采用水蒸气蒸馏法和气体吸附法提取黑杨萎蔫叶片的挥发性气味物质,用二氯甲烷做萃取剂,气相色谱_质谱联用技术分离和鉴定萃取到的气味物质的化学组分,结果表明,水蒸气蒸馏法共得到19个峰,鉴定出结构的有16个峰,未鉴明的3个峰;主要组分为4号峰的(Z)_3_己烯醇,相对含量为28.71%,其次为14号峰的邻羟基苯甲醛,相对含量为10.35%。气体吸附法共分离到20个峰,鉴定出结构的有17个峰;主要组分分别为6、7、8号峰的(E)_2_己烯醛、(Z)_3_己烯醇、(E)_2_己烯醇,相对含量分别为20.78%、29.12%、29.22%。两种方法所得到的相同组分为9种,相似性为46.15%。 相似文献

目的:研究巴戟天中的挥发性成分。方法:利用水蒸气蒸馏法提取巴戟天(Radix morindae offtcinalis)挥发油,用GC—MS进行测定,结合计算机检索技术对分离的化合物进行结构鉴定,应用色谱峰面积归一化法计算各成分的相对百分含量。结果:鉴定出34个化学成分,其中相对百分含量大于2%的分别确定为L-龙脑(Bomeol L)29.28%,α-姜烯(Alpha—Zingiberene)4.88%,2-甲基-6-对甲基苯基-2-庚烯(Ar-Cureumene)4.49%,1-己醇(1-Hexanol)3.40%,β-倍半水芹烯(beta—sesquiphellandrene)3.34%,2-戊基呋喃(2-Amylfuran)3.32%,正壬醛(n—nonanal)2.17%,樟脑(L—camphor)2.07%,β-没药烯(bete—Bisabolene)2.06%。结论:34个挥发性成分均为首次从该植物中得到。 相似文献

目的:研究柔毛路边青(Herba gei)挥发油成分,为有效控制中成药蓝止安脑胶囊质量及中药二次开发提供理论依据。方法:利用水蒸气蒸馏法提取柔毛路边青挥发油,用GC/MS进行分离测定,结合计算机检索技术对分离的化合物进行结构鉴定,应用色谱峰面积归一化法计算各成分的相对百分含量。结果:分离出59个成分,确定了43个化合物,占挥发油总量的87.51%。结论:柔毛路边青挥发油成分大于5%的为α-蒎烯10.73%,松金娘烷醇13.63%,松金娘烯醛7.19%,松金娘烯醇5.03%,二环[3,1,1,]-6,6-二甲基-3-亚甲基庚烷6.83%,丁子香酚15.82%。其中含量最高的物质为丁子香酚。 相似文献

利用水蒸气蒸馏法提取小花清风藤中的挥发性成分,通过气相色谱质谱联用技术(GC-MS)并结合计算机分析鉴定提取物中挥发油芳香成分,共鉴定出17种化合物,其中有12种烷烃类化合物,1种芳香类化合物,1种醚类化合物,1种杂环类化合物,1种环戊二烯和1种酰胺类化合物。用面积归一化法测定挥发油中各种成分的相对含量,合计占总峰面积的8.8%。实验结果表明:用水蒸气蒸馏提取小花清风藤挥发油时,提取率为4.18%,其最主要成分为烷烃类化合物。 相似文献

采用顶空吸附法对健康松、衰弱松的针叶进行了活体挥发性物质收集。通过GC-MS联用技术和计算机信息检索法对两种状态下松树的针叶挥发物化学成分进行了分离鉴定,用离子流峰面积归一法通过化学工作站数据处理系统确定了各成分的相对含量。比较结果表明:云南松由健康松转向衰弱松的过程中挥发性成分及含量发生了很大变化。两种状态下针叶挥发性物质中共有15种相异化合物成分,18种相同化合物成分,相同化合物成分在含量上存在很大差异。 相似文献

以番茄(Lycopersicon esculentum)叶为材料.分别采用溶媒萃取、水蒸气蒸馏、气体吸附法收集挥发性物质;气相色谱-质谱联用技术分离鉴定挥发组分。结果表明,3种收集方式共获得19种挥发性组分.其中的14种组分相同.且多为主要组分。水蒸气蒸馏法获得的挥发性物质总含量最高,为27.13μg FW;其次是气体吸附法.为23.38μg FW,溶媒萃取法获得的最少,仅为12.40μg FW。溶媒萃取法共获得15种挥发组分.含量最高的组分为β-水芹烯。水蒸气蒸馏法获得18种组分.含量最高的组分为(E)-2-己烯醛。气体吸附法获得17种组分.含量最高的组分为(E)-2-己烯醛。不同方法所获各组分的相对含量和绝对含量均不同.可根据研究目的综合分析运用。 相似文献

The chemical components of the volatiles from Vetiveria zizanioides were analyzed by SPME and GC-MS. In the roots, the main component was valencene (30.36%), while in the shoots and leaves, they were 9-octadece-namide (33.50 % ), 2, 6, 10, 15, 19, 23-hexamethyl-2, 6, 10, 14, 18, 22-tetracosahexaene (27.46% ), and 1,2-benzendicarboxylic acid, diisooctyl ester(18.29% ). The results showed that there were many terpenoids in the volatils. In shoot volatiles, there existed 3 monoterpenes, 2 sesquiterpenes and 1 triterpene. Most of the volatiles in roots were sesquiterpenes. 相似文献

Cuttings of Populus nigra L. cv. Loenen and Populus maximowiczii Henri × Populus nigra L. cv. Rochester were exposed for 6 weeks in open-top chambers to investigate effects of realistic mixtures of ozone, sulphur dioxide, and nitrogen oxides on carbohydrate levels. Whereas the total main nonstructural carbohydrates in the leaves were reduced, those in the roots were nearly not affected. The reduction in leaf carbohydrates was associated with a significant decrease in starch. In contrast, sucrose and especially glucose were increased, causing a shift in the starch to sucrose and the starch to glucose ratios. The effects were strongest in the older leaves of both cultivars, with cv. Loenen, especially, responding to ozonic mixtures. In the petioles, sucrose and glucose were unchanged or, in some cases, decreased. The alterations in carbohydrate levels were coupled to a reduction in both total fresh weight of leaves and total dry matter of roots, and were attributed to an air pollution-dependent demand for energy and carbon in leaves. 相似文献

小黑杨是人们以小叶杨和欧洲黑杨为亲本培育出的杂交种,兼具双亲生长速度快、抗性强的优点。本研究通过二代测序技术,对小黑杨进行全基因组测序,初步组装出小黑杨基因组,并以此为基础,识别分析其SSR序列,为小黑杨品种划分、表型性状关联等提供参考。结果表明,共组装得到了366 876条总计368.96 Mbp的contig序列;对其中不小于2 000 bp的21 788条的非冗余contig进行SSR分析,共识别出18 111条SSR序列,其中一、二、三核苷酸重复基序较多。对得到的SSR序列设计引物,共得到12 838对引物,供今后实验使用。 相似文献

A survey of amplified fragment length polymorphism (AFLP) was conducted to examine genetic diversity in 146 individuals of British, native black poplar ( Populus nigra subsp. betulifolia ) and three individuals considered by collectors to be non- betulifolia poplars. Using two primer pairs, a total of 147 bands were detected of which 82 (56%) were polymorphic in at least one individual. Cluster analysis and principal coordinates analysis of the calculated similarity matrix revealed a low level of genetic diversity, although a loose clustering into five groups could be identified, one of which contained the non- betulifolia individuals. Examination of the spatial distribution of the other four groups (all betulifolia ) revealed a general correlation between geographic proximity and genetic similarity. On the basis of the polymorphism observed, it was possible to identify a small number of individual plants which exhibit maximum diversity and might therefore be suitable for inclusion in a replanting programme designed to maintain at least the existing low level of polymorphism observed within British black poplar. 相似文献

以小黑杨(Populus simonii×p.nigra)花药培养植株无菌苗叶片为外植体,通过根癌农杆菌(Agrobacterium tumefaciens)介导法将胆碱氧化酶基因(codA)导入小黑杨中,共获得4株转化株系,PCR扩增和Southern杂交检测结果全部呈阳性,表明codA基因已整合到小黑杨花药培养植株基因组中。荧光定量RT-PCR检测证明,codA基因在小黑杨花药培养植株中获得表达。耐盐实验结果显示,各转基因株系在0.6%的NaCl浓度下能够生长,而非转基因对照小黑杨受盐害严重,说明codA基因的导入提高了转基因植株的耐盐性。 相似文献

As the evolutionary significance of hybridization is largely dictated by its extent beyond the first generation, we broadly surveyed patterns of introgression across a sympatric zone of two native poplars ( Populus balsamifera , Populus deltoides ) in Quebec, Canada within which European exotic Populus nigra and its hybrids have been extensively planted since the 1800s. Single nucleotide polymorphisms (SNPs) that appeared fixed within each species were characterized by DNA-sequencing pools of pure individuals. Thirty-five of these diagnostic SNPs were employed in a high-throughput assay that genotyped 635 trees of different age classes, sampled from 15 sites with various degrees of anthropogenic disturbance. The degree of admixture within sampled trees was then assessed through Bayesian clustering of genotypes. Hybrids were present in seven of the populations, with 2.4% of all sampled trees showing spontaneous admixture. Sites with hybrids were significantly more disturbed than pure stands, while hybrids comprised both immature juveniles and trees of reproductive age. All three possible F1s were detected. Advanced-generation hybrids were consistently biased towards P. balsamifera regardless of whether hybridization had occurred with P. deltoides or P. nigra. Gene exchange between P. deltoides and P. nigra was not detected beyond the F1 generation; however, detection of a trihybrid demonstrates that even this apparent reproductive isolation does not necessarily result in an evolutionary dead end. Collectively, results demonstrate the natural fertility of hybrid poplars and suggest that introduced genes could potentially affect the genetic integrity of native trees, similar to that arising from introgression between natives. 相似文献

The European black poplar (Populus nigra L.) has been used as agermplasm resource for the breeding of new poplar varieties around the world. Theidentification and screening of its high nitrogen use efficiency genotypes couldenable the breeding of new resource-efficient poplar varieties. The accessions werescreened using MALDI-TOF MS genotyping technology for ammoniumtransporter (AMT) and nitrate transporters (NRT) genes against phenotypic data forseedling height and ground diameter traits, in both low and high nitrogenenvironments. Allele re-sequencing of seven genes related to root developmentwas carried out using the minisequencing method. By cluster analysis, 101accessions of black poplar were divided into 4 populations, and it was concludedthat Central Europe is the origin of the evolution of low-nitrogen and highefficiencypopulations of European black poplar. Association study between SNPtyping and seedling height and ground diameter traits showed that there weresignificant correlations between four SNP loci and growth traits under thecontrasting N levels. We found that SNP3 and SNP4 in the PttAMT1;3 gene weresignificantly associated with seedling height traits, and that SNP2 and SNP7 in thePttAMT1;2 and PttAMT1;5 genes, respectively, were significantly associated withground diameter traits. Thus, considerable allelic diversity is present within thecandidate genes studied and can be utilized to develop functional markers to selectfor poplars with improved growth under N stress conditions. 相似文献
