To elucidate the role of ethylene in the production of flavor compounds by tomato fruits, wild-type tomato (Lycopersicon esculentum L., cv. Lichun) and its transgenic antisense LeACS2 line with suppressed ethylene biosynthesis were used. The metabolism of individual sugars was ethylene-independent. However, citric acid and malic acid were under ethylene regulation. The content of these acids was higher in transgenic tomato fruits and returned to normal level after transgenic fruits were treated with ethylene. Because most of amino acids, which are important precursors of volatiles, were shown to be correlated with ethylene, we surmise that amino acid-related aroma volatiles were also affected by ethylene. Headspace analysis of volatiles showed a significant accumulation of aldehydes in wild-type tomato fruits during fruit ripening and showed a dramatic decrease in most aroma volatiles in transgenic tomato fruits as compared with wild-type fruits. The production of hexanal, hexanol, trans-2-heptenal, cis-3-hexanol, and carotenoid-related volatiles, except β-damascenone and β-ionone, was inhibited by suppression of ethylene biosynthesis. No remarkable differences were observed in the concentrations of cis-3-hexenal and trans-2-hexenal between transgenic and wild-type tomato fruits, indicating these two volatiles to be independent of ethylene. Recognition and acceptance of a suitable host plant by phytophagous insects requires the integration of visual, physical and chemical cues. The present study investigates the host cues that a specialist insect integrates to optimize oviposition decisions and whether these cues are weighted in a specific way. The study also determines whether the tomato fruit borer Neoleucinodes elegantalis (Guenée) (Lepidoptera: Crambidae), an important pest on Solanaceae in Brazil, shows a preference for oviposition sites that differ in physical and/or chemical cues. When styrofoam balls are provided as artificial fruits, N. elegantalis deposit significantly more eggs on rough artificial fruits than on smooth ones. Hexane fruit extracts applied to the artificial fruits stimulate female oviposition strongly. Physical and chemical cues also affect the oviposition of females when offered together. Furthermore, certain parts of the artificial fruits are prefered, irrespective of the presence of chemical cues. Both physical and chemical cues affect oviposition decisions; hence, the fruit borer relies on cues of different sensory modalities.  相似文献

Ethylene at 10 and 100 μl/litre stimulated germ-tube elongation of Botrytis cinerea spores incubated within normal and non-ripening nor tomato fruits, but had little influence on the total percent of germination. Values of germ-tube length within the mature-green normal fruits and the mature-green or mature nor fruits were similar to those recorded within the normal mature fruits when held in air. Exposure of the normal and the mutant fruits to 100 μl/litre ethylene immediately after inoculation with B. cinerea insignificantly increased lesion development, but resulted in increased sporulation. When tomato fruits were exposed to ethylene for 3 days before inoculation a marked stimulatory effect on rot development was exhibited on the mature-green normal fruits but not on the nor mutant fruits. The results indicate that exogenous ethylene may directly stimulate germ tube growth of B. cinerea in both normal and mutant fruit, but that it may affect subsequent fungal growth indirectly, via stimulation of the ripening process, only in preclimacteric normal tomato fruit.  相似文献

Gas exchanges, chlorophyll (Chl) a fluorescence and carboxylation activities of ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO) and phosphoenolpyruvate carboxylase (PEPC) were determined in tomato (Lycopersicon esculentum Mill.) fruits picked at different developmental stages (immature, red-turning, mature, and over-ripe). The fruits did not show signs of CO2 fixation. However, photochemical activity was detectable and an effective electron transport was observed, the values of Chl fluorescence parameters in green fruits being similar to those determined in the leaves. The RuBPCO activity, which was similar to those recorded in the leaves at the immature stage of the fruit, decreased as the fruit ripened. PEPC activity was always higher than RuBPCO activity.  相似文献

A greenhouse test was carried out to examine the effects on tomato growth of application of purple non-sulfur bacterium Rhodopseudomonas sp. which had enhanced germination and growth of tomato seed under axenic conditions. The shoot length of tomato plant inoculated by Rhodopseudomonas sp. KL9 increased by 34.6% compared to that of control in 8 weeks of cultivation. During the same period, this strain increased 120.6 and 78.6% of dry weight of shoot and root of tomato plants, respectively. The formation ratio of tomato fruit from flower was also raised by inoculation of KL9. In addition, Rhodopseudomonas sp. KL9 treatment enhanced the fresh weight and lycopene content in the harvested tomato fruits by 98.3 and 48.3%, respectively compared to those of the uninoculated control. When the effect on the indigenous bacterial community and fate of the inoculated Rhodopseudomonas sp. KL9 were monitored by denaturing gradient gel electrophoresis analysis, its application did not affect the native bacterial community in tomato rhizosphere soil, but should be repeated to maintain its population size. This bacterial capability may be applied as an environment-friendly biofertilizer to cultivation of high quality tomato and other crops including lycopene-containing vegetables and fruits.  相似文献

Low doses of the exogenous flavonoid quercetin increased the content of sugars in tomato fruits of various cultivars. The content of glucose in tomato fruits of cv.Ukrainskii teplichnyi increased from 3.62 to 11.24% per unit dry weight. Increases in the content of glucose were found in all tomato cultivars examined. An analysis of qualitative and quantitative compositions of amino acids showed that their levels were markedly decreased in fruits of quercetin-treated plants. Our studies and data found in the literature suggest that this effect is due to the synthesis of sugars from amino acids by gluconeogenesis. The reverse process of sugar hydrolysis does not occur because exogenous quercetin inhibits the activity of pyruvate kinase.  相似文献

B. cinerea and C. atramentarium rotted wound-inoculated green tomato fruits and wounded or intact ripe fruits while G. cingulata developed rots only in ripe fruits. Pectic en-zymes were extracted from the fruit tissue rotted by B. cinerea and C. atramentarium but no pectic enzymes attributable to the fungus were detected in ripe fruits rotted by G. cingulata. G. cingulata produced endo-PG and endo-PL in vitro, C. atramentarium produced endo-PL in vitro and in vivo and B. cinerea produced exo-PG in vitro and in green fruits but endo-PG and endo-PL in ripe fruits. Well ripened tomato fruits contained high levels of endogenous PG. All three fungi produced proteolytic enzymes in vitro and in vivo. Proteases produced by G. cingulata and C. atramentarium had optimum activity at pH 9 to 10 and were not trypsin-like or chymotrypsin-like in nature. Protease produced by B. cinerea had optimum activity at pH 7 and showed both trypsin and chymotrypsin-like activity. Proteins extracted from the cell walls of tomato fruits inhibited both the endo-PG and endo-PL produced by G. cingulata and the endo-PL produced by B. cinerea but did not in-hibit the activity of PGs produced by B. cinerea, the endo-PL produced by C. atramentarium or the endogenous PG from tomato fruits. The cell wall proteins also contained trypsin and chymotrypsin inhibitor activity which inhibited 70 % of the activity of the protease produced by B. cinerea, but had little effect on the proteases produced by G. cingulata, C. atramentarium or the tomato endogenous protease. Enzymes produced in vitro by G. cingulata macerated green tomato tissue more slowly than enzymes produced in vitro by C. atramentarium and B. cinerea and the rate of mation was further reduced in the presence of added cell wall proteins. Excess inhibitor of the little effect on the rate of maceration by the enzymes produced by C. atramentarium of the cinerea.  相似文献

We present a complementary resource for trait fine‐mapping in tomato to those based on the intra‐specific cross between cultivated tomato and the wild tomato species Solanum pennellii, which have been extensively used for quantitative genetics in tomato over the last 20 years. The current population of backcross inbred lines (BILs) is composed of 107 lines derived after three backcrosses of progeny of the wild species Solanum neorickii (LA2133) and cultivated tomato (cultivar TA209) and is freely available to the scientific community. These S. neorickii BILs were genotyped using the 10K SolCAP single nucleotide polymorphism chip, and 3111 polymorphic markers were used to map recombination break points relative to the physical map of Solanum lycopersicum. The BILs harbor on average 4.3 introgressions per line, with a mean introgression length of 34.7 Mbp, allowing partitioning of the genome into 340 bins and thereby facilitating rapid trait mapping. We demonstrate the power of using this resource in comparison with archival data from the S. pennellii resources by carrying out metabolic quantitative trait locus analysis following gas chromatography–mass spectrometry on fruits harvested from the S. neorickii BILs. The metabolic candidate genes phenylalanine ammonia‐lyase and cystathionine gamma‐lyase were then tested and validated in F2 populations and via agroinfiltration‐based overexpression in order to exemplify the fidelity of this method in identifying the genes that drive tomato metabolic phenotypes.  相似文献

Inoculations with both Botrytis cinerea and Geotrichum candidum stimulated ethylene evolution in the pre-climacteric normal tomato fruit and the non-ripening nor mutant which did not show any rise in ethylene when uninfected. In the post-climacteric normal fruits, new peaks in ethylene production were formed. The rise in ethylene evolution in all types of infected fruits has already been detected during the incubation period of the disease. Ethylene peaks were detected earlier and were higher in fruits infected with B. cinerea than with G. candidum, coinciding with the faster rate of growth of the former. Mechanical wounding also stimulated ethylene synthesis by the non-ripening fruits, production being directly proportional to wound dimension. Considerably higher rates of ethylene were recorded for infected fruits than for mechanically-injured fruits in which wound dimensions were similar to those of lesion development. Applying aminoxyacetic acid at the site of inoculation inhibited ethylene production by 55–60 % in the normal fruits and by about 80 % in the nor mutant fruits. A similar pathway of ethylene synthesis was suggested for normally ripening tomato fruit and non-ripening infected tissues.  相似文献

Enzymes of Botrytis cinerea were detected in vitro using various carbon sources. Pectin-pectate as a sole carbon source induced both polygalacturonase (PG) and pectin lyase (PL) activity, whereas carboxymethylcellulose served as an inducer for cellulase (Cx) activity. PG activity appeared earlier than Cx activity when induced by their respective sources. Both PG and PL activities were detected earlier and their level was higher on cell walls of the normal tomato fruit, than of the nor mutant, and in each case activity was higher on cell walls of the mature fruits than of the mature-green ones. Whereas relatively high rates of PG and PL activity were recorded on autoclaved tomato homogenate (TH) of both the normal and the nor fruits, only trace levels of PG activity were recorded on unautoclaved media, except for those prepared from ripe normal fruits, and no PL activity was detected on either of the unsterilized media. Botrytis-infection resulted in PG activity in the enzyme-less rin and nor mutant fruits at both stages of maturity and in the normal and hybrid fruits at their mature-green stage. In the ripe normal and hybrid fruits, infection increased the level of PG activity recorded prior to inoculation. An association was drawn between the low PG activity recorded in the nor mutant and its hybrid at initial stages of invasion and their resistance to infection. Following infection an increase in the level of Cx activity over that recorded in healthy fruits was found in all the tomato genotypes, whereas no PL was recorded in either healthy or infected fruits.  相似文献

Alcobaca is commonly regarded as an abnormally ripening mutant of the tomato (Lycopersicon esculentum Mill.). Alcobaca fruits were found to be similar to cv. Rutgers fruits in the following characteristics: time between full anthesis and the onset of ripening, response to ethephon, flavor, pH and concentrations of titratable acids, total soluble solids and reducing sugars. The pattern of CO2 and ethylene climacteric are similar in the two plant types, but the peak levels were lower and occurred later in alcobaca than in ‘Rutgers’. The mutant fruits differed from fruits of normal varieties in their greatly prolonged shelf life, their relatively low activity of polygalacturonase (PG) and polymethylgalacturonase (PMG), and their low level of endogenous ethylene. Fruits of the mutant harvested before the onset of ripening failed to reach normal pigmentation and remained yellow. Fruits harvested at the onset of ripening reached an orange color, while fruits ripened while attached to the plant reached almost normal pigmentation. These results suggest that alcobaca is a slow ripening mutant and does not belong to the category of non-ripening mutants.  相似文献
