研究了通过共享同一寄主卵,短管赤眼蜂Trichogramma pretiosum体内共生的Wolbachia被水平传递到拟澳洲赤眼蜂T. confusum体内后,Wolbachia对新宿主拟澳洲赤眼蜂的影响。结果表明: Wolbachia的侵染能使拟澳洲赤眼蜂进行不完全的产雌孤雌生殖,增加拟澳洲赤眼蜂子代雌性比例,但却导致了雌蜂寿命缩短和繁殖力降低的生理损失。Wolbachia感染的当代处女蜂及其建立的种群连续5代(F1~F5),其子代雌蜂百分率分别为79.17%、76.60%、68.66%、72.58%、68.15%和64.06%,基本上呈现出逐代降低的趋势,并越来越接近对照的63.85%。处女蜂及F1~F5代雌蜂的平均寿命分别为4.33、5.50、5.60、6.68、7.32和7.50天,而未感染交配雌蜂寿命为7.59 天;处女蜂及F1~F5代雌蜂平均产卵量分别为11.33、70.00、86.41、93.90、102.92和124.38粒/雌,除F5代外,均显著低于未感染交配雌蜂的产卵量134.81粒/雌。用四臂嗅觉仪测定了Wolbachia新宿主拟澳洲赤眼蜂对寄主小菜蛾的嗅觉反应,结果表明Wolbachia的侵染具轻微干扰拟澳洲赤眼蜂嗅觉反应的负面影响。未感染Wolbachia的拟澳洲赤眼蜂及Wolbachia供体短管赤眼蜂对寄主小菜蛾具较强的嗅觉反应,其雌蜂在小菜蛾腹部鳞片正己烷提取液和小菜蛾卵表正己烷提取液处理区内滞留时间显著或极显著长于对照区。而感染Wolbachia的拟澳洲赤眼蜂F2代和F3代雌蜂,尽管在小菜蛾腹部鳞片正己烷提取液处理区内滞留时间比对照区长,但没有达到显著水平;其F2代雌蜂在小菜蛾卵表正己烷提取液处理区内滞留时间与对照区相比,也没有达到显著水平。随Wolbachia在拟澳洲赤眼蜂种群中垂直感染代数的增加,拟澳洲赤眼蜂对寄主小菜蛾的嗅觉反应恢复正常,在Wolbachia感染后的 F4~F6代,雌蜂在两种提取液处理区内的滞留时间均显著或极显著长于对照区。  相似文献

为了了解阿维菌素和哒螨灵对土耳其斯坦叶螨Tetranychus turkestani Ugarov et Nikolski实验种群的亚致死效应, 为新疆土耳其斯坦叶螨的综合防治提供理论依据, 我们在室内通过叶碟饲养的方法, 利用生命表技术研究了阿维菌素和哒螨灵亚致死剂量对土耳其斯坦叶螨实验种群成螨和卵的影响。结果表明: 阿维菌素和哒螨灵LC20、LC10剂量处理成螨后, 可使成螨的产卵量、平均寿命和卵孵化率显著降低; 卵期、幼螨期、若螨期和产卵前期明显延长, 而成螨期和雌螨寿命又明显低于对照; 对次代种群的影响表现在净生殖率(R0)、 周限增长率(λ)降低、生存率和平均每雌日产卵率明显降低, 内禀增长率(rm)由0.37降低至0.17~0.29, 平均世代历期(T)除阿维菌素LC20处理时长于对照外, 其他处理均低于对照, 种群倍增时间(Dt)延长。两种药剂亚致死剂量处理卵后, 内禀增长率(rm)由0.32降低至0.11~0.22, 净生殖率(R0)降低, 平均世代历期(T)和周限增长率(λ)降低, 而种群加倍时间(Dt)增长; 幼螨期、若螨期和产卵前期明显长于对照, 成螨期和雌螨寿命显著低于对照; 生存率和平均每雌日产雌率明显降低。这些结果证明, 在亚致死剂量下, 阿维菌素和哒螨灵能够降低土耳其斯坦叶螨种群的发育速率, 这对土耳其斯坦叶螨的综合防治策略的制定有积极意义。  相似文献

为了了解共生菌Cardinium对朱砂叶螨Tetranychus cinnabarinus (Boisduval)的生殖调控作用,从检测到感染Cardinium的种群中选取山西运城和甘肃兰州2个感染率较高的种群作为实验材料,对Cardinium在寄主生殖调控方面的作用进行研究。通过人工培养100%感染Cardinium的种群和100%不感染的种群,并从中挑取不同感染类型的雌雄螨单独进行杂交实验,设立感染Cardinium(♀)×感染Cardinium(),感染Cardinium(♀)×不感染(),不感染(♀)×感染Cardinium(),不感染(♀)×不感染() 4种杂交组合。结果发现,抗生素处理后不感染的雌螨和受感染雄螨的交配表现出了单向的胞质不亲和,而且2个种群表现出了2种类型:在山西种群不亲和的组合中,CI表现为卵孵化率显著降低,多数雌螨都产生出F1雌螨后代,但是F1代雌螨数量相对于其他3种组合明显减少; 而在甘肃种群不亲和的组合中则表现为卵全部孵化且幼螨100%存活,但全部为雄螨后代。2个种群中Cardinium的亲缘关系非常相近,而它们却能够在不同的地理种群中表现出不同的CI表型,这提示亲缘关系非常接近的品系也可以产生不同的表型。研究结果为深入揭示Cardinium的感染机制和生殖调控作用提供重要基础。  相似文献

共生细菌Wolbachia对宿主的生殖起多种调控作用。以往研究表明, Wolbachia基因组中广泛存在插入序列(insertion sequence, IS), 它们对宿主基因组的可塑性、 多样性和进化起重要作用。稻水象甲Lissorhoptrus oryzophilus Kuschel在东亚是一种外来水稻害虫, 在原产地北美营两性生殖, 而在所有入侵地均营孤雌生殖。本研究采用PCR法从河北唐海孤雌生殖型稻水象甲体内克隆获得了Wolbachia的2条IS序列, 即ISWosp4和ISWosp6; 从美国德克萨斯州两性生殖型稻水象甲成虫体内克隆获得了Wolbachia的2条IS序列, 即ISWosp3和ISWosp5。碱基序列比对显示: ISWosp3和ISWosp4属于IS3家族IS3组成员, ISWosp5为IS4家族IS231组成员, ISWosp6为IS5家族IS1031组成员。对这些IS的ORF结构、 所编码氨基酸序列的结构等进行了分析, 推测ISWosp5具有潜在转座活性。所得结果增进了我们对Wolbachia IS3, IS4和IS5家族插入序列的认识, 同时为今后从IS的角度探讨Wolbachia与稻水象甲生殖的关系奠定了基础。  相似文献

根据毕赤酵母Pichia pastoris密码子偏爱性,不改变毒素蛋白质一级结构,设计合成了昆虫神经毒素BjαIT基因,并分别克隆至大肠杆菌Escherichia coli融合表达载体pPET30-a(+)和毕赤酵母分泌表达载体pPIC9K。在IPTG的诱导下,神经毒素在大肠杆菌中融合表达,表达产物利用镍亲和层析柱纯化,纯化产物用于制备抗血清和活性测试。采用斑点杂交,筛选得到了较高水平分泌表达重组BjαIT的酵母转化子,摇瓶条件下,毒素表达量最大可达约20 mg/L。大肠杆菌BjαIT表达产物对东亚飞蝗Locusta migratoria manilensis和德国小蠊Blattela germanica没有活性,但酵母表达产物经注射东亚飞蝗和德国小蠊表现出杀虫活性。  相似文献

棉铃虫P450基因CYP6AE12和CYP9A18的克隆与mRNA表达水平   总被引:2,自引:1,他引:1  
采用RT-PCR和RACE技术克隆到2个新的棉铃虫细胞色素P450基因:CYP6AE12CYP9A18CYP6AE12的cDNA编码区长1 569 bp,编码523个氨基酸;CYP9A18的cDNA编码区长1 590 bp,编码530个氨基酸。用实时定量PCR技术分析了这2个基因在棉铃虫YS敏感品系和YS-FP抗性品系(由氰戊菊酯加辛硫磷混剂筛选YS品系而得) 6龄幼虫脂肪体和中肠中mRNA的表达水平。结果表明:CYP6AE12CYP9A18的mRNA表达具有组织特异性,CYP6AE12在脂肪体中表达量较高,而CYP9A18在中肠中的表达量较高。与相对敏感品系YS相比,CYP6AE12在YS-FP抗性品系中肠和脂肪体中的mRNA表达量分别为YS品系的3.6倍和1.3倍;CYP9A18在YS-FP品系中肠和脂肪体的mRNA表达量分别为YS品系的0.3倍和1.0倍。CYP6AE12的过量表达与YS-FP品系棉铃虫的抗药性可能有一定关系。  相似文献

不同杀虫剂选育对家蝇抗药性水平及kdr基因频率的影响   总被引:4,自引:0,他引:4  
采用杀虫剂(溴氰菊酯和甲基嘧啶磷)筛选及不接触药物自然衰退的方法,研究了家蝇Musca domestica氯氟氰菊酯高抗品系(Cyh-R)对杀虫剂的抗性变化,探讨蝇类抗药性治理的方法。用点滴法测定氯氟氰菊酯对不同家蝇品系的毒力,比较抗药性的变化,结合特异性等位基因PCR扩增(PASA)技术检测了不同家蝇品系的kdr基因频率,探讨kdr基因频率与抗性水平之间的关系。结果表明:甲基嘧啶磷筛选后,氯氟氰菊酯对第2~8代Cyh-R品系的LD50呈递减趋势,从F0的2.8434 μg/蝇降为F8的0.4404 μg/蝇,但第8~18代Cyh-R品系的LD50呈逐代递增趋势;溴氰菊酯筛选后,氯氟氰菊酯对Cyh-R品系第2~16代的LD50呈上升趋势,从F0的2.8434 μg/蝇升至F16的24.3249 μg/蝇;表明了施用有机磷杀虫剂可降低其对氯氟氰菊酯的抗药性,而施用拟除虫菊酯药剂则有助于其对氯氟氰菊酯抗药性的增长;不使用任何杀虫剂也能降低其对氯氟氰菊酯的抗药性,但下降速率低于甲基嘧啶磷。PASA技术检测表明Cyh-R品系的kdr抗性基因频率为88.8%,不经过任何药剂筛选其kdr抗性基因频率下降程度最大,达到69.7%;甲基嘧啶磷筛选后其结果降为78.8%,而经溴氰菊酯筛选后kdr抗性基因频率则明显升高,达到98.9%。通过对kdr抗性基因频率和抗性水平进行相关和回归分析表明kdr抗性基因频率与家蝇对氯氟氰菊酯的LD50呈对数关系,即LD50值高的品系其kdr抗性基因频率相应的也较高。建议在家蝇防治中考虑轮换用药。  相似文献

家蚕细胞色素P450基因Bmcyp6u1的克隆、序列分析与表达谱   总被引:3,自引:0,他引:3  
细胞色素P450第6亚家族基因为昆虫所特有,与抗性相关。为了检测家蚕Bombyx mori cyp6u1基因是否与耐氟性相关,首先克隆了cyp6u1基因。采用生物信息学方法获得与黑腹果蝇Drosophila melanogaster cyp6u1基因同源的家蚕B. mori cyp6u1基因序列, 预测该序列的开放阅读框(ORF)为1 476 bp, 编码491个氨基酸, 推定的蛋白质分子质量为56.15 kD, 等电点为9.23。以家蚕5龄第3天幼虫精巢cDNA为模板, 设计特异引物PCR扩增出一条约1 500 bp的条带, 大小与家蚕cyp6u1序列的ORF预测值接近, 命名为Bmcyp6u1基因(GenBank登录号:HM130560)。同源性分析表明, Bmcyp6u1基因与蜜蜂Apis mellifera的同源基因cyp6AS13的相似性为56%, 与拟南芥Arabidopsis thalianacyp72A82的相似性为48%, 与人Homo sapienscyp3A7基因的相似性为50%。芯片数据分析表明, Bmcyp6u1基因在家蚕5龄第3天幼虫各组织表达量很低, 只在精巢组织(5龄第3天)稍有表达, 推测该基因具有组织特异性。  相似文献

The discontinuous gas exchange cycle (DGC) was described in the German cockroach, Blattella germanica (L.) (Dictyoptera: Blattellidae) for the first time. Also, the effect of the DGC on water loss was investigated. The CO(2) emission pattern in both insecticide resistant and susceptible B. germanica varied with temperature. At 10, 15, and 20 degrees C the pattern was discontinuous. Cycle frequency increased at 25 and 30 degrees C, and at 35 degrees C the pattern became cyclic. In most DGCs, there was no clear distinction between the closed and flutter phases in both strains thus data for these phases were combined and analyzed as the interburst phase. The probability that B. germanica would breath discontinuously varied with temperature. Most cockroaches (62.8%) displayed DGCs at 10 degrees C, therefore measurement of metabolic rate and water loss was carried out at this temperature. Using repeated measures of analysis of variance, the interburst and burst V(.)(CO(2))(ml h(-1)) were not significantly different between the two strains. The variability in CO(2) emission during the interburst and burst phases over time was not significantly different from cycle to cycle or between strains. Overall metabolic rate during the entire recording was not significantly different between both strains. There was a significant difference in the duration of the interburst and burst phases between the strains. The susceptible strain had significantly longer interburst and burst phase durations during a complete DGC than the resistant strain. The interburst and burst phase durations were 5.01+/-0.19 and 6.21+/-0.13 min, respectively, for the resistant strain, whereas the durations were 7.16+/-0.37 and 6.73+/-0.17 min, respectively, for the susceptible strain. This resulted in a DGC of significantly longer duration (13.89+/-0.44 min) in the susceptible strain compared with the resistant strain (11.23+/-0.26 min). The duration of the interburst phase was significantly different from the open phase duration in the resistant strain such that during a single DGC lasting approximately 11.23 min, 43.5% consisted of the interburst phase while the burst phase made up 56.5% of the cycle. The cuticular permeability at 10 degrees C and 0% RH was 2.26 microg cm(-2) h(-1) mmHg(-1) for the resistant strain and 3.42 microg cm(-2) h(-1) mmHg(-1) for the susceptible strain. In both strains, cuticular transpiration accounted for approximately 95% of total water loss. The significantly longer duration of the interburst phase of the susceptible strain was not important in reducing water loss.  相似文献

【目的】在前期研究高温胁迫下意大利蝗Calliptamus italicus呼吸代谢的变化特征基础上,为探明相关变化机制,本研究进一步从不连续气体交换循环(DGC)呼吸周期历时阐明其对高温胁迫的响应机制。【方法】以源自新疆伊犁察布查尔蝗区野生种群的意大利蝗成虫为材料,应用多通道昆虫呼吸仪测定意大利蝗DGC暴发间期、暴发期和呼吸周期三者历时随温度变化(21~42℃区间,3℃温度梯度)的响应特征。【结果】雌雄意大利蝗DGC呼吸周期中暴发间期历时随温度升高而缩短,21℃时暴发间期历时最长,雌雄分别为7.29±0.31和7.05±0.37 min,与24,27,30和33℃差异不显著(P0.05),超过33℃后迅速缩短;至39℃时最短,雌雄分别为0.42±0.01和0.70±0.03 min,与其他温度下的历时差异显著(P0.05)。暴发期历时亦随温度升高而缩短,21和39℃时,雌虫暴发期历时分别达到最长和最短,依次为1.55±0.14和0.45±0.01 min,与其他温度下的历时差异显著(P0.05);24和39℃时,雄虫暴发期历时分别达到最长和最短,依次为1.61±0.10和0.68±0.02 min,与其他温度下的历时差异显著(P0.05)。随温度升高,意大利蝗DGC呼吸周期历时呈减少趋势,且主要由暴发间期历时减少所致。21~33℃范围内,每升高3℃雌雄虫暴发间期历时分别平均减少0.174和0.121 min,DGC呼吸频率增幅一致,均为0.0022 Hz,不同温度间差异不显著(P0.05);超过33℃后,每升高3℃雌雄虫暴发间期历时分别平均减少2.197和2.189 min,DGC呼吸频率分别增加0.0120和0.0085 Hz,不同温度间差异显著(P0.05);39℃时暴发间期历时接近零,42℃时DGC呼吸模式消失。【结论】随温度升高,意大利蝗呈现出O2吸收量和CO2释放量增加的变化特征,这是通过减少暴发间期历时和增加DGC呼吸频率等机制而实现的。  相似文献

Cuticular hydrocarbon (CH) profiles of four Periplaneta species were compared and their role in aggregation and interspecific recognition was evaluated. CH profiles are species specific and include from 19 to 25 hydrocarbons. P. brunnea, P. fuliginosa and P. australasiae have more CH components in common with one another than with P. americana. P. americana hydrocarbons include components from 24 to 43 carbon atoms but hydrocarbons for the three other species range from 21 to 41 atoms. The major compound in CH profiles in P. americana is 6,9-heptacosadiene that is absent from the three other species. The major compound in CH profiles of P. Brunnea and P. fuliginosa is 13-methyl pentacosane that is not found in the CH profile of P. americana. Major compounds in P. australasiae species are tricosene and 13-methyl pentacosane, respectively, in males and females. Binary choice tests demonstrated that specific CH extracts induced aggregation in the four species. High CH amounts were required to induce aggregation of P. americana and P. brunnea on conditioned sites, whereas low amounts were sufficient to induce aggregation of P. fuliginosa and P. australasiae. These results suggest that CHs are involved in aggregation and interspecific recognition.  相似文献

Sexual dimorphism in the distribution of antennal sense organs is common among adults of the genus Periplaneta. In three out of the four strains of Periplaneta americana examined, adult males had more contact chemoreceptors than females. In the fourth strain of P. americana and in P. australasiae, P. brunnea, P. fuliginosa, and P. japonica, no statistically supportable sexual dimorphism of contact chemoreceptors was found. However, in all strains and species of Periplaneta examined, sexual dimorphism was found in the total number and/or density of olfactory sensilla. Male adults had nearly twice as many olfactory sensilla as female adults. These observations are consistent with the behavioral observation that males within the genus Periplaneta rely on the reception of an airborne pheromone for the initiation of courtship behavior. In P. americana, where sexual dimorphism was found in the contact chemoreceptors, contact stimuli release the full wing raising display and presentation in males during courtship.  相似文献

Toxicity of fipronil to German and American cockroaches   总被引:2,自引:0,他引:2  
Topical and oral toxicity of fipronil, compared to chlorpyrifos, was determined for the German cockroach, Blattella germanica (L.), and American cockroach, Periplaneta americana (L.). Fipronil and Combat bait matrices were evaluated for their attractancies to both species. In the topical toxicity tests, LD50's of fipronil, at 72 h after topical application, were 0.03 and 0.02 µg/g for adult B. germanica and P. americana, respectively. Fipronil was significantly more toxic than topically applied chlorpyrifos (LD50's were 0.06 and 0.16 µg/g for B. germanica and P. americana, respectively). The oral toxicity of fipronil and chlorpyrifos in Petri dish experiments, against both species, was affected by stage (for B. germanica), diet concentration, and feeding assay. Fipronil caused higher mortality of B. germanica than chlorpyrifos in two feeding assays (continuous and abbreviated). Both compounds were equally toxic to adult males of P. americana at all rates. Fipronil caused higher nymphal mortality than chlorpyrifos 48–72 h after exposure in both feeding assays. In large population chamber tests, fipronil bait was more effective and faster in killing P. americana than Raid and Combat. LT50's were 0.8, 2.4, and 7.6 d for fipronil, Raid (a.i. = chlorpyrifos), and Combat (a.i. = hydramethylnon) baits, respectively. Mortality reached 96.5, 93.4, and 84.6%, respectively, at the end of the 14 d test. In the bait attractancy tests, both strains of B. germanica were attracted similarly to fipronil and Combat bait matrices. P. americana were attracted more to fipronil than to Combat bait matrix or to other alternative foods.  相似文献

The discontinuous gas exchange cycle (DGC) of the pseudoscorpion Garypus californicus is characterized by periodic bursts of CO(2) emission and by high rates of interburst CO(2) emission. We investigated the mechanism that triggers the burst phase by manipulating ambient oxygen partial pressures (Po(2)). The ventilatory trigger in most land animals is hypercapnia; in insects, for example, the burst phase is triggered when endotracheal Pco(2) reaches about 4 kPa. In insects with a DGC, hypoxia induces prolonged interburst phases because spiracular conductance is elevated to supply oxygen to the tissues, thus delaying the onset of the hypercapnia-triggered burst phase because CO(2) accumulates more slowly. In G. californicus, hypoxia induced a decrease in interburst phase length, while hyperoxia increased its duration relative to normoxia. This is opposite to the condition in insects. In addition, CO(2) emission fell during the interburst phase as ambient Po(2) rose, also opposite to the condition in insects. Thus, the burst phase is triggered in G. californicus (and presumably in other pseudoscorpions) not by hypercapnia but by hypoxia, a situation that is seldom encountered in terrestrial animals.  相似文献

Sclerotized oothecae from four species of cockroaches, Periplaneta americana, P. fuliginosa, Blatta orientalis and Blattella germanica, were examined by solid-state 13C-nuclear magnetic resonance and chemical analyses. The oothecae were composed of protein, water, calcium oxalate, diphenolic compounds, lipid, and uric acid. Calcium oxalate was the major soluble component in egg cases of P. americana, P. fuliginosa, and B. orientalis. Oothecae of B. germanica had approx. 10-fold less calcium oxalate and extractable diphenols than the other species. The major diphenolic compound extracted in cold dilute perchloric acid was 3,4-dihydroxybenzoic acid. Exuviae from P. americana, B. germanica, Gromphadorhina portentosa, Blaberus craniifer, and Leucophaea maderae also were examined by solid-state 13C-NMR. They contained protein, diphenols, and lipid, as well as chitin, which accounted for 30–42% of the organic content, depending upon the species.  相似文献

To assess the roles of the German cockroach (Blatella germanica) and the American cockroach (Periplaneta americana) in the transmission of Sarcocystis muris and of 3 other coccidia of cats-Toxoplasma gondii, Isospora felis, and Isospora rivolta, cockroaches exposed to feces containing these coccidia were periodically fed to mice, as was a portion of the fecal matter. Sarcocystis muris sporocysts, which in feces remained infectious for at least 20 days, were also transmitted to mice by P. americana for at least 20 days and by B. germanica for 5 days after exposure to infectious feces. Toxoplasma gondii oocysts were transmitted by P. americana intermittently up to 10 days, but by B. germanica only immediately after exposure to feces. Oocysts of 2 species of Isospora, when associated with fecal matter, remained infectious for 20 days. Those of I. rivolta were transmitted by both cockroach species for 10 days, but I. felis was transmitted only by by B. germanica, and for only 2 days.  相似文献

The LT50s for the German cockroach ranged from 4.4 to 1.7 d for males, from 9.0 to 2.4 d for females, and from 4.4 to 1.6 d for nymphs for bait concentrations of abamectin between 0.0025 and 0.0500%. The LC50s of abamectin were 0.0110 and 0.0040% from males, 0.0240 and 0.0090% for females, and 0.0200 and 0.0080% for nymphs at 3 and 6 d, respectively. The LT50 values of 0.0550% abamectin bait were 3.4, 3.4, 2.4, 7.5, 2.9, and 4.5 d for Periplaneta americana (L.), P. fuliginosa (Serville), P. brunnea Burmeister, P. australasiae (F.), Blatta orientalis L., and Supella longipalpa (Serville). Although the bait was effective against various cockroach species, time to death for the larger species was longer than for the German cockroach. In preference tests in which male German cockroaches were allowed to feed on rat chow or abamectin bait, all died within 5 d of exposure to abamectin bait. Abamectin bait consumption was not significantly lower than that of untreated rat chow. Arena tests with 0.0550% abamectin bait resulted in 31-75% mortality of German cockroaches after 9 d, with most control being achieved by treating harborages with the bait. The hydramethylnon standard resulted in 65% mortality after 9 d.  相似文献
