At Helgoland, in the North Sea, growth of the high sublittoral brown macroalga Dictyota dichotoma (Hudson) Lamoroux was examined in October (the time of tetraspore release) in an outdoor tank by exposing 2-day-old germlings to four solar radiation treatments achieved with different filter materials or an additional artificial light source: photosynthetically active radiation (PAR; 395–700 nm), PAR plus ultraviolet (UV)-A (320–700 nm), full solar spectrum, or solar spectrum plus artificial UV radiation (UVR). Based on length measurements over a period of 3 weeks, the growth rate in germlings strongly decreased in conditions with UVR compared to PAR: by 14% under PAR+UV-A, by 31% under the full solar spectrum and by 65% with additional UVR. Although growth rates of germlings under UVR were reduced mainly in the first week, the plants did not regain the size of the untreated plants even after 9 weeks. Regardless of the exposure, no defects in morphology or anatomy including the exposed apical meristem were detected, except for a reduction in cell division rates perhaps due to additional cost for photoprotective or repair mechanisms. Depending on the actual position of D. dichotoma plants in the natural habitat, individuals in high positions receive substantial amounts of the more harmful UV-B while those lower down might only receive UV-A during part of the day, thus the effect of UV-B on the growth of D. dichotoma will depend on its position in the field. The effects of tidal variation of the light climate and the implications of our results for the zonation of D. dichotoma are discussed. Guinea pig cochleae were perfused with high-potassium solutions to depolarize hair cells artificially and induce the release of afferent neurotransmitter. Sequential injections of artificial perilymph containing 5 mM KCl, then 50 mM KCl, and finally 5 mM KCl were made into the scala tympani. This injection sequence was conducted under either normal divalent-cation conditions (2.0 mM CaCl2, 1.0 mM MgCl2) or calcium-deficient conditions intended to antagonize evoked transmitter release (0.1 mM CaCl2, 20.0 mM MgCl2). The levels of 21 endogenous primary amines in effluent collected from the scala vestibuli were determined by gradient-elution, reverse-phase HPLC using o-phthaldialdehyde-thiol adducts with fluorescence detection. Analyses indicated effluent concentrations of glutamate, taurine, and a coeluting taurine-gamma-aminobutyrate (GABA) fraction (but not GABA alone) increased significantly after exposure to 50 mM KC1 and returned to baseline levels after reintroduction of 5 mM KC1 under normal divalent-cation conditions. Correspondent changes in the release of these constituents were significantly attenuated under calcium-deficient conditions. This was not the case for potassium-induced changes in the release of arginine, aspartate, and isoleucine. These data are consistent with the hypothesis that the receptoneuronal transmitter is glutamate and further suggest a calcium-dependent mechanism involving taurine.  相似文献

The mutagenic effects of ultraviolet and solar irradiation are thought to be due to the formation of DNA photoproducts, most notably cyclobutane pyrimidine dimers (CPDs) and pyrimidine (6-4) pyrimidone photoproducts ((6-4)PPs). Experimental systems for determining the levels and sequence dependence of photoproduct formation in DNA have often used high doses of short-wave (UVC) irradiation. We have re-assessed this issue by using DNA sequencing technologies and different doses of UVC as well as more physiologically relevant doses of solar irradiation emitted from a solar UV simulator. It has been questioned whether hot alkali treatment can detect (6-4)PPs at all sequence positions. With high UVC doses, the sequence distribution of (6-4)PPs was virtually identical when hot alkali or UV damage endonuclease (UVDE) were used for detection, which appears to validate both methods. The (6-4)PPs form at 5'-TpC and 5'CpC sequences but very low levels are seen at all other dipyrimidines including 5'-TpT. Contrary to expectation, we find that (6-4) photoproducts form at almost undetectable levels under conditions of irradiation for up to five hours with the solar UV simulator. The same treatment produces high levels of CPDs. In addition, DNA glycosylases, which recognize oxidized and ring-opened bases, did not produce significant cleavage of sunlight-irradiated DNA. From these data, we conclude that cyclobutane pyrimidine dimers are at least 20 to 40 times more frequent than any other DNA photoproduct when DNA or cells are irradiated with simulated sunlight.  相似文献

Broadcast spawning corals release gametes into the oceans with extraordinarily accurate timing. While the date of spawning is set by the lunar cycle, the hour/minute of spawning is set by the solar cycle. In this report, we describe experiments that test whether the time of spawning is regulated by an entrained biological clock or whether it is directly controlled by the solar cycle. Montastraea franksi samples were collected on the morning of the predicted spawning. Fragments from colonies were kept under three different lighting conditions and spawning monitored. The three conditions were sunset times of 0, 1 or 2 h earlier than normal. Fragments from the same colony spawned differently under these three conditions, with an early sunset causing a corresponding early shift in spawning. These results indicate that spawn timing is not controlled by a circadian rhythm and that it is directly controlled by local solar light cycle.  相似文献

Parietal cells are a major source of gastric mucosal prostaglandins in various species. We examined cholinergic stimulation of prostaglandin E2 (PGE2) release from human parietal cells; using activators of the protein kinase C we attempted to get an indirect insight into cellular mechanisms which control PGE2 release. Gastric mucosal specimens were obtained at surgery and the cells were dispersed by collagenase and pronase E. Parietal cells were enriched to 65-80% by a Percoll gradient, and were incubated for 30 min. PGE2 release into the medium (radioimmunoassay) was 74-126 pg/10(6) cells/30 min under basal conditions and was 2.6-fold increased by carbachol (10(-5) and 10(-4) M). Similarly, PGE2 release was stimulated by phospholipase C (20-200 mU/ml, 364% above basal), 1-oleoyl-2-acetyl-sn-glycerol (10(-9)-10(-5) M, 229%), 12-O-tetradecanoylphorbol-13-acetate (TPA; 10(-9)-10(-5) M, 283%) and calcium ionophore A23187 (10(-7)-10(-5) M, 219%). Simultaneous presence of A23187 and TPA synergistically induced stimulation which was slightly higher than the sum of the individual responses. N-(6-aminohexyl)-5-chloro-1-naphthalene sulfonamide W-7, a putative calmodulin antagonist, inhibited TPA-induced PGE2 release at concentrations regarded specific for blocking calmodulin (IC50 = 1.5 X 0(-6) M). We conclude that in human parietal cells PGE2 is released upon cholinergic stimulation and that phospholipase C and protein kinase C are involved in the control of PGE2 release. We speculate that calmodulin might interact with a protein phosphorylated by protein kinase C to cause PGE2 release.  相似文献

Three field manipulation experiments were carried out during 1993–1995 on the Northern Pennines to investigate the influences of temperature, solar radiation and rainfall on the release of dissolved organic carbon (DOC) from vegetated soil cores using zero‐tension lysimeters. The cores were manipulated by being translocated to four sites down a climatic gradient, by artificial soil heating or being exposed to double normal rainfall. In each experiment three soil types, a brown earth, a micropodzol and a peaty gley, with differing organic matter content and distribution within the profile, were studied. DOC data, expressed as mg C m?2 day?1, were analysed following log10 transformation, by a repeated measures analysis of variance procedure, using climatic variables measured concurrently with sampling, and 1 and 2 months before sampling. DOC release was dominated by rainfall but was also associated with solar radiation and temperature. With each of the three climatic variables, rainfall, solar radiation and temperature, both positive and negative effects on DOC release have been found significant, indicating that the concurrent and delayed effects of the same variable may be different. DOC release was positively related with all three soils to concurrent rainfall, indicating rainfall's primary leaching action. DOC release was also negatively related to rainfall of the previous month indicating that its action depletes the leachable pool of DOC in the soil. DOC release was positively associated with solar radiation 2 months earlier, indicating that DOC's main source is that of primary production; DOC peaks closely followed peaks of annual primary production. DOC release was linked with temperature, the strongest association being with temperature 2 months earlier, indicating that temperature effects both primary production and DOC regeneration via organic matter decomposition. A conceptual model, relating our findings to those processes known to govern DOC release from soils, has been presented.  相似文献

Colorless phenylpropanoid derivatives are known to protect plants from ultraviolet (UV) radiation, but their photoregulation and physiological roles under field conditions have not been investigated in detail. Here we describe a fast method to estimate the degree of UV penetration into photosynthetic tissue, which is based on chlorophyll fluorescence imaging. In Arabidopsis this technique clearly separated the UV-hypersensitive transparent testa (tt) tt5 and tt6 mutants from the wild type (WT) and tt3, tt4, and tt7 mutants. In field-grown soybean (Glycine max), we found significant differences in UV penetration among cultivars with different levels of leaf phenolics, and between plants grown under contrasting levels of solar UV-B. The reduction in UV penetration induced by ambient UV-B had direct implications for DNA integrity in the underlying leaf tissue; thus, the number of cyclobutane pyrimidine dimers caused by a short exposure to solar UV-B was much larger in leaves with high UV transmittance than in leaves pretreated with solar UV-B to increase the content phenylpropanoids. 由大气臭氧层减薄导致的UV-B辐射变化将直接影响到凋落物的分解。目前,有关UV-B辐射影响木本植物凋落物分解的研究还很少,在国内还没有开展。采用分解袋法开展了马尾松凋落叶在自然环境和UV-B辐射滤减两种辐射环境下的分解试验。结果表明:在UV-B辐射滤减环境下的马尾松凋落叶年分解速率比对照环境减慢了47.74%。UV-B辐射极显著(p<0.01)地加快了马尾松凋落叶的分解速率,促进了凋落叶中碳、磷、钾的释放和木质素的降解,对氮的释放无明显影响。研究结果意味着UV-B辐射将加快马尾松林的营养循环速度,降低马尾松林凋落物层的碳储量。  相似文献

Controllable storage and release of solar energy has always been a highlighted scientific issue for its benefit of mankind. Solar thermal fuels (STFs) supply a closed cycle and renewable energy‐storage strategy by transforming solar energy into chemical energy stored in the conformation of molecular isomers, such as cis/trans‐azobenzene, and releasing it as heat under various stimuli. Although the potential high energy density of the STFs which are based on the hybrids of azobenzene derivatives and carbon nanomaterials has been reported the solvent‐assistant charging hinders their practicability. In this study, a solid‐state STF device is designed and fabricated by compositing one photoliquefiable azobenzene (PLAZ) derivative with a flexible fabric template. The photoinduced phase transition of the PLAZ derivative enables the charging of the flexible STFs to be totally solvent‐free. Interestingly, the energy‐storage capacity (energy density ≈201 J g?1) of flexible PLAZ STFs has been improved by the soft fabric template. The exothermic situation is monitored with one infrared camera, which shows 4 °C temperature difference between charged and discharged samples under blue light stimulus. The flexible STFs are may be used in practice as heating equipment.  相似文献

Abstract: In vivo microdialysis was used to determine whether the 3,4-methylenedioxymethamphetamine (MDMA)-induced release of serotonin (5-HT) in vivo involves a carrier-mediated process and to investigate further the state-dependent interaction between 5-HT and dopamine. MDMA produced a dose-dependent increase in the extracellular concentration of 5-HT in the striatum and prefrontal cortex that was attenuated by treatment with fluoxetine but not by tetrodotoxin. Suppression by fluoxetine of the MDMA-induced release of 5-HT was accompanied by a suppression of the MDMA-induced release of dopamine. Administration of MDMA to rats treated with carbidopa and l -5-hydroxytryptophan resulted in a synergistic elevation of the extracellular concentration of 5-HT that was much greater than that produced by either treatment alone. The MDMA-induced release of dopamine by MDMA also was potentiated in 5-hydroxytryptophan-treated rats. These data are consistent with the view that MDMA increases the extracellular concentration of 5-HT by facilitating carrier-mediated 5-HT release, which can be enhanced greatly under conditions in which 5-HT synthesis is stimulated. Moreover, these data are supportive of a state-dependent, stimulatory role of 5-HT in the regulation of dopamine release.  相似文献

The decomposition of dissolved organic matter (DOM) in pelagic ecosystems is mediated primarily by heterotrophic bacteria, but transformation by short-wave solar radiation may play an important role in surface waters, in particular when humic substances constitute a substantial fraction of the DOM pool. Most of the studies examining bacterial decomposition and photochemical transformation of DOM stem from limnetic and coastal marine systems and much less information is available from oceanic environments. To examine the bacterial decomposition of humic and non-humic DOM in the Southern Ocean we carried out microcosm experiments in which we measured bacterial growth on isolated fractions of humic and non-humic DOM of the size classes <3 kDa and >3 kDa. Experiments carried out at the Polar Front showed a preferential bacterial growth on non-humic DOM and in particular on the size fraction <3 kDa. Bacterial growth, measured as bacterial biomass production, on non-humic DOM accounted for 74% to 88% of the total growth on all four DOM fractions. In experiments in the Antarctic circumpolar current and the coastal current under pack ice, bacterial growth was 6× lower than at the Polar Front, and humic and non-humic DOM was consumed to equal amounts. The size fraction <3 kDa was always preferred. Experiments examining the effect of solar radiation on the release of dissolved amino acids (DAA) and carbohydrates (DCHO) and their subsequent bacterial utilization showed a stimulating effect on glucose uptake and the release of DAA at the Polar Front but an inhibition in the eastern Weddell Sea. Ultraviolet-B was the most effective component of the solar radiation spectrum tested. Effects of UV-B on glucose uptake and release of DAA were positively correlated with concentrations of humic-bound DAA. The data imply that at low concentrations, e.g., <100 nM (amino acid equivalent), UV-irradiation reduces, whereas at concentrations >100 nM UV-irradiation stimulates glucose uptake and release of DAA as compared to dark conditions.  相似文献

Photovoltaic solar panels represent one of the most promising renewable energy sources, but are strong reflectors of horizontally polarized light. Polarized light pollution (PLP) associated with solar panels causes aquatic insects to prefer to oviposit on panels over natural water bodies, with potential to negatively impact their global populations as solar energy expands. We evaluate the hypothesis that anti-reflective coatings (ARCs) used to increase the energy efficiency of solar panels will reduce the amount of PLP they reflect, and their attractiveness to aquatic insects. We created artificial test surfaces that mimicked the optical properties of coated and uncoated solar panels and exposed them to wild populations of polarotactic mayflies (Ephemeroptera), horseflies (Tabanidae) and non-biting midges (Chironomidae) used as indicators of PLP. We evaluated the reflection-polarization properties of test surfaces from four different angles of view and under sunny and overcast skies in the visible and ultraviolet parts of the spectrum. Matte (i.e. ARC-coated) sunlit solar panels were strong sources of horizontally polarized light only when the sun was afront and behind, in contrast to uncoated panels which exceeded common polarization-sensitivity thresholds for aquatic insects from all four viewing directions. As predicted by these polarization patterns, horsefly numbers and water-seeking behaviors were significantly reduced by ARCs. Under overcast skies, both matte and shiny (i.e. uncoated) panels were insect-detectible sources of PLP. Matteness modestly reduced the degree of polarization of reflected light, but not sufficiently such that fewer chironomids were attracted to them. Mayflies actually preferred matte panels under overcast skies. ARCs are most likely to reduce PLP and benefit aquatic insects under sunny skies and when used in conjunction with white non-polarizing gridding, but may actually exacerbate the severity of their negative effects under overcast conditions. Consequently, even current ARC technology has a role to play in aquatic insect conservation, but strategic deployment of solar panels away from water bodies and temperate regions may trump these benefits.  相似文献

Recent studies showed that photochemical breakdown (photodegradation) of plant material accounts for a substantial portion of litter decomposition and subsequent trace gas release in ecosystems under high radiative load and low precipitation. In the absence of solar radiation, thermal degradation may also cause trace gas release at temperatures below the ignition point. These observations suggest that the abiotic processes of photodegradation and thermal degradation of plant litter may be important in understanding global trace gas budgets. In a laboratory incubation study, we performed a simultaneous carbon (C) accounting of CO2, CO, and CH4 produced as a byproduct of photodegradation and thermal degradation of six different plant litter types that varied in chemical composition. The patterns of trace gas release during photodegradation and thermal degradation differed considerably across the six plant materials, suggesting that chemical composition of litter may influence the rates of abiotic degradation. There was a strong positive correlation between the rates of trace gas release during photodegradation and temperature. A significant portion of trace gases were produced during low temperature (< 100 °C) thermal degradation of litter in the absence of solar radiation, which was also positively correlated to temperature. In addition, both thermal degradation and photodegradation occurred in the absence of O2. This indicates that the mechanism formerly accepted as photo‐oxidation may only be one of several photodegradation processes. We speculate that the direct breakdown of chemical groups such as carboxyl, carbonyl, and methoxyl groups may result in CO2, CO, and CH4 release. We suggest that the combined processes of thermal and photodegradation of litter may be a previously under accounted source of C‐based trace gases from terrestrial systems.  相似文献

The involvement of glutamate receptors in GABA release in ischemia was investigated in hippocampal slices from adult (3-month-old) and developing (7-day-old) mice. For in vitro ischemia, the slices were superfused in glucose-free media under nitrogen. Ionotropic glutamate receptor agonists failed to affect the ischemia-induced basal GABA release at either age. The K(+)-stimulated release in the immature hippocampus was potentiated by N-methyl-D-aspartate receptors, whereas in adults this release was reduced by both kainate and 2-amino-3-hydroxy-5-methyl-4-isoxazoleproprionate receptor activation. The group I metabotropic receptor agonist (1+/-)-1-aminocyclopentane-trans-1,3-dicarboxylate enhanced the basal ischemic GABA release in a receptor-mediated manner in adults, this being concordant with the positive modulation of GABAergic neurotransmission by group I metabotropic glutamate receptors. (1 +/-)-1-Aminocyclopentane-trans-1,3-dicarboxylate and (S)-3,5-dihydroxyphenylglycine also enhanced the K(+)-stimulated release in the developing hippocampus in a receptor-mediated manner. Because group I receptors generally increase neuronal excitability, the enhanced GABA release may attenuate hyperexcitation or strengthen inhibition, being thus neuroprotective, particularly under ischemic conditions. Group III metabotropic glutamate receptors were not at all involved in ischemic GABA release in the immature mice, but in adults their activation by O-phospho-L-serine potentiated the basal release and reduced the K(+)-stimulated release. These opposite effects were abolished by the antagonist (RS)-2-cyclopropyl-4-phosphonophenylglycine. Metabotropic glutamate receptors, namely group I and III receptors, are able to modify the release of GABA from hippocampal slices under ischemic conditions, both positive and negative effects being discernible, depending on the age and type of receptor activated.  相似文献

At higher latitudes, most organisms use the periodic changes in day length to time their annual life cycle. At the equator, changes in day length are minimal, and it is unknown which cues organisms use to synchronize their underlying circannual rhythms to environmental conditions. Here, we demonstrate that the African stonechat (Saxicola torquatus axillaris)-an equatorial songbird-can use subtle solar cues for the annual timing of postnuptial moult, a reliable marker of the circannual cycle. We compared four groups that were kept over more than 3 years: (i) a control group maintained under constant equatorial day length, (ii) a 12-month solar time group maintained under equatorial day length, but including a simulation of the annual periodic change in sunrise and sunset times (solar time), (iii) a 14-month solar time group similar to the previous group but with an extended solar time cycle and (iv) a group maintained under a European temperate photoperiod. Within all 3 years, 12-month solar time birds were significantly more synchronized than controls and 14-month solar time birds. Furthermore, the moult of 12-month solar time birds occurred during the same time of the year as that of free-living Kenyan conspecifics. Thus, our data indicate that stonechats may use the subtle periodic pattern of sunrise and sunset at the equator to synchronize their circannual clock.  相似文献

BACKGROUND AND AIMS: Hard-seeded (physical) dormancy is common among plants, yet mechanisms for dormancy release are poorly understood, especially in the tropics. The following questions are asked: (a) whether dormancy release in seed banks of the tropical shrub Parkinsonia aculeata (Caesalpiniaceae) is determined by wet heat (incubation under wet, warm to hot, conditions); and (b) whether its effect is modified by microclimate. METHODS: A seed burial trial was conducted in the wet-dry tropics (northern Australia) to compare dormancy release across different habitats (open, artificial cover, ground cover and canopy cover), burial depths (0, 3 and 20 cm) and burial durations (2, 6 and 14 weeks). Results were compared with predictions using a laboratory-derived relationship between wet heat and dormancy release, and microclimate data collected during the trial. KEY RESULTS: Wet heat (defined as the soil temperature above which seeds were exposed to field capacity or higher for a cumulative total of 24 h) was 43.6 degrees C in the 0 cm open treatment, and decreased with increasing shade and depth to 29.5 degrees C at 20 cm under canopy cover. The dormancy release model showed that wet heat was a good predictor of the proportion of seeds remaining dormant. Furthermore, dormancy release was particularly sensitive to wet heat across the temperature range encountered across treatments. This resulted in a 16-fold difference in dormancy levels between open (<5 % of seeds still dormant) and covered (82 %) microhabitats. CONCLUSIONS: These results demonstrate that wet heat is the principal dormancy release mechanism for P. aculeata when conditions are hot and wet. They also highlight the potential importance of microclimate in driving the population dynamics of such species.  相似文献

The entomopathogenic fungus, Beauveria bassiana, is highly susceptible to the damaging effects of solar radiation. This study demonstrates protection from simulated solar radiation by coating B. bassiana (GHA) spores with lignin and effects on pathogenicity to Lygus lineolaris (Palisot de Beauvois) adults in direct spray and contact uptake bioassays. Spores were coated with either lignin or cross-linked lignin by spray drying to produce spore coatings of high and low water solubility, respectively. Non-coated spores and the two spore coating treatments were suspended in either water (0.04% Silwet L77) or oil (Orchex 692) to produce six formulations. Rates of loss in spore viability under simulated solar radiation were approximately ten times lower for the three formulations in which spores remained coated in suspension (cross-linked lignin in water, cross-linked lignin in oil, and lignin in oil). However, these three formulations were the least pathogenic. Estimates of LC50 and LC75 values for the most pathogenic formulation (non-coated in oil) were 5 and 12 times lower, respectively, than the least pathogenic formulation (cross-linked lignin in oil), but these differences were not significant. The three lignin-coated formulations that provided the greatest UV protection were also the least pathogenic based on LT values, which were significant. Overall, L. lineolaris mortality was approximately 80 times less when exposed to treated broccoli rather than sprayed directly, based on LC50 values. If the contribution of spore uptake from plant surfaces to L. lineolaris mortality is similarly low under field conditions, then improving persistence may be less important for improving efficacy. However, under control conditions where solar radiation significantly impacts mycoinsecticide efficacy, the improved persistence of lignin coating formulations may outweigh negative effects on pathogenicity.  相似文献

研究了日光温室连作黄瓜土壤酚类物质变化及其对黄瓜根系抗病性相关酶的影响.结果表明,随着连作年限的增加,对羟基苯甲酸、阿魏酸、苯甲酸以及总量呈现明显的积累特征:种植5年、7年、9年的含量显著高于1年、3年,且9年的是1年的2倍.在低浓度处理下(40和80 μg·g-1),前期(2、3叶期)诱导多酚氧化酶、过氧化物酶、苯丙氨酸转氨酶活性不同程度地升高,植株表现高抗性,在较高浓度下(120和160 μg·g-1),处理前期多酚氧化酶、过氧化物酶、苯丙氨酸转氨酶活性迅速降低,甚至丧失酶活性.连作土壤酚类物质浓度变化是影响日光温室黄瓜抗病性的主要因素之一.  相似文献

Insulin-like growth factor-II (IGF-II) is an autocrine modulator of epiphyseal chondrogenesis in the fetus. The cellular availability of IGFs are influenced by the IGF-binding proteins (IGFBPs). In this study, we investigated the control of expression and release of IGFBPs from isolated epiphyseal growth plate chondrocytes from the ovine fetus by hormones and growth factors implicated in the chondrogenic process. Chondrocytes were isolated from the proliferative zone of the fetal ovine proximal tibial growth plate and maintained in monolayer culture at early passage number. Culture media conditioned by chondrocytes under basal conditions released IGFBPs of 24, 34, and 29 kDa, and a less abundant species of 39-43 kDa that were identified immunologically as IGFBP-4, IGFBP-2, IGFBP-5, and IGFBP-3, respectively. Messenger RNAs encoding each species were identified by Northern blot analysis within chondrocytes, as was mRNA encoding IGFBP-6. Exposure to IGF-I or IGF-II (13 or 26 nM) caused an increase in expression and release of IGFBP-3. The release of IGFBP-2 and IGFBP-5 were also potentiated without changes to steady state mRNA, and for IGFBP-5 this was due in part to a release from the cell membrane in the presence of IGF-II. Insulin (16.7 or 167 nM) selectively increased mRNA and the release of IGFBP-3, while cortisol (1 or 5 microM) inhibited both mRNA and release of IGFBP-2 and IGFBP-5. Transforming growth factor-beta1 (TGF-beta1) (0.1 or 0.2 nM) increased the expression and release of IGFBP-3, and caused an increase in mRNAs encoding IGFBP-2 and IGFBP-5. Neither growth hormone (GH), fibroblast growth factor-2, nor thyroxine (T(4)) had any effect on IGFBP expression or release. The results suggest that IGFBP expression and release within the developing growth plate can be modulated by IGF-II and other trophic factors, thus controlling IGF availability and action.  相似文献

In its role as a mobile receptor for peroxisomal matrix cargo containing a peroxisomal targeting signal called PTS1, the protein Pex5 shuttles between the cytosol and the peroxisome lumen. Pex5 binds PTS1 proteins in the cytosol via its C-terminal tetratricopeptide domains and delivers them to the peroxisome lumen, where the receptor·cargo complex dissociates. The cargo-free receptor is exported to the cytosol for another round of import. How cargo release and receptor recycling are regulated is poorly understood. We found that Pex5 functions as a dimer/oligomer and that its protein interactions with itself (homo-oligomeric) and with Pex8 (hetero-oligomeric) control the binding and release of cargo proteins. These interactions are controlled by a redox-sensitive amino acid, cysteine 10 of Pex5, which is essential for the formation of disulfide bond-linked Pex5 forms, for high affinity cargo binding, and for receptor recycling. Disulfide bond-linked Pex5 showed the highest affinity for PTS1 cargo. Upon reduction of the disulfide bond by dithiothreitol, Pex5 transitioned to a noncovalent dimer, concomitant with the partial release of PTS1 cargo. Additionally, dissipation of the redox balance between the cytosol and the peroxisome lumen caused an import defect. A hetero-oligomeric interaction between the N-terminal domain (amino acids 1–110) of Pex5 and a conserved motif at the C terminus of Pex8 further facilitates cargo release, but only under reducing conditions. This interaction is also important for the release of PTS1 proteins. We suggest a redox-regulated model for Pex5 function during the peroxisomal matrix protein import cycle.  相似文献
