The distribution of genetic diversity in a local population of the trematode Schistosoma mansoni was determined within and between individual wild rats at a microspatial geographic scale of a standing water transmission site. Using RAPD markers, molecular variance and canonical correspondence analysis were performed to test the significance of genetic differentiation between infrapopulations. Of total gene diversity, 8 and 11% was partitioned between hosts trapped at few metres distance from each other. Significant temporal differentiation (2%) was also detected among schistosomes sampled at 6 month intervals with more infrapopulation pairs differentiated during the dry season of parasite transmission than during the rainy season (45 and 12%, respectively). A combination of factors such as restricted displacement of rats, patchy spatial aggregation of infected snails and limited cercarial dispersion in standing water are likely to promote the genetic differentiation observed between infrapopulations at this microgeographic scale.  相似文献

Dipteronia is an endemic genus to China and includes only two species, Dipteronia sinensis and D. dyeriana. Based on random amplified polymorphic DNA (RAPD) markers, a comparative study of the genetic diversity and genetic structure of Dipteronia was performed. In total, 128 and 103 loci were detected in 17 D. sinensis populations and 4 D. dyeriana populations, respectively, using 18 random primers. These results showed that the proportions of polymorphic loci for the two species were 92.97% and 81.55%, respectively, indicating that the genetic diversity of D. sinensis was higher than that of D. dyeriana. Analysis, based on similarity coefficients, Shannon diversity index and Nei gene diversity index, also confirmed this result. AMOVA analysis demonstrated that the genetic variation of D. sinensis within and among populations accounted for 56.89% and 43.11% of the total variation, respectively, and that of D. dyeriana was 57.86% and 42.14%, respectively. The Shannon diversity index and Nei gene diversity index showed similar results. The abovementioned characteristics indicated that the genetic diversity levels of these two species were extremely similar and that the interpopulational genetic differentiation within both species was relatively high. Analysis of the genetic distance among populations also supported this conclusion. Low levels of interpopulational gene flow within both species were believed to be among the leading causes for the above-mentioned phenomenon. The correlation analysis between genetic and geographical distances showed the existence of a remarkably significant correlation between the genetic distance and the longitudinal difference among populations of D. sinensis (p < 0.01), while no significant correlation was found between genetic and geographical distances among populations of D. dyeriana. This indicated that genetic distance was correlated with geographical distances on a large scale rather than on a small scale. This result may be related to differences in the selection pressure on species by their habitats with different distribution ranges. We suggest that in situ conservation efforts should focus on establishing more sites to protect the natural populations and their habitats. 为揭示我国东部归化水仙(Narcissus tazetta var. chinensis)的群体遗传多样性,利用2个叶绿体基因mat K和trn H-psb A片段对采自沪、浙、闽的5个代表群体的49株水仙进行了评估。结果表明,双基因联合序列的总长为1443bp,共定义6个单倍型,各归化群体的遗传多样性水平为DLSYPTDNJD=ZZCMD。AMOVA分析表明,群体内变异为遗传变异的主要来源(91.98%),群体间的遗传分化较低(Fst=0.080 22)。群体物种水平上的谱系结构不显著(Nst=0.020Gst=0.031;P0.05)。Mantel检验表明水仙群体间的遗传距离与地理距离呈显著的线性相关(r=0.929,P=0.02 0.05)。中性检验和错配分布分析结果均暗示水仙群体背离了快速扩张模型的假设。单倍型分布的中介网络图结合系统发育NJ树均将所有群体划分为2大分支。因此,我国东部沿海水仙归化群体整体遗传多样性水平较低,各群体间遗传分化较弱,遗传变异主要来自群体内,物种近期未经历扩张事件,可能是基因流受海岛隔离、自身生物学特征、生境异质性与及人为干扰的综合作用影响。  相似文献

唐古特大黄(Rheum tanguticum)是中国传统的中藏药材,近几年由于生境的严重破坏,已濒临灭绝,并被列入濒危植物名单。为了探索唐古特大黄物种濒危的原因并保护其野生资源,本研究采集了9个居群87个个体的唐古特大黄样本,基于该物种的叶绿体基因trn S-G序列对其进行了遗传多样性研究。结果表明,唐古特大黄物种具有较高的遗传多样性水平(Ht=0.694),其中95.97%的遗传分化来自于居群间(G_(ST)=0.960),4.03%的遗传分化来自于居群内(Hs=0.028)。AMOVA分析也显示唐古特大黄居群间基因流较小(N_m=0.01),存在较高的遗传分化(F_(ST)=0.9631)。唐古特大黄较高的遗传多样性水平可能与该物种较长的进化史和生活史有关,居群间较高的遗传分化可能与高山地区特殊的地理环境和人类活动有关。根据研究结果,建议对唐古特大黄所有野生居群进行就地保护,同时收集种质资源开展异地繁殖工作,以保护物种的遗传多样性,维持其进化潜力。  相似文献

为了阐明东北三省近年育成水稻品种的遗传基础,该研究通过对2021年育成的256个水稻新品种的系谱进行分析,确定了20个材料为育种核心亲本,并利用44对多态性SSR分子标记和34对InDel籼粳特异性分子标记分析了它们的遗传多样性和籼粳分化程度。结果表明:(1)SSR标记能够高效评价参试材料的多样性水平,共检测到130个等位基因,每个位点检测等位基因(Na)变异为2~5个,平均为2.95个;基因多样性指数(H)变异为0.10~0.75,平均为0.41;多态性信息含量(PIC)变异为0.09~0.70,平均为0.36。(2)遗传多样性分析表明,东北三省20份水稻核心亲本的整体多样性水平较低,黑龙江省核心亲本多样性略高于辽宁省和吉林省。(3)NJ聚类分析表明,20份核心亲本可以划分为3个类群,分别为辽宁类群、吉林类群和黑龙江类群。(4)遗传结构分析表明,20份核心亲本可划分为2个稳定的类群,其中辽宁省核心亲本均处于A类群,黑龙江省核心亲本均处于B类群。(5)籼粳分化分析表明,20份核心亲本遗传背景均为粳稻类型,但都含有少数籼型位点,其籼型基因频率F在0.01~0.19范围内。研究认为,东北三省水稻育种核心亲本遗传基础狭窄、使用频率较高是导致育成品种遗传多样性水平较低的可能原因,在育种中亟待挖掘新种质、引入新资源,丰富遗传背景。  相似文献

以华北落叶松控制授粉群体的全部子代为材料,母本相同的个体视为同一家系,利用18对SSR分子标记对7个家系257个个体进行扩增,分析其遗传多样性及其遗传分化水平。结果表明:(1)18个SSR位点共检测到72个等位基因,平均等位基因数4个,有效等位基因数(Ne)为1.247~3.411。(2)7个家系的平均有效等位基因数为2.135个,观测杂合度(Ho)为0.518,期望杂合度(He)为0.502,Shannon信息指数0.846,其中55号家系遗传多样性水平最高,56号的遗传多样性水平最低。(3)遗传分化系数(GST)为0.113,各家系群体处于中等遗传分化水平,AMOVA分析结果显示82%的遗传变异存在于家系内,18%的遗传变异存在于家系间。(4)聚类分析结果表明,55号与59号家系的遗传距离最近,具有较近的亲缘关系,49号家系与其他家系的遗传距离最远。(5)结合遗传多样性及遗传分化水平结果,估算获得选择核心家系数及核心个体数,选择5个家系均可获得96%以上的遗传多样性,对于个体数较少的家系,选择15~20个个体;对于个体数较多的家系,选择35个个体,即可获得96%以上的遗传多样性。该研究结果对华北落叶松种子园育种群体的选择及其遗传多样性的保护具有重要意义。  相似文献

为了解海南岛油茶(Camellia oleifera)种质资源的遗传多样性,采用SRAP分子标记,对海南岛油茶主要分布区的31个居群进行了遗传多样性和亲缘关系分析。结果表明,海南岛油茶资源的遗传多样性低,物种水平的多态性百分率(PPB)为98.30%,Nei’s基因多样性(H)为0.222 8,Shannon信息指数(I)为0.353 8;居群水平的PPB=40.96%,观测等位基因数(Na)为1.409 6,有效等位基因数(Ne)为1.237 1, H=0.138 5, I=0.208 3,这与海南岛油茶丰富的表型多样性水平不一致。海南岛油茶资源遗传分化较大,居群间基因交流有限,不同居群间的遗传分化指数(Gst)为0.380,基因流(Nm)为0.813 91。遗传变异主要发生在居群内,有38.05%的变异存在居群间,61.95%存在于居群内。遗传距离为0.022 6~0.276 4,平均为0.107 7,居群间的亲缘关系较近。UPGMA聚类分析表明,在遗传距离为0.11处,可将31个油茶居群聚为6类,表现出明显的行政区域性,而与地理距离关系不大。因此,海南岛油茶资源遗传多样性低,亲缘关系近可能导致自交或近交不亲和,可能是海南油茶林分花量大而结实低的主要内在原因。  相似文献

Molecular markers have been used only rarely to characterize the population genetic structure of nematodes. Published studies have suggested that different taxa may show distinct genetic architectures. Isoenzyme and RAPD markers have been used to investigate geographic variation of Ascaris suum at the level of infrapopulations (nematodes within individual hosts), within localities, and among geographic regions. Independent estimates of genetic differentiation among population samples based on isoenzyme and RAPD data showed similar patterns and substantial correlation. Heterozygote deficiencies within infrapopulations and large values for inbreeding coefficients among infrapopulations suggested that the composition of these populations was not consistent with a model of random recruitment from a large panmictic pool of life-cycle stages. Both isoenzyme and RAPD markers revealed moderate levels of genetic differentiation among samples representing infrapopulations and localities. Of total gene diversity, 9.4% (isoenzyme) and 9.2% (RAPD) was partitioned among infrapopulations. Geographic localities accounted for 7.8% (isoenzyme) and 6.2% (RAPD) of total diversity. Only infrapopulations from the same farm had low levels of differentiation.  相似文献

淡黄金花茶(Camelia flavida)是一种具有淡黄色花瓣的金花茶植物,为喀斯特典型的耐阴灌木。基于广西弄岗喀斯特季节性雨林15 hm2(300 m×500 m)监测样地,利用13对微卫星(SSR)标记,探讨喀斯特地貌对淡黄金花茶的精细空间遗传结构(SGS)、基因流和小尺度范围遗传分化的影响。结果表明:(1)淡黄金花茶在50 m距离内产生显著的SGS,在灌木类型和依赖种子传播的物种中具有中等程度的SGS强度(Sp=0.0248);(2)淡黄金花茶种子和花粉传播平均距离较短,分别为12.47 m和29.03 m,72.2%花粉和81.0%种子的传播距离均主要是在20 m内;(3)淡黄金花茶在小尺度范围(种群间距离1 km)的4个种群,甚至相距100 m的两个斑块产生了显著的遗传分化。喀斯特生境地貌对淡黄金花茶的基因流产生阻碍作用,从而导致淡黄金花茶在小尺度范围产生遗传分化。  相似文献

为揭示中国橄榄(Canarium album)种质资源的遗传多样性,采用ISSR和RAPD标记对橄榄主要分布区的86份种质资源进行遗传多样性分析并构建核心种质。结果表明,基于UPGMA遗传相似系数,86份种质资源可分为3个大类;基于STRUCTURE模型聚类,可分为4个类群,这基本符合橄榄的地域性分布规律。采用ISSR和RAPD获得的中国橄榄种质资源的整体遗传多样性水平分别为0.284±0.169和0.244±0.163,多态性位点百分率分别为92.56%和100%,总遗传分化系数分别为0.127和0.142,基因流分别为3.423和3.025,群体间遗传相似系数分别为0.930和0.939,个体间遗传相似系数分别为0.736和0.732。因此,中国橄榄种质资源丰富的遗传多样性主要来源于个体间的遗传分化或变异,且这种遗传多样性存在明显的地域性差异。  相似文献

为了解铁皮石斛(Dendrobium officinale)种质间的亲缘关系,利用ISSR技术对34份铁皮石斛种质资源进行亲缘关系和遗传多样性分析。结果表明,9条ISSR引物在34份种质中共扩增出78条带,多态位点百分率达100%。UPGMA聚类分析表明,种质的相似系数为0.61~0.92,在相似系数0.626处,福建省泰宁的野生铁皮石斛与栽培铁皮石斛分为两大类。泰宁野生铁皮石斛种群的Nei’s基因多样性(H)和遗传分化系数(Gst)分别为0.3111和0.4609,均高于栽培种群(0.3056和0.4204),表明泰宁野生铁皮石斛具有较丰富的多样性和较高的种群分化系数。AMOVA分析表明,铁皮石斛种群内变异指数为74%,种群间变异指数为26%,表明不同种群间可能存在基因交流。这些为不同地域的野生铁皮石斛资源的有效保护及利用提供理论依据及技术参考。  相似文献

该研究利用 ISSR 分子标记,对不同生境下的6个河口莲座蕨种群进行遗传多样性分析。结果表明:从44条ISSR引物中筛选出8条能够扩增出清晰、稳定条带的引物,对6个河口莲座蕨种群进行基因组 DNA扩增,共扩增出144条带,其中多态性条带119个,多态性条带比率为93.7%,Nei’s遗传多样性指数(H)为0.296,Shannon多样性指数(I)为0.457,遗传分化指数(Gst)为0.1520,种群遗传一致度和遗传距离分别为0.9138~0.9548和0.0584~0.0901;UPGMA聚类分析表明,种群间遗传距离与空间距离和生境类型有关。河口莲座蕨在不同干扰程度的生境中,种群具有高水平的遗传多样性,一定强度或频率的干扰生境中,种群遗传多样性较高,与海拔、坡度和坡向无相关性。  相似文献

Tissue-samples from 161 bank voles (Clethrionomys glareolus) collected in three forests (five sampling localities) situated in eastern Jutland (Denmark) were analysed by nine microsatellite loci. The genetic diversity found within the populations was high (He=0.753–0.806).Bank voles have specific habitat requirements favouring woodlots, hedgerows and deciduous forests as their prime living area. Hence, a natural or human-induced fragmentation of the forest may cause a sub-structuring of the populations and thereby a restriction of dispersal among populations. The sub-structuring indicated by the observed significant genetic differentiation among the five geographically distinct localities (Fst =0.033, P<0.05) could either result from habitat fragmentation or a combination of home range behaviour and different tree composition in the forests. A road situated between two adjacent forests was not found to exert any barrier effect to the gene flow of bank voles. In one out of five localities investigated, genetic evidence for a recent bottleneck-like situation was found. Bank voles are known to exhibit sometimes huge density fluctuations not only from year to year but also from season to season. The bottleneck-like situation found could therefore be due to the low number of individuals during the low-density phase.  相似文献

应用ISSR分子标记技术,对云南南部7个地区的野生大叶千斤拔( Flemingia macrophylla)居群进行了遗传多样性分析。结果表明:云南野生大叶千斤拔具有较高的遗传多样性。在物种水平上,平均每个位点的多态位点百分率(PPL)为94.85%,有效等位基因数(Ne)为1.4627,Nei’s基因多样性指数(He)为0.2815, Shannon’s多样性信息指数(Ho)为0.4337;在居群水平上,PPL =43.44%,Ne =1.2981,He =0.1704,Ho =0.2499。基于Nei’ s遗传多样性分析可得出,居群间的遗传分化系数( Gst)为0.3975,表明居群内的遗传变异为60.25%,居群间的遗传变异为39.75%,这说明居群间的遗传分化要低于居群内的遗传分化。根据遗传多样性分析和聚类结果,应在大叶千金拔遗传多样性较高的勐腊易武( MY)、丘北( QB)和宁洱( NE)地区,设立保护点对其进行就地保护。  相似文献

通过线粒体控制区序列的分析,研究采自中国南海及东海5个群体102尾细鳞鯻的遗传多样性。发现在962 bp序列中有205个变异位点,其中135个为简约信息位点,共定义102个单倍型。中国近海细鳞鯻总体呈现出较高的遗传多样性特征(Hd=1.000,Pi=0.022),其中博鳌最高(Hd=1.000,Pi=0.028),平潭最低(Hd=1.000,Pi=0.014)。不同地理群体间无明显分化,基因交流频繁(Fst=-0.014—0.041,P0.05);中性检验均为显著负值,推测在16.9万年—5.06万年前,即中-晚更新世出现种群扩张。系统邻接树和单倍型网络图均出现3个显著分化的谱系(谱系间Fst=0.508—0.698,P0.001;净遗传距离Da=0.024—0.031),且各谱系中均有不同地理来源的群体。3个谱系间分歧时间大约在1.07百万年—0.24百万年前,推测可能是更新世冰期边缘海的出现导致群体隔离而产生分化。谱系A(Lineage A)包含85.3%的个体,其总体遗传多样性较高(Hd=1.000,Pi=0.012),其中平潭最高(Hd=1.000,Pi=0.014),合浦最低(Hd=1.000,Pi=0.010);群体间Fst在-0.021—0.068之间,P0.005;AMOVA分析显示只有1.97%的变异来自于种群间,表明群体间也无明显分化;中性检验均为显著负值,推测在25.4万年—7.6万年前出现种群扩张。中国近海细鳞鯻主要受到中-晚更新世海侵和海退的影响而出现种群扩张使得谱系间发生二次接触,最终形成具有显著谱系结构但无地理分化的情况。  相似文献

大明松是广西和贵州特有的高山松类,具有很高的经济价值和生态价值,其自然种群长期没有得到充分的保护和利用,不利于该树种长期稳定发展。为了合理保护和开发利用大明松天然遗传资源,该文利用12个SSR分子标记对大明松3个天然群体进行遗传多样性研究,分析其群体间的遗传分化和基因流,为该物种的保护策略提供参考。研究结果表明:(1)12对引物共检测到37 个等位基因,多态位点百分率为100%。每个位点平均观察等位基因数(Na)为3.08,平均有效等位基因数(Ne)为1.68, 不同位点间有效等位基因数差异较大; 每个位点平均观察杂合度(Ho)为0.35,平均期望杂合度(He)为0.40,平均多态信息含量(PIC)为0.31。(2)3 个群体的Shannon''s多样性指数变化范围为0.48~0.65,Nei''s基因多样度的变化范围为0.27~0.39,与其他近缘种松类相比遗传多样性偏低。群体平均观察杂合度为0.40,平均期望杂合度为0.33,平均有效等位基因数为1.58。群体间基因分化系数(Gst)为0.10,群体间的遗传分化水平较低,大部分变异均存在群体内。群体间的基因流(Nm)为2.74,说明大明松群体间的基因交流比较充分。该研究可为大明松生物多样性保护提供重要参考依据,为科学利用大明松资源打下基础。  相似文献

岛屿具有独特的生态系统,常被生态学家和进化生物学家视为研究生物进化的天然实验室,岛屿生物地理学也受到了越来越多科学家的关注。对舟山群岛8个面积不等岛屿的黄毛鼠(Rattus losea)种群进行了调查,分析了8个种群的遗传变异特征,对探讨岛屿理论中的种群动态和种群分化具有重要意义。采用线粒体分子标记技术,利用PCR扩增得到D-loop区基因序列815 bp,在330个黄毛鼠样本中共识别出15个单倍型,平均核苷酸多样性(P_i)为0.001,平均单倍型多样性(H_d)为0.364,表明舟山群岛黄毛鼠种群的遗传多样性较低。Tajima′s D中性检验显示除了小盘峙种群,均为显著负值(P0.01),表明种群受到了自然选择的作用,历史上发生过种群扩张。AMOVE显示,群体间的遗传分化指数平均值为0.745,处于较高的分化水平,表明遗传变异主要来自种群间,占74.5%。基于线粒体D-loop区序列构建的系统发育树和中值网络都表明8个岛屿的黄毛鼠种群起源于两个母系。此外,Mental检验显示不同岛屿种群间的遗传距离与岛屿间地理距离之间存在显著正相关关系(r=0.6077,P=0.004),种群遗传多样性与岛屿面积并未发现显著相关性(r=0.6255,P=0.1840)。研究结果可为岛屿黄毛鼠种群的微观演化以及一些岛屿物种的进化理论提供参考。  相似文献

薄叶金花茶、小花金花茶和小瓣金花茶是三种濒危金花茶植物,为了解珍稀濒危植物遗传多样性和遗传结构,该研究利用微卫星标记对他们的7个种群共184个个体进行了遗传多样性和遗传结构分析。结果表明:11个位点共检测到等位基因92个。在物种水平上,小瓣金花茶平均等位基因数(N_A)为3.9、有效等位基因数(N_E)为2.328、观测杂合度(H_o)为0.520、期望杂合度(H_e)为0.501,高于薄叶金花茶和小花金花茶。在种群水平上,有效等位基因数(N_E)在1.788~2.466之间,期望杂合度(H_e)在0.379~0.543之间;种群间遗传分化系数(FST)在0.143 7~0.453 3之间,种群间基因流(N_m)在0.301 5~1.488 9之间。AMOVA分子变异分析显示65.72%的变异存在于种群内。三种金花茶具有较低水平的遗传多样性和高水平的种群间遗传分化。STRUCTURE和PCoA种群遗传结构分析结果将取样种群分为2组,即薄叶金花茶和小花金花茶大部分个体分为一组,小瓣金花茶大部分个体分为一组。现存所有种群应根据实际情况尽快采取就地保护或迁地保护措施。  相似文献
