长、干重、叶绿素含量、F_v/F_o、q P和Yield显著降低。接种AM真菌显著提高了高丹草的生长和光合效率。与其他处理相比,在基质S1、S3和S4上,接种摩西球囊霉显著增加了根长、干重、叶绿素含量、F_v/F_o、q P和Yield,在基质S2上,接种地表球囊霉显著增加了根长、干重,接种地表球囊霉和摩西球囊霉+地表球囊霉(G.mv)处理间叶绿素荧光参数均无显著差异。这表明在煤矿废弃物基质的复合逆境中高丹草生长和光合作用显著受到抑制,AM真菌可通过提高高丹草叶绿素含量,改善叶片叶绿素荧光和光合作用,促进植物生长,来缓解该复合逆境对高丹草造成的伤害,增强其对煤矿废弃物不良环境的抗逆性,提高煤矿区植被恢复效果。接种摩西球囊霉对粉煤灰以及粉煤灰和煤矸石混合基质上高丹草的促进作用最佳,而接种地表球囊霉更适于煤矸石基质上高丹草的生长。  相似文献

Summary Root-colonizing pseudomonads capable of inhibiting seedling winter wheat (Triticum aestivum L.) root growth in an agar seedling bioassay also significantly inhibited wheat root growth in vermiculite; however, the inhibitory trait is quite labile in laboratory culturing. The extent of inhibition in both the agar and vermiculite medium depended on inoculum level. These pseudomonads were found to produce a toxin capable of inhibiting growth ofEscherichia coli C-la andBacillus subtilis. Field isolates that strongly inhibit growth of indicator bacteria also inhibited root growth. Toxin production by the bacteria appeared necessary for inhibition of root growth and indicator bacteria as toxin-negative (TOX) mutants no longer inhibited either. Antibiosis towardsE. coli as well as wheat seedling root inhibition in agar was reversed by L-methionine, providing further evidence that a toxin, produced by these organisms, is involved in growth retardation.Contribution in cooperation with the College of Agric. Green cocoa pod husk agar (GCPA), ripe cocoa pod husk agar (RCPA), green cocoabean agar (GCBA), ripe cocoa bean agar (RCBA), green cocoa mucilage agar (GCMA)and ripe cocoa mucilage agar (RCMA) were prepared and assessd for their clarity andfor potential to support mycelial growth and sporulation of P. palmivora. Oatmeal agar (OMA), potato-dextrose agar (PDA), vegetable 8 juice agar (V8JA) and pineapple crown agar (PCA) were included for comparison. The highest radial growth rates of 8.3 and 7.2 mm/day were recorded, respectively, on OMA and GCPA but these were not significantly different (P ≤ 0 05) from each other. The two media also supported good aerial mycelial growth but were not clear. Radial mycelial growth rates of 6.5, 7.0 and 6.6 mm/day were obtained on GCMA, RCPA and V8JA, respectively, and these rates were also not significantly different from each other. Of the three media, only the GCMA was clear and supported the best aerial mycelial growth. In comparison, the RCMA supported a significantly lower radial growth (4.6 mm/day) of P. palmivora than the three media. Growth rates were least on RCBA, PCA and PDA but sporulation was poorest on PDA, PCA and V8JA. GCMA was found to be the best medium based on all the growth parameters and media characteristics. GCMA has been used effectively to isolate/detect P. palmivora from infected cocoa pod tissues. Apart from differences in radial growth rate, both the GCMA and RCMA were similar in all other respects and are recommended for culturing P. palmivora. Guar gum, a galactomannan, has been reported to be an inexpensive substitute of agar for microbial culture media. However, its use is restricted probably because of (1) its highly viscous nature even at high temperatures, making dispensing of the media to Petri plates difficult and (2) lesser clarity of the guar gum gelled media than agar media due to impurities present in guar gum. To overcome these problems, three guar gum derivatives, carboxymethyl guar, carboxymethyl hydroxypropyl guar and hydroxypropyl guar, were tested as gelling agents for microbial growth and differentiation. These were also evaluated for their suitability for other routine microbiological methods, such as, enumeration, use of selective and differential media, and antibiotic sensitivity test. For evaluation purpose, growth and differentiation of eight fungi and eight bacteria grown on the media gelled with agar (1.5%), guar gum (4%) or one of the guar gum derivatives (4%), were compared. All fungi and bacteria exhibited normal growth and differentiation on all these media. Generally, growth of most of the fungi was better on guar gum derivatives gelled medium than on agar medium. The enumeration carried out for Serratia sp. and Pseudomonas aeruginosa by serial dilution and pour plate method yielded similar counts in all the treatments. Likewise, the selective succinate medium, specific for P. aeruginosa, did not allow growth of co-inoculated Bacillus sp. even if gelled with guar gum derivatives. The differential medium, Congo red mannitol agar could not differentiate between Agrobacterium tumefaciens and Rhizobium meliloti on color basis, if gelled with guar gum or any of its derivatives However, for antibiotic sensitivity tests for both Gram-positive and -negative bacteria, guar gum and its derivatives were as effective as agar.  相似文献

Factors excreted into the medium by various bacteria and fungi exert a significant effect on the growth and revertibility of bacterial L-forms. Studies with L-forms ofHaemophilus influenzae have been facilitated by a strain ofNeisseria perflava, which not only promotes the growth of the L-forms on media of normal osmolarity (horse blood agar), but also enhances the revertibility of the L-forms to the parent bacterium. L-forms derived fromN. perflava also exhibit this growth-enhacing property, an effect that does not appear to be related to X and V factors. Preliminary characterization of the active component produced byNeisseria perflava indicates that it is present in broth filtrate, diffusible through agar, heat labile, and of large molecular size.  相似文献

The diatom population structure of a salt marsh epiphytic community growing on Enteromorpha intestinalis was studied at one station throughout the summer. A total of 218 species or varieties were recognized. Six species— Fragilaria construens, Cocconeis scutellum, Cocconeis placentula, Achnanthes hauckiana varieties, Achnanthes pinnata, and Amphora coffeaeformis (var. acutiuscula)–dominated the Enteromorpha epiphytic community during the summer months and comprised ∼ 40% of the total populations. Melosira nummuloides, Opephora martyi, Synedra fasiculata var. tabulata, S. affinis, Navicula platyventris, and N. pavillardi were also very common species (13% of the total population). The distribution of many species in the community was seasonal. A series of differential media have been developed which are effective as tools for the isolation and nutritional characterization of the algae and bacteria from the community. Many diatom species can be recognized by their colony type or growth pattern on solidified media. A key and illustrated plates aid in identification. Changes in population structure of the community were reflected by changes in the nutritional patterns as judged by differences in the growth of diatoms and bacteria on the differential media tested. The nutritional requirements, selectivity, and rank order of media for individual species is given. Almost half of the colony types (32–33) grew on either unenriched seawater from the collection site or basal synthetic seawater. Erdschreiber was a poorer medium for the isolation of algae than seawater alone. Few algal colonies grew on either marine nutrient agar or trypticase soy agar. Nitrite inhibited algal growth. Media enriched with thiamine, biotin, or nitrate, phosphate, and B12 were stimulatory. Soil extract or an acetone extract of Enteromorpha tripled growth; a complex vitamin mixture, or glycerol, or mannitol, or an autoclaved extract of Enteromorpha doubled colony numbers. The greatest numbers of diatom types (43) and total colonies (655) were recovered on media enriched with acetone extract of Enteromorpha, suggesting a possible nutritional relationship between Enteromorpha and its epiphytes. The representativeness of the isolations from the natural community as a function of time was also considered. Concentrations of selected dissolved potential growth-stimulating nutrients were measured within and external to Enteromorpha communities. We conclude that the algal assemblage growing epiphytically on Enteromorpha intestinalis has a diverse auxotrophic profile which contributes to the productivity and stability within this important component of the epiphytic community and that much of the organic substrates used by individual species originate within and are recycled among the community members.  相似文献

The purpose of this study was to select, identify and characterise bacteria as a disease control measure in the rearing of marine fish larvae (turbot, Scophthalmus maximus). Thirty-four out of 400 marine bacterial strains exhibited in vitro anti-bacterial activity against three fish larval pathogens. Two strains originated from culture collections and thirty two strains were isolated directly from turbot larvae rearing units using a pre-selection procedure to facilitate detection of antagonists. Approximately 8,500 colonies from colony-count plates were replica-plated on agar seeded with Vibrio anguillarum, and 196 of them caused zones of clearing in the V. anguillarum agar layer. Of these, 32 strains exhibited reproducible antibacterial properties in vitro when tested against the fish pathogens V. anguillarum 90-11-287, V. splendidus DMC-1 and a Pseudoalteromonas HQ. Seventeen antagonists were identified as Vibrio spp. and four of twelve tested were lethal to yolk-sac larvae. The 15 remaining strains were identified as Roseobacter spp. based on phenotypic criteria and 16S rDNA gene sequence analysis of two strains representing the two major RAPD groups. Most of the remaining 164 strains selected in the initial replica plating were identified as Vibrionaceae or Pseudoalteromonas. Roseobacter spp. were not lethal to egg yolk sac turbot larvae and in two of three trials, the mortality of larvae decreased (p > 0.001) in treatments where 10(7) cfu/ml Roseobacter sp. strain 27-4 was added, indicating a probiotic potential.  相似文献
