转基因抗虫棉根系分泌物对棉花黄萎病菌生长的影响   总被引:4,自引:0,他引:4  
转基因抗虫棉的抗病性下降已成为制约我国棉花生产的重要因素之一.以转基因抗虫棉及其亲本非转基因棉花对照为材料,研究转基因抗虫棉根系分泌物对棉花黄萎病菌孢子萌发和菌丝生长的影响,并对其根系分泌物中氨基酸及糖类的组成和含量进行了测定.结果表明:与亲本非转基因棉相比,2种转基因抗虫棉对棉花黄萎病菌的抗性下降,转基因抗虫棉的根系分泌物对黄萎病菌孢子萌发和菌丝生长均具有促进作用.与亲本常规棉中23相比,转基因双价抗虫棉中41根系分泌物中多了甲硫氨酸和赖氨酸,并且天冬氨酸、谷氨酸、丝氨酸、丙氨酸、缬氨酸、亮氨酸、酪氨酸的含量显著升高.泗棉三号与单价抗虫棉GK12的根系分泌物中虽然含有相同的氨基酸组分,但GK12的根系分泌物中酪氨酸、缬氨酸、亮氨酸的含量显著下降.中41根系分泌物中检测出4种糖类,而中23中仅检测出葡萄糖.抗虫棉GK12和泗棉三号根系分泌物中都检测出4种糖类,但其含量差异明显.  相似文献

Summary From root exudates of three cultivars of chilli (Capsicum annuum L.) 12 amino acids and 7 sugars were detected. Methionine, d-1- phenylalanine, citrulline and d-xylose were detected only from the root exudates of resdistant cultivars. The root exudates of resistant variety inhibited spore germination of the pathogen (Fusarium oxysporum f. sp.capsici), but that of susceptible variety enhanced spore germiantion of the same. Spore germiantion of antagonistic fungi (Trichderma viride andAspergillus sydowi) was also influenced by the root exudates of resistant and susceptible varieties, but the influence was different.Spore germiantion of a number of rhizosphere fungi was studied and in general root exudate of susceptible cultivar enhanced spore germiantion of majority of fungi, but spore germination of antagonistic fungi against the pathogen was inhibited. However, root exudate of resistant cultivar stimulated spore germination of antagonistic fungi.  相似文献

Summary Seed and root exudates from three varieties of both broad bean and cotton grown aseptically were analyzed for amino acids and sugars. Broad bean varieties were found to excrete considerable amounts of amino compounds with 15 different amino acids, while cotton varieties excreted less consisting of 12 amino acids. Five sugars were identified in the seed and root exudates from broad bean varieties, while three were present in the seed and root exudates from cotton varieties. Seed and root exudates stimulated spore germination and growth of fungi isolated from the rhizosphere.  相似文献

Root exudates have a key role in communication between plants and microbes in the rhizosphere. Fusarium wilt of watermelon, caused by Fusarium oxysporum f. sp. niveum (Fusarium oxysporum), drastically reduces watermelon yields in continuous cultivation systems, but it can be significantly alleviated using watermelon/aerobic rice intercropping system as shown by the research carried out in this laboratory. It is important to evaluate the interaction between root exudates from the two crops and the pathogen and thus to clarify the mechanism of disease suppressiveness in the intercropping system. The effects of phenolic acids, sugars and free amino acids in root exudates from watermelon (REW) and rice (RER) on the growth of Fusarium oxysporum were studied. The results obtained are listed as follows: (1) REW significantly increased spore germination and sporulation, whereas RER had inhibitory effects on those two parameters. (2) HPLC analysis showed that salicylic acid, p-hydroxybenzoic acid and phthalic acid were identified in exudates from both plants, but p-coumaric acid was only detected in rice and ferulic acid only in watermelon. Moreover, of the total rice exudates a high proportion (37.9 %) of p-coumaric acid was detected and the total amount of phenolic acids was 1.4-fold as high as that in watermelon. (3) Considerable differences in the components and contents of both sugars and amino acids were found between REW and RER exudates. (4) Exogenously applied alanine (Ala) increased spore germination and sporulation. In contrast, addition of exogenous p-coumaric acid reduced spore germination and sporulation, relative to controls. It was concluded that the rice root exudates had anti-fungal properties while that from watermelon promoted pathogen growth. This discovery provided a scientific basis for practicing watermelon/aerobic rice intercropping to control Fusarium wilt in watermelon.  相似文献

Summary Root exudates from healthy and diseased lentil plants (Lens culinaris) have been investigated in relation to the wilt disease caused byFusarium oxysporum f.lentis. In all ten amino acids and five sugars have been detected. The spore germination of the pathogen in root exudates indicated that 21-days root exudate was inhibitory. Glycine and phenylalanine were detected in 21-days exudate and were found to have an inhibitory effect upon the germination of the spores of the pathogen which may partly explain the lesser disease incidence when plants of more than 3 weeks are inoculated with the pathogen.  相似文献

Summary Ninetten aminoacids, twelve sugars, eleven organic acids and ten phenols were detected in the leaf exudates of three cultivars of chilli. The number of aminoacids, sugars, organic acids and phenols increased as the plants grew older. More aminoacids and sugars were detected in the exudate from the susceptible cultivar (Malwa). More organic acids and phenols were detected from the resistant cultivar (Simla). The leaf exudate of the resistant cultivar (Simla) inhibited spore germination of the pathogen (Alternaria solani) while that of susceptible (Malwa) stimulated spore germination. The cultivar ‘Patna’ which is moderately resistant, occupied an intermediate position. Spore germination of the isolated fungi was enhanced in leaf exudate of susceptible cultivar (Malwa), while leaf exudates of the moderately resistant (Patna) and resistant (Simla) inhibited spore germination of the majority of fungi isolated. Most of the antagonistic fungi were not isolated from the susceptible cultivar and the percentage spore germination of these fungi was less in leaf exudate of the susceptible cultivar, while leaf exudates of resistant cultivars enhanced the percentage spore germination of antagonistic fungi,viz Aspergillus flavus, A. fumigatus, A. versicolor, Penicillium citrinum, P. restrictum andTrichoderma viride.  相似文献

In earlier studies, biological control of Fusarium wilt of cucumber induced by Fusarium oxysporum f. sp. cucumerinum was demonstrated using nonpathogenic strains C5 and C14 of Fusarium oxysporum. Strain C14 induced resistance and competed for infection sites whether roots were wounded or intact, whereas strain C5 required wounds to achieve biocontrol. In the current work, additional attributes involved in enhanced resistance by nonpathogenic biocontrol agents strains to Fusarium wilt of cucumber and pea were further investigated. In pre-penetration assays, pathogenic formae specials exhibited a significantly higher percentage of spore germination in 4-day-old root exudates of cucumber and pea than nonpathogens. Also, strain C5 exhibited the lowest significant reduction in spore germination in contrast to strain C14 or control. One-day-old cucumber roots injected with strain C14 resulted in significant reduction in germ tube orientation towards the root surface, 48–96 h after inoculation with F. o. cucumerinum spores, whereas strain C5 induced significantly lower spore orientation of the pathogen and only at 72 and 96 h after inoculation. In post-penetration tests, passive transport of microconidia of pathogenic and nonpathogens in stems from base to apex were examined when severed plant roots were immersed in spore suspension. In repeated trials, strain C5, F. o. cucumerinum and F. o. pisi were consistently isolated from stem tissues of both cucumber and pea at increasing heights over a 17 days incubation period. Strain C14 however, was recovered at a maximum translocation distance of 4.6 cm at day 6 and later height of isolation significantly declined thereafter to 1.2 cm at day 17. In pea stem, the decline was even less. Significant induction of resistance to challenge inoculation by the pathogen in cucumber occurred 72 and 96 h after pre-inoculation with biocontrol agents. Nonetheless, strain C14 induced protection as early as 48 h and the maximum resistance was reached at 96 h. The presented data confirm the previous findings that attributes important for nonpathogenic fusaria to induce resistant are: rapid spore germination and orientation in response to root exudate; active root penetration and passive conidia transport in stem to initiate defence reaction without pathogenicity and enough lag period between induction and challenge inoculation. Strain C14 possesses all these qualifications and hence its ability to enhance host resistance is superior than strain C5.  相似文献

Abstract

In this study we assessed microconidia germination of the tomato pathogens F. oxysporum f. sp. lycopersici (Fol) and F. oxysporum f. sp. radicis-lycopersici (Forl) in the presence of root exudates. Tomato root exudates stimulated microconidia germination and the level of stimulation was affected by plant age. Treatment of root exudates with insoluble polyvinylpolypyrrolidone, which binds phenolic compounds, indicated that tomato root exudates contain phenolic compounds inhibitory to F. oxysporum microconidia germination. Our study indicates that tomato root exudates similarly stimulate microconidia germination of both Fol and Forl. However, individual F. oxysporum strains differ in the degree of germination response to the root exudates. Furthermore, root exudates from non-host plants also contain compounds that stimulate microconidia germination of Fol. In general, the effects of root exudates from non-host plants did not differ considerably from those of tomato. The ability of phenolic compounds to inhibit germination of Fol seems not to be plant-specific.  相似文献

Summary Cotton-field soils from a wilt-free zone (KPT) and from a wilt-sick zone (PLD), and root exudates of diploid and amphidiploid cotton strains grown in KPT and PLD soils were analyzed for 13 free amino acids employing microbiological assay techniques. Test organisms used wereLactobacillus arabinosus 17/5 andLeuconostoc mesenteroides P. 60.Arginine, proline, lysine, serine, glycine, cystine, and tyrosine occurred in both the soils; amounts of these were higher in PLD soil than in KPT soil.Seven to eleven amino acids were found in the root exudates of cotton. Aspartic acid, threonine (in one), phenylalanine and leucine were found in root exudates but not in the control soils. Diploid strains were found to exude greater amounts of amino acids than the amphidiploid strains. Comparing the effect of the two soils tested, it was found that amino acids in exudates of plants grown in PLD soil were higher except in the case of one diploid strain.In the inoculated series, amino acids in the exudates were markedly lower than the respective healthy controls in the diploid strains. In the exudates of amphidiploid strains also, amino acid contents were reduced but to a lesser extent.Significance of occurrence of cystine in the exudates of some strains is indicated.Part of Doctoral thesis, University of Madras.  相似文献

Arbuscular mycorrhizal fungi (AMF) can control soilborne diseases such as Fusarium oxysporum f.sp. lycopersici (Fol). Root exudates play an important role in plant–microbe interactions in the rhizosphere, especially, in the initial phase of these interactions. In this work, we focus on (i) elucidating dynamics in root exudation of Solanum lycopersicum L. in an intercropping system due to AMF and/or Fol; (ii) its effect on Fol development in vitro; and (iii) the testing of the root exudate compounds identified in the chromatographic analyses in terms of effects on fungal growth in in vitro assays. GC‐MS analyses revealed an AMF‐dependent increase in sugars and decrease in organic acids, mainly glucose and malate. In the HPLC analyses, an increase in chlorogenic acid was evident in the combined treatment of AMF and Fol, which is to our knowledge the first report about an increase in chlorogenic acid in root exudates of AM plants challenged with Fol compared with plants inoculated with AMF only, clearly indicating changes in root exudation due to AMF and Fol. Root exudates of AMF tomato plants stimulate the germination rate of Fol, whereas the co‐inoculation of AMF and Fol leads to a reduction in spore germination. In the in vitro assays, citrate and chlorogenic acid could be identified as possible candidates for the reduction in Fol germination rate in the root exudates of the AMF+Fol treatment because they proved inhibition at concentrations naturally occurring in the rhizosphere.  相似文献

Sterile root exudates from wilt susceptible and wilt resistant pea cultivars showed no differential effects on spore germination of Fusarium oxysporum Schl. f.pisi (Linf.) Snyd. & Hans, races 1 and 2 which could be correlated with the pathogenicity of a particular isolate to a given cultivar. Uniformly high percentages of germination were obtained with conidia of the two races in aseptic shake culture with exudates collected from resistant or susceptible plants of various ages. Chlamydospores of the two races incubated with exudates under sterile conditions germinated to uniformly high levels irrespective of exudate origin. Conidia and chlamydospores of Fusarium solani (Mart.) Sacc. f. pisi (Jones) Snyd. & Hans., used for comparative purposes, also germinated to high levels in the presence of exudate solutions of all cultivars. Non-specific germination of the two races of F. oxysporum f. pisi occurred in soil when the exudates were supplied to populations of chlamydospores via diffusion units. Germination was lower than that recorded under sterile conditions and was rapidly followed by germling lysis.  相似文献

Summary The amounts of amino acids in seed exudates were generally higher than in root exudates of the same plant. The spectra and relative abundance of amino acids in both plants were similar but they were generally more abundant in cowpea exudates than in sorghum. Glutamic acid and alanine were the most abundant amino acids in the seed and root exudates of both plants. The proportions of the amino acids in the seed exudates were comparable to that stored in the seeds. Many of the major amino acids identified in the exudates were also found to support thein vitro growth ofFusarium spp. isolated from the rhizosphere and rhizoplane. This suggests that the amino acids exuded might contribute signficantly to Fusaria nutrition and its consequent predominance around the root. 盐碱胁迫下植物根系分泌物包含丰富生化信息并具有重要生态作用。为了探讨耐盐碱牧草湖南稷子(Echinochloa frumentacea) 在盐碱胁迫下根系分泌物组成,揭示其在盐碱胁迫下的生理及生态作用,以湖南稷子为试验对象,在人工气候室开展水培试验,并在苗期分别进行中性盐(NaCl+Na2SO4 100 mmol/L)、碱性盐(NaCl+NaHCO3 100 mmol/L)和碱(Na2CO3+NaHCO3 50 mmol/L)处理。在处理3 d后,利用液质联用仪(LC-MS/MS)检测对照组和处理组根系分泌物的化合物成分。结果表明,盐碱胁迫下湖南稷子根系分泌物共有334种化合物。依据正交偏最小二乘法判别分析(OPLS、|DA),重要值(VIP)得分及t检验的P值, 发现对照比SaSo100(碱性盐处理 100 mmol/L),对照比Soda50(碱处理50 mmol/L)和对照比Salt100(中性盐处理100 mmol/L)分别有22、15和21个差异根系分泌物。其中碱性盐和碱处理下根系分泌物组成相近,包括脂质、酚酸,生物碱,苯酞类,氨基糖,萜类,醌类,氨基酸及其衍生物;中性盐处理下有脂质、酚酸,生物碱,苯酞类,萜类。京都基因与基因组百科全书注释及富集发现,盐碱胁迫下根系分泌物不仅含有三羧酸循环代谢产生的碳水化合物、核苷酸,氨基酸,脂肪酸,类脂和维生素等物质,而且与瓦博格效应、膜运输,信号传导以及遗传信息处理等途径有关。研究表明,湖南稷子通过根系分泌物渗出,调节自身代谢物浓度,加强或改变碳同化、呼吸作用、信号传导等提高对盐碱胁迫的适应性。  相似文献

以5种对枯萎病不同抗性黄瓜品种为试材,对其根系分泌物氨基酸组分进行测定,并对氨基酸组分与黄瓜品种枯萎病抗病性之间的相关性进行了分析。结果表明:在中抗品种根系分泌中检测到的16种氨基酸:半胱氨酸Cys、苏氨酸Thr、丙氨酸Ala、缬氨酸Val、异亮氨酸Lle、天冬氨酸Asp、亮氨酸Leu、苯丙氨酸Phe、甘氨酸Gly、甲硫氨酸Met、组氨酸His、谷氨酸Glu、酪氨酸Tyr、赖氨酸Lys、丝氨酸Ser和精氨酸Arg。其中的精氨酸在感病品种中没有被检出,组氨酸和精氨酸组分在抗病品种中没有被检出。根系分泌物中总氨基酸含量随品种抗性的增加而降低;精氨酸、丝氨酸和赖氨酸的含量与品种对枯萎病的病情指数呈负相关,其他13种氨基酸组分含量与品种对枯萎病的病情指数呈正相关,其中苯丙氨酸含量与病情指数呈显著正相关。丝氨酸与苯丙氨酸、天冬氨酸、丙氨酸、甘氨酸的比值均与品种对枯萎病的病情指数呈显著负相关,其中Ser/Phe与品种对枯萎病的病情指数呈极显著负相关。  相似文献

Summary Extract from onion bulbs and diffusate from roots of onion seedlings were fractionated by column chromatography. The stimulatory effects of the different fractions of onion extract on sclerotial germination ofSclerotium cepivorum were studied. The sugar fraction was the most stimulatory, whereas, the amino acid fraction was not effective. Paper chromatographic analysis revealed the presence of glucose, fructose and no amino acids in the root diffusate. These two sugars and 13 amino acids were identified in the onion extract. When various sugars and amino acids were supplied individually to autoclaved soil, only glucose, fructose, mannose and maltose effectively induced sclerotial germination. Partial stimulation occured in nonsterile soil amended with high glucose concentrations. Studies on the antibiotic effect of the different fractions against some soil fungi by the spore germination method showed that, the sugar fraction inhibits completely the spore germination of all the fungi, tested, whereas, the amino acid fraction was non-inhibitory. Both fractions did not show antibiotic activity when tested by the filter paper disc method. Attempts to extract inhibitory substances from soil which inhibit sclerotial germination were unsuccessful. It was suggested that onion extract plays a twofold role in stimulating sclerotial germination in natural soil: (a) a direct nutritional influence; (b) an antibiotic effect on soil mycoflora which reduces competition for nutrients.  相似文献

The influence of boron starvation on the root exudates content in soybean seedlings (Glycine max. L. Merr.) and the effect of exudates pretreatment on the pre-infection processes in symbiotic system Br. japonicum strain 636 and soybean were investigated. Root cell membrane stability of boron starved soybean plants (-B) decreased compared to the control. The concentrations of all analyzed metabolites (reducing sugars, free amino acids, organic acids, soluble phenols and total flavonoids) from root exudates of -B plants were lower than the control concentrations. Analysis of polyphenols after HPLC chromatography of root exudates showed significant difference of peak numbers between chromatograms of exudates obtained from boron starved and from control plants. Bacterial culture treatment with root exudates from -B plants showed decreased growth, chemotaxis and attachment ability toward the host root compared to the control exudate treatments. These changes were accompanied by decreased nodulation and acetylene reduction activity of boron starved soybean plants.  相似文献

Root colonization by arbuscular mycorrhizal (AM) fungi reduces stimulation of seed germination of the plant parasite Striga (Orobanchaceae). This reduction can affect not only host plants for Striga, resulting in a lower parasite incidence, but also false hosts or trap crops, which induce suicidal Striga seed germination, thereby diminishing their effectiveness. In order to better understand these AM-induced effects, we tested the influence of root colonization by different AM fungi on the seed-germination activity of root exudates of the Striga hermonthica nonhost plants cowpea and cotton on S. hermonthica. We also tested the effect of AM fungi on the seed-germination activity of the Striga gesnerioides host plant cowpea on S. gesnerioides. Moreover, we studied whether mycorrhization affects the transport of seed-germination activity to above-ground plant parts. Mycorrhization not only resulted in a lower seed germination of S. gesnerioides in the presence of root exudates of the S. gesnerioides host cowpea but also seed germination of S. hermonthica was also lower in the presence of root exudates of the S. hermonthica nonhosts cowpea and cotton. Downregulation of the Striga seed-germination activity occurs not only in root exudates upon root colonization by different AM fungi but also in the compounds produced by stems. The lowered seed-germination activity does not appear to depend on the presence of seed germination inhibitors in the root exudates of mycorrhizal plants. The implication for Striga control in the field is discussed.  相似文献

Verticillium wilt is a plant vascular disease caused by the soilborne fungus Verticillium dahliae that severely limits cotton production. In a previous study, we screened Bacillus cereus YUPP-10, an efficient antagonistic bacterium, to uncover mechanisms for controlling verticillium wilt. Here, we report a novel antimicrobial cyclodextrin glycosyltransferase (CGTase) from YUPP-10. Compared to other CGTases, six different conserved domains were identified, and six mutants were constructed by gene splicing with overlap extension PCR. Functional analysis showed that domain D was important for hydrolysis activity and domains A1 and C were important for inducing disease resistance. Direct effects of recombinant CGTase on V. dahliae included reduced mycelial growth, spore germination, spore production, and microsclerotia germination. In addition, CGTase also elicited cotton's innate defence reactions. Transgenic Arabidopsis thaliana lines that overexpress CGTase showed higher resistance to verticillium wilt. Transgenic CGTase A. thaliana plants grew faster and resisted disease better. CGTase overexpression enabled a burst of reactive oxygen species production and activated pathogenesis-related gene expression, indicating that the transgenic cotton was better prepared to protect itself from infection. Our work revealed that CGTase could inhibit the growth of V. dahliae, activate innate immunity, and play a major role in the biocontrol of fungal pathogens.  相似文献
