Summary Specific secretory cells in the hypophyseal pars tuberalis of Djungarian hamsters maintained under different photoperiods were investigated immunocytochemically by means of the colloidal gold technique using antibodies against rat thyrotropin (TSH). Secretory cells of animals kept under long photoperiods (LD16:8) showed positive staining of secretory granules (diameters 90–130 nm), whereas other intracellular structures were free of immunoreactivity. In animals kept under short photoperiods (LD8:16) secretory cells displayed increased numbers of secretory granules, but these organelles were devoid of immunoreactivity. In contrast, immunoreactivity of thyrotropes in the pars distalis did not differ between the two groups of animals investigated. The present results confirm earlier light-microscopical studies that in the pars tuberalis specific secretory cells show TSH-like immunoreactivity; however, they differ in their reactivity pattern from classical thyrotropes in the pars distalis.  相似文献

The effect of different photoperiods on the specific secretory cells of the pars tuberalis was examined in male chicks. Animals were placed in one of three different photoperiod regimens: (1) normal control (light:dark = 12 h:12 h), (2) continuous light (L:D = 24 h:0), and (3) extended darkness (L:D = 1 h:23 h). The levels of common alpha-subunit mRNA in the pars tuberalis were examined by Northern blot analysis and compared with those in the pars distalis. In chicks exposed to continuous light for 1 week, alpha-subunit mRNA level in the pars tuberalis was decreased, although the level in the pars distalis was increased. Exposure to continuous light for 30 days also induced a decrease in alpha-subunit mRNA level in the pars tuberalis. On the other hand, in chicks exposed to extended darkness for 1 week, the alpha-subunit mRNA level of the pars tuberalis was markedly increased. In situ hybridization with digoxigenin-labeled common alpha-subunit cRNA probe also showed that the hybridization signals for alpha-subunit mRNA in the pars tuberalis cells become weak under continuous light for 30 days but they are very intense under extended darkness. Thus, the synthesis of alpha-subunits in the chick pars tuberalis was inhibited by continuous light but stimulated by extended darkness. These results were confirmed by semiquantitative electron-microscopic analyses. After exposure to continuous light for 30 days, many pars tuberalis (PT)-specific cells were filled with enlarged secretory granules, showing the reduction of secretory activity. On the contrary, extended darkness for 30 days induced hypertrophy of the PT-specific cells; the areas of cytoplasm and nucleus were significantly increased. In addition, secretory granules became small in size and exocytotic features were more frequent. Mitochondria and lysosomes were also increased in number. Thus, the synthetic and secretory activities of the PT-specific cells were increased under extended darkness. The data indicate that the specific cells of the pars tuberalis are responsive to photoperiodic changes in the chick.  相似文献

Summary Certain secretory cells in the hypophysial pars tuberalis of the Djungarian hamster display marked circannual structural alterations. The present investigation deals with the immunohistochemical properties of this cell group. A distinct TSH-like immunoreactivity was found in secretory cells of this type in the pars tuberalis of animals exposed to long photoperiods, whereas under short photoperiods the TSH-like immunoreactivity was nearly absent. In the pars distalis, the number and distribution of TSH-positive cells did not differ significantly between animals maintained under long and under short photoperiods. LH-and FSH-positive cells could not be detected in the pars tuberalis, but they are clearly present in the pars distalis of both groups of hamsters. Our immunocytochemical results suggest that photoperiodic stimuli influence the secretory activity of TSH-like immunoreactive cells in the pars tuberalis. Summary The pars tuberalis of the hypophysis of Rana temporaria presents the general structural and the cytological characteristics of an endocrine gland. It is composed of elongated cells with long, branching processes ending on the external basement membrane of the pericapillary space. The pars tuberalis cells produce secretory granules which are accumulated in the pericapillary endings of the processes.Corresponding to its separate localization, the pars tuberalis of Rana temporaria has a separate vascularization of which the efferent capillaries anastomose with the capillary plexus of the median eminence. The general direction of the blood flow of the pars tuberalis is towards the capillaries of the median eminence. Also, the secretory products of the pars tuberalis pass into the blood stream of the hypophysial portal system.Several characteristics of the pars tuberalis show that its function must be different from that of the pars distalis of the hypophysis. Moreover, in contrast with the pars distalis, the activity of the pars tuberalis is not regulated by neurohumoral factors.The results show that a role of the pars tuberalis in the regulation of the activity of the pars distalis of the hypophysis is not excluded.  相似文献

In an earlier study onHeteropneustes fossilis, evidence of secretory activity in the pinealocytes had been demonstrated at the electron microscopic (EM) level and it was found to exist in two phases: a secretory phase (light cells) and a storage phase (dark cells). In the present investigation,H. fossilis was subjected to artificial photoperiods of continuous illumination and continuous darkness for a period of ten days and the effect on the secretory pinealocytes was studied at the EM level. Marked results were observed within the short period of ten days emphasizing the role of environmental photoperiod on the secretory activity of the pinealocytes. During continuous illuminated phase, both light and dark cells were observed: the light cells showed intense secretory activity and dark cells a storage one. During the dark phase both types of cells were present but in different metabolic states and neither of the cells demonstrated synthetic nor storage activity. Light cells were metabolically active but not secretory active and dark cells showed a necrotic condition. Phagocytotic activity of the dark cells was also seen. Intense neural activity was also observed during exposure to both the artificial photoperiods. The results highlight the role of light on the secretory activities of the pinealocytes of the catfish pineal organ.  相似文献

Summary The secretory dynamics of the Crustacean Hyperglycemic Hormone (CHH)-producing cells in the eyestalk of the crayfish Astacus leptodactylus were studied during the daily cycle (12 h light/12 h dark). The different secretory stages of individual cells were determined by means of immunocytochemistry combined with morphometric analysis at the light-microscopic level. The data obtained were correlated with the 24-h rhythmicity of blood glucose concentration. The results suggest the following hypothesis. The synthetic activity of the CHH cells receives a stimulus 2 h before the beginning of the dark period, resulting in a pronounced transfer of CHH granules into the axons. These CHH granules reach the axon terminals after the onset of the dark period. At that time a burst of exocytotic activity occurs, causing a strong release of CHH into the hemolymph. Four hours later this CHH release results in hyperglycemia. The same process, though with less intensity, is repeated and causes a second smaller glucose peak at the beginning of the light period.  相似文献

Onset of sexual maturation was determined in weanling male collared lemmings exposed to one of three experimental regimens of different photoperiods before and after weaning. Animals gestated in photoperiods of either 16 h light:8 h dark or 8 h light:16 h dark. Those from 16 h light:8 h dark were transferred at 19 days of age to either 20 h light:4 h dark or 8 h light:16 h dark; those gestated under 8 h light: 16 h dark remained in that photoperiod throughout the experiment. After exposure for 15, 20, 25 or 30 days to the postweaning photoperiod, animals were killed and the following parameters assessed: body weight, testes weight, seminal vesicle weight, the presence or absence of epididymal spermatozoa and serum concentrations of prolactin, testosterone and corticosterone. All parameters except serum testosterone were significantly influenced by photoperiod. Animals housed under 8 h light:16 h dark had significantly greater body weights than those housed under 20 h light:4 h dark, a response that differs from that reported for other arvicoline rodents. The group gestated on 16 h light:8 h dark and transferred on day 19 to 8 h light:16 h dark had lower testes and seminal vesicle weights than the other two groups, and mature spermatozoa in the epididymides appeared 5 days later than in the 20 h light:4 h dark group. Serum prolactin was largely undetectable in animals from both 8 h light:16 h dark groups, but all males housed in 20 h light:4 h dark had 2.0-15.0 ng prolactin ml-1. Concentration of serum corticosterone was higher in animals weaned into long photoperiod, and decreased with age. These data indicate that weanling male D. groenlandicus are reproductively photoresponsive, but use a decrease in photoperiod, not static short-photoperiod exposure, to alter the rate of development. Prolactin was largely undetectable in animals exposed to short photoperiod, indicating that high concentrations of this hormone are not important for maturation. Low prolactin concentrations in animals in short photoperiods may mediate the annual moult to white pelage. The short-photoperiod-mediated decrease in corticosterone may play a role in seasonal changes in body weight and composition.  相似文献

 The pars tuberalis mainly consists of the secretory cells specific to this portion of the pituitary. We examined the localization and development of luteinizing hormone (LH) and chromogranin A in the chicken pars tuberalis by immunohistochemistry. The vast majority of the chicken pars tuberalis was occupied by cells immunoreactive for both LH and chromogranin A. Furthermore, immunoblot analysis of chicken pars tuberalis extracts with LH antiserum demonstrated that two bands, the large α-subunit and small β-subunit of the LH molecule, were expressed in this tissue as well as in the pars distalis. A band for chromogranin A was also detected in pars tuberalis extracts with chromogranin A antiserum. In contrast to the cells of mammalian species that contain only a few small secretory granules, the specific cells of the chicken pars tuberalis were characterized by the presence of many secretory granules ranging from 90 to 400 nm in diameter. Postembedding immunogold labeling showed that gold particles representing immunoreactivity for LH were densely located on all secretory granules of the secretory-specific cells. Many secretory granules, especially the large ones, of the cells were also loaded with immunogold particles for chromogranin A. Double immunogold labeling confirmed that LH and chromogranin A were colocalized on the same secretory granules. During embryonic development, the primordium of the pars tuberalis was first detected at 8 days of incubation as a small group of cells containing LH- and chromogranin-immunoreactive cells. In the pars distalis, the onset of LH and chromogranin expression occurred earlier, at 6 days of incubation. At 10 days of incubation, the pars tuberalis primordium became large cell masses consisting of LH- and chromogranin-immunoreactive cells, which were located close to the median eminence. Subsequently, the primordium extended along the median eminence progressively with age. At 14 days of incubation, it reached to the rostral end and surrounded the median eminence as slender cell cords. These results indicate that specific cells of the chicken pars tuberalis synthesize a glycoprotein hormone related to the LH molecule, which is stored in the secretory granules together with chromogranin A. The pars tuberalis may be involved in the regulation of gonadal function in a different way from that of the pars distalis. Accepted: 26 August 1997  相似文献

Summary N-acetyltransferase (NAT) activity in pineal glands exhibits a circadian rhythm with peak activity occurring in the dark-time. We previously showed that inGallus domesticus chicks pretreated with LD12:12, NAT activity was increased by dark exposure (peak dark sensitivity occurred during the expected dark-time) or decreased by light at night (peak light sensitivity occurred early in the night during the time of dark sensitivity). In this study we mapped dark sensitivity vs time (for NAT activity increase in response to 2 h dark pulses), and light sensitivity vs time (for NAT activity decrease in response to 10 min or 30 min light pulses) over a cycle for 3-week old chicks,Gallus domesticus, pretreated with long (LD16:8) or short photoperiod (LD8:16). Sensitivity to light was increased in the second 8 h after L/D by LD8:16. Abstract:  The influence of three photoperiods, viz. short day [8 : 16 h; light : dark (L : D)], long day (16 : 8 h; L : D) and continuous light (24 : 0 h; L : D) and four wavelengths of light, viz. white (control), yellow, blue and red on pre-imaginal development and reproductive performance of a ladybird, Coelophora (= Lemnia ) saucia (Mulsant) (Col., Coccinellidae) have been investigated under laboratory conditions. Long-day photoperiod and white light resulted in low immature stage mortality. Total larval period was longest under short-day and red-light conditions. Females kept at long-day photoperiod and under white light showed better reproductive performance than those placed under other photoperiods and wavelengths.  相似文献

The comparison of the hypophyseal pars tuberalis in active and in hibernating hedgehogs reveals different cytological characteristics in the specific secretory cells. In active animals, these cells show oval nuclei and the cytoplasm contains numerous secretory granules near or attached to the cell membrane, suggesting exocytotic release. In hibernating animals, the specific secretory cells are characterized by irregularly shaped and often invaginated nuclei. Clusters of secretory granules lying within the cytoplasm are often joined by lysosomes. These areas are observed to be encircled by cisterns of the endoplasmic reticulum. Degenerative processes in the structures sequestered in this way are interpreted as signs of crinophagy which has no equivalent in the cell types of the pars distalis.  相似文献

The importance of photoperiods in the regulation of annual testicular events in the carp Catla catla was evaluated by subjecting them to either long (16 h light : 8 h dark) or short (8 h light : 16 h dark) photoperiods for 30 days during the preparatory, prespawning, spawning and postspawning phases of an annual gonadal cycle. In each reproductive phase, testicular responsiveness to subjected photoperiods was determined by comparing the gonadal status in corresponding groups of control or natural photoperiodic fish. The values of testicular weight, gametogenic index, as well as testicular activity of two steroidogenic enzymes (Δ53β‐, and 17β‐hydroxysteroid dehydrogenase), and the serum titre of testosterone were considered as the indices of functional status of the testis in the fish concerned. During the prespawning phase, exposure of fish to a daily long photoperiod schedule resulted in precocious maturation of testis, while retardation of testicular growth was noted under the influences of short photoperiod. However, none of the employed photo‐schedules could influence the gametogenic and steroidogenic functions of the testis in the remaining part of the gonadal cycle. Collectively, the present study provides evidence for the first time that in the case of a commercially important carp, Catla catla, artificial, long photoperiods may be used for advanced testicular maturation, while reductions in maturation‐associated growth and deterioration in flesh quality may be avoided by submitting the fish to shorter day lengths during the prespawning phase of the reproductive cycle.  相似文献

The non-heterocystous cyanobacterium Oscillatoria sp. strain 23 fixes nitrogen under aerobic conditions. If nitrate-grown cultures were transferred to a medium free of combined nitrogen, nitrogenase was induced within about 1 day. The acetylene reduction showed a diurnal variation under conditions of continuous light. Maximum rates of acetylene reduction steadily increased during 8 successive days. When grown under alternating light-dark cycles, Oscillatoria sp. fixes nitrogen preferably in the dark period. For dark periods longer than 8 h, nitrogenase activity is only present during the dark period. For dark periods of 8 h and less, however, nitrogenase activity appears before the beginning of the dark period. This is most pronounced in cultures grown in a 20 h light – 4 h dark cycle. In that case, nitrogenase activity appears 3–4 h before the beginning of the dark period. According to the light-dark regime applied, nitrogenase activity was observed during 8–11 h. Oscillatoria sp. grown under 16 h light and 8 h dark cycle, also induced nitrogenase at the usual point of time, when suddenly transferred to conditions of continuous light. The activity appeared exactly at the point of time where the dark period used to begin. No nitrogenase activity was observed when chloramphenicol was added to the cultures 3 h before the onset of the dark period. This observation indicated that for each cycle, de novo nitrogenase synthesis is necessary.  相似文献

Summary Preembedding visualization of exocytosis by tannic acid treatment and postembedding immunocytochemical identification of cell types were combined to demonstrate the release of secretory products by exocytosis of characterized cell types. Treatment with tannic acid was carried out by perfusion with Ringer containing tannic acid, followed by fixation, dehydration and embedding. For electron microscopical immunocytochemistry protein A-gold was used as marker. In this study, exocytotic release was demonstrated for prolactin by cells in the pars distalis, and for oxytocin by axon terminals in the pars nervosa of the pituitary gland of the rat.  相似文献

Summary The effects of brief light pulses on photoperiodic reaction were examined in male Djungarian hamsters (Phodopus sungorus). Half of the animals came from long photoperiods and, accordingly, were in physiological summer state with large and functional testes. The other half had lived in short natural photoperiods before and was in physiological winter condition with involuted testes. These animals were exposed to long (LD 168) or to short (LD 816) photoperiods, or to short photoperiods in which the 16 h of darktime were interrupted by 1 min of light each night, either 4, 8 or 12 h after light off.Light pulses at midnight had the same effect as long photoperiods, they maintained gonadal size and activity or caused rapid recrudescence. If the light pulses were applied 4 or 12 h after light-off, however, a marked difference in effect ensued, depending on previous state and light regime. In animals coming from long photoperiods, these schedules induced regression, while in hamsters coming from short photoperiods the same schedules stimulated recrudescence, though significantly less than full long photoperiods or schedules with light-interruptions at midnight. Changes in body weight and pelage colour closely paralleled the changes in gonadal state.  相似文献

Locomotor activity rhythm was examined at various temperatures under a 16 h light : 8 h dark photoperiod (LD 16:8) or LD 12:12 using adults of the burying beetle Nicrophorus quadripunctatus. At 20°C, the locomotor activity of the beetles showed a bimodal daily pattern with two peaks around lights on and lights off under both photoperiods. This bimodal activity rhythm persisted under constant darkness; therefore, the activity of adult N. quadripunctatus is controlled by a circadian clock. Adults showed a bimodal activity pattern for temperatures ranging from 15 to 25°C. The evening peak of the daily activity was earlier at lower temperatures. These findings suggest that in the field, N. quadripunctatus shows crepuscular activity, and is active earlier in the afternoon in cooler seasons. In this species, therefore, temperature appears to play an important role in the determination of daily activity patterns.  相似文献

The harpacticoid copepod Tisbe holothuriae Humes was reared for several generations under different photoperiods (white light) and spectral quality (continuous illumination). The main life cycle parameters were measured and demographic variables were determined. Development time was retarded under green wavelengths and at photoperiods different from LD 12:12, especially under continuous white light. Total body length of females and males was shortest under LD 12:12 and longest under constant dark. Photoperiods different from LD 12:12 prolonged the interval between broods of egg sacs. Short photoperiods caused higher abortion rate, while long photoperiods retarded the maturation time of the egg sacs. Red wavelengths stimulated the abortion rate. Photoperiod LD 12:12 and blue wavelengths increased the production of offspring and the life span of females. Highest intrinsic rates of natural increase were estimated under LD 12:12 (rm =0.304) and blue wavelengths (rm = 0.254). These light conditions proved the most favourable for rearing Tisbe holothuriae.  相似文献
