Summary Using fresh frozen (with and without semipermeable membranes), freeze-dried or sections from aldehyde fixed material and hexazotized p-rosaniline for simultaneous coupling more than 20 different unsubstituted or substituted l-amino acid naphthylamides are split especially in the microvilli and/or stereocilia of the small intestine, kidney and epididymis from rats. Further sites of positive reactions can be revealed by l-alanyl, l-leucyl, l-lysyl, ,l-glutamyl, ,l-glutamyl, l-asparaginyl, N-benzoyl-l-arginyl, N-carbobenzoxy-l-arginyl and N-benzoyl-l-phenylalanyl 2-naphthylamide. Among the substituted and unsubstituted peptide 2-naphthylamides l-prolyl-l-arginyl 2-naphthylamide is not hydrolysed in visible amounts; l-arginyl-l-arginyl, l-alanyl-l-arginyl-l-arginyl, l-alanyl-l-leucyl-l-tyrosyl, l-histidyl-l-seryl, l-seryl-l-tyrosyl and l-glycyl-l-phenylalanyl 2-naphthylamide are metabolized in the renal and intestinal brush border; the reaction pattern obtained with N-carbobenzoxy-l-glycyl-l-glycyl-l-arginyl 2-naphthylamide differs from that of N-carbobenzoxy-l-arginyl 2-naphthylamide. In addition l-glycyl-l-prolyl, l-leucyl-l-alanyl, l-lysyl-l-alanyl and l-alanyl-l-phenylalanyl-l-prolyl 2-naphthylamide are also split in the lysosomes of many organs and the secretion granules of gland cells.

Zusammenfassung Mit hexazotiertem p-Rosanilin als Simultankuppler werden in frischen (mit und ohne semipermeable Membranen), gefriergetrockneten und in Schnitten von aldehyd-fixiertem Material mehr als 20 unsubstituierte oder substituierte Aminosäure-2-naphthylamide nahezu ausschließlich in den Mikrovilli und/oder Stereocilien von Dünndarm, Nebenhoden und Niere von Ratten umgesetzt. Unter den Peptid-2-naphthylamiden wird l-Prolyl-l-arginyl-2-naphthylamid nicht in sichtbarer Menge hydrolysiert; l-Alanyl-l-arginyl-l-arginyl-, l-Arginyl-l-arginyl-, l-Asparaginyl-l-arginyl-, l-Alanyl-l-leucyl-l-tyrosyl-, l-Histidyl-l-seryl-, l-Seryl-l-tyrosyl- und l-Glycyl-l-phenylalanyl-2-naphthylamid setzen der renale und intestinale Bürstensaum um. N-Carbobenzoxy-l-glycyl-l-glycyl-l-arginyl-2-naphthylamid liefert verglichen mit Benzoyl- und N-Carbobenzoxy-l-arginyl-2-naphthylamid zusätzliche Resultate. Mit l-Glycyl-l-prolyl-, l-Leucyl-l-alanyl-, l-Lysyl-l-alanyl-und l-Alanyl-l-phenylalanyl-l-prolyl-2-naphthylamid reagieren die Lysosomen zahlreicher Organe und Sekretgranula positiv.

Summary Cell extracts ofCandida guilliermondii grown ind-xylose,l-arabinose,d-galactose,d-glucose,d-mannose and glycerol as sole carbon sources possessed NADPH-dependent aldose reductase activity, but no NADH-dependent activity was detected.d-xylose andl-arabinose were the best inducers of aldose reductase activity. The highest enzyme activity ind-xylose orl-arabinose-grown cells was observed first withl-arabinose followed byd-xylose as substrates of the enzymatic reaction. However, only low activity was found ind-glucose,d-mannose andd-galactose-grown cells, indicating that these carbon sources cause catabolite repression. Enzyme activities induced ind-xylose-grown cells were twice as high as those obtained from the cells under resting conditions. Furthermore, the level of induction of aldose reductase activity depended on the initial concentration ofd-xylose. The present study shows that aldose reductase activity may be efficiently induced by pentose sugars of hemicellulosic hydrolysates and weakly by hemicellulosic hexoses.  相似文献

The ability of Aspergillus fumigatus l-amino acid oxidase (l-aao) to cause the resolution of racemic mixtures of dl-amino acids was investigated with dl-alanine, dl-phenylalanine, dl-tyrosine, and dl-aspartic acid. A chiral column, Crownpak CR+ was used for the analysis of the amino acids. The enzyme was able to cause the resolution of the three dl-amino acids resulting in the production of optically pure d-alanine (100% resolution), d-phenylalanine (80.2%), and d-tyrosine (84.1%), respectively. The optically pure d-amino acids have many uses and thus can be exploited industrially. This is the first report of the use of A. fumigatus l-amino acid oxidase for racemic resolution of dl-amino acids.  相似文献

The phototrophic bacterium Rhodobacter capsulatus E1F1 assimilates ammonia and other forms of reduced nitrogen either through the GS/GOGAT pathway or by the concerted action of l-alanine dehydrogenase and aminotransferases. These routes are light-independent and very responsive to the carbon and nitrogen sources used for cell growth. GS was most active in cells grown on nitrate or l-glutamate as nitrogen sources, whereas it was heavily adenylylated and siginificantly repressed by ammonium, glycine, l-alanine, l-aspartate, l-asparagine and l-glutamine, under which conditions specific aminotransferases were induced. GOGAT activity was kept at constitutive levels in cells grown on l-amino acids as nitrogen sources except on l-glutamine where it was significantly induced during the early phase of growth. In vitro, GOGAT activity was strongly inhibited by l-tyrosine and NADPH. In cells using l-asparagine or l-aspartate as nitrogen source, a concerted induction of l-aspartate aminotransferase and l-asparaginase was observed. Summary L-Phenylalanine is a potent inhibitor of growth in a marine species of blue-green bacteria, Agmenellum quadruplicatum. The growth inhibition is reversed by many amino acids when added to the culture medium simultaneously with L-phenylalanine. The most effective L-phenylalanine antagonists are L-tyrosine, L-alanine, L-leucine, L-methionine, L-tryptophan, and L-isoleucine. However, L-tyrosine is the only effective L-phenylalanine antagonist when growth is inhibited by L-phenylalanine for two or more hours prior to addition of an equimolar concentration of the compound tested as an antagonist. Various explanations that could account for inhibition of growth by L-phenylalanine are discussed. Inhibition of growth by L-phenylalanine is not a feature peculiar to the general physiology of blue-green bacteria. For example, the growth of Anacystis nidulans, a fresh water species, was not inhibited by L-phenylalanine, although a different pattern of metabolite sensitivity was found.  相似文献

A putative ribose-5-phosphate isomerase (RpiB) from Streptococcus pneumoniae was purified with a specific activity of 26.7 U mg−1 by Hi-Trap Q HP anion exchange and Sephacryl S-300 HR 16/60 gel filtration chromatographies. The native enzyme existed as a 96-kDa tetramer with activity maxima at pH 7.5 and 35°C. The RpiB exhibited isomerization activity with l-lyxose, l-talose, d-gulose, d-ribose, l-mannose, d-allose, l-xylulose, l-tagatose, d-sorbose, d-ribulose, l-fructose, and d-psicose and exhibited particularly high activity with l-form monosaccharides such as l-lyxose, l-xylulose, l-talose, and l-tagatose. With l-xylulose (500 g l−1) and l-talose (500 g l−1) substrates, the optimum concentrations of RpiB were 300 and 600 U ml−1, respectively. The enzyme converted 500 g l−1 l-xylulose to 350 g l−1 l-lyxose after 3 h, and yielded 450 g l−1 l-tagatose from 500 g l−1 l-talose after 5 h. These results suggest that RpiB from S. pneumoniae can be employed as a potential producer of l-form monosaccharides.  相似文献

l-Aspartyl l-amino acid methyl ester was synthesized using a mutant of a thermostable leucine aminopeptidase from Streptomyces cinnamoneus, D198 K SSAP, obtained in previously. A peptide of high-intensity sweetener, l-aspartyl-l-phenylalanine methyl ester, was selected as a model for demonstrating the synthesis of l-aspartyl l-amino acid methyl ester. The hydrolytic activities of D198 K SSAP toward l-aspartyl-l-phenylalanine and its methyl ester were, respectively, 74-fold and fourfold higher than those of wild type. Similarly, the initial rate of the enzyme for l-aspartyl-l-phenylalanine methyl ester synthesis was over fivefold higher than that of wild-type SSAP in 90% methanol (v/v) in a one-pot reaction. Furthermore, other l-aspartyl l-amino acid methyl esters were synthesized efficiently using D198 K SSAP. Results show that the substitution of Asp198 of SSAP with Lys is effective for synthesizing l-aspartyl l-amino acid methyl ester.  相似文献

Summary. To mutant ddY/DAO mice lacking D-amino-acid oxidase activity and normal ddY/DAO+ mice, five D-amino acids (D-Asp, D-Ser, D-Ala, D-Leu and D-Pro) were orally administered for two weeks, and the D-amino acid levels were examined in seven brain regions. The levels of D-Asp markedly increased in the pituitary and pineal glands in both strains. In the ddY/DAO+ mice, the levels of the other D-amino acids did not significantly change in most of the brain regions. While in the ddY/DAO mice the levels of D-Ser significantly increased in most of the brain regions except for the cerebrum and hippocampus. The levels of D-Ala and D-Leu increased in all regions but the levels of D-Pro did not significantly change. The same five D-amino acids were intravenously injected into Wistar rats and the D-amino acid levels in their brains were examined for 60 min after the administration. The levels of D-Asp markedly increased in the pineal gland 3 min after the administration, while the levels of D-Ser, D-Ala, and D-Pro increased both in the pineal and pituitary glands, the levels of D-Leu increased in all brain regions. These results are useful for the elucidation of the origins and regulation of D-amino acids in the mammalian body.  相似文献

Summary l-Threonine hyper-producing mutants were obtained fromEscherichia coli W strain KY-8366, by reducingl-threonine degradation activity and enhancingl-threonine biosynthetic activity. Anl-threonine degradation reaction test using resting cells of KY-8366 suggested that the main pathway ofl-threonine degradation by KY-8366 is via glycine. A strain with reducedl-threonine degradation activity was obtained among those mutants that could not utilizel-threonine as sole nitrogen source. Rifampicin-resistant mutants andl-lysine plus methionine-insentitive mutants were isolated. These mutants showed enhanced aspartokinase levels and accumulated morel-threonine than the parental strains. Mutant H-4290 accumulated 58 g/l ofl-threonine.  相似文献

N-Acyl-D-glutamate amidohydrolase (D-AGase) was inhibited by 94 % when 1 mol/l N-acetyl-DL- glutamate was used as a substrate. The addition of 1 mM Co2+ stabilized D-AGase. Moreover, the substrate inhibition was weakened to 88% with the addition of 0.4 mM Co2+ to the reaction mixture. Although D-AGase is a zinc-metalloenzyme, the addition of Zn2+ from 0.01 to 10 mM did not increase the D-glutamic acid production in the saturated substrate. Under optimal conditions, 0.38 M D-glutamic acid was obtained from N-acyl-DL-glutamate with 100% of the theoretical yield after 48 h.  相似文献

l-Arabinose is the second most abundant pentose beside d-xylose and is found in the plant polysaccharides, hemicellulose and pectin. The need to find renewable carbon and energy sources has accelerated research to investigate the potential of l-arabinose for the development and production of biofuels and other bioproducts. Fungi produce a number of extracellular arabinanases, including α-l-arabinofuranosidases and endo-arabinanases, to specifically release l-arabinose from the plant polymers. Following uptake of l-arabinose, its intracellular catabolism follows a four-step alternating reduction and oxidation path, which is concluded by a phosphorylation, resulting in d-xylulose 5-phosphate, an intermediate of the pentose phosphate pathway. The genes and encoding enzymes l-arabinose reductase, l-arabinitol dehydrogenase, l-xylulose reductase, xylitol dehydrogenase, and xylulokinase of this pathway were mainly characterized in the two biotechnological important fungi Aspergillus niger and Trichoderma reesei. Analysis of the components of the l-arabinose pathway revealed a number of specific adaptations in the enzymatic and regulatory machinery towards the utilization of l-arabinose. Further genetic and biochemical analysis provided evidence that l-arabinose and the interconnected d-xylose pathway are also involved in the oxidoreductive degradation of the hexose d-galactose.  相似文献
