The purpose of this study was to investigate criteria for maximal effort in middle-aged men and women undertaking a maximal exercise test until they were exhausted if no measurements of oxygen uptake are made. A large group of 2164 men and 975 women, all active in sports and aged between 40 and 65 years, volunteered for a medical examination including a progressive exercise test to exhaustion on a cycle ergometer. In the 3rd min of recovery a venous blood sample was taken to determine the plasma lactate concentration ([la]p, 3min). Lactate concentration and maximal heart rate (f c, max) were lower in the women than in the men (P<0.001). Multiple regression analyses were performed to assess the contribution of sex to [la]p, 3 min, independent of age and f c max, It was found that [la]p,3 min was about 2.5 mmol·l–1 lower in women than in men of the same age and f c, max. In our population 88% of the men and 85% of the women met a combination of the following f c, max and [la]p, 3min criteria: f c, max equal to or greater than 220 minus age beats·min–1 and/or [la]p, 3min equal to or greater than 8 mmol·l–1 in the men and f c, max equal to or greater than 220 minus age beats·min–1 and/or [la]p, 3min equal to or greater than 5.5 mmol·1–1 in the women.  相似文献

A group of 14-healthy men performed anisotonic isometric contractions (AIC), for 60 s, at an intensity of 100% maximal voluntary contraction force (MVC) during handgrip (HG) and leg extension (LE). Heart rate (f c), stroke volume index (SVI) and cardiac output index (QcI) were measured during the last 10 s of both AIC by an impedance reography method. Force (F) exerted by the subjects was recorded continuously and reported as a relative force (F r) (% MVC). The F generated during MVC was greater for LE than for HG (502.I N compared to 374.6 N, P < 0.001). The rate of decrease in F r was significantly slower for LE than HG for the first 25 s of the exercise (phase 1 of AIC). The F r developed by the subjects at the end of AIC was 40% MVC for both LE and HG. The increase in f c was greater for LE (63 beats · min–1) than for HG (52 beats · min–1), P < 0.01. The SVI decreased significantly from the resting level by 17.0 ml · m–2 and by 18.2 ml · m–2 for LE and HG, respectively. The QcI increased insignificantly for HG by 0.091 · min–1 · m–2 andsignificantly forLE by 0.561 · min–1 · m–2 (P < 0.001). It was concluded that although both AIC caused a significant decrease in SVI, greater increases in f c and Qc were observed for LE than for HG. The greater f c and Qc reported during LE was probably related to the greater relative force exerted by LE during phase 1 of AIC. It seems, therefore that central command might have dominated for phase 1 of AIC but that the muscle reflex also contributed significantly to the control of the cardiac response to the high intensity AIC.  相似文献

To gain information on extended flight energetics, quasi-natural flight conditions imitating steady horizontal flight were set by combining the tetheredflight wind-tunnel method with the exhaustion-flight method. The bees were suspended from a two-component aerodynamic balance at different, near optimum body angle of attack and were allowed to choose their own speed: their body mass and body weight was determined before and after a flight; their speed, lift, wingbeat frequency and total flight time were measured throughout a flight. These values were used to determine thrust, resultant aerodynamic force (magnitude and tilting angle), Reynolds number, total flight distance and total flight impulse. Flights in which lift was body weight were mostly obtained. Bees, flown to complete exhausion, were refed with 5, 10, 15 or 20 l of a 1.28-mol·l-1 glucose solution (energy content w=18.5, 37.0, 55.5 or 74.0 J) and again flown to complete exhaustion at an ambient temperature of 25±1.5°C by a flight of known duration such that the calculation of absolute and relative metabolic power was possible. Mean body mass after exhaustion was 76.49±3.52 mg. During long term flights of 7.47–31.30 min similar changes in flight velocity, lift, thrust, aerodynamic force, wingbeat frequency and tilting angle took place, independent of the volume of feeding solution. After increasing rapidly within 15 s a more or less steady phase of 60–80% of total flight time, showing only a slight decrease, was followed by a steeper, more irregular decrease, finally reaching 0 within 20–30 s. In steady phases lift was nearly equal to resultant aerodynamic force; tilting angle was 79.8±4.0°, thrust to lift radio did not vary, thrust was 18.0±7.4% of lift, lift was somewhat higher/equal/lower than body mass in 61.3%, 16.1%, 22.6% of all totally analysable flights (n=31). The following parameters were varied as functions of volume of feeding solution (5–20 l in steps of 5 l) and energy content. (18.5–74.0 J in steps of 18.5 J): total flight time, velocity, total flight distance, mean lift, thrust, mean resultant aerodynamic force, tilting angle, total flight impulse, wingbeat frequency, metabolic power and metabolic power related to body mass, the latter related to empty, full and mean (=100 mg) body mass. The following positive correlations were found: L=1.069·10-9 f 2.538; R=1.629·10-9 f 2.464; P m=7.079·10-8 f 2.456; P m=0.008v+0.008; P m=18.996L+0.022; P m=19.782R+0.021; P m=82.143T+0.028; P m=1.245·bm f 1.424 ; P mrel e=6.471·bm f 1.040 ; =83.248+0.385. The following negative correlations were found: V=3.939–0.032; T=1.324·10-4–0.038·10-4. To elucidate the role of factors other than the nervous system in heart rate (f c) control during exercise, the kinetics off c and plasma catecholamine concentrations were studied in ten heart transplant recipients during and after 10-min cycle ergometer exercise at 50 W. Thef c did not increase at the beginning of the exercise for about 60 s. Then in the eight subjects who completed the exercise it increased following an exponential kinetic with a mean time constant of 210 (SEM 22) s. The two other subjects were exhausted after 5 and 8 min of exercise during whichf c increased linearly. At the cessation of the exercise,f c remained unchanged for about 50 s and then decreased exponentially with a time constant which was unchanged from that at the beginning of exercise. In the group of eight subjects plasma noradrenaline concentration ([NA]) increased after 30 s to a mean value above resting of 547 (SEM 124) pg · ml–1, showing a tendency to a plateau, while adrenaline concentration ([A]) did not increase significantly. In the two subjects who became exhausted an almost linear increase in [NA] occurred up to about 1,300 pg · ml–1 coupled with a significant increase in [A]. During recovery an immediate decrease in [NA] was observed towards resting values. The values of thef c increase above resting levels determined at the time of blood collection were linearly related with [NA] increments both at the beginning and end of exercise with a similar slope, i.e. about 2.5 beats · min–1 per 100 pg · ml–1 of [NA] change. These findings would seem to suggest that in the absence of heart innervation the increase inf c depends on plasma [NA].  相似文献

The dorsal skin of the leech Hirudo medicinalis was used for electrophysiological measurements performed in Ussing chambers. The leech skin is a tight epithelium (transepithelial resistance = 10.5±0.5 k· cm-2) with an initial short-circuit current of 29.0±2.9 A·cm-2. Removal of Na+ from the apical bath medium reduced short-circuit current about 55%. Ouabain (50mol·l-1) added to the basolateral solution, depressed the short-circuit current completely. The Na+ current saturated at a concentration of 90 mmol Na+·l-1 in the apical solution (K M=11.2±1.8 mmol·l-1). Amiloride (100 mol·l-1) on the apical side inhibited ca. 40% of the Na+ current and indicated the presence of Na+ channels. The dependence of Na+ current on the amiloride concentration followed Michaclis-Menten kinetics (K i=2.9±0.4 mol·l-1). The amiloride analogue benzamil had a higher affinity to the Na+ channel (K i=0.7±0.2 mol·l-1). Thus, Na+ channels in leech integument are less sensitive to amiloride than channels known from vertebrate epithelia. With 20 mmol Na+·l-1 in the mucosal solution the tissue showed an optimum amiloride-inhibitable current, and the amiloride-sensitive current under this condition was 86.8±2.3% of total short-circuit current. Higher Na+ concentrations lead to a decrease in amiloride-blockade short-circuit current. Sitmulation of the tissue with cyclic adenosine monophosphate (100 mol·l-1) and isobutylmethylxanthine (1 mmol·l-1) nearly doubled short-circuit current and increased amiloride-sensitive Na+ currents by 50%. By current fluctuation analysis we estimated single Na+ channel current (2.7±0.9 pA) and Na+ channel density (3.6±0.6 channels·m-2) under control conditions. After cyclic adenosine monophosphate stimulation Na+ channel density increased to 5.4±1.1 channels·m-2, whereas single Na+ channel current showed no significant change (1.9±0.2 pA). These data present a detailed investigation of an invertebrate epithelial Na+ channel, and show the similarities and differences to vertebrate Na+ channels. Whereas the channel properties are different from the classical vertebrate Na+ channel, the regulation by cyclic adenosine monophosphate seems similar. Control of fed-batch culture of hybridoma cells was investigated based on two approaches optimal control theory and feedback control. Experiments were conducted for both approaches-with a feed enriched in glutamine. The optimal feed trajectory, a decreasing one, yielded a final monoclonal antibody (MAb) concentration of 170 mg/l, a three-fold increase compared to a typical batch operation.The feedback strategy relied on the on-line estimation of the net specific growth rate of cells from the measurement of the CO2 production rate with a mass-spectrometer. A PI controller was then used to maintain the growth rate at a desired value by adjusting the dilution rate to the reactor. For the chosen set-point (0.1 d–1), the final MAb concentration achieved was about 100 mg/1. It was found that there was a delay in the assimilation of the glutamine that should be included in the model to explain the lower MAb production in feedback mode. The growth of vegetative and reproductive shoots of Nicotiana tabacum L. cv. Xanthi is analyzed with the plastochron index to estimate the relationship between corolla growth and time. The plastochron of leaves 9 through 20 declines steadily at each successive node. The flower plastochron increases steadily during the growth of an individual cyme, with the most distal flower to open having the longest plastochron. Variation in the flower plastochron is the result of variation in the rate of flower initiation, not the growth rate of individual flowers. The corolla has an extended phase of approximately constant relative growth in length (between 0.2 · d–1 and 0.3 · d–1) until a peak of growth (0.5 · d–1) 2–3 d before anthesis. Corollas also have periodic peaks and troughs of growth that are low in amplitude (0.1 · d–1), but persist throughout most of corolla development. Summary The BMR (6.00 ml O2·min–1) and thermal conductance (0.235 ml O2·min–1·°C–1) ofAmazona viridigenalis, a medium sized parrot, are close to allometrically predicted values for nonpasserine birds, but theT 1c of 26.5 °C is 8.5 °C higher than predicted (Fig. 1). Minimal respiratory frequencies measured in four species of birds average 60% of the rate predicted by a previous equation and yield the relationship, breaths·min–1= 10.3 kg–0.32. Frequencies are very dependent upon the methods used to obtain the data (Fig. 3). Resting values of respiratory parameters are poorly defined in the existing literature, and there are no single resting values within the TNZ analogous to a BMR. Rather values change within, as well as below and above, the TNZ. Minimal values of different parameters occur at differentT a's, not necessarily within the TNZ (Figs. 2, 4, 5). For clarity, resting respiratory parameters should be reported as standard values, analogous to standard metabolic rates, withT a specified. In birds the pattern of ventilation (f andV T) changes asT a changes resulting in different extraction efficiencies at a given minute volume (Figs. 6, 7). A brief survey of the literature on manifestations of myo-electric fatigue has disclosed a surprisingly sharp conflict between early studies, focusing on neuromotor regulatory mechanisms, and more recent studies which stress the determinant influence of local metabolism and skewed homeostasis. Favoured explanations concerning changes in the electromyographic (EMG) spectrum were synchronization/grouping of motor unit (MU) firing and conduction velocity (CV) decreases of the action potential propagation. The notion of mutual exclusivity interwoven with these theories prompted us to reinvestigate the EMG of moderate level, static endurance contraction. Ten men in their twenties performed isometric elbow flexion (elbow angle 135°) at 30%6 maximal voluntary contraction (MVC), and the surface EMG of the brachioradialis (BR) and biceps brachii (BB) muscles was recorded. Initially the CV — determined by cross-correlation — was 4.3 m · s–1 (BR) and 4.6 m · s–1 (BB). At exhaustion the CV of the BR muscle had declined by 33%, roughly twice the decrease of the BB CV. Substantially larger relative median frequency (f m) reductions of 50% (BR) and 43% (BB) were found. Simultaneously, the root-mean-square amplitudes grew by 150% (BR) and 120% (BB). All changes during contraction reached the same level of significance (P<0.001, both muscles). From the largely uniform relative increases inf m and CV during the last 4 min of a 5-min recovery period, variations in CV were suggested to produce equivalent shifts inf m. The gradually increasing discrepancies between relative decreases inf m and CV during contraction presumably reflected centrally mediated regulation of MU firing patterns (notably synchronization). After the 5-min recovery another 11 endurance contractions at 30% MVC were executed, separated by 5-min intervals. The series of contractions reduced the endurance time to one-third of the 153 s initially sustained, while the terminal CV recordings increased by 1.0 (BR) and 0.6 (BB) m · s–1, and the terminalf m increased by 24 (BR) and 14 (BB) Hz. The relative CV decreased in direct proportion to the endurance time and thef m decreases varied with the CV; the findings did not support a causal link between CV decrease (signifying impaired fibre excitability) and the force failure of exhaustion.  相似文献

The hydraulic conductivities of excised whole root systems of wheat (Triticum aestivum L. cv. Atou) and of single excised roots of wheat and maize (Zea mays L. cv. Passat) were measured using an osmotically induced back-flow technique. Ninety minutes after excision the values for single excised roots ranged from 1.6·10-8 to 5.5·10-8 m·s-1·MPa-1 in wheat and from 0.9·10-8 to 4.8·10-8 m·s-1·MPa-1 in maize. The main source of variation was a decrease in the value as root length increased. The hydraulic conductivities of whole root systems, but not of single excised roots, were smaller 15 h after excision. This was not caused by occlusion of the xylem at the cut end of the coleoptile. The hydraulic conductivities of epidermal, cortical and endodermal cells were measured using a pressure probe. Epidermal and cortical cells of both wheat and maize roots gave mean values of 1.2·10-7 m·s-1·MPa-1 but in endodermal cells (measured only in wheat) the mean value was 0.5·10-7 m·s-1·MPa-1. The cellular hydraulic conductivities were used to calculate the root hydraulic conductivities expected if water flow across the root was via transcellular (vacuole-to-vacuole), apoplasmic or symplasmic pathways. The results indicate that, in freshly excised roots, the bulk of water flow is unlikely to be via the transcellular pathway. This is in contrast to our previous conclusion (H. Jones, A.D. Tomos, R.A. Leigh and R.G. Wyn Jones 1983, Planta 158, 230–236) which was based on results obtained with whole root systems of wheat measured 14–15 h after excision and which probably gave artefactually low values for root hydraulic conductivity. It is now concluded that, near the root tip, water flow could be through a symplasmic pathway in which the only substantial resistances to water flow are provided by the outer epidermal and the inner endodermal plasma membranes. Dithiothreitol (DTT), an inhibitor of violaxanthin de-epoxidation and zeaxanthin formation in chloroplasts, inhibited blue-light-stimulated stomatal opening in epidermal peels of Vicia faba L. in a concentration-dependent fashion. Complete inhibition was observed at 3 mM DTT. The DTT effect was specific for the stomatal response to blue light, and the red-light-stimulated opening, which depends on photosynthetic reactions in the guard cells, was unaffected. Preirradiation of stomata in epidermal peels with increasing photon fluence rates of red light, prior to an incubation in 10 mol·m-2·s-1 of blue light and 100 mol·m-2·s-1 red light, resulted in a DTT-sensitive, blue-light-stimulated opening that was proportional to the fluence rate of the red light pre-treatment. Guard cells in epidermal peels and guard-cell protoplasts irradiated with red light showed increases in their zeaxanthin content that depended on the fluence rate of red light, or on the incubation time. The increases in zeaxanthin concentration were inhibited by DTT. Heart rate (beats · min–1;f c) measured during marching with a load is often used to predict the oxygen cost (1·min–1; VO2) of the activity. The prediction comes from thef c/VO2 relationship determined from laboratory measures off c and VO2 during treadmill running. Studies in men have suggested that this may not be appropriate although this has yet to be examined in women. This study, therefore, compared thef c/VO2 relationship between loaded marching and maximal running protocols in women. Sixteen female subjects [mean (SD), age 21.9 (2.3) years, height 6 (0.06) m, weight 62.6 (7.6) kg] had theirf c (from three-lead chest electrodes) and VO2 measured first during standard treadmill run protocols, and again 1 week later during loaded marching protocols. The slopes and intercepts determined from linear regression off c on VO2 for each individual for each protocol were compared as were the maximalf c(f cmax), VO2 and ratings of perceived exertion (RPE) from the last work period of each protocol in pairedt-tests. The VO2 slopes (P < 0.01) and intercepts (P < 0.05) differed significantly between loaded marching and running.f cmax for loaded marching were 90% off cmax for running (P < 0.01) and VO2 for loaded marching were 80% of those for running (P < 0.01). However, RPE at the final levels for the two protocols were not significantly different. The data suggest that in women the VO2 relationships for loaded marching and for running are different. This difference is similar to that found in men when speed is held constant and the load and gradient are varied. The results suggest that it would be erroneous to usef c and VO2 measured during running protocols in the laboratory to estimate energy expenditure and work intensity during loaded marching in the filed.  相似文献

Summary Abiotic factors, standing crop and photosynthetic production were studied in the equatorial alkaline-saline closed-basin Lake Nakuru (cond. 10,000–160,000 S). Meteorological conditions and abiotic factors offer suppositions for a high primary productivity: mean solar radiation is 450–550 kerg·cm-2·s-1, with little seasonal variation, regular winds circulate the lake every day and nutrient concentrations are usually high (>100 g P–PO4·l-1). Oxygen concentrations near sediments were <1 gO2·m-3 for at least 6 h·d-1 in 1972/73, resulting in a release of 45 mg P–PO4·m-2·d-1. Attenuation coefficients vary from 3.6–16.5 according to algal densities and mean depth from 0–400 cm. Algal biomass was 200 g·m-3 (d.w.) in 1972/73, due to a lasting Spirulina platensis bloom (98.5% of algal biomass). In 1974 algal biomass suddenly dropped to 50 g·m-3 (d.w.). Spirulina and several consumer organisms almost vanished, but coccoid cyanobacteria, Anabaenopsis and diatoms increased. Several causes for this change in ecosystem structure are discussed. The use of the light/dark bottle method to measure photosynthetic production in eutrophic alkaline lakes is discussed and relevant experiments were done. Oxygen tensions of 2–35 gO2·m-3 do not influence primary production rates. Net photosynthetic rates (mgO2·m-3·h-1; photosynthetic quotient=1.18) reached 12–17.7 in 1972/73 and 2–3 in 1974, but vertically integrated rates were only 1–1.4 in 1972/73 and 0.8 in 1974, and daily net photosynthetic rates (gO2·m-3·24 h-1) 3.5 in 1972/73 and 1 in 1974. 50% of areal rates were produced within the 10 most productive cm of the depth profile. The disproportion between high algal standing crops and relatively low production rates is due to self-shading of the algae, reducing the euphotic zone to 35 cm in 1972/73 and 77 cm in 1974. Efficiency of light utilization is 0.4–2%, varying with time of day and phytoplankton density. In situ efficiencies show an inverse relationship to light intensities. Photosynthetic rates of L. Nakuru remain within the range of other African lakes (0.1–3 gO2·m-2·h-1). The relation of O2 produced/Chl a of the euphotic zone is 50% lower then in tropical African freshwater lakes and conforms to lakes of temperate regions.  相似文献

Summary Physical characteristics, namely floc density function, floc size distribution, and relative floc strength, of a number of flocculent yeast types were measured. A straight-line relationship was found to exist between log values of size and density for the yeasts examined. Each yeast type had coefficients from this relationship which could be used to interpret settling behaviour. Isometric flexion of the right elbow at 15% of the maximal voluntary contraction (MVC) was maintained to the limit of endurance (elbow angle 135°). The surface electromyogram (EMG) of the brachioradialis (BR) and biceps brachii (BB) muscles was recorded for calculation of conduction velocity (CV) by the cross-correlation method, and determination of median frequency (fm) and root mean square (rms) amplitude. Perceived exertion was rated for both muscles, and heart rate and blood pressure were measured. The EMG of ten brief 15% MVC contractions distributed over a 30-min recovery period was also recorded. Eleven males in their twenties volunteered for the investigation. The average endurance time was 906 (SD 419) s. Mean CV for the unfatigued muscles was 4.2 (SD 0.41) m·s–1 (BR), and 4.3 (SD 0.29) m·s–1 (BB). The contraction caused a significant decrease in CV of BR (12%,P<0.001) whereas CV variation of BB remained insignificant. Concurrently the meanf m of both muscles dropped to approximately 66% of their initial values and their average rms amplitudes grew by approximately 380% (BR and BB:P<0.001, both parameters). The 1st min of recovery lowered the rms amplitudes by approximately 60% (BR and BB:P<0.01), while thef m increased to approximately 88% of the initial recording (BR,P<0.01; BB,P<0.05). The accompanying small increases in CV were beyond the level of significance. Over the next 29 min a significant parallel restitution inf m and CV took place; changes inf m evidenced a simple one to one reflection of relative CV variation. A similar uncomplicated linear causality between relative changes in CV andf m was hypothesized for the endurance contraction. Consequently, the 12% CV decrease of the BR accounted for only one-third of the fatigue inducedf m reduction of 33%, while two-thirds were assumed to be attributable to centrally mediated regulatory interventions in motor unit (MU) performance. Independent of contributions from the virtually unchanged CV, thef m of the BB muscle decreased by 35%; from one subject exhibiting a remarkably manifest burst-type pattern of MU activity it is argued that synchronization/grouping of MU firing predominantly determined the power redistribution in the BB spectrum.  相似文献

The classical induction of Crassulacean acid metabolism (CAM) in Mesembryanthemum crystallinum L. by water stress is observed within one week when fourto five-week-old plants (grown under a 16/8 h photoperiod at ca. 600 mol quanta · m–2 · s–1) are irrigated with 350 mM NaCl. The induction of CAM was evaluated by measuring phosphoenolpyruvate carboxylase (PEPCase, EC 4.1.1.31) and NADP-malic enzyme (NADP-ME, EC 4.1.1.82) activities and nocturnal increases in malate content and titratable acidity of leaf extracts, and the daily pattern of CO2 exchange and stomatal conductance during the 7-d induction period. Three growth regulators, abscisic acid (ABA), farnesol (an antitranspirant and analog of ABA), and benzylaminopurine (BAP), were found to substitute for NaCl for induction of CAM when fed to plants in nutrient media. Daily irrigation with solutions containing micromolar levels (optimum ca. 10 micromolar) of these growth regulators led to the induction of CAM similar to that by high salt. Application of the growth regulators, like NaCl, caused large increases in the activity of NADP-ME and the activity and level of PEPCase, which are components of the biochemical machinery required for CAM. Western immunoblotting showed that the increased activity of PEPCase on addition of ABA, farnesol and BAP was mainly due to increased levels of the CAM-specific isoforms. Also, dehydration of cut leaves over 8.5 h under light resulted in a severalfold increase in PEPCase activity. An equivalent increase in PEPCase activity in excised leaves was also obtained by feeding 150 mM NaCl, or micromolar levels of ABA or BAP via the petiole, which supports results obtained by feeding the growth regulators to roots. However, the increase in PEPCase activity was inhibited by feeding high levels of BAP to cut leaves prior to dehydration, indicating a more complex response to the cytokinin. We showed that transposon P{GUS · p53.259H}, mapped to chromosome 3 and carrying a dominant mutation p53 259H.GUS, has a positive effect on the frequency of spontaneous and carcinogen-induced tumor mosaic clones warts in Drosopila melanogaster heterozygotes for the tumor suppressor gene warts located in the same chromosome. The transposon effect could be explained either by the arrest of apoptosis in the cells expressing mutant p53 259H.GUS gene and containing carcinogen-induced pre-mutations, and/or by genetic instability introduced into chromosome 3 by the P{GUS · p53.259H} transposon itself. The effect of the P{GUS · p53.259H} appeared to be carcinogen-specific. It substantially increased the frequency of tumors induced by supermutagenic platinum complex, oxoplatin, and did not increase the frequencies of tumors induced by polycyclic aromatic hydrocarbons, benzo()pyrene and pyrene. In the spectrum of mutations induced by all carcinogens tested, somatic recombination events prevailed over somatic mutations. Hence, carcinogen-specificity of the P{GUS · p53.259H} effect cannot be explained by preferential induction of somatic mutations or somatic recombination by one of the carcinogens. Organ-specificity of the increased frequency of mosaic warts clones induced by P{GUS · p53.259H} was established.  相似文献

A modified Rotating Biological Contactor (RBC) was used for the treatability studies of synthetic tapioca wastewaters. The RBC used was a four stage laboratory model and the discs were modified by attaching porous nechlon sheets to enhance biofilm area. Synthetic tapioca wastewaters were prepared with influent concentrations from 927 to 3600 mg/l of COD. Three hydraulic loads were used in the range of 0.03 to 0.09 m3·m–2·d–1 and the organic loads used were in the range of 28 to 306 g COD· m–2·d–1. The percentage COD removal were in the range from 97.4 to 68. RBC was operated at a rotating speed of 18 rpm which was found to be the optimal rotating speed. Biokinetic coefficients based on Kornegay and Hudson models were obtained using linear analysis. Basolateral K+ channels and their regulation during aldosterone- and thyroxine-stimulated Na+ transport were studied in the lower intestinal epithelium (coprodeum) of embryonic chicken in vitro. Isolated tissues of the coprodeum were mounted in Ussing chambers and investigated under voltage-clamped conditions. Simultaneous stimulation with aldosterone (1 mol·l-1) and thyroxine (1 mol·l-1) raised short-circuit current after a 1- to 2-h latent period. Maximal values were reached after 6–7 h of hormonal treatment, at which time transepithelial Na+ absorption was more than tripled (77±11 A·cm-2) compared to control (24±8 A·cm-2). K+ currents across the basolateral membrane with the pore-forming antibiotic amphotericin B and application of a mucosal-to-serosal K+ gradient. This K+ current could be dose dependently depressed by the K+ channel blocker quinidine. Fluctuation analysis of the short-circuit current revealed a spontaneous and a blocker-induced Lorentzian noise component in the power density spectra. The Lorentzian corner frequencies increased linearly with the applied blocker concentration. This enabled the calculation of single K+ channel current and K+ channel density. Single K+ channel current was not affected by stimulation, whereas the number of quinidine-sensitive K+ channels in the basolateral membrane increased from 11 to 26·106·cm-2 in parallel to the hormonal stimulation transepithelial Na+ transport. Summary Daily rates of gross and net primary production were calculated in the Scotia-Weddell Sea sector of the Southern Ocean during spring 1988 (EPOS, Leg 2) on the basis of kinetic experiments, which combine radiotracer technology and classic biochemical procedures, and by taking into account the light regime, the physical structure of the water column, the vertical distribution of chlorophyll a, and the protozoan grazing pressure. From these calculations, three distinct sub-areas were identified: the Closed Pack Ice Zone (CPIZ), characterized by the lowest average gross primary production (0.36 gC · m–2 · day–1); the Marginal Ice Zone (MIZ) with a maximum mean value of 1.76 gC · m–2 · day–1; and the Open Ocean Zone off the ice edge (OOZ) with an intermediate mean value of 0.87 gC · m–2 · day–1. Net primary production fluctuated nearly in the same proportions, averaging 0.55, 0.2 and 1.13 gC · m–2 · day–1 in the OOZ, CPIZ and MIZ respectively, representing 53% of the total photo-assimilated carbon under heavy ice cover (CPIZ) and 64% in the two other areas. Available light, strongly dependent on the ice cover, was shown to control the level of primary production in the sea ice associated sub-areas, whilst protozoa grazing on phytoplankton determined the moderate primary production level characteristic of the well illuminated OOZ area.Data presented here were collected during the European Polarstern Study (EPOS) sponsored by the European Science Foundation  相似文献
