Participation was studied of central serotonin receptors of the first and second types in behaviour change of animals selected by the character of defensive reaction to man. Serotonin receptors were determined by radioligand method by binding of the brain preparations 3H-serotonin and 3H-spiperone. An increase of C2 receptors number was found in the frontal brain cortex of the tame brown rats in comparison with the aggressive ones. Differences were not found in specific C1-receptor binding in the frontal brain cortex of tame and aggressive brown rats, silver foxes and American minks in various relatively early selection stages. It is supposed that disappearance of aggressive reaction to man at domestication is connected with an increase of C2 receptors number.  相似文献

Studies have been made on the density of receptors and dissociation constants for dopamine D1- and D2-receptors in the striatum, n. accumbens with the olfactory tubercles and in the frontal cortex of Wistar rats, Norway rats and silver foxes. D1 binding was found to be significantly higher than D2 one in all the analysed brain structures of the animals studied, especially in the striatum. Although the analysis of D1- and D2-receptor binding kinetics revealed differences in Wistar and Norway rats, more significant differences were found between rats and silver foxes.  相似文献

Studies have been made on the content of catecholamines (noradrenaline and dopamine) as well as metabolites of dopamine (3,4-dihydroxyphenylacetic and homovanillic acids) in the brain structures of silver foxes which differ in their handling reactions. The level of noradrenaline was found to be significantly higher in the anterior hypothalamus of domesticated animals; no differences in noradrenaline content were found in the frontal cortex, hippocamp, posterior hypothalamus and midbrain in animals from aggressive and domesticated groups. Dopamine content was higher in the tuberculum olfactorium of domesticated animals, being lower in the striatum and n. accumbens. Metabolite level remained unaffected which is presumably due to changes in dopamine synthesis in the investigated structures. It was concluded that domestication of animals favours the specimens with an altered state of catecholaminergic system of the brain.  相似文献

Selective agonist of 1A subtype of serotonine receptors ipsapirone inhibited manifestation of affective kinds of aggression in wild and domesticated rats. Administration of ipsapirone (10 mg/kg) decreased the number of aggressive attacks of wild and domesticated rats in the test of shock-induced aggression and blocked manifestation of defensive reaction to the experimenter in wild rats. Neophobia in wild rats decreased under the influence of ipsapirone. At the same time ipsapirone did not change mouse-killing behaviour either in wild or in domesticated rats. Probably, 5-HT1A receptors the aggressive regulate reaction, which are parts of the complex of defensive behaviour of the wild animals.  相似文献

The activity of tryptophan hydroxylase, the key enzyme of serotonin biosynthesis, was determined in the brain of silver foxes and wild rats selected, according to domestic or aggressive behavior, in respect to man. Significant increase of enzyme activity in midbrain of both domesticated rats and domesticated foxes was found, in comparison with that of aggressive animals. It was suggested that genetic mechanisms of the selection according to aggressive behavior, involve the changes of genes responsible for the synthesis of serotonin, the brain neurotransmitter which inhibits this type of behavior.  相似文献

Studies on 7-day-old and 8-month-old female silver foxes indicate that additional photostimulation affects to a various extent morphological and functional condition of the adrenal cortex in animals selected and unselected for the domestication type of behaviour. In relatively wild foxes, additional illumination stimulated the activity of the reticular zone, whereas in domesticated animals the same photoperiod increased the activity in the fascicular zone. It is suggested that selection of silver foxes for the domestication type of behaviour affects the sensitivity of the adrenals to external illumination.  相似文献

The review summarizes the results of long-term studies on the role of the brain mediator serotonin and genetic predisposition to various types of defensive behavior. The involvement of the serotonergic brain system in the mechanisms of genetic control of both active and passive defensive responses has been established using silver foxes, Norway rats of S40 selection for low and high aggressiveness to humans, aggressive mice with genetic knockout of monoaminoxidase A, and S40 rats selected for predisposition to passive defensive response of freezing (catalepsy). The changes in the serotonergic 5-HT1A-brain receptors of rats genetically predisposed to different strategies of defensive behavior were similar. However, the activity of the key enzyme of serotonin biosynthesis and the brain structures, in which serotonin metabolism was altered, significantly differed with regard to the preferred strategy. The conclusion was drawn that the 5-HT1A-receptors and enzymes of serotonin metabolism in the brain are involved in implementing genetic control of defensive behavior. Expression of the 5-HT1A-brain receptors was suggested to determine the levels of fear and anxiety and, consequently, the predisposition to defensive behavior, whereas the preferred strategy of defensive response (active or passive defensive) depends on genetically determined features of serotonin metabolism in the brain structures.  相似文献

The hypothesis was tested that one of the critical mechanisms underlying genetically determined aggressiveness involves brain serotonin 5-HT(1A)-receptors. The expression of 5-HT(1A)-receptor mRNA in brain structures and functional correlate for 5-HT(1A)-receptors identified as 8-OH-DPAT-induced hypothermia were studied in Norway rats bred over the course of 59 generations for the low and high affective (defensive) aggressiveness with respect to man and in highly aggressive (offensive) MAO A-knockout mice (Tg8 strain). Considerable differences between the aggressive and the nonaggressive animals were shown. Agonist of 5-HT(1A)-receptor 8-OH-DPAT (0.5 mg/kg for rats and 2.0 mg/kg for mice, i.p.) produced a distinct hypothermic reaction in nonaggressive rats and mice and did not affect significantly the body temperature in aggressive animals. In aggressive rats, a significant reduction of the expression of 5-HT(1A)-receptor mRNA was found in the midbrain. In Tg8 mice, 5-HT(1A)-receptor mRNA level was increased in the frontal cortex and amygdala and not changed in the hypothalamus and the midbrain. The results provide support for the idea that brain 5-HT(1A)-receptors contribute to the genetically determined individual differences in aggressiveness.  相似文献

5-HT receptors are predominantly located in the brain and are involved in pancreatic function and cell proliferation through sympathetic nervous system. The objective of this study was to investigate the role of hypothalamic 5-HT, 5-HT1A and 5-HT2C receptor binding and gene expression in rat model of pancreatic regeneration using 60% pancreatectomy. The pancreatic regeneration was evaluated by 5-HT content, 5-HT1A and 5-HT2C receptor gene expression in the hypothalamus of sham operated, 72 h and 7 days pancreatectomised rats. 5-HT content was quantified by HPLC. 5-HT1A receptor assay was done by using specific agonist [3H]8-OH DPAT. 5-HT2C receptor assay was done by using specific antagonist [3H]mesulergine. The expression of 5-HT1A and 5-HT2C receptor gene was analyzed by RT-PCR. 5-HT content was higher in the hypothalamus of 72 h pancreatectomised rats. 5-HT1A and 5-HT2C receptors were down-regulated in the hypothalamus. RT-PCR analysis revealed decreased 5-HT1A and 5-HT2C receptor mRNA expression. The 5-HT1A and 5-HT2C receptors gene expression in the 7 days pancreatectomised rats reversed to near sham level. This study is the first to identify 5-HT1A and 5-HT2C receptor gene expression in the hypothalamus during pancreatic regeneration in rats. Our results suggest the hypothalamic serotonergic receptor functional regulation during pancreatic regeneration.  相似文献

The contribution of genetic and environmental components in phenotype variety of corticosteroid adrenal function was studied in undomesticated and domesticated silver foxes during postnatal ontogenesis. The variation of basal and stress plasma corticosteroid level in animals aged 2, 4, 6, 8 months and in vitro secretion of the adrenal gland at the age 8 months was analysed. Significant genotype-depended variability was only demonstrated in undomesticated foxes under stress stimulation conditions. This phenomenon is manifested from the fourth month of life. However, significant genotype-depended variability was already revealed under basal conditions in domesticated foxes, on initial steps of postnatal ontogenesis. The peculiarities of genetic variability of adrenocortical function in foxes selected for domestication are discussed.  相似文献

The study deals with the mechanisms that bring about a directional asymmetry in the expression of some morphological traits observed in some animals subjected to experimental domestication. The key role in the integration of development is attributed to the genetic systems controlling the activity of brain neurotransmitter systems. Therefore, the investigation of directional asymmetry of morphological traits began with the analysis of interhemispheral differences in neurotransmitter activity in animal lines selected for domestic and aggressive behavior. Experiments on silver foxes reveal interhemispheral differences in the dopaminergic system emerging in the striatum. An increased dopamine level is observed in the right half of the striatum of aggressive foxes and in both right and left halves of the striata of domestic foxes. On the basis of the literature data, it is suggested that the considerable increase in the dopamine level in the right halves of the striata of both aggressive and domestic animals is related to a genetic increase in the manifestation of emotional response in both lines, whereas its increase in the left half of the striatum of domesticated foxes may be related to a correlated deterioration of the function of the pituitary-adrenal system.  相似文献

Domestication affects behavioral and vocal responses, involved in communication with humans; in particular, those that attract human attention. In this study, we found that silver foxes of Tame strain, experimentally domesticated for a few tenses of generation, displayed bursts of vocal activity during the first minute after appearance of an unfamiliar human, that faded quickly during the remaining time of the test, when the experimenter stayed passively before the cage. Distinctively, foxes of Aggressive strain, artificially selected for tenses of generation for aggressive behavior toward humans, and the control group of Unselected for behavior silver foxes kept steady levels of vocal activity for the duration of the tests. We found also that Aggressive foxes vocalized for a larger proportion of time than Unselected foxes for all 5 min of the test. We discuss the obtained data in relation to proposal effects of domestication on mechanisms directed to involving people into human-animal interactions and structural similarity between human laughter and vocalization of Tame foxes.  相似文献

1.Rat hypothalamic 5-hydroxytryptamine (5-HT) and 5-hydroxyindole acetic acid (5-HIAA) concentrations are transiently sexually differentiated in the second week postpartum (pp), with higher levels in the female. In this report we investigate the possibility that 5-HT receptors may also exhibit sexual dimorphism in the neonatal period.2.5-HT1A and 5-HT2A receptors were quantitated by radioligand binding of [3H]ketanserin and [3H]8-OH DPAT, respectively, in hypothalamus and amygdala from male and female rats at days 8–16 pp.3.There was no sexual dimorphism or change in the density of 5-HT2A binding in hypothalamus or amygdala over days 8–16 pp. There was also no sexual dimorphism of 5-HT1A receptors.4.There was an increase in 5-HT1A receptor density in both the hypothalamus and the amygdala. In the hypothalamus, but not the amygdala, this increase was interrupted on day 14 by a decrease in 5-HT1A receptors, which we suggest may be of physiological significance in modifying the eventual pattern of adult agonistic activity.5.The results suggest that the sexual dimorphism in 5-HT turnover is predominantly presynaptic, relating to altered synthesis and/or release, and is not of sufficient magnitude or duration to produce adaptive responses in postsynaptic 5-HT1A or 5-HT2A receptors.  相似文献

Two groups of silver foxes, selected according their behaviour with respect to man and non-selected ones, were exposed to restriction-induced stress. It was found that changes in the level of brain serotonin and its main metabolite, 5-hydroxyindolacetic acid, and elevation of plasma corticosteroids concentration in domesticated (tame) silver foxes were much less pronounced than in non-domesticated (non-tame) animals. Positive relationship between type of behaviour, brain serotonin metabolism and pituitary-adrenal axis response to stress was observed. It is suggested that such correlative pattern as changed pituitary-adrenal axis responses in domesticated animals may be due to changes in metabolism of controlling this axis brain transmitter serotonin.  相似文献

This paper concentrates on involvement of protein elements in the brain neurotransmitter serotonin system (key enzymes in serotonin metabolism and 5-HT(1A) receptors) in the genetic control of behaviour. The data were obtained using Norway rats selected for more that 50 generations for lack of aggressive response and for aggressive behaviour towards humans (fear-induced aggression), inbred mouse strains, and MAO A knockout mice. The review provides converging line of evidence that: 1) brain serotonin contributes to critical mechanism underlying genetically defined individual differences in aggressiveness, and 2) genes encoding pivotal enzymes in serotonin metabolism (tryptophan hydroxylase, MAO A) and 5-HT(1A) receptors belong to a group of genes that modulate aggressive behaviour.  相似文献

Functional state of pineal gland in female Norway rats selected for tame and aggressive behavior was analyzed using stereometric technique. Activity of epiphysis in tame rats (the study was performed in November, during diestrus stage) was evaluated higher than in aggressive rats. 5-HT1 receptors of the amygdala, midbrain's central gray substance, hippocampus and frontal cortex of the rats were investigated during the memory trace retrieval, using the model of "psychogenic" amnesia. It was found that specific binding of 3H 5-HT in amygdala and central gray substance was decreased in the rats with passive avoidance retention. 3H 5-HT binding in the amnesic rats didn't differ from the control animals. Absence of decrease of 5-HT1 receptors number could be considered as one of the possible mechanisms of the memory trace non-retrieval due to amnesia or its consequence.  相似文献

We studied the effects of selective agonists of 5-HT1A receptors 8-OH-DPAT and flesinoxan on aggressive behavior of C57BL/6 male mice in the "resident-intruder" test and on defensive aggression of Norway rats toward man. 8-OH-DPAT (0.4 mg/kg, i.p.) significantly reduced the intermale aggression in mice and defensive aggression in rats (0.1-0.5 mg/kg, i.p.). In the dose of 0.5 mg/kg, flesinoxan inhibited the aggressive behavior in mice. These results suggest that activation of 5-HT1A receptors reduces different kinds of affective aggression. The results are discussed in terms of interaction between the well-known anxiolytic effects of 5-HT1A agonists and their antiaggressive properties.  相似文献

Domestication has led to similar changes in morphology and behavior in several animal species, raising the question whether similarities between different domestication events also exist at the molecular level. We used mRNA sequencing to analyze genome-wide gene expression patterns in brain frontal cortex in three pairs of domesticated and wild species (dogs and wolves, pigs and wild boars, and domesticated and wild rabbits). We compared the expression differences with those between domesticated guinea pigs and a distant wild relative (Cavia aperea) as well as between two lines of rats selected for tameness or aggression towards humans. There were few gene expression differences between domesticated and wild dogs, pigs, and rabbits (30–75 genes (less than 1%) of expressed genes were differentially expressed), while guinea pigs and C. aperea differed more strongly. Almost no overlap was found between the genes with differential expression in the different domestication events. In addition, joint analyses of all domesticated and wild samples provided only suggestive evidence for the existence of a small group of genes that changed their expression in a similar fashion in different domesticated species. The most extreme of these shared expression changes include up-regulation in domesticates of SOX6 and PROM1, two modulators of brain development. There was almost no overlap between gene expression in domesticated animals and the tame and aggressive rats. However, two of the genes with the strongest expression differences between the rats (DLL3 and DHDH) were located in a genomic region associated with tameness and aggression, suggesting a role in influencing tameness. In summary, the majority of brain gene expression changes in domesticated animals are specific to the given domestication event, suggesting that the causative variants of behavioral domestication traits may likewise be different.  相似文献
